The cell envelopes of Cryptomonas and Chroomonas exhibited significant fluorescence using FITC-labelled concanavalin A and wheat germ agglutinin when the cells were fixed prior to lectin binding. The periplast became intensely labelled in Chroomonas whereas Cryptomonas showed fluorescing granula in its gullet/furrow region and on the cell surface. Lectin labelling followed by fixation showed only label of periplast remnants of lysed cells and of the flagella of Chroomonas. Isolated periplasts of Cryptomonas and Chroomonas were intensively labelled with both concanavalin A and wheat germ agglutinin. Glycostaining of gels, onto which total cell protein extracts were loaded, showed a glycoprotein of high molecular weight for Cryptomonas and Chroomonas and an additional glycoprotein for Cryptomonas species.  相似文献

Summary A comparative action spectroscopical study was made on phototaxis in two genera of cryptomonads (cryptophyte flagellate algae), namely,Cryptomonas (rostratiformis) andChroomonas (nordstedtii andcoeruled). The two genera differ in their characteristic phycobilin pigmentation and, among three species, onlyChroomonas coerulea possesses an eyespot. The two species with no eyespot,Cryptomonas rostratiformis andChroomonas nordstedtii, exhibited positive phototaxis, showing very similar action spectra characterized by a broad band in the region from 450 nm to 650 nm, with an action maximum at about 560 nm; these features are essentially the same as those observed previously forCryptomonas strain CR-1. InCryptomonas rostratiformis, a small peak was also found at 280 nm in the UV-B/C region.Chroomonas coerulea, with eyespot, did not exhibit distinct positive phototaxis in a wide spectral region at any given, even very low, light intensity, but exhibited negative phototaxis of spectral sensitivity maximal at 400–450 nm. These results indicate that the positive phototaxis ofCryptomonas (rostratiformis and CR-1) andChroomonas nordstedtii is mediated by the same, yet unidentified photoreceptor(s).Chroomonas nordstedtii, possessing no phycoerythrin absorbing at 545 nm, also exhibits positive phototaxis at ca. 560 nm, and this result disfavors the so far proposed possibility that the positive phototaxis of the cryptophytes may be mediated by phycobilin pigments. On the other hand, the spectral characteristics of negative phototaxis ofChroomonas coerulea can possibly be ascribed to the presence of an eyespot.  相似文献

The fluorescence of purified biliproteins (phycocyanin 645, phycocyanin 612, and phycoerythrin 545) from three cryptomonads, Chroomonas species, Hemiselmis virescens, and Rhodomonas lens, and C-phycocyanin from Anacystis nidulans has been time resolved in the picosecond region with a streak camera system having less than or equal to 2-ps jitter. The fluorescence lifetimes of phycocyanins from Chroomonas species and Hemiselmis virescens are 1.5 +/- 0.2 ns and 2.3 +/- 0.2 ns, respectively, regardless of the fluence of the 30 ps, 532-nm excitation pulse. (Fluence [or photons/cm2] = f intensity [photons/cm2s]dt.). In contrast, that of C-phycocyanin is 2.3 +/- 0.2 ns when the excitation fluence is 8.2 X 10(11) photons/cm2 and decreases to a decay approximated by an exponential decay time of 0.65 +/- 0.1 ns at 7.2 X 10(16) photons/cm2. The cryptomonad phycoerythrin fluorescence decay lifetime is also dependent on intensity, having a decay time of 1.5 +/- 0.1 ns at low fluences and becoming clearly biphasic at higher fluences (greater than 10(15) photons/cm2). We interpret the shortening of decay times for C-phycocyanin and phycoerythrin 545 in terms of exciton annihilation, and have discussed the applicability of exciton annihilation theories to the high fluence effects.  相似文献

以亚热带黄村水库流域为研究对象,探讨了浮游植物群落的季节变化及其与环境因子的关系。共鉴定出浮游植物51种,种类最多的是绿藻门(18种)(35.29%),其次是硅藻门(15种)(29.41%)和蓝藻门(11种)(21.57%)。从浮游植物群落季节组成差异来看,春季(62.96%)和夏季(71.03%)蓝藻门丰度最高,秋季(56.11%)和冬季(80.74%)硅藻门丰度最高。黄村水库流域优势种尖尾蓝隐藻(Chroomonas acuta)和曲丝藻(Achnanthidium sp.)在春夏秋冬季节均存在。多样性指数表明该流域水体处于中营养状况。温度、电导率和营养盐是影响浮游植物群落组成最主要的环境因素。  相似文献

Clones of Cryptomonas phaseolus Skuja , Cryptomonas rostratiformis ( Skuja ) Skuja in Huber-Pestalozi, and Cryptomonas undulata Gervais were isolated from the deep chlorophyll maximum near the oxic/anoxic boundary layer of the mesoeutrophic lake Schlachtensee, Germany. Different autecological features of these species were studied in batch culture experiments . Cryptomonas cf . ovata Ehrenberg and Chroomonas sp. that never dominated in the deep chlorophyll layer were also isolated from Schlachtensee to study their light-dependent growth in comparison to the deep-living species . Cryptomonas undulata, C. cf . ovata, and C. phaseolus had a very low light compensation point ( 5–7 μmol.m-2.s-1 ), whereas the growth rate of Chroomonas sp. and C. rostratiformis was positive above 16 and 24 μmol.m-2.s-1 . Cryptomonas phaseclus and Chroomonas sp. became photoinhibited above photon flux densities of 92 and 116 μmol.m-2.s-1 . Cryptomonas rostratiformis, C. cf . ovata, and C. undulata reached a maximum growth rate at a considerably higher photon flux density (198–250 μmol.m-2.s-1 ). Cryptomonas phaseolus grew fastest under light-limiting conditions . Chyptomonas phaseolus and C. undulata were best able to suruive prolonged periods of darkness . Cryptomonas phaseolus, C. rostratiformis, and C. undulata did not show any uptake of fluorescent latex beads. When labeled glucose was provided in naturally occurring concentrations, carbon uptake by C. phaseolus, C. rostratiformis, and C. undulata was negligibly small in comparison to cellular carbon content. I suggest that the adaptation to a low-light environment is an important preadaptation for the dominance of C. phaseolus and C. undulata near the freshwater chemocline .  相似文献

The species composition of phytoflagellates in the Salton Sea has recently been the subject of intense investigation as part of an analysis of the Salton Sea ecosystem. The Salton Sea, an inland sea occupying 980 km2 in southern California, has become a major stopping point for migratory birds along the Pacific Flyway. The increasing salinity of the sea, currently at 44 gm L−1, and its eutrophic condition (average depth is nine meters, with a high nutrient load contributed by agricultural drainage from the surrounding farmlands) have contributed to a stressed ecosystem. Massive fish kills and bird kills, including such endangered birds as the brown pelican, have become a recurring problem. Although previous investigations have noted the presence of at least two phytoflagellates implicated in fish mortality, little attention has been paid the to the identities of the smaller flagellates observed growing in the sea and their possible contribution to the fish and bird population mortality. Using freshly collected field samples as well as enrichment culture techniques, we report the occurrence of several genera of cryptomonads in the Salton Sea, including representatives from the genera Chroomonas , Hemiselmis , Leucocryptos , Plagioselmis , Storeatula and Teleaulax.  相似文献

The trophic interactions of the marine rotifer Synchaeta cecilia were investigated by determining its feeding and growth rates on a wide variety of marine phytoplankton and by determining its susceptibility to predation by the calanoid copepod, Acartia tonsa. Reproduction of S. cecilia was sustained in four-day feeding trails by 13 of 37 algal species tested. Growth-supporting species included species of Cryptophyceae, Dinophyceae, Chlorophyceae and Haptophyceae in sizes from 4 to 47 μm. Within these taxa, other species in the acceptable size range failed to support growth. No species of Cyanophyceae, Bacillariophyceae, or Chrysophyceae supported growth of the rotifer. S. cecilia can be maintained on unialgal cultures of Cryptophyceae but growth is enhanced by a combination of two or three species; a mixture of Chroomonas salina (Cryptophyceae), Heterocapsa pygmaea (Dinophyceae), and Isochrysis galbana (Haptophyceae) has sustained laboratory stocks of S. cecilia for over four years. The expected response of S. cecilia to food quantity was observed: as food concentration was increased from 58 to 1154 μg C 1−1, the population growth constant increased from 0.17 to 0.60 d−1 at 20°C. This is equivalent to population doubling times of 4.0 and 1.1 days at H. pygmaea densities of 500 and 104 cells ml−1, respectively. The susceptibility of S. cecilia to predation was investigated by determining its rate of capture by the omnivorous marine copepod Acartia tonsa. At prey densities of 5 to 35 μg C 1−1 (0.3 to 1.9 individuals 1−1), A. tonsa readily ingested S. cecilia at rates up to 3 μg C copepod−1 day−1.  相似文献

Phytoplankton were collected over five austral summers (1987–88through 1991–92) to examine seasonal and annual fluctuationsin species composition and biovolume in Lake Fryxell, a perenniallyice-covered lake located in the Dry Valleys of South VictoriaLand, Antarctica. Lake Fryxell has perennial gradients of salinity,dissolved oxygen and nutrients. We found that algal speciesdiversity was low (56 taxa were collected), confirming the resultsof previous short-term studies. The phytoplankton consistedprimarily of cryptophyte and chlorophyte flagellates, and filamentouscyanobacteria. The presence of filamentous cyanobacteria, whichhave not been reported as abundant in this lake by previousworkers, may represent a significant ecological change. Eachaustral summer, one dominant species contributed >70% ofthe total biovolume; Chroomonas lacustris was dominant in 1987–88,while Cryptomonas sp. dominated the phytoplankton in the remaining4 years. No species succession occurred during the austral summer.Some common taxa were vertically stratified (Oscillatoria limnetica,Phormidium angustissimum, Pyramimonas sp., Oscillatoria sp.),while others showed no distinct vertical stratification (Chlamydomonassubcaudata, Cryptomonas sp.). The stratification of the phytoplanktonreflects the gradients of nutrients and light, and the stabilityof the water column.  相似文献

A quick-freezing technique for freeze fracturing was used to determine periplast plate types in 20 cryptomonads. With this technique cells are frozen so rapidly that major artifacts are eliminated. We propose that periplast plates are attached to the cell membrane by intramembrane particles (IMP's), consequently plate shapes are outlined by IMP distribution in fractured membranes. Round to oval, sometimes slightly angular, plates occur in Cryptomonas ovata, Cryptomonas tetrapyrenoidosa, Cryptomonas parapyrenoidifera, Cryptomonas obovata, Cryptomonas erosa and two unidentified species of Cryptomonas; large rectangular plates occur in Chroomonas pochmannii, Chroomonas coerulea and Hemiselmis sp.; small rectangular plates were found in Cryptomonas sp. (Strain SDB); square to slightly rounded plates occur in Cryptomonas chrysoidea and a single continuous plate or sheet, perforated by ejectisome pores, was observed in Cryptomonas caudata, Cryptomonas rostratiformis, Cryptomonas marssonii, Cryptomonas platyuris, Cryptomonas curvata, Cryptomonas ozolini, Chilomonas paramecium and Rhodomonas sp. Oval and square plates are described for the first time in Cryptomonas. Plate IMP's may be morphologically modified in size and shape, depending upon their location in relation to the plate, the plate ridges, and ejectisome chambers. Conformational changes in plate shapes, to form hexagons or polygons, may be induced when cells are subjected to fixation, desiccation, cryoprotectants or centrifugation.  相似文献

A new kleptoplastidal dinoflagellate, Gymnodinium myriopyrenoides sp. nov., was described using light microscopy, electron microscopy and phylogengetic analysis based on partial LSU rDNA sequences. Cells were dorsiventrally flattened, elongate-elliptical in ventral view. There was no displacement of the cingulum encircling the anterior part of the cell. The cingulum was curved posteriorly at the terminal junction with the sulcus. The sulcus was generally narrow but expanded in the posterior end. The epicone possessed an apical groove made of one and one-half counterclockwise revolutions. Phylogenetic analysis based on LSU rDNA showed that the sequence of G. myriopyrenoides was included in the Gymnodiniales sensu stricto clade and had special affinities with the species Amphidinium poecilochroum and Gymnodinium acidotum, which also harbor kleptochloroplasts. Phylogenetic analysis based on plastid-encoded SSU rDNA and ultrastructural observations suggested that the symbionts of G. myriopyrenoides were cryptophytes of the genus Chroomonas or Hemiselmis. Organelles including the nucleus, the nucleomorph, mitochondria, Golgi bodies and large chloroplasts remained in the cytoplasm of the symbionts, but not the periplast, ejectosomes or flagellar apparatus. The symbiotic level of G. myriopyrenoides was estimated to be a relatively early stage in the unarmored kleptoplastidal dinoflagellates.  相似文献

A specimen collected in southern Tibet in 1928, hitherto identified only as "Arenanu sp.", is found to represent a new species of the subgenus Odontostemma . The new species resembles A. littledalei , the species that forms the basis of the monotypic genus Gooringia , characterized by a dwarf habit, a tetramerous perianth and only two or three stamens. Several other species of Arenaria subgenus Odontostemma resemble A. littledalei in habit and one ( A. saginoides ) is tetramerous, but hitherto none has been found with its extremely reduced androecium. The new species has an identical androecium to A. littledalei and its discovery leaves no doubt that Gooringia should not be recognized taxonomically at any rank. The new species is described as A. pharensis McNcill & Majumdar and a table compares its diagnostic features with those of A. littledalei and five other dwarf Arenaria species of sibgrnus Odontostemma from the mountains of Sikkim, Tibet, Yunnan and Tsinghai.  相似文献

SYNOPSIS. A new carotenoid, manixanthin , was isolated from a bleached, autolysed culture of the marine cryptomonad Chroomonas salina grown on glycerol in light, after the culture showed disappearance of the chlorophylls and carotenoids normally produced. Manixanthin lacked carbonyl, ester, and epoxy groups in chemical tests, whilst its spectroscopic properties indicated a conjugated nonaene chromophore. In its chromatographic mobility, manixanthin closely resembled alloxanthin, the major normal xanthophyll of the cryptomonad. The iodine-catalysed photostereomutation of manixanthin produced the same equilibrium mixture of cis -isomers as that given by similar treatment of alloxanthin. This mixture of isomers completely lacked the all-trans alloxanthin and contained ∼60% manixanthin. It was concluded that manixanthin is an unusually stable cis -isomer of alloxanthin, and is presumably produced in the culture medium by the action of light on alloxanthin released by autolysed cells of the cryptomonad. The unusual stability of manixanthin relative to alloxanthin was investigated by effecting partial stereoisomerizations under conditions using either of heat, light, or iodine, and evidence was obtained for a series of at least 10 isomers with a trend of stability increase from alloxanthin to manixanthin. It was inferred that manixanthin may be 9,9'-di cis -alloxanthin.  相似文献

We investigated the empirical relationship between mean abundance and its variance, known as Taylor’s power law, in fleas parasitic on small mammals. It has been suggested that the exponent of this function, b, represents a true biological character of a species and, dependent on the level of host specificity, varies among species. Other empirical and theoretical studies suggest that exponent b depends on interspecific competition and varies intraspecifically. We tested these hypotheses using data from central and eastern Slovakia. We demonstrate that the slope of Taylor’s relationship (a) is repeatable within a flea species, i.e. the slope represents a true species character; (b) increases with an increase of the degree of flea host specificity; and (c) decreases with an increase in flea community size. Fifteen freshwater cryptomonad species were freeze-dried and examined with the scanning electron microscope. Surveys of cell surfaces revealed four general cell types. Chroomonas type cells lack a furrow but possess a shallow vestibular depression where the flagella are inserted. The presence of a gullet could not be detected. Cryptomonas spp. displayed three morphological types, all lacking gullets. The first type of Cryptomonas has a simple, shallow furrow with ridges that apparently can close to form a raphe but an oval opening or stoma remains at the posterior end and an opening from the vestibulum is formed at the anterior end. The second Cryptomonas type consists of a complex furrow with furrow ridges and folds that extend almost two-thirds of the cell length. A sloma is present in the central region of the closed furrow. The folds apparently can separate thereby exposing the underlying furrow. The third type of Cryptomonas possesses a simple, non-closing furrow. At the anterior end there is a vestibular ligule which extends from the dorsalleft side of the cell and covers the region of the vestibulum where the contractile vacuole discharges.  相似文献

2008年12月至2009年10月按季度对养殖湖泊大通湖浮游植物群落结构及与环境因子关系进行了初步研究,并通过与1960年数据的纵向比较,探讨了集约化养殖对大通湖水域环境的影响,为其渔业可持续发展提供理论依据.调查期间,共发现浮游植物98种,隶属于7门54属.优势种类包括二角多甲藻、尖尾蓝隐藻、小球藻、卵形隐藻、梅尼小环藻、啮蚀隐藻、卷曲鱼腥藻、铜绿微囊藻和固氮鱼腥藻,优势种的季节更替明显.浮游植物细胞密度的年平均值为1.84×106 cells·L-1,其中夏季最高,为16.4×106 cells·L-1,其他3个季度在1.71×106 ~ 1.98×106 cells·L-1.浮游植物丰富度指数(D)全年在2.01 ~4.55,Shannon多样性指数(H)在1.26 ~2.69,均匀度指数(J)在0.69~1.27.典范对应分析表明,水深、水温、透明度、总磷、氧化还原电位、电导率是影响大通湖浮游植物群落结构的主要因子.  相似文献

Bunias orientalis is an alien species that has spread greatly in recent decades in Central Europe. This coloniser is particularly well (pre)adapted to anthropogenic disturbances and human-mediated dispersal, allowing it to spread mainly along roads and railways. Here, its distribution and population structure is investigated for the sub-montane region located in the northern foothills of the Tatra Mts. Based on 70 phytosociological samples of invading vegetation, the relationships between B. orientalis cover and species diversity, together with the composition of vegetation, were analysed using regression analysis. Comparison of the current distribution of B. orientalis within the investigated area with published records indicates that the expansion rate of this alien species is not particularly high, despite its increased frequency in anthropogenic habitats and meadows. Meadows invaded by this species revealed that increased B. orientalis cover was accompanied by a significant reduction in the values of the Inverse Simpson diversity index and Pielou’s measure of evenness. Reduced diversity was caused by considerable decrease in cover by species of the Molinio-Arrhenatheretea class. However, for anthropogenic habitats, no statistically significant relationships were found to occur between the abundance of B. orientalis, species diversity and vegetation composition.  相似文献
