Since differences in metal accumulation may exist between bivalve species, the aim of this study was to assess the impact of cadmium (Cd) on Ruditapes decussatus and Ruditapes philippinarum. For this, the Cd accumulation, mortality rates and biochemical responses were analysed in the two species after 5days of exposure, under laboratory-controlled conditions. The concentration of Cd that caused 50% of mortality on clams was two-times higher in R. decussatus than in R. philippinarum. For both species, higher percentage (84.5-98.2%) of the Cd was in the insoluble fraction, but the Cd concentration in solution was 3 to 8 times higher in R. decussatus. Nevertheless, R. philippinarum presented higher oxidative stress and higher CAT activity. The paradox observed between the two clams can be explained by the higher capacity of R. decussatus to increase the expression of MTs when exposed to Cd.  相似文献

Excised maize (Zea mays L.) root tips were used to study the early metabolic effects of glucose (Glc) starvation. Root tips were prelabeled with [1-13C]Glc so that carbohydrates and metabolic intermediates were close to steady-state labeling, but lipids and proteins were scarcely labeled. They were then incubated in a sugar-deprived medium for carbon starvation. Changes in the level of soluble sugars, the respiratory quotient, and the 13C enrichment of intermediates, as measured by 13C and 1H nuclear magnetic resonance, were studied to detect changes in carbon fluxes through glycolysis and the tricarboxylic acid cycle. Labeling of glutamate carbons revealed two major changes in carbon input into the tricarboxylic acid cycle: (a) the phosphoenolpyruvate carboxylase flux stopped early after the start of Glc starvation, and (b) the contribution of glycolysis as the source of acetyl-coenzyme A for respiration decreased progressively, indicating an increasing contribution of the catabolism of protein amino acids, fatty acids, or both. The enrichment of glutamate carbons gave no evidence for proteolysis in the early steps of starvation, indicating that the catabolism of proteins was delayed compared with that of fatty acids. Labeling of carbohydrates showed that sucrose turnover continues during sugar starvation, but gave no indication for any significant flux through gluconeogenesis.  相似文献

The response of Vibrio cholerae to low nutrient levels was determined by measuring the concentrations of lipids, carbohydrates, DNA, RNA, and proteins over a 30-day starvation period. Ultrastructural integrity was observed by transmission electron microscopy. Total lipids and carbohydrates declined rapidly within the first 7 days, while DNA and protein exhibited a more constant decline over the 30 days of starvation. In contrast, RNA showed little decrease upon starvation. Although neutral lipids were lost, the percentage of neutral lipids did not decline as rapidly as the phospholipids. Detectable levels of poly-beta-hydroxybutyrate disappeared completely by 7 days. Carbohydrate profiles revealed the relative loss of the five-carbon sugar ribose and N-acetylglucosamine and a relative increase in the total six-carbon sugars, especially glucose. Morphologically, ribosomes appeared to exhibit no structural change, while inclusion bodies and mesosomelike structures disappeared completely, and cell wall and membrane integrity was lost. The data suggest that V. cholerae differs somewhat from other marine vibrios in its response to low nutrients but shares some characteristics in common with them. The data also suggest that certain lipids and carbohydrates may provide the endogenous energy sources needed for dormancy preparation and cell maintenance under nutrient starvation.  相似文献

This study evaluates the effect of temperature, coupled with ingestion rate, on the dynamics of biochemical components and lipid classes in R. philippinarum. The data are discussed with regard to sexual development and energy balance. Experimental protocol developed in the present study used two groups of the clam R. philippinarum: L (temperatures of 14 degrees C and 18 degrees C) and H (temperatures of 18 degrees C and 22 degrees C). The intra-group ingestion level was similar, although the ingestion level of the clams in the group H was 2.4 times higher than group L. We observed that R. philippinarum conditioned at 18 degrees C (18L) shows higher protein content, furthermore an important loss of organic weight was observed after 48 days. In such a situation, the clams use their own reserves (carbohydrates and glycogen) for sexual development while in situations without food stress (positive energy balance) and low temperature (14 degrees C) an accumulation of reserves is produced. Strikingly dissimilar behaviour in biochemical composition was observed for the 18H and 22H treatments, both with a positive energy balance. Despite similar protein content, the highest levels of carbohydrates were observed at the lower temperature (18 degrees C). Glycogen was also higher for the 18 degrees C treatment, although the differences were significant only in the males. Although the total lipids in R. philippinarum showed no significant differences in any treatment, they became apparent and related to sex when considering the individual lipid classes. There was no variation in lipid classes in the males between the 14L and 22H treatments despite the large disparity in the degree of sexual development. However, in the females significant differences in lipid classes (phospholipids, triglycerides) were observed. The results of this study show that a positive energy balance permits R. philippinarum gonadal development and accumulation of reserves both in low and high temperature conditions. In low temperature situations, gonadal development is slower and the energy reserves are accumulated in the form of carbohydrates. When the clams are conditioned at high temperatures, gonadal development is fast and complete, carbohydrates are consumed and lipids are accumulated.  相似文献

The effect of cadmium (Cd) on the reproduction of Ruditapes decussatus was monitored over a period of 12 months, from June 2001 to June 2002. Two sites "Bordj d'Ungha" and "El Hofra" differing by their degree of cadmium contamination were chosen in the Gulf of Gabès area (Tunisia). Annual mean concentrations of Cd in the whole soft tissues of clams from the site El Hofra were more than 4 times higher than those from the site Bordj d'Ungha (reference site). The gametogenic cycle of the clam R. decussatus was also investigated by histological examination and monthly observations of gonadal sections in a population of clams from both sites. The results show that gametogenesis occurred from March to December in clams from both sites. Nevertheless, spawning and emission of gametes were synchronized in both sexes from only the clams of the reference site. Although this species is considered as gonochoristic, 6.6% of hermaphroditic cases were observed in clams from both sites in which gametes of both males and females were in ripe stage. Moreover, the period of ripening of sexual products led to an increase of condition index and to a decrease of Cd concentrations in the whole soft tissues of clams from both sites, hence reflecting the phenomenon of "biological dilution".  相似文献

The hypothesis was tested that animals near their extreme Southern limits, due to high temperatures, have a high respiration rate, whereby they reach an extreme low weight-index and ultimately disappear. At estuarine stations the respiration rate of Macoma balthica (L.) (Baltic clam) did not show interseasonal changes, indicating seasonal acclimation, but within the season the respiration increased with increasing temperature, indicating the absence of short-term acclimation. In clams translocated from the Netherlands towards the Bidasoa estuary, 200 km South of their Southern distribution limit, their respiration rate was higher and weight-index lower than in specimens living in Dutch estuaries. Irrespective of an effect of the temperature, clams exposed in experiments to water from Bidasoa showed a higher respiration than clams exposed to water from the other stations. Moreover, at non-estuarine stations with a low food content, the clams showed reversed acclimation, i.e., the respiration rates in winter were much lower than summer rates, most probably a strategy to conserve energy by means of a depressed metabolism. A weight index of 5 mg DW/cm(3) and glycogen content of 2% DW are suggested as the minimal values below which the metabolic energy balance of Baltic clams becomes more negative and the clam population disappears. It was concluded that factors other than temperature influenced the respiration and weight-index of clams, and hence their presence or absence, e.g., food concentration, innate seasonal cycles, and possible pollutants in the water.  相似文献

This work compared the effect of challenge with Vibrio tapetis, the etiologic agent of brown ring disease (BRD) in clams, and other bacterial strains on defence-related factors in four bivalve species: Ruditapes philippinarum (highly susceptible to BRD), R. decussatus (slightly susceptible to BRD), Mercenaria mercenaria and Crassostrea virginica (both non-susceptible to BRD). Results show that bacterial challenge modulated defence-related factors, namely total and differential haemocyte counts, percentage of viable haemocytes, and lysozyme activity, both in haemolymph and extrapallial fluid. Injection with bacteria induced a response that was dependent upon the bacterial and bivalve species investigated, and upon the site of inoculation: external (pallial cavity), pseudo-internal (extrapallial space), or internal compartment (adductor muscle). The most conspicuous changes were systematically measured in R. philippinarum injected with V. tapetis, indicating a bacterial pathogenicity particular to the host in which it causes a specific disease syndrome. Alterations of defence-related factors were maximal in haemolymph of clams injected with V. tapetis in the muscle, and in the extrapallial fluid when the bacteria were injected into the pallial or the extrapallial cavity. Resistance to the development of the BRD symptom was not related to the extent of the haemocyte reaction measured following in vivo challenge.  相似文献

In this paper, we review the current knowledge about the usage of carbohydrates, lipids and proteins as energy source by marine crustaceans during starvation. Crustaceans are a large and diverse group including some economically important species. The efforts to culture them for human consumption has prompted the interest to understand the preferences of energy sources to be applied for feed formulation and cost reduction. Important differences have been found among species and appear to be related not only to the biochemistry and physiology of nutrition, but also to the living environment of the crustaceans. Furthermore, crustaceans undergo morphological, physiological and behavioral changes due to their natural growing process that affect their feeding habits, an aspect that should be carefully considered. We discuss the current information on marine crustaceans about energy usage and describe areas of future research, where starvation studies render important insights.  相似文献

The digestive enzyme activities were determined in Adriatic sturgeon and rainbow trout during starvation and refeeding period. Overall, the digestive enzyme activities are affected in the same sense in both species. The protease and lipase activities were decreased later than amylase activity. Even after 1 month of starvation, both species would be prepared to digest protein and lipids in an effective way. After 72 days of starvation, the digestive machinery of the sturgeon and of the trout shows an altered capacity to digest macronutrients. The capacity to digest proteins and lipids, after 60 days of refeeding, begins to become re-established in sturgeon and trout. In contrast, in this period, the capacity to digest carbohydrates remains depressed in both species.  相似文献

This review is a survey of progress in the field of helminth intermediary metabolism since ICOPA V. Data on the catabolism and synthesis of carbohydrates, lipids, amino acids, proteins, nucleotides and nucleic acids are presented and discussed in relation to previous work. The large number of references cited reflects a continued interest of parasite biochemists in this field, although many fundamental problems of helminth metabolism remain unsolved. Important areas for future research are identified and the potential application of new biochemical techniques, especially those in molecular biology, are emphasised.  相似文献

The gulf of Gabès is well known for its multiple contaminations which are closely associated with the past and present industrial and anthropogenic activity. In order to evaluate in situ the cadmium bioaccumulation in the clam Ruditapes decussatus sampled in this area, we measured cadmium concentrations in the whole soft tissues and in the digestive gland during one year. Energy reserves (proteins, glycogen, lipids) were monitored during gametogenesis in the remainder (mantle, adductor muscle, foot, siphons) of clams. Sex steroids (progesterone, testosterone, estradiol-17beta) were also measured by radio-immunoassay (RIA) in the gonads. The results showed that cadmium varied according to the reproductive cycle. High values were recorded at the resting stage in both sexes, during the gamete development and maturation in males and at the ripening and partially spawn stages in females. No significant changes in protein levels were observed in both females and males. However, high levels of glycogen and lipid reserves were recorded during the gametogenesis activity (from the development to the partially spawn stages). For steroid analyses, the results showed no important changes in testosterone levels all along the reproductive cycle. Fluctuations of sex steroids (especially progesterone and estradiol-17beta) could suggest their possible role as endogenous modulators of gametogenesis in R. decussatus. Correlation matrix and multiple regression models were also proposed to describe negative and significant relationship between cadmium and (glycogen, progesterone, testosterone and estradiol-17beta). These correlations may suggest the involvement of cadmium in the dysfunction of energy status and the endocrine disruption which could impede reproduction.  相似文献

The response to oxidative stress involves numerous genes and mutations in these genes often manifest in pleiotropic ways that presumably reflect perturbations in ROS-mediated physiology. The Drosophila melanogaster SOD1-null allele (cSODn108) is proposed to result in oxidative stress by preventing superoxide breakdown. In SOD1-null flies, oxidative stress management is thought to be reliant on the glutathione-dependent antioxidants that utilize NADPH to cycle between reduced and oxidized form. Previous studies suggest that SOD1-null Drosophila rely on lipid catabolism for energy rather than carbohydrate metabolism. We tested these connections by comparing the activity of carbohydrate metabolizing enzymes, lipid and triglyceride concentration, and steady state NADPH:NADP(+) in SOD1-null and control transgenic rescue flies. We find a negative shift in the activity of carbohydrate metabolizing enzymes in SOD1-nulls and the NADP(+)-reducing enzymes were found to have significantly lower activity than the other enzymes assayed. Little evidence for the catabolism of lipids as preferential energy source was found, as the concentration of lipids and triglycerides were not significantly lower in SOD1-nulls compared with controls. Using a starvation assay to impact lipids and triglycerides, we found that lipids were indeed depleted in both genotypes when under starvation stress, suggesting that oxidative damage was not preventing the catabolism of lipids in SOD1-null flies. Remarkably, SOD1-nulls were also found to be relatively resistant to starvation. Age profiles of enzyme activity, triglyceride and lipid concentration indicates that the trends observed are consistent over the average lifespan of the SOD1-nulls. Based on our results, we propose a model of physiological response in which organisms under oxidative stress limit the production of ROS through the down-regulation of carbohydrate metabolism in order to moderate the products exiting the electron transport chain.  相似文献

Brown ring disease, caused by Vibrio tapetis, is an important pathological problem in different species of cultured clams. In order to evaluate the genetic diversity of the pathogen, twenty-seven isolates of V tapetis with different origin were screened by ribotyping (RT), pulsed-field gel electrophoresis (PFGE) and randomly amplified polymorphic DNA PCR (RAPD). Restriction with PvuII, SalI, and SmaI gave 2 RT patterns, differentiating in all cases the strain 0202RD isolated from carpet-shell clams (Ruditapes decussatus) from the other strains tested. The use of NotI generated strain specific PFGE profiles, which could be grouped in two main clusters. Cluster 1 grouped all but one strain and was subdivided into six PFGE subtypes (1a to 1f) which joined at a similarity level of 75.6%. Cluster 2 included again only the isolate 0202RD. RAPD analysis yielded the same results with three different primers, this method being able to differentiate the isolates from R. decussatus from those isolated from other clam species. Of the three techniques evaluated, PFGE was the most discriminating of the three techniques evaluated, followed in discriminating power by RAPD and RT tests. On the basis of the results obtained, we conclude that the RAPD procedure, which is more rapid and easier to perform than the other techniques, shows to be very useful to analyze large amounts of strain collections from an epidemiological monitoring stanpoint. In addition, PFGE is of great utility to evaluate the genetic diversity of strains involved in an outbreak and to study the spreading of a specific clone.  相似文献

The rRNA locus of Perkinsus atlanticus from the clam Ruditapes decussatus cultivated on the Atlantic coast of Spain was cloned and sequenced. Sequences of the internal transcribed spacer (ITS) from the rRNA locus were compared to sequences reported earlier for a P. atlanticus isolate from Portugal and to those from other Perkinsus species. The ITS I sequence of the Spanish P. atlanticus isolate was identical to the Portuguese P. atlanticus sequence and had 76.6% identity to the ITS1 of Perkinsus marinus. The ITS2 sequence had 99.7% identity to the Portuguese P. atlanticus ITS2, 92.5% identity to the P. marinus ITS2, and 99.5% identity to the Perkinsus olseni ITS2. We report for first the time the small subunit (SSU) and nontranscribed spacer (NTS) of P. atlanticus. The P. atlanticus SSU sequence was 99.6% identical to that of an unidentified Perkinsus species from the Australian clam Anadara trapezia and 98.0% identical to that of P. marinus. Further, our results support the proposal that P. atlanticus, P. olseni, and the Perkinsus sp. from A. trapezia constitute a subgroup of Perkinsus species distributed in the Pacific and eastern Atlantic, different from P. marinus that is distributed along the western edge of the Atlantic. Based on the NTS sequence of P. atlanticus from Spain and the differences with P. marinus NTS (62.2% identity), we developed a polymerase chain reaction (PCR)-based diagnostic assay with a lowest limit of detection of 0.01 amol of cloned NTS DNA as assessed on ethidium bromide-stained agarose gels. Specificity of the PCR-based assay was tested with samples from the clams R. decussatus, Ruditapes philippinarum, and Venerupis pullastra collected in P. atlanticus-enzootic areas of Spain. The specificity and sensitivity demonstrated for this NTS-based PCR assay validate its use as a tool for assessment of P. atlanticus in molluscs.  相似文献

Zusammenfassung In frisch gefangenen Flußkrebsen und solchen, die 15 bzw. 41 Tage gehungert hatten, wurde der Bestand an Lipiden, Kohlenhydraten, freien Aminosäuren und Proteinen bestimmt. Die Meßergebnisse sind in Tabelle 1 wiedergegeben. Es wird geschlossen, daß der Energiebedarf in den ersten 15 Hungertagen überwiegend aus den Lipiden, anschließend aus Proteinen gedeckt wird. Die Kohlenhydrate spielen als energieliefernde Substanzen eine untergeordnete Rolle; die beobachtete Abnahme der Gesamtkohlenhydrate betrifft vor allem die Trehalose.

Summary The amounts of lipids, carbohydrates, free amino acids, and proteins were determined in freshly caught crayfishes and in crayfishes that had starved for 15 or 41 days. The results are summarized in Table 1. It is concluded that in the first two weeks of starvation the lipids are the main source of energy production, afterwards the proteins. Carbohydrates are of minor importance in energy metabolism. Of all carbohydrates the amount of trehalose shows the greatest reduction during starvation.

This study reports on the polymorphism of 15 allozyme loci in Ruditapes decussatus clams collected from 11 locations along the Tunisian coasts. We concentrated our sampling effort around the Siculo-Tunisian region to verify if any population structuring exists in this region and to identify the factors that have shaped this structure. Measurements of genetic diversity were quantified both within and between populations, and the geographic variability of gene frequencies was analyzed. Our study shows that the Siculo-Tunisian Strait is an important genetic boundary between eastern and western regions, which agrees with findings for a variety of other species. We suggest that vicariance is a predominant factor shaping the current distribution of genetic diversity of R. decussatus, and the mixing of divergent gene pools from the eastern and western regions still seems to be limited by some physical and/or biological factors.  相似文献

Obesity has a strong genetic component, but few of the genes that predispose to obesity are known. Genetic screens in invertebrates have the potential to identify genes and pathways that regulate the levels of stored fat, many of which are likely to be conserved in humans. To facilitate such screens, we have developed a simple buoyancy-based screening method for identifying mutant Drosophila larvae with increased levels of stored fat. Using this approach, we have identified 66 genes that when mutated increase organismal fat levels. Among these was a sirtuin family member, Sir2. Sirtuins regulate the storage and metabolism of carbohydrates and lipids by deacetylating key regulatory proteins. However, since mammalian sirtuins function in many tissues in different ways, it has been difficult to define their role in energy homeostasis accurately under normal feeding conditions. We show that knockdown of Sir2 in the larval fat body results in increased fat levels. Moreover, using genetic mosaics, we demonstrate that Sir2 restricts fat accumulation in individual cells of the fat body in a cell-autonomous manner. Consistent with this function, changes in the expression of metabolic enzymes in Sir2 mutants point to a shift away from catabolism. Surprisingly, although Sir2 is typically upregulated under conditions of starvation, Sir2 mutant larvae survive better than wild type under conditions of amino-acid starvation as long as sugars are provided. Our findings point to a Sir2-mediated pathway that activates a catabolic response to amino-acid starvation irrespective of the sugar content of the diet.  相似文献

Storage of energy metabolites has been investigated in different sets of laboratory selected desiccation or starvation resistant lines but few studies have examined such changes in wild-caught populations of Drosophila melanogaster. In contrast to parallel selection of desiccation and starvation tolerance under laboratory selection experiments, opposite clines were observed in wild populations of D. melanogaster. If resistance to desiccation and starvation occurs in opposite directions under field conditions, we may expect a trade-off for energy metabolites but such correlated changes are largely unknown. We tested whether there is a trade-off for storage as well as actual utilization of carbohydrates (trehalose and glycogen), lipids and proteins in D. melanogaster populations collected from different altitudes (512-2500 m). For desiccation resistance, darker flies (> 50% body melanization) store more body water content and endure greater loss of water (higher dehydration tolerance) as compared to lighter flies (< 30% body melanization). Based on within population analysis, we found evidence for coadapted phenotypes i.e. darker flies store and actually utilize more carbohydrates to confer greater desiccation resistance. In contrast, higher starvation resistance in lighter flies is associated with storage and actual utilization of greater lipid amount. However, darker and lighter flies did not vary in the rate of utilization of carbohydrates under desiccation stress; and of lipids under starvation stress. Thus, we did not find support for the hypothesis that a lower rate of utilization of energy metabolites may contribute to greater stress resistance. Further, for increased desiccation resistance of darker flies, about two-third of total energy budget is provided by carbohydrates. By contrast, lighter flies derive about 66% of total energy content from lipids which sustain higher starvation tolerance. Our results support evolutionary trade-off for storage as well as utilization of energy metabolites for desiccation versus starvation resistance in D. melanogaster.  相似文献

Most of our understanding about the physiology of fasting and starvation comes from studies of vertebrates; however, for ethical reasons, studies that monitor vertebrates through the lethal endpoint are scant. Insects are convenient models to characterize the comparative strategies used to cope with starvation because they have diverse life histories and have evolved under the omnipresent challenge of food limitation. Moreover, we can study the physiology of starvation through its natural endpoint. In this study we raised populations of five species of insects (adult grasshoppers, crickets, cockroaches, and larval beetles and moths) on diets labeled with either 13C-palmitic acid or 13C-leucine to isotopically enrich the lipids or the proteins in their bodies, respectively. The insects were allowed to become postabsorptive and then starved. We periodically measured the δ13C of the exhaled breath to characterize how each species adjusted their reliance on endogenous lipids and proteins as energy sources. We found that starving insects employ a wide range of strategies for regulating lipid and protein oxidation. All of the insects except for the beetle larvae were capable of sharply reducing reliance on protein oxidation; however, this protein sparing strategy was usually unsustainable during the entire starvation period. All insects increased their reliance on lipid oxidation, but while some species (grasshoppers, cockroaches, and beetle larvae) were still relying extensively on lipids at the time of death, other species (crickets and moth larvae) allowed rates of lipid oxidation to return to prestarvation levels. Although lipids and proteins are critical metabolic fuels for both vertebrates and insects, insects apparently exhibit a much wider range of strategies for rationing these limited resources during starvation.  相似文献
