过氧化物酶是植物抗病过程中的关键酶,也参与植物抗低温胁迫以及一些正常的植物生长发育生理过程。冷调控蛋白是植物抗低、高温的重要蛋白。近年的研究表明植物抗病、某些正常生长发育过程。冷调控蛋白是植物细胞程序性死亡有关。以玉米自效系黄早四为材料,采用生物素标记,利用原位杂效技术绎玉米中过氧化物酶和冷调控蛋白编码基因px与cld进行了原位杂效定位,DAB和荧光检测得到了一致的结果。在2号和7号染色体长臂同时 相似文献

玉米基因mir1编码一种抗秋季黏虫的半胱氨酸蛋白酶。利用RFLP作图mir1基因被定位在玉米第 6号染色体短臂上 ,但它在第 6号染色体短臂上的物理位置还不知道。实验以mir1和 4 5SrDNA为探针 ,通过双色荧光原位杂交技术确定了mir1基因在玉米细胞分裂中期和粗线期第 6号染色体上的物理位置。Southern杂交结果表明 ,在薏苡基因组中存在mir1基因的同源序列 ,进一步利用荧光原位杂交的方法确定mir1基因的同源序列定位于薏苡第 7号染色体长臂的近末端 ,其信号与着丝粒的百分距离为 73 33± 0 15。 相似文献

Abstract: The effect of chilling on the intercellular distribution of mRNAs for enzymes of assimilatory sulfate reduction, the activity of adenosine 5'-phosphosulfate reductase (APR), and the level of glutathione was analysed in leaves and roots of maize ( Zea mays L). At 25 °C the mRNAs for APR, ATP sulfurylase, and sulfite reductase accumulated in bundle-sheath only, whereas the mRNA for O-acetylserine sulfhydrylase was also detected in mesophyll cells. Glutathione was predominantly detected in mesophyll cells; however, oxidized glutathione was equally distributed between the two cell types. Chilling at 12 °C induced oxidative stress which resulted in increased concentrations of oxidized glutathione in both cell types and a prominent increase of APR mRNA and activity in bundle-sheath cells. After chilling, mRNAs for APR and sulfite reductase, as well as low APR activity, were detected in mesophyll cells. In roots, APR mRNA and activity were at higher levels in root tips than in the mature root and were greatly increased after chilling. These results demonstrate that chilling stress affected the levels and the intercellular distribution of mRNAs for enzymes of sulfate assimilation. 相似文献

A maize genome library was constructed using maize W22 DNA from leaf tissue nuclei and bacteriophage λCh4 as the vector. cDNA clones of zein mRNA were used to identify homologous genomic sequences in the Ch4 maize library. Each of the genomic clones identified has homology to a family of mRNAs in the zein mRNA population. 在很多转录因子中发现的锌指结构,被认为在人类心脏的发育和相关疾病的发生过程中发挥重要的作用。本文报道了克隆和表达分析人类新的锌指蛋白基因ZNF18。该基因cDNA长2 767 bp,编码一个有549个氨基酸的蛋白,这一蛋白含有一个SCAN结构域,一个KRAB结构域和5个连续的C2H2型锌指结构域。ZNF18蛋白与小鼠Zfp535有77%的同源性。ZNF18基因定位于人染色体17p12~p13,包含9个外显子和8个内含子。以ZNF18全长编码区为探针进行Northern杂交,结果显示ZNF18在成体小鼠各组织中广泛表达,但在心脏中低丰度表达。整体原位杂交结果显示,ZNF18基因在小鼠胚胎的表达有很高的动态性。ZNF18主要在E7.5小鼠胚胎的胚外组织表达,E8.5出现了胚胎躯干前端表达。ZNF18从E9.0开始在胚胎的心脏和尾部表达,尤其在E10.5胚胎的心脏高丰度表达。这提示ZNF18基因与心脏发育过程可能有密切的关系。 相似文献

The differential distribution of beta tubulin mRNAs in individual mammalian brain cells 总被引:2,自引:0,他引:2
It has been shown by in vitro translation of polyadenylated messenger RNAs (poly(A)+ mRNAs) that the mRNAs encoding both alpha and beta tubulin isotypes are present at much higher relative levels in the developing rat brain than they are in the adult, suggesting that the requirements for tubulin subunits vary with cell type and/or with the developmental stages of a particular cell type. The postnatally developing rat cerebellum, with its readily identifiable cell populations that perform the gamut of developmental tasks, is a suitable model for analyzing specific cellular mRNA distributions during development. In this report, by in situ hybridization techniques it is shown that, by comparison to total cellular poly(A)+ mRNA levels, there is relatively more of the total beta tubulin mRNAs in mitotically active external granule layer cells than in those in the internal granule layer. These results show that migration and differentiation of these granule cells is accompanied by a decrease in their beta tubulin mRNA levels relative to the levels in granule cells of the external granule cell layer. Furthermore, the relative levels of beta tubulin mRNA both in the prenatally formed Purkinje cells and the postnatally formed stellate cells are two to fourfold less than in the granule cells of the internal granule cell layer. 相似文献

Knobs are blocks of heterochromatin present on chromosomes of maize (Zea mays L.) and its relatives that have effects on the frequency of genetic recombination, as well as on chromosome behavior.Knob heterozygosity and instability in six maize inbred lines and one Z. diploperennis Iltis Doebley line were investigated using the fluorescence in situ hybridization (FISH) technique with knob-associated tandem repeats (180 bp and 350 bp (TR-1)) as probes. Signals of seven heterozygous knobs containing 180-bp repeats and of one heterozygous knob containing TR- 1 were captured in chromosomes of all materials tested according to the results of FISH, which demonstrates that the 180-bp repeat is the main contributor to knob heterozygosity compared with the TR-1 element. In addition, one target cell with two TR-1 signals on one homolog of chromosome 2L, which was different from the normal cells in the maize inbred line GB57,was observed, suggesting knob duplication and an instability phenomenon in the maize genome. 相似文献

We used fluorescence in situ hybridization to identify and map the position of B chromosomes (supernumerary chromosomes) within
maize sperm cells. Observations on over 1,000 sperm cells from several genotypes show that, on average, the B chromosomes
are positioned in the tip one-fourth of the sperm nucleus two-thirds of the time. In contrast, the centromeres and knobs of
the A chromosomes (the normal set) are not restricted to the tip portion of the nucleus. To our knowledge, this is the first
example of specific chromosome positioning within a plant gamete. Studies on nuclear architecture of somatic cells in both
plants and animals suggest that chromosome behavior and gene expression may correlate with chromosome position within the
nucleus. The functional significance of nonrandom positioning of the B chromosomes within maize sperm is as yet unclear.

In situ hybridization using biotinylated cDNA probes of 18S rRNA, 5S rRNA, tRNAfMet, tRNAcys, tRNAAsn was performed on ultra-thin sections of K4M-embedded maize root tip. After hybridization, the biotinylated hybrids were detected with avidin coupled to 10 nm gold particles and then examined under the electron microscopy. The results showed that 18S rRNA, 5S rRNA and tRNA all existed in the metaphase chromosomes at random. They were distributed not only in the interior of the chromosomes, but also in the periphery of the chromosomes. Three tRNAs and 5S rRNA in the chromosomes were equal in amount to that in the cytoplasm, but the amount of 18S rRNA in the chromosomes was much higher than that in the cytoplasm. These results indicated that a part of the RNAs in the chromosomes came from the nucleolus, while others came from the nucleoplasm. 相似文献

为了探索东亚三角涡虫Djp53基因在涡虫组织中的表达和功能,利用整体原位杂交、RT-PCR技术,检测了涡虫Djp53基因在组织中的表达分布特点,结果表明,Djp53基因在再生1、3、5天的胚基中具有较强的阳性信号,且3天的表达量最高;而在再生7、10天和成虫的实质组织中表达较弱.RNA干扰后的RT-PCR检测显示,Djp53表达量显著下降,涡虫不能正常再生或出现眼点缺陷.由此推断,东亚三角涡虫Djp53基因在早期胚基发育阶段,通过调节多功能干细胞的迁移和增殖分化影响早期胚基的形成,是涡虫早期胚基发育必不可少的一个基因,并且在涡虫成体和再生后期对多功能干细胞的维持具有重要的作用. 相似文献

Copy DNAs corresponding to a highly repetitive, proline-rich protein from maize have been cloned by differential screening of a coleoptile cDNA library. The deduced amino acid sequence contains a single repetitive element of carrot extensin (Ser-Pro-Pro-Pro-Pro). The related mRNAs have a defined distribution in tissues of the plant and are accumulated mainly in the coleoptile node and root tip. A peptide that corresponds to one of the repetitive elements of the protein has been synthesized and antisera have been obtained in rabbits. These antibodies react against crude preparations of coleoptile cell wall and against polypeptides extracted following the protocols described for the extraction of extensin. From these data it is concluded that the cDNAs correspond to a family of cell wall glycoproteins from maize. 相似文献
