Previous work has established that experienced male ring doves (Streptopelia risoria) can maintain prolactin-dependent crop growth and readiness to incubate by observing an incubating partner. We report that this is also true for female ring doves. The role of experience in this phenomenon was examined in separate experiments with males and females. Observation of an incubating mate from 3 days after completion of egg laying is sufficient to maintain crop growth and incubation in both male and female ring doves in their second, but not in their first, reproductive cycle. Male and female doves in their first cycle must incubate for a greater part of the cycle before observation of an incubating mate is an effective stimulus; there are no differences between first and second cycle doves separated by a glass plate from the mate and nest 8 days after laying. Experience obtained within the first cycle apparently ensures that previously neutral stimuli come to elicit prolactin secretion. The effectiveness of these stimuli is reinstated early in a second cycle.  相似文献

In Ramos cells, a human Burkitt's lymphoma cell line, stimulation of the B cell antigen receptor with anti-IgM antibody (Ab) induces apoptosis as indicated by a decrease in cell viability and an increase in DNA fragmentation and cell surface exposure of phosphatidylserine. Furthermore, these changes are suppressed by incubating the cells in alpha(1)-acid glycoprotein (AGP)-coated tissue culture plates. Here, we found that, during Anti-IgM Ab-induced apoptosis in Ramos cells, caspase-3 is activated downstream of caspase-8 and the mitochondrial pathway is activated, as indicated by a loss of mitochondrial membrane potential, an increase in the release of cytochrome c to the cytoplasm, and enhanced Bax expression. Anti-IgM Ab-induced apoptosis of neuraminidase-treated Ramos cells was suppressed by incubating the cells on plates coated with AGP, which contains a high concentration of alpha2,6-linked sialic acid. The incubation on plates coated with AGP also suppressed anti-IgM Ab-stimulated caspase-3 activity and increased the level of X-linked inhibitor of apoptosis protein (XIAP), but it did not affect caspase-8 activity, the mitochondrial membrane potential, cytochrome c release, or Bax expression. The results indicate that the interaction of Ramos cells with immobilized alpha2,6-linked sialic acid enhances XIAP expression, directly or indirectly suppressing caspase-3 activity and inhibiting anti-IgM Ab-induced apoptosis.  相似文献

To assess the possibility that vasoactive intestinal polypeptide (VIP) plays a role in naturally occurring changes in prolactin secretion in ring doves, we used immunohistochemical techniques to measure VIP-like immunoreactivity in the brain as a function of stage of the reproductive cycle. Differences between parental and nonparental birds in VIP profiles were detected in the ventral portion of the infundibular region. More specifically, there is an increase in cell size and staining intensity in the ventral infundibulum of breeding birds compared to simultaneously processed tissue taken from control animals. In both sexes, an increase in size of VIP-like immunoreactive cells is detectable during courtship and early incubation, anticipating the increase in plasma prolactin levels. VIP cell size remains elevated from about Incubation Day 14 to Brooding Day 14, and a steady decrease is observed during the remaining posthatching period, as squab begin to feed independently. Compared to parents rearing one squab, those with two young have a prolonged interval of increased infundibular VIP immunoreactivity. Furthermore, doves with no previous experience of a breeding cycle exhibit prolonged VIP-like immunoreactivity compared to experienced parents, paralleling previously described differences between these groups in parental behavior.  相似文献

A soluble antigen isolated from Eimeria stiedai merozoites with a molecular mass of 49 kDa was detected in the bile of infected rabbits. Rabbits immunized with the antigen shed a lower number of oocysts than did nonimmunized rabbits postchallenge (p.c.). The immunized rabbits showed a marked and transient increase of alanine-aminotransferase (ALT) activity on day 8 p.c. The blood indocyanine green (ICG) clearance and r-glutamyltransferase (GGT) activity showed no change throughout the experiment However, nonimmunized rabbits showed a gradual increase of ALT and GGT in the plasma and a delay of ICG p.c. Many merozoites were observed in the biliary ducts of the nonimmunized rabbits on day 8 p.c. using standard histology. In contrast, in the immunized rabbits, many inflammatory cells were observed around the biliary ducts, but there were few parasites in the tissue. These results suggest that the 49-kDa soluble protein antigen detected in the bile of the infected rabbits was a merozoite-specific antigen, and the immune reaction to the antigen may induce protective effects against the infection.  相似文献

The effects of eccentric exercise on changes innumbers of circulating leukocytes, cell activation, cell adhesion, andcellular memory function were investigated in 12 men, aged 22-35yr. The immunologic effects of postexercise epidermal treatment withmonochromatic, infrared light were also evaluated. Blood was drawnbefore and 6, 24, and 48 h after exercise for phenotyping and analysisof creatine kinase activity. There was an increase in leukocyte, monocyte, and neutrophil number, no change in the number of basophils, eosinophils, B cells, and T cells, and a decrease in natural killer cell number postexercise. Some markers of lymphocyte and monocyte activation remained unchanged or decreased, whereas the expression ofadhesion molecules 62L and 11b increased on monocytes. It is concludedthat eccentric exercise induced decreased activation, and increasedcell adhesion capacity, of monocytes. Altered trafficking of cellsbetween lymphoid tissue and blood, selective apoptosis, orattachment/detachment from the endothelial wall can explain theobserved phenotypic changes. Treatment with monochromatic, infraredlight did not significantly affect any of the investigated variables.Correlations between immunologic and physiological parameters indicatea role of the immune system in adaptation to physical exercise.  相似文献

The cellular activity of an antigen is understood as its power to cause plasma cells to accumulate in the regional lymph node. Two plasma cell units (PCU) is the dose causing one plasma cell addition (as compared with the “background”), whereas 1 PCU causes neither increase nor decrease in plasma cell number in the regional lymph node. Injections of antigen in less than 1 PCU causes plasma cells to decrease in number. Interrelation between antigen and plasma cells changes with time in different regions of the lymphatic system.  相似文献

为了解HCV感染后细胞免疫在其中的作用,对31例慢丙肝及20例正常献血员以MTT法检测外周血单核细胞(PBMC)对HCVE2/NS1相对保守区多肽抗原及C22、NS5的增殖反应。结果表明与正常人相比,慢性丙型肝炎患者PBMC对HCVC22、E2/NS1抗原有明显的增殖反应(P<005),而对NS5无明显增殖,其中以C22抗原性最强。作者认为慢性丙型肝炎患者存在针对与HCV相关抗原的细胞免疫,这种细胞免疫不能消除HCV感染,而且与疾病的状态无关。  相似文献

Passive immunization of incubating ring doves with daily injections of sheep anti-chicken vasoactive intestinal polypeptide (cVIP) serum prevented the proliferation of crop sac tissue observed in control doves given nonimmune serum. Daily injections of anti-cVIP serum did not prevent crop sac development in nonbreeding doves simultaneously treated with ovine prolactin. The concentrations of plasma prolactin were significantly depressed in birds given anti-cVIP serum although this effect became progressively less pronounced during the course of the 7- or 14-day treatment periods. Body weights and weights of regressed reproductive organs were unaffected by treatment with anti-cVIP serum and did not differ significantly from control birds. Doves showing a decreased prolactin response to anti-cVIP serum treatment developed an immune response to sheep serum which may have immunoneutralized the administered antibody. Concentrations of plasma LH were not consistently affected by anti-cVIP serum administration and were low throughout the study. The depression in plasma LH normally seen in females after their young hatch was not observed in females treated with anti-cVIP serum. No effect of treatment was observed upon the birds' incubation behavior or in their readiness to feed and brood their young. These results suggest that in the ring dove, VIP is the physiological prolactin-releasing factor responsible for stimulating prolactin secretion and consequently the development of the crop sac, during incubation. They further indicate that increased concentrations of plasma prolactin may not be essential for gonadal regression or the maintenance of full incubation and brooding behavior in ring doves under laboratory conditions.  相似文献

In order to understand the mechanism of the shock reaction that can occur in Dirofilaria immitis-infected dogs, following treatment with diethylcarbamazine (DEC), a variety of blood parameters were measured. A decrease in red blood cell parameters occurred in reactive dogs, followed by an increase, suggesting a possible sequestration of erythrocytes followed by peripheral vasoconstriction. Changes in leucocytes were consistent with stress reactions. Thrombocytopenia occurred, but no evidence of coagulopathy was seen. Alanine aminotransferase and creatinine phosphokinase (especially CK1 and CK3) concentrations increased suggesting cell damage and possibly liver cell necrosis, possibly due to hypoxia. Complement was not activated during the reaction and there was no increase in circulating immune complexes. No changes in plasma concentrations or histamine, 5-hydroxytryptamine, kininogen or prostaglandin F metabolite were found. Bradycardia was not caused by changes in potassium or calcium ion concentrations. These results suggest that the shock reaction is not an anaphylactic Type 1 reaction and is not caused by mast cell degranulation or by immune complex activation of mediators. The most likely cause of the reaction is that DEC causes the liberation of substances from microfilariae or adult worms which are able to constrict the hepatic vein directly or via its sympathetic innervation.  相似文献

Previous field studies of hunter-harvested mourning doves (Zenaida macroura) have reported the percentage of birds with ingested lead shot as 0.2–6.5%. To reduce the uncertainty concerning the number of doves that ingest shot, we conducted an experiment to test the proportion of mourning doves that ingested lead shot on the bare soil of a disked field (typical of a managed dove field) to simulate more natural feeding conditions. In each of 3 treatment groups of 80 birds, we exposed 35 birds to low-density lead shot (1.5 million shot/ha), and35 birds to high-density lead shot (29.5 million shot/ha), and 10 birds served as controls (no shot). We dosed 5 positive control birds with 2 lead shot each in trials 2 and 3. We scattered lead shot and mixed seed on the loosely packed soil of treatment cages and after 4 days of exposure, 2.9% of doves voluntarily ingested ≥1 lead shot. The proportion of birds that ingested shot when exposed to the high-density shot treatment (4.9%) was not different (P = 0.098) from that of the low-density shot treatment (1.0%). Lead concentrations in liver, kidneys, and blood reached maxima of 94.402 ppm, 346.033 ppm, and 13.883 ppm wet mass, respectively. Differences in delta-aminolevulinic acid dehydratase (ALAD) activity, packed cell volume, and heterophil:lymphocyte ratio (H:L) were greater posttreatment in doves that had ingested shot than in those that did not. The risk posed to mourning doves from lead shot ingestion can be reduced by banning lead shot on management areas or dove fields or disking fields after hunting season to reduce shot availability. © 2011 The Wildlife Society.  相似文献

Moderate, habitual physical activity improves health, possibly because of beneficial changes in immune function. For example, physical activity can increase natural killer cell cytotoxicity, T cell proliferation, and macrophage function but has minimal impact on antigen-driven B-2-mediated immunoglobulin (Ig) responses. The following studies tested whether physical activity selectively impacts nonantigen-driven B-1-natural IgM (nIgM) but not antigen-driven B-2 Ig. Adult male, pathogen-free Sprague-Dawley rats in a barrier facility voluntarily ran in wheels from 7 to 56 days or were housed in an enriched environment for 56 days. Rats received either no antigen or keyhole limpet hemocyanin (KLH) to assess the B-2 response. Blood samples assessed serum nIgM, total IgG, total serum protein, anti-KLH IgM, and anti-KLH IgG. Physically active rats had higher serum nIgM after 7 days of running, and nIgM remained elevated over 56 days of running. In contrast, free-wheel running produced no changes in total IgG, total serum protein, anti-KLH IgM, and anti-KLH IgG. Environmental enrichment did not alter immune measures from controls. These results suggest that B-1, not B-2, cell responses are selectively impacted by physical activity. Because nIgM is important in multiple aspects of the immune response, an elevation in this innate humoral component could contribute to improved immunity in physically active organisms.  相似文献

The influence of an antigen injected at different periods of the circadian mitotic activity on the cell proliferation in the thymus cortex was studied. The duration of mitosis and the number of cells entering division were determined. A pronounced stimulating effect of immunization with sheep red blood cells entering mitosis increased, while the duration of mitosis diminished (colchamine mitotic index was 29.79 percent in control mice, and 47.99 percent in the immunized ones. The duration of mitosis was 0.4 hours in control animals, and 0.24 hours in the immunized ones. When compared with intact mice, the animals immunized in the evening showed no significant changes in the parameters studied.  相似文献

Different variants of the popliteal lymph node (PLN) assay have been published. Here we describe the adjuvant popliteal lymph node assay, an immune response assay to study the adjuvant activity of soluble substances as well as particulate matter. The substance to be studied for adjuvant activity is injected into the hind footpad of mice or rats together with an antigen. Adjuvant activity is determined as the increase in PLN weight and cell numbers in animals receiving antigen together with the substance under study, compared with PLN weight and cell numbers in animals given the antigen without the substance in question, and animals given the putative adjuvant alone. Because lymph node weight and cell numbers are immunologically non-specific parameters, specific immune response assays like serum antibody responses or antibody-forming cell numbers should additionally be performed. Different antigens and immune response assays may be used, depending on the research question asked. In relation to respiratory (or food) allergy, the assays should as a minimum include determination of specific IgE in serum, and preferably also IgG1 (mouse). Serum specific IgG2a antibody determination may be added to get an indication of the Th1-Th2-balance of the response. The adjuvant PLN assay, with cellular response assays performed in the draining popliteal lymph node and antibody determinations in serum, requires small amounts of test material. The assay offers a practical, sensitive and reproducible method to determine the adjuvant activity of soluble substances as well as particulate material, with the possibility to also perform mechanistic studies.  相似文献

Many birds and mammals show changes in the hypothalamo-pituitary-gonadal (HPG) axis in response to social or sexual interactions between breeding partners. While alterations in GnRH neuronal activity play an important role in stimulating these changes, it remains unclear if acute behaviorally-induced alterations in GnRH release are accompanied by parallel changes in GnRH synthesis. To investigate this relationship, we examined changes in the activity of GnRH neurons in the brains of male ring doves following brief periods of courtship interactions with females. Such interactions have been previously shown to increase plasma LH in courting male doves at 24 h, but not at 1 h, after pairing with females. In the first study, males allowed to court females for 2 h had 60% more cells that showed immunocytochemical labeling for GnRH-I in the preoptic area (POA) of the hypothalamus than did control males that remained isolated from females. To determine whether an increase in GnRH gene expression preceded this increase in GnRH immunoreactivity in the POA, changes in the number of cells with detectable GnRH-I mRNA in the POA were measured by in situ hybridization following a 1 h period of courtship interactions with females. In this second study, courting males exhibited 40% more cells with GnRH-I in this region than did isolated control males. GnRH-immunoreactive neurons in two other diencephalic regions failed to show these courtship-induced changes. Plasma LH was not elevated after 1 or 2 h of courtship. These results demonstrate that the release of GnRH-I in the POA that is presumably responsible for courtship-induced pituitary and gonadal activation is accompanied by a rapid increase in GnRH synthesis that occurs before plasma LH levels increase. We suggest that this increase in GnRH synthesis is necessary to support the extended period of HPG axis activation that is seen in this species during the 5–10 day period of courtship and nest building activity.  相似文献

Previous studies have indicated that there is a cardiodepressant factor in the medium incubating the posterior pituitary lobe in situ. The cardiodepressant activity of the medium incubating the posterior pituitary lobe before and during stimulation of the vagus nerves was tested on isolated auricles of the right heart atrium of a two-day-old rat. It was found that the medium incubating the posterior pituitary lobe collected before stimulation decreased the contraction rate of the auricle by 34%, while that collected during the intermittent stimulation of the central ends of the cut vagus nerves caused a decrease of the auricle contractions frequency by 52%. The addition of cholinergic, serotoninergic, histaminergic receptor blockers or prostaglandin synthetase into Ringer-Lock's solution bathing the auricle has no effect on the changes of the contraction rate caused by the incubation medium.  相似文献

This review examines possible role of progesterone receptor (PR) and androgen receptor (AR) "cross-talk" in the expression of courtship behaviour in the ring dove (Streptopelia risoria). In doves, although androgen has been mostly associated with aggressive courtship behaviour and progesterone with the initiation of incubation, progesterone administration to courting birds terminates the aggressive component of courtship whilst having no effect on nesting behaviour. Recent results in doves have identified a high density of androgen receptor and progesterone receptor immunoreactivity (AR-ir and PR-ir) in the hypothalamus of both sexes in regions known to be directly involved in courtship and incubation behaviour. Nuclear AR-ir in courting birds is widespread throughout the brain. Nuclear PR-ir is only localized in discrete regions of the preoptic hypothalamus of both sexes. In the anterior and posterior hypothalamus of courting birds an increase number of AR-ir and PR-ir neurons colocalizes (70-90%) in the nucleus preopticus anterior (POA), nucleus preopticus medialis (POM), nucleus preopticus paraventricularis magnocellularis (PPM), nucleus hypothalami lateralis posterioris (PLH), and tuberal hypothalamus (Tu). A lower percentage of colocalization is seen in birds at other stages of the breeding cycle. The high percentage of AR-ir and PR-ir colocalization in the preoptic hypothalamus of courting doves supports previous reports involving progesterone acting in these brain regions to terminate the androgen-dependent aggressive courtship behaviour in male doves. The increase in PR-ir staining intensity in AR-ir neurons in courting birds suggests that this progesterone-dependent termination of aggressive courtship display in males occurs at the receptor level and may be orchestrated by central oestrogen.  相似文献

A number of naturally occurring small organic molecules, primarily involved in maintaining osmotic pressure in the cell, display chaperone-like activity, stabilizing the native conformation of proteins, and protecting them from various kinds of stress. Most of them are sugars, polyols, amino acids or methylamines. Similar to molecular chaperones, most of these compounds have no substrate specificity, but some specifically stabilize certain proteins. In the present work, the capacity of trehalose and glycerol, two well-known osmolytes, to stabilize and renature inorganic pyrophosphatase is demonstrated. Both trehalose and glycerol significantly protect pyrophosphatase against thermoinactivation achieved by incubating the enzyme at temperatures up to 95 degrees C, and allow the enzyme already inactivated in the presence of these osmolytes to renature upon incubation at low temperatures. To the best of our knowledge, there are no data on the effects of these compounds on renaturation of thermoinactivated proteins. The correlation between the recovery of enzyme activity and structural changes indicated by fluorescence spectroscopy contribute to better understanding of the protein stabilization mechanism.  相似文献
