The sterol composition of Palaemon adspersus is studied. It is established that the nonsaponifiable fraction of Palaemon adspersus contains cholesterol, 7-dehydrocholesterol, desmosterol, substance "240" and inconsiderable amounts of other still nonidentified sterols.  相似文献

Summary Six sterols were extracted from lipids produced by Candida curvata yeast. Ergosterol (main component) and five minor compounds: ergosta-7, 24(28)-dien-3 -ol, ergosta-5, 7-dien-3 -ol, ergosta-5, 7, 9, (11), 22-tetraen-3 -ol, ergosta 7, 22-dien-3 -ol, ergost-7-en-3 -ol were identified by gas chromafography/mass spectrometry (GCMS) and quantitated by GLC.  相似文献

The sterols of 27 species of Porifera have been examined by GC-MS; this study includes 23 members of the class Demospongiae and four species of the hitherto uninvestigated class Calcarea. The trends observed earlier for the Demospongiae are supported by the results of the present survey. Sterols of the Calcarea have not proved diverse but, of those present, the majority are of the C27 type.  相似文献

The Demospongiae (Porifera) have yielded a wide range of novel and conventional sterols; a sample of 55 species screened and reported on have yielded 45 distinct sterol structures. The taxonomic relevance of the occurrence of particular sterols or overall sterol complement is evaluated and has proved to be informative in the case of the Verongida, Nepheliospongida, Axinellida and Hadromerida. A possible relationship between the occurrence of 26-methyl sterols and oviparous reproductive patterns within the Ceractinomorpha is noted. Sterol complement is observed to be qualitatively stable despite season and location of collection. High molecular weight sterols, present as major components, are probable membrane constituents. They afford an interesting parallel to the occurrence of diverse, high carbon fatty acids in sponge membranes.  相似文献

Six groups of nystatin resistant mutants of C. maltosa and of haploid and diploid Saccharomyces cerevisiae strains were obtained with the help of genetic and biochemical analysis. It has been shown that every group of the mutants was characterized by a specific level of resistance to nystatin. The dependence of the resistance level upon sterol content has been established. It has been shown that the more the structure of the sterol present differed from ergosterol the higher was the resistance level. The results obtained in vivo permit to make conclusions about the role of different functional groups of sterol molecule in the interaction with nystatin.  相似文献

The sterol content in Saccharomyces cerevisiae mutants defective in the synthesis of cyclic ergosterol precursors has been studied. It was found that strains with mutational blocks involving the stages of zymosterol side chain methylation at C24 and delta 8----delta 7 isomerization accumulated twice more sterols as compared to parent strains. Regulation of the ergosterol biosynthesis is discussed.  相似文献

Nystatin-resistant yeast Candida tropicalis was obtained after UV illumination and plating on nystatin-containing media. The mutant contained no ergosterol in the plasma membrane but bound nystatin to a degree similar to that of the wild strain (1.2 vs. 1.5 nmol per mg dry solid). Respiration of the mutant on glucose was reduced by 36% in the presence of 25 microM nystatin. This corresponded to a 25-43% decrease of the uptake of monosaccharides. Transport of amino acids was reduced by nystatin in the mutant by 44-86%, as compared with a 84-95% reduction in the wild strain. The intracellular ATP content was reduced by nystatin equally in the wild strain and in the mutant (by 43 and 47%). Nystatin appears to affect specifically membrane transport processes of nonelectrolytes while both the H+-extruding ATPase and the membrane potential are unaffected.  相似文献

The sterol and fatty acid composition of three Adriatic Sea sponges (Geodia cydonium and two unidentified Tedania sp.), collected at the same time and same place, was established. Twenty-four sterols and forty fatty acids were identified. The identical ecological conditions, including the diet, allowed us to apply the results obtained for taxonomical conclusions, based on the biodiversity of the investigated sponges. On the basis of the sterol composition they can be separated into two groups: Tedania and Geodia sponges. The sterol and fatty acid composition indicates that the two investigated Tedania samples might be different species or subspecies.  相似文献

Summary Non-saponifiable cell extracts of wild type and sterol mutants of N. crassa were analysed by means of gas-liquid chromatography. The wild-type contained ergosterol and episterol in a 10:1 ratio. None of the mutants was able to synthesize ergosterol. Three of the mutants carry single recessive gene mutations causing blocks in the terminal steps of ergosterol biosynthesis: erg-1 has an inactive 8 7 isomerase, erg-2 has an inactive 24(28) hydrogenase, and erg-4 has an inactive C-24 methyl transferase. Some of the mutants accumulated novel sterols as a result of their enzyme defects. The genes erg-1 and erg-2 were mapped close to inl on the right arm of chromosome V.  相似文献

Sterol composition of four clinical isolates of Aspergillus fumigatus resistant to itraconazole was determined by gas chromatography--mass spectrometry and compared with that of four susceptible strains. For all strains, the major sterol was ergosterol. Sterol compositions were qualitatively and quantitatively similar for the resistant and susceptible strains. These results suggest that itraconazole resistance is not related, for the strains studied, to alterations in the ergosterol synthesis pathway.  相似文献

Sterols in silkworm larvae were analyzed. Cholesterol was predominantly detected in all tissues examined. Dietary phytosterols and desmosterol, a putative biosynthetic intermediate from phytosterols to cholesterol, were also detected, indicating that imperfect intestinal conversion from phytosterols to cholesterol influences the sterol composition in larval tissues.  相似文献

Several drugs that interact with membrane sterols or inhibit their syntheses are effective in clearing a number of fungal infections. The AIDS-associated lung infection caused by Pneumocystis jirovecii is not cleared by many of these therapies. Pneumocystis normally synthesizes distinct C28 and C29 24-alkylsterols, but ergosterol, the major fungal sterol, is not among them. Two distinct sterol compositional phenotypes were previously observed in P. jirovecii. One was characterized by delta7 C28 and C29 24-alkylsterols with only low proportions of higher molecular mass components. In contrast, the other type was dominated by high C31 and C32 24-alkylsterols, especially pneumocysterol. In the present study, 28 molecular species were elucidated by nuclear magnetic resonance analysis of a human lung specimen containing P. jirovecii representing the latter sterol profile phenotype. Fifteen of the 28 had the methyl group at C-14 of the sterol nucleus and these represented 96% of the total sterol mass in the specimen (excluding cholesterol). These results strongly suggest that sterol 14alpha-demethylase was blocked in these organisms. Twenty-four of the 28 were 24-alkylsterols, indicating that methylation of the C-24 position of the sterol side chain by S-adenosyl-L-methionine:sterol C-24 methyl transferase was fully functional.  相似文献
