Cotyledons of Lupinus luteus were sampled from 1 to 21 days after sowing and processed for light microscopy and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE). Length, width, and surface area of the cotyledons increased gradually until day 10. The thickness of the cotyledons increased from day 7 to day 12 and decreased thereafter. Morphometric analyses showed that the increase in length, width, and thickness of the cotyledon was due to cell expansion, and the decrease in thickness of the cotyledon was due to the decrease in the length of abaxial cells and in the total number of cells. Mesophyll development accompanied schizogenous and lysigenous air space formation. There were two structurally distinct types of protein bodies. Protein bodies in five to six layers of cells on the abaxial side did not contain globoids, while globoids were prominent in protein bodies in the center and adaxial side. Storage protein mobilization occurred first in the abaxial side of the cotyledon and proceeded toward the adaxial side. SDS-PAGE analysis showed that proteins ranged from 97 to 14 kD. High molecular weight α- and ß-conglutinins were more abundant in the abaxial region, whereas γ-conglutinin occurred in both abaxial and adaxial regions. In addition, there were five minor bands between 97 and 43 kD unique to abaxial region and five minor bands between 43 and 14 kD unique to adaxial region in the nonreduced protein profiles. The α- and ß-conglutinins began to decrease after imbibition and disappeared by day 7 after sowing. The amount of gibberellin-like substances in the seed of Pharbitisnil increased in parallel with the growth of the seed, and attained20 days after anthesis to a maximum of 0.115 µg gibberellinA3 eqivalent per seed, when the seed reached its maximum freshweight or four-fifths of its final dry weight. At this maximumlevel, 0.03 µg and 0.1 µg gibberellin A3 equivalentswere localized in the embryo and in the "endosperm", respectively.Three gibberellin-like substances (Factors I, II and III) wereseparated upon paper chromatography. In view of the changesin amount of the factors with respect to the seed maturation,these factors, especially Factor II, in the "endosperm" andembryo were assumed to participate in the initial or the mainpart of growth of the embryo. Dwarf rice seedling and maizedwarfs 1, 3 and 5 responded to the three factors nearly in thesame way. Pharbitis dwarf, however, responded only to FactorI, but not to Factors II and III. Two gibberellin-like substances were separated chromatographicallyfrom the ethanol-extract of germinating cotyledons of Pharbitisnil. In the dark-grown seedling, these substances in the cotyledonincreased in amount gradually with the slow increase in freshweight and area of the tissue, and attained, 4 days after sowing,a maximum level of 0.03 µg gibberellin A3 equivalent pera couple of cotyledon; in the light-grown seedling, they increasedrapidly with the rapid increase in the fresh weight and area,and attained a maximum content of 0.07 µg gibberellinA3 equivalent per cotyledon on the 6th day. When the etiolatedseedling was exposed to light, the amount of the gibberellin-likesubstances in the cotyledon increased in parallel with openingof the hypccotyl hook and expansion of the cotyledon. The openingof the hook and increase of the substances occurred in red lightas well as in far-red. Relatively small amounts of the gibberellin-like substanceswere also contained in the germinating hypocotyl and root. The growth of cotyledonary disks was promoted by gibberellinA3 and the gibberellin-like substances from the expanding cotyledon. 应用微量分析方法检测了山黧豆胚胎发育过程中ODAP毒素含量和核酸、蛋白质、糖类等大分子物质的含量变化。结果表明:每粒种子的ODAP含量随着胚的发育而增加。每粒种子DNA量随着细胞的迅速分裂而增加,R、蛋白质、淀粉含量随着胚的发育而成倍地增加,当进入心形胚时这些物质的增加更为迅速。如以每克干重中的含量来表示,那么ODAP、DNA及可溶性糖含量则随胚的发育而下降,其它大分子物质含量在胚发育前期升高,进入心形胚时,这些物质达到最高峰;到鱼雷胚时,这些物质含量开始下降,直到胚基本分化完全时,降到最低点;只有酸性蛋白质含量一直保持增长。 相似文献

The average number of ovules produced per individual of Lupinus texensis is much greater than the average number of seeds per plant. Each plant produces approximately 2,000 ovules but only 2.5% develop into seeds. One fourth of the seeds is lost due to abortion and 0.3% is lost due to predation on the plant. Mature seeds from this population exhibit a five-fold range in weight, from 10 to 56 mg. The distribution of seed weights in the field population is skewed and leptokurtic. Seed wt is positively correlated with both seed germination and seedling survivorship. Heritability of seed wt is 0.09. There is no correlation between average seed wt per plant and total number of seeds per plant, seeds per pod, or legumes per plant. 相似文献

红蕉苹果后熟过程中其果肉内ABA含量开始迅速上升,比乙烯释放量的上升时间略早。在采后第12~14天,ABA水平达到高峰,尔后快速下降。当乙烯释放量在采后第15天出现高峰时,近果心的果肉ABA含量已降到其高峰值的1/4。青蕉苹果ABA和乙烯的变化趋势与前者相似,只是达到高峰所需要的时间稍长于前者。而较耐贮藏的国光苹果,所需时间更长,ABA水平也比前两个品种为低。 相似文献

Illus. 1959.—Photometric measurements were made of the densities of catechol tannin test colors histochemically developed with a nitrous acid reaction in fresh sections of fruits. The measured values showed a linear relationship with both section thickness and tannin concentration. When average cell size, section thickness. and the volume of tissue photometrically illuminated are known, it is possible to calculate an amount of phenolic substance per cell. Such values calculated for the naturally occurring catechol tannins in developing and ripening peaches revealed very large changes in these substances per cell. Catechol tannins so estimated increased about 90-fold from the beginning of cell enlargement until the fruit was slightly over half its mature size, a period of about 16-fold increase in volume of the average cell. After a maximum tannin content had been reached, a decrease of about 8-fold per cell was found as the fruit matured and ripened. The early changes in tannin content, when expressed on a unit volume basis, were much less pronounced (about 5-fold increases) and agreed in general with published chemical data for increases in chlorogenic acid on a weight basis during a comparable growth period in a sweet cherry. Myelin osmiophilia has been shown to develop significantly later than myelin staining by Luxol fast blue and Sudan black, in the developing kitten optic nerve. These histological changes are accompanied by alterations in the lipid composition of the optic nerve. Although myelination commences at about 10 days post partum in the nerve the appearance of cerebrosides is unexpectedly delayed. Changes in fatty acid chain length and lipid composition of optic nerve are consistent with the suggestion that ‘early’ myelin may be unchanged glial plasma membrane. 相似文献

应用光镜技术、组织化学定位及植物化学方法,研究了人参皂甙在绞股蓝营养器官中的积累分布状态以及不同生长期、不同器官、不同性别之间的绞股蓝总皂甙含量的动态变化。结果表明,绞股蓝人参皂甙主要分布在营养器官的同化组织及韧皮部薄壁细胞中,厚角组织、表皮及周皮的栓内层也有少量分布,木质部和髓薄壁组织中无皂甙分布;叶中皂甙积累最多,茎次之,根最少。绞股蓝在营养生长期→花果期→枯萎期的生长发育过程中,其地上部分的皂甙含量呈现出低→高→低的变化趋势;叶的含量高于茎,雄株的含量高于雌株。从而认为在9-10月的花果期采收绞股蓝的地上部分而保留地下茎和根,有利于药材品质和产量的提高,又有利于药用资源的可持续开发利用。 相似文献

Blastomyces luteus has been shown to be antagonistic in culture to both Verticillium albo-atrum and V. dahliae over a wide range of temperature and irrespective of the pH value of the medium. When B. luteus is grown on Dox's and potato-dextrose solutions it produces an exudate which, added to agar media, has an inhibiting effect upon the growth of these two species of Verticillium. The effectiveness of this inhibiting material is slightly reduced by boiling for 15 min.
When 'spent' Dox's liquid medium, which has supported the growth of B. luteus , is injected into tomato and antirrhinum seedlings inoculated with Verticillium no control of the disease is effected, but when B. luteus mycelium is added to soil infected with Verticillium , marked reduction in the incidence of disease results when the antagonistic organism and the pathogens have co-existed in the soil a sufficient length of time for the exudate of the former to be effective upon the development of the latter. 相似文献
