Abstract: The N-methyl-d -aspartate (NMDA) receptor possesses two distinct amino acid recognition sites, one for glutamate and one for glycine, which appear to be allosterically linked. Using rat cortex/hippocampus P2 membranes we have investigated the effect of glutamate recognition site ligands on [3H]glycine (agonist) and (±)4-trans-2-car-boxy-5,7-dichloro-4-[3H]phenylaminocarbonylamino-1,2,3,4-tetrahydroquinoline ([3H]l -689,560; antagonist) binding to the glycine site and the effect of glycine recognition site ligands on l -[3H]glutamate (agonist), dl -3-(2-carboxypiperazin-4-yl)-[3H]propyl-1 -phosphonate ([3H]-CPP; “C-7” antagonist), and cis-4-phosphonomethyl-2-[3H]piperidine carboxylate ([3H]CGS-19755; “C-5” antagonist) binding to the glutamate site. “C-7” glutamate site antagonists partially inhibited [3H]l -689,560 binding but had no effect on [3H]glycine binding, whereas “C-5” antagonists partially inhibited the binding of both radioligands. Glycine, d -serine, and d -cycloserine partially inhibited [3H]CGS-19755 binding but had little effect on l -[3H]-glutamate or [3H]CPP binding, whereas the partial agonists (+)-3-amino-1-hydroxypyrrolid-2-one [(+)-HA-966], 3R-(+)cis-4-methyl-HA-966 (l -687,414), and 1-amino-1-carboxycyclobutane all enhanced [3H]CPP binding but had no effect on [3H]CGS-19755 binding, and (+)-HA-966 and l -687,414 inhibited l -[3H]glutamate binding. The association and dissociation rates of [3H]l -689,560 binding were decreased by CPP and d -2-amino-5-phosphonopentanoic acid (“C-5”). Saturation analysis of [3H]l -689,560 binding carried out at equilibrium showed that CPP had little effect on the affinity or number of [3H]l -689,560 binding sites. These results indicate that complex interactions occur between the glutamate and glycine recognition sites on the NMDA receptor. In addition, mechanisms other than allosterism may underlie some effects, and the possibility of a steric interaction between CPP and [3H]l -689,560 is discussed.  相似文献

Abstract— A manual cladistic analysis, subsequently expanded with a PAUP computer analysis, was performed on 21 genera of the monophyletic taxon Pandaloidea. Morphological data were obtained from the literature for 146 of the 152 known species-group taxa and from specimens belonging to 11 genera and 15 species—those of Pantomus parvulus extending the known range from the North Western Atlantic to Uruguay. The taxon Physetocaridoidca was synonymized with Pandaloidea, and the genus Pandalopsis with “Pandalus”. I have rejected reversal hypotheses indicated by the computer for four transformation series and chosen a final cladogram of slightly different topology which is six steps longer than the shortest tree. The cladogram for 20 terminal taxa is based on 108 apomorphic characters, resolved into 155 steps (72 synapomorphies and 83 homoplasies and reversals). ABSTRACT

The objective of the present study was to assess the effect of combining CBT-I with wearing blue-light blocking glasses 90 min prior to bedtime on subjective and objective sleep parameters and daily symptoms (anxiety, depression, hyperarousal). Thirty patients (mean age 48.1 ± 16.13 years, range 21–71, 15 men/15 women) completed a CBT-I group therapy program, with groups randomly assigned to either “active” (blue-light filtering glasses) condition or “placebo” (glasses without filtering properties) condition. Patients were continually monitored by wristwatch actigraphy, kept their sleep diaries and completed a standard questionnaire battery at admission and after the end of the program. Statistical analyses showed a greater reduction of BAI score in “active” (4.33 ± 4.58) versus “placebo” (?0.92 ± 3.68) groups of patients [F = 6.389, p = .019, Cohen’s d = 1.26] and significant prolongation of subjective total sleep time in “active” (?36.88 ± 48.68 min.) versus “placebo” (7.04 ± 47.50 min.) [F = 8.56, p < .01, d = 0.91] group. When pre- and post-treatment results were compared in both groups separately, using paired-samples t-tests, significant differences were observed also in the active group for BDI-II score (t = 3.66, p = .003, Cohen’s d = 0.95) and HAS score (t = 2.90, p = .012, Cohen’s d = 0.75). No significant differences were found in the placebo group. In active group, there was also a significant reduction of subjective sleep latency (t = 2.65, p = .021, d = 0.73) and an increase of subjective total sleep time (t = ?2.73, p = .018, d = ?0.76) without change in objective sleep duration which was significantly shortened in the placebo group. We provide further evidence that blocking short-wavelength light in the evening hours may be beneficial for patients suffering from insomnia.  相似文献

The free ecdysteroid titre determined by radioimmunoassay in adult female Boophilus microplus showed a peak just prior to full engorgement and detachment of the ticks and decreased subsequently to a very low value. In contrast, the titre of polar ecdysteroid conjugates was very low. Ecdysone was the major ecdysteroid at peak titre and was accompanied by much lower levels of 20-hydroxyecdysone. In newly detached ticks, injected [3H]ecdysone was metabolized primarily (80%) into much less polar compounds, which could be resolved into at least three groups by reversed-phase h.p.l.c. These [3H] “apolar” metabolites were transferred to the newly laid eggs, where they accounted for the vast preponderance of ecdysteroids, the level of free hormone being low. Hydrolysis of the three groups of compounds with an esterase preparation from porcine liver yielding [3H]ecdysone, together with the release of [3H] ecdysteroid and fatty acids upon alkaline saponification of the compounds, suggests that they are of a fatty acyl ester nature. The chemical transformation of these “esters” into the corresponding acetonide derivatives indicates that the 2- and 3-hydroxyls of ecdysone remain unsubstituted in these compounds. Several tick tissues, including Malpighian tubules, ovaries, gut, and fat body, metabolized [3H]ecdysone in vitro forming the “apolar esters” as major products. The maternal ecdysteroid “esters” may function as storage forms of hormone (presumably hormonally inactive), which could be hydrolysed enzymically during embryogenesis releasing free ecdysteroids. Such enzymic hydrolysis of [3H]ecdysone “esters” by homogenates from developing eggs of B. microplus has been demonstrated.  相似文献

Although environmental DNA surveys improve our understanding of biodiversity, interpretation of unidentified lineages is limited by the absence of associated morphological traits and living cultures. Unidentified lineages of marine stramenopiles are called “MAST clades”. Twenty‐five MAST clades have been recognized: MAST‐1 through MAST‐25; seven of these have been subsequently discarded because the sequences representing those clades were found to either (1) be chimeric or (2) affiliate within previously described taxonomic groups. Eighteen MAST clades remain without a cellular identity. Moreover, the discarded “MAST‐13” has been used in different studies to refer to two different environmental sequence clades. After establishing four cultures representing two different species of heterotrophic stramenopiles and then characterizing their morphology and molecular phylogenetic positions, we determined that the two different species represented the two different MAST‐13 clades: (1) a lorica‐bearing Bicosoeca kenaiensis and (2) a microaerophilic flagellate previously named “Cafeteria marsupialis”. Both species were previously described with only light microscopy; no cultures, ultrastructural data or DNA sequences were available from these species prior to this study. The molecular phylogenetic position of three different “C. marsupialis” isolates was not closely related to the type species of Cafeteria; therefore, we established a new genus for these isolates, Cantina gen. nov.  相似文献

The effects of varying concentrations of monovalent cation chlorides on the fluorescence of nine proteins were studied. These are discussed in terms of “direct” or “indirect” interactions with the aromatic amino acid residues. Cs+ is the only cation that quenches fluorescence of proteins due to “direct” interaction with aromatic amino acid residues. Quenching is due to collisional processes. An agreement with the Stern-Volmer relationship is shown and the values of [(KQ)eff] and [(fa)eff] are calculated. These values confirm that the fraction of fluorescence accessible to Cs+ belongs to the “exposed” fluorophors. The mechanism of quenching by Cs+ is due to the heavy-atom effect because phosphorescence enhancement is also seen at the same time. The chlorides of Na+, K+, Rb+, NH4+, and Li+ do not have a similar effect on the fluorescence of all proteins. For a given protein a gradation of the same effect (i.e., quenching or dequenching) is seen. Interactions with factors that “inderectly” affect fluorescence of any protein are involved and the structural features of the protein are responsible for such “indirect” effects. The results indicate that neutral salts can act in more than one manner. The changes in fluorescence are indicative of electrostatic and lyotropic effects of ions. Only electrostatic interactions which occur in the vicinity of tryptophan in proteins are reflected. Li+ shows strong interactions with proteins. In 4 M LiCl, BSA, papain, and trypsin show fluorescence changes that are indicative of changes in protein structure.  相似文献

While Roma are both Europe’s largest minority, there is no “homeland” state that claims to guard their interest. The lack of “an external national homeland” [Brubaker, R. 1996. Nationalism Reframed: Nationhood and the National Question in the New Europe. Cambridge: Cambridge University Press] to watchdog and safeguard their rights has a particular effect on how Roma engage as political actors and subjects. International donors/foundations have assumed the role of “external homeland”. This article explores the effect that donors’ funding priorities have on Romani advocacy – specifically Romani journalism. Drawing upon multi-sited fieldwork in five countries, extensive document analysis, and interviews, Idemonstrate that the change in Romani media content over the past two decades reflects the shift in funders’ priorities, particularly in relation to the European project. “The Roma” become a tool for donors and European institutions to build a “European” identity while Romani-led advocacy becomes increasingly marganilized.  相似文献

Several species of African grasses brought to Brazil as cattle forage have spread widely, outcompeting native herb species. The open forms of Brazilian savanna (“campo cerrado” and “campo sujo”) are the most affected by such invasions, because their structure is open, permitting enough sunlight into the lower strata. The invasion of alien forage grasses occurs in almost every cerrado nature reserve. This study was carried out in the “Cerrado de Hmas Biological Reserve”, Pirassununga, São Paulo State, Brazil, with the following objectives: (a) to compare the abundance of native and alien grass species; (b) to verify the importance of such alien grasses in the community; (c) to identity distribution patterns for the alien grass species in a gradient from the edge (highly disturbed) to the center (less disturbed) of the reserve; and (d) to explore the distribution of native and alien grasses in the search for possible competitive interactions. Using the “point method,” a total of 260 points was sampled and 52 species were recorded. The four most frequent species (FA = absolute frequency) were two native (Echinolaena inflexa [Poir.J Chase [FA = 38.85%] and Diandrostachya chrysotrix [Nees] Jacues Felix [FA = 15.38%]) and two alien African species (Melinis minutiflora Beauv. [FA = 33.08%] and Brachiaria decumbens Stapf [FA = 13.85%]). M. minutiflora and E. inflexa had higher values of absolute vigor (67.69 and 59.62%, respectively), relative vigor (28.16 and 24.80%, respectively), and cover (100.77 and 98.47, respectively), indicating higher biomasses and densities and their dominance in the community. B. decumbens presented the highest number of contacts per point, showing the highest stratification. To detect possible edge–center distribution gradients, correspondence analysis was done, initially using all the recorded species and subsequently only the four more frequent grasses, with similar results: (a) the alien grasses, especially M. minutiflora, did not show a distinct distribution gradient from edge to center, but occurred over the whole reserve; (b) no distinct ecotonal band around the reserve (edge–belt) was detected, the whole reserve seeming to be “ecotonal”; and (c) E. inflexa and M. minutiflora showed similar phytosociological patterns, and spatial distribution; association between these two species was statistically significant.  相似文献

An original set of experimental and modeling tools is used to quantify the yield of each of the physical processes leading to photocurrent generation in organic bulk heterojunction solar cells, enabling evaluation of materials and processing condition beyond the trivial comparison of device performances. Transient absorption spectroscopy, “the” technique to monitor all intermediate states over the entire relevant timescale, is combined with time‐delayed collection field experiments, transfer matrix simulations, spectral deconvolution, and parametrization of the charge carrier recombination by a two‐pool model, allowing quantification of densities of excitons and charges and extrapolation of their kinetics to device‐relevant conditions. Photon absorption, charge transfer, charge separation, and charge extraction are all quantified for two recently developed wide‐bandgap donor polymers: poly(4,8‐bis((2‐ethylhexyl)oxy)benzo[1,2‐b:4,5‐b′]dithiophene‐3,4‐difluorothiophene) (PBDT[2F]T) and its nonfluorinated counterpart poly(4,8‐bis((2‐ethylhexyl)oxy)benzo[1,2‐b:4,5‐b′]dithiophene‐3,4‐thiophene) (PBDT[2H]T) combined with PC71BM in bulk heterojunctions. The product of these yields is shown to agree well with the devices' external quantum efficiency. This methodology elucidates in the specific case studied here the origin of improved photocurrents obtained when using PBDT[2F]T instead of PBDT[2H]T as well as upon using solvent additives. Furthermore, a higher charge transfer (CT)‐state energy is shown to lead to significantly lower energy losses (resulting in higher VOC) during charge generation compared to P3HT:PCBM.  相似文献

Molecular and “adaptive” evolution, cladistic and phylogeny. Supplementations to a working hypothesis The methods of molecular biology and morpholo for the taxonomical assessment have given in several cases very different results. The following examples are discussed: Ailuropoda melanoleuca as an ursoid member of the Carnivora (Ursidae or Ailuropodidae?), Homo saiens and his position to the African apes (Pan and Gorilla) (Pongidae or Hominidae?), Theroipithecus gelada and its affinities to the baboons (Papionini or Theropithecini ?) and Oreopithecus bambolii from the late Miocene of Italy and its taxonomical position within the catarrhine primates. For the three extant species not only the “two level evolution” (molecular and morphological-adaptative evolution) is evident, but also the working hypothesis, that the “adaptative” evolution is more rapidly as the molecular evolution, as the author is suggesting since 1969. The results of molecular methods (immunoloy, amino acid sequences, electrophoresis, DNA-DNA hybridization) are more important for the phylogeny than for the taxonomy. For the cladistic method it is clear, that a cladogram is not a phylogenetic tree, as suggested Peter Ax (1984).  相似文献

The envelope membrane glycoprotein gC of HSV-1 was purified from Triton X-100 extracts of virus-infected BHK-21 or HEp-2 cells by a single step immuno-affinity column using monoclonal anti-gC antibody. The analysis of the purified [3H]G1cN labeled glycoprotein gC (by gel filtration on Bio-Gel P4) before and after digestion with endo-β-N-acetylglucosaminidase (endo D) indicated that gC contains Asn-linked “complex type” oligosaccharides. No “high mannose” type oligosaccharides were detected. Fractionation of radio-labeled glycopeptides of gC on a column of concanavalin A-sepharose suggested that glycopeptides have “diantennary” and “triantennary” and/or “tetra antennary” structures. Tunicamycin inhibited the incorporation of [14C]GalN or [3H]GlcN into gC in HSV-1 infected BHK-21 or HEp-2 cells. Gel filtration analysis of [3H]GlcN labeled gC following β-elimination reaction failed to indicate O-glycosidically linked oligosaccharides.  相似文献

Circulating schistosome antigens (CSA) and circulating immune complexes (CIC) were investigated during the course of Schistosoma mansoni infection in mice. The radioimmunoprecipitation-polyethylene glycol (PEG) assay (RIPEGA) with [125I]anti-S. mansoni antibodies or [125I] anti-antigen “4” antibodies detected, respectively, total CSA and antigen “4” in serum and in 3% polyethylene glycol-precipitated CIC from infected mice. Complement fixation test and [125I] C1q-binding test revealed, respectively, an anticomplementary activity and the presence of C1q-binding CIC. All these substances appeared in infected mice at approximately the same period, i.e., between the 40th- and the 55th- day postinfection. No correlation was observed between the detection of anticomplementary active substances and C1q-binding CIC. In contrast, a close relationship was noticed between CSA and complement-activating material during the course of the infection. This suggests that substances with anticomplementary activity in serum from infected mice could be one or various CSA. A close correlation was also observed between C1q-binding CIC and free or “complexed” antigen “4.” This observation supports well the possibility that antigen “4” is one of the major complexed circulating antigen present in schistosomiasis. The immunoglobulins G1, G2a, M, and A were also characterized in 3% PEG-precipitated CIC from infected mice during the period in which we detected C1q-binding CIC. The roles played by specific S. mansoni CIC in either schistosomal nephropathy or protective mechanisms to a challenge infection in mice are discussed.  相似文献

In the Second Manifesto of Surrealism, issued by André Breton in 1929, surrealism was described as “a total recuperation of our psychic strength by a means of none other than vertiginous descent into ourselves, systematic illumination of the hidden places and progressive darkening of the other places, a perpetual walk in the forbidden zone”. Surrealism sought to represent the unconscious and forbidden zones of the psyche, of the body, of the noumenal world within, which offered access, for surrealists, to energies and intuitions repressed by “civilized” modes of perception. For Jean Rouch, the significance of surrealism, of automatic writing and ciné‐transe, rested in the potential escape they offered from the formal constraints of conventional film and of conventional perception and observation. In his celebration of ciné‐transe, and of the technological apparatus that makes it possible, it is possible to detect his desire for a freeing‐up of the constraints of consciousness—a desire to “write with the body”, to dream, to tap the unexplored power of the unconscious in its overturning of “reality”, of system, and of convention. As a phaneroscopic “wide‐angle lens”, surreality aimed to document the scientifically unexplainable, the immense experiential overload of ritual possession. It attempted to make visible, in the movement between observation and participation and across disjunctive points of view; the crossings‐over into the unconscious world by which possessed Songhay dancers gained access to powers of phaneroscopic perception. By adopting filmic techniques which follow the surrealist practice of creating “verbal and visual collage”, in which randomly‐generated images, emerging out of a trance‐like state (of “automatic writing” or "ciné‐transe"), are juxtaposed in indeterminate and polyphonic relations with each other in an attempt to disturb or destroy patterns of perception which are confining, rationalistic, linear, or restricted to conscious phenomena, Rouch believed he could create powerful representations of the unknowable. This paper relates the phaneroscopic practice of ethnographic surrealism to psychoanalytic models of the unconscious. In a discussion of Rouch's interpretation of the Hauka spirit cult in his film Les Maîtres Fous, the paper argues that the neo‐Freudian paradigm which allowed him to depict the Sohghay's weekend Hauka rites as a parodic reversion to “savagery” (which both reversed the hierarchy of colonizer/colonized and enabled participants to experience a therapeutic release from the traumas of colonization) has been challenged by Lacanian and post‐Lacanian “re‐readings” of Freud that call into question the extent to which the unconscious can be equated with a pre‐linguistic state characterized by disjunctive “primitive” and “instinctive” energies. The surrealist longing for a rupture of the symbolic order of Western rationalism and a return to the “imaginary order” of the unconscious is confounded in the Lacanian conception of the unconscious as a zone inhabited by the “discourse of the Other”. However, the work of Gillès Deleuze and Félix Guanari [1977] provides a means of conceptualizing the unconscious in terms that avoid simplistic binary logic (phenomenon/noumenon; signifier/signified; subject/object; conscious/unconscious; civilization/savagery). The unconscious is not the “unrepresentable” Other of consciousness; it is a schizophrenic phaneron, a signifying “machine”, a transgressive producer of “group fantasy”. Rejecting both Freud's Oedipal model (the unconscious as primal imagery or “ghostly signifieds"), and the Lacanian notion of the unconscious (as a play of “empty signifiers"). Deleuze and Guattari argue that the unconscious cannot be accounted for in terms of the individual child and its entry into language any more than it can be conceived of as the domain of the primitive. On the contrary, the unconscious is constituted as “group fantasy”, as collective public memory which need not be reduced to elemental (Oedipal) signifiers: “all delirium possesses a world‐historical, political and racial content”. The schizophrenic embodies the public nature of unconscious meaning, since schizophrenia is primarily a communication disorder in which an individual never sees himself in terms of a linguistically‐generated “selfhood”, and fails to adopt the “false” identity which is offered to him in the language of the Other. Schizophrenia is characterized by a refusal to treat some meanings as superior to others, to remain within the bounds of a stable identity, or to distinguish between material (noumenal) things and actions and their (phenomenal) meanings. The schizophrenic unconscious treats all experience as signs, registering language in the same way as the body registers physical stimuli. Thus “meanings in the unconscious are simply meanings as workings of the body” [Harland 1987:174–175]. The schizophrenic as a model for the unconscious holds several implications for those interested in representing the experiential power of public rituals, for the public meanings in circulation during such rituals are material and noumenal, and are registered on the bodies of the dancers as they transgress boundaries and pass beyond consciousness. Rouch's surreality attempted to inscribe this unconscious production of public meaning as it was manifested in the movements of the ritual and in the movements of the camera‐body.  相似文献

Water-soluble polypeptides of L -valyl and L -isoleucyl residues flanked with DL -lysyl blocks [poly(DL Lys · HCl)x–poly(L Val)y–poly(DL Lys · HCl)x, poly(DL Lys · HCl)x–poly-(L Ile)y–poly(DL Lys · HCl)x] and homopoly(L -threonine) were prepared. The β conformation of these polymers in water, as well as in aqueous methanol, was confirmed by infrared spectroscopy and circular dichroism studies. The optical properties of these valyl and isoleucyl polypeptides were quite different from those of previously reported synthetic homopolypeptides in the β structure. Their differences could be explained by the presence of a “single extended β chain” without either intra- or interchain association.  相似文献

The Tropical West African-Congolese spiny Cleome previously referred to the neotropical “C. spinosa” is the indigenous, distinct, here newly described C. afrospina, most closely related to the southern South American C. trachycarpa of the otherwise endemic neotropical Section Tarenaya. Early collections (from 1816) and widespread distribution in wet forests and riversides also indicate it to be indigenous and not adventive. Related also is the widespread Brazilian-Argentinian C. hassleriana Chod. (“C. spinosa” auct. non Jacq.; “C. pungens” auct. non Willd.?), the commonly cultivated “Spider Flower,” once recently collected in the Congo (cultivated?). As the only indigenous spiny Old World Cleome, C. afrospina is a remarkable neotropical element in the African flora. Historical explanations for such disjunctions (continental drift, land bridges) are rejected. Avian long-distance dispersal is indicated by semi-aquatic successional habitats of high ecological receptivity, generally annual habit, other highly disjunct vicariads, and self-compatibility within the group. Similar disjunctions occur in the distribution patterns of many aquatic birds (e.g., whistling ducks—Dendrocygna) and of various tropical plants of ecologically receptive “open” habitats.  相似文献

In 2004, the Italian Parliament passed a controversial law on medically assisted reproduction (Law 40/2004). The Law obliged clinicians to create a maximum of three embryos during one in vitro fertilization (IVF) cycle and transfer them simultaneously into the patient’s uterus. With this “three embryo” standard, the Parliament sought to secure the realization of rights of IVF embryos. Drawing on the concepts of boundary-work [Gieryn, T. F. 1983. “Boundary-Work and the Demarcation of Science from Non-Science: Strains and Interests in Professional Ideologies of Scientists.” American Sociological Review 48 (6): 781–795] and bioconstitutionalism [Jasanoff, S., ed. 2011. Reframing Rights: Bioconstitutionalism in the Genetic Age. Cambridge, MA: MIT Press.], this article explores the role that the constitutional obligations of the Italian State towards its citizens, including IVF embryos as its new “citizen subjects,” played in how it envisaged and demarcated the professional boundaries of medical expertise. It argues that the latter depended upon how it balanced its commitments to protect the rights of IVF embryos and those of adult citizens. As such, the demarcation of the jurisdictional boundaries of medical expertise, and the definition of constitutional rights, formed two sides of the same governing project.  相似文献
