Little is known about the effect of sugars in controlling secondary metabolism. In this study, sugars alone or in combination with their analogs were used to investigate their role in the production of the antimalarial drug, artemisinin, in Artemisia annua L. seedlings. Compared to sucrose, a 200% increase in artemisinin by glucose was observed. Different ratios of fructose to glucose yielded artemisinin levels directly proportional to increases in relative glucose concentration. When the glucose analog, 3-O-methylglucose, was added with glucose, artemisinin production was dramatically decreased, but hexokinase activity was significantly increased compared to glucose alone. In contrast, neither mannose nor mannitol had any significant effect on artemisinin yield. In comparison with 30 g/l sucrose, artemisinin levels were significantly reduced by 80% in the presence of 27 g/l sucrose + 3 g/l palatinose, which cannot be transported into cells through the sucrose transporter. Together these results suggest that both monosaccharide and disaccharide sugars are likely acting not only as carbon sources but also as signals to affect the downstream production of artemisinin, and that the mechanism of these effects appears to be complex.  相似文献

The effects of sugars on root growth and on development of adventitious roots were analyzed in Arabidopsis thaliana. Seeds were sown on agar plates containing 0.0–5.0% sugars and placed vertically in darkness (DD) or under long day (LD, 16 h:8 h) conditions, so that the seedlings were constantly attached to the agar medium. In the sucrose-supplemented medium, seedlings showed sustained growth in both DD and LD. However, only dark-grown seedlings developed adventitious roots from the elongated hypocotyl. The adventitious roots began to develop 5 days after imbibition and increased in number until day 11. They could, however, be initiated at any position along the hypocotyl, near the cotyledon or the primary root. They were initiated in the pericycle in the same manner as ordinary lateral roots. Sucrose, glucose and fructose greatly stimulated the induction of adventitious roots, but mannose or sorbitol did not. Sucrose at concentrations of 0.5–2.0% was most effective in inducing adventitious roots, although 5.0% sucrose suppressed induction. 1,2-O-(R)-Trichloroethylidene-beta-D-mannofuranose (1) was obtained from the reaction of D-mannose with chloral. Reaction of 1 with potassium tert-butoxide (3 Mequiv) gave the thermodynamically stable 1,2,5-O-orthodichloroacetyl-beta-D-mannofuranose as the sole product whereas 1.5 Mequiv of reagent gave the kinetically controlled 1,2,3-O-orthodichloroacetyl-beta-D-mannofuranose (10) as the main product. Orthoester 10 gave the 5,6-isopropylidene derivative, which was also obtained from the reaction of 5,6-O-isopropylidene-1,2-O-(R)-trichloroethylidene-beta-D-mannofuranose with potassium tert-butoxide (1.5 Mequiv). These novel orthoesters are expected to prove useful as protecting groups and as building blocks in the formations of new mannofuranisidic units.  相似文献

Grape juice contains about equal amounts of glucose and fructose, but wine strains of Saccharomyces cerevisiae ferment glucose slightly faster than fructose, leading to fructose concentrations that exceed glucose concentrations in the fermenting must. A high fructose/glucose ratio may contribute to sluggish and stuck fermentations, a major problem in the global wine industry. We evaluated wine yeast strains with different glucose and fructose consumption rates to show that a lower glucose preference correlates with a higher fructose/glucose phosphorylation ratio in cell extracts and a lower K m for both sugars. Hxk1 has a threefold higher V max with fructose than with glucose, whereas Hxk2 has only a slightly higher V max with glucose than with fructose. Overexpression of HXK1 in a laboratory strain of S. cerevisiae (W303–1A) accelerated fructose consumption more than glucose consumption, but overexpression in a wine yeast strain (VIN13) reduced fructose consumption less than glucose consumption. Results with laboratory strains expressing a single kinase showed that total hexokinase activity is inversely correlated with the glucose/fructose (G/F) discrepancy. The latter has been defined as the difference between the rate of glucose and fructose fermentation. We conclude that the G/F discrepancy in wine yeast strains correlates with the kinetic properties of hexokinase-mediated sugar phosphorylation. A higher fructose/glucose phosphorylation ratio and a lower K m might serve as markers in selection and breeding of wine yeast strains with a lower tendency for sluggish fructose fermentation.  相似文献

Summary In field experiments conducted in 1981 on deep loess soils a significant correlation was obtained between EUF-N content in the topsoil and N in sugar beet roots as well as in whole plants. A very close correlation was found to exist between EUF-N values in June 1981 and -amino N in sugar beet roots at harvest. Also other quality criteria such as the contents of sugar, K and Na in roots and yield parameters correlated with EUF-N contents in topsoils.The inorganic and organic EUF-N fractions changed during the vegetation period due to both N uptake by the crop and mineralization. Therefore it appears to be expedient to use both fractions when calculating N fertilizer requirements.The correlation between EUF-N (topsoil samples drawn in the year preceding sugar beet) and sugar yield increases obtained by N fertilization that was found in Austria12 was confirmed in our experiments with soils from Southern Lower Saxony.  相似文献

A series of anomeric phosphates and ADP-activated L-glycero- and D-glycero-D-manno-heptopyranoses has been prepared in high overall yields, which provided model compounds and substrates in the elucidation of biosynthetic pathways and glycosyl transfer reactions of nucleotide-activated bacterial heptoses. The alpha-anomers of the heptosyl phosphates were obtained in high yield and selectivity using the phosphoramidite procedure, whereas the beta-phosphates were formed preferentially employing acylation of reducing heptoses with diphenyl phosphorochloridate. An efficient route to the formation of the nucleotide diphosphate sugars was elaborated by coupling of the O-acetylated phosphates with AMP-morpholidate followed by alkaline deprotection to furnish ADP-L- and D-glycero-alpha-D-manno-heptose in 84 and 89% yield, respectively. Deacetylation of the O-acetylated beta-configured ADP heptoses was conducted at strictly controlled conditions (-28 degrees C at pH 10.5) to suppress formation of cyclic heptose-1,2-phosphodiesters with concomitant release of AMP. Isolation of the unstable beta-configured ADP-heptoses by anion-exchange chromatography and gel-filtration afforded ADP L- and D-glycero-beta-D-manno-heptose in high yields.  相似文献

目的:研究胃癌患者血清糖链抗原125(CA125)、糖链抗原724(CA724)、癌胚抗原(CEA)、糖链抗原199(CA199)水平的表达及与临床病理特征的关系。方法:选取2016年5月-2017年8月我院收治的胃癌患者94例记为胃癌组,胃部良性病变患者82例记为良性病变组,另取同期于我院接受体检的健康志愿者80例记为对照组。分别测定三组受试者血清CA125、CA724、CEA、CA199水平,并分析上述指标与胃癌患者临床病理特征的关系,并观察胃癌患者各指标单独检测和联合检测的阳性率。结果:三组受试者血清CA125、CA724、CEA、CA199水平整体比较差异有统计学意义(P0.05),胃癌组、良性病变组、对照组的血清CA125、CA724、CEA、CA199水平呈逐渐降低趋势,两两对比差异有统计学意义(P0.05)。不同性别的胃癌患者CA125、CA724、CEA、CA199水平比较差异无统计学意义(P0.05),年龄60岁、TNM分期为Ⅲ-Ⅳ期、肿瘤大小≥5 cm2的胃癌患者CA125、CA724、CEA、CA199水平均高于年龄≤60岁、TNM分期为Ⅰ-Ⅱ期、肿瘤大小5 cm2的胃癌患者,差异有统计学意义(P0.05)。联合检测的胃癌阳性率高于CA125、CA724、CEA、CA199单独检测,且CA724单独检测高于CA125、CEA、CA199单独检测,差异有统计学意义(P0.05)。结论:胃癌患者血清CA125、CA724、CEA、CA199水平较高,与患者的年龄、TNM分期、肿瘤大小等因素有关,且联合检测的胃癌检出率较高,可为胃癌的早期诊断提供指导作用。  相似文献

During the last 2 years, several novel genes that encode glucose transporter-like proteins have been identified and characterized. Because of their sequence similarity with GLUT1, these genes appear to belong to the family of solute carriers 2A ( SLC2A, protein symbol GLUT). Sequence comparisons of all 13 family members allow the definition of characteristic sugar/polyol transporter signatures: (1) the presence of 12 membrane-spanning helices, (2) seven conserved glycine residues in the helices, (3) several basic and acidic residues at the intracellular surface of the proteins, (4) two conserved tryptophan residues, and (5) two conserved tyrosine residues. On the basis of sequence similarities and characteristic elements, the extended GLUTfamily can be divided intothree subfamilies, namely class I (the previously known glucose transporters GLUT1-4), class II (the previously known fructose transporter GLUT5, the GLUT7, GLUT9 and GLUT11), and class III (GLUT6, 8, 10, 12, and the myoinositol transporter HMIT1). Functional characteristics have been reported for some of the novel GLUTs. Like GLUT1-4, they exhibit a tissue/cell-specific expression (GLUT6, leukocytes, brain; GLUT8, testis, blastocysts, brain, muscle, adipocytes; GLUT9, liver, kidney; GLUT10, liver, pancreas; GLUT11, heart, skeletal muscle). GLUT6 and GLUT8 appear to be regulated by sub-cellular redistribution, because they are targeted to intracellular compartments by dileucine motifs in a dynamin dependent manner. Sugar transport has been reported for GLUT6, 8, and 11; HMIT1 has been shown to be a H + /myo-inositol co-transporter. Thus, the members of the extended GLUT family exhibit a surprisingly diverse substrate specificity, and the definition of sequence elements determining this substrate specificity will require a full functional characterization of all members.  相似文献

The base catalyzed conjugate Michael addition of the 1-thiosugar, 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranose, 1, to a new highly reactive enone 4-deoxy-1,2-O-isopropylidene-L-glycero-pent-4-enopyranos-3-ulose, 2, proceeds steroselectively with formation of adduct 3 in 94% yield. Convenient stereoselective reduction of the C-3 keto function of 3 with L-Selectride followed by in situ acetylation produces thiodisaccharide 4 in good 82% yield. Cleavage of the 1,2-O-isopropylidene protecting group with p-toluenesulfonic acid in methanol, followed by de-O-acetylation, produced an inseparable anomeric mixture of methyl 4-deoxy-5-C-(beta-D-glucopyranosyl)-thio-alpha/beta-L-ribo-pyranoside 5 in 72% overall yield. This approach constitutes a new general two-step click chemistry route to the previously unknown class of 4-deoxy-(1-->5)-5-C-thiodisaccharides as stable and biologically important glycomimetics.  相似文献

The change in population density of Cavelerius saccharivorus was studied in the sugar cane field and the Miscanthus habitats around it. This species spent 2 (or partly 3) generations a year. It was suggested that the density rose in the sugar cane field where the density had been low enough, when the population density increased in the nearby sugar cane fields. Likely, the density in the Miscanthus habitat increased with the density in the nearby sugar cane field. It seems that these are mainly due to flying movement of adults, and that the movement takes place in every season. It was found through the investigation in the sugar cane field that the percentage of long-winged adults increased with the population density. These adults seemed to emmigrate to the nearby sugar cane fields and Miscanthus habitats. The remarkable increase in the percentage of long-winged adults in the sugar cane field was probably due to the immigration into there. In the Miscanthus habitat the adult population was consisted mainly of long-winged ones. Through all the observations, it was suggested that the adult movement was associated with the population regulation and performed chiefly by long-winged adults.  相似文献

Sugar beets are a raw material for the production of sugar and ethanol. The decision on which end product to pursue could be facilitated by fast and reliable means of predicting the potential ethanol yield from the beets. A Near Infrared (NIR) Spectroscopy-based approach was tested for the direct prediction of the potential bioethanol production from sugar beets. A modified partial least squares (MPLS) regression model was applied to 125 samples, ranging from 21.9 to 31.0 gL(-1) of bioethanol in sugar beet brei. The samples were analyzed in reflectance mode in a Direct Contact Food Analyser (DCFA) FOSS-NIRSystems 6500 monochromator, with standard error of cross validation (SECV), standard error of prediction (SEP), coefficient of determination (r(2)) and coefficient of variation (CV) of 0.51, 0.49, 0.91 and 1.9 gL(-1), respectively. The NIR technique allowed direct prediction of the ethanol yield from sugar beet brei (i.e. the product obtained after sawing beets with a proper machine) in less than 3 min.  相似文献

Mesophyll cells and most types of storage cells harbor large central vacuoles representing the main cellular store for sugars and other primary metabolites like carboxylic- or and amino acids. The general biochemical characteristics of sugar transport across the vacuolar membrane are already known since a couple of years but only recently the first tonoplast sugar carriers have been identified on the molecular level. A candidate sucrose carrier has been identified in a proteomic approach. In Arabidopsis, the tonoplast monosaccharide transporters (TMT) represent a small protein family comprising only three members, which reside in the vacuolar membrane. Two of three tmt genes are induced upon cold, drought or salt stress and tmt knock out mutants exhibit altered monosaccharide levels upon cold induction. These observations indicate that TMT proteins represent the first examples of tonoplast sugar carriers involved in the cellular response upon osmotic stress stimuli.  相似文献

糖类是影响植物组织培养成功与否的关键之一。迄今为止,已用于植物组织培养的糖类有50多种。在植物体细胞组织培养中,蔗糖一直作为标准碳源,然而其他糖类物质如:葡萄糖、麦芽糖和山梨醇对植物体细胞培养也产生了一定的影响。在植物花药培养中,蔗糖较好,但应注意麦芽糖对花药培养的促进作用。  相似文献
