Two groups of microbiologists have independently isolated 'Walsby's square bacterium' from salt crystallizer ponds; its growth depends on pyruvate. Genetic analysis shows that the squares, discovered 25 years ago on the Sinai Peninsula, are archaea rather than bacteria. These transparent tile-like cells might have been dismissed as surface artefacts of salt crystals but for their gas vesicles--structures peculiar to prokaryotic organisms. Paradoxically, the square archaea are the dominant microorganisms in some hypersaline environments and might be globally important.  相似文献

Cultivation of Walsby's square haloarchaeon   总被引:10,自引:0,他引:10  
The square haloarchaea of Walsby (SHOW group) dominate hypersaline microbial communities but have not been cultured since their discovery 25 years ago. We show that natural water dilution cultures can be used to isolate members of this group and, once in pure culture, they can be grown in standard halobacterial media. Cells display a square morphology and contain gas vesicles and poly-beta-hydroxybutyrate (PHB) granules. The 16S rRNA gene sequence was >99% identical to other SHOW group sequences. They prefer high salinities (23-30%), and can grow with a doubling time of 1-2 days in rich media. The ability to culture SHOW group organisms makes it possible to study, in a comprehensive way, the microbial ecology of salt lakes.  相似文献

Square arrays are prominent structures in plasma membranes of brain, muscle, and kidneys with an unknown function. So far, the analysis of these arrays has been restricted to freeze fracture preparations, which have shown square arrays to contain the water channel Aquaporin-4 (AQP4). Using Blue-Native PAGE immunoblots, we provide evidence that higher-order AQP4 complexes correspond to square arrays, with the AQP4 isoform M23 playing a dominant role. Our data are consistent with the idea that square arrays consist of aggregates of AQP4 tetramers complexed with multiples of dimers. By comparison, Aquaporin-1 and Aquaporin-9 form tetramers, but not higher-order complexes. AQP4 square arrays are stable under several biochemical purification steps. Analyzing the internal composition of the higher-order complexes by 2D gels, we demonstrate that the square arrays in addition to M23 also invariably contain AQP4, M1, and a novel AQP4 isoform that we call Mz. The visualization AQP4 square arrays by a rapid, biochemical assay provides new insight in the molecular organization of square arrays and gives further proof of the heterogeneity of AQP4 square arrays in vivo.  相似文献

Response of cotton to prebloom square loss   总被引:2,自引:0,他引:2  
In 1996 and 1997, various intensities of prebloom square removal were applied to three cultivars of cotton grown in Mississippi. With the exception of one cultivar in 1997, all cultivars were B. thuringiensis (Bt)-transgenic cotton. At harvest, the number of bolls and seed cotton weight was recorded for all plants in each square removal treatment. All cultivars responded similarly to square loss. A yield increase (overcompensation) was observed in the treatment where all squares were removed from the plant one week after squaring began. Only the treatment where all squares were removed before bloom significantly reduced yield and caused a large (>7 d) delay in crop maturation. Otherwise, moderate levels of square removal (approximately 20-50% of prebloom squares) had little impact on overall lint production. However, the patterns of cotton production on the plants were significantly influenced by the square removal treatments. The removal of relatively more or larger squares increased seed cotton production in late-season fruiting cohorts and on 'vegetative' branches. Compensation for square loss occurred by increasing the relative number and weight of bolls produced subsequent to early-season square removal. Typically, early-season square loss increased the value of later-season fruiting cohorts, especially the midseason cohorts and bolls on vegetative branches. The implications of prebloom square loss, including the compensatory ability of the cotton plant, on insect management are discussed.  相似文献

Oxygen transfer rate and the corresponding power requirement to operate the rotor are vital for design and scale-up of surface aerators. Present study develops simulation or scale-up criterion correlating the oxygen transfer coefficient and power number along with a parameter governing theoretical power per unit volume (X, which is defined as equal to F(4/3)R(1/3), where F and R are impellers' Froude and Reynolds number, respectively). Based on such scale-up criteria, design considerations are developed to save energy requirements while designing square tank surface aerators. It has been demonstrated that energy can be saved substantially if the aeration tanks are run at relatively higher input powers. It is also demonstrated that smaller sized tanks are more energy conservative and economical when compared to big sized tanks, while aerating the same volume of water, and at the same time by maintaining a constant input power in all the tanks irrespective of their size. An example illustrating how energy can be reduced while designing different sized aerators is given. The results presented have a wide application in biotechnology and bioengineering areas with a particular emphasis on the design of appropriate surface aeration systems.  相似文献

In 1980, A. E. Walsby described a square halophilic archaeon. This archaeon is of specific interest because of its unique shape and its abundance in hypersaline ecosystems, which suggests an important ecophysiological role. Ever since its discovery, the isolation and cultivation of 'Walsby's square archaeon' has been a holy grail for many microbiologists working on halophiles. Despite their abundance and easy recognition by microscopy, all cultivation attempts have failed up to now, marking the organism as one of the unculturables. Cultivation of the square archaeon is essential to understand their ecophysiological role and the nature of their unique morphologically features. Here, we report the isolation and cultivation of the enigmatic square archaeon that we propose to name Haloquadratum walsbyi. Pure cultures are easily maintained in simple artificial hypersaline media. Initial growth experiments revealed a tolerance to high concentrations of MgCl(2) (>2 M) in the presence of 3.3 M NaCl. Fresh cultures contained extraordinary large cells (>40 x 40 microm) without any visible division structures, ranking them among the largest prokaryotes known to date. The genome was estimated to contain approximately three million basepairs.  相似文献

Square bacteria are shown to have right-handed helical (RH) flagella. They swim forward by clockwise (CW), and backwards by counterclockwise (CCW) rotation of their flagella. They are propelled by several or single filaments arising at several or single points on the cell surface. When there are several filaments a stable bundle is formed that does not fly apart during the change from clockwise to counterclockwise rotation or vice versa. In addition to the flagella attached to the cells, large amounts of detached flagella aggregated into thick super-flagella, can be observed at all phases of growth.  相似文献

In the design of the total replacement of the human joint, the joint center of rotation is often important. This paper describes an analytical technique to study joint kinematics, especially the instantaneous center of rotation. The empirical data for the analysis were obtained by the sonic digitizer system which works acoustically and electronically.
