The influence on embryonic development of nicotine and caffeine at dose levels approximating human consumption was investigated in Sprague-Dawley rats. One group of animals received nicotine administered subcutaneously by an Alzet mini-osmotic pump from gestational day 6 through 12 (25 mg over 7 days; rate 149 micrograms/hr). Control animals received physiological saline in a similar manner. A second group received a single intravenous injection of caffeine (25 mg/kg) on gestational day 6. Control animals were treated with physiological saline. A further group received both nicotine and caffeine on gestational day 6 as described for the two previous groups. There were no significant differences among any of the groups with respect to maternal weight gain, litter size, embryolethality, fetal weight, or crown-rump length. The offspring of nicotine treated animals showed a significantly higher incidence of hydrocephaly when compared to the controls, but in the combined treatment group no malformed fetuses were observed. Light microscopic examination of maternal liver, kidney and placentas revealed changes in the hepatic sinusoids, glomeruli and intervillous spaces after nicotine and combined treatment. In addition, the decidua basalis was poorly developed compared to the controls. Chorionic villi and fetal kidney appeared normal in all groups. A coteratogenic effect is not evident from these findings.  相似文献

BACKGROUND : Scutellaria baicalensis is a perennial herbaceous plant widely distributed in Oriental areas. Its roots, a commonly used medicinal source, reputedly calm fetuses in pregnant women; however, there is no sufficient evidence to date to assess its safety during pregnancy. This study aims to evaluate the effects of S. baicalensis aqueous extract on embryonic development in ICR mice. METHODS : Aqueous extract of S. baicalensis roots was prepared in accordance with clinical application. Pregnant mice were randomly divided into four groups, i.e., mice treated by gavage with water as negative control, with aqueous extract of 2 (1.8 times of human daily dose), 8 or 32 g/kg/day from gestation day (Gd) 6 to 15 as low-, middle-, and high-dose groups, respectively. The parameters of live and dead fetuses, resorptions, external and skeletal malformed fetuses, maternal body weight, maternal liver, kidneys, and heart weights were evaluated on Gd 18. RESULTS : There was no significant difference in fetal parameters among four groups. Maternal absolute liver and kidneys weights in the high-dose group were significantly higher than those in negative control (p<0.05). Relative liver and kidneys weights in this group were significantly higher than those in any other group (p<0.05). CONCLUSIONS : Oral administration of aqueous extract of S. baicalensis roots at or below 32 g/kg/day to ICR mice during organogenesis did not cause significant fetal external or skeletal malformations. 龟足(Capitulum mitella Linnaeus)在我国主要分布于长江口以南海浪剧烈冲击的暴露型岩相海岸的中、高潮区,是一种颇具养殖潜力和市场前景的新品种。研究温度(24、27、30、33℃)和盐度(28,31,34)对龟足胚胎发育和幼虫生长的协同影响,可为龟足的人工育苗提供依据。结果如下(1):33℃-28温盐度组合胚胎发育时间最短144h,27℃-28温盐度组合胚胎相对孵化率最高。温度与盐度对胚胎发育时间没有显著影响;但温度和盐度对胚胎孵化率有极显著影响,温度与盐度间的交互作用显著。胚胎发育最适宜的温盐度组合是27℃-28。(2):27℃的3个盐度组、30℃-31温盐度组合无节幼虫持续时间最短。在同一盐度条件下以27℃的存活率较高,在同一温度条件下以盐度31的存活率较高,其中以27℃-31温盐度组合的存活率最高;存活率1和存活率2分别高达99.0%、90.7%。27℃-28、27℃-31温盐度组合变态率最高,变态率分别为81.8%、73.7%。34高盐组幼虫的存活率和变态率均很低甚至为零。温度和盐度对幼虫存活率和变态率有极显著影响,两者的交互作用极为显著。综合无节幼虫持续时间、存活和变态情况,27℃-31温盐度组合为幼虫生长发育的最佳组合条件。龟足胚胎发育、无节幼虫的生长和变态对温度盐度的敏感性有所不同,这是由龟足的自身繁殖特点及生活环境决定的。  相似文献

Atherosclerosis is one of the diseases related to metabolic syndrome which is caused by obesity. Previous reports have shown that green tea and its components have anti-obesity effect. We examined whether catechins and caffeine can prevent the development of atherosclerosis by oral administration, singly or in combination to the atherosclerosis model mice. Results demonstrated that the number of atherosclerotic regions in the aorta was significantly reduced by the combined treatment, and the atherosclerotic area was also improved. Serum HDL-C increased by caffeine single treatment, but no effect on the TG and TC by any treatments. Moreover, ECG illuviated to atheromatous lesions in aorta and the illuviation was enhanced by caffeine. The mRNA expression levels of LOX-1 and TNF-α showed a tendency to suppress by the combined treatment. These results indicated that the combined administration of catechins and caffeine has the inhibitory effect on the development of atherosclerosis in mice.  相似文献

Administration of nickel chloride to the pregnant mice on the seventh to eleventh day of their gestational period, resulted in significant embryotoxic effects in terms of an increased resorption rate, a decreased fetal weight, delay in skeletal ossification and high incidence of malformation. Among the cases of fetal malformation, the following malformations were observed to occur at a higher rate of incidence: acephalia, exencephaly, cerebral hernia, open eyelid, cleft palate, micromelia, ankylosis of the extremity, club foot and skeletal anomalies. Most skeletal anomalies were in the form of vertebral and/or rib fusions and were found mostly at thoracic and lumbar levels. The concentration of nickel retained in embryonic tissues was 800 times higher in the exposed compared to control groups and indicated that increased tissue levels of nickel chloride had a toxic influence on the developing embryo.  相似文献

Teratogenic to subteratogenic doses of x-ray, mitomycin C, MNNG, thio-TEPA, cyclophosphamide, and chlorambucil were administered to pregnant ICR mice together with caffeine at doses of 12.5, 25, or 50 mg/kg on day 11 of gestation. Fetuses were examined for gross malformations on day 18 of gestation. The teratogenicity of mitomycin C was significantly potentiated by caffeine at a dose as low as 12.5 mg/kg. The teratogenicity of chlorambucil was also significantly potentiated by caffeine at 50 mg/kg, but similar potentiation was not observed for x-ray, MNNG, thio-TEPA, and cyclophosphamide.  相似文献

The influence of both alcohol and caffeine on early embryonic development was investigated in pregnant rats. Compared to the corresponding controls, a high incidence of resorptions and abnormal embryos was induced following treatment of the animals with alcohol (0.015 ml/g body weight, 12.5% v/v, i.p.) on gestational days 6 through 12 and with caffeine (25 mg/kg body weight, i.v.) on gestational day 10. In addition, embryonic growth was severely affected. Reduction of placental blood circulation and impairment of cellular proliferation may account for the observed deleterious effects on the embryo.  相似文献

It is important to evaluate the health effects of low-dose-rate or low-dose radiation in combination with chemicals as humans are exposed to a variety of chemical agents. Here, we examined combined genotoxic effects of low-dose-rate radiation and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK), the most carcinogenic tobacco-specific nitrosamine, in the lung of gpt delta transgenic mice. In this mouse model, base substitutions and deletions can be separately analyzed by gpt and Spi- selections, respectively. Female gpt delta mice were either treated with gamma-irradiation alone at a dose rate of 0.5, 1.0 or 1.5 mGy/h for 22 h/day for 31 days or combined with NNK treatments at a dose of 2 mg/mouse/day, i.p. for four consecutive days in the middle course of irradiation. In the gpt selection, the NNK treatments enhanced the mutation frequencies (MFs) significantly, but no obvious combined effects of gamma-irradiation were observable at any given radiation dose. In contrast, NNK treatments appeared to suppress the Spi- large deletions. In the Spi- selection, the MFs of deletions more than 1 kb in size increased in a dose-dependent manner. When NNK treatments were combined, the dose-response curve became bell-shaped where the MF at the highest radiation dose decreased substantially. These results suggest that NNK treatments may elicit an adaptive response that eliminates cells bearing radiation-induced double-strand breaks in DNA. Possible mechanisms underlying the combined genotoxicity of radiation and NNK are discussed, and the importance of evaluation of combined genotoxicity of more than one agent is emphasized.  相似文献

The probability that a dose of ionizing radiation kills a cell is about 10,000 times the probability that the cell will be transformed to malignancy. On the other hand, the number of cells killed required to significantly impact health is about 10,000 times the number that must be transformed to cause a late malignancy. If these two risks, cell killing and malignant transformation, are about equal, then the risk that occurs during a mission is more significant than the risk that occurs after a mission. The latent period for acute irradiation effects (cell killing) is about 2-4 weeks; the latent period for malignancy is 10-20 years. If these statements are approximately true, then the impact of cell killing on health in the low-gravity environment of space flight should be examined to establish an estimate of risk. The objective of this study is to synthesize data and conclusions from three areas of space biology and environmental health to arrive at rational risk assessment for radiations received by spacecraft crews: (1) the increased physiological demands of the space flight environment; (2) the effects of the space flight environment on physiological systems; and (3) the effects of radiation on physiological systems. One physiological system has been chosen: the immune response and its components, consisting of myeloid and lymphoid proliferative cell compartments. Best-case and worst-case scenarios are considered. In the worst case, a doubling of immune-function demand, accompanied by a halving of immune capacity, would reduce the endangering dose to a crew member to around 1 Gy.  相似文献

Micriwave radiations are being increasingly used in radar, space research, telecommunication, medicine and in ovens. Microwaves have been found to induce chromatid and chromosomal aberrations in human lymphocyte cultures [1], mice spermatogonia [8], Chinese hamster corneal epithelial cells [13], root tips of garlic [7] and Vicia faba [4]. In the present study we have investigated the mutagenic potential of 2450 MHz microwave radiation in mice using the dominant lethal test and sperm abnormality assay.  相似文献

Background  

μ-calpain and m-calpain are ubiquitously expressed proteases implicated in cellular migration, cell cycle progression, degenerative processes and cell death. These heterodimeric enzymes are composed of distinct catalytic subunits, encoded by Capn1 (μ-calpain) or Capn2 (m-calpain), and a common regulatory subunit encoded by Capn4. Disruption of the mouse Capn4 gene abolished both μ-calpain and m-calpain activity, and resulted in embryonic lethality, thereby suggesting essential roles for one or both of these enzymes during mammalian embryogenesis. Disruption of the Capn1 gene produced viable, fertile mice implying that either m-calpain could compensate for the loss of μ-calpain, or that the loss of m-calpain was responsible for death of Capn4 -/- mice.  相似文献

The features of the repair processes of DNA damage and their implementation at the chromosome level in marrow cells of experimental animals (mices of the CBA and C3H lines) with different radiation sensitivity were studied. It is shown that the radiomodifying efficiency of moderate hyperthermia (HT) is higher for animals of the radioresyst line. At the same time, it was possible to observe intensive elimination of chromosomal damages, the level of which in later observations almost was not distinguished from the control values. For mices of more radiosensitive lines, a long-term potentiation damaging action of irradiation at the chromosomal level was observed as an additional HT effect.  相似文献
