微波高压罐水解法在氨基酸分析中的应用简报   总被引:1,自引:0,他引:1  
本文报导了采用微波高压罐水解技术快速水解食物蛋白质,并测定其17种氨基酸含量的方法。研制的高压罐具有密闭性、微波穿透性好,耐热、耐压、耐腐蚀等特点。采用该高压罐研究了微波强度及作用时间等条件对氨基酸稳定性的影响,在微波输出功率100W35~45分钟水解了3份食品样品,其氨基酸含量测定结果与传统的110℃22左水解基本相符,17种氨基酸5次平行测定结果CV值均<10%。  相似文献

用氨基酸自动分析仪测定了蝉幼的氨基酸含量,结果表明:蝉幼的氨基酸总含量在17%左右,其中含有八种人体必需氨基酸,其氨基酸的构成比例与鸡蛋相近,为一富含蛋白质的食品。  相似文献

本文主要测定了山糜子幼苗中氨基酸、无机元素。结果表明,其幼苗含16种氨基酸,总含量为188.28g/kg,有7种是人体必需的氨基酸,其中亮氨酸和赖氨酸含量较高。同时发现,山糜子所含17种无机元素中,9种是人体必需的,而锌、锰、锶等对人体有重要生理意义的必需微量元素含量突出。本文还对该植物中蛋白质、氨基酸和微量元素的营养、食疗方面进行了评价。  相似文献

对α-淀粉酶、麦芽四糖淀粉酶和葡萄糖淀粉酶进行的氨基酸序列比较表明,它们之间的氨基酸等同性相当低。本文在对这三种淀粉酶的氨基酸残基进行疏水性分析的基础上,采用国际上近期发展的疏水簇分析方法对这三种淀粉酶的氨基酸序列进行了二维描述,其结果清楚地显示了它们之间的同源性差异。这也被测定的三维结构所证实。这一研究结果表明,与蛋白质三维结构密切相关的蛋白质的二级结构及其相互配置主要取决于蛋白质序列中不同类型氨基酸残基的排列。  相似文献

在对黄皮种子氨基酸的组成及蛋白质的含量进行分析的基础上,应用模糊识别法和氨基酸比值系数法,对其营养价值进行了全面评价,并与8种植物种子蛋白质进行对照比较。结果表明:黄皮种子(干品)蛋白质含量为8.625%,含有17种氨基酸,种类齐全,其含量为762.2 mg.g-1蛋白质,必需氨基酸(EAA)占总氨基酸量的35.5%,第一限制性氨基酸为含硫氨基酸(M et+Cys),贴近度为0.8536,营养价值与高粱米、西瓜子等接近。  相似文献

为了弄清不同闽产李(Prunus salicina)果实氨基酸含量、组成及营养价值,采用氨基酸自动分析仪 对不同李果实样品中各种氨基酸的含量进行检测,并对其组成进行比较分析。结果表明,李果实含有18种蛋白质氨基酸和3种非蛋白质氨基酸。田黄李的牛磺酸含量与蛋白质氨基酸含量均最高分别为26.54 mg/100 g和496.14 mg/100 g;皇后李的鸟氨酸、药用氨基酸、酸味氨基酸含量以及特殊功效蛋白质氨基酸的比重均最高分别为0.36 mg/100 g、364.67 mg/100 g、298.28 mg/100 g和73.45%;早熟胭脂李的γ-氨基丁酸含量最高为7.98 mg/100 g;大胭脂李的支链氨基酸含量最高为46.77 mg/100 g。李果实中氨基酸种类齐全、含量存在一定的差异,可根据不同的开发利用目标选择合适的品种。  相似文献

研究测定了五种食用菌中氨基酸的含量,并利用五种非生物学指标对蛋白质的营养价值进行评价。五种食用菌氨基酸种类齐全,必需氨基酸占氨基酸总量的百分比分别达到45.94%、37.62%、42.45%、41.54%、35.98%。茶树菇和姬松茸的限制性氨基酸为半胱氨酸和甲硫氨酸,鸡腿菇、香菇和黑木耳的限制性氨基酸是缬氨酸。不同的评价指标对蛋白质进行营养评价,评价结果会有所差异。  相似文献

每种蛇毒含有多种蛋白质组分,每种蛋白质分子有其自已的一级结构和相应的氨基酸残基的组成。因此蛇毒的氢谱是其所有组成的谱图的加和,对不同产地和种属的20多种冷冻干燥的蛇毒进行了测定,结果每种蛇毒均显示其特征的核磁共振氢谱,提示各种蛇毒的氨基酸残基组成是不同的。  相似文献

酸雨是中国重要的环境问题,为研究酸雨对小麦籽粒品质的可能影响,以小麦品种扬麦15和汶农17为材料开展盆栽试验,研究了不同酸度(pH2.5、p H4.0和p H5.6)酸雨对小麦花后氮硫代谢关键酶活性和籽粒蛋白质含量及组分的影响。结果显示:酸雨处理抑制叶片硝酸还原酶(NR)活性,提高了扬麦15整个灌浆期及汶农17灌浆中后期叶片谷氨酰胺合成酶(GS)活性,促进了叶片蛋白的降解,降低了叶片可溶性蛋白含量。不同酸度酸雨提高了成熟期籽粒中蛋白质含量,酸度越强,增加幅度越大,籽粒中各蛋白组分含量和大部分氨基酸含量也有明显提高。酸雨提高了扬麦15叶片丝氨酸乙酰转移酶(SAT)和O-乙酰丝氨酸硫裂解酶(OAS-TL)活性,但对汶农17硫代谢关键酶活性影响较小,酸雨处理还提高了籽粒中二硫键和含硫氨基酸含量。可见酸雨对小麦氮硫代谢有不同程度影响,进而影响了小麦籽粒蛋白质含量和组成,酸度越强影响越大,但不同品种对酸雨响应有一定差异。  相似文献

The role of silent position in the codon on the protein structure is an interesting and yet unclear problem. In this paper, 563 Homo sapiens genes and 417 Escherichia coli genes coding for proteins with four different folding types have been analyzed using variance analysis, a multivariate analysis method newly used in codon usage analysis, to find the correlation between amino acid composition, synonymous codon, and protein structure in different organisms. It has been found that in E. coli, both amino acid compositions in differently folded proteins and synonymous codon usage in different gene classes coding for differently folded proteins are significantly different. It was also found that only amino acid composition is different in different protein classes in H. sapiens. There is no universal correlation between synonymous codon usage and protein structure in these two different organisms. Further analysis has shown that GC content on the second codon position can distinguish coding genes for different folded proteins in both organisms.  相似文献

An original modification of the standard ELISA procedure for differential determination of different forms of abscisic acid (ABA) is proposed. It is shown that endogenous forms of ABA may be quantitatively determined in plant tissues subjected to minimal treatment, without purification of the hormones and their chemical modification. The modification has been approved when analyzing changes in the content of different ABA forms in plant tissues differing in physiological activity. Quantitative differential determination of changes in the content of different ABA forms has been performed in ovaries of Triticum aestivum L. and Taraxacum officinale Web. in the period of activity of the ovule (from the moment of its activation to the beginning of division). It is shown that, despite the different types of reproduction in the species studied (amphimixis and apomixis), the time course of changes in the content of different forms of ABA in ovaries is similar, which is suggestive of a correlation between the activity of endogenous hormonal system and chronology of main events (e.g., the beginning of endospermogenesis) of the reproductive cycle.  相似文献

杜仲(EucommiaulmoidesOliv)是我国特有的雌雄异株植物,具有极高的药用价值、工业价值和观赏价值。主要依靠种子繁殖。为了有计划地进行定性栽培,就需要对幼龄植株进行早期性别鉴定。许多生理指标都可能作为性别鉴定的依据。通过对杜仲中氨基酸含量和组成的分析表明,不同性别植株树皮和叶片中均含有被分析的17种氨基酸,其中胱氨酸和蛋氨酸在雌雄株间差异明显,雄株树皮和叶片中胱氨酸的含量分别高出雌株76.98%和45.04%;树皮和叶片中蛋氨酸含量雌株分别高出雄株20.50%和18.77%。这是否反映了杜仲雌雄的性别差异,值得探讨。必需氨基酸总量在树皮和叶片中均表现为雌株高于雄株,且叶片中必需氨基酸总量高于树皮中的含量。  相似文献

Summary A number of experiments was carried out with pea plants growing in a nutrient solution of low pH, inhibitory to nodule formation but not to plant growth when combined nitrogen was available. It has been found that on transferring plants from a neutral culture solution to an acid solution of pH 4.5 at different periods of time after inoculation, nodulation is reduced when the exposure to low pH takes place during the first days after inoculation, and particularly on the second or third day. Once nodulation has been initiated, nodule growth and nitrogen fixation proceed normally in an acid solution. Failure of nodulation at low pH, under the conditions of these experiments, was not due to the inhibition of bacterial growth. From these results an acid-sensitive period, occurring during the second or third day after inoculation, was demonstrated.  相似文献

1. Phytophagous insects frequently manipulate their host‐plant to improve their immediate environment. This generally implies substantial modifications of host metabolism, and sometimes an alteration of nitrogen allocation within the host‐plant. However, the outcome of plant manipulation on amino acid or protein content can be modulated by environmental factors and host‐plant traits. 2. It was investigated whether the pseudogall induced by the aphid Phloeomyzus passerinii (Signoret) (Aphididae: Phloeomyzinae) in the bark of its host‐plant affects the amino acid content in bark tissues, and whether the strength of the modification is modulated by the fertilisation regime and/or the resistance level of the host‐plant. The development of aphid colonies on a resistant and a susceptible poplar genotype, under three fertilisation regimes, was studied. After the development of colonies, the free and protein‐bound amino acid content of the infested bark were quantified. 3. Fertilisation enhanced poplar growth and increased the free amino acid content of bark tissues. Infestation also triggered accumulations of both free and protein‐bound amino acids in the feeding sites, but in the susceptible genotype only. The increase in amino acid content was more pronounced when fertilisation was low, and fertilisation did not enhance aphid development. 4. In conclusion, infestation by P. passerinii triggers an accumulation of amino acids, but the effect is influenced by both the fertilisation regime and the resistance level of the host‐plant. This suggests that P. passerinii could affect the allocation of nutrients within trees during outbreaks.  相似文献

By the method of quantitative cytophotometry, the content of ribonucleic acid in neurons and mantle gliocytes of the cat vagus caudal node has been studied after a single maximal overloading of 10 units. It has been stated that a decreased RNA content in the mantle gliocytes is less pronounced; that could be explained by their different nutritional conditions (owing to their various position towards the microvessels of the node), on the one hand, and by different functional loading experienced by the two types of cells under the effect of the extreme factor studied, on the other hand. RNA content in gliocytes is restored more quickly owing to its less decrease, and also owing to a more favourable protein synthesis in glycocytes--"for itself".  相似文献

In order to meet the protein nutrition needs of the world population, greater reliance on plant protein sources will become necessary. The amino acid balance of most plant protein sources does not match the nutritional requirements of monogastric animals, limiting their nutritional value. In cereals, the essential amino acid lysine is deficient. Maize is a major component of human and animal diets worldwide and especially where sources of plant protein are in critical need such as sub-Saharan Africa. To improve the amino acid balance of maize, we developed transgenic maize lines that produce the milk protein α-lactalbumin in the endosperm. Lines in which the transgene was inherited as a single dominant genetic locus were identified. Sibling kernels with or without the transgene were compared to determine the effect of the transgene on kernel traits in lines selected for their high content of α-lactalbumin. Total protein content in endosperm from transgene positive kernels was not significantly different from total protein content in endosperm from transgene negative kernels in three out of four comparisons, whereas the lysine content of the lines examined was 29–47% greater in endosperm from transgene positive kernels. The content of some other amino acids was changed to a lesser extent. Taken together, these changes resulted in the transgenic endosperms having an improved amino acid balance relative to non-transgenic endosperms produced on the same ear. Kernel appearance, weight, density and zein content did not exhibit substantial differences in kernels expressing the transgene when compared to non-expressing siblings. Assessment of the antigenicity and impacts on animal health will be required in order to determine the overall value of this technology.  相似文献

A simple and efficient process for the production of a ruminant feed supplement, rich in crude protein (defined as total N X 6.25), by bacterial fermentation of cheese whey has been developed. The lactose in unpasteurized whey is fermented to lactate acid by Lactobacillus bulgaricus at a temperature of 43 degrees C and pH 5.5. The lactic acid produced is continually neutralized with ammonia to form ammonium lactate. The fermented product is concentrated by evaporation to a solids content of about 70% and adjusted to pH 6.8 with additional ammonia. The concentrated product contains about 55% crude protein. Approximately 6 to 8% of the crude protein is derived from bacterial cells. 17% from whey proteins, and 75 to 77% from ammonium lactate. The efficiency of conversion of lactose to lactic acid usually exceeds 95%. The fermentation time is greatly reduced upon the addition of 0.2% yeast extract or 0.1% corn steep liquor as a source of growth factors. Whey containing lactose at concentrations up to 7% can be fermented efficiently, but at higher concentrations lactose is fermented incompletely. The process has been scaled up to a pilot plant level, and 40 tons of concentrated product were produced fro animal feeding trials, without ever encountering putrefactive spoilage.  相似文献
