By means of electron probe analysis, the effects of significant amounts of accumulation of silicon on the accumulation of calcium, potassium, magnesium, manganese, phosphorous, iron, and sodium in the silica cells of rice leaves are described. The silica cells of both the surfaces of the leaf blade and leaf sheath were studied. Silicon accumulation in the silica cells appears to decrease the amount of accumulation of potassium on both the surfaces of the leaf blade and sheath. The effect of significant amounts of silicon accumulation on the accumulation of other elements in a particular cell varies in different organs or on different surfaces of the organ of the same plant. Magnesium, manganese, iron, and phosphorus could not be detected in the adaxial epidermis of the leaf sheath and magnesium and iron in the adaxial epidermis of the leaf blade. Manganese, magnesium, and phosphorus were not detected in the abaxial epidermis of the leaf blade nor iron in the abaxial epidermis of the leaf sheath. Sodium was not revealed in either surface of the leaf blade and leaf sheath. Possible mechanisms for the effects of silicon accumulation on the accumulation of these elements in rice leaf epidermal cells are discussed.  相似文献

A method for the isolation of the Golgi apparatus from stem tissues of onion is described. Preparations that consisted mainly of morphologically identifiable Golgi apparatus have been obtained. The best preparations were obtained from tissue homogenized under conditions of minimum shear, and in the presence of sucrose and certain additives which aid in preservation of the integrity of the Golgi membranes. Those additives, which had a pronounced stabilizing effect on the isolated apparatus, included both monovalent and divalent ions (sodium and calcium) and dextran. A large portion of the Golgi apparatus did not appear to change microscopic appearance upon isolation, but were observed to fuse into large aggregate structures not unlike those occurring naturally in certain animal or insect cells (12). Fusion occurred both at the edges of the cisternae and in register, but the integrity of the individual cisternae was not destroyed. The major contaminants of the Golgi apparatus fraction were numerous small and large spherical vesicles. At least some of these vesicles appeared to have been derived from the Golgi apparatus; others may have been fragments of the cell membrane, the endoplasmic reticulum, or other cell debris. By utilizing this procedure, it has been possible to obtain fractions of Golgi apparatus from plant tissues other than onion stem. However, at the present time it is only with onion that the Golgi apparatus has been isolated in a form that would warrant further purification for biochemical analysis.  相似文献

Inclusions were found in epidermal plastids of Beta vulgaris L., by the use of the electron microscope. One type of inclusion resembles the membrane-bound plastid inclusions found by others in various meristematic plant cells; the other does not appear to have been described before. The second type of inclusion is not membrane-bound and appears elongated and fibrillar in longisection. The elongated plastid inclusion sometimes becomes quite large. Details are given of the elimination of mitochondria which occurs during the first 1 to 2 hours of the life of the egg of the fern Pteridium aquilinum (L.) Kuhn. During this phase the only mitochondria present are swollen and appear degenerate, but subsequently the cytoplasm of the egg becomes filled with large mitochondria containing numerous villi. Accompanying the appearance of these mitochondria, many, if not all of which have a peculiar umbo-like form, is the production by the nucleus of conspicuous and complex evaginations. The umbo-mitochondria are believed to be new, and a mechanism is suggested by which they may be generated from the complex evaginations of the nucleus.  相似文献

Lens-shaped wall papillae, resembling those known to form in response to fungi or mechanical damage, occur in root epidermal cells of axenically grown seedlings of Zea mays. Papillae are most common in the tabular epidermal cells but also occur in younger cells. Not all tabular cells have papillae, and they are more frequent in some seedlings. Where present, there is usually only one papilla per cell and it lies against the outer periclinal wall just proximal to an emerged root hair or near the position where a hair would be expected to form. Electron micrographs show that a papilla is structurally heterogeneous. Papillae fluoresce strongly in the presence of aniline blue even in freeze-substituted material.  相似文献
