Effective management of coastal and marine resources requires knowledge of how community sensitivity varies spatially. With this in mind, we developed a benthic sensitivity index (SI), based on the distribution and abundance of five ecological groups that can be used to assess community tolerance to organic enrichment and other disturbances. The index, projected as a high-resolution map, ranks communities from those dominated by sensitive and ecologically important species (i.e. low SI values) to those composed mainly of tolerant and/or opportunistic species (i.e., high SI values). Applying our model to a multiple-use case study in southeast Brazil, we were able to show considerable variability in the sensitivity of communities across the study area that was relatively stable over time. This allowed us to evaluate the possible direct (i.e., spatially overlapping) and indirect effects (i.e., cumulative changes to the physical environment) of a range of activities on sensitive and ecologically diverse benthic communities. Our approach and the resulting high-resolution maps hold promise for a range of spatial planning applications, including the development of coastal infrastructure, assessments of the representativeness of marine protected areas and other activities such as the selection of appropriate locations for dredge spoil dumping. Overall, we present a novel and transparent way of extrapolating limited survey data to provide spatial and temporal information on the sensitivity of benthic communities in multiple-use coastal and marine areas.  相似文献

Objective: To determine whether cardiorespiratory fitness and/or physical activity (PA) were related to measures of insulin sensitivity and secretion independent of body composition in overweight Hispanic children. Research Methods and Procedures: Ninety‐five Hispanic children (n = 55 boys; n = 40 girls; 8 to 13 years old) participated in this investigation. The frequently sampled intravenous glucose tolerance test was used to determine the insulin sensitivity index (SI), the acute insulin response, and the disposition index. Cardiorespiratory fitness [maximal oxygen uptake (Vo2max)] was evaluated using a treadmill protocol, and PA was determined by an interviewer‐administered questionnaire. Body composition was measured using DXA. Results: Unadjusted correlations indicated that Vo2max (milliliters of O2 per minute) was negatively related to SI (r = ?0.46, p < 0.05) and disposition index (r = ?0.31, p < 0.05) and positively associated with fasting insulin (r = 0.29, p < 0.05), but these relationships were no longer significant once gender, Tanner stage, fat mass, and soft lean tissue mass were included as covariates (all p > 0.05). Multivariate linear regression analysis showed that body fat mass explained 53% of the variance in SI and that Vo2max (milliliters of O2 per minute) was not independently related to SI. Cardiorespiratory fitness was positively related to both fat mass (r = 0.43, p < 0.001) and soft lean tissue mass (r = 0.89, p < 0.001). PA was not related to any measure of insulin sensitivity and secretion. Discussion: Cardiorespiratory fitness, as determined by Vo2max (milliliters of O2 per minute), was not independently related to insulin sensitivity or secretion, suggesting that Vo2max influences insulin dynamics indirectly through fat mass.  相似文献

Objective: The aim of the study was to evaluate the diagnostic performance of a new ultrasound elastography (USE) parameter based on the measurement of the percentage of maximal stiffness within a nodule as compared with the already established elastographic strain index (SI) and to investigate their diagnostic performance according to nodule size.Methods: The study included 218 nodules. Each nodule underwent conventional ultrasound (US), USE evaluation, and fine-needle aspiration cytology (FNAC). Thyroid nodules were further stratified into 4 subgroups (G) according to their size (G1, <1 cm; G2, 1–2 cm; G3, >3 cm). USE evaluation comprised the measurement of the percentage of the areas included in the region of interest corresponding to the maximal stiffness (% Index) and of the SI.Results: The % Index and of the SI were significantly higher in malignant than in benign thyroid nodules, and both measurements displayed a good diagnostic performance (SI sensitivity and specificity, 0.66 and 0.90, respectively; % Index sensitivity and specificity, 0.76 and 0.89, respectively). Compared with SI, the % Index was more informative, both in the whole group of thyroid nodules (odds ratio [OR], 18.68; 95% confidence interval [CI], 6.06 to 63.49; P<.0001 versus OR, 26.15; 95% CI, 8.01 to 102.87; P<.0001, respectively) and in the G1 and G2 subgroups.Conclusion: The % Index is a stronger predictor of nodule malignancy than both the SI and the conventional US signs. ObjectiveIn patients with primary aldosteronism, adrenal venous sampling (AVS) is performed to determine the presence of unilateral or bilateral adrenal disease. During AVS, verification of catheter positioning within the left adrenal vein (AV) and the right AV by comparison of AV and inferior vena cava (IVC) cortisol levels can be variable. The objective of this study was to determine the utility of AV epinephrine levels in assessing successful AV cannulation.MethodsThis was a single institution, retrospective review of patients who underwent AVS with cosyntropin stimulation for primary aldosteronism between 2009 and 2018. Successful cannulation of the AV was defined by an AV/IVC cortisol ratio selectivity index (SI) ≥3:1. Epinephrine thresholds to predict catheter placement in the AV were determined using logistic regression. The calculated epinephrine thresholds were compared with previously published thresholds.ResultsAVS was performed on 101 consecutive patients and, based on the SI, successful cannulation of the left AV and right AV occurred in 98 (97%) and 91(90%) patients, respectively. The calculated optimal epinephrine threshold to predict AV cannulation was 364 pg/mL (sensitivity, 92.1%; specificity, 94.6%) and the calculated optimal AV/IVC epinephrine ratio threshold was 27.4, (sensitivity, 92.1%; specificity, 91.3%). Among the 14 patients with failed AV cannulation, 3 patients would have been considered to have successful AVS using AV epinephrine levels >364 pg/mL and AV/IVC epinephrine ratio >27.4 thresholds.ConclusionObtaining 2 right AV samples routinely as well as AV and IVC epinephrine levels during AVS could prevent unnecessary repeat AVS in patients with failed AV cannulation based on cortisol-based SI <3:1.  相似文献

Previous studies have shown that the hemodynamic response of the primary somatosensory cortex (SI) to electrical median nerve stimulation doubles in strength when the stimulus rate (SR) increases from 1 to 5 Hz. Here we investigated whether such sensitivity to SR is homogenous within the functionally different subareas of the SI cortex, and whether SR sensitivity would help discern area 3b among the other SI subareas. We acquired 3-tesla functional magnetic resonance imaging (fMRI) data from nine healthy adults who received pneumotactile stimuli in 25-s blocks to three right-hand fingers, either at 1, 4, or 10 Hz. The main contrast (all stimulations pooled vs. baseline), applied to the whole brain, first limited the search to the whole SI cortex. The conjunction of SR-sensitive contrasts [4 Hz − 1 Hz] > 0 and [10 Hz − 1 Hz] > 0 ([4Hz − 1Hz] + [10Hz − 1Hz] > 0), applied to the SI cluster, then revealed an anterior-ventral subcluster that reacted more strongly to both 10-Hz and 4-Hz stimuli than to the 1-Hz stimuli. No other SR-sensitive clusters were found at the group-level in the whole-brain analysis. The site of the SR-sensitive SI subcluster corresponds to the canonical position of area 3b; such differentiation was also possible at the individual level in 5 out of 9 subjects. Thus the SR sensitivity of the BOLD response appears to discern area 3b among other subareas of the human SI cortex.  相似文献

火山熔岩湿地作为湿地系统中重要组成部分,广泛分布于海南岛北部火山岩地区,其生态安全状况直接影响着与其相连的水陆生态系统的安全。基于Landsat遥感影像、DEM数据以及植被覆盖指数,通过构建稳定性及敏感性评价体系,在景观斑块尺度上对海南岛北部火山熔岩湿地分布区1988-2018年的稳定性动态变化及敏感性现状进行分析,旨在探索该区域生态安全分布格局。结果表明:(1)1988和2018年两个时间节点的湿地斑块稳定性均以府城镇、城西镇、龙泉镇等熔岩湿地主要分布区域的稳定性最高,西南部农业用地区域及北部沿海地区斑块稳定性最弱;(2)1988-2018年期间研究区域熔岩湿地稳定性整体呈增长趋势,面积重心和稳定性重心在向南转移的同时在东西方向上呈收缩状态;(3)研究区域极度敏感和高度敏感区域主要分布在湿地密集分布区域,不敏感区域则主要分布在研究区域北部的海口市区及开发利用较多的沿海乡镇;(4)稳定性与敏感性结果呈正相关关系,湿地斑块稳定性低的沿海及城镇地区,在生态上多表现为不敏感和低敏感,而湿地斑块稳定性高的区域通常表现出高敏感性。这也说明自然生态系统相较人工生态系统,虽然其结构更为完善稳定,抗干扰能力相对较强,但也更容易遭到破坏,恢复周期长,因而在城镇发展等人为活动中需要慎重对待高敏感区域。  相似文献

Nickel (Ni) exposure through the intestinal mucosa may cause a hypersensitivity reaction recently defined as allergic contact mucositis (ACM). This condition is identifiable by the oral mucosa patch test (omPT), a qualitative and subjective examination that requires clinical expertise. Our aim was to evaluate if a peripheral blood lymphocyte typing performed before and after the omPT for Ni may be able to objectify this examination for diagnostic purposes. Thirty patients with symptoms referable to the ingestion of Ni-rich foods were subjected to omPT for Ni. Before and after the omPT, each patient underwent blood sampling for the typing of total lymphocytes and their subsets (T, T helper or Th, T cytotoxic or Tc, B, natural killer or NK). Statistical analysis was performed by Student t test and receiver operating characteristic (ROC) curve analysis. According to the omPT outcomes, 18 patients were defined as Ni-sensitive and the remaining 12 as controls. In Ni-sensitive patients, the number of total, T, Th, Tc, and B lymphocytes/μL whole blood increased after the omPT (p?<?0.0001 for the first three, p?=?0.0004 and p?=?0.0001 for the last two lymphocyte types). No omPT-dependent lymphocyte increase was observed in controls. The post/pre omPT cell ratio, especially if calculated for Th lymphocytes, appears to be an effective index for diagnostic purposes (sensitivity?=?100 %, specificity?=?83.3 %, Youden index?=?0.833, area under curve (AUC)?=?0.926, p?<?0.0001). In conclusion, the peripheral blood lymphocyte typing with calculation of post/pre omPT cell ratio has the potential to support the omPT in diagnosing ACM, with the advantage of providing quantitative and objective data.  相似文献

In vitro sensitivity to pentavalent antimony (SbV) as meglumine antimoniate (Glucantime) of Leishmania of the Viannia subgenus isolated prior to treatment from patients with uncomplicated cutaneous leishmaniasis was evaluated for intracellular amastigotes in the U-937 human monocytic cell line and log phase promastigotes. The 50% effective dose (ED50) of pharmaceutical and additive-free formulations of Glucantime were determined based on the kinetics of the response of Leishmania Viannia to SbV in vitro. ED50 to SbV was inversely related to time of exposure to drug. The potency of the additive-free formulation of Glucantime was significantly greater than that of the pharmaceutical formulation, irrespective of the parasite form. In vitro sensitivity to SbV ranged from < 5.3 micrograms/ml to > 170.0 micrograms/ml. Under the conditions used, 11 (39%) of 28 strains were sensitive to clinically achievable serum concentrations of SbV. No correlation was observed between the total amount of SbV required for healing of lesions and the in vitro response to the pharmaceutical formulation of Glucantime. In contrast, a significant correlation (P = 0.001) was observed between clinical response and the in vitro sensitivity of promastigotes to the additive-free formulation of Glucantime. The greater potency of the additive-free formulation of Glucantime, the correlation of in vitro sensitivity of promastigotes to this formulation and the clinical response to treatment, and the effect of time of exposure to SbV demonstrate the importance of assay conditions on the outcome and interpretation of in vitro evaluation of drug sensitivity.  相似文献

Ecosystem dynamics and the responses to climate change in mangrove forests are poorly understood. We applied the biogeochemical process model Biome-BGC to simulate the dynamics of net primary productivity (NPP) and leaf area index (LAI) under the present and future climate conditions in mangrove forests in Shenzhen, Zhanjiang, and Qiongshan across the southern coast of China, and in three monocultural mangrove stands of two native species, Avicennia marina and Kandelia obovata, and one exotic species, Sonneratia apetala, in Shenzhen. The soil hydrological process of the model was modified by incorporating a soil water (SW) stress index to account for the impact of the effective SW availability in the coastal wetland. Our modified Biome-BGC well predicted the dynamics of NPP and LAI in the mangrove forests at the study sites. We found that the six mangrove systems differed in sensitivity to variations in the effective SW availability. At the ecosystem level, however, soil salinity alone could not entirely explain the limitation of the effective SW availability on the productivity of mangrove forests. Increasing atmospheric CO2 concentration differentially affected growth of different mangrove species but only had a small impact on NPP (<7%); whereas a doubling of atmospheric CO2 concentration associated with a 2°C temperature rise would increase NPP by 14–19% across the three geographically separate mangrove forests and by 12% to as much as 68% across the three monocultural mangrove stands. Our simulation analysis indicates that temperature change is more important than increasing CO2 concentration in affecting productivity of mangroves at the ecosystem level, and that different mangrove species differ in sensitivity to increases in temperature and CO2 concentration.  相似文献

当前,城市化与全球变化背景叠加,海岸带生态系统发生了巨大的变化,沿海城市在全球变化下的脆弱性、敏感性与适应能力等问题已经成为政府和科学界关注的重要问题.土地利用变化是导致生态系统敏感性的重要因素之一.以厦门市作为沿海城市的典型代表,对近20 a土地利用变化下的生态敏感性进行系统研究.分析了城市建设用地扩张、围填海等主要土地利用活动对生态敏感性的影响机制;其次建立土地利用变化下的生态敏感性指数,并分析其变化过程;最后通过ArcGIS图层叠加计算功能,获得生态敏感性空间分布状况.结果表明:1987-2007年厦门市城市建设用地面积由1987年的67.48 km2,增加为2007年的308.21 km2,扩张了4.57倍,主要为蚕食农业用地和林地而来;海岸线长度由1987年的290.19 km,增加为1992年的343.23 km,而后减小为2007年的299.93 km,围垦养殖活动使得岸线变得曲折,填海造地活动导致岸线变得平滑.厦门市土地利用强度由1987年的2.44逐年增加为2007年的2.52,生态系统服务价值总体呈现减小的趋势,由1987年的7.39×109元减少到2007年的7.02× 109元,土地利用强度与生态系统服务价值呈现负相关关系.土地利用变化下的生态敏感性指数由1992年的1.50增加为1997年的4.94,而后减小为2002年的4.12,再增加为2007年的4.47.整体而言,近20a来厦门市生态系统对土地利用变化的敏感性灵敏响应程度不很剧烈,生态敏感性高的区域主要位于沿海区域.  相似文献

Rock barrens support rare plant species but may be threatened by forest expansion. We determined the extent of forest expansion onto open coastal barrens and identified environmental correlates of dynamic versus persistent barrens in Nova Scotia, Canada. We used aerial photos to quantify the amount of forest expansion over the last 70 years at five coastal barrens sites and GIS to derive topographic and other environmental predictors to differentiate persistent coastal barrens compared with persistent forests or barrens that succeed to forests. Linear discriminant and classification tree analyses identified the variables associated with each class of habitat. Coastal barrens decreased by an average of 7.9% (from 4.2 to 24.6% depending on the site) in the last 70 years due to forest expansion. The best predictors of persistent barrens were elevation and distance to coast. Environmental factors such as topographical heterogeneity and evidence of fire varied among sites. Climatic and edaphic conditions near the coast and in exposed inland areas may protect coastal barrens vegetation from forest expansion. Evidence of fire was not found at all barrens sites, thus at least some of the persistent open barrens are likely maintained by shallow soils, salt spray, and wind exposure. All three classes of habitat had distinct vegetation, and the only rare species was found in a persistent barren. Management of human activities in such landscapes should take into account the dynamic nature of barrens vegetation, while prioritizing conservation efforts in persistent barrens.  相似文献

Sensitivity of 159 isolates of Zymoseptoria tritici collected from durum wheat fields in Tunisia in 2012 was analysed towards pyraclostrobin, fluxapyroxad, epoxiconazole, metconazole, prochloraz and tebuconazole using microtiter tests. All isolates were found to be highly sensitive to pyraclostrobin with EC50 <0.01 mg/l with the exception of three isolates from the same field with higher EC50 values (>0.5 mg/l). These three isolates carried a mutation in the cytochrome b gene encoding the G143A substitution. This is the first report of quinone outside inhibitors (QoI) resistance in Z. tritici in Tunisia. Sensitivity towards r fluxapyroxad was in a narrow range with EC50 values ranging between 0.013 and 0.125 mg/l, which can serve as baseline sensitivity data for the future. Demethylation inhibitors sensitivity varied across a broad range with the data indicating a slight shift in sensitivity when compared to a previous study on the 2010 population. No highly sensitive strains were isolated from samples from fields, which had received three or four DMI applications.  相似文献

Sensitivity of African biomes to changes in the precipitation regime   总被引:4,自引:0,他引:4  
Aim Africa is identified by the Inter‐governmental Panel on Climate Change (IPCC) as the least studied continent in terms of ecosystem dynamics and climate variability. The aim of this study was (1) to adapt the Lund‐Postdam‐Jena‐GUESS (LPJ‐GUESS) ecological modelling framework to Africa by providing new parameter values for tropical plant functional types (PFT), and (2) to assess the sensitivity of some African biomes to changes in precipitation regime. Location The study area was a representative transect (0–22° N and 7–18° E) through the transition from equatorial evergreen forests to savannas, steppes and desert northwards. The transect showed large latitudinal variation in precipitation (mean rainfall ranged from 50 to 2300 mm year?1). Methods New PFT parameters used to calibrate LPJ‐GUESS were based on modern pollen PFTs and remote sensed leaf area index (LAI). The model was validated using independent modern pollen assemblages, LAI and through comparison with White's modern potential vegetation map. Several scenarios were developed by combining changes in total rainfall amount with variation in the length of the dry season in order to test the sensitivity of African biomes. Results Simulated vegetation compared well to observed data at local and regional scales, in terms of ecosystem functioning (LAI), and composition (pollen and White's vegetation map). The assessment of the sensitivity of biomes to changes in precipitation showed that none of the ecosystems would shift towards a new type under the range of precipitation increases suggested by the IPCC (increases from 5 to 20%). However, deciduous and semi‐deciduous forests may be very sensitive to small reductions in both the amount and seasonality of precipitation. Main conclusions This version of LPJ‐GUESS parameterized for Africa simulated correctly the vegetation present over a wide precipitation gradient. The biome sensitivity assessment showed that, compared with savannas and grasslands, closed canopy forests may be more sensitive to change in precipitation regime due to the synergetic effects of changed rainfall amounts and seasonality on vegetation functioning.  相似文献

流域生态敏感性是流域生态系统遇到干扰时产生生态失衡与生态环境问题的难易程度和可能性大小,生物多样性是其影响最为重要的生态因子。主要考虑物种丰富度、珍稀程度、濒危程度、保护等级和生态系统类型等生物多样性敏感因子,借助GIS强大的空间数据采集和建模分析能力,在专家打分求取敏感因子权重基础上,通过空间模型计算生物多样性敏感度综合得分。基于二级流域综合得分最大值,采用ARCGIS自然间断点法实现敏感度分区,结果表明:极敏感区域主要分布在长江区的岷沱江、金沙江石鼓以下、金沙江石鼓以上、宜宾至宜昌、嘉陵江流域,珠江区的郁江、红柳河和西江流域,西南诸河区的澜沧江、红河和怒江及伊洛瓦底江流域,黄河区的龙羊峡以上流域等,这些区域水生生物特有种和受威胁物种丰度大,国家级自然保护区密集,是陆地生物多样性最丰富的地区。不敏感区域主要分布在松花江区除第二松花江以外流域,西北诸河区的塔里木盆地荒漠、古尔班通古特荒漠、中亚西亚内陆河、塔里木河干流等荒漠区,松花江区低温高寒,具有大森林、大草原、大湿地、大农田和大水域的特点,库坝工程对其生物多样性产生影响的可能性较小,西北诸河的荒漠区生境严酷,生物多样性贫乏,也不具备修建大型水库的条件,生物多样性敏感度也较低。其它区域介于二者之间,因其所处的生态系统类型、物种丰富度、珍稀程度、濒危程度和保护等级不一样,生物多样性敏感度各异。对流域库坝工程产生的生物多样性敏感度进行辨识,可为未来流域水资源合理和适度开发提供科学依据。  相似文献

生态保护重要性是表征区域生态系统结构和功能重要性程度的综合指标,确定生态保护空间范围是协调保护与发展、保障生态服务持续供给的基础。在明确福建省生态本底和关键生态问题的基础上,综合分析生态服务功能重要性和生态敏感性,构建福建省生态保护重要性评价指标体系,识别具有重要保护意义的生态极重要地区,能够为优化生态保护策略、划定生态红线和主体功能区划提供技术支持。研究结果表明,福建省生态保护极重要区占福建省陆地总面积的38.94%,生态重要性空间格局基本沿福建省闽西大山带、闽中大山带与海岸带分布,其中生态服务功能极重要区面积为3.96万km~2,以生物多样性维护功能和水源涵养功能为主;生态极敏感性区面积占福建省陆地总面积的9.71%,水土流失是主要的生态问题,占福建省陆地总面积的8.93%,土地沙化极敏感区集中在海岸带附近,与海岸侵蚀极敏感区空间范围基本一致。研究建议,生态保护极重要区作为划定生态红线和重点生态功能区的依据,支撑福建省开展有针对性的生态保护,保障和维护生态安全。  相似文献

Whereas with advancing age, peak heart rate (HR) and cardiac index (CI) are clearly reduced, peak stroke index (SI) may decrease, remain constant or even increase. The aim of this study was to describe the patterns of HR, SI, CI, arteriovenous difference in oxygen concentration (C a-vO2), mean arterial pressure (MAP), systemic vascular resistance index (SVRI), stroke work index (SWI) and mean systolic ejection rate index (MSERI) in two age groups (A: 20–30 years, n = 20; B: 50–60 years n = 20. After determination of pulmonary function, an incremental bicycle exercise test was performed, with standard gas-exchange measurements and SI assessment using electrical impedance cardiography. The following age-related changes were found: similar submaximal HR response to exercise in both groups and a higher peak HR in A than in B[185 (SD 9) vs 167 (SD 14) beats · min−1, P < 0.0005]; increase in SI with exercise up to 60–90 W and subsequent stabilization in both groups. As SI decreased towards the end of exercise in B, a higher peak SI was found in A [57.5 (SD 14.0) vs 43.6 (SD 7.7) ml · m−2, P < 0.0005]; similar submaximal CI response to exercise, higher peak CI in A [10.6 (SD 2.5) vs 7.2 (SD 1.3) l · min−1 · m−2, P < 0.0005]; no differences in C a-vO2 during exercise; higher MAP at all levels of exercise in B; higher SVRI at all levels of exercise in B; lower SWI in B after recovery; higher MSERI at all levels of exercise in A. The decrease in SI with advancing age would seem to be related to a decrease in myocardial contractility, which can no longer be compensated for by an increase in preload (as during submaximal exercise). Articular cartilage is optimised for bearing mechanical loads. Chondrocytes are the only cells present in mature cartilage and are responsible for the synthesis and integrity of the extracellular matrix. Appropriate joint loads stimulate chondrocytes to maintain healthy cartilage with a concrete protein composition according to loading demands. In contrast, inappropriate loads alter the composition of cartilage, leading to osteoarthritis (OA). Matrix metalloproteinases (MMPs) are involved in degradation of cartilage matrix components and have been implicated in OA, but their role in loading response is unclear. With this study, we aimed to elucidate the role of MMP-1 and MMP-3 in cartilage composition in response to mechanical load and to analyse the differences in aggrecan and type II collagen content in articular cartilage from maximum- and minimum-weight-bearing regions of human healthy and OA hips. In parallel, we analyse the apoptosis of chondrocytes in maximal and minimal load areas. Because human femoral heads are subjected to different loads at defined sites, both areas were obtained from the same hip and subsequently evaluated for differences in aggrecan, type II collagen, MMP-1, and MMP-3 content (enzyme-linked immunosorbent assay) and gene expression (real-time polymerase chain reaction) and for chondrocyte apoptosis (flow cytometry, bcl-2 Western blot, and mitochondrial membrane potential analysis). The results showed that the load reduced the MMP-1 and MMP-3 synthesis (p < 0.05) in healthy but not in OA cartilage. No significant differences between pressure areas were found for aggrecan and type II collagen gene expression levels. However, a trend toward significance, in the aggrecan/collagen II ratio, was found for healthy hips (p = 0.057) upon comparison of pressure areas (loaded areas > non-loaded areas). Moreover, compared with normal cartilage, OA cartilage showed a 10- to 20-fold lower ratio of aggrecan to type II collagen, suggesting that the balance between the major structural proteins is crucial to the integrity and function of the tissue. Alternatively, no differences in apoptosis levels between loading areas were found – evidence that mechanical load regulates cartilage matrix composition but does not affect chondrocyte viability. The results suggest that MMPs play a key role in regulating the balance of structural proteins of the articular cartilage matrix according to local mechanical demands.  相似文献

In this study, we investigated the causal relationship between chronic cold exposure and insulin resistance and the mechanisms of how DNA methylation and histone deacetylation regulate cold-reduced insulin resistance. 46 adult male mice from postnatal day 90–180 were randomly assigned to control group and cold-exposure group. Mice in cold-exposure group were placed at temperature from -1 to 4 °C for 30 days to mimic chronic cold environment. Then, fasting blood glucose, blood insulin level and insulin resistance index were measured with enzymatic methods. Immunofluorescent labeling was carried out to visualize the insulin receptor substrate 2 (IRS2), Obese receptor (Ob-R, a leptin receptor), voltage-dependent anion channel protein 1 (VDAC1), cytochrome C (cytC), 5-methylcytosine (5-mC) positive cells in hippocampal CA1 area. Furthermore, the expressions of some proteins mentioned above were detected with Western blot. The results showed: ① Chronic cold exposure could reduce the insulin resistance index (P < 0.01) and increase the number of IRS2 positive cells and Ob-R positive cells in hippocampus (P < 0.01). ② The expressions of mitochondrial energy-relative proteins, VDAC1 and cytC, were higher in cold-exposure group than in control group with both immunohistochemical staining and Western blot (P < 0.01). ③ Chronic cold exposure increased DNA methylation and histone deacetylation in the pyramidal cells of CA1 area and led to an increase in the expression of histone deacetylase 1 (HDAC1) and DNA methylation relative enzymes (P < 0.01). In conclusion, chronic cold exposure can improve insulin sensitivity, with the involvement of DNA methylation, histone deacetylation and the regulation of mitochondrial energy metabolism. These epigenetic modifications probably form the basic mechanism of cold-reduced insulin resistance.  相似文献

The sensitivity of S23 Lolium perenne L. to 11 parts 10–8SO2 was investigated at two different wind speeds. At the higherwind speed of 25 m min–8, SO2 caused significant reductionsin leaf area, root/shoot ratio, and all dry weight fractionsmeasured. At the lower wind speed of 10 m min–1 growthreductions were not found. The differences in sensitivity ofplants to SO2 at different wind speeds is discussed in relationto boundary layer resistances of leaves. It is concluded thatthe sensitivity of a plant to a particular pollutant shouldno longer be measured in terms of only the concentration andlength of exposure.  相似文献

Species of the harpacticoid copepod genus Microsetella are commonly reported to occur in Arctic and sub-Arctic coastal waters, but nothing has yet been reported on their sensitivity towards toxic substances. Effects of the polyaromatic hydrocarbon pyrene on Microsetella spp. from Western Greenland were investigated by looking at survival of females, feeding status, and nucleic acid content after a 96-h exposure. Less than 10% survived at 100 nM exposure concentration, and a 50% reduced survival was also evident at 0.1 and 10 nM. The RNA:DNA ratio was significantly higher, 2.1 ng/individual, at the lowest concentration of 0.01 nM compared to 0.9 ng/individual in the control, which indicates increased metabolic activities. A reduced DNA content in higher exposure concentrations of 1–100 nM suggests inhibition of egg development. Gut content was significantly declining with increasing exposure concentrations with only 2% of exposed females displaying full guts at 100 nM pyrene. Together with reduced feeding activity, effects at the population level can be anticipated, but would have to be confirmed in future experiments. The data suggests a higher sensitivity of Microsetella spp. compared to other Arctic copepods, which implies more severe effects from oil on the pelagic food web in the areas and periods where Microsetella spp. dominates Arctic plankton food webs.  相似文献
