The susceptibility of Botrytis cinerea to copper sulphate in liquid media increased when the volume, and therefore the depth, of the medium in culture bottles exceeded certain values; when the volume was 40 ml. the maximum concentration allowing growth was 300 p.p.m.
By growing mycelium in media containing progressively higher concentrations of copper sulphate a strain was produced which grew at a concentration of 750 p.p.m.
In high concentrations of copper sulphate growth always started at the edge of the liquid, and inocula grew only if they were placed in this position.
In germination tests spores from the resistant strain were more resistant to copper sulphate than were spores of the parent strain.
The resistance of mycelium, and to a lesser extent of spores, was retained after growth of the resistant strain for six months in fungicide-free media.
Spore and mycelial inocula grew in much higher concentrations of copper sulphate when nutrient media were solidified with agar.
The strain resistant in liquid media was no more resistant than the parent strain on agar media.
The resistance of the fungus was not increased after growth for long periods on agar containing high concentrations of copper sulphate. The resistance of the strain resistant in liquid media was not lost after growth on agar media for 3 months.
Attempts to produce strains more resistant than the parent to mercuric chloride were unsuccessful.
The results obtained with phenyl-mercuric acetate were essentially similar to those obtained with copper sulphate, but relatively much more resistant strains were produced.  相似文献

Six lines of the bacterium Escherichia coli were propagated for 2,000 generations in a temporally varying environment. The imposed environmental regime consisted of alternating days at 32°C and 42°C, with rapid transitions between them. These derived lines are competitively superior to their ancestor in this variable temperature regime. We also measured changes in the fitness of these lines, relative to their common ancestor, in both the constant (32°C and 42°C) and transition (from 32°C to 42°C and from 42°C to 32°C) components of this temporally varying environment, to determine whether the bacteria had adapted to the particular constant temperatures or the transitions between them, or both. The experimentally evolved lines had significantly improved fitness in each of the constant environmental components (32°C and 42°C). However, the experimental lines had not improved in making the sudden temperature transitions that were a potentially important aspect of the temporally variable environment. In fact, fitness in making at least one of the transitions (between 32°C and 42°C) unexpectedly decreased. This reduced adaptation to the abrupt transitions between these temperatures is probably a pleiotropic effect of mutations that were responsible for the increased fitness at the component temperatures. Among the six experimental lines, significant heterogeneity occurred in their adaptation to the constant and transition components of the variable environment.  相似文献

Electroencephalography is a valuable aid in diagnosis and prognosis of diseases affecting the brain. It has definite limitations.Abnormal electroencephalograms are either paroxysmal or non-paroxysmal. Both of these changes may be generalized or focal.Epilepsy is primarily associated with paroxysmal activity. Electroencephalography aids in the diagnosis and in determining the severity, course and response to treatment.Non-paroxysmal activity, although less specific in pattern, indicates either generalized or focal pathologic change or some alteration of the normal structure of the brain. Non-paroxysmal activity may give supportive evidence referable to conditions, systemic or intracranial, which disturb the state of consciousness.  相似文献

Photoperiodic responses of collections of Xanthium strumarium L., grown from seed obtained in nature at various localities in North America between latitudes 20° and 45.5° N, were examined. The critical night length was found to vary noticeably with latitude of origin, from about 7.5 hr in the northernmost strains to above 10 hr in the southern strains. These differences are considered to represent genetic adaptation of the reproductive system to environmental variables as a result of natural selection. Several strains showed a quantitative short day response rather than the more usual qualitative response. Strains from Hawaii exhibited a surprisingly tardy and erratic short-day response with a critical night length of about 11 hr, which may be an adaptation to a tropical climate.  相似文献

The concentrations of thiram preventing germination of spores of Botrytis cinerea in drops of a 1% solution of sucrose, and on the surface of a sucrose-nitrate agar have been determined. Thiram had much less effect on germination in the agar medium, even when a purified agar was used. There was no growth on sucrose-nitrate agar if the concentration of thiram exceeded 31 p.p.m. Attempts to obtain strains able to grow at higher concentrations were unsuccessful.
Similar results were obtained with ziram, nabam and zineb.
Ferbam also was more effective in preventing spore germination in spore drops than on agar media; this effect was obtained with ordinary and with purified agar.
On a sucrose-nitrate agar generally there was no growth if the concentration of ferbam exceeded 125 p.p.m., but in one of forty-eight plates containing 250 p.p.m. ferbam, five slowly growing colonies were produced, and from these colonies arose mycelium which grew and sporulated rapidly on 500 p.p.m. ferbam agar. Agar disk inocula were transferred from these cultures to agar containing higher concentrations of ferbam and in this way, and by repeating the process, a strain was obtained which grew slowly but continuously, and sporulated on agar containing 5000 p.p.m. ferbam. However, the poor solubility of this fungicide made it difficult to assess quantitatively the degree of adaptation.
A proportion of the spores from this strain germinated in drops containing about twice the concentration of ferbam which prevented germination of parent spores.
The resistance of the mycelium of the resistant strain was not lost after repeated subculture on fungicide-free agar. The resistant strain was as susceptible as the parent strain to thiram, ziram, nabam and zineb.
Attempts to obtain strains of Venturia inaequalis resistant to thiram, ferbam, ziram and zineb were unsuccessful.  相似文献

Genetic correlations between traits can constrain responses to natural selection. To what extent such correlations limit adaptation depends on patterns of directional selection. I derive the expected rate of adaptation (or evolvability) under randomly changing selection gradients. When directional selection gradients have an arbitrary covariance matrix, the average rate of adaptation depends on genetic correlations between traits, contrary to the isotropic case investigated in previous studies. Adaptation may be faster on average with more genetic correlation between traits, if these traits are selected to change jointly more often than the average pair of traits. However, natural selection maximizes the long‐term fitness of a population, not necessarily its rate of adaptation. I therefore derive the average lag load caused by deviations of the mean phenotype from an optimum, under several forms of environmental changes typically experienced by natural populations, both stochastic and deterministic. Simple formulas are produced for how the G matrix affects long‐term fitness in these contexts, and I discuss how their parameters can be estimated empirically.  相似文献

Following an environmental change, the course of a population's adaptive evolution may be influenced by environmental factors, such as the degree of marginality of the new environment relative to the organism's potential range, and by genetic factors, including constraints that may have arisen during its past history. Experimental populations of bacteria were used to address these issues in the context of evolutionary adaptation to the thermal environment. Six replicate lines of Escherichia coli (20°C group), founded from a common ancestor, were propagated for 2000 generations at 20°C, a novel temperature that is very near the lower thermal limit at which it can maintain a stable population size in a daily serial transfer (100-fold dilution) regime. Four additional groups (32/20, 37/20, 42/20, and 32–42/20°C groups) of six lines, each with 2000 generation selection histories at different temperatures (32, 37, 42, and daily alternation of 32 and 42°C), were moved to the same 20°C environment and propagated in parallel to ascertain whether selection histories influence the adaptive response in this novel environment. Adaptation was measured by improvement in fitness relative to the common ancestor in direct competition experiments conducted at 20°C. All five groups showed improvement in relative fitness in this environment; the mean fitness of the 20°C group after 2000 generations increased by about 8%. Selection history had no discernible effect on the rate or final magnitude of the fitness responses of the four groups with different histories after 2000 generations. The correlated fitness responses of the 20°C group were measured across the entire thermal niche. There were significant tradeoffs in fitness at higher temperatures; for example, at 40°C the average fitness of the 20°C group was reduced by almost 20% relative to the common ancestor. We also observed a downward shift of 1–2°C in both the upper and lower thermal niche limits for the 20°C selected group. These observations are contrasted with previous observations of a markedly greater rate of adaptation to growth near the upper thermal limit (42°C) and a lack of trade-off in fitness at lower temperatures for lines adapted to that high temperature. The evolutionary implications of this asymmetry are discussed.  相似文献

We investigate how different rates of environmental change affect adaptive outcomes and dynamics by selecting Chlamydomonas populations for over 200 generations in environments where the rate of change varies. We find that slower rates of environmental change result in end populations that grow faster and pay a lower cost of adaptation than populations that adapt to a sudden change of the same magnitude. We detected partial selective sweeps in adapting populations by monitoring changes in marker frequency in each population. Although populations adapting to a sudden environmental change showed evidence of mutations of large effect segregating early on, populations adapting to slow rates of change showed patterns that were consistent with mutations of relatively small effect occurring at less predictable times. This work suggests that rates of environmental change may fundamentally alter adaptive dynamics and outcomes of adaptation by changing the size and timing of fitness increases. We suggest that using mutations of smaller effect during adaptation may result in lower levels of pleiotropy and historical constraints, which could in turn result in higher fitness by the end of the experiment.  相似文献

在海拔4500m处,对26名18~43岁男性移居人进行了用增压车增压辅助对高原适应的研究。在增压30kPa时,心肺储备能力增强。除压速度>0.44m/s,会产生明显适应不良反应;除压速度<0.33m/s,不发生明显不适,并使增压过程中的心率减慢效应持续到除压后数小时。夜间睡眠增压,能消除睡眠中的潮式呼吸,改善若干高原适应不良症状。在33d内经18次断续增压处理的4名对象未损害正常的习服能力。结论:在4500m 高度用合理的增压,可辅助人体对高原的适应。  相似文献

Populations evolve in response to the external environment, whether abiotic (e.g., climate) or biotic (e.g., other conspecifics). We investigated how adaptation to biotic, heritable environments differs from adaptation to abiotic, nonheritable environments. We found that, for the same selection coefficients, the coadaptive process between genes and heritable environments is much faster than genetic adaptation to an abiotic nonheritable environment. The increased rate of adaptation results from the positive association generated by reciprocal selection between the heritable environment and the genes responding to it. These associations result in a runaway process of adaptive coevolution, even when the genes creating the heritable environment and genes responding to the heritable environment are unlinked. Although tightening the degree of linkage accelerates the coadaptive process, the acceleration caused by a comparable amount of inbreeding is greater, because inbreeding has a cumulative effect on reducing functional recombination over generations. Our results suggest that that adaptation to local abiotic environmental variation may result in the rapid diversification of populations and subsequent reproductive isolation not directly but rather via its effects on heritable environments and the genes responding to them.  相似文献

Rising temperatures associated with global warming present a challenge to the fate of many aquatic organisms. Although rapid evolutionary response to temperature-mediated selection may allow local persistence of populations under global warming, and therefore is a key aspect of evolutionary biology, solid proof of its occurrence is rare. In this study, we tested for genetic adaptation to an increase in temperature in the water flea Daphnia magna , a keystone species in freshwater systems, by performing a thermal selection experiment under laboratory conditions followed by the quantification of microevolutionary responses to temperature for both life-history traits as well as for intraspecific competitive strength. After three months of selection, we found a microevolutionary response to temperature in performance, but only in one of two culling regimes, highlighting the importance of population dynamics in driving microevolutionary change within populations. Furthermore, there was an evolutionary increase in thermal plasticity in performance. The results of the competition experiment were in agreement with predictions based on performance as quantified in the life table experiment and illustrate that microevolution within a short time frame has the ability to influence the outcome of intraspecific competition.  相似文献

Understanding the effects of sex and migration on adaptation to novel environments remains a key problem in evolutionary biology. Using a single‐cell alga Chlamydomonas reinhardtii, we investigated how sex and migration affected rates of evolutionary rescue in a sink environment, and subsequent changes in fitness following evolutionary rescue. We show that sex and migration affect both the rate of evolutionary rescue and subsequent adaptation. However, their combined effects change as the populations adapt to a sink habitat. Both sex and migration independently increased rates of evolutionary rescue, but the effect of sex on subsequent fitness improvements, following initial rescue, changed with migration, as sex was beneficial in the absence of migration but constraining adaptation when combined with migration. These results suggest that sex and migration are beneficial during the initial stages of adaptation, but can become detrimental as the population adapts to its environment.  相似文献

1. It has been shown that a fairly constant fraction of the total number of bacteria in a fresh-water culture of E. coli can reproduce on direct transfer to a saline medium with a definite NaCl concentration, as judged from the viable count determinations in such a medium. 2. The absolute value of this fraction depends on a number of factors other than the salt content of the test medium, such as the hydrogen ion and yeast autolysate concentrations, aeration, and the physiological condition of the bacteria. 3. A method for testing the degree and rate of adaptation of the bacteria to saline environment, depending on the analysis of changes in the value of the salt-viable fraction, was developed. 4. Maximum adaptability to saline environments was found during the early stationary phase of NaCl-free cultures. Low adaptability accompanied the logarithmic phase and the senescence of the cultures. 5. The limits of variation could be extended by treatment of non-dividing cells with gradually increasing concentrations of salt or by subjecting them to a single intermediate NaCl concentration. This acclimatization was independent of reproduction. The number of bacteria becoming capable of reproducing in a hitherto unfavorable environment increased with the period of exposure to intermediate salt concentrations until a maximum value was reached. 6. This maximum value was shown to depend on the salinity of the test medium, the age of the bacterial culture, and the method of preliminary treatment. "Optimal acclimatization" could be effected by subjecting the organisms to a single fairly low intermediate NaCl concentration. 7. The rate of the individual acclimatization process was shown to be greater at higher than at lower temperatures. 8. Acclimatized bacteria rapidly lost their increased ability to reproduce in saline media upon return to a salt-free environment, although no reproduction of the cells could be detected. This was interpreted as an indication that the processes involved are readily reversible. 9. Studies on the reproduction of E. coli in strongly saline broth indicated that only those cells originally acclimatized to the salt concentration of the medium could divide. All cells produced in such a medium could continue to reproduce. The propagation in the altered medium was not accompanied by any further acclimatization throughout five subcultures. 10. Both the division rate and the maximum crop of cultures in saline broth were considerably lower than of those in a fresh-water medium. No change in either occurred throughout five successive subcultures. The morphology of the organisms was also altered by the presence of salt. 11. The division rate, maximum crop, morphology, and adaptive power returned immediately to normal on re-transfer of bacteria grown in an NaCl-containing medium to "salt-free" broth. 12. The entire adaptive response of the bacteria to a considerable increase in the salinity of the environment could thus be separated into two components: an acclimatization, independent of reproduction, and a selection of those cells with the widest range of potentialities.  相似文献

Many animal species face periods of chronic nutritional stress during which the individuals must continue to develop, grow, and/or reproduce despite low quantity or quality of food. Here, we use experimental evolution to study adaptation to such chronic nutritional stress in six replicate Drosophila melanogaster populations selected for the ability to survive and develop within a limited time on a very poor larval food. In unselected control populations, this poor food resulted in 20% lower egg‐to‐adult viability, 70% longer egg‐to‐adult development, and 50% lower adult body weight (compared to the standard food on which the flies were normally maintained). The evolutionary changes associated with adaptation to the poor food were assayed by comparing the selected and control lines in a common environment for different traits after 29–64 generations of selection. The selected populations evolved improved egg‐to‐adult viability and faster development on poor food. Even though the adult dry weight of selected flies when raised on the poor food was lower than that of controls, their average larval growth rate was higher. No differences in proportional pupal lipid content were observed. When raised on the standard food, the selected flies showed the same egg‐to‐adult viability and the same resistance to larval heat and cold shock as the controls and a slightly shorter developmental time. However, despite only 4% shorter development time, the adults of selected populations raised on the standard food were 13% smaller and showed 20% lower early‐life fecundity than the controls, with no differences in life span. The selected flies also turned out less tolerant to adult malnutrition. Thus, fruit flies have the genetic potential to adapt to poor larval food, with no detectable loss of larval performance on the standard food. However, adaptation to larval nutritional stress is associated with trade‐offs with adult fitness components, including adult tolerance to nutritional stress.  相似文献
