Total lipid and total fatty acid composition was studied in twelve marine microalgae species, belonging to the Eustigmatophyceae, Chlorophyceae, Haptophyceae and Prasinophyceae, cultured under comparable conditions.Species of the same class showed a particular fatty acid composition, indicating specific fatty acid bioconversion modes. Twelve species of microalgae, isolated from north Australian marine, freshwater and hypersaline environments, were grown under controlled conditions of temperature, pH, photon flux density and salinity, and analysed for ash, total protein, water soluble carbohydrates, chlorophylla, total lipids, total fatty acids and fatty acid composition. Highest levels of the polyunsaturated fatty acid eicosapentaenoic acid [20:5(n-3)] were found in the marine diatoms.Nitzschia (frustulum) andN. closterium (23.1% and 15.2% of total fatty acids, respectively). None of the species studied had levels of docosahexaenoic acid [22:6(n-3)] greater than 1.1 % of total fatty acids. None of the chlorophyte species contained significant levels of either 20:5(n-3) or 22:6(n-3). The highest total fatty acid concentration of all species in the study was found in the freshwater chlorophyte speciesScenedesmus dimorphus (105 mg g–1 dry wt). The hypersaline speciesDunaliella salina had the highest total lipid content (28.1% dry wt), followed byN. closterium, N. (frustulum) andNavicula sp. (24.2–27.8% dry wt).Chlamydomonas sp. had the highest protein content (66.9% dry wt).N. The influence of salinity on the growth, gross chemical composition and fatty acid composition of three species of marine microalgae,Isochrysis sp.,Nannochloropsis oculata andNitzschia (frustulum), was investigated. There was no significant change in growth rate ofIsochrysis sp. andN. (frustulum) over the experimental range of salinity (10–35 ppt), whileN. oculata had a significantly slower growth rate only at 35 ppt. The ash content of all three species increased with increasing salinity. Two species,Isochrysis sp. andN. oculata, showed significant linear increases in total lipid content with increasing salinity over the range 10 to 35 ppt.N. (frustulum) showed significant linear decrease in total lipids, with the highest percentage at low salinity within the range 10–15 ppt. Variation in salinity had only a slight effect on the total protein, the soluble carbohydrate and chlorophylla content of all species. All species responded to change in salinity by modifying their cellular fatty acid compositions. Significant positive correlations were observed between increase in salinity and increase in the percentage ofcis-9-hexadecenoic acid [16:1 (n-7)] over the entire experimental range inN. (frustulum) and between 25–35 ppt inN. oculata. There were curved relationships between salinity and percentage of hexadecanoic acid [16:0] inN. oculata andN. (frustulum), with maxima within the range 25–30 ppt for both species. A curved relationship was found between salinity and percentage of eicosapentaenoic acid [20–5(n-3)], forN. (frustulum), with lowest percentages of the fatty acid within the range 25–30 ppt. There was no consistent pattern in the percentages of other major fatty acids as functions of salinity. The Northern Territory isolateN. (frustulum) was unusual in having a substantial increase in total fatty acids with decreasing salinity (85 mg g–1 dry wt at 10 ppt compared with 33 mg g–1 at 35 ppt). The optimum salinities for the production of maximum amount of lipids and the essential fatty acids 20:5(n-3) and/or 22:6(n-3) were as follows:25 ppt forIsochrysis sp. The total protein, carbohydrate, lipid and ash compositions, and fatty acid contents of two species of marine microalgae, the eustigmatophyte Nannochloropsis oculata (formerly ‘Chlorella sp., Japan’) and the chrysophyte Isochrysis sp. (Tahitian) used in tropical Australian mariculture, were studied. The microalgae were grown under a range of culture conditions (41 and 601 laboratory culture, 3001 bag culture, and 80001 outdoor culture) and four light regimes (100 to 107 μ E m−2 s−1, 240 to 390 μ E m−2 s−1, 340 to 620 μ E m−2 s−1, and 1100 to 1200 μE m−2 s−1 respectively) to determine the effect of light intensity on the chemical composition of large scale outdoor cultures. Laboratory and bag cultures were axenic and cultured in Walne medium while outdoor cultures were grown in a commercial medium designed for optimum nutrition in tropical outdoor aquaculture operations. Change in growth medium and photon flux density produced only small changes in the proximate biochemical composition of both algae. N. oculata and Isochrysis sp. both showed a trend towards slightly lower carbohydrate and higher chlorophyll a in shaded outdoor culture. Isochrysis sp. showed significant concentrations of the essential polyunsaturated fatty acid 22:6(n−3) (docosahexaenoic acid) from 5.3 to 10.3% of total fatty acid, and 20:5(n−3) (eicosapentaenoic acid) ranged from 0.6 to 4.1%. In contrast, N. oculata had high concentrations of 20:5(n−3) (17.8 to 39.9%) and only traces of 22:6(n−3). The fatty acid composition of Isochrysis sp. grown at high photon flux density (1100–1200 μE m−2 s−1) under outdoor culture showed a decrease in the percentage of several highly unsaturated fatty acids, including 20:5(n−3), and an increase in 22:6(n−3). N. oculata showed a similar decrease in the percentage of 20:5(n−3). High light intensity caused a decrease in the ratio of total C16 unsaturated fatty acids to saturated 16:0 in N. oculata, and a decrease in the ratio of total C18 unsaturated fatty acids to saturated 18:0 together with a decrease in the ratio of total unsaturated fatty acids to total saturated fatty acids in both microalgae.  相似文献

A variety of invertebrates are commonly fed to insectivorous animals by both zoos and hobbyists, but information as to the nutrient composition of most commercially raised species is limited. Adult house crickets, house cricket nymphs (Acheta domesticus), superworms (Zophobas morio larvae), giant mealworm larvae, mealworm larvae and adult mealworms (Tenebrio molitor), waxworm larvae (Galleria mellonella), and silkworm larvae (Bombyx mori) were analyzed for moisture, crude protein, crude fat, ash, acid detergent fiber (ADF), neutral detergent fiber (NDF), minerals, amino acids, fatty acids, and vitamins. Earthworms (Lumbricus terresstris) were analyzed for moisture, crude protein, crude fat, ash, ADF, NDF, minerals, amino acids, and vitamins A and D3. Proximate analyses were variable, with wide ranges found for moisture (57.9–83.6%), crude protein (9.3–23.7%), crude fat (1.6–24.9%), ADF (0.1–7.4%), NDF (0.0–11.5%), and ash (0.6–1.2%). Energy content ranged from a low of 674 kcal/kg for silkworms to 2,741 kcal/kg for waxworms.Using an amino acid scoring pattern for rats, the first limiting amino acid for all invertebrates tested was the total sulfur amino acid methionine+cystine. Deficiencies by nutrient (% of samples deficient vs. NRC requirements for rats on a dry matter (DM) basis) were as follows: calcium (100%), vitamin D3 (100%), vitamin A (89%), vitamin B12 (75%), thiamin (63%), vitamin E (50%), iodine (44%), manganese (22%), methionine‐cystine (22%), and sodium (11%). Deficiencies by invertebrate species (number of nutrients deficient vs. the NRC requirements for rats on a DM basis) were as follows: waxworms (9), superworms (8), giant mealworm larvae (7), adult mealworms (6), mealworm larvae (5), adult house crickets (4), house cricket nymphs (4), silkworms (4), and earthworms (4). These data provide a basis for determining nutrient intake of captive insectivores, and will aid in the development of gut‐loading diets to provide captive insectivorous animals with appropriate levels of necessary nutrients. Biomass and lipid productivity, lipid content, and quantitative and qualitative lipid composition are critical parameters in selecting microalgal species for commercial scale‐up production. This study compares lipid content and composition, and lipid and biomass productivity during logarithmic, late logarithmic, and stationary phase of Nannochloropsis sp., Isochrysis sp., Tetraselmis sp., and Rhodomonas sp. grown in L1‐, f/2‐, and K‐medium. Of the tested species, Tetraselmis sp. exhibited a lipid productivity of 3.9–4.8 g m?2 day?1 in any media type, with comparable lipid productivity by Nannochloropsis sp. and Isochrysis sp. when grown in L1‐medium. The dry biomass productivity of Tetraselmis sp. (33.1–45.0 g m?2 day?1) exceeded that of the other species by a factor 2–10. Of the organisms studied, Tetraselmis sp. had the best dry biomass and/or lipid production profile in large‐scale cultures. The present study provides a practical benchmark, which allows comparison of microalgal production systems with different footprints, as well as terrestrial systems. Biotechnol. Lipid classes and their fatty acids were studied in the major lipoprotein fractions from canine, in comparison with human, plasma. In dogs, high-density-lipoprotein (HDL), the main carrier of plasma phospholipid (PL), cholesterol ester (CE) and free cholesterol, was the most abundant lipoprotein, followed by low and very-low density lipoproteins (LDL and VLDL). Notably, LDL and VLDL contributed similarly to the total dog plasma triacylglycerol (TG). The PL composition was similar in all three lipoproteins, dominated by phosphatidylcholine (PC). Even though the content and composition of lipids within and among lipoproteins differed markedly between dog and man, the total amount of circulating lipid was similar. All canine lipoproteins were relatively richer than those from humans in long-chain (C20-C22) n-6 and n-3 polyunsaturated fatty acids (PUFA) but had comparable proportions of total saturated and monoenoic fatty acids, with 18:2n-6 being the main PUFA in both mammals. The fatty acid profile of canine and human lipoproteins differed because they had distinct proportions of their major lipids. There were more n-3 and n-6 long-chain PUFA in canine than in human plasma, because dogs had more HDL, their HDL had more PC and CE, and both these lipids were richer in such PUFA.  相似文献

The purpose of the present study was to compare the influence of adding no or 8% fat of varying sources (coconut oil, fish oil, rapeseed oil and sunflower oil) to diets for sows 1 week prior to farrowing and during lactation on the composition of fatty acids in plasma and tissues of the progeny while sucking and 3 weeks after weaning from the sow. A control diet without supplemental fat and four diets supplemented with 8% of coconut oil, rapeseed oil, fish oil or sunflower oil were provided to lactating sows (n = 15), and during the post-weaning period the same weaner diet was provided to all piglets (n = 15 litters), which were housed litterwise. The dietary ratio of n-6:n-3 fatty acids of the maternal diets largely influenced the progeny, as the ratio varying from 1.2 (fish oil) to 12.2 (sunflower oil) in the sow milk was reflected in plasma and adipose tissues of the sucking progeny. The liver showed similar variations according to dietary treatments, but a lower n-6:n-3 fatty acids ratio. From day 4 to later on during the suckling period, the concentration of C14:0, C16:0 and C18:1 in the liver of the piglets decreased, irrespective of the dietary treatments of sows. In plasma and liver, the total concentration of saturated fatty acids (SAFA), monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA) did not differ markedly in piglets sucking sows fed different dietary fatty acids, whereas the adipose tissue of piglets sucking sows fed sunflower oil and coconut oil showed the highest proportion of PUFA and SAFA, respectively. Weaning lowered the concentration of lipid-soluble extracts in plasma and the concentration of fatty acids in the liver of the piglets. Within the post-weaning period, dietary treatments of sows, rather than age of piglets, influenced the fatty acid composition of plasma and adipose tissue of the piglets, whereas the hepatic fatty acid profile was more affected by the age of the piglets during the post-weaning period. This study shows that the fatty acid profile of plasma and tissues of the progeny is highly dependent on the maternal dietary composition, and that the dietary impact persists for up to 3 weeks after the suckling period.  相似文献

Galaxias maculatus eggs and larvae obtained from broodfish captured either in an estuarine or a freshwater environment, as well as from cultured broodstock were analysed to compare their lipid and fatty acid profiles. Results showed a lower lipid content in embryos and larvae from estuarine populations than those from fresh water, denoting the influence of environmental conditions. The n-3:n-6 ratio was higher in eggs from estuarine and cultured populations, being in the range of marine fishes, whereas for eggs from freshwater fish was lower and typical of freshwater fishes. The polyunsaturated fatty acids (PUFA), particularly docosahexaenoic acid (22:6n-3) and eicosapentaenoic acid (20:5n-3), were higher in eggs and larvae of broodstock coming from culture or estuarine environments than in those from fresh water. Moreover, these fatty acids markedly increased after hatching in larvae coming from estuarine populations, suggesting the effect of the environment on fatty acid profiles to physiologically prepare the larvae to adapt to higher salinity conditions. Linoleic acid (18:2n-6) content was higher in fresh water fish and its reduction during embryo and larval development was accompanied by a significant increase of arachidonic acid (20:4n-6), which was not observed in embryos or larvae from broodstock fish from estuary or aquaculture origin. Atlantic herring larvae ( Clupea harengus ) were fed two enriched Artemia diets with different contents of (n-3) highly unsaturated fatty acids (HUFA), one containing low levels of 20: 5(n-3) and no 22: 6(n-3), the other containing substantial levels of both 20: 5(n-3) and 22: 6(n-3). After 30 days of culture, fatty acid compositions of lipid classes in the heads, bodies and eyes of the larvae were analysed. Fish fed Artemia with the low (n-3) HUFA diet lacking 22: 6(n-3) had lower amounts of total (n-3)HUFA and, in particular, of 22:6(n-3) in individual phospholipids and total neutral lipids of heads, bodies and eyes as compared to fish fed Artemia with high levels of (n-3)HUFA. The amount of 22: 6(n-3) in the fatty acids of phosphatidyl-ethanolamine of eyes was particularly susceptible to dietary depletion. The implications of these findings are discussed, particularly in relation to dietary requirements for 22: 6(n-3) during development of neural tissue in predatory fish iarvae.  相似文献

The fatty acid metabolism in fish is influenced by various factors, including fish species, water temperature, water environment and diet supply. The aim of present work is to investigate the fatty acid composition of yolk‐stage Siberian sturgeon larvae reared at three different temperatures. Fertilized Siberian sturgeon eggs were transferred to the Lodi Aquaculture Research Center of the University of Milan, divided in three aquaria, each containing three incubators and incubated at 16°C. After hatching the temperature was switched to 16, 19 and 22°C. Larvae sampling was performed at the end of yolk sac reabsorption. No feed was dispensed during the trial. Eggs and larvae were weighed and fatty acid profile was determined by GC‐FID analysis after lipid extraction by chloroform/methanol mixture and fatty acid transesterification by methanolic hydrogen chloride. The fertilized eggs had a weight of 23.27 mg and a lipid content of 2.67 mg/egg. At hatching, the weight was 12.2 (0.17 SD) mg and lipid content 1.9 (0.6 SD) mg/larva. At the end of the trial, larvae mean weight was 33.6 (3.6 SD), 34.7 (1.8 SD) and 36.9 (1.1 SD) mg, while lipid content was 2.0 (0.3 SD), 2.1 (0.3 SD) and 2.0 (0.2 SD) mg for larvae reared at 16, 19 and 22°C respectively, without statistically significant difference. Larvae subjected to the highest water temperature showed a faster yolk‐sac absorption. No differences were found across temperatures regarding survival rates and regarding ontogenic development. The fatty acid composition of larvae was affected by the temperature. Larvae reared at 16°C had the lowest amount of saturated fatty acids, mainly due to a lower palmitic acid content, that was offset by a higher level of linolenic and linoleic acid, if compared with larvae reared at 19°C and 22°C. The study suggests that at a lower temperature sturgeon spare unsaturated fatty acid consuming preferably saturated fatty acids, increasing our knowledge of the fatty acid metabolism in this species.  相似文献

Results are given for their gross chemical composition (water content, crude protein, total carbohydrates, lipids, nucleic acids etc.) and contents of macrominerals, trace elements, fatty acids, amino acids and neutral sugars. 【目的】阐明家蝇 Musca domestica 幼虫对食物中各种多不饱和脂肪酸的富集能力以及代谢转化情况,并探究各种多不饱和脂肪酸对家蝇幼虫生长的影响。【方法】在基础饲料中添加不同浓度(3%, 6%和12%)的多不饱和脂肪酸(亚油酸、α-亚麻酸、花生四烯酸和二十二碳六烯酸)饲养经过脱脂传代培养的家蝇幼虫;提取家蝇幼虫的总脂肪酸,利用气相色谱仪进行检测和分析;测定统计幼虫体重,以分析多不饱和脂肪酸对家蝇幼虫生长的影响。【结果】亚油酸、α-亚麻酸和花生四烯酸在家蝇幼虫体内均能被富集,且它们的富集程度随着食物中多不饱和脂肪酸的添加浓度的升高而增加,其中亚油酸、α-亚麻酸和花生四烯酸在幼虫体内富集的最高含量(占体内总脂肪酸的比例)分别为21.93%, 16.13%和9.68%,而二十二碳六烯酸不能在家蝇幼虫体内富集,提示家蝇幼虫食物中添加的各种多不饱和脂肪酸经过代谢后并没有在其体内产生新的脂肪酸,而食物中添加的二十二碳六烯酸在家蝇幼虫体内被分解代谢后消除。饲喂α-亚麻酸及花生四烯酸后家蝇幼虫体重增长较为明显,其中6%α-亚麻酸添加组的幼虫体重显著高于对照组(取食脱脂饲料)和3%和12%α-亚麻酸添加组,3%和6%花生四烯酸添加组的幼虫体重显著高于对照组和12%花生四烯酸添加组。【结论】家蝇幼虫体内能够从食物中富集部分多不饱和脂肪酸,多不饱和脂肪酸碳链越长其富集程度越低直至不能富集,富集的多不饱和脂肪酸对家蝇幼虫生长有不同程度的影响。  相似文献

以意大利蜜蜂Apis mellifera ligustica雌性幼虫为试验材料,用鲜王浆、葡萄糖、果糖和酵母抽提物分别配制不同日龄的蜂王幼虫食物和工蜂幼虫食物,采用室内人工饲养蜜蜂幼虫技术饲养12h以内的蜂王幼虫和工蜂幼虫,直到幼虫出现排便和吐丝行为,记录幼虫生长发育期间消耗的总食物量。结果表明:1只蜂王幼虫在生长发育期平均消耗196.8mg蜂王幼虫食物,而1只工蜂幼虫在生长发育期平均消耗139.4mg工蜂幼虫食物。  相似文献

Emerging aquatic insects, including mosquitoes, are known to transfer to terrestrial ecosystems specific essential biochemicals, such as polyunsaturated fatty acids (PUFA). We studied fatty acid (FA) composition and contents of dominant mosquito populations (Diptera: Culicidae), that is, Anopheles messeae, Ochlerotatus caspius, Oc. flavescens, Oc. euedes, Oc. subdiversus, Oc. cataphylla, and Aedes cinereus, inhabited a steppe wetland of a temperate climate zone to fill up the gap in their lipid knowledge. The polar lipid and triacylglycerol fractions of larvae and adults were compared. In most studied mosquito species, we first found and identified a number of short‐chain PUFA, for example, prominent 14:2n‐6 and 14:3n‐3, which were not earlier documented in living organisms. These PUFA, although occurred in low levels in adult mosquitoes, can be potentially used as markers of mosquito biomass in terrestrial food webs. We hypothesize that these acids might be synthesized (or retroconverted) by the mosquitoes. Using FA trophic markers accumulated in triacylglycerols, trophic relations of the mosquitoes were accessed. The larval diet comprised green algae, cryptophytes, and dinoflagellates and provided the mosquitoes with essential n‐3 PUFA, linolenic, and eicosapentaenoic acids. As a result, both larvae and adults of the studied mosquitoes had comparatively high content of the essential PUFA. Comparison of FA proportions in polar lipids versus storage lipids shown that during mosquito metamorphosis transfer of essential eicosapentaenoic and arachidonic acids from the reserve in storage lipids of larvae to functional polar lipids in adults occurred.  相似文献

The effect of temperature from 10 °C to 35 °C on the growth, total lipid content, and fatty acid composition of three species of tropical marine microalgae, Isochrysis sp., Nitzschia closterium, N. paleacea (formerly frustulum), and the Tahitian Isochrysis sp. (T.ISO), was investigated.Cultures of N. closterium, Isochrysis sp. and T.ISO grew very slowly at 35 °C, while N. closterium did not grow at temperatures higher than 30 °C or lower than 20 °C. N. paleacea was low-temperature tolerant, with cells growing slowly at 10 °C. N. paleacea produced the highest percentage of lipids at 10 °C, while the other species produced maximum amounts of lipid at 20 °C. None of the species maintained high levels of polyunsaturated fatty acids (PUFAs) at high growth temperature and there was a significant inverse relationship between the percentage of PUFAs and temperature for N. paleacea. A curved relationship was found between temperature and percentage of PUFA for N. closterium and tropical Isochrysis sp., with the maximum production of PUFA at 25 °C and 20 °C, respectively. The two Nitzschia species produced higher levels of the essential fatty acid eicosapentaenoic acid [20:5(n-3)] at lower growth temperatures, but the two Isochrysis species had little change in percentage of 20:5(n-3) with temperature. Recent studies suggest that the use of vegetable oils at expense of fish oil in aquaculture feeds might have potential negative effects on fish redox homeostasis and adiposity. Resveratrol (RESV) is a lipid-soluble phytoalexin present in fruits and vegetables with proven in vivo antioxidant function in animals. The present study aims to assess the potential use of RESV in Atlantic salmon feeds. To this end, post-smolt salmons with an initial BW of 148±3 g were fed four experimental diets for 15 weeks. A diet low in fish oil served as a control and was supplemented with 0, 0.5, 1.5 and 2.5 g/kg of RESV, respectively. The effect of the experimental diets on animal performance, tissue fatty acid composition, and the expression of genes encoding proteins involved in antioxidant signalling, lipid peroxidation, and metabolism were studied. Resveratrol significantly reduced feed intake and final BW of the salmon. Feeding RESV did not affect the sum of saturated and monounsaturated fatty acids or total lipids in the fillet. While the content of total polyunsaturated fatty acids was not affected, the percentages of some fatty acids in the liver and fillet were changed by RESV. Furthermore, in liver, the relative expression of glutathione peroxidase 4b, nuclear factor-like 2, and arachidonate 5-lipoxygenase remained unchanged across treatment groups. In conclusion, the negative impact of dietary RESV on FI and hence reduction of the BW discourages its inclusion in low fish oil diets for Atlantic salmon.  相似文献

To study the effect of feeding silages with different botanical composition, on rumen and lamb fat, 30 male lambs were assigned to five different silage groups for 11 weeks: botanically diverse silage (BDS); white clover silage (WCS); red clover silage (RCS), intensive English ryegrass silage (IRS) and crushed linseed and maize silage (MSL). Besides the silages, animals received organic wheat and barley and the MSL group additionally received bicarbonate (15 g/day). Silages were sampled when the bales were opened and analysed for fatty acid (FA) content and chemical composition. At slaughter, ruminal contents were sampled and 24 h after slaughter, longissimus muscle and subcutaneous (SC) fat were sampled. All samples were analysed for FA composition. The MSL group ingested the highest amount of FA (35.8 g/day v. 13.5, 19.4, 17.2 and 30.4 g/day for MSL v. BDS, WCS, RCS and IRS, respectively) and the sum of the major polyunsaturated FA, C18:2 n-6 and C18:3 n-3, was similar for groups BDS, WCS, RCS and MSL (61.3 g/100 g, 62.3 g/100 g, 62.3 g/100 g, 63.7 g/100 g of FA methylesters (FAME), respectively), while group IRS ingested higher proportions of these FA (74.5 g/100 g of FAME). Rumen data showed that animals fed BDS presented higher proportions of biohydrogenation intermediates, particularly C18:1 t11 and CLA c9t11, suggesting partial inhibition of rumen biohydrogenation. In the MSL group, the content of C18:3 n-3 in the rumen was highest, most probably due to reduced lipolysis and hence biohydrogenation through the combined effect of esterified C18:3 n-3 and seed protection. Additionally, C18:3 n-3 proportions were higher in rumen contents of RCS animals compared with WCS animals, which could be due to the activity of the polyphenol oxidase enzyme in the RC silages. Proportions of C18:3 n-3 were similar between treatments both for SC and intramuscular (IM) fat, whereas CLA c9t11 content was higher in the SC fat of BDS animals and lower in the IM fat of IRS animals compared with the other forage groups. No differences were found for C20:4 n-6, C20:5 n-3, C22:5 n-3 and C22:6 n-3 in the IM fat of the animals. Nevertheless, indices for desaturation and elongation activity in muscle of BDS animals suggest some stimulation of the first three steps of desaturation and elongation (Δ6-desaturase, elongase and Δ5-desaturase) of long-chain FA.  相似文献

1. While the balance of light and nutrients is known to influence the food quality of herbivores by altering algal phosphorus and nitrogen content, the combined effects of light and nutrients on fatty acid synthesis in freshwater periphyton are relatively unknown. In this study, we manipulated light and phosphorus concentration in large, flow‐through experimental streams to examine their effects on both elemental stoichiometry and fatty acid content in periphyton. 2. Two levels of phosphorus (4 and 80 μg L?1) and three of light (17, 40, 110 μmol photons m?2 s?1) were applied in a factorial design in two separate experiments. Diatoms dominated periphyton communities in both experiments, comprising >95% of algal biovolume. Periphyton growth in the streams was simultaneously affected by both resources, even at low rates of supply. 3. Periphyton C/P and C/N ratios increased with light augmentation and decreased with phosphorus enrichment, and consistent with the light : nutrient hypothesis (LNH). Light effects were strongest in streams with low phosphorus concentrations. 4. Periphyton fatty acids reflected the dominance of diatoms : palmitic (16 : 0), palmitoleic (16 : 1ω7) and eicosapentanoic (20 : 5ω3) were the principal saturated (SAFA), monounsaturated (MUFA) and polyunsaturated fatty acids (PUFA), respectively. Linoleic (18 : 2ω6) and linolenic (18 : 3ω3) acids, characteristic of chlorophytes and cyanophytes, were rare, comprising <2% of total fatty acids. 5. Periphyton fatty acid profiles were highly sensitive to light and phosphorus. The proportion of fatty acids comprised by SAFA and MUFA increased with light augmentation and decreased with phosphorus enrichment, whereas PUFA decreased with light and increased with phosphorus. Light effects on fatty acid composition were strongest in phosphorus‐poor streams. PUFA declined with increasing light/phosphorus ratios in the streams, whereas ‘energy’ fatty acids (16 : 0 and 16 : 1) increased. The ratio of SAFA/PUFA was strongly and positively correlated with C/P and C/N ratios. SAFA and MUFA, normalised to dry mass, increased two‐ to threefold with increasing light, while PUFA normalised to dry mass was not significantly affected by light. 6. Similarities in the responses of fatty acids and elemental stoichiometry to light and phosphorus treatments suggested that they were influenced by a common mechanism. Both components of food quality appeared to be sensitive to light‐regulated rates of carbon fixation which, when coupled with insufficient supplies of phosphorus, caused diatom cells to store surplus carbon in SAFA, MUFA and other carbon‐rich compounds that diluted both essential fatty acids and mineral nutrients.  相似文献
