Preference-aversion for heavy metals in ddy mice was examinedusing the two-bottle preference technique. Mice preferred MnCl2,CoCl2 at certain concentration ranges, but avoided NiCl2, CuCl2,ZnCl2 and CdCl2. The avoidance threshold for the salts becamelower in the order of Mn > Co > Ni > Cd > Cu >Zn, though the degree of rejection for CuCl2 was larger thanthat for ZnCl2. Preferences for sucrose and Na saccharin werereduced by low concentrations of CuCl2 or ZnCl2. However, preferencefor glycine was scarcely affected by addition of either saltat 1 mM. Electrophysiological experiments in mice on the inhibitionby heavy metal salts of chords tympani nerve responses to thefour basic taste stimuli revealed that CuCl2 and ZnCl2 at 1mM inhibited the response to sucrose strongly and those to guinineHCl (QHCl) and NaCl moderately, but had no effect on the HClresponse. On the other hand, MnCl2, CoCl2 and NiCl2 at 1 mMscarcely inhibited the response to sucrose but suppressed thoseto other stimuli. CoCl2 moderately inhibited responses to allthe stimuli. In general, metal salts producing a larger inhibitionof neural responses to sucrose and QHCL are avoided by miceat a lower concentration.  相似文献

Catalase plays a major role in protecting cells against toxic reactive oxygen species. Here, Catalase was purified from larvae of the camel tick Hyalomma dromedarii and designated TLCAT. It was purified by ammonium sulfate precipitation and chromatography on DEAE-cellulose, Sephacryl S-300 and CM-cellulose columns. Gel filtration and SDS-PAGE of the purified TLCAT indicated that the protein has a native molecular weight of 120 kDa and is most likely a homodimer with a subunit of approximately 60 kDa. The Km value of TLCAT is 12 mM H2O2 and displayed its optimum activity at pH 7.2. CaCl2, MgCl2, MnCl2 and NiCl2 increased the activity of TLCAT, while FeCl2, CoCl2, CuCl2 and ZnCl2 inhibited the activity of TLCAT. Sodium azide inhibited TLCAT competitively with a Ki value of 0.28 mM. The presence of TLCAT in cells may play a role in protecting H. dromedarii ticks against oxidative damage. This finding will contribute to our understanding of the physiology of these ectoparasites and the development of untraditional methods to control them.  相似文献

A highly purified adenosine 3′,5′-monophosphate-dependent protein kinase from bovine brain has been found to catalyze its own phosphorylation. The incorporated phosphate was shown to be associated with the cyclic AMP-binding subunit (R-protein) of the protein kinase. The catalytic subunit exhibited no detectable incorporation of phosphate into itself, but was required for the phosphorylation of R-protein. The molecular weight of R-protein was determined by polyacrylamide gel electrophoresis to be about 48,000 in the presence of sodium dodecyl sulfate. Cyclic AMP strikingly inhibited the rate of autophosphorylation observed in the presence of ZnCl2, CaCl2, NiCl2, or FeCl2, but had no significant effect in the presence of MgCl2 or CoCl2. The concentration of cyclic AMP required to give half-maximal inhibition of phosphorylation was 3 × 10?7m in the presence of either CaCl2 or ZnCl2. Guanosine 3′,5′-monophosphate was far less effective under the same experimental conditions than cyclic AMP. R-protein appears to be similar to a phosphoprotein recently discovered in synaptic membrane fractions from rat and bovine cerebral cortex.  相似文献

The effects of HgCl2, NiCl2 and CoCl2 on Nostoc linckia (Roth) Born. et Flah. were studied. Low level (0.2 p.p.m.) of mercury increased heterocyst frequency, stimulated nitrate reductase and ammonium uptake, but significantly inhibited acetylene-reducing and glutamine-synthetase activities. In contrast, NiCl2 and CoCl2 greatly inhibited all of the above processes.  相似文献

Four triazine herbicides: amitrole, metribuzin, prometryn and terbutryn, and the bipyridal compound diquat dibromide have been evaluated for genotoxicity in the wing somatic mutation and recombination test of Drosophila melanogaster, following standard procedures. Third-instar larvae trans-heterozygous for the third chromosome recessive markers multiple wing hairs (mwh) and flare-3 (flr(3)) were chronically fed with different concentrations of the test compounds. Feeding ended with pupation of the surviving larvae. Genetic changes induced in somatic cells of the wing's imaginal discs lead to the formation of mutant clones on the wing blade. Point mutation, chromosome breakage and mitotic recombination produce single spots; while twin spots are produced only by mitotic recombination. Exposure to 0.5 mM and 1 mM of amitrole clearly increased the frequency of small single, large single and total spots. Terbutryn, at the concentration of 5 mM, induced a slight increase in the frequency of small single and total spots, but this result could be false positive. The other three herbicides tested did not show any genotoxic effect. When heterozygous larvae for mwh and the multiple inverted TM3 balancer chromosomes were treated, significant increases in the frequency of mutant spots were only detected for amitrole. The observed spot frequencies were lower than those found in mwh/flr(3)50%) of the total spot induction was due to mitotic recombination.  相似文献

DNA interstrand crosslinks (ICLs) are the primary mechanism for the cytotoxic activity of many clinical anticancer drugs, and numerous strategies for forming ICLs have been developed. One such method is using crosslink-forming oligonucleotides (CFOs). In this study, we designed a 4-amino-6-oxo-2-vinylpyrimidine (AOVP) derivative with an acyclic spacer to react selectively with guanine. The AOVP CFO exhibited selective crosslinking reactivity with guanine and thymine in DNA, and with guanine in RNA. These crosslinking reactions with guanine were accelerated in the presence of CoCl2, NiCl2, ZnCl2 and MnCl2. In addition, we demonstrated that the AOVP CFO was reactive toward 8-oxoguanine opposite AOVP in the duplex DNA. The structural analysis of each guanine and 8-oxoguanine adduct in the duplex DNA was investigated by high-resolution NMR. The results suggested that AOVP reacts at the N2 amine in guanine and at the N1 or N2 amines in 8-oxoguanine in the duplex DNA. This study demonstrated the first direct determination of the adduct structure in duplex DNA without enzyme digestion.  相似文献

Precipitation of human fibrinogen in 0.15 m NaCl occurred at pH 7.4 (Tris-HCl buffer) when ZnCl2, CuCl2, NiCl2, or CoCl2 were added beyond their respective critical concentrations. The critical concentrations were about 4 × 10?5m ZnCl2, 6 × 10?5m CuCl2, 3 × 10?4m NiCl2 and 1 × 10?3m CoCl2. At pH 5.8 2-(N-morpholino)-ethane sulphonic acid buffer, the critical concentrations were found only for CuCl2 and ZnCl2, and were about 3 × 10?5and 3 × 10?4m, respectively. CaCl2 and MgCl2 were not effective up to 1 × 10?2and 2 × 10?2m at pH 7.4 and 5.8, respectively. At pH 7.4, precipitation was better in 0.015 m NaCl than in 0.15 m NaCl for both CuCl2 and ZnCl2. Little or no conformational change was indicated on binding Cu2+ ions. The fluorescence of tryptophan was quenched only by CuCl2, while other metal ions (ZnCl2, NiCl2, CoCl2 and CaCl2) were ineffective as quenchers.  相似文献

The newly developed somatic mutation and recombination test, utilizing the wing-hair mutations mwh and flr3, was used to evaluate the genotoxicity of the base analog 5-azacytidine in larvae of Drosophila melanogaster. Third instar larvae were fed media wetted with various concentrations of the compound, and wings of surviving adults were removed and scored for the presence of clones of cells possessing malformed hairs. Wings of exposed flies trans-dihybrid for mwh and flr3 had significantly increased frequencies of twin spots, small single spots and large single spots. Significant linear regression of twin-spot frequencies upon concentration was also obtained. Induction of twin spots by 5-azacytidine unambiguously demonstrates its recombinogenic activity in somatic cells of Drosophila. Significantly increased frequencies of large single spots on wings of inversion-heterozygous flies were also observed and suggest that 5-azacytidine may also be inducing somatic gene mutations (or deletions).  相似文献

The ability of ascorbic acid (Vitamin C) to modulate the genotoxic action of several mutagens was investigated in the wing spot test of Drosophila melanogaster. In this assay, 3-day-old transheterozygous larvae for the multiple wing hairs (mwh, 3-0.3) and flare (flr, 3-38.8) genes were treated with three reference mutagenic compounds, namely cobalt chloride (CoCl2), 4-nitroquinoline 1-oxide (4-NQO) and potassium dichromate (K2Cr2O7). The results obtained show that the three reference mutagens tested were clearly genotoxic in the Drosophila wing somatic mutation and recombination test (SMART). None of the three concentrations tested of ascorbic acid (25, 75 and 250 mM) induced significant increases in the frequency of the mutant clones recorded. When co-treatment experiments with ascorbic acid were carried out, different results were found. Thus, ascorbic acid was effective in reducing the genotoxicity of K2Cr2O7 virtually to the control level; on the contrary, it did not show any antigenotoxic effect on the genotoxicity of 4-NQO. Finally, co-treatments with CoCl2 and ascorbic acid show a significant increase in the frequency of mutant clones over the values obtained with CoCl2 alone.  相似文献

Nucleoli from Novikoff hepatoma ascites cells contain phosphatase activity that acts upon 32P-labeled nucleolar protein substrates. The activity is optimal near pH 7.0 and is inhibited by increasing concentrations of NaCl. The divalent cations CaCl2, MnCl2 and CoCl2 at 6 mM inhibited phosphatase activity from 30–60%. ZnCl2 completely inhibited the activity above 2 mM while EDTA and MgCl2 had little effect. The activity was stimulated by dithiothreitol and inhibited by N-ethylmaleimide indicating a requirement for free sulfhydryl groups.  相似文献

It has been proposed that mitotic recombination is involved in tumor promotion. To test this idea, we investigated the effect of a tumor promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA), on spontaneous and mitomycin C (MMC)-induced mitotic recombination in Drosophila melanogaster. The test system used was the wing-spot assay. Third instar larvae (mwh+/+flr) were treated with MMC (0-0.3 mM) for 6 h and fed media containing TPA (0-10 micrograms/ml), and the wings of surviving adults were analyzed for the presence of mutant spots. The results are: (1) MMC induced twin spots as well as single spots dose dependently (0.03-0.3 mM). (2) TPA itself did not induce twin spots or single spots in the range of doses examined (0.1-10 micrograms/ml). (3) TPA did not enhance the frequencies of twin or single spots induced by MMC. These results indicate that TPA induced neither mitotic recombination nor mutations under these experimental conditions. Thus the results would not support the mitotic recombination theory in tumor promotion. Rather, in this study, TPA posttreatment resulted in reduced frequencies of mutant single spots induced by MMC.  相似文献

An extracellular lethal toxin produced by Vibrio alginolyticus strain Swy originally isolated from diseased kuruma prawn (Penaeus japonicus) was partially purified by Fast Protein Liquid Chromatography with hydrophobic interaction (Phenyl Sepharose High Performance) chromatography and gel filtration columns. The toxin is an alkaline serine protease, inhibited by phenyl-methylsulfonyl fluoride (PMSF), and showed maximal activity at pH 10, having a molecular weight of about 33 kDa estimated by SDS-PAGE and gel filtration chromatography. In addition, the toxin was also completely inhibited by FeCl2 but partially inhibited by CaCl2, CuCl2, CoCl2, MnCl2, and ZnCl2, and not inhibited by ethylenediamine tetraacetic acid (EDTA), ethylene glycol-bis(β-amino-ethyl ether) N,N,N′,N′-tetraacetic acid (EGTA), iodoacetamide, pepstatin A, sodium dodecyl sulfate (SDS), and N-tosyl-l-phenyl-alanine chloromethyl ketone (TPCK). Both the crude extracellular products (ECP) and the partially purified toxin are lethal for kuruma prawn at LD50 values of 0.30 and 0.27 μg protein/g body weight, respectively. The addition of PMSF completely inhibited the lethal toxicity of both the ECP and the partially purified toxin, indicating that this serine protease is a lethal factor produced by the bacterium. The 33-kDa protease is, therefore, suggested to be a new toxic protease produced by V. alginolyticus strain Swy. Received: 12 April 1996 / Accepted: 31 July 1996  相似文献

Twenty metallic compounds were assayed for their genotoxic mutagenic activity by the bioluminescence test restoration of the luminescence of dark mutant of the luminous bacterium Photobacterium fischeri). The activity of the metals was tested in a liquid medium as well as on a solid medium. K2Cr2O7, MnCl2, BeCl2, KH2AsO4, ZnCl2 and Na2WO4 showed strong activity in liquid medium while AgNO3, Cd(OOCCH3)2, CoCl2, CuCl2, HgCl2, Na2SeO3 and Pb(NO3)2 were more active in the solid medium test. BaCl2, Na2MoO4, NaAsO2, NiSO4, Na2SeO4, RbCl, and SnCl2 were not active in the bioluminescence test. The correlation between the genotoxic activity of the tested metallic compounds in the bioluminescence test and other bacterial tests for genotoxic agents as well as the correlation between these results and the carcinogenicity of these compounds is discussed.  相似文献

In search for new conglomerates, seven stereochemically labile complexes between MCl2 (M = Co, Cu, Ni, Zn) and bidentate ligands, the commercially available N,N,N′-trimethylethane-1,2-diamine (trimeda) and the somewhat bulkier N-isopropyl-N,N′,N′-trimethylethane-1,2-diamine (itmeda), have been synthesized and characterized using single crystal X-ray diffraction. The trimeda and itmeda ligands exhibit chirogenic nitrogen centers and may form chiral metal complexes that are candidates for total spontaneous resolution. Copper(II) chloride forms the dimeric meso complexes [{CuCl2(trimeda)}2] (1) and [{CuCl2(itmeda)}2] (2), while [CoCl2(trimeda)2] (3) and [NiCl2(trimeda)2] (4) exhibit six-coordinate but chiral (R,R)- and (S,S)-complexes. Three examples of the chiral target complex, comprising four-coordinate stereochemically labile monomers, was successfully prepared, viz. [NiCl2(itmeda)] (5), [ZnCl2(itmeda)] (6), and [CoCl2(itmeda)] (7).In all seven complexes, the λ-conformation of the five-membered trimeda-metal chelate ring corresponds to the (S)-configuration at nitrogen, and vice versa. Supramolecular interactions in 3 and 4 form hydrogen-bonded heterochiral ribbons. However, crystals of 5-7 display homochiral interactions resulting in polar phases. Weak CH-Cl interactions in 5 and 6 form homochiral layers. In 7, interactions form homochiral helices along the a-axis.  相似文献

Fibers of the frog glossopharyngeal nerve (water fibers) thatare sensitive to water also respond to CaCl2, MgCl2 and NaCl.In the present study, interaction among cations (Ca2+, Mg2+and Na+) on taste cell membrane in frogs was studied using transitionmetals (NiCl2, CoCl2 and MnCl2), which themselves are barelyeffective in producing neural response at concentrations below5 mM. Unitary discharges from single water fibers were recordedfrom fungiform papillae with suction electrode. Transition metalions (0.05–5.0 mM) had exclusively enhancing effects onthe responses to 50 mM Ca2+, 100 mM Mg2+ and 500 mM Na+. Theeffects of transition metal ions were always reversible. Therank order of effectiveness of transition metals at 1 mM inthe enhancement of the responses to 50 mM CaCl2, 100 mM MgCl2and 500 mM NaCl was NiCl2 > CoCl2 > MnCl2. The concentrationof transition metal ions effective to enhance salt responsewas almost the same among Ca2+, Mg2+ and Na+ responses. Theresults suggest that a common mechanism is involved in the enhancementof Ca2+, Mg2+ and Na+ taste responses. The enhanced Mg2+ responseand the enhanced Na+ response were greatly inhibited by theaddition of Ca2+ ions, and the enhanced Ca2+ response was inhibitedby the addition of Mg2+ or Na+ ions, suggesting that competitiveantagonism occurs between Ca2+ and Mg2+ ions and between Ca2+and Na+ ions in the presence of Ni2+ ions. Ni2+ ions had a dualeffect on the Ca2+ response induced by low concentration (0.1mM) of CaCl2: enhancement at lower concentrations (0.02–0.1mM) of NiCl2 and inhibition at higher concentrations (0.5–5mM)of NiCl2. The present results suggest that transition metalions do not affect the receptor-antagonist complex, but affectonly the receptor-agonist complex.  相似文献

Summary Discophrya collini is a suctorian protozoan with contractile tentacles containing a microtubule-lined canal and microfilaments. The effects of a range of cations on tentacle contraction and ultrastructure have been determined. Treatment with 80 mM CaCl2 and 95 mM MgCl2 causes contraction to 28% and 57% of the control length respectively. Re-extension takes over 4 hours in the culture medium, but CaCl2-treated tentacles are re-extended after a 5 minutes treatment with 10–2 M EDTA or 5 × 10–3 M EGTA. CuCl2 causes a significant contraction at 10–5 M (to 77%); LaCl3 at 10–4 M (to 65%); ZnCl2 at 10–2 M (to 65%), but BaCl2, CoCl2, MnCl2, NiCl2, and SrCl2 cause significant changes only at 10–1 M.The cytoplasm of CaCl2-treated cells contains two forms of membraneous structures when viewed in TEM; that of MgCl2-treated cells reveals granular areas of medium electron density. None of these features are seen in control cells. The microtubules of the tentacle canal appear to be intact upon its retraction into the cell with no change occurring in the numbers or relative positions of the microtubules. The tentacle cortex is wrinkled. It is suggested from this and previous work that tentacle contraction may be mediated by a microfilament-based mechanism, and that calcium may be involved.  相似文献

The pore complex-lamina fraction obtained from nuclear envelope contains a protein phosphokinase activity capable of phosphorylating endogenous and exogenous protein substrates. Its specific activity in the presence of MgCl2 is approximately twice that of intact nuclear envelope. However, when MgCl2 is replaced by CoCl2 in the reaction mixture, a 7 to 12-fold increase in incorporation of 32P from γ-32P-ATP into protein substrate occurs. This appears not to be due to an effect of the divalent cation on the substrate, or to inhibition of a phosphoprotein phosphatase activity. Substitution of CuCl2, MnCl2, CaCl2, and ZnCl2 for MgCl2 results in a 20 to 30% decrease in incorporation of 32P. Cyclic AMP and cyclic GMP at 1 μM were without apparent effect. Approximately 40% of the total protein phosphokinase activity of the nuclear envelope is associated with the pore complex-lamina fraction.  相似文献

A 3′ -phosphoadenosine 5′ -phosphosulfate (PAPS):desulfoglucosinolate sulfotransferase (EC 2.8.2-) was extensively purified from light-grown cress (Lepidium sativum L.) seedlings by gel filtration and concanavalin A-Sepharose 4B, Matrex Gel Green A, and Mono Q fast protein liquid chromatography. The purified enzyme, which required bovine serum albumin for stabilization, had a native molecular weight of 31,000 ± 5,000 and an apparent isoelectric point of 5.2. Using PAPS (Km 60 micromolar) as sulfur donor, it catalyzed the sulfation of desulfobenzylglucosinolate (Km 82 micromolar), desulfo-p-hydroxybenzylglucosinolate (Km 670 micromolar), and desulfoallylglucosinolate (Km 6.5 millimolar) at an optimal pH of 9.0. All other potential substrates tested, including flavonoids, flavonoid glycosides, cinnamic acids, and phenylacetaldoxime, were not sulfated. Sulfotransferase activity was stimulated by MgCl2, MnCl2 and reducing agents and inhibited by ZnCl2, PbNO3 NiCl2 and the reaction product PAP. The thiol reagents N-ethylmaleimide, p-chloromercuriphenylsulfonic acid, and 5,5′ -dithio-bis-(2-nitrobenzoic acid) were also potent inhibitors, but the enzyme was protected from covalent modification by β-mercaptoethanol. The kinetics of desulfobenzylglucosinolate sulfation were consistent with a rapid equilibrium ordered mechanism with desulfobenzylglucosinolate binding first and PAPS second.  相似文献

This study evaluated different concentrations of selective serotonin-reuptake inhibitors (citalopram and sertraline) for genotoxicity by use of the somatic mutation and recombination test (SMART) in Drosophila melanogaster. Three-day-old larvae, trans-heterozygous for the multiple wing hairs (mwh) and flare (flr3) genes were treated with these two compounds. Two recessive markers were located on the left arm of chromosome 3, i.e. 'multiple wing hairs' (mwh) in map position 0.3 and 'flare-3' (flr3) at 38.8, while the centromere was located in position 47.7. SMART is based on the loss of heterozygosity, which may occur through various mechanisms, such as mitotic recombination, mutation, deletion, half-translocation, chromosome loss, and non-disjunction. Genetic changes occurring in somatic cells of the wing's imaginal discs, cause the formation of mutant clones on the wing blade. The results of this study show that citalopram had a genotoxic effect in the Drosophila SMART. Sertraline, however, did not show any genotoxic effect in balancer heterozygous wings. This study concluded that more information is needed to be certain regarding the mutagenic effects of sertraline.  相似文献

Crystalline D-glucose-isomerizing enzyme from Bacillus coagulans, strain NH–68 has been shown to consist of subunits by the method of electrophoresis on sodium dodecyl sulfate (SDS) polyacrylamide gels.

The dissociation behavior of the enzyme has been characterized. The enzyme dissociates into inactive subunits by the preincubation with 0.05% SDS in the presence of 5 × 10?3M MnCl2 or CoCl2, but not in the absence of these metal salts. In 8 м urea, however, the enzyme does not dissociate into subunits and the activity is completely recovered by dilution of the urea. Metal salts, such as MnCl2 and CoCl2, also do not affect activity in the presence of urea.  相似文献
