Heat shock protein 70 (HSP70) is a chaperone that maintains protein conformation during heat stress. It has recently been observed that HSP70 may be released from cells in response to increased energy demand (e.g., exercise) and/or oxidative stress. Since HSP70 levels should change in response to athletic training, we have investigated whether blood HSP70 levels in young women handball players change over a complete training season. Thirty women handball players (12-24 years old) were divided into low (≥30 pg mL(-1)) (LE) and normal (30-330 pg mL(-1)) (NE) estradiol groups. HSP70 levels in lymphocytes and plasma and blood redox parameters were evaluated over 1 year (2009), with sampling at the beginning, middle, and end of the season. We observed no changes in superoxide dismutase activity or protein carbonyl or extracellular HSP70 levels, while catalase activity increased at the middle of the season in the NE group, and the thiobarbituric acid species levels in both groups were higher at the beginning of the season than at the middle or end. The lymphocyte HSP70 content was higher at the middle and end than at the beginning of the season in the NE group and also higher in the LE group than in the NE group at the beginning of the season. These results suggest that plasma estradiol levels may play an important role in exercise training and that the intracellular HSP70 content, a biomarker for inflammation, is affected by both estradiol levels and exercise-induced oxidative stress.  相似文献

Regular training has been claimed to increase the activity of antioxidant enzymes and, consequently, augments the resistance to oxidative stress; however, large volumes of training performed by elite sportsmen could lead to a chronic oxidative stress state. The aim of our study was to assess the oxidative status of elite athletes at the beginning of the preparatory and the beginning of the competition training phases, so that the influence of three months of programmed physical activity on redox status could be determined. The chronic effects of exercise on the redox state of the athletes were compared to the effects of a single bout of karate training. Thirty elite karate athletes, 16-30 years old, were subjected to maximal graded exercise test to estimate their aerobic capacity; blood sampling was also performed to measure levels of superoxide anion radical (O??), hydrogen peroxide (H?O?), superoxide dismutase activity (SOD) and catalase activity (CAT). The only significant change after the three-month training process was found in the significantly decreased CAT activity (X ± SE: 7.95 ± 0.13 U/g Hb × 103 in the preparatory period, 6.65 ± 0.28 U/g Hb × 103 in the competition stage; P < 0.01). After a single karate training session, there was statistically significant decrease of O??(X ± SE: 32.7 ± 4.9 nmol/ml in the preparatory period, 24.5 ± 2.5 nmol/ml in the competition stage; P < 0.05) and increase of H?O?(X ± SE: 11.8 ± 1.0 nmol/ml in the preparatory period, 14.2 ± 0.9 nmol/ml in the competition stage; P < 0.01), as well as significant CAT increase (X ± SE: 6.6 ± 0.6 U/g Hb × 103 in the preparatory period, 8.5 ± 0.5 U/g Hb × 103 in the competition stage; P < 0.05). Although the three-month training process induced, at the first sight, negative changes in the redox state, expressed through the decrease in CAT activity, adequate response of the antioxidant system of our athletes to acute exercise was preserved.  相似文献

Four distinct stages of diapause development in pupae of Mamestra configurata held at 20°C can be recognized by means of the ‘β-ecdysone sensitivity index’. The latter refers to the ED50 of injected β-ecdysone required to break diapause in half of the treated pupae. Stage 1 begins with the newly-formed pupa, lasts about 3.5 days and is characterized by a rapidly falling ED50. Stage 2 lasts about four weeks during which the ED50 increases by almost 20-fold, from 0.27 μg/g at the beginning to 4.9 μg/g at the end. Stage 3 begins when the pupae are about 4 weeks old and lasts for about six weeks. Stage 3 is the stable diapausing state and is characterized by a virtually unchanging ED50. The onset of stage 4 occurs when the pupae are about 10 weeks old and is recognized by the beginning of a decline in the ED50. Stage 4 precedes the completion of diapause development and may signal the transition of the endocrine system to its active state.Pupal diapause deepens with time in M. configurata. The deepening process evidently occurs during stage 2 of diapause development. Pupae that were transferred to 5°C at the beginning of stage 2 failed to make the transition to stage 3 and were trapped in a very shallow diapause.  相似文献

The development of the cells in the posterior silk gland of the silkworm, Bombyx mori, during the fourth larval instar has been studied. In the early stages of this instar, the wet weight of the gland and the amounts of RNA, DNA, and protein per animal increase logarithmically until they reach a stationary state at about 72 hr. At around 96 hr of the fourth instar, the larvae enter the molting state, which lasts for about 24 hr until the fourth ecdysis. Towards the end of the molt stage, the growth of the silk gland is resumed. Electron microscopical observation shows that in the early intermolt stage the cytoplasm is filled with free ribosomes and with rough endoplasmic reticulum (ER), first of the lamellar type (0–6 hr) and then of the vesicular or tubular type. The Golgi apparatus also is well developed. At the beginning of the molt stage (90–96 hr), however, most of the ER becomes lamellar in type, concentric lamellar structures being occasionally observed, and the Golgi vacuoles disappear. Autophagosomes and lysosomes increase markedly and the apical portion of the cytoplasm becomes extensively vacuolated; this suggests that the secretory activities are completely depressed, and pronounced degenerative changes appear during the molt stage. Towards the end of the molt stage, large lamellar ER elements are fragmented into smaller lamellae and there is a pronounced increase in the number of free ribosomes.  相似文献

The effects of the watch system of job management on central hemodynamic parameters and the constant potential of the brain were studied in sailors during a long voyage. Twenty crew members were examined during a 120-day translatitudinal voyage. The studies were performed on days 4–6, 40–42, 73–75, and 107–109 of the voyage. It was found that the role of the peripheral regulatory loop in maintaining physiological functions increases during the second half of the voyage and that the functional activity before a watch determines the changes in the parameters by the end of the watch. For each stage of the study, the baseline value of the constant potential of the brain is an important element of functional activity. The system of ratios between parameters before and after a watch that is formed in the early period of the voyage breaks up during the second month, the general reactivity of the body after a watch being independent of the reactivity before the watch; the reactivity is stochastic and is mainly controlled by central mechanisms. In the second month of the voyage, the first signs of a strategy aimed at making the functioning more economical are observed in crew members. The second and third months of the voyage are characterized by a considerable strain of the mechanisms of adaptation, activity, and reactivity in sailors. Cytolytic processes in posterior silk gland cells of the silkworm, Bombyx mori, during metamorphosis from larva to pupa have been studied. During this stage, the wet weight and the amounts of RNA and protein of the gland decrease rapidly and markedly, while the amount of DNA decreases slowly and slightly. The ultrastructural changes observed at the beginning of the prepupal stage consist of the appearance or the increase in the number of autophagosomes containing endoplasmic reticulum (ER), or "early autophagosomes" as we have called them, which seem to be gradually transformed to autolysosomes. A number of usual lysosomes, which frequently contain myelin figures, also appear in the cytoplasm. Sometimes they fuse with each other to form large conglomerates. In the middle of the prepupal stage, a number of smooth membrane-bounded vacuoles appear in cytoplasm. Towards the end of the prepupal stage the partition or sequestration of cytoplasm was observed. Thus large autophagosomes containing cytoplasmic organelles such as rough ER and/or mitochondria are formed. The nucleus is partitioned in a similar way by smooth membranes, and then autophagosomes containing condensed chromatin blocks are formed. These various kinds of autophagosomes, or "late autophagosomes" as we have generally called them, are continuously released into the hemolymph until the gland is completely disintegrated.  相似文献

The biological activity of the catholyte and anolyte of bidistilled water in experiments with the germination of wheat grains in the period from March to May has been studied. The activity of solutions, which was characterized by the grain germination index, was high at the beginning of the period, then it gradually decreased and was equal to zero at the middle of the period; at the end of the period it gradually increased almost to initial values. It has been established that the effectiveness of bidistilled water anolyte was as a rule higher than that of catholyte throughout the observation period. At the beginning and end, the stimulating effect of anolyte was 5-5.5 times greater than that of catholyte. The seasonal changes in the biological activity of M 9 medium catholyte were compared with those of bidistilled water anolyte and catholyte. The stimulating effect of M 9 catholyte was estimated by changes in the growth of E. coli cells. The stimulating effect, which was estimated from an increase in the optical density of cell suspension in the initial period at a cultivation temperature of 20 degrees C was 55-60% relative to control (untreated medium). Then it decreased almost to zero in the middle of the period to increase again approximately to the initial values. The assumption has been made that the physicochemical causes of the influence of catholyte and anolyte of bidistilled water on wheat grains and of the culture medium catholyte on E. coli cells are of different nature.  相似文献

Changes in social hierarchy were evaluated in a herd of 3-year-old Hanoverian geldings. One group (n = 5) was exposed to a training programme, the other (n = 5) remained untrained. After 6 months, the groups were reversed. Hierarchical positions were evaluated by field observations and/or paired-feeding tests at the beginning, the middle, the end of the first and at the end of the second training period. Both methods yielded identical results. Almost all horses changed position in only one direction: either up or down. Neither increase in aggression nor mutual injuries were recorded during the whole experiment. No statistically verified differences in dominance ranking occurred between trained and untrained groups, but apparent differences were consistent. Thus, if horses are kept in the same group for a longer period of time, exercise induced changes in hierarchy are probably of minor importance and are unlikely to increase the incidence of injuries. This may have implications for the promotion of group-housing for sport horses.  相似文献

To explain the decrease of hydrogen production rate in the batch culture of Rhodobacter sphaeroides S, the activities of enzymes related to the TCA cycle, nitrogenase in cell-free extracts, ATP generation by chromatophores, and ferredoxin were examined at the beginning, middle and end of the hydrogen production phase of batch culture. The activities of TCA cycle enzymes, nitrogenase and ATP generation were found to remain at almost the same level throughout the culture, while ferredoxin activity decreased linearly with time. In addition, by bubbling N2 gas into the culture broth at the end of the culture, the hydrogen production rate was restored to the initial level through the increase of the ferredoxin activity. Although the decrease of ferredoxin activity and its restoration by bubbling N2 gas remained unexplained, ferredoxin activity was considered to be a key function in the nitrogenase system for H2 production by this photosynthetic bacterium.  相似文献

The development, generation number and diapausing state of threeCyclops species dominant in the open water of Lake Balaton werestudied by means of vials placed back into the lake, in naturalfeeding conditions. The males of all three species turned intoadults earlier than the females. The copepodite V stage wasof the longest duration. Three generations of Cyclops vicinusdeveloped yearly; in spring, autumn and winter. About 50% ofthe copepodite stage IV of the spring generation entered activediapause at the end of May, beginning of June, then entereddormancy between the middle of June and middle of July. Consideringthe literary data as well, it seems that the generation numberof the species is the function of temperature and of the water'strophic state. The Acanthocydops robustus f. limnetica is fondof warm water (> 14°C). Seven generations of this speciesdeveloped completely yearly, during an average of 2-week intervalsbetween May and September. The eightli generation died in thenaupliar stage. The generation number of A. robustus f. limneticadepends on the average temperature of the lake. Inactive diapausecould not be observed for this species. Only one generationof Mesocyclops latckarti developed in autumn. The copepoditeV of the winter generation entered active diapause below a watertemperature of 10°C, and inactive diapause from the endof January, which state ended in March.  相似文献

The history of Stare Biele paleolake (northeast Poland) has been reconstructed using subfossil Cladocera remains and pollen and spores of aquatic and mire plants from a sediment core. Sediment accumulation began approximately 12,000 years ago during the Older Dryas chronozone. Throughout the entire Late Glacial period, the basin was a small, low-trophic state lake with a developed open-water zone. A well-recorded increase in the trophic state started at the beginning of Holocene. The lake reached its highest trophic level during the early and middle Atlantic chronozone. The first human activity in the lake catchment area occurred at this time, as recorded by fern spores and numerous charcoal grains. Repeated rises in lake water level are documented at the beginning and throughout the early part of the Younger Dryas. Two clear events of decreasing lake water levels are recorded, first during the middle part of the Younger Dryas and second in the Preboreal. In the Seno de Reloncaví, southern Chile, seasonal changes in dry weight (DW) and elemental composition (CHN) were studied in embryo (initial embryonic stage), newly hatched zoeae, and newly settled megalopae of a porcelain crab, Petrolisthes laevigatus. Samples were taken throughout the seasons of egg laying (March-December), hatching (August-February), and settlement (October–February). Values of DW and CHN per embryo or larva, respectively, were consistently minimum in the middle of each season and maximum near its beginning and end. Patterns of seasonal variation in early embryonic biomass may thus be carried over to larvae at hatching and, possibly, to the settlement stage. Such carry-over effects may be selectively advantageous, as zoeae released at the beginning or near the end of the hatching season face conditions of poor planktonic food availability in combination with low winter temperatures or decreasing temperatures at the end of summer (enforcing long development duration). Hence, an enhanced female energy allocation into egg production may subsequently translate to enhanced yolk reserves remaining at hatching, allowing for a larval development under unfavourable winter conditions. In summer, by contrast, plankton productivity and temperatures are generally high, allowing for fast larval growth and development. This coincides with minimal biomass and energy contents both at hatching and settlement. In conclusion, our data suggest that seasonal patterns in the biomass of early developmental stages of P. laevigatus may reflect phenotypic variability as an adaptive response to predictable variations in environmental conditions, allowing this species to reproduce in temperate regions with marked seasonality in water temperature and plankton productivity.  相似文献

We determined the changes in hemocyte titer and in the abundance of hemocyte types of the tobacco hornworm Manduca sexta during the fourth and fifth larval stadium and the beginning of the pupal stadium. As we analyzed the samples of individual insects at daily intervals, we were able to correlate phenotypical features, body weight, as well as total protein content and lysozyme activity in the hemolymph with the observations on hemocytes. In the course of the fifth larval stadium, the hemocyte titer decreased slightly and declined further after pupation. Using calculated values for total hemocyte numbers, females had about five times and males three times more hemocytes in the circulating population at the beginning of the wandering stage (in the middle of the fifth larval stadium) than immediately after the last larval--larval molt (from the fourth to the fifth larval stadium). This sexual difference was mainly due to an increase in the number of plasmatocytes, which was more prominent in females than in males. Granular cells were dominant in early fifth larval stadium while plasmatocytes were the most abundant cells in pupae. Oenocytoids and spherule cells disappeared during the wandering stage. Lysozyme activity in the hemolymph rose to a maximum during the wandering stage, with females having lysozyme values twice as high as those for males. These changes in lysozyme activity, however, did not correlate with the increase of total hemolymph protein titer which occurred already at the beginning of the wandering stage. We postulate that changes in hemocyte titers are under direct hormonal control, which has to be proven in future experiments.  相似文献

Sea travel mandates changes in the control of the body. The process by which we adapt bodily control to life at sea is known as getting one''s sea legs. We conducted the first experimental study of bodily control as maritime novices adapted to motion of a ship at sea. We evaluated postural activity (stance width, stance angle, and the kinematics of body sway) before and during a sea voyage. In addition, we evaluated the role of the visible horizon in the control of body sway. Finally, we related data on postural activity to two subjective experiences that are associated with sea travel; seasickness, and mal de debarquement. Our results revealed rapid changes in postural activity among novices at sea. Before the beginning of the voyage, the temporal dynamics of body sway differed among participants as a function of their (subsequent) severity of seasickness. Body sway measured at sea differed among participants as a function of their (subsequent) experience of mal de debarquement. We discuss implications of these results for general theories of the perception and control of bodily orientation, for the etiology of motion sickness, and for general phenomena of perceptual-motor adaptation and learning.  相似文献

Altitudinal variation of the demographic structure of populations of Poecilus fortipes (Chaud.) was studied in four mountain localities in the south of Eastern Siberia, and the life cycle of the species was reconstructed for each locality. The size of adults of both sexes decreased along the altitudinal gradient: the beetles from the Selenga middle mountains (600–650 m above sea level) were much larger, while those at the upper boundary of the forest-steppe belt in the Eastern Sayan (1500–1700 m asl) were smaller than others. Their linear dimensions decreased more distinctly in the transitional zone between middle and high mountains. The duration of the reproductive period did not depend on the altitude. However, the onset of oviposition shifted along the altitudinal gradient to the beginning of the vegetation season, due to its shortening and a decrease in the sum of effective temperatures. As the altitude increased, the mean number of eggs per female declined significantly from 15.9 ± 6.1 to 8.6 ± 5.8, whereas the maximum number of eggs increased from 26–28 to 40–43. On the contrary, the dates of emergence shifted gradually from the beginning and middle of the vegetation period to its middle and end. Populations from the Selenga middle mountains, the Vitim Plateau, and the lower margin of the forest-steppe belt in the Eastern Sayan mountains were characterized by a relatively low catch index and the absence of rebreeding old individuals, both these features increasing the risk of spontaneous local extinction. The annual life cycle with summer breeding was replaced along the altitudinal gradient with an obligate biennial life cycle with summer and early-summer breeding. This transition was observed as the altitude increased from 600–650 to 900–950 m asl while the sum of effective temperatures dropped below 1400°C, i.e., the value needed for implementation of the annual life cycle.  相似文献

Burnout and stress-related mental disorders (depression, anxiety) occur in medical students and physicians with a significantly higher prevalence than in the general population. At the same time, the learning of coping mechanisms against stress is still not an integral part of medical education. In this pilot study we developed an elective course for learning relaxation techniques and examined the condition of the students before and after the course. 42 students participated in the semester courses in 2012 and 2013 as well as in a survey at the start and end of each course. The students were instructed in autogenic training (AT) and progressive muscle relaxation according to Jacobsen (PMR) with the goal of independent and regular exercising. At the beginning and the end of the semester/course the students were interviewed using standardized, validated questionnaires on burnout (BOSS-II) and anxiety (STAI-G), depression (BDI), quality of life (SF-12) and sense of coherence (SOC-L9). We compared the results of our students participating in Relacs with results from eight semester medical students (n = 88), assessed with the same questionnaires at similar points of time within their semester. Participating students showed a significant decline in cognitive and emotional burnout stress and in trait anxiety. Furthermore, they showed a reduction in state anxiety and a conspicuous decrease in mean depression. The sense of coherence increased at the same time. A comparative cohort of medical students of 8th semester students, showed lower values for the specified measurement parameters at the beginning, but showed no progressive changes. Our course introducing AT and PMR led to a significant reduction of burnout and anxiety within the participating group of medical students. Even the course attendance for just one semester resulted in significant improvements in the evaluated parameters in contrast to those students who did not attend the course.  相似文献

Locusta migratoria larvae were submitted to electrical stimulation of the protocerebral neurosecretory cells (median neurosecretory cells of the pars intercerebralis and lateral neurosecretory cells), during the last larval instar. The effects of the treatment were observed both on the duration of the stage and on the variations in haemolymph ecdysone levels. In untreated larvae, there was an initial ecdysone peak at the beginning of day 5, which was followed by 4 larger peaks between days 6 and 8. Stimulation of the median neurosecretory cells at the beginning of the instar resulted in the formation of one very large hormonal peak at the end of day 3: a day and a half earlier than in the control groups. Moulting was likewise accelerated. Stimulation also increased the size of the peaks, as compared with the controls. Stimulation of the lateral neurosecretory cells had a weaker ecdysiotropic effect; neither the number nor the size of the peaks were changed, though, like ecdysis, they occurred earlier. Stimulation of the deutocerebrum had no effect on either ecdysone titres or moulting. Electrical stimulation of the median neurosecretory cells at the end of day 5, that is after the occurrence of the first ecdysone peak, shortened the larval stage while having no significant effect on ecdysone levels in the haemolymph. The neuroendocrine control of ecdysis in Locusta is discussed.  相似文献

When an extract of the corpora cardiaca/corpora allata from two species of wingless stick insects, Carausius morosus and Cuniculina impigra, which cause no adipokinetic or hyperglycaemic effect when injected into the donor insects themselves, is injected into adult Locusta migratoria it resulted in an increase in the haemolymph lipid concentration. The lipid elevation was time dependent, with a maximum effect about 90–180 min after injection, and was also dose-dependent. About 0.001–0.002 (C. morosus) and 0.01 (C. impigra) gland equivalents were needed to produce a significant increase; a maximal effect was reached with approx. 0.075 (C. morosus) and 0.25 (C. impigra) gland equivalents. Carausius extract was also able to elevate carbohydrate concentration in the haemolymph of Periplaneta americana. However, the effect was weak and no maximal response was reached even with a dose of 0.5 gland equivalents. Adipokinetic hormone activity was present in CC/CA extracts of larval Carausius; the activity was about 30 times lower in 1-day-old 2nd instar individuals, and approx. 5 times less at the beginning of the 6th instar than that found in adults. In both stages the hormone levels increased gradually from the beginning to the end of the instar. No age-related changes were observed during the adult stage. Further studies on the lipid-mobilising factor of C. morosus revealed that it was stored entirely in the CC and not in other nervous tissue, e.g. brain, CA, suboesophageal ganglion, thoracic and abdominal cord. The factor was heat stable for at least 1 hr at 100°C and retained its adipokinetic activity after incubation with trypsin and the exopeptidases such as carboxypeptidase A and leucine aminopeptidase. However, activity was abolished when incubated with thermolysin and α-chymotrypsin. From these experiments a close resemblance to the locust AKH, a blocked decapeptide, is suggested.  相似文献

Determinations of total protein and RNA at various times after the beginning of the pharate pupal stage of Drosophila hydei, revealed an increase in both substances during the first 25 hr and a sudden decrease thereafter until 52 hr. From this time on the total amount of both protein and RNA increases slightly until emergence of the flies at 160 hr after the beginning of the pharate pupal stage. A similar pattern of changes was recorded for the total radioactivity as well as the specific activity of RNA labelled with 3H-uridine after the injection of the isotope immediately before the beginning of the pharate pupal stage.The migration profile of RNA labelled with 3H-uridine during larval development, revealed that shortly after the onset of the pharate pupal stage an essentially normal larval pattern consisting of major fractions of 28, 18, 8 to 9, and 4 to 7 S RNA. At 52 hr only the low molecular weight RNA was present. The ‘normal’ pattern was restored at the time of emergence of the flies, indicating re-utilization of degradation products of previously labelled RNA.  相似文献

We used tensor-based morphometry (TBM) to: 1) map gray matter (GM) volume changes associated with motor learning in young healthy individuals; 2) evaluate if GM changes persist three months after cessation of motor training; and 3) assess whether the use of different schemes of motor training during the learning phase could lead to volume modifications of specific GM structures. From 31 healthy subjects, motor functional assessment and brain 3D T1-weighted sequence were obtained: before motor training (time 0), at the end of training (two weeks) (time 2), and three months later (time 3). Fifteen subjects (group A) were trained with goal-directed motor sequences, and 16 (group B) with non purposeful motor actions of the right hand. At time 1 vs. time 0, the whole sample of subjects had GM volume increase in regions of the temporo-occipital lobes, inferior parietal lobule (IPL) and middle frontal gyrus, while at time 2 vs. time 1, an increased GM volume in the middle temporal gyrus was seen. At time 1 vs. time 0, compared to group B, group A had a GM volume increase of the hippocampi, while the opposite comparison showed greater GM volume increase in the IPL and insula in group B vs. group A. Motor learning results in structural GM changes of different brain areas which are part of specific neuronal networks and tend to persist after training is stopped. The scheme applied during the learning phase influences the pattern of such structural changes.  相似文献
