Summary Fed-batch culture ofE.coli B was carried out to obtain high concentration of biomass. After requirement of oxygen was met by sparging the pure oxygen, physiological constraints were delineated. High partial pressure of CO2 caused the decrease of the maximum specific growth rate, whereas fermentative byproducts caused the decrease of biomass yield as well as the maximum specific growth rate.  相似文献

Based on the well-known fact thatKluyveromyces fragilis strains show sub-optimal performance when grown in concentrated whey permeate, previously optimized medium was investigated for possible limitations appearing at high concentrations. Shaken flask cultures showed that no additional vitamin or mineral sources were required when the optimized amount of yeast extract was added to the concentrated permeate. Several aspects of the ethanol inhibition of the growth ofK. fragilis NRRL 665 were investigated in continuous culture. The maximum ethanol concentration tolerated by this yeast, i.e. 45 g/l, was much lower than commonly reported for other strains. Ethanol and biomass production were also influenced by the increased ethanol concentration of the medium. At 31 g/l of alcohol product yield was reduced to 0.23 g/g whereas biomass yield was 0.05 g/g. Some evidence suggested that residence time and residual lactose concentration played a significant role in modulating the toxic effect of ethanol.  相似文献

Summary The production of -carotene by the biomass ofRhodotorula strain var.glutinis, during the stationary phase of growth and in non-proliferating conditions was assayed. Summary Three pentose fermenting yeast strains ofCandida shehatae and three ofPichia stipitis were examined for their ability to produce ethanol from cellobiose and from sugars liberated by hydrolysis of lignocellulosic biomass. All of thePichia strains tested produced some ethanol;P. stipitis CBS 5776 gave the highest yield: 10.3 g/L on complete fermentation of 25 g/L cellobiose within 48 hours. Summary Three strains ofPichia stipitis and three ofCandida shehatae were compared withPachysolen tannophilus in their abilities to ferment xylose at concentrations as high as 200 g/L when subjected to both aerobic and microaerophilic conditions. Evaluations based on accumulated ethanol concentrations, ethanol productivities, xylose consumption, and ethanol and xylitol yields were determined from batch culture time courses. Of the strains considered,P.stipitis NRRL Y-7124 seemed most promising since it was able to utilize all but 7 g/L of 150 g/L xylose supplied aerobically to produce 52 g/L ethanol at a yield of 0.39 g per gram xylose (76% of theoretical yield) and at a rate comparable to the fastest shown byC.shehatae NRRL Y-12878. For all strains tested, fermentation results from aerobic cultures were more favorable than those from microaerophilic cultures.The mention of firm names or trade products does not imply that they are endorsed or recommended by the U.S. Department of Agriculture over other firms or similar products not mentioned.  相似文献

Summary The possibility of usingZymomonas mobilis as the microorganism, in solid-state fermentation of sugar-beet particles was investigated. The major factors affecting the process were investigated and related to ethanol yield and productivity. Ethanol yield of 0.48 g/g sugar, volumetric productivity of 12 g/L h, and final ethanol concentration of 130 g/L show the good performance ofZ.mobilis in a solid-state fermentation.  相似文献

Summary A locally isolated oleaginous strain ofRhodotorula glutinis strain IIP-30 produced a growth associated extracellular emulsifying agent while utilizing glucose during fed batch fermentation under nitrogen limitation at 30°C and pH 4. 0. Rhodotorula glutinis, an oil producing strain, can utilize monosodium glutamate (MSG) wastewater as a raw material for lipid production. The effects of ammonium-N in the MSG wastewater (ammonium 15,000–25,000?mg/L, COD 30,000–50,000?mg/L) on cell growth, lipid accumulation and malic enzyme activity of R. glutinis have been studied. Four initial ammonium sulfate concentrations in the medium were set, which were 20, 60, 100, and 140?g/L. With an increase in the ammonium sulfate concentration, the uptake of ammonia nitrogen and lipid accumulation increased while the biomass decreased at 72?h. The maximum value of ammonia nitrogen consumption reached 5.77?g/L for an initial ammonium sulfate concentration of 140?g/L at 72?h. In addition, 60?g/L ammonium sulfate concentration may be an appropriate concentration for R. glutinis cultivation. The activity of the malic enzyme was measured and the results showed that there was a linear relationship between the intracellular lipid content and the total malic enzyme activity.  相似文献

Summary A strain ofSaccharomyces cerevisiae, Y18, has been isolated and characterized for the production of ethanol and biomass from the sap of Palmyra palm. The ethanol and biomass produced by Y18 in batch cultures without aeration were 70 g/L and 6 g/L respectively, from saps containing 150 g/L of sucrose. Under aerobic conditions, the biomass produced was 0.5 g/g sucrose, with crude and true protein contentsentrations of 57 and 52 % w/w respectively.  相似文献

Summary Batch and continuous culture studies have been carried out on a strain ofZ.mobilis (ZM6306) which can convert lactose directly to ethanol. Previous strain development has established that thelac operon encoded on the transposon Tn951 can be expressed inZ.mobilis. Using a medium containing 80 g/l glucose and 40 g/l lactose, it was found that strain ZM6306 could convert about 13 g/l lactose to 4 g/l ethanol and 6 g/l galactose in continuous culture. Further lactose conversion is likely with increased cell concentration using a cell recycle system.  相似文献

Summary A culture technique, based on the growth of a microorganism on inert porous particles (e. g. pozzolano) impregnated and continuously fed with substrate is applied to the growth and spore production ofPenicillium roqueforti. Summary Some 19 strains ofAspergillus niger,A. oryzae, andPaccilomyces spp. are tested for their ability to grow on the supernatant remaining after the expressed juice from sugar beet tops and meadow grass has been heat-treated to precipitate crude leaf protein, and supplemented as required by glucose or ammonium sulphate. With effective strains ofA. niger,A. oryzae,P. elegans orP. variotii and an optimized carbohydrate/nitrogen ratio, over 70% of the organic content of the supernatant is rapidly converted into mycelial biomass of high protein content.  相似文献

Protoplasts of several wildLinum species were isolated enzymatically from roots, hypocotyls and cotyledons of seedlings, and also from theirin vitro grown shoots and cell suspension cultures. When cultured all these protoplasts divided to produce callus but only good plant regeneration capability was evident in the case ofLinum lewissii and to a much lesser extent forL. strictum. Only rhizogenesis was observed withL. alpinum, L. narbonense, L. grandiflorum andL. altaicum. Summary A computer-assisted on-line glucose analyzer was developed for feed-back control of cell growth. Using this system the glucose consumption rate for Escherichia coli was determined to be linear during batch culture at 0.37 g/hr. On-line feed-back control of glucose concentration at 1.5±0.5 g/L was used with fed-batch cultures to produce 31.2 g dry weight of E. coli cells/L in 12 h.  相似文献

Microbial oil produced by the oleaginous yeast Rhodosporidium toruloides ATCC 204091 (formerly referred to as Rhodotorula glutinis) has a similar fatty acid composition to the vegetable oils and represents a potential alternative for biodiesel production. Finding strategies to improve the oil production by this yeast is desirable, as it is one of this nutrient’s limitations during the accumulation phase, as well as one of the main factors influencing the process. Therefore, the effect of single or combined nutrient limitation on lipid accumulation by R. toruloides was investigated. Biomass production and lipid accumulation by R. toruloides was improved using experimental designs in a two-step batch culture on a chemically-defined culture medium with high initial glucose concentration. For the first culture step, a Box–Behnken design was applied to optimize the main medium components’ concentrations, while maintaining a high biomass production. A biomass concentration of 44.3 g/L was reached with a medium composed of (g/L): glucose, 100; KH2PO4, 4.6; NaNO3, 13.4; MgSO4 .7H2O, 0.2; and CaCl2 .2H2O, 0.11. For the second culture step, the biomass was transferred to lipid accumulation media. A 23 factorial experimental design was conducted to investigate the effect of N, P and S limitations (individually or jointly) on lipid production from glucose (100 g/L). Lipid accumulation on dry cell mass was 77.04, 65.42, 70.13 and 69.84% for N, P, S and simultaneous nutrients’ limitations, respectively.  相似文献

Summary Callus culture ofDioscorea deltoidea produced diosgenin and sterols during stationary phase. Ammonium nitrate (420 mg Nitrogen/l) as sole nitrogen source supported better growth than a combination of ammonium nitrate and potassium nitrate (totally equivalent to 840 mg Nitrogen/l). The production of diosgenin increased under low phosphate concentration (100 mg/l) whereas high phosphate concentration (240 mg/l) promoted growth. Micronutrients, when used at 1 1/2 strength, enhanced growth and diosgenin production. Depletion of nitrogen increased the diosgenin synthesis by a factor of 2. Adoption of a two stage culture method enhanced the diosgenin production in cultured cells by eight-fold.  相似文献

Summary Lithospermum erythrorhizon was immobilized in a dual hollow fiber bioreactor (DHFBR) to maintain high cell density and to operate continuously. The cells grew well and its dry biomass density was 325 g/L of the void volume for the cell growth. Volumetric and specific productivities of phenolics were 221 mg/L.day and 0.68 mg/g.dry wt.day, respectively, which are 58 and 2 times of those of shake flask cultures.  相似文献

Summary Cells ofPichia farinosa were immobilised in calcium alginate and K-Carrageenan and their ability to produce glycerol from glucose under aerobic conditions with acidic as well as alkaline pH was investigated. An average glycerol production rate of 0.07 g/l.h was obtained with immobilised cells (IMC) in shake flasks. Continuous glycerol production in a fluidised bed reactor (FBR) under steady state operation gave a glycerol concentration of 13.5 g/l in the product stream.  相似文献
