In mammalian blood plasma, serum albumin acts as a transport protein for free fatty acids, other lipids and hydrophobic molecules including neurodegenerative peptides, and essential metal ions such as zinc to allow their systemic distribution. Importantly, binding of these chemically extremely diverse entities is not independent, but linked allosterically. One particularly intriguing allosteric link exists between free fatty acid and zinc binding. Albumin thus mediates crosstalk between energy status/metabolism and organismal zinc handling. In recognition of the fact that even small changes in extracellular zinc concentration and speciation modulate the function of many cell types, the albumin-mediated impact of free fatty acid concentration on zinc distribution may be significant for both normal physiological processes including energy metabolism, insulin activity, heparin neutralisation, blood coagulation, and zinc signalling, and a range of disease states, including metabolic syndrome, cardiovascular disease, myocardial ischemia, diabetes, and thrombosis.  相似文献

The predominant strategy for using algae to produce biofuels relies on the overproduction of lipids in microalgae with subsequent conversion to biodiesel (methyl-esters) or green diesel (alkanes). Conditions that both optimize algal growth and lipid accumulation rarely overlap, and differences in growth rates can lead to wild species outcompeting the desired lipid-rich strains. Here, we demonstrate an alternative strategy in which cellulose contained in the cell walls of multicellular algae is used as a feedstock for cultivating biofuel-producing microorganisms. Cellulose was extracted from an environmental sample of Cladophora glomerata-dominated periphyton that was collected from Lake Mendota, WI, USA. The resulting cellulose cake was hydrolyzed by commercial enzymes to release fermentable glucose. The hydrolysis mixture was used to formulate an undefined medium that was able to support the growth, without supplementation, of a free fatty acid (FFA)-overproducing strain of Escherichia coli (Lennen et. al 2010). To maximize free fatty acid production from glucose, an isopropyl β-D-1-thiogalactopyranoside (IPTG)-inducible vector was constructed to express the Umbellularia californica acyl-acyl carrier protein (ACP) thioesterase. Thioesterase expression was optimized by inducing cultures with 50 μM IPTG. Cell density and FFA titers from cultures grown on algae-based media reached 50% of those (~90 μg/mL FFA) cultures grown on rich Luria-Bertani broth supplemented with 0.2% glucose. In comparison, cultures grown in two media based on AFEX-pretreated corn stover generated tenfold less FFA than cultures grown in algae-based media. This study demonstrates that macroalgal cellulose is a potential carbon source for the production of biofuels or other microbially synthesized compounds.  相似文献

Seasonal variations of lipid fatty acids of boreal freshwater fish species   总被引:2,自引:0,他引:2  
1. The fatty acid levels of muscle and liver lipids of perch, vendace and rainbow trout (Salmo gairdneri) cultivated in the same area (for comparison) were monitored. 2. The total lipid content in the muscle of perch (Perca fluviatilis) and vendace (Coregonus albula) was less than 50% of that in rainbow trout and a seasonal variation was clear only in vendace. 3. The relative amounts of polyunsaturated fatty acids as well as omega-3 acids were higher in vendace and perch than in cultivated rainbow trouts. Arachidonic acid content was much higher in vendace and perch than in rainbow trout. The content of monoenes was considerably higher in rainbow trout than in free freshwater fish. 4. The seasonal variations in the degree of unsaturation were small in fish muscle. 5. In the muscle of rainbow trout the relative amount of polyunsaturated fatty acids diminished with the increase of total lipids.  相似文献

Endproducts of anaerobic fermentation, volatile fatty acids (VFAs), occur in the intestines of fish but no direct comparisons exist between VFA levels in fish with differing feeding ecologies during different seasons. We measured intestinal concentrations of six types of VFA in the upper and lower intestines of two freshwater omnivores ( Cyprinus carpioi and Dorosoma cepedianum ), and one piscivore ( Micropterus salmoides ) during the spring, summer and autumn. Acetate occurred in all species, and was highest in M. salmoides . In all species, concentrations were similar between upper and lower guts and higher during the summer. All three species contained anerobic bacteria and C. carpio and D. cepedianum contained cellulolytic types. Scanning electron microscopy revealed extensive colonization and suggested microbial breakdown of digesta in M. salmoides . In radio-tracer experiments, C. carpio dosed orally with [2–14C] acetate contained label in liver, muscle and blood tissues. Amounts of intestinal VFA did not appear to increase in species with refractile diets, and low VFA in D. cepedianum suggests fermentation plays a minimal role in the nutrition of this species. Low levels of intestinal VFA during cool seasons are consistent with the hypothesis that temperature limits fermentation in these Species.  相似文献

The pine moth Dendrolimus pini effectively resists many insecticides, but it can be controlled by the use of bioinsecticides such as entomopathogenic fungi. In the use of microbial agents for the biocontrol of D. pini, it is important to identify the cuticular lipids of this pest if we are to understand the factors responsible for the preferential adhesion or selective repulsion of entomopathogenic fungi that are potentially useful in biocontrol. In this work the qualitative and quantitative analyses of free fatty acids in two exuviae extracts (petroleum ether and dichloromethane) and two developmental stages (larval-larval and larval-pupal molts) were studied. The free fatty acid composition of the epicuticular lipids from exuviae of D. pini was characterized chemically using gas chromatography (GC) and gas chromatography-electron impact mass spectrometry (GC-MS). Structural analyses of the dichloromethane extracts from larval-larval exuviae (LLE) and larval-pupal exuviae (LPE) revealed that the carbon numbers for the major acid moieties ranged from C8:0 to C34:0. Only C23:0 was not identified in the LPE extract. The relative contents of fatty acids in the extracts varied from trace amounts to 34%. The fatty acids extracted by dichloromethane were essentially the same as those in the petroleum ether extract. We also identified dehydroabietic acid in the exuviae of D. pini. The respective quantities of dehydroabietic acid obtained from D. pini LLE and LPE were 1763 ± 103 μg/g exuviae and 11521 ± 1198 μg/g of exuviae.  相似文献

The effects of nicotinic acid on plasma free fatty acid levels and turnover and on plasma glycerol levels were studied in the first few hours after administration to man and the sheep. In both species a fall in all parameters studied was followed by a rise above basal level, interpreted as due to an increase in lipolysis above resting level. P/O ratio, state 3 and 4 respiration rates, and acceptor control index (ACI) were assessed in rat liver mitochondria following an overnight fast and single bout of treadmill exercise of 30-180 min. P/O was unaffected by fasting and 30 min of exercise; however, ACI was reduced because of an increase in state 4 respiration. Fasting, followed by running for 1 h or more decreased P/O approx. 40% and ACI by 50%, an effect that could be attributed to a reduction in state 3 respiration. The decrease in P/O was reversed 15 min after the cessation of exercise, whereas ACI remained depressed. All these functional alterations were mimicked by incubation of isolated mitochondria with palmitate and reversed by washing them with albumin. No direct correlation between plasma free fatty acids and the alterations in mitochondrial respiration was apparent. These data demonstrate that the decrease in the normal coupling of oxidation and phosphorylation in liver mitochondria produced by fasting/exercise is reversed rapidly in vivo. Furthermore, it is apparent that, if fatty acids act as a regulatory agent under these conditions, they do not do so solely on the basis of their plasma concentration.  相似文献

Morphometric‐based condition indices are widely used to assess proximate body composition and, collaterally, feeding and living conditions of fish. However, the exact relationship between condition indices and proximate body composition of fish and its relatedness to life history traits and seasonality has yet to be fully explored. Therefore, the aim of this study was to examine how the Fulton's condition factor (K‐factor) is related to the chemical composition (i.e. lipid, protein, water and carbon content, and molar carbon : nitrogen ratio), length and gonadal development of fish, and how these relationships are influenced by gender and seasonality in three freshwater fish species: Amur sleeper (Perccottus glenii), pumpkinseed (Lepomis gibbosus) and rudd (Scardinius erythrophthalmus). It was found that the strength and direction of association between the K‐factor and proximate body composition can vary markedly among fish species. The K‐factor correlated positively with gonadal development in pumpkinseed and Amur sleeper, while no such relationship existed in rudd. Condition factor can be a reliable measure of lipid content; however, the relationship was stronger in species with higher and more variable lipid contents. Moreover, a striking and consistently negative linkage was found between the K‐factor and water content of the fish body, which corresponds to the findings of several other studies. In turn, any relationship between the K‐factor and the protein content of fish was not detected. Gender seemed to exert a negligible effect on the relationship between the K‐factor and proximate body composition, while seasonal variance was obvious in most relationships.  相似文献

1. The irreversible disposal rate coefficient for free fatty acids in the plasma of fed and starved rats was determined after a single intravenous injection of [1-(14)C]palmitic acid into each rat. The dose of labelled palmitic acid was given as a complex with (131)I-labelled albumin in rat serum. The total amount of [1-(14)C]palmitic acid remaining in the plasma was determined at short times after injection from the (14)C/(131)I ratio in the injected serum and in the collected plasma. The rate coefficient was determined from the area under the curve that describes the disappearance of [1-(14)C]palmitic acid with time from the plasma. Possible sources of error in these determinations are discussed. 2. The irreversible disposal rate coefficient was significantly higher in fed rats (2.07min(-1)) than in rats which had been starved for 24h (1.53min(-1)). The possible relationship between this difference and the processes whereby free fatty acids are removed from the plasma is discussed briefly. 3. An estimate of the irreversible disposal rate for free fatty acids in plasma was made from the concentration of free fatty acids in plasma and from the volume of distribution of (131)I-labelled albumin. The irreversible disposal rate was significantly lower in the fed state than in the starved.  相似文献
