The fluorometry of urinary total estrogens ofBrown et al. (1968) has been modified and successfully applied to the routine measurement of urinary estrogens in Japanese monkeys. Serial analysis during the menstrual cycle of the Japanese monkey has shown typical changes in urinary total estrogens with a distinct midcycle peak which preceded the day of ovulation by 0–2 days. The technique is valuable in an accurate estimation of the time of ovulation for successful programmed breeding of the Japanese monkey.  相似文献

The TUNEL reaction was used to study induced apoptosis in the tumor cells of urinary bladder cancer (UBC) patients. It has been shown that an increase in the values of induced apoptosis correlates well with the indicators showing a tumor’s corresponding response to the application of neoadjuvant chemotherapy, using the gemcitabine-cisplatin regimen. It has been concluded that assessing the level of induced apoptosis in UBC tumor cells using the TUNEL assay enables making a prognosis for the efficacy of chemotherapy at the single-cell level in patients with this type of cancer.  相似文献

This paper describes an original dot-enzyme-linked immunosorbent assay (ELISA) for predicting ovulation in women, based on the detection of the pre-ovulatory estrogen peak in urine. A monoclonal anti-estrogen antibody is used which recognizes not only free estrogens but also some of their urinary metabolites (17-glucuro- and sulfo-conjugates) allowing a direct assay on early morning urines. Antigen is immobilized as a spot on a nitrocellulose membrane which is immersed in urine in the presence of this antibody. A peroxidase-labeled second antibody allows the detection of the first antibody bound to the membrane. Antigen and anti-estrogen antibody concentrations are chosen to obtain a maximal enzymatic coloration of spots corresponding to basal urinary estrogen levels and no coloration corresponding to the pre-ovulatory surge. Six menstrual cycles were studied, comparing dot-ELISA results with patterns of: (1) urinary estrogens measured by RIA either directly or after hydrolysis and extraction, and (2) basal body temperatures. The validity of the pre-ovulatory signal obtained and the requirements for an adaptation of this methodology to a reliable home kit are discussed.  相似文献

A highly specific chemical procedure for the quantitative determination of 2-hydroxyestrone in the urine of pregnant women is described. The assay consists of the following steps: 1) Hot acid hydrolysis of 20 ml of urine, 2) purification of 2-hydroxyestrone by “reducing chromatography” on paper and silica gel column, 3) conversion of 2-hydroxyestrone to the phenazine compound, 4) purification of the phenazine derivative by alumina column chromatography, and 5) spectroscopic quantitation of the phenazine. For internal yield correction [4-14C]2-hydroxyestrone is added after urine hydrolysis. High specificity of the method is especially guaranteed by the specific transformation of 2-hydroxyestrone to a stable phenazine derivative and by rigorous chromatographic purification of the estrogen as well as of the phenazine. The method can be used for the determination of amounts of less than 1 μg of 2-hydroxyestrone/20 ml of urine. From the data obtained the coefficient of variation is calculated to be ±3.7%. The urinary excretion of 2-hydroxyestrone in late pregnancy was found to vary within a wide range of 30–800 μg of 2-hydroxyestrone/24 hours.It seems possible to extend this method to the determination of other 2-substituted estrogens present in urine.  相似文献

The use of acetylcholinesterase inhibitors to decrease the breakdown of the neurotransmitter acetylcholine has been the main symptomatic therapy for mild to moderate Alzheimer’s patients, though the etiology of Alzheimer’s disease remains unclear and seems to involve multiple factors. Further evidence has indicated that some of these acetylcholinesterase inhibitors also have non-cholinergic functions on the pathogenesis of Alzheimer’s disease including the formation and deposition of β-amyloid. Huperzine A, a potent and reversible inhibitor of acetylcholinesterase that was initially isolated from a Chinese herb, has been found to improve cognitive deficits in a broad range of animal models and has been used for Alzheimer’s disease treatment in China. The novel neuroprotective effects of huperzine A might yield beneficial effects in Alzheimer’s disease therapy and provide a potential template for the design of new selective and powerful anti-Alzheimer’s drugs. The present paper gives an overview on the neuroprotective effects of huperzine A beyond its acetylcholinesterase inhibition. These effects include regulating β-amyloid precursor protein metabolism, protecting against β-amyloid-mediated oxidative stress and apoptosis. The structure–function relationship of huperzine A is also discussed.  相似文献

Although conductance-based neural models provide a realistic depiction of neuronal activity, their complexity often limits effective implementation and analysis. Neuronal model reduction methods provide a means to reduce model complexity while retaining the original model’s realism and relevance. Such methods, however, typically include ad hoc components that require that the modeler already be intimately familiar with the dynamics of the original model. We present an automated, algorithmic method for reducing conductance-based neuron models using the method of equivalent potentials (Kelper et al., Biol Cybern 66(5):381–387, 1992) Our results demonstrate that this algorithm is able to reduce the complexity of the original model with minimal performance loss, and requires minimal prior knowledge of the model’s dynamics. Furthermore, by utilizing a cost function based on the contribution of each state variable to the total conductance of the model, the performance of the algorithm can be significantly improved.  相似文献

Most of these speculations have wandered significantly far from the original formulation; yet, they may have been useful for a critical examination of methodological issues. As molecular genetics and its complexities evolve amid a backdrop of technological innovation, new “uncertainties” may have emerged. Summary Increasing concerns over the effects of environmental estrogens on wildlife and humans have highlighted the need for screening systems to assess potentially estrogenic effects of test compounds. As a result, in vitro screening methods such as cell proliferation assays using the estrogen-responsive human breast cancer cell line, MCF-7, have been developed. The present study describes an alternative in vitro approach for the assessment of such xenoestrogens, based on estrogenic rescue of MCF-7 cells from antiestrogen-induced cytotoxicity. This method measures the ability of various estrogenic compounds to compete with a known estrogen-receptor-mediated antihormonal drug, 4-hydroxytamoxifen, using the 1-[4,5-dimethylthiazol-2-yl]-3,5-diphenylformazan (MTT) assay to assess mitochondrial activity. Because 4-hydroxytamoxifen treatment of cells results in a dramatic decrease in mitochondrial dehydrogenase activity which is directly related to their estrogen-receptor content, inhibition of this effect with estrogenic compounds represents an estrogen-receptor interaction, or estrogenic rescue. The estrogenic compounds tested include a weak xenoestrogen, bisphernol A (BPA), and two biological estrogens, 17α- and 17β-estradiol. Competitive inhibition of 4-hydroxytamoxifen-induced cytotoxicity by BPA was compared to that of the biological estrogens. The results indicate that the biological estrogens can successfully compete with the antiestrogen in a dose-dependent manner. In addition, the assay is sensitive enough to detect estrogenic rescue by even the very weak xenoestrogen, BPA, albeit at high BPA concentrations. This simple in vitro method could be used as an alternative or second-line screen for potential xenoestrogens.  相似文献

Apoptosis of human breast carcinoma cells (SKBR-3, MCF-7, and MDA-468) has been observed after treatment of these cells with anti-cancer drug cis-platin and glycosphingolipid biosynthesis inhibitor L- and D-PPMP, respectively. These drugs initiated apoptosis in a dose-dependent manner as measured by phenotypic morphological changes, by binding of a fluorescent phophatidyl serine-specific dye (PSS-380) onto the outer leaflet of the cell membranes, and by activation of caspases, −3, −8, and −9. It was observed that in two hours very little apoptotic process had started but predominant biochemical changes occurred after 6 h. DNA degradation started after 24 hours of drug treatment. However, very little is known about the stability of the ‘`Replication Complexes’’ during the apoptotic process. DNA helicases are motor proteins that catalyze the melting of genomic DNA during its replication, repair, and recombination processes. Previously, DNA helicase-III was characterized as a component of the replication complexes isolated from embryonic chicken brains as well as breast and colon carcinoma cells. Helicase activities were measured by a novel method (ROME assay), and DNA polymerase-α activities were determined by regular chain extension of the nicked ACT-DNA, by determining values obtained from +/− aphidicolin-treated incubation mixtures. In all three breast carcinoma cell lines, a common trend was observed: a decrease of activities of DNA polymerase-α and Helicase III. A sharp decrease of activities of the glycolipid sialyltransferases: SAT-2 (CMP-NeuAc; GD3 α2-8 sialyltransferase) and SAT-4 (CMP-NeuAc: GM1a α2-3 sialyltransferase) was observed in the apoptotic carcinoma cells treated with L-PPMP compared with cis-platin.  相似文献

Daily sperm production in rats and monkeys were estimated by extended histometric method. Individual seminiferous tubules were separated, fixed, processed and embedded. Semi-thin sections were stained with toluidine blue and the different cell types quantitated with Chalkley’s eye piece. The daily sperm production estimated was compared with that obtained by the homogenization method. The results indicate that daily sperm production obtained by both the methods were comparable. The present histometric method has an added advantage of requiring very small amount of biopsy material to give precise daily sperm production estimations.  相似文献

Summary When comparison was made of directions given for three salt solutions commonly used in cell culture preparations and identified as Dulbecco’s, Earle’s, and Hanks’, variations in composition were found. Some significantly alter the suitability of the materials for the intended use. Other differences may have less effect. This brief review reveals a tendency among researchers to follow procedures obtained from colleagues for the preparation of laboratory reagents and media, to cite the original publication rather than their colleagues’ work as the source of the information, and to fail to compare the two for differences. Some failures in cell culture propagation may be attributed to similar instances with other published but incorrectly cited work. Tables are provided that facilitate comparison of the correct original formulations with variants from selected published sources. In this paper, a methodology for the determination of three naturally occurring estrogens (estradiol, estrone and estriol) in pregnant women's urine has been described. The procedure included immunoaffinity column (IAC) extraction of 4 mL of urine sample and subsequent analysis of the extraction by micellar electrokinetic chromatography (MEKC). A multi-target polyclonal antibody that has high affinity to three estrogens was produced. Then the IAC was developed by coupling polyclonal antibody to CNBr-activated Sepharose 4B. The IAC showed high affinity for these estrogens. Recoveries of three estrogens from human serum matrix were greater than 92% with R.S.D. less than 4.5%. The final elute of urine sample was diluted with running buffer and then quantitated with MEKC. The experimental results demonstrated that IAC was a useful technique for extraction and concentration of estrogens from biological samples. Three estrogens levels in six pregnant women's urine were measured by both the present method and enzyme-linked immunoadsorbent assay (ELISA). The results of this method have been found to correlate well with those of ELISA.  相似文献

Experiments show that deflections of microcantilever-DNA chip can be induced by many factors, such as grafting density, hybridization efficiency, concentration, length and sequence of DNA molecules, buffer salt concentration, time, and temperature variation. However, there are few theoretical works on microcantilever-DNA chips. The present paper is aimed to study the influence of counterion effects of single-stranded DNA (ssDNA) polyelectrolyte solution on the nanomechanical behaviors of microcantilever-based ssDNA chips during packing process. First, the effect of osmotic pressure induced by ingress of counterions into DNA brush structures is studied with Hagan’s model for a cylindrical polyelectrolyte brush system on the basis of Poisson-Boltzmann distribution hypothesis. Second, Zhang’s two-variable method for a laminated cantilever is used to formulate a four-layer energy model for ssDNA chips with weak interactions. Third, the influence of grafting density, ssDNA chain length, and salt concentration on packing deflection is investigated using the principle of minimum energy. The predictive tendency is qualitatively similar to those observed in some related ssDNA chip experiments. The difference between the four-layer model and the simplified two-layer model for ssDNA chips is also discussed.  相似文献

A radioimmunoassay method for urinary catechol estrogens is described. The specific nature of the antisera allows direct analyses of acid hydrolyzed urine. A LH-20 Sephadex column chromatography can be employed for individual determinations of 2-hydroxyestrone and 2-hydroxyestradiol. The excretion of catechol estrogens during menstrual cycles ranged from 14.48 to 50.15 μg per 24 hours, whereas, during the last trimester of pregnancies, the values ranged from 129.30 to 1758.20 μg per 24 hours.  相似文献

Atomic force microscopy is a common technique used to determine the elastic properties of living cells. It furnishes the relative Young’s modulus, which is typically determined for indentation depths within the range 300–500 nm. Here, we present the results of depth-sensing analysis of the mechanical properties of living fibroblasts measured under physiological conditions. Distributions of the Young’s moduli were obtained for all studied cells and for every cell. The results show that for small indentation depths, histograms of the relative values of the Young’s modulus described the regions rich in the network of actin filaments. For large indentation depths, the overall stiffness of a whole cell was obtained, which was accompanied by a decrease of the modulus value. In conclusion, the results enable us to describe the non-homogeneity of the cell cytoskeleton, particularly, its contribution linked to actin filaments located beneath the cell membrane. Preliminary results showing a potential application to improve the detection of cancerous cells, have been presented for melanoma cell lines.  相似文献

Summary Sembene’s technique allows for African models of reality that are not solely in opposition to our concept of civilization. We are not presented with Conrad’s single emissaries into the exotic heartland of Africa who then return to us with a message. For Sembene, large numbers of people are characterized in a revolutionary act and large numbers of people are reacting to colonial control not just within the grid of traditional thought that reinterprets new contacts in terms of a concept of “untouched” communalism, but as mirroring cleavages in society that are reflective of historical interaction and of African literature’s basis in oral tradition. What happens when you get something like a heinneman series (a London publishing concern which controls the literature that reaches a Western audience) is that authors like Sembene move to different mediums. Sembene now writes exclusively in Woloof.God’s Bits of Wood ends optimistically. The “Vatican” is the city’s living compound that is created by the whites, but it is the blacks who name it. The “Vatican” symbolizes the white’s inability to control the blacks. The final confrontation involves Isnard’s wife, Beatrice, who upon realizing that the new order born out of the union member’s solidarity will cause their recall to the metropole, culminates in the black’s destruction of the European’s sanctuary. The fallen wife sybolizes the resistance on the part of the Africans, to European attempts to deny their history “Tell them” she screamed at the terrified woman. “Tell them that you like Monsieur! Say it in the name of God, say it! [86]. I have attempted to present in my examination of one aspect of African literature the idea that when shared symbolic elements are highlighted over time, the units of analysis that have been utilized in the past (e.g., tribes; extrapolated to the concept of the modern nation state) must gain a wider frame for analysis. Class formation has not been seen as a basis of cleavage in Africa’s past. The perspective for Africa has been that of the closed social system; the only perceived basis for cleavage becomes ethnicity, interaction with external forces is that of cultural substitution and ultimately supplication to the steamroller of Imperialism. It is at this level, and at this level alone, that the analysis of Oral and Literate traditions has been seen as dichotomous. But if one perceives the openness in African systems of thought and chronicles the fluctuations of these systems over time as representative of their ability to generate alternative hypotheses, one can obtain a more sophisticated, more appropriate, dichotomy between Oral and Literate traditions, that can become the genesis of creating new cultural forms and new social structures. The purpose of the following research was to improve the original Celsior solution in order to obtain a higher degree of stability and effectiveness. The solution was modified by the addition of selected antioxidants such as vitamin C, cysteine, and fumaric acid in the following concentrations: 0.1, 0.3, and 0.5 mmol/l. The solution’s stability was estimated using an accelerated stability test based on changes in histidine concentrations in the solution using Pauly’s method for determining concentrations. Elevated temperatures, the factor accelerating substances’ decomposition reaction rate, were used in the tests. The research was conducted at four temperatures at intervals of 10°C: 60 ± 0.2°C, 70 ± 0.2°C, 80 ± 0.2°C, and 90 ± 0.2°C. It was stated that the studied substances’ decomposition occurred in accordance with the equation for first-order reactions. The function of the logarithmic concentration (log%C) over time was revealed to be rectilinear. This dependence was used to determine the kinetics of decomposition reaction rate parameters (the rate constant of decomposition k, activation energy E a, and frequency factor A). On the basis of these parameters, the stability of the modified solution was estimated at +5°C. The results obtained show that the proposed antioxidants have a significant effect on lengthening the Celsior solution’s stability. The best results were reached when combining two antioxidants: vitamin C and cysteine in 0.5 mmol/l concentrations. As a result, the Celsior solution’s stability was lengthened from 22 to 299 days, which is 13.5 times. Vitamin C at a concentration of 0.5 mmol/l increased the solution’s stability by 5.2 times (t 90 = 115 days), cysteine at a concentration of 0.5 mmol/l caused a 4.4 times stability increase (t 90 = 96 days), and fumaric acid at a concentration of 0.5 mmol/l extended the stability by 2.1 times (t 90 = 48 days) in relation to the original solution.  相似文献
