The internal colony-forming bacterial flora of the schistosome intermediate host snailBiomphalaria glabrata (Say) has been characterized in ca. 500 individual snails from Puerto Rico, Guadeloupe, and St. Lucia, and from laboratory aquaria. Freshly captured wild snails harbor 2–40×106 CFU·g–1, and healthy aquarium snails harbor 4–16×107 CFU·g–1, whereas moribund individuals have 4–10 times as many bacteria as healthy individuals from the same habitats.Pseudomonas spp. are the most common predominant bacteria in normal snails, whereasAcinetobacter, Aeromonas, andMoraxella spp. predominate in moribund snails. External bacterial populations in water appear to have little effect on the composition and size of the flora in any snail. In addition to normal (healthy) and moribund snails, a third group of snails has been distinguished on the basis of internal bacterial density and predominating genera. These high-density snails may have undergone stresses and may harbor opportunistic pathogens. The microfloras of wild and laboratory-reared snails can be altered and stimulated to increase in density by crowding the snails or treating them with antibiotics.  相似文献

  • 1.1. Rainbow trout maintained in fresh water or Actapted to sea-water for 24 hr were fed casein-based dry diet. After feeding, fish were kept in fresh water (FW) or transferred to artificial sea-water (SW) and sacrificed after 10 or 20 hr.
  • 2.2. The digestive tract was separated into five parts: stomach, pyloric caeca region, middle intestine and two equal lengths of rectum.
  • 3.3. The content of these parts was analysed for ions Na+, K+, Cl, Mg2+ and for free, peptide and total amino acids.
  • 4.4. In the fish stomach all ions, with the exception of Ca2+, indicate drinking of sea-water. In the pyloric caeca region Na+ appears to be efficiently absorbed in SW fish but influxed in FW fish. In the rectum of SW fish K+ appears to be reabsorbed but Na+ concentrated in faeces.
  • 5.5. Free amino acid concentrations were always higher in gut lumen of SW than in FW fish in respect to time after feeding and portion of intestinal content. Free amino acids constitute at most 7.4–8.7% of total amino acids in the content of pyloric caeca region.
  • 6.6. Peptide amino acids, being mostly di-, tri- and tetra-peptides, increased in stomach content from 14.7 to 28.4% of the total, from 6 to 10 hr after a meal in SW fish. Peptide amino acids constituted 80.3–89.0% of total amino acids in intestinal content of the pyloric caeca region. These peptide portions decreased in the mid-intestine (47.5–52.5%) and increased again in the rectum (73.6–76.0%).
  • 7.7. It was concluded that in rainbow trout fed in both sea- or fresh water, ion concentrations do not seem to interfere with protein digestion and nutrient absorption in alimentary tract.

The proliferative activity of the pyloric caeca of Asterias rubens was investigated. Autoradiographic experiments using intracoelomically injected (methyl-3H)-thymidine were performed throughout the year and incorporation into pyloric caeca and into gonads was studied. Tritiated thymidine was found to be incorporated mainly in the coelomic lining of both organs. Cell divisions in the coelomic lining may be necessary for the growth of these organs, for the production of coelomocytes or, in the case of the pyloric caeca, for growth of the digestive epithelium. Proliferative activity of the digestive epithelium of the pyloric caeca was only observed in the median duct. It is hypothesized that new cells, arising from mitosis, grow from the median duct to the side lobes and differentiate into storage cells, for example. The existence of a mitosis-inducing or mitosis-stimulating substance is discussed. In the ovaries follicle cells were found to incorporate (methyl-3H)-thymidine; in the testis, proliferation of the germinal epithelium occurred simultaneously in all spermatogenic columns. First, the spermatogonia and then later the spermatocytes became labeled. Absorption of substances from the coelomic fluid is discussed.  相似文献

Summary The biochemical composition of the pyloric caeca of femaleAsterias rubens was studied during the annual reproductive cycle. The amounts of nucleic acids, DNA and RNA were measured in addition to the amounts of lipids, glycogen, other reducing carbohydrates, free amino acids and proteins.A positive correlation of the pyloric caeca index to the biochemical constituents at the cellular level was established. The total DNA content of the pyloric caeca was found to increase with the pyloric caeca index.The relation of the biochemical composition of the pyloric caeca to the size changes of the organ is discussed. The hypothesis that the changes in size of the organ are due to changes both in the number and size of the cells is confirmed. Comparative analysis was performed of morphological characters and coloration elements in goatfish of the genus Mullus from collections of the Zoological Museum (Moscow State University, Russian Academy of Sciences), on the basis of which individuals from Atlantic waters off the coasts of western Africa were identified as M. argentinae. Brief characteristics of all valid species of the genus are presented; the main diagnostic characters include: ratio between the length of the barbels and pectoral fins, number of pyloric caeca, and the presence of a dark spot (or stripe) in the upper part of dorsal fin. The Mediterranean populations of M. surmuletus are separated as a special subspecies M. surmuletus fuscatus, and M. argentinae from eastern Atlantic is described as a new subspecies M. argentinae africanus.  相似文献

Summary In vivo electrolyte transport and water absorption from the caeca of dehydrated, low-NaCl diet hens are reported. In the absence of luminal glucose or acetate, net electrolyte transport rates and water absorption are small. When physiological concentrations of acetate (40 mM) are included in the perfusate, Na+ transport and water absorption increase significantly (P<0.01): 38±7 eqNa+/caecum kg·h and 256±33 l H2O/caecum · kg · h.A similar increase in water absorption occurs with the inclusion of 15 mM glucose in the perfusate (219±30 l H2O/caecum · kg · h), however both net Na+ and Cl absorption increase: 28±6 eq Na+/caecum · kg · h and 21±5 eq Cl/caecum kg · h.These pronounced increases in electrolyte and water absorption are not accompanied by any significant increase in transmural potential difference.The data presented establish caeca as important sites in the recuperation of water and electrolytes in dehydrated, low-NaCl diet hens.Abbreviations ECPD electrochemical potential difference - PD (transmural) potential difference - PEG polyethylene glycol  相似文献

Summary The fermentation by Candida shehatae and Pichia stipitis of xylitol and the various sugars which are liberated upon hydrolysis of lignocellulosic biomass was investigated. Both yeasts produced ethanol from d-glucose, d-mannose, d-galactose and d-xylose. Only P. stipitis fermented d-cellobiose, producing 6.5 g·l-1 ethanol from 20 g·l-1 cellobiose within 48 h. No ethanol was produced from l-arabinose, l-rhamnose or xylitol. Diauxie was evident during the fermentation of a sugar mixture. Following the depletion of glucose, P. stipitis fermented galactose, mannose, xylose and cellobiose simultaneously with no noticeable preceding lag period. A similar fermentation pattern was observed with C. shehatae, except that it failed to utilize cellobiose even though it grew on cellobiose when supplied as the sole sugar. P. stipitis produced considerably more ethanol from the sugar mixture than C. shehatae, primarily due to its ability to ferment cellobiose. In general P. stipitis exhibited a higher volumetric rate and yield of ethanol production. This yeast fermented glucose 30–50% more rapidly than xylose, whereas the rates of ethanol production from these two sugars by C. shehatae were similar. Synopsis The effect of southern flounder,Paralichthys lethostigma, predation on the survival and size-distribution of spot,Leiostomus xanthurus, was investigated using feeding experiments in outdoor tanks and estuarine ponds. In the outdoor tank experiments, southern flounder, 143 mm average standard length (SL), fed for 48 h on spot (25–30 mm SL), over a range of densities (4–20 m–2). In these trials, flounder showed a Type II functional response to spot density. In the estuarine pond experiment, flounder density (2 or 4 flounders 70 m–2) contributed significantly to spot mortality relative to predator-free controls. The effect of flounder density on spot mortality was non-linear; in the 4-flounder treatment the daily instantaneous mortality due to flounder (0.0177) was 3.5 times that in the 2-flounder treatment (0.0051). The total spot mortality rate in the 4-flounder treatment (0.028) was similar to that observed from published field observations. Flounder also altered the size distribution of spot relative to predator-free controls. Predation by flounder resulted in fewer intermediate-sized spot at the end of the experiment when compared to predator-free controls. Flounder also significantly reduced survival and mean length of two killifishes,Cyprinodon variegatus andFundulus heteroclitus, that invaded the pond. Flounder predation was a significant size-structuring force on the prey fish assemblage in the pond. Results from both the outdoor tank experiment and the pond experiment indicate that flounder may represent a significant source of mortality for juvenile estuarine fishes.  相似文献

The turbellarian Triloborhynchus psilastericola n.sp. (family Pterastericolidae) occurs in the starfish Psilaster andromeda. On approaching maturation it moves from the host's coelom into the glandular pockets of the pyloric caeca with simultaneous loss of almost the entire ciliation. Specimens of a little-known nematode, Spinitectus oviflagellis Fourment, 1884, the type-species of Spinitectus Fourment, 1884, were collected mainly from the pyloric caeca of a marine deep-water fish, the onion-eye grenadier Macrourus berglax Lacépède (a new host record), in the eastern Greenland Sea, North Atlantic Ocean. Studies using light and scanning electron microscopy revealed some taxonomically important, previously unreported features of S. oviflagellis, such as the detailed structure of the cephalic end, the position of the excretory pore and the presence of ventral pre-anal cuticular ridges (area rugosa) in the male, which indicated a certain degree of intraspecific biometrical variability in this species. S. oviflagellis is compared with similar congeneric species parasitising marine fishes.  相似文献

  • 1.1. Levels of progesterone and oestrone were estimated by means of RIA in the gonads and pyloric caeca of male specimens of Asterias rubens throughout the reproductive cycle.
  • 2.2. The patterns obtained appeared to be sex-specific on comparison with those of female specimens of A. rubens and it was concluded that progesterone and oestrone in the testes are involved in the regulation of gametogenesis.
  • 3.3. The levels of progesterone in the pyloric caeca were about ten times higher than those in the testes; in the same sequence levels of oestrone were only slightly higher.
  • 4.4. The onset of a new reproductive cycle, taking the abrupt decrease of the pyloric caeca-index as a marker, coincides with a strong decrease of the progesterone level (P < 0.001) and an increase of the oestrone level (P < 0.01) in the pyloric caeca.

To gain information on extended flight energetics, quasi-natural flight conditions imitating steady horizontal flight were set by combining the tetheredflight wind-tunnel method with the exhaustion-flight method. The bees were suspended from a two-component aerodynamic balance at different, near optimum body angle of attack and were allowed to choose their own speed: their body mass and body weight was determined before and after a flight; their speed, lift, wingbeat frequency and total flight time were measured throughout a flight. These values were used to determine thrust, resultant aerodynamic force (magnitude and tilting angle), Reynolds number, total flight distance and total flight impulse. Flights in which lift was body weight were mostly obtained. Bees, flown to complete exhausion, were refed with 5, 10, 15 or 20 l of a 1.28-mol·l-1 glucose solution (energy content w=18.5, 37.0, 55.5 or 74.0 J) and again flown to complete exhaustion at an ambient temperature of 25±1.5°C by a flight of known duration such that the calculation of absolute and relative metabolic power was possible. Mean body mass after exhaustion was 76.49±3.52 mg. During long term flights of 7.47–31.30 min similar changes in flight velocity, lift, thrust, aerodynamic force, wingbeat frequency and tilting angle took place, independent of the volume of feeding solution. After increasing rapidly within 15 s a more or less steady phase of 60–80% of total flight time, showing only a slight decrease, was followed by a steeper, more irregular decrease, finally reaching 0 within 20–30 s. In steady phases lift was nearly equal to resultant aerodynamic force; tilting angle was 79.8±4.0°, thrust to lift radio did not vary, thrust was 18.0±7.4% of lift, lift was somewhat higher/equal/lower than body mass in 61.3%, 16.1%, 22.6% of all totally analysable flights (n=31). The following parameters were varied as functions of volume of feeding solution (5–20 l in steps of 5 l) and energy content. (18.5–74.0 J in steps of 18.5 J): total flight time, velocity, total flight distance, mean lift, thrust, mean resultant aerodynamic force, tilting angle, total flight impulse, wingbeat frequency, metabolic power and metabolic power related to body mass, the latter related to empty, full and mean (=100 mg) body mass. The following positive correlations were found: L=1.069·10-9 f 2.538; R=1.629·10-9 f 2.464; P m=7.079·10-8 f 2.456; P m=0.008v+0.008; P m=18.996L+0.022; P m=19.782R+0.021; P m=82.143T+0.028; P m=1.245·bm f 1.424 ; P mrel e=6.471·bm f 1.040 ; =83.248+0.385. Summary Cellobiose oxidase from Phanerochaete chrysosporium was used for continuous monitoring of cellulase action on microcrystalline cellulose (Avicel). Two protocols are described, the parameter monitored being either the decline in electrode potential as ferricyanide is reduced or consumption of dioxygen. Most experiments used a commercial cellulase preparation from Trichoderma reesei and ferricyanide as acceptor. Within 1 min of an addition of cellulase, ferricyanide reduction reached a steady rate. This was converted into a rate of production of substrate for celobiose oxidase, in mol·min–1. Experiments were conducted either with a constant concentration of cellulase and increasing Avicel, or with constant Avicel and increasing cellulase. Kinetic analysis of the experiments with constant cellulase indicated a K mof 4.8 ± 1.0 (g cellulose)·1–1, which was close to the value predicted from binding studies. The specific activity of the cellulase was measured as 375±25 mol·(g cellulase)–1·min–1 in experiments with a high cellulose concentration, but was less than half this value when the cellulose was saturated with cellulase. The effects of light intensity, oxygen concentration, and pH on the rates of photosynthesis and net excretion by metalimnetic phytoplankton populations of Little Crooked Lake, Indiana, were studied. Photosynthetic rates increased from 1.42 to 3.14 mg C·mg–1 chlorophylla·hour–1 within a range of light intensities from 65 to 150E·m–2·sec–1, whereas net excretion remained constant at 0.05 mg C·mg–1 chlorophylla·hour–1. Bacteria assimilated approximately 50% of the carbon released by the phytoplankton under these conditions. Excreted carbon (organic compounds either assimilated by bacteria or dissolved in the lake water) was produced by phytoplankton at rates of 0.02–0.15 mg C·mg–1 chlorophylla·hour–1. These rates were 6%–13% of the photosynthetic rates of the phytoplankton. Both total excretion of carbon and bacterial assimilation of excreted carbon increased at high light intensities whereas net excretion remained fairly constant. Elevated oxygen concentrations in samples incubated at 150E· m–2·sec–1 decreased rates of both photosynthesis and net excretion. The photosynthetic rate increased from 3.0 to 5.0 mg C·mg–1 chlorophylla· hour–1 as the pH was raised from 7.5 to 8.8. Net excretion within this range decreased slightly. Calculation of total primary production using a numerical model showed that whereas 225.8 g C·m–2 was photosynthetically fixed between 12 May and 24 August 1982, a maximum of about 9.3 g C·m–2 was released extracellularly.  相似文献

Four new Co(II) coordination complexes, [Co(o-phta)(pz)2]n1, [Co(PTA)2(Imh)2]·(HPTA)·H2O 2, {[Co(pdc)2(H2O)]·(ppz)·2H2O}n3, [K2Co2(ox)(btec)(CH3OH)2]n4, (H2phta = o-phthalic acid, pz = pyrazole, HPTA = p-toluic acid, ppz = piperazine, Imh = imidazole, H2pdc = pyridine-2,5-dicarboxylic acid, H2(ox) = oxalic acid, H4btec = 1,2,4,5-benzenetetracarboxylic acid), were hydrothermally synthesized and characterized by X-ray single crystal diffraction, IR, UV–Vis absorption spectrum, TG analysis and elemental analysis. The surface photovoltage properties of the four Co(II) complexes were investigated by the surface photovoltage spectroscopy (SPS). The structural analyses indicate that complexes 1 and 3 are 1D coordination polymers and complex 2 is a mononuclear molecular complex. Complexes 1, 2 and 3 are connected into 2D supramolecules by hydrogen bonds, respectively. Complex 4 is a coordination polymer with 3D structure, exhibiting a 4-nodal(4,5,6,12)-connected topology with a Schläfli symbol of (410)2(424·632·810)(45·6)2(49·65·8). The results of SPS show the four complexes exhibit obvious photovoltaic responses in 300–800 nm, which indicates they all possess photo-electric conversion properties. By the comparative analysis of the SPS, it is found that structure of the complex, species of ligand and coordination micro-environment of the Co(II) ion affect the SPS. The relationships between SPS and UV–Vis absorption spectra are discussed.  相似文献

The life history characteristics, population dynamics and production of Pontoporeia hoyi in Lake George, New York, were studied from May 1981 through October 1982. P. hoyi, in terms of both density and standing crop, is the most prevalent member of the deep water macrobenthos of Lake George. It reproduces in the winter, with young being released in the late winter-early spring. At the southernmost study site, young released in the spring grew to 6–7 mm in length and bred during their first winter. At the remaining sites, P. hoyi required two years to complete its life cycle. This difference in life history characteristics can be related to food availability and temperature differences. The open waters of the south end of Lake George are not only more productive but are also more closely associated with the littoral zone, providing a wealth of bacteria-rich detritus for benthic deposit feeders. The greater food availability in the south basin of Lake George is reflected in significantly larger brood sizes and smaller size at maturity for P. hoyi populations from the south end of the lake.The southernmost study site has significantly greater P. hoyi density and standing crop than all other sites. The cohort of the year dominated density and standing crop at the southern site while the cohort of the previous year dominated standing crop at the other sites. Peak abundance ranged from 600 · m–2 at the north site to 2 900 · m–2 at the south site. Cohort production ranged from 2g · m–2 at the north site to 15g · m–2 at the south site.  相似文献

Evidence of smolting was studied in Danish hatchery-reared brown trout Salmo trutta L. Twenty-four hour seawater (SW) challenge tests (28‰, 10°C) at regular intervals showed that maximal hypo-osmoregulatory ability developed within a 3–4-week period in March and April. The improved ability to regulate plasma osmolality, muscle water content and plasma total [Mg] developed asynchronously, indicating that developmental changes in the gill, the gastrointestinal system and the kidney may not necessarily concur during smolting. Gill Na+, K+-ATPase activity peaked in April at the time of optimal hypo-osmoregulatory ability. Na+, K+-ATPase a -subunit mRNA level in gills was unchanged from January until April, but decreased in May in parallel with a decrease in the activity of the enzyme. In the middle region of the intestine, Na+, K+-ATPase activity increased in February and remained high until April. In the posterior region of the intestine, the activity was stable from January until April after which it decreased. In vitro fluid transport capacitity, Jv, in the middle intestine fluctuated throughout the spring. In the posterior intestine, Jv was low until late March, when it increased fivefold until early May. Drinking rate in fish transferred to SW for 24 h surged during spring. Na+, K+-ATPase activity in the pyloric caeca was elevated from March until May, and increased in response to SW transfer in June, suggesting a hypo-osmoregulatory function of the pyloric caeca. Plasma GH levels surged in FW trout during spring, concurring with the increase in gill Na+, K+-ATPase activity and SW tolerance, but peaked in May when gill Na+, K+-ATPase activity and SW tolerance were regressing. GH levels were generally low in SW-challenged fish, and there was no consistent effect of 24-h SW exposure on GH levels. In wild anadromous trout, gill Na+, K+-ATPase activity varied seasonally as in hatchery-reared fish, but peaked at higher levels suggesting a more intense smolting in fish living in their natural environment.  相似文献

This study examined the restoration of the digestive capacity of Atlantic cod (Gadus morhua Linnaeus) following a long period of food deprivation. Fifty cod (48 cm, 1 kg) were food-deprived for 68 days and then fed in excess with capelin (Mallotus villosus Müller) on alternate days. Ten fish were sampled after 0, 2, 6, 14 and 28 days and the mass of the pyloric caeca, intestine and carcass determined. Two metabolic enzymes (cytochrome c oxidase and citrate synthase) were assayed in white muscle, pyloric caeca and intestine, and trypsin activity was measured in the pyloric caeca. A delay of 14 days was required before body mass started to increase markedly, whereas most of the increase in mass of both the pyloric caeca and intestine relative to fish length occurred earlier in the experiment. By day 14, the activities of trypsin and citrate synthase in the pyloric caeca as well as citrate synthase in the intestine had reached maxima. The growth of the digestive tissues and restoration of their metabolic capacities thus occur early upon refeeding and are likely required for recovery growth to take place. The phenotypic flexibility of the cod digestive system is therefore remarkable: increases in trypsin activity and size of pyloric caeca resulted in a combined 29-fold increase in digestive capacity of the fish during the refeeding period. Our study suggests that Atlantic cod are able to cope with marked fluctuations in food availability in their environment by making a rapid adjustment of their digestive capacity as soon as food availability increases.  相似文献

Relatively little is known about salinity acclimation in the primitive groups of fishes. To test whether physiological preparative changes occur and to investigate the mechanisms of salinity acclimation, anadromous green sturgeon, Acipenser medirostris (Chondrostei) of three different ages (100, 170, and 533 dph) were acclimated for 7 weeks to three different salinities (<3, 10, and 33 ppt). Gill, kidney, pyloric caeca, and spiral intestine tissues were assayed for Na+, K+-ATPase activity; and gills were analyzed for mitochondria-rich cell (MRC) size, abundance, localization and Na+, K+-ATPase content. Kidneys were analyzed for Na+, K+-ATPase localization and the gastro-intestinal tract (GIT) was assessed for changes in ion and base content. Na+, K+-ATPase activities increased in the gills and decreased in the kidneys with increasing salinity. Gill MRCs increased in size and decreased in relative abundance with fish size/age. Gill MRC Na+, K+-ATPase content (e.g., ion-pumping capacity) was proportional to MRC size, indicating greater abilities to regulate ions with size/age. Developmental/ontogenetic changes were seen in the rapid increases in gill MRC size and lamellar length between 100 and 170 dph. Na+, K+-ATPase activities increased fourfold in the pyloric caeca in 33 ppt, presumably due to increased salt and water absorption as indicated by GIT fluids, solids, and ion concentrations. In contrast to teleosts, a greater proportion of base (HCO3 and 2CO3 2−) was found in intestinal precipitates than fluids. Green sturgeon osmo- and ionoregulate with similar mechanisms to more-derived teleosts, indicating the importance of these mechanisms during the evolution of fishes, although salinity acclimation may be more dependent on body size.  相似文献
