The humoral innate immune response of sharpsnout seabream Diplodus puntazzo against the myxozoan Enteromyxum leei was studied. Enteromyxosis was transmitted by cohabitation and a group of uninfected fish served as control. At 5, 12, 19, 26, 40 and 55 days post-exposure (p.e.), control and recipient fish were sampled to determine the prevalence of infection and some humoral innate immune parameters (antiprotease, antitumoral and peroxidase activities). Prevalence of infection was high from day 12 p.e. and reached 100% at days 40 and 55, when intensity of infection was medium to severe. The antiprotease activity was significantly increased in E. leei-exposed fish with respect to control fish at days 12 and 19 p.e. The serum antitumoral activity was slightly lower in recipient than in control fish at all sampling times, except at 40 days p.e., though no statistically significant differences were observed. Serum peroxidases were higher in all recipient fish than in control ones, with the highest stimulation index at 40 days p.e. Within recipient fish, no differences were detected between sampling times in any of the measured activities. The possible implication of these immune factors in the high susceptibility of D. puntazzo to this enteromyxosis is discussed.  相似文献

Nonacclimated and Cu-acclimated rainbow trout (Oncorhynchus mykiss) exhibited equally rapid clearance of a single bolus of injected (64)Cu (3,780 nmol/kg) from the plasma (32-40 min to half- concentration). Eight hours after Cu injection, approximately 80% of the injected Cu was found in the liver. However, when Cu labeled with (64)Cu was presented intravascularly via continuous infusion at a rate of 158 nmol x kg(-1) x h(-1) for 72 h, Cu-acclimated fish cleared plasma Cu more effectively than nonacclimated fish. The use of chronically implanted cystic bile duct cannulas revealed a fourfold increase in hepatobiliary Cu excretion in Cu-acclimated fish during infusion, demonstrating the important homeostatic role of the liver in Cu metabolism. Extrahepatobiliary Cu excretion, likely through the gills and apparently exceeding biliary Cu excretion, was evident from appearance of (64)Cu in the ambient water but was not altered by Cu acclimation. Cu accumulation in white muscle also played an important a role in copper homeostasis.  相似文献

The transient rise in plasma triacylglycerol fatty acids after single-dose ingestion of fish oil as triacylglycerols, free acids, or ethyl esters with linseed oil as an absorption standard was used to determine the relative absorption of fish oil fatty acids in eight men. As free acids, the fish oil fatty acids were well absorbed (greater than or equal to 95%). As triacylglycerols, eicosapentaenoic acid (1.00 g) and docosahexaenoic acid (0.67 g) were absorbed only 68% and 57% as well as the free acids. The ethyl esters were absorbed only 20% and 21% as well as the free acids. The incomplete absorption of eicosapentaenoic and docosahexaenoic acids from fish oil triacylglycerols correlates well with known in vitro pancreatic lipase activity.  相似文献

1. Intracellular distribution of a muscle alkaline proteinase was investigated on four kinds of fish. 2. The total activity of the muscle proteinase of carp, Cyprinus carpio, was larger in both myofibrillar (Mf) and microsomal (Mic) fractions than the activity in mitochondrial, lysosomal, and supernatant fractions. The activity found in Mf fraction seemed to due to the Mic enzyme which was not separated from Mf fraction. 3. The relative specific activity was mostly found in Mic fraction in the species tested. 4. The results indicate that the distribution pattern of fish muscle alkaline proteinase is different from those of cathepsin D and acid phosphatase. 5. The Mic fraction hydrolyzed Mf and sarcoplasmic proteins. The rates were 40 and 55%, respectively, of the rate when casein was used as a substrate.  相似文献

The effects of supplemental oral (0, 40, and 400 ppm) and parenteral iron (0 and 2.72 mg Fe iv given initially as a single dose) on iron absorption, excretion, and retention were determined in 30 rats. Endogenous fecal iron excretion was determined by the radioisotope dilution technique after im injection of 80 kBq Fe-59, using blood and certain body tissues as reference sources for the estimation of the specific activity (Bq Fe-59/micrograms Fe) of endogenous iron. The basal diet contained 3.6 ppm Fe. Fe(III)-hydroxide-polymaltose was used as the sole iron source in oral, iv, and im iron treatments. Iron balance as determined from day 14 to 20 of the experiment was not significantly affected by iv iron administration. Nevertheless, a temporarily reduced retention should have occurred, since differences in final body iron contents were lower than 2.72 mg, as compared to the respective untreated groups. Apparent iron absorption and iron retention increased with surplus oral iron, and the efficiency rates were highest with adequate iron supply (40 ppm). True absorption rates of iron were similar without any, and with 40 ppm Fe amounting 40 to 50% of the intake. In the iron deficient rats, half of the actually absorbed iron (about 16 micrograms/d) was lost by endogenous fecal re-excretion, and another 3 micrograms/d by the urinary route. Endogenous loss with feces and with urine increased with further oral iron supply, but at a considerably lower rate as total fecal excretion. Parenterally administered iron did not affect endogenous loss at all. The results indicate that endogenous excretion cannot be regarded as a means to eliminate excessive iron, and might actually be an inevitable loss.  相似文献

The chemotactic efficiency of macrophages isolated from the kidney of spot, Leiostomus xanthurus , and hogchoker, Trinectes maculatus. was determined in fish captured from the York River and the heavily polluted Elizabeth River (Virginia). Chemotactic activity was quantified in Boyden chambers using Escherichia coli as the chemotactic stimulus. Macrophage chemc-taxis was found to be markedly reduced in the Elizabeth River fish as compared to York River controls. Chemotactic migrations of macrophages at 90 min were 55% and 33% for control and experimental spot, respectively. Values for control and experimental hogchoker were 85% and 56%, respectively. The macrophage chemotactic activity of Elizabeth River fish returned to normal (spot, 56%; hogchoker, 80%) after the fish were held in clean water for 3 weeks. This indicates that the decreased chemotactic activity was related to exposure to Elizabeth River pollutants but may be reversible.  相似文献

Drinking rate and rectal fluid production of juvenile Atlantic salmon (1-2 g) in freshwater were investigated in unfed fish and recently fed fish. Drinking was also investigated following activation of the renin-angiotensin system (RAS) by two hypotensive agents, a nitric oxide (NO) donor sodium nitroprusside (SNP) and bacterial lipopolysaccharide (LPS). In unfed fish the basal drinking rate was 0.13 microL g(-1) h(-1) and rectal fluid production was 0.076 microL g(-1) h(-1). In recently fed fish both drinking rate and rectal fluid production increased significantly by about fivefold compared to unfed fish, and similar values were obtained for fish exposed to PS for 24 h. Exposure to SNP resulted in about a tenfold elevation of drinking rate and rectal fluid production, compared to unfed fish. Absorption of water by the gut was in the range 35-60% for all treatments. Drinking may have a role in processing food in the gut and the fluid in the gut may subjected to absorptive and secretory processes. The most likely route for removal of water absorbed by the gut is excretion via the kidney and this would result in an increased osmoregulatory burden on the fish. In polluted waters drinking could be increased through stimulation of the endogenous RAS by vasodilators, e.g., LPS and the gut could be a significant target for toxin exposure.  相似文献

The profiles of biliary, fecal and urinary excretion of tritium labeled prostaglandins (PG's) of differing biological activity were investigated in the rat. The PG's (10 micrograms/kg: 2 to 50 microCi/rat, in 1 ml polyethylene glycol-400) were administered intragastrically. Excretion data were expressed as a percentage of the total administered radioactivity. For the orally administered PG's 11R-methyl-16R-fluoro-15R-hydroxy-9-oxoprosta-ci s-5-trans-13-dienoic acid and its methyl ester, excretion was equally divided between urine and feces. The fecal and urinary profile of excretion of 3H after prostacyclin (PGI2) was similar to that following administration of 11R, 16, 16-trimethyl-15R-hydroxy-9-oxoprosta-cis-5-trans-13-dienoic acid (trimoprostil), a PG with antisecretory-antiulcer potential. However, PGI2 was very poorly absorbed from the intestine, while the absorption of trimoprostil was very efficient. Biliary excretion, with little entero-porto-hepatic biliary circulation, was the main route of elimination of trimoprostil, thereby resulting in rapid elimination of drug-related products and diminishing the potential for systemic liability in the rat.  相似文献

以体重为(4.86±0.02)g的虹鳟稚鱼为实验对象,在室内循环水养殖系统中进行为期90d的生长实验。用1000 U/kg植酸酶预处理大豆分离蛋白,然后以预处理的大豆分离蛋白(Soy protein isolate,SPI)分别替代0、20%、40%、60%、80%和100%的鱼粉蛋白,配制成6种等氮(粗蛋白为44.97%)、等能(粗脂肪为13.42%)的饲料,以评估饲料中鱼粉替代量对虹鳟生长、体组成、消化率及氮磷排泄的影响。90d的养殖实验结果表明,大豆分离蛋白替代鱼粉对虹鳟生长性能和饲料利用率产生显著的影响(P<0.05)。当大豆蛋白替代水平由0增加到40%时,虹鳟的特定生长率(SGR)显著提高,随着饲料中SPI替代水平由40%增加到100%时,虹鳟的SGR显著降低(P<0.05),在S0、S20和S60试验组之间差异不显著(P>0.05)。增重率(WGR)和蛋白质效率(PER)分别在处理组之间的差异关系与特定生长率(SGR)在各组之间的变化趋势相类似;当大豆蛋白分别替代40%时,其饲料系数(FCR)显著低于其他替代组(P<0.05),当替代量超过40%后,随着替代水平的增加,FCR显著增加(P<0.05)。随着替代水平由0升高到100%,磷排泄显著降低,与之相反的是,磷沉积率显著升高。随着替代水平由0升高至40%,氮排泄逐渐降低,当替代水平由40%升高至100%,氮排泄逐渐升高,随着替代水平升高至40%,氮沉积率升高。试验证明,大豆分离蛋白替代量达到40%时,虹鳟的生长及饲料转化率达到最佳,而高比例的SPI对虹鳟的生长性能和饲料效率产生负面影响。  相似文献

Eight different strains ofBacillus were isolated from fermented fish (Budu) and their proteolytic enzyme activities were determined after 18 h cultivation at room temperature (35° C). Four isolates possessed high protease activities. Optimum pH for these enzymes was between 7.0 and 8.0 and the optimal temperature was 55° C. The proteases retained 40% of their original activity after 20 min at 55° C but lost all activity at 65° C. Three of the four isolates were identified asBacillus subtilis, the fourth asBacillus licheniformis.  相似文献

We present the first data on the effect of hypoxia on the specific dynamic action (SDA) in a teleost fish. Juvenile cod (Gadus morhua) were fed meals of 2.5% and 5% of their wet body mass (BM) in normoxia (19.8 kPa Po(2)) and 5% BM in hypoxia (6.3 kPa Po(2)). Reduced O(2) availability depressed the postprandial peaks of oxygen consumption, and to compensate for this, the total SDA duration lasted 212.0+/-20 h in hypoxia, compared with 95.1+/-25 h in normoxia. The percentage of energy associated with the meal digestion and assimilation (SDA coefficient) was equivalent between the different feeding rations but higher for fish exposed to hypoxia. Comparing peak oxygen consumption during the SDA course with maximum metabolic rates showed that food rations of 2.5% and 5% BM reduced the scope for activity by 40% and 55%, while ingestion of 5% BM in hypoxia occupied 69% of the aerobic scope, leaving little energy for other activities.  相似文献

The knowledge about the combined effects of higher temperature and dietary nutrient quality on the diurnal nitrogenous excretion rates is very limited in farmed fish species including European sea bass. Therefore this study investigated the combined effects of increasing levels (30 vs. 60 %) of dietary fish oil replacement by equal mixture of cotton seed oil (CSO) and canola oils (CO) and two different ambient temperature (24 vs. 30 °C) on diurnal total ammonia and Urea–N excretion rates in European sea bass (Dicentrarchus labrax). Experimental diets were fed to fish three times (08:30–13:30–18:30 h) at a fixed rate of 3 % BW.d−1. The daily consumed nitrogen and energy intake of fish were similar during the investigation in different dietary treatments. However, the daily excretion rates of TAN, total nitrogen (TAN+Urea–N) and total nitrogen expressed as a proportion of consumed nitrogen by the European sea bass maintained at 30 °C were found to be significantly (P<0.001) higher (40 to 50%) than fish maintained at 24 °C in all the dietary treatments suggesting higher rates of deamination of ingested amino acids with increased temperature. Daily urea–nitrogen excretion of fish accounted for between 20–30 % of total ammonia–nitrogen excretion rates for each dietary treatment at 24 and 30 °C and appeared to be slightly increased by the temperature but neither the temperature nor the amount of plant oil mixture inclusion in diets or the interaction of these two factors had a significant effect on the urea nitrogen excretion rates of fish in different dietary treatments. However, Urea–N excretion rates in fish fed fish oil only (FO) diet were significantly higher (P<0.05) compared to that of fish fed diets containing increasing amount of plant oil mixture (VOM30 and VOM 60) during the light-on phase of the sampling period at 24 °C indicating periodic enhancement of permeability for urea at excretion sites. Further research is needed in order to elucidate the mechanism of nitrogenous excretion in European sea bass fed plant oil containing diets under extreme summer time temperatures employing total dietary fish oil replacement to reveal the possible effects of alteration in cell membrane phospholipid composition on enzymes responsible for nitrogenous excretion and/or detoxification.  相似文献

The present study was conducted to extend the understanding of the combined physiological effects of different food rations in combination with sublethal levels of copper in common carp (Cyprinus carpio). Fish acclimated to low (0.5% body weight) and high (5% body weight) food rations were exposed to 1 microM copper for a period of 28 days and kept for a further 14 days in copper free water to examine their recovery. Measurements of oxygen consumption, ammonia excretion and ammonia accumulation in plasma and muscle were done at various time intervals during the experimental period. Overall, oxygen consumption and ammonia excretion rates were significantly affected by food ration in both copper free and copper exposed fish. Additional challenges, such as copper exposure and/or exercise, significantly increased plasma and muscle ammonia in the fish fed a high food ration. Muscle ammonia levels in general responded slower (first increase after 72 h) and recovered within 2 weeks of exposure. There was a significant correlation between plasma ammonia levels, muscle ammonia levels and ammonia excretion rates. Influence of copper in terms of ammonia excretion and plasma ammonia accumulation was observed in high ration fish but low ration fish remained unaffected. This clearly indicates that ammonia metabolism was significantly influenced by copper in this group of fish showing that during unfavourable environmental conditions a high amount of food supply may turn deleterious to fish.  相似文献

本研究探讨了通过收集器不同粪便收集时间和饲料中肉骨粉(MBM)含量对异育银鲫干物质、蛋白、能量、磷的表观消化率 (ADC) 影响.不同梯度(0, 20, 40, 60, 80 ,100%)的肉骨粉替代鱼粉(FM) 蛋白配制成六种等氮 (粗蛋白: 410 g/kg) 等能 (总能: 18 kJ/g) 的饲料,通过11周的饲养实验,实验开始后2周开始收集粪便,收集时间分别是:排粪后 1 min, 投喂后4h和16h. 结果表明,干物质、蛋白、能量、磷的消化率明显随粪便收集时间增加而升高(p<0.05),但在高肉骨粉替代饲料中差异不显著(p>0.05). 干物质、蛋白、能量、磷的消化率随饲料中肉骨粉含量的增加呈线性或近线性下降.因此,在消化率测定中应该尽快收集排出的粪便以保证消化率的真实性.消化率是影响肉骨粉利用一个因素.  相似文献

Furosemide (FD; Lasix®) is a loop diuretic which strongly increases both urine flow and electrolyte urinary excretion. Healthy volunteers were administered 40 mg orally (dissolved in water) and concentrations of FD were determined in serum and urine for up to 6 h for eight subjects, who absorbed water at a rate of 400 ml/h. Quantification was performed by HPLC with fluorescence detection (excitation at 233 nm, emission at 389 nm) with a limit of detection of 5 ng/ml for a 300-μl sample. The elution of FD was completed within 4 min using a gradient of acetonitrile concentration rising from 30 to 50% in 0.08 M phosphoric acid. The delay to the peak serum concentration ranged from 60 to 120 min. FD was still easily measurable in the sera from all subjects 6 h after administration. In urine, the excretion rates reached their maximum between 1 and 3 h. The total amount of FD excreted in the urine averaged 11.2 mg (range 7.6–14.0 mg), with a mean urine volume of 3024 ml (range 2620–3596 ml). Moreover, the urine density was lower than 1.010 (recommended as an upper limit in doping analyses to screen diuretics) only for 2 h. An additional volunteer was administered 40 mg of FD and his urine was collected over a longer period. FD was still detectable 48 h after intake. Gas chromatography—mass spectrometry with different types of ionization was used to confirm the occurrence of FD after permethylation of the extract. Negative-ion chemical ionization, with ammonia as reactant gas, was found to be the most sensitive method of detection.  相似文献

The excretion of mutagens in the urine of cigarette smokers was studied as a model for absorption and elimination of complex carcinogenic and mutagenic mixtures in humans. Urine was collected from an occasional smoker who smoked 1 cigarette (17 mg tar/cigarette) and from a heavy smoker (smokes approximately 20 cigarettes/day) who quit for 2 days and then resumed smoking. Urine samples were collected for 6 days, including a 2-day pre-smoking period for the occasional smoker and pre-abstention period for the heavy smoker, respectively. Mutagen excretion patterns were determined by extracting the mutagens in each urine sample with XAD-2 resin and testing the extract in a microsuspension modification of the Salmonella/microsome liquid-incubation assay using bacterial strain TA98 with metabolic activation. Peak mutagenic activity of the urine collected from the two smokers appeared 4-5 h after the beginning of smoking. Activity decreased to pre-smoking "baseline' levels in approximately 12 h for the occasional smoker, and the activity for the heavy smoker approached the occasional smoker's 'baseline' in approximately 18 h after the cessation of smoking. The mutagen excretion patterns of the occasional smoker after smoking a single cigarette suggests that, the mutagens, as detected by the Salmonella assay, are absorbed rapidly (3-5 h) and are eliminated from the body following first order kinetics. The excretion rate constant for the occasional smoker was approximately 0.1 h-1 and the half-life (T1/2) was approximately 7 h.  相似文献

The effects of three fungicides of a new generation were studied for cyprinid young of the year. Acute toxicity (96 h of exposure) has an extremely negative effect on fish. The minimal lethal concentrations of fungicides (LC16) caused metabolic disorders and the activation of detoxication processes in carp liver at the very beginning of exposure to toxins (24–96 h). The increase in exposure led to the lipid peroxidation process intensifying and to a simultaneous decrease in the effectiveness of antioxidant protection (AOP) and detoxication. The inhibition of the activity of superoxide dismutase, catalase, and glutathione-S-transferase was observed, and the depletion of reduced glutathione reserves in the liver was found. The progression of intoxication over 15 days resulted in morphological and functional alterations in the fish liver and death in 49% of fish exposed to toxins.  相似文献

Feeding experiments with juvenile kuruma shrimp (Marsupenaeus japonicus) were conducted to understand the effects of supplemental levels of crystalline arginine hydrochloride on the growth and assimilation of arginine. In experiment 1 the juvenile shrimp were maintained on diets with and without arginine supplements. The addition of 3.0% arginine to a casein-based diet was slightly effective in improving the growth of the juveniles. In experiment 2, tracer experiments using [14C] arginine were conducted to clarify the ingestion and assimilation of arginine, 9 and 24 h after feeding, at different levels of supplemental arginine. Tracer experiments showed that the leaching rate of arginine 1 h after feeding ranged from 16 to 26% in the diets with different levels (0, 0.1, 1.5, 3.0 and 6.0%) of supplemental arginine; that is, 74-84% of the given arginine was actually ingested by the shrimp fed the diets. However, with increasing levels of supplemental arginine the incorporation rate of arginine into the whole body decreased and the utilization of absorbed arginine for body protein synthesis was reduced, whereas the excretion of absorbed arginine was increased. Thus, the absorbed arginine was not effectively utilized for body protein synthesis when large amounts of arginine were supplemented to the diets.  相似文献

The concentrations of urea and ammonia were measured in the portal and arterial blood simultaneously to the blood flow rate in the portal vein during the postprandial period (8 hrs.) after ingestion of a normal protein diet with 3% urea (10 meals) or without urea (12 meals) in conscious pigs (mean body weight: 55.5 +/- 2.3 kg). When no urea was present in the diet, there was a slight and permanent uptake of blood urea by the gut (570 mg/h, i.e. 9,5 mmoles/h) as well as a permanent appearance of ammonia in the portal vein (258 mg/h i.e. 15.2 mmoles/h), increasing with time (P less than 0.05). The absorbed ammonia nitrogen represented a maximum of 70% of urea nitrogen taken up. 2. Addition of urea to the diet brought about a large absorption of that substance (73% of the ingested amount) followed by a rather large excretion (960 mg/h, i.e. 16 mmoles/h), 5-6 hrs. after the meal and led to an increase (P less than 0.05) in the absorption of ammonia.  相似文献
