The concentrations of Fe, Zn and Co were determined in up to five successive needle age classes in 54 individual Norway spruce trees from eight different sites (soil pH 3.1–7.7). Fe concentrations (12–25 μg in needles from the current year) were lower than most published values, due to the removal of surface contamination prior to analysis. Fe showed a significant positive correlation with Al. Successive needle age classes either had constant values or showed an increase for Fe concentrations; individual trees on a given site were rather uniform in their behaviour. Zn concentrations were 19–40 μg/g. On acid sites, they showed a positive correlation with total soil concentrations. The majority of trees showed decreasing Zn concentrations in successive needle age classes, but constant or increasing concentrations were also found; site homogeneity was less than with Fe. Co concentrations differed between trees on a neutral soil (12 ng/g) and on acid soils (41–174 ng/g). They showed a significant positive correlation with Mn needle concentrations. The changes of Co with needle age in most, but not all, trees were similar to those of Zn. The different changes of Fe, Zn and Co with needle age may be due to a different retranslocation. A modest retranslocation of Fe as opposed to a high retranslocation of Zn and Co (in most trees) is consistent with the observed behaviour. Embryogenic cell lines of Norway spruce ( Picea abies ) varying in growth habit and morphology were compared as regards profiles of extracellular proteins. Similar proteins were detected in the culture medium by SDS PAGE and in vivo labeling experiments, indicating that the proteins were secreted. Approximately 20 protein bands could be detected in the medium of each cell line. Three of the bands represented glycosylated proteins, as revealed by Concanavalin A staining. Some of the secreted proteins were similar for all tested embryogenic lines of Norway spruce, others were either specific for a group of cell lines or for individual cell lines. A correlation was observed between the morphology of the somatic embryos in a cell line and the presence of secreted proteins. The embryogenic cell lines of Norway spruce can be divided into two main groups. A and B, where A is characterized by somatic embryos with dense embryoheads and B by somatic embryos with loosely aggregated cells in their embryoheads. When proteins secreted from a cell line belonging to group A were added to cell lines belonging to group B, the somatic embryos of the B type developed further and became more similar in morphology to A-type embryos. These observations indicate that cell lines belonging to group A secrete certain proteins to the culture medium that are essential for the development of somatic embryos of Norway spruce.  相似文献

Bioindication can be carried out at different hierarchical levels, eg. cell, organism, and ecosystem. While the monitoring of damage by visible criteria (e.g. loss of needles) is connected with the organism as a whole, the monitoring of damage by biochemical indicators is above all connected with cell metabolism.
The degree of vitality of a tree can be ascertained through the integration of a number of biochemical parameters. Furthermore, a differential diagnosis of a particular stress pattern can be carried out because of the feedback pattern of several biochemical indicators. In order to describe and interpret biochemical or physiological changes that have been caused by a number of factors, multivariate statistical methods are being used more frequently. Apart from cluster and discriminant analysis, it is especially factor analysis which provides a helpful tool when dealing with problems in the field of environmental analysis. Factor analysis can be used for an integrating as well as a differentiating assessment.
Within the framework of forest damage research, numerous changes at the level of cell metabolism have been detected to which a bioindicative character can be attached. A number of physiological and biochemical parameters with bioindicative character concerning Norway spruce are presented.  相似文献

In northern boreal forests, the diversity of ectomycorrhizal (ECM) species is much greater than that of their host trees. This field study investigated the role of individual trees in shaping the ECM community. We compared ECM communities of eight Norway spruce (Picea abies) clones planted in a clear-cut area in 1994 with a randomized block design. In 2003, the ECM fungi were identified from randomly sampled root tips using denaturing gradient gel electrophoresis (DGGE) and rDNA internal transcribed spacer (ITS) sequence similarity. ECM diversity varied among clone groups, showing twofold growth differences. Moreover, according to detrended correspondence analysis (DCA), ECM community structure varied not only among but also within slow-growing or fast-growing clones. Results suggest that ECM diversity and community structure are related to the growth rate or size of the host. A direct or indirect influence of host genotype was also observed, and we therefore suggest that individual trees are partly responsible for the high diversity and patchy distribution of ECM communities in boreal forests.  相似文献

The morphology of somatic embryos of Norway spruce ( Picea abies ) varies among different cell lines, from less developed somatic embryos with small embryonic regions (group B) to well developed embryos with large embryonic regions (group A). Only well developed somatic embryos will undergo a maturation process after a treatment with ABA and develop into mature somatic embryos, which is required for plant regeneration. We have previously shown that the presence of specific extracellular proteins can be correlated with the morphology of the somatic embryos. In the present study we show that extracellular proteins concentrated from group A cell lines can stimulate group B embryos to develop further and that seed extract can stably convert B embryos into A embryos. The arabinogalactan protein (AGP) fraction of the extracellular proteins and of the seed extract was shown to be an active component for stimulating B embryos to develop further. Furthermore, the amount and type of extracellular AGPs, as detected with β-glucosyl Yariv reagent and monoclonal antibodies, varied among different types of tissues and cell lines. The data show that development of somatic embryos in Norway spruce is associated with particular extracellular AGPs, which have a regulatory function.  相似文献

Total protein was extracted from zygotic embryos and from somatic embryos of Picea abies (L.) Karst. (Norway spruce) cultured in vitro at different times during their development. An analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and 2-dimensional gel electrophoresis of the protein extracts showed that protein composition and the temporal changes in protein abundance were very similar in the two embryo types. Both zygotic and somatic embryos accumulated storage proteins in abundance during their maturation phase of growth; the somatic embryos when cultured on medium containing 90 m M sucrose and 7.6 μ M ABA. The major storage proteins are composed of polypeptides with molecular masses of about 22, 28, 33 and 42 kDa and they are identical in both embryo types according to their molecular mass and average isoelectric points. These proteins are also the most abundant proteins in the female gametophytic tissue of the mature seed.  相似文献

Short photoperiod induces growth cessation in seedlings of Norway spruce ( Picea abies (L.] Karst.). Application of different gibberellins (GAS) to seedlings growing under a short photoperiod show that GA9 and GA20 can not induce growth. In contrast application of GA, and GA4 induced shoot elongation. The results indicate that 3β-hydroxylation of GA9 to GA4 and of GA20 to GA1 is under photoperiodic control. To confirm that conclusion, both qualitative and quantitative analyses of endogenous GAs were performed. GA1, GA3, GA4, GA7, GA9, GA12, GA15, GA15, GA20, GA29, GA34 and GA51 were identified by combined gas chromatography-mass spectrometry in shoots of Norway spruce seedlings. The effect of photoperiod on GA levels was determined by using deuterated and 14C-labelled GAs as intermal standards. In short days, the amounts of GA9, GA4 and GA1 are less than in plants grown in continuous light. There is no significant difference in the amounts of GA3, GA12, and GA20 between the different photoperiods. The lack of accumulation of GA9 and GA20 under short days is discussed.  相似文献

The reliability of arbitrarily primed amplification products was tested. The segregation analysis of 266 amplification products obtained using 17 different 10-mer oligonucleotides in 34 megagametophytes from a single tree of Picea abies was carried out. Fifty-four out of the 165 variable bands fit the 1:1 segregation ratio expected for Mendelian traits. The segregation ratio of a subset of six RAPD markers in five other individuals from the same population confirmed their genetic nature. Our results strengthen the evidence previously reported that RAPDs markers can be considered Mendelian traits useful in the detection of genetic variability among both different individuals and populations.  相似文献

In order to optimize transient gene expression in Norway spruce pollen after DNA delivery with particle bombardment, effects of different conditions during homhardmenl were analysed using β-glucuroniduse (GUS) driven by the rice Act I promoter and Inciferase (LUS) driven by the tomato !at 52 promoter as reporter genes. Transient gene expression was significantly increased hy using two bombardments. Also the distance from the stopping plate to the sample was critical to gam maximum gene expression. There was no significant difference between gold and tungsten particles, and the number of positively stained pollen increased with increasing DNA concentration, from 5 to 40 pg DNA added in the DNA/tungsten solution The DNA delivery to Norway spruce pollen was most efficient at a chamber pressure above 70 kPa.  相似文献

Highly purified chloroplasts were isolated from spruce ( Picea abies [L.] Karst.) needles by free-flow electrophoresis. The chloroplast crude suspension was separated into two structurally different populations. The two populations could not be distinguished according to their protein/chlorophyll ratios and protein patterns from two-dimensional gel electrophoresis. The O2 evolution, however, showed differences: the chloroplast population deflected towards the cathode contained a major part of structurally intact chloroplasts in contrast to the population deflected towards the anode. The two populations were not contaminated by endoplasmatic membranes or mitochondria.  相似文献

 Vacuolar ATPase (EC 3.6.1.3) and PPase (EC 3.6.1.1) were studied in suspension cells and seedlings from spruce [Picea abies (L.) Karst. Proton transport activity and uncoupler (1 μM nigericin) stimulated substrate hydrolysis were measured in tonoplast enriched membrane vesicles. In suspension cells the vacuolar PPase exhibited 1.8-fold activity of the ATPase. In roots and needles from 12-week-old spruce seedlings the vacuolar PPase was inactive, whereas the ATPase was active. Therefore, we investigated whether the preparation of spruce tonoplast vesicles from roots and needles inactivates the vacuolar PPase but not the ATPase. For this purpose, maize (Zea mays L.) tonoplast membranes exhibiting vacuolar PPase as well as ATPase activity were used as a probe and added to the homogenization medium prior to the preparation of spruce vesicles. The preparation of spruce vesicles was more inhibitory to the vacuolar ATPase than to the PPase. The comparison of vacuolar PPases from spruce suspension cells and maize roots revealed similar enzymatic properties. After isopycnic centrifugation on continuous sucrose gradients the vacuolar PPase from spruce suspension cells co-purified with the vacuolar ATPase. Together, these data show: (1) vacuolar PPases from spruce suspension cells and maize roots are similar, (2) the preparation of tonoplast vesicles from spruce roots and needles does not inactivate the vacuolar PPase, (3) tonoplasts of suspension cultured cells and seedlings from spruce are differentially energized by the vacuolar pyrophosphatase that may indicate a difference in pyrophosphate metabolism between embryogenic and differentiated spruce cells, and (4) tonoplast vesicles from spruce seedlings may allow investigations of the effect of pyrophosphate on the vacuolar ATPase in the absence of vacuolar PPase activity.  The seasonal variation in maximum photochemical efficiency of photosystem II (Fv/Fm) and the relationship between Fv/Fm and climatic factors such as irradiance, frost-nights and daily mean temperature was studied in young Norway spruce trees for 4 years in northern Sweden. As a result of night frost, the Fv/Fm-ratio gradually decreased during the autumn. There was between-year variation in the pattern of Fv/Fm in fully exposed shoots during autumn and spring, largely as an effect of differing temperature conditions. During spring, there was a strong apparent relationship between daily mean temperature and Fv/Fm within the temperature range –3 to 12°C. The light regime to which the needles were exposed during winter affected Fv/Fm, and moderately shaded shoots from the bottom of the canopy generally had a higher Fv/Fm-ratio than fully exposed shoots from the top of the canopy.  Nod factors are lipochitooligosaccharides (LCOs) secreted by rhizobia. Nod factors trigger the nodulation programme in a compatible host. A bioassay was set up to test how crude (NGR234) and purified (NodS) Nod factors influence cell division and somatic embryogenesis in a conifer, Norway spruce (Picea abies). The Nod factors promoted cell division in the absence of auxin and cytokinin. More detailed studies showed that NodS stimulates development of proembryogenic masses from small cell aggregates and further embryo development. However, stimulation was only observed in low-density cell cultures. Our data suggest that rhizobial Nod factors substitute for conditioning factors in embryogenic cultures of Norway spruce. The objective of the present study was to investigate whether peak concentrations of ozone can deplete the apoplastic ascorbate pool of needles from Norway spruce trees (Picea abies L. Karst.) and, thereby, contribute to damage to forest trees. Twigs of forest trees grown at high altitude (1950m above sea level; Mt Patscherkofel, Austria) were enclosed in situ in chambers and fumigated for 5-5 or 17 h with ozone concentrations ranging from 60 to 798 nmol mol?1. Adjacent branches were fumigated with filtered air. Ozone influx into the foliage ranging from 1-7 to 17nmolm?2s?1 had little effect on whole-needle ascorbate or glutathione contents. However, apoplastic ascorbate decreased by about 30% when the needles were exposed to environmentally relevant ozone concentrations and increased about 3-fold at higher ozone concentrations. This response suggests the induction of ascorbate as a protective system and may also be important under field conditions. Needles of spruce trees from high altitude that were exposed to chronically increased ozone concentrations contained significantly higher apoplastic ascorbate concentrations than needles from spruce trees from lower altitudes with lower mean atmospheric ozone concentrations. The results show that peak concentrations of ozone do not act in spruce via a depletion of the apoplastic ascorbate pool.  相似文献

Norway spruce [ Picea aides (L.) Karst.] seed lots were obtained from populations growing on an acid soil in the Black Forest, West Germany (acid), and a calcareous soil in the Schwabische Alb, West Germany (calc). Seedlings were grown in sterile perlite culture containing 0–6 mM aluminium. Hypocotyl extension was inhibited by aluminium in the calcareous seedlings, hut not in the acidic seedlings. In a longer term experiment acidic and calcareous plants were grown for 10 weeks in perlite. Some of the tubes were inoculated with the fungus Paxillus involutus Fr. (designated F +) and some were not (F -). Aluminium sulphate solutions were then added to the tubes to raise the aluminium concentrations to 0–6 mM. Plants were harvested after a further 10 weeks. Fungus was associated with the roots in F+ plants, but mycorrhizas did not form. Growth of acid(F -) was somewhat stimulated by aluminium treatment, but that of calc(F -) was greatly reduced, and the plants were chlorotic. The presence of a rhizospherie fungus (F +) enhanced the growth of the calcareous plants, but had little effect on the acidic plants. Shoot analyses suggested that the greater aluminium sensitivity of the calcareous plants involves an inability to exclude aluminium or to maintain normal levels of calcium and magnesium uptake in its presence. The presence of rhizospheric fungi reduced the effects of aluminium.  相似文献

 Trees were regenerated from six white spruce embryogenic clones after cryopreservation for 3 and 4 years, respectively. Genetic stability was evaluated using randomly amplified polymorphic DNA (RAPD) fingerprints. Somaclonal variation was detected in some in vitro embryogenic cultures 2 and 12 months after they were re-established following cryopreservation but not in the corresponding regenerated trees. These results suggest that trees regenerated from cryopreserved cultures in subsequent years are primarily genetically stable in the genomic regions tested and that variation observed due to the in vitro culture process infrequently affects trees regenerated from normally maturing and germinating somatic embryos. However, trees regenerated from somatic embryos that matured or germinated abnormally in in vitro culture exhibited altered RAPD fragment patterns.  Genetic relationship within a population can be measured by average coancestry. This can also be expressed as an effective number which represents the relative genetic diversity of the population. The goal of breeding can be formulated to maximise genetic value minus average coancestry times a constant (the “penalty constant”). An iterative search algorithm can then be used to find the best selections for meeting this goal. Two such algorithms, one for a fixed number of selections and the other for a variable optimum number, were applied to select a mixture of field-tested Norway spruce clones with known parents. The results were compared with those from the conventional method of restricting parental contributions to the selected population as a means to control diversity. Coancestry-adjusted selection always yielded more gain than restricted selection at a given effective population size (except under circumstances where the methods were equivalent). Expressed another way, at any given level of gain, coancestry-adjusted selection maintained a larger effective population size than did restricted selection. The relative superiority of coancestry-adjusted selection declined when the effective population size approached the lowest value, that at which no penalty or restriction was applied. The method was extended by the second search algorithm to optimise the selected number of clones. The optimal number of clones can be rather large when diversity is heavily valued, but the reduction in genetic gain becomes large. Received: 7 April 1997 / Accepted: 9 June 1997  相似文献
