Heterotrophic cell suspension cultures of soya (Glycine max) and photomixotrophic cell suspension cultures of rape (Brassica napus) were incubated with cis-9-[1-14C]octadecenol for 3–48 h. It was found that under aerobic conditions large proportions of the alcohol are oxidized to oleic acid, which is incorporated predominantly into phospholipids, whereas up to 30% of the substrate is esterified to wax esters. This is true for both the heterotrophic and the photomixotrophic cell suspension cultures, but the metabolic rates are much higher in the latter. Under anaerobic conditions only small proportions of the radioactively labeled alcohol are oxidized to oleic acid, whereas a major portion of the alcohol is esterified to wax esters both in heterotrophic and photomixotrophic cultures. Incubations of homogenates of photomixotrophic rape cells with labeled cis-9-octadecenol showed that pH 6 is optimum for the formation of wax esters. This monounsaturated alcohol is preferred as a substrate over saturated longchain alcohols, whereas short-chain alcohols, cholesterol, and glycerol are not acylated. Incubations of an enzyme concentrate from a homogenate of rape cells with unlabeled cis-9-octadecenol and [1-14C]oleic acid, or [1-14C]stearoyl-CoA, or di[1-14C]palmitoyl-sn-glycero-3-phosphocholine showed that acylation of the longchain alcohol proceeds predominantly through acyl-CoA. Direct esterification of the alcohol with fatty acid as well as acyl transfer from diacylglycerophosphocholine could be demonstrated to occur to a much smaller extent.  相似文献

Long-chain aldehydes, alcohols, hydrocarbons and wax esters were major components of the external lipids of adult Aleyrodes singularis. In exuviae, acetate esters replaced the hydrocarbons as a major component. The major long-chain alcohol and aldehyde from adults were C32 and were essentially the exclusive components of the wax particles. The major alcohol from exuviae was C26 and the aldehydes were C26, C28, C30 and C32. The major acetate esters were C28 and C30 in both adults and exuviae. There were wax esters of similar carbon number in adults and exuviae although the exuviae had a greater amount of wax esters with unsaturated fatty acids. The fatty acid and alcohol composition of the wax esters differed markedly between adults and exuviae. Wax esters of adults had similar amounts of C16, C18, C20, C22 and C24 fatty acids while those from exuviae contained largely C16 and C18. The major alcohol in the wax esters of adults was C22 and those of exuviae were C26 and C28. The distribution of fatty acids and alcohols among wax esters of varying chain length also differed between adults and exuviae: in adults C22 was the major fatty acid found in the dominant wax ester, C44 and the C22 alcohol was the major alcohol and found in wax esters C42 and C44. In exuviae C16 and C18 were the major fatty acids found in most wax esters and a C28 alcohol was the major alcohol found in wax esters C44 and C46, the two dominant wax esters in exuviae. It was clear that the difference in chemistry of the wax esters between the adults and exuviae is not evident unless the acid and alcohol moieties are characterized.  相似文献

The surface of plants is covered by cuticular wax, which contains a mixture of very long-chain fatty acid (VLCFA) derivatives. This wax surface provides a hydrophobic barrier which reduces non-stomatal water loss. One component of the cuticular wax is the alkyl esters, which typically contain a VLCFA esterified to an alcohol of a similar length. As part of an EST project, we recently identified an acyltransferase with 19% sequence identity (amino acid) to a bacterial ‘bifunctional’ wax-ester synthase/diacylglycerol acyltransferase (WS/DGAT). Northern analysis revealed that this petunia homologue was expressed predominantly within the petals. The cDNA encoding the WS/DGAT homologue was introduced into a yeast strain deficient in triacylglycerol biosynthesis. The expressed protein failed to restore triacylglycerol biosynthesis, indicating that it lacked DGAT activity. However, isoamyl esters of fatty acids were detected, which suggested that the petunia cDNA encoded a wax-synthase. Waxes were extracted from petunia petals and leaves. The petal wax extract was rich in VLCFA esters of methyl, isoamyl, and short-to-medium straight chain alcohols (C4–C12). These low molecular weight wax-esters were not present in leaf wax. In-vitro enzymes assays were performed using the heterologously expressed protein and 14C-labelled substrates. The expressed protein was membrane bound, and displayed a preference for medium chain alcohols and saturated very long-chain acyl-CoAs. In fact, the activity would be sufficient to produce most of the low molecular wax-esters present in petals, with methyl-esters being the exception. This work is the first characterization of a eukaryotic protein from the WS/DGAT family. The biosynthesis of wax esters has been investigated in maturing seeds of Sinapis alba. Exogenous long-chain alcohols are incorporated exclusively into alkyl moieties of wax esters. Oxidation of the long-chain alcohols is not detected. Exogenous fatty acids are incorporated into acyl moieties of wax esters to a low extent. A reduction of fatty acids to alcohols is not observed. Synthesis of wax esters is localized exclusively in the testa; both outer and inner integument are equally active in wax ester biosynthesis. The biosynthesis of wax esters is specific with regard to both chain length and degree of unsaturation of long-chain alcohols. Exogenous and endogenous sterols are not esterified.  相似文献

Utilizing freeze-fracturing and conventional electron microscopy methods, we have studied the details of morphogenesis and construction of the wax layer envelope from Oregon R and mutants of Drosophila melanogaster egg' s during oogenesis. The wax layer is synthesized and secreted by the follicular cells in the 10b of lipid vesicles during static 10b. During secretion (stages 10b, 11 and 12) the lipid vcsicles are accumulated on the vitelline membrane surface and become flat. At the late stage of choriogenisis (stages 13, 14) the lipid vesicles are compressed tightly between the vitelline membrane and the other already constructed eggshell layers, so the wax layer becomes very thin and is hardly seen in crossfractured views.  相似文献

The sub-cellular distribution of chitin synthetase was studied in homogenates of Saccharomyces cerevisiae protoplasts. Use of a mild disruption method minimized rupture of vacuoles and ensuing contamination of subcellular fractions by vacoular proteinases. After fractionation of whole or partially purified homogenates through an isopycnic sucrose gradient chitin synthetase activity was found to be distributed between two distinct particulate fractions with different buoyant density and particle diameter. When whole homogenates were used, about 52% of the chitin synthetase loaded was localized in a microvesicular population identified as chitosomes (diameter 40–110 nm; bouyant density (d) = 1.146 g/cm3). Another vesicular population containing 26% of the activity was identified as plasma membrane vesicles because of its large mean diameter (260 nm), its high buoyant density (d = 1.203 g/cm3) and by the presence of the vanadate-sensitive ATPase activity. Moreover, after surface labeling of protoplasts with 3H-concanavalin A, the label cosedimented with the presumed plasma membrane vesicles. There was a negligible cross-contamination of the chitosome fraction by yeast plasma membrane markers. In both the plasma membrane and the chitosome fractions, the chitin synthetase was stable and essentially zymogenic. Activation of the chitosome fraction produces microfibrils 100–250 nm in length. Our results support the idea that chitosomes do not originate by plasma membrane vesiculation but are defined sub-cellular organelles containing most of the chitin synthetase in protoplasts of Saccharomyces cerevisiae.  相似文献

The physiology of lipid production in Streptomyces avermitilis MA-4680 with regard to the fatty acid composition of the accumulated lipids and their cellular distribution was analyzed. Cells were able to accumulate about ten to 30 lipid granules with diameters between 100 and 500 nm filling about 70–80% of the cell cytoplasm. Gas chromatography/mass spectrometry analyses of total cellular lipids and from isolated triacylglycerols (TAG) confirmed a similar fatty acid composition with a large portion of iso- and anteiso-methyl-branched fatty acids. De novo biosynthesis of wax esters (WE) appeared only during cocultivation on glucose and hexadecanol as carbon source. Homology alignments with the wax ester synthase/acyl-CoA:diacylglycerol acyltransferase (WS/DGAT; AtfA) from Acinetobacter baylyi strain ADP1 yielded one open reading frame in the genome databases of S. avermitilis MA-4680 referred to as SAV7256 with 25.3% homology. The highly conserved HHAxxDG active site motif found in AtfA, which is present in SAV7256, as well as the similar hydrophobicity profiles of AtfA and SAV7256 indicate a similar structure and function of both proteins. High acyl-CoA:diacylglycerol acyltransferase activity (DGAT; 143 pmol (mg min)−1) but low wax ester synthase activity (WS; 1.3 pmol (mg min)−1) were detected in crude extracts of S. avermitilis, which were consistent with the high TAG and negligible WE content of the cells. This indicates that TAG accumulation in S. avermitilis MA-4680 is mediated by the classical acyl-CoA-dependent DGAT pathway. Heterologous expression experiments in recombinant Escherichia coli BL21(DE3) demonstrated both WS and DGAT enzyme activity of SAV7256. The phosphotungstic acid-chromic acid (PTA-CrO3) stain, putatively specific for the plasma membrane of plants, has been used in an attempt to monitor the distribution of this membrane in a 20,000 x g particulate fraction from Cucurbita hypocotyl hooks. On discontinuous sucrose gradients, the relative distributions of the phytochrome and PTA-CrO3-positive vesicles present in this fraction appear to be correlated. When intact tissue is stained, however, other components, in addition to the plasma membrane, react positively to the stain. These components include prolamellar-body membranes, lipid droplets, and ribosomes. This lack of specificity calls into question the reliability of the technique for the unequivocal identification and accurate quantitation of plasma-membrane fragments in isolated particulate fractions. Both photogeneration and quenching of singlet oxygen by monomeric and aggregated (dimeric and oligomeric) molecules of bacteriochlorophyll (BChl) d have been studied in solution and in chlorosomes isolated from the green photosynthetic bacterium Chlorobium vibrioforme f. thiosulfatophilum. The yield of singlet-oxygen photogeneration by pigment dimers was about 6 times less than for monomers. Singlet oxygen formation was not observed in oligomer-containing solutions or in chlorosomes. To estimate the efficiency of singlet oxygen quenching an effective rate constant for 1O2 quenching by BChl molecules (kq M) was determined using the Stern-Volmer equation and the total concentration of BChl d in the samples. In solutions containing only monomeric BChl, the kq M values coincide with the real values for 1O2 quenching rate constants by BChl molecules. Aggregation weakly influenced the kq M values in pigment solutions. In chlorosomes (which contain both BChl and carotenoids) the kq M value was less than in solutions of BChl alone and much less than in acetone extracts from chlorosomes. Thus 1O2 quenching by BChl and carotenoids is much less efficient in chlorosomes than in solution and is likely caused primarily by BChl molecules which are close to the surface of the large chlorosome particles. The data allow a general conclusion that monomeric and dimeric chlorophyll molecules are the most likely sources of 1O2 formation in photosynthetic systems and excitation energy trapping by the long wavelength aggregates as well as 1O2 physical quenching by monomeric and aggregated chlorophyll can be considered as parts of the protective system against singlet oxygen formation.Abbreviations BChl bacteriochlorophyll - MBpd methyl bacteriopheophorbide - Chl chlorophyll - TPP meso-tetraphenylporphyrin - TPPS meso-tetra (p-sulfophenyl) porphyrin  相似文献

Adult whiteflies are characterized by the presence of copious amounts of wax particles covering all surfaces of the body except the eyes. The lipid composition was determined for wax particles removed from the surfaces of the sweetpotato whitefly, Bemisia tabaci (Gennadius), and the greenhouse whitefly, Trialeurodes vaporariorum (Westwood). The lipid components in the wax particles of both species were mostly mixtures of long-chain aldehydes and long-chain primary alcohols. The major wax particle components for B. tabaci were C34 aldehyde and C34 alcohol and small amounts of C32 aldhyde and alcohol. For the wax particles from T. vaporariorum, C32 aldehyde and C32 alcohol were the major components with lesser amounts of the C30 components. These findings were compared to the surface lipids of fully-waxed B. tabaci and T. vaporariorum adults that contained, in addition to the major amounts of long-chain aldehydes and alcohols, quantities of long-chain wax esters. Wax esters were not present in lipid extracts from the surface of B. tabaci whiteflies at the time of adult emergence (prior to deposition of wax particles). Thus, the appearance of wax esters on the cuticular surfaces occurred during the period of deposition of wax particles. The quantities of wax esters in the surface lipid extracts of wing tissues separated from the bodies of adult whiteflies indicated that the wing surfaces were a major site of wax ester deposition.  相似文献

Membranes from roots of Lepidium sativum L. were investigated in situ and after fractionation by applying morphological and biochemical methods. After freeze-fracture combined with filipin labelling the tonoplast and the plasma membrane could be easily characterized by the frequency of intramembranous particles and the arrangement of filipin-induced lesions. On tonoplast vesicles, the filipin-induced lesions were arranged in clusters of different size whereas they were evenly distributed on plasma membrane vesicles. Enrichment of tonoplast and plasma membrane in different fractions was documented by filipin labelling, phosphotungstic acid staining and by the profiles of marker enzyme activities and ATP-dependent H+-transport. Additionally, the presence of rightside-out and inside-out vesicles of both tonoplast and plasma membrane could be demonstrated. Using linear sucrose gradients, particulates derived from pea (Pisum sativum L. cv. Alaska) epicotyls have been fractionated and examined for marker enzyme activity. The coincidence of three reputed plasma-membrane markers [cellulase (EC 3.2.1.4), K+-stimulated Mg2+-ATPase, and glucan synthetase] at the same position on sucrose density gradients, in combination with electron microscopic evidence reported by G. Shore and G. Maclachlan (J. Cell Biol. 64, 557–571; 1975), indicates that plasma membrane of pea epicotyl has a buoyant density of about 1.13 g/cm3. This density disagrees with those usually reported for plant plasma membranes and also with recent reports for Pisum. It is, however, shown to be distinct from the equilibrium densities of enzymic markers for particulate components derived from Pisum endoplasmic reticulum (1.10–1.11 g/cm3), Golgi (1.12 g/cm3) and mitochondria (1.18 g/cm3). Furthermore, other recent literature indicates that the 1.13 g/cm3 buoyant density may be characteristic of the plasma membrane of many members of the Leguminosae. Our data indicate that the conditions of differential centrifugation (time, centrifugal force), coupled with the amount of protein utilized, affect the resolution and interpretation of profiles of marker enzymes on sucrose gradients (e.g. glucan synthetase and K+-stimulated Mg2+-ATPase were sometimes found to be associated not only with particles of 1.13 g/cm3 density, but with particles of higher densities as well). Particulate cellulase was found to be associated only with particles with equilibrium densities of about 1.13 g/cm3. Cellulase thus proved to be the most useful marker for establishing a differential centrifugation regime which would permit examination of the 1.13 g/cm3 particulate components with minimal contamination by particles of higher densities.  相似文献

Continuous cultures of Chloroflexus aurantiacus were cultivated in a chemostat in the light with varying bacteriochlorophyll (BChl) a/c ratios by changing the growth rate. Under these culture conditions all cells were homogeneously and reproducibly equipped with chlorosomes. In order to determine the number and size of chlorosomes in relation to different BChl contents morphometric measurements were performed on electron micrographs. The linear increase of BChl a contents coincided with an increasing number of chlorosomes per membrane area and per bacterium rather than with an enlargement of the average size of chlorosomes. The numbers of chlorosomes and therefore the percentage of chlorosome-covered cytoplasmic membrane increased linearly with increasing BChl a contents. The average size of the baseplates was largely constant in all cultures (mean 3,222±836 nm2). However, within individual cells the size of baseplates varied by a factor of 3.0, especially by the variation of the length. The exponential increase in BChl c contents coincided with an increasing number of chlorosomes (up to a factor of 2.3) and an enlargement of the average chlorosome volume (up to a factor of 1.9). The number of BChl a molecules per chlorosome was about 1,484±165, thus the number of reaction centers per chlorosome was 58±12. The data suggest, firstly, that BChl a is confined to areas (cytoplasmic membrane plus baseplate) as represented by the chlorosome attachment sites; secondly, that the degree of packing of BChl c molecules within chlorosomes increases with increasing BChl c contents.  相似文献

The plasma membrane (PM) vesicles from Populus euphratica (P. euphratica) callus were isolated to investigate the properties of the PM H+-ATPase. An enrichment of sealed and oriented right-side-out PM vesicles was demonstrated by measurement of the purity and orientation of membrane vesicles in the upper phase fraction. Analysis of pH optimum, temperature effects and kinetic properties showed that the properties of the PM H+-ATPase from woody plant P. euphratica callus were consistent with those from herbaceous species. Application of various thiol reagents to the reaction revealed that reduced thiol groups were essential to maintain the PM H+-ATPase activity. In addition, there was increased H+-ATPase activity in the PM vesicles when callus was exposed to NaCl. Western blotting analysis demonstrated an enhancement of H+-ATPase content in NaCl-treated P. euphratica callus compared with the control.  相似文献

Candida rugosa lipase (CRL) was applied in a non-solvent esterification reaction to yield twelve wax esters. All products were obtained in nearly 100% yield for 10 h at 50°C when immobilized PEG2000-activated C. rugosa lipase was added to the reaction mixture. The surfactant had also a beneficial effect on the stability of the biocatalytic preparation with 83% of its activity conserved after the seventh run of repeated batch reactions.  相似文献

Whole cells, chlorosome-membrane complexes and isolated chlorosomes of the green mesophilic filamentous bacterium Oscillochloris trichoides, representing a new family of the green bacteria Oscillochloridaceae, were studied by optical spectroscopy and electron microscopy. It was shown that the main light-harvesting pigment in the chlorosome is BChl c. The presence of BChl a in chlorosomes was visualized only by pigment extraction and fluorescence spectroscopy at 77 K. The molar ratio BChl c: BChl a in chlorosomes was found to vary from 70:1 to 110:1 depending on light intensity used for cell growth. Micrographs of negatively and positively stained chlorosomes as well as of ultrathin sections of the cells were obtained and used for morphometric measurements of chlorosomes. Our results indicated that Osc. trichoides chlorosomes resemble, in part, those from Chlorobiaceae species, namely, in some spectral features of their absorption, fluorescence, CD spectra, pigment content as well as the morphometric characteristics. Additionally, it was shown that similar to Chlorobiaceae species, the light-harvesting chlorosome antenna of Osc. trichoides exhibited a highly redox-dependent BChl c fluorescence. At the same time, the membrane B805–860 BChl a antenna of Osc. trichoides is close to the membrane B808–866 BChl a antenna of Chloroflexaceae species. Summary We have examined the cortex of the teleost (Brachydanio rerio) egg before and during exocytosis of cortical granules by scanning, transmission, and freeze-fracture electron microscopy. In the unactivated egg, the P-face of the plasma membrane exhibits a random distribution of intramembranous particles, showing a density of 959/m2 and an average diameter of 8 nm. Particles over P- and E-faces of the membranes of cortical granules are substantially larger and display a significantly lower density. An anastomosing cortical endoplasmic reticulum forms close associations with both the plasma membrane of the egg and the membranes of cortical granules. Exocytosis begins with cortical granules pushing up beneath the plasma membrane to form domeshaped swellings, coupled with an apparent clearing of particles from the site of contact between the apposed membranes. A depression in the particle-free plasma membrane appears to mark sites of fusion and pore formation between cortical granules and plasma membranes. Profiles of exocytotic vesicles undergo a predictable sequence of morphological change, but maintain their identity in the egg surface during this transformation. Coated vesicles form at sites of cortical granule breakdown. Differences in particle density between cortical granules and egg plasma membranes persist during transformation of the exocytotic profiles. This suggests that constituents of the 2 membrane domains remain segregated and do not intermix rapidly, lending support to the view that the process of membrane retrieval is selective (i.e., cortical granule membrane is removed).  相似文献

Ilex leaves can utilize sucrose and mevalonate for the synthesis of triterpene esters. Mevalonate is also used for the synthesis of free triterpenoid alcohols, but sucrose is not. The selectivity of precursor utilization indicates separate sites for triterpenol and triterpenol-ester synthesis. The sites of synthesis are not found at the main locations of accumulation of triterpenols (the epicuticular wax) or triterpene esters (the cytoplasmatic lipid globules). Summary Plasma membrane vesicles, which are mostly right side-out, were isolated from corn leaves by aqueous two-phase partitioning method. Characteristics of Ca2+ transport were investigated after preparing inside-out vesicles by Triton X-100 treatment.45Ca2+ transport was assayed by membrane filtration technique. Results showed that Ca2+ transport into the plasma membrane vesicles was Mg-ATP dependent. The active Ca2+ transport system had a high affinity for Ca2+(K m (Ca2+)=0.4 m) and ATP(K m (ATP)=3.9 m), and showed pH optimum at 7.5. ATP-dependent Ca2+ uptake in the plasma membrane vesicles was stimulated in the presence of Cl or NO 3 . Quenching of quinacrine fluorescence showed that these anions also induced H+ transport into the vesicles. The Ca2+ uptake stimulated by Cl was dependent on the activity of H+ transport into the vesicles. However, carbonylcyanidem-chlorophenylhydrazone (CCCP) and VO 4 3– which is known to inhibit the H+ pump associated with the plasma membrane, canceled almost all of the Cl-stimulated Ca2+ uptake. Furthermore, artificially imposed pH gradient (acid inside) caused Ca2+ uptake into the vesicles. These results suggest that the Cl-stimulated Ca2+ uptake is caused by the efflux of H+ from the vesicles by the operation of Ca2+/H+ antiport system in the plasma membrane. In Cl-free medium, H+ transport into the vesicles scarcely occurred and the addition of CCCP caused only a slight inhibition of the active Ca2+ uptake into the vesicles. These results suggest that two Ca2+ transport systems are operating in the plasma membrane from corn leaves, i.e., one is an ATP-dependent active Ca2+ transport system (Ca2+ pump) and the other is a Ca2+/H+ antiport system. Little difference in characteristics of Ca2+ transport was observed between the plasma membranes isolated from etiolated and green corn leaves.  相似文献
