A radioisotope tracer technique and quantitative PCR were used to study the mechanisms and regulation of transepithelial transport of the type II organic anion methotrexate (MTX) by the Malpighian tubules of Drosophila melanogaster. Transport of MTX was saturable and Na+-independent; the kinetic parameters Jmax and Kt were 437 fmol min−1 and 23.5 μM, respectively. The transport of MTX was competitively inhibited by phenol red and probenecid; non-competitively inhibited by salicylate, verapamil and MK-571; and uncompetitively inhibited by Texas Red. Dietary exposure to 0.1 mM MTX led to dramatic increases in gene expression for several members of the ABC family of transporters in both the Malpighian tubules and the gut. Our results suggest that multiple transporters are upregulated in response to dietary exposure to MTX. Increased levels of the protein products which may result from expression of these genes may enhance elimination of toxic compounds such as MTX or its metabolites.  相似文献

A transepithelial potential of +8.74±0.29 mV (n = 85) has been recorded across the Malpighian tubules of Locusta. The effect of varying the Na+ and K+ concentration in the bathing medium on the transepithelial potential has been determined. The data show that the transepithelial potential does not obey the Nernst equation for K+. Ouabain, ethacrynic acid and amiloride all inhibit the transepithelial potential. The results are discussed in relation to the nature of the mechanisms of cation transport across the Malpighian tubules.  相似文献

The Malpighian tubules play a major role in haemolymph calcium homeostasis in insects by sequestering excess Ca2+ within the biomineralized granules that often accumulate in the tubule cells and/or lumen. Using the scanning ion‐selective microelectrode technique, measurements of basolateral Ca2+ transport are determined at several sites along the length of the Malpighian tubules isolated from the eight insects representing seven orders: Drosophila melanogaster (Diptera), Aedes aegypti (Diptera), Tenebrio molitor (Coleoptera), Acheta domesticus (Orthoptera), Trichoplusia ni (Lepidoptera), Periplaneta americana (Blattodea), Halyomorpha halys (Hemiptera) and Pogonomyrmex occidentalis (Hymenoptera). Ca2+ transport is specific to tubule segments containing Ca‐rich granules in D. melanogaster and A. aegypti, whereas Ca2+ transport is relatively uniform along the length of whole tubules in the remaining species. Generally, manipulation of second messenger pathways using cAMP and thapsigargin has little effect on rates of basolateral Ca2+ transport, suggesting that previous effects observed across midtubules of A. domesticus are unique to this species. In addition, the present study is the first to provide measurements of basolateral Ca2+ across single principal and secondary tubule cells, where Ca2+ uptake occurs only across principal cells. Estimated times for all tubules to eliminate the entire haemolymph Ca2+ content in each insect range from 6 min (D. melanogaster) to 19 h (H. halys) or more, indicating that rates of Ca2+ uptake by the Malpighian tubules are not always rapid. The results of the present study suggest that the principal cells of the Malpighian tubules contribute to haemolymph calcium homeostasis by sequestering excess Ca2+, often within specific tubule segments.  相似文献

5-Hydroxytryptamine (5-HT, serotonin) acts as a diuretic hormone in Rhodnius prolixus, where it increases to 0.1 μM in the haemolymph during feeding and stimulates the fluid secretion in isolated Malpighian tubules. The ouabain-sensitive (Na++K+)ATPase activity present in homogenates of Malpighian tubules from unfed Rhodnius prolixus is inhibited 60% by 0.01 μM 5-HT. This inhibition is reversed by ketanserin, a 5-HT2 receptor antagonist in mammals, and also by GDPβS, a competitive inhibitor of G-protein GTPase activity. GTPγS, a nonhydrolysable analog of GTP, and cholera toxin, a Gs-protein activator, also inhibit the ouabain-sensitive (Na++K+)ATPase activity, while pertussis toxin, a Gi-protein inhibitor, has no effect. The (Na++K+)ATPase activity is inhibited 55% by 0.4–100 μM dibutyryl-cAMP in the presence of IBMX, a phosphodiesterase inhibitor, which also potentiates the effect of a low concentration of 5-HT. The cAMP-dependent protein kinase inhibitor peptide abolishes the 5-HT effect. These data suggest that the (Na++K+)ATPase activity in Malpighian tubules is inhibited by 5-HT through activation of Gs-protein and a cAMP-dependent protein kinase.   • 1.1. The study was carried out on 22 species of insects from 5 orders. The osmolality of their hemolymph varied from 319 to 421 mOsm/kg H2O, concentration of Na+ 4.6 to 118 mM/l, K+ 6.3 to 73mM/l, Ca2+ 3.6 to 12.9 mM/l, Mg2+ 2.3 to 76 mM/l. The most abundant cation in the hemolymph of insects from higher orders is either K+ or Mg2+.
  • 2.2. In the muscles of lower and higher insects K+ is usually within 80–120 mM/kg wet wt.
  • 3.3. Most Ca2+ and Mg2+ in hemolymph is bound with protein and low molecular anions, concentration of free Ca2+ is 0.9-2.1mM/l Mg2+ 3.7–8.0 mM/l.

It is concluded that, in insects, potassium hemolymph, cell volume regulation and accumulation of ions in the cell, are ensured by an increased osmolality of hemolymph due to a high percentage contribution of low molecular organic substances which are retained in the hemolymph due to the absence of filtration apparatus in the Malpighian tubules.

Hemolymph calcium homeostasis in insects is achieved by the Malpighian tubules, primarily by sequestering excess Ca2+ within internal calcium stores (Ca‐rich granules) most often located within type I (principal) tubule cells. Using both the scanning ion‐selective electrode technique and the Ramsay secretion assay this study provides the first measurements of basolateral and transepithelial Ca2+ fluxes across the Malpighian tubules of an Orthopteran insect, the house cricket Acheta domesticus. Ca2+ transport was specific to midtubule segments, where 97% of the Ca2+ entering the tubule is sequestered within intracellular calcium stores and the remaining 3% is secreted into the lumen. Antagonists of voltage‐gated (L‐type) calcium channels decreased Ca2+ influx ≥fivefold in adenosine 3′,5′‐cyclic monophosphate (cAMP)‐stimulated tubules, suggesting basolateral Ca2+ influx is facilitated by voltage‐gated Ca2+ channels. Increasing fluid secretion through manipulation of intracellular levels of cAMP or Ca2+ had opposite effects on tubule Ca2+ transport. The adenylyl cyclase‐cAMP‐PKA pathway promotes Ca2+ sequestration whereas both 5‐hydroxytryptamine and thapsigargin inhibited sequestration. Our results suggest that the midtubules of Acheta domesticus are dynamic calcium stores, which maintain hemolymph calcium concentration by manipulating rates of Ca2+ sequestration through stimulatory (cAMP) and inhibitory (Ca2+) regulatory pathways.  相似文献

The presence of a Na+K+-activated, Mg2+-dependent ATPase (E.C. 3.6.1.3) has been demonstrated in microsomal preparations from the Malpighian tubules of Locusta. The effects of sodium and potassium ions, and different concentrations of ouabain, have been studied in relation to the activity of this enzyme and the ability of in vitro Malpighian tubule preparations to secrete fluid. From these studies it seems highly likely that a Na+K+ activated ATPase ‘pump’ is involved in fluid transport across the walls of the tubules.  相似文献

Recent studies have identified paracrine and endocrine cells in the midgut of larval Drosophila melanogaster as well as midgut and hindgut receptors for multiple neuropeptides implicated in the control of fluid and ion balance. Although the effects of diuretic factors on fluid secretion by isolated Malpighian tubules of D. melanogaster have been examined extensively, relatively little is known about the effects of such factors on gut peristalsis or ion transport across the gut. We have measured the effects of diuretic hormone 31 (DH31), drosokinin and allatostatin A (AST‐A) on both K+ transport and muscle contraction frequency in the isolated gut of larval D. melanogaster. K+ absorption across the gut was measured using K+‐selective microelectrodes and the scanning ion‐selective electrode technique. Allatostatin A (AST‐A; 1 μM) increased K+ absorption across the anterior midgut but reduced K+ absorption across the copper cells and large flat cells of the middle midgut. AST‐A strongly inhibited gut contractions in the anterior midgut but had no effect on contractions of the pyloric sphincter induced by proctolin. DH31 (1 μM) increased the contraction frequency in the anterior midgut, but had no effect on K+ flux across the anterior, middle, or posterior midgut or across the ileum. Drosokinin (1 μM) did not affect either contraction frequency or K+ flux across any of the gut regions examined. Possible functions of AST‐A, DH31, and drosokinin in regulating midgut physiology are discussed.  相似文献

The Malpighian (renal) tubules play important roles in ionic and osmotic homeostasis in insects. In Lepidoptera, the Malpighian tubules are structurally regionalized and the concentration of Na+ and K+ in the secreted fluid varies depending on the segment of tubule analyzed. In this work, we have characterized fluid and ion (Na+, K+, H+) transport by tubules of the larval stage of the cabbage looper Trichoplusia ni; we have also evaluated the effects of fluid secretion inhibitors and stimulants on fluid and ion transport. Ramsay assays showed that fluid was secreted by the iliac plexus but not by the yellow and white regions of the tubule. K+ and Na+ were secreted by the distal iliac plexus (DIP) and K+ was reabsorbed in downstream regions. The fluid secretion rate decreased > 50% after 25 μM bafilomycin A1, 500 μM amiloride or 50 μM bumetanide was added to the bath. The concentration of K+ in the secreted fluid did not change, whereas the concentration of Na+ in the secreted fluid decreased significantly when tubules were exposed to bafilomycin A1 or amiloride. Addition of 500 μM cAMP or 1 μM 5-HT to the bath stimulated fluid secretion and resulted in a decrease in K+ concentration in the secreted fluid. An increase in Na+ concentration in the secreted fluid was observed only in cAMP-stimulated tubules. Secreted fluid pH and the transepithelial electrical potential (TEP) did not change when tubules were stimulated. Taken together, our results show that the secretion of fluid is carried out by the upper regions (DIP) in T. ni Malpighian tubules. Upper regions of the tubules secrete K+, whereas lower regions reabsorb it. Stimulation of fluid secretion is correlated with a decrease in the K+/Na+ ratio.  相似文献

The effects of monovalent cations, inhibitors of metabolism dinitrophenol (DNP), carbonyl cyanide-p-trifluoromethoxyphenylhydrazone (FCCP), and KCN and temperature variations upon Ca2+ fluxes in intact roots of barley (Hordeum vulgare L. cv. Fergus and Herta) seedlings were investigated. 45Ca2+ influx was depressed in CaSO4-grown (low-salt) plants by the presence of NH4+, K+, or Na+ in the uptake medium. In contrast Ca2+ influx was slightly increased by Li+. In low-salt roots pretreated with KCN and in roots preloaded with K+ (high-K+ plants), the presence of K+ in the medium had no significant effect on Ca2+ influx, while in roots preloaded with Na+, the presence of K+ in the medium depressed Ca2+ influx. In absolute terms, Ca2+ influx was significantly greater in high-salt (both K+ or Na+ preloaded) than in low-salt roots.Patterns of 45Ca2+ efflux in the absence and in the presence of K+, NH4+, or Li+ in the external medium showed that these monovalent cations caused stimulation of 45Ca2+ efflux both from the cytoplasmic and vacuolar phases.It was noted that these modifications of Ca2+ fluxes by monovalent cations are transient and characteristic of a transitional stage of cation uptake by low-salt roots. We conclude that, together with stimulated active H+ efflux (another characteristic of this transitional stage), modifications of Ca2+ fluxes during monovalent cation uptake by low-salt roots is a response directed towards the maintenance of electrical neutrality.Determination of net fluxes revealed that the plants were close to Ca2+ flux equilibrium in the growth medium (0.5 mM CaSO4). Transfer of these plants to 0.5 mM CaSO4 + 0.25 mM K2SO4 caused a net release of CA2+ into the external medium.  相似文献

1. The sidedness of Ca2+-pump activation by Na+ and K+ was studied by atomic absorption spectrophotometry in human erythrocyte ghosts, which had been prepared in dextran solutions and resealed to alkali cations. 2. When ghosts were incubated in an all-choline medium, the increase in Nai+ elicited an inhibitory-stimulatory effect on Ca2+ extrusion. By contrast, only a stimulatory action was induced when choline was replaced by Na0+. 3. A dual effect on active Ca2+ efflux was also produced by increasing Ki+ or K0+. The biphasic response to the latter, however, was absent from high-K+ ghosts. Furthermore, the stimulation obtained at high K0+ was additive to that elicited by Ki+. 4. The results suggest that Na+ and K+ stimulate the Ca2+ pump of human red cells through two different mechanisms. The first one appears to be an electric coupling between Ca2+ efflux and the external activating cation. The other seems associated with the molecular reactions of the Ca2+-pump protein.  相似文献

Ionic composition of the haemolymph and electrophysiological responses of tarsal taste hairs were determined for individual flies of the species Calliphora vicina, in order to test the hypothesis that electrophysiological response values of individual flies are correlated with the ionic composition. Instead of flame photometry we used isotachophoresis to determine the ionic composition; only non-bound ions contribute to the result. Because of this we found lower concentration values than those reported so far. There was a strong correlation between the concentrations of the cations Na+, K+ and Mg2+, while Ca2+ was not related with any other cation. A significant correlation was shown to exist between the response of taste cells and the Na+, K+ and Ca2+ content in the haemolymph. These correlations explain, at least partly, the systematic differences in taste cell responses between flies, as reflected in interindividual variability.  相似文献

The Na+/Ca2+ antiporter was purified from beef heart mitochondria and reconstituted into liposomes containing fluorescent probes selective for Na+ or Ca2+. Na+/Ca2+ exchange was strongly inhibited at alkaline pH, a property that is relevant to rapid Ca2+ oscillations in mitochondria. The effect of pH was mediated entirely via an effect on the Km for Ca2+. When present on the same side as Ca2+, K+ activated exchange by lowering the Km for Ca2+ from 2  to 0.9 μM. The Km for Na+ was 8 mM. In the absence of Ca2+, the exchanger catalyzed high rates of Na+/Li+ and Na+/K+ exchange. Diltiazem and tetraphenylphosphonium cation inhibited both Na+/Ca2+ and Na+/K+ exchange with IC50 values of 10 and 0.6 μM, respectively. The Vmax for Na+/Ca2+ exchange was increased about fourfold by bovine serum albumin, an effect that may reflect unmasking of an autoregulatory domain in the carrier protein.  相似文献

Inward-rectifying K+ (Kir) channels play critical physiological roles in a variety of vertebrate cells/tissues, including the regulation of membrane potential in nerve and muscle, and the transepithelial transport of ions in osmoregulatory epithelia, such as kidneys and gills. It remains to be determined whether Kir channels play similar physiological roles in insects. In the present study, we sought to 1) clone the cDNAs of Kir channel subunits expressed in the renal (Malpighian) tubules of the mosquito Aedes aegypti, and 2) characterize the electrophysiological properties of the cloned Kir subunits when expressed heterologously in oocytes of Xenopus laevis. Here, we reveal that three Kir subunits are expressed abundantly in Aedes Malpighian tubules (AeKir1, AeKir2B, and AeKir3); each of their full-length cDNAs was cloned. Heterologous expression of the AeKir1 or the AeKir2B subunits in Xenopus oocytes elicits inward-rectifying K+ currents that are blocked by barium. Relative to the AeKir2B-expressing oocytes, the AeKir1-expressing oocytes 1) produce larger macroscopic currents, and 2) exhibit a modulation of their conductive properties by extracellular Na+. Attempts to functionally characterize the AeKir3 subunit in Xenopus oocytes were unsuccessful. Lastly, we show that in isolated Aedes Malpighian tubules, the cation permeability sequence of the basolateral membrane of principal cells (Tl+ > K+ > Rb+ > NH4+) is consistent with the presence of functional Kir channels. We conclude that in Aedes Malpighian tubules, Kir channels contribute to the majority of the barium-sensitive transepithelial transport of K+.  相似文献

This study examined the status of sarcolemmal Na+/K+-ATPase activity in rat heart under conditions of Ca2+-paradox to explore the existence of a relationship between changes in Na+/K+-pump function and myocardial Na+ as well as K+ content. One min of reperfusion with Ca2+ after 5 min of Ca2+-free perfusion reduced Na+/K+-ATPase activity in the isolated heart by 53% while Mg2+-ATPase, another sarcolemmal bound enzyme, retained 74% of its control activity. These changes in sarcolemmal ATPase activities were dependent on the duration and Ca2+ concentration of the initial perfusion and subsequent reperfusion periods; however, the Na+/K+-ATPase activity was consistently more depressed than Mg2+-ATPase activity under all conditions. The depression in both enzyme activities was associated with a reduction in Vmax without any changes in Km values. Low Na+ perfusion and hypothermia, which protect the isolated heart from the Ca2+-paradox, also prevented reperfusion-induced enzyme alterations. A significant relationship emerged upon comparison of the changes in myocardial Na+ and K+ content to Na+/K+-ATPase activity under identical conditions. At least 60% of the control enzyme activity was necessary to maintain normal cation gradients. Depression of the Na+/K+-ATPase activity by 60-65% resulted in a marked increase and decrease in intracellular Na+ and K+ content, respectively. These results suggest that changes in myocardial Na+ and K+ content during Ca2+-paradox are related to activity of the Na+/K+-pump; the impaired Na+/K+-ATPase activity may lead to augmentation of Ca2+-overload via an enhancement of the Na+/Ca2+-exchange system.  相似文献

During adaptation to brackish water the young great sturgeon Huso huso is able to regulate its serum osmolarity and ion concentrations. After transfer from fresh water to brackish water the ion concentrations in the urine increase and the urine becomes isoosmotic to the blood serum after 24h. The Na+ and K+ concentrations in the urine increase during the first 12 h by 4.4 and 7.7 times, respectively, later decreasing again. The Mg2+ and Ca2+ concentrations in the urine increase by 3.4 and 14 times during the first 72h in brackish water and remain high thereafter. These results suggest that the kidneys play an important part in the regulation of serum osmolarity and in the removal of Ca2+; however, in contrast to teleosts, Mg2+ must be removed extrarenally. During adaptation to a hyperosmotic medium the diameters of the Malpighian bodies, the glomeruli and the diameter of the tubules initially all decrease, but the distal tubules become morphologically differentiated into two regions and the diameter of the distal section later increases again. It is suggested that this is the site of Ca2+ secretion into the urine.  相似文献

We have previously demonstrated mobilization of Ca2+ in the efflux of Rb+ (K+) from isolated hamster brown adipocytes as a consequence of norepinephrine stimulation. We have now investigated the adrenoceptor subtype specificity of these responses and found them both to be of theα1-subtype. Futher, we have found that the Rb+ (K+) effux was dependent upon a primary Ca2+mobilization event in response to the α1-adrenergic stimulation, since the Rb+ efflux could also be demonstrated by the addition ionophore A23187 to the cells. The norepinephrine- and A23187-stimulated Rb+ effluxes were both inhibited by the Ca2+-dependent K+ -channel blocker apamin. Apamin also significantly attenuated Ca2+ mobilization in cells in response to a submaximal concentration of norepinephrine. We conclude that α1-adrenergic stimulation of brown fat cells leads to a mobilization of intracellular Ca2+ which, in itself or via other mechanisms, leads to an increase in cytosolic Ca2+ concentration which, in turn, activates a Ca2+ -dependent K+ channel leading to a K+ release from these cells. A possible role for this channel to sustain and augment the response toα1-adrenergic stimulation is discussed.  相似文献

In vitro preparations of Locusta Malpighian tubules are able to transport K+ against its concentration gradient. The ‘urine’ is slightly hyper-osmotic with respect to the bathing solution and the rate of secretion is inversely dependent on the osmotic pressure of the latter. The rate of fluid secretion increases with increasing temperature; being maximal at approx 40°C. The ionic composition of the secreted fluid, as indicated by Na+/K+ ratios, is altered by the presence of 1 mM ouabain in the bathing solution. Fluid secretion is inhibited by 1 mM ouabain. In addition, oxygen consumption by the Malpighian tubules is inhibited by either the presence of 1 mM ouabain or the absence of K+ in the bathing solution. The relationship between respiration, active transport and the Na+K+-activated ATPase is discussed.  相似文献

The existence of [Ca2+]i-activated K+-channels in the pancreatic β-cell membrane is based in two observations: quinine inhibits K+-permeability and, increasing intracellular Ca2+ stimulates it. The changes in K+-permeability of the β-cell have been monitored electrically by combining measurements of the dependence of the membrane potential on external K+ concentration and input resistance. The changes in the passive 42K and 86Rb efflux from the whole islet have been measured directly. Intracellular Ca2+ has been increased by various means, including increasing extracellular Ca2+, addition of the Ca2+-ionophore A23187 or noradrenaline and application of mitochondrial uncouplers and blockers. In addition to quinine, many other substances have been found to inhibit or modulate the [Ca2+]i-activated K+-channel. The most important of these is the natural stimulus for insulin secretion, glucose. Glucose may inhibit K+-permeability by lowering intracellular Ca2+. Glibenclamide, a hypoglycaemic sulphonylurea, is about 25 times more active than quinine in blocking the K+-channel in β-cells. The methylxanthines, c-AMP, various calmodulin inhibitors and Ba2+ also inhibit K+-permeability. Genetically diabetic mice have been studied and show an alteration in the [Ca2+]i-activated K+-channel.It is concluded that the [Ca2+]i-activated K+-channel plays a major role in the normal function of the pancreatic β-cell. The study of its properties should prove valuable for the understanding and treatment of diabetes.  相似文献

Our previous studies suggested the cross talk of nitric oxide (NO) with Ca2+ in regulating stomatal movement. However, its mechanism of action is not well defined in plant roots. In this study, sodium nitroprusside (SNP, a NO donor) showed an inhibitory effect on the growth of wheat seedling roots in a dose-dependent manner, which was alleviated through reducing extracellular Ca2+ concentration. Analyzing the content of Ca2+ and K+ in wheat seedling roots showed that SNP significantly promoted Ca2+ accumulation and inhibited K+ accumulation at a higher concentration of extracellular Ca2+, but SNP promoted K+ accumulation in the absence of extracellular Ca2+. To gain further insights into Ca2+ function in the NO-regulated growth of wheat seedling roots, we conducted the patch-clamped protoplasts of wheat seedling roots in a whole cell configuration. In the absence of extracellular Ca2+, NO activated inward-rectifying K+ channels, but had little effects on outward-rectifying K+ channels. In the presence of 2 mmol L−1 CaCl2 in the bath solution, NO significantly activated outward-rectifying K+ channels, which was partially alleviated by LaCl3 (a Ca2+ channel inhibitor). In contrast, 2 mmol L−1 CaCl2 alone had little effect on inward or outward-rectifying K+ channels. Thus, NO inhibits the growth of wheat seedling roots likely by promoting extracellular Ca2+ influx excessively. The increase in cytosolic Ca2+ appears to inhibit K+ influx, promotes K+ outflux across the plasma membrane, and finally reduces the content of K+ in root cells.  相似文献
