Local and chemical distribution of phlorotannins in brown algae   总被引:4,自引:2,他引:2  
The local and chemical distribution of phlorotannins among the Japanese Laminariaceae, Eisenia bicyclis, Ecklonia cava and Ecklonia kurome, was investigated. As a result of light microscopy observations with vanillin-HCl staining, phlorotannins were found to be accumulated within the vegetative cells of the outer cortical layer of the thalli, regardless of the species, stage of growth or organ. Crude phlorotannins comprised about 3.0% of the algal powder for each of the algae. High-performance liquid chromatography (HPLC) showed that the phlorotannins of E. bicyclis were composed of phloroglucinol (0.9%), phloroglucinol tetramer (4.4%), eckol (7.5%), phlorofucofuroeckol A (21.9%), dieckol (23.4%), and 8,8'-bieckol (24.6%), plus some other unknown phenolic compounds (17.3%). The composition of the phlorotannins differed little among the Laminariaceae, except for a significantly larger amount of the tetramer, MW 478, in E. bicyclis.  相似文献

We investigated, for the first time in a microscopic study, the accumulation of phlorotannins as a possible inducible chemical defence against herbivory. Ecklonia radiata (C. Agardh) was mechanically wounded with a cork borer, simulating grazer action, and at intervals of 0, 1, 2, 3, 5, 7 and 9 days after wounding the distribution of phlorotannins and other structural changes were examined by light, fluorescence and electron microscopy. In brown algal cells, phlorotannins (polyphenolic compounds) occur in vesicles known as physodes. In E. radiata, most of the physodes were found in the outer epidermal cell layer, but some were present in the cortical cells and in the innermost medullary cells (sieve elements and hyphal cells). The wound-healing process could be divided into three stages: (i) closing of the medulla by the formation of new medullary cells, (ii) accumulation of phlorotannins (physodes) at the wound area (first in the medullary cells and then in the cortical cells) and in the medullary tissue further away from the wound, and (iii) formation of a new epidermis. The accumulation of phlorotannins started on day 1 and was evident from day 3 on. Our results show structural wound-healing and support wound-sealing functions for phlorotannins and the view that phlorotannins might be considered as inducible anti-herbivory agent in E. radiata. Our results strongly demonstrate the importance of detailed microscopic studies, in addition to chemical analysis, for revealing the localised nature of the brown algal response to wounding.  相似文献

The chemical structures of ultraviolet (UV)- absorbing substances secreted from the healthy living brown algae, Eisenia bicyclis and Ecklonia kurome, were demonstrated. The living activity of algal cells was critically examined using a confocal laser-scanning microscope after incorporation of fluorescein diacetate (FDA) into the cells. Using thin-layer chromatography (TLC), reversed-phase three-dimensional high-performance liquid chromatography (RP-3D-HPLC) and gas chromatography-mass spectrometry (GC-MS), it was found that the UV-absorbing substances (λmax 265–270 nm) secreted from the living brown algae mainly consisted of the three monomeric bromophenols, 2,4-dibromophenol, 2,4,6-tribromophenol and dibromo-iodophenol, but not phloroglucinol or phlorotannins. The other minor compounds detected in the secretions were as follows: benzothiazole, fatty acids (14:1, 16:0 and 18:0 acids), franesol, 3-hydroxy-2,4,4-trimethylpentyl 2-methylpropanoate and squalene. Exudation of phloroglucinol and phlorotannins was ascertained to take place only after the cell death of these algae. These results indicate that, whilst the algae are alive, polymeric phlorotannins are strictly kept within the algal body, and only monomeric bromophenols are secreted into the seawater medium.  相似文献

To screen algal phlorotannins with antioxidative activities, 50% ethanol extracts of 25 Japanese marine algae were evaluated. Scavenging activity against superoxide anion radicals was frequently found with a high content of total phenolic compounds. Among these, the extract from the brown seaweed, Sargassum ringgoldianum, showed the strongest scavenging activity. The active fraction contained a mixture of high molecular weight polyphenols, phlorotannins that were found to be polymerized bifuhalol, as analyzed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). The scavenging activity of the fraction against superoxide anion radicals was estimated to be 1.0 μg/ml (IC50), which were approximately five times stronger than that of catechin.  相似文献

The crude phlorotannins from the brown alga Eisenia bicyclis showed inhibitory activity against 10 of 13 kinds of glycosidases present in the viscera of the turban shell Turbo cornutus. Phloroglucinol and its oligomers – eckol (a trimer), phlorofucofuroeckol A (a pentamer), dieckol and 8,8′-bieckol (hexamers), and an unidentified tetramer – were isolated from the crude phlorotannins by column and thin-layer chromatography. Phlorofucofuroeckol A, dieckol and 8,8′-bieckol inhibited α-fucosidase, β-galactosidase and β-mannosidase partially purified from T. cornutus, while phloroglucinol, eckol and the unidentified tetramer were weakly active. Dieckol was a competitive inhibitor of α-fucosidase with an inhibition constant (K i) of 0.12?mM. The amounts of phlorotannins released after the immersion of freshly collected E. bicyclis in seawater or deionized water were estimated by high-performance liquid chromatography. Nearly all the phlorotannins were exuded into the medium following the death of the algae, whereas no phlorotannins were detected in the medium of living algae. These findings indicate that the phlorotannins deter the feeding of marine herbivorous gastropods by inhibiting the glycosidases.  相似文献

The antioxidant activities of brown algal phlorotannins were evaluated using the inhibition of phospholipid peroxidation in the liposome system, and by determining radical scavenging activities against the superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH). Oligomers of phloroglucinol (1,3,5-trihydroxybenzene), eckol (a trimer), phlorofucofuroeckol A (a pentamer), dieckol and 8,8′-bieckol (hexamers), isolated from the Laminarian brown algae Eisenia bicyclis, Ecklonia cava and Ecklonia kurome, showed potent inhibition of phospholipid peroxidation at 1 μM in the liposome system. The phlorotannins had significant radical scavenging activities against the superoxide anion (50% effective concentration values: 6.5–8.4 μM) and DPPH (50% effective concentration values: 12–26 μM), and were more effective compared to ascorbic acid and α-tocopherol. For the purpose of using phlorotannins as functional food ingredients, the antioxidant activity of a complex of crude phlorotannins and soybean protein was examined. The complex had a pronounced DPPH-radical scavenging activity. These results suggest that phlorotannins are potent anti-inflammatory substances, and that the Laminariaceous brown algae, which are abundant in phlorotannins, may be useful as a new functional foodstuff or supplement with anti-inflammatory activity.  相似文献

The inhibitory effects of high molecularweight phlorotannins (HMP) from Sargassum kjellmanianum on mouse liverlipid peroxidation were investigated byspectrophotometric methods. The content ofmalondialdehyde (MDA) in liver samples wasmeasured by TBA (thiobarbituric acid)assay. It showed that HMP significantlyinhibited the generation of MDA invivo and in situations induced byCCl4 and Fe2+-Vc (ascorbic acid),and significantly decreased membraneswelling of mouse liver mitochondria,compared with controls (p<0.01). It has been proposed that seaweed secondary metabolites, e.g. brown algal phlorotannins, may have an ecologically important function as a chemical defence against epiphytes, by acting against colonisation of epiphytic organisms. We tested whether the low epiphytic abundance on the invasive brown seaweed Fucus evanescens, compared to the congeneric F. vesiculosus, is due to a more effective chemical defence against epiphyte colonisation. A field survey of the distribution of the common fouling organism Balanus improvisus (Cirripedia) showed that the abundance was consistently lower on F. evanescens than on F. vesiculosus. However, contrary to expectations, results from experimental studies indicated that F. vesiculosus has a more effective anti-settlement defence than F. evanescens. In settlement experiments with intact fronds of the two Fucus species, both species deterred settlement by barnacle larvae, but settlement was lower on F. vesiculosus both in choice and no-choice experiments. Phlorotannins from F. vesiculosus also had a stronger negative effect on larval settlement and were active at a lower concentration than those from F. evanescens. The results show that Fucus phlorotannins have the potential to inhibit settlement of invertebrate larvae, but that settlement inhibition cannot explain the lower abundance of the barnacle Balanus improvisus on F. evanescens compared to F. vesiculosus. Assessment of barnacle survival in the laboratory and in the field showed that this pattern could instead be attributed to a higher mortality of newly settled barnacles. Observation suggests that the increased mortality was due to detachment of young barnacles from the seaweed surface. This shows that the antifouling mechanism of F. evanescens acts on post-settlement stages of B. improvisus.  相似文献

The inhibitory effects of brown algal phlorotannins on secretory phospholipase A2s (sPLA2s), lipoxygenases (LOXs) and cyclooxygenases (COXs) were determined with an in vitro assay. Oligomers of phloroglucinol; eckol (a trimer), phlorofucofuroeckol A (a pentamer), dieckol (a hexamer) and 8,8-bieckol (a hexamer) isolated from the brown alga Eisenia bicyclis had pronounced inhibitory effects on sPLA2 from porcine pancreas and bee venom (IC50 100–200 M). The phlorotannins inhibited LOX activity more effectively than the well-known LOX inhibitors; resveratrol and epigallocatechin gallate. 8,8-Bieckol, the strongest LOX inhibitor in this study, inhibited soybean LOX and 5-LOX with IC50 values of 38 and 24 M, respectively. Negligible or very weak effects of the phlorotannins on COX-1 and COX-2 were found, except for an inhibitory effect of dieckol on COX-1 (74.7%) and of eckol on COX-2 (43.2%) at 100 M.  相似文献

Aqueous extracts from 33 species of marine algae were assessed for their methyl mercaptan-trapping activity by gas chromatography to search for novel natural oral deodorants. Brown algae belonging to the Laminariales such as Eisenia bicyclis, Ecklonia cava and Ecklonia kurome were found to show remarkable deodorizing action against methyl mercaptan. The effective components in Eisenia bicyclis were identified as a phlorotannin, a group of molecules which are characteristic components of Laminariales. In addition phlorotannins extracted from E. bicyclis were more effective at reducing methyl mercaptan than conventional natural deodorants such as chlorophyll and sodium copper chlorophyllin.Author for Correspondence  相似文献

We examined factors affecting the abundance and distribution of epiphytes (fouling) on the sublittoral kelp Ecklonia radiata. We first assessed the importance of phlorotannins as chemical defences against epiphytes by (a) correlating epiphyte loads on different parts of the thallus with the phlorotannin content of those tissues, and (b) experimentally testing the effects of variation in phlorotannin concentration against the settlement and growth of gametes of Ulva lactuca, a common epiphyte in the system. Tissue phlorotannin content was, at best, only weakly related to epiphyte loads, with r 2 values typically <0.10. Inhibition of Ulva gametes only occurred at concentrations >10 mg l−1, which is 5 orders of magnitude greater than levels of phlorotannins in the water column around beds of E. radiata, and 1–3 orders of magnitude greater than estimated levels in the boundary layer at the surface of the plant. We concluded that phlorotannins have a negligible impact on patterns of epiphytism on E. radiata, and proceeded to investigate other factors influencing the distribution and abundance of epiphytes. In our samples the relative age of different parts of the thallus was strongly correlated with epiphyte abundance, with epiphyte densities greatest on the oldest tissue and least on the youngest. Distal parts of the thalli also had greater epiphyte loads than basal parts. Field experiments in which kelp tissue was suspended at two heights in an E. radiata forest for varying lengths of time confirmed the importance of the length of time that the tissue was in the water, and its height in the water column, to the development of an epiphyte community. Comparison of epiphyte loads on tissue from primary (smooth) and secondary (rough) laminae in these experiments indicated that surface rugosity also affected fouling. Macroherbivores were rare on E. radiata, and abundances of mesofauna and epiphytes were positively related, suggesting that grazers were not important determinants of patterns of epiphyte abundance. Although phlorotannins have been previously suggested to play an important role as defences against epiphytes, we suggest that water-soluble compounds such as phlorotannins are less likely to be effective defences against epiphytes than non-polar metabolites, which can adhere to the surface of the producing organism.  相似文献

Phlorotannins are metabolites found only in the Phaeophyceae (brown algae) and have a variety of metabolic roles, including both primary (e.g. cell wall construction) and secondary (e.g. herbivore defence and UV protection). This study aimed to establish the level of variation of phlorotannins in two species of dominant Antarctic macroalgae (Desmarestia anceps and D. menziesii). Thirteen samples were taken from specific locations throughout the thallus of multiple individuals at two depths at three locations near Anvers Island, Antarctica. Overall average concentrations were 0.117±0.003 g g–1dwt in D. anceps and 0.052±0.002 g g–1dwt in D. menziesii. Concentrations varied greatly at the 13 sampling locations in each individual, but not in any consistent manner. Significant variation occurred between locations in both species and between depths in D. anceps.  相似文献

Phlorotannins are polyphenoloic metabolites occurring only in the Phaeophyceae that have numerous putative primary roles (e.g. cell‐wall construction and storage) as well as secondary metabolic roles, which include herbivore feeding deterrence and protection from UV radiation. The proposed role of phlorotannins in the defense against UV radiation is of particular importance in the Antarctic due to depletion of the stratospheric ozone layer in that area. Several studies of brown algae have found evidence of an induction response (the production of defensive metabolites, including phlorotannins) after grazing by various mesograzers, after simulated grazing/wounding, and after exposure to increases in UV radiation. This study aimed to determine if phlorotannin production or other defenses in two dominant, endemic Antarctic species (Desmarestia menziesii Montagne and Desmarestia anceps J. Agardh) could be induced by an increase in exposure to UV radiation or by natural and artificial grazing. An in situ experiment failed to detect any effect of UV radiation on phlorotannin concentrations in either species or on subsequent palatability in feeding bioassays. A laboratory‐based experiment did not detect any effect of mesoherbivore grazing or simulated grazing (wounding) on palatability or the concentration of phlorotannins in D. menziesii. Instead, phlorotannin concentrations increased in all treatments in both experiments, consistent with an increase in overall resource availability due to an increase in available PAR compared with the in situ irradiance at the algal collection sites.  相似文献

The present work investigates protein tyrosine phosphatase 1B (PTP1B) and the α-glucosidase inhibitory activities of two edible brown algae, Ecklonia stolonifera and Eisenia bicyclis, as well as in their isolated phlorotannins. Since the individual extracts and fractions showed significant inhibitory activities, column chromatography was performed to isolate six phlorotannins, phloroglucinol (1), dioxinodehydroeckol (2), eckol (3), phlorofurofucoeckol-A (4), dieckol (5), and 7-phloroeckol (6). Phlorotannins 3–6 were potent and noncompetitive PTP1B inhibitors with IC50 values ranging from 0.56 to 2.64 μM; 4–6 exhibited the most potent α-glucosidase inhibition with IC50 values ranging from 1.37 to 6.13 μM. Interestingly, 4 and 6 were noncompetitive, while 5 exhibited competitive inhibition in an α-glucosidase assay. E. stolonifera and E. bicyclis as well as their isolated phlorotannins therefore possessed marked PTP1B and α-glucosidase inhibitory activities; this could lead to opportunities in the development of therapeutic agents to control the postprandial blood glucose level and thereby prevent diabetic complications.  相似文献

Most theories for the evolution of plant chemical defences assume that defences are costly. In order to investigate the potential cost of phlorotannin production in the brown alga (kelp) Ecklonia radiata, I measured the correlation between changes in phlorotannin levels and growth rates for E. radiata in two seasons, spring and autumn. In spring, when both phlorotannin levels and growth rates in E. radiata were highest, there was a significant negative relationship between changes in phlorotannin levels and growth, consistent with a cost of phlorotannin production. No relationship was evident in autumn, indicating that moderate levels of phlorotannins and growth could be maintained simultaneously. These and related results suggest that the cost of defence will vary as a function of (1) endogenous patterns of growth or metabolite production and (2) variation in available resources. Evidence for a cost of phlorotannins in E. radiata adds to the paradox of the maintenance of high levels of phlorotannins in Australasian brown algae.  相似文献

Herbivory on marine macroalgae (seaweeds) in temperate areas is often dominated by relatively small gastropods and crustaceans (mesoherbivores). The effects of these herbivores on the performance of adult seaweeds have so far been almost exclusively investigated under artificial laboratory conditions. Furthermore, several recent laboratory studies with mesoherbivores indicate that inducible chemical resistance may be as common in seaweeds as in vascular plants. However, in order to further explore and test the possible ecological significance of induced chemical resistance in temperate seaweeds, data are needed that address this issue in natural populations. We investigated the effect of grazing by littorinid herbivorous snails (Littorina spp.) on the individual net growth of the brown seaweed Ascophyllum nodosum in natural field populations. Furthermore, the capacity for induced resistance in the seaweeds was assessed by removing herbivores and assaying for relaxation of defences. We found that ambient densities of gastropod herbivores significantly reduced net growth by 45% in natural field populations of A. nodosum. Seaweeds previously exposed to grazing in the field were less consumed by gastropod herbivores in feeding bioassays. Furthermore, the concentration of phlorotannins (polyphenolics), which have been shown to deter gastropod herbivores, was higher in the seaweeds that were exposed to gastropod herbivores in the field. This field study corroborates earlier laboratory experiments and demonstrates that it is important to make sure that the lack of experimental field data on marine mesoherbivory does not lead to rash conclusions about the lack of significant effects of these herbivores on seaweed performance. The results strongly suggest that gastropods exert a significant selection pressure on the evolution of defensive traits in the seaweeds, and that brown seaweeds can respond to attacks by natural densities of these herbivores through increased chemical resistance to further grazing.  相似文献

Phlorotannins, C-based defence compounds in brown seaweeds, show a high degree of spatial and temporal variation within seaweed species. One important model explaining this variation is the Carbon Nutrient Balance Model (CNBM), which states that the relative supply of carbon and limiting nutrients will determine the level of defence compounds in plants. Nitrogen is often considered to be the limiting nutrient for marine macroalgal growth and the CNBM thus predicts that when the carbon:nitrogen ratio is high, photosynthetically fixed carbon will be allocated to production of phlorotannins. In the present study, we evaluated the effects of light (i.e. carbon) and nitrogen on the phlorotannin content of two intertidal brown seaweeds, Ascophyllum nodosum and Fucus vesiculosus. This was done in an observational field study, as well as in a manipulative experiment where plants from habitats with different light regimes were subjected to different nitrogen and light treatments, and their phlorotannin content was measured after 14 days. The results showed that there was a negative relationship between tissue nitrogen and phlorotannin content in natural populations of F. vesiculosus, but not in A. nodosum. In the short term, the phlorotannin content in both algal species was not affected by changes in nitrogen availability. Exposure to sunlight had a positive effect on the phlorotannin content in natural populations of both algal species but, in the manipulative experiment, only F. vesiculosus showed a rapid response to changes in light intensities. Plants subjected to sunlight contained higher phlorotannin content than shaded plants. In conclusion, the results imply that nitrogen availability explains some of the natural variation in the phlorotannin content of F. vesiculosus, but the light environment has greater importance than nitrogen availability in predicting the phlorotannin content of each species.  相似文献

Chronic exposure of the skin to ultraviolet B (UVB) radiation induces oxidative stress, which plays a crucial role in the induction of skin aging. In this study, potential protective effect of extracts of six species of brown seaweeds on UVB radiation-induced cell damage was assessed via cell viability in HaCaT cells. The Ecklonia cava extract showed a profound protective effect against UVB radiation-induced cell damage, compared to that exhibited by other brown seaweed extracts. Thus, E. cava was selected for use in further experiments and was extracted with different solvents. The protection effect was evaluated via DCFH-DA, MTT, and morphological changes in HaCaT cells. Profound protective effect against UVB radiation-induced cell damage was detected in the 100% methanol extract. Partitioning of the 100% methanol extract with different organic solvents revealed a pronounced protective effect in the ethyl acetate fraction. The isolated active compounds were phlorotannins, especially dieckol, as identified using ultra-performance liquid chromatography-tandem mass spectrometry. Among the phlorotannins, dieckol showed a higher protective effect on UVB-induced cell damage in HaCaT cells than the other phlorotannins. Therefore, UVB protection of dieckol was evaluated via DCFH-DA, DAF-FM DA, acridine orange, and morphological changes in zebrafish model. Reactive oxygen species, nitric oxide, and cell death in live zebrafish induced by UVB radiation were reduced by the addition of dieckol. These results indicated that dieckol has potential protective effects on UVB-induced skin damage, which might be useful in pharmaceutical and cosmetic formulations.  相似文献

Herbivory is typically intense in marine littoral environments; thus, macrophytes are expected to evolve defenses against grazing. Although putative defenses of macrophytes are widely studied, there is lack of studies demonstrating the main premises of defense adaptations: the consequences of herbivory to macrophytes, genetic variation of defense traits and the costs and benefits of defenses in natural environment. We conducted a factorial experiment, where we manipulated amount of herbivory, growing depth and nutrient availability, and measured resistance to herbivory as well as genetic variation and costs of phlorotannins, putative defensive secondary metabolites, in the brown alga Fucus vesiculosus . Herbivory on algae varied with depth: grazing did not cause losses close to the surface, but, most of the algal production was consumed at the deeper end of the algal belt. The higher the genotypic phlorotannin content the less damage the genotype received implying that phlorotannins acted as a resistance trait. Production of phlorotannins was associated with costs for growth. Consistent with the prediction that the cost of defense will be greatest when resources are limiting, the cost appeared only in the deep end of the algal belt where growth was slowed down. Phlorotannins displayed phenotypic plasticity; the three factors influenced phlorotannins interactively, with the main tendencies of nutrient enrichment decreasing and herbivory and increasing depth increasing phlorotannins. Despite this plasticity, variation of phlorotannins was mainly due to the genotype of algae. These results emphasize the role of herbivory as a selective agent for algal defenses and the importance of genetic variation in the constitutive level of phlorotannins in interactions with natural enemies. The cost of phlorotannins may constrain the evolution of resistance in environments where growth is limited by light availability.  相似文献
