When B10.D2 (H-2d) mice are immunized with lymphoid cells from C57B1/10 (H-2 d ) and their antisera tested against B10.A (H-2 a ) target cells, only antibodies to H-2.5 are measured. The same is true for immunization of DBA/2 (H-2 d ) mice when their antisera are absorbed with B10.D2 cells prior to testing. Irrespective of the dose of immunogen administered, the primary hemagglutinin response of B10.D2 mice is significantly lower than that of DBA/2 mice and (B10.D2 × DBA/2)F1 hybrids, but the secondary responses are similar. The low responsiveness of B10.D2 mice appears to be determined by a single dominant gene with incomplete penetrance; the gene is not linked to eitherH- 2, Hc, or the immunoglobulin allotype loci. In addition, the H-2.5 hemagglutinin response is susceptible to nongenetic influences. When antisera from B10.D2, devoid of H-2.5 hemagglutinins, were assayed in a complement-mediated cytotoxic test, they contained almost as much anti-H-2.5 activity as did the antisera from DBA/2 mice or (B10.D2 × DBA/2)F1 hybrids. The possibility is discussed that the locus responsible for the deficient primary hemagglutinin response of B 10.D2 may not be determinant-specific but may affect hemagglutinin responses in general.  相似文献

In a preceding report, the detection of an H-2-linked immune response to the H-X d antigen on the P815-X2 mastocytoma was demonstrated by the significantly increased survival of (C57BL/6 × DBA/2)F1 (B6D2F1) male hybrids (H-X b ) compared with female siblings (H-X b/H-X d ) after injection with the histocompatible tumor (H-X d ). This interpretation was supported by the absence of this sex effect in reciprocal D2B6F1 hybrids (H-X d and H-X d/H-X b ). Additional findings presented in this paper support the conclusion that this sex effect is due to a true immunological response to H-X d : (a) Reciprocal (DBA/2 × C57BL/6 H-2 mutant)F1 hybrids, as well as D2B6F1, failed to exhibit the sex effect: (b) the demonstration of the sex effect in (BALB/c × DBA/2)F1 and (BALB/c-H-2 dm2 × DBA/2)F1 hybrids and in (C57BL/10 × DBA/2)F1 hybrids was consistent with the known H-X incompatibilities between the strains BALB/c and DBA/2 and C57BL/10 and DBA/2, respectively, previously demonstrated by skin grafting; and (c) the sex effect was not abrogated by castration of male B6D2F1 hybrids. Variability in the presence or absence of the sex effect was observed in various [recombinant inbred (RI) × DBA/2]F1 hybrids and may be attributed to the influence of a regulatory non-H-2 gene which is closely linked to the gene coding for mouse kidney-androgen-regulated protein (KAP) but androgen-independent, or to variability in inheritance of the H-X b allele among the RI lines. It is proposed that the P815-X2 model may be utilized to type RI lines derived from a cross between C57BL/6 and DBA/2 for their H-X genotypes.Abbreviations B C57BL/6 origin allele - B6 C57BL/6 - B10 C57BL/10 - B6D2F1 (C57BL/6 × DBA/2)F1 - B6 m D2F1 (C57BL/6 H-2 mutant × DBA/2)F1 - bm10 B6.C-H-2 bm10 - C BALB/c - D DBA/2 origin allele - D2 DBA/2 - dm2 BALB/c-H-2 dm2 - H-X X chromosome-determined histocompatibility antigen of the mouse - Ir gene, immune response gene - KAP kidney androgenregulated protein - MST median survival time - RI recombinant inbred - SDP strain distribution pattern  相似文献

We have studied the influence of DBA/2 non-H-2 antigens on the lethal graft-versus-host reaction (GVHR) developed across an H-2 barrier. (DBA/2 x B10.D2)F1 x B10.D2 (H-2 d) backcross (BC) mice were typed for their allelic constitution at nine genetically independent chromosome markers and used as individual cell donors simultaneously for two to three (DBA/2 X B10.D2)F1 recipients incompatible for DBA/2 non-H-2 antigens alone and two to three (DBA/2 x B10.BR)F1 recipients incompatible for DBA/2 non-H-2 antigens and H-2k. The results showed that, when compared with that developed in a control group incompatible for H-2 kalone [B10.D2(B10.D2xB10.BR)F1], the GVHR mortality seen in the presence of an additional incompatibility for DBA/2 non-H-2 antigens [(DBA/2 X B10.BR)F1recipients] is significantly delayed but only in female mice. An analysis of individual BC donors indicated that this protective effect of DBA/2 non-H-2 antigens correlates with incompatibility for gene(s) linked to the Pgm-1 chromosome marker. In contrast, incompatibility for gene(s) linked to Mod-1 and Es-3 markers accelerates GVHR mortality, but only in male mice. Finally, the results obtained with (DBA/2 x B10.D2)F1 and (DBA/2 x B10.BR)F1 recipients were compared; they showed that the intensity of the GVHR developed by cells from individual BC donors against a given set of DBA/2 non-H-2 antigens correlates well with that developed by the same BC donor against the same set of non-H-2 antigens plus H-2k. We conclude that certain non-H-2 genes (and antigens) can modulate the intensity of the GVHR developed across an H-2 barrier. The number of such genes is probably great; their effects are strong and complex, and can be sex-dependent.  相似文献

Inbred strains of mice were studied for their susceptibility to the induction of experimental allergic orchitis after sensitization with mouse testicular homogenate in complete Freund's adjuvant accompanied by injections of extract from Bordetella pertussis. Susceptibility to autoimmune orchitis was found to be linked to the major histocompatibility complex in BALB/c and C57BL/10 mice and mapped to genes encoded within the H-2D dregion. In five of six groups of bidirectional (susceptible × resistant) F1 hybrids, H-2D d-linked susceptibility was inherited as a dominant autosomal trait. However, in (BALB/cByJ × DBA/2J)F1 and (DBA/2J × BALB/cByJ)F1 hybrids, dominant autosomal resistance to the induction of autoimmune orchitis was observed. Backcross analysis between the resistant F1 hybrid and the susceptible BALB/cByJ parent suggests that a single independently segregating DBA/2J locus is capable of negating H-2D d-linked susceptibility, and controls resistance to the induction of autoimmune orchitis.Abbreviations used in this paper BP extract Bordetella pertussis extract - CFA complete Freund's adjuvant - EAO experimental allergic orchitis - Ir immune response - MHC major histocompatibility complex - MLH mouse liver homogenate - MTH mouse testis homogenate - PI pathology index  相似文献

Summary Patterns of genetic control of hybrid resistance to the BALB/c plasmacytoma LPC-1 were studied for comparison with those to MPC-11, a plasmacytoma investigated previously. The overall patterns of hybrid resistance to the two tumors were similar, i.e., hybrids between BALB/c and BALB congenic resistant (CR) strains, A and A CR strains, SJL and DBA/2 were as susceptible to LPC-1 as BALB/c mice themselves, whereas hybrids between BALB/c and AKR, C57BL/Ks, DBA/1, C57BL/6 (B6), C57BL/10 (B10) and B10 CR strains were resistant to LPC-1 as previously shown with MPC-11. Heterozygosity within the H-2 complex alone was insufficient for resistance to either tumor. Among hybrids between BALB/c and the B10 CR strains, however, the presence of certain H-2 haplotypes influenced the degree of resistance seen and this H-2 effect was different for the two tumors. A sex effect on resistance to LPC-1, but not to MPC-11, was seen among F1 hybrids between BALB/c and DBA/1 although not in any other F1 hybrids. Among ((B10×BALB/c)F1×BALB/c) and (BALB/c×(B10×BALB/c)F1) and ((BALB/c×B10)F1×BALB/c) and ((BALB/c×B10)F1×BALB/c) backcross mice, however, significantly more males than females were resistant to LPC-1 and the results of this study are compatible with the idea that in F1 hybrids between BALB/c and B10, resistance to LPC-1 is controlled by two dominant autosomal genes, one of which is sex-limited and neither of which is linked to H-2. In contrast, hybrid resistance to MPC-11 in this cross is controlled by a single gene. Cross-protection experiments indicated that the two tumors share at least one tumor-associated transplantation antigen.  相似文献

TwoH-2 d mutants,H-2 dm2 (H-2L loss mutation) andH-2dm1 (gainplus-loss mutation involving bothH-2L andH-2D) were evaluated for any change in the immunogenicity of marrow stem cells. Grafts of 2 or 4 × 106 BALB/c(C) or BALB/c-H-2dm2 (C-H-2 dm 2) marrow cells were accepted by lethally irradiated B10.D2(H-2 d ) recipients and were rejected by irradiated B10(H-2 b ) recipients. Moreover, both (B6 × C)F1 and (B6 × C-H-2 dm 2)F1 mice, as irradiated recipients, resisted the growth of parental-strain B6(H-2 b ) marrow cells but accepted grafts from C or C-H-2 dm 2 parental-strain donors. Thus, theH-2 mutation involvingH-2L but notH-2D did not affect the expression ofH-2 d -associated Hemopoietic or Hybrid(Hh) antigens of marrow stem cells. Grafts of 2 to 8 × 106 B10.D2 or B10.D2-H-2 dm 1 marrow cells were rejected by B10.BR(H-2 k ) and B6 hosts and were accepted by B10.D2 hosts. However, B10.D2-H-2 dm 1 marrow cells grew to a much greater extent than B10.D2 cells in irradiated (B6 × B10.D2)F1 or (B6 × B10.D2-H-2 dm 1)F1 host mice. Therefore, theH-2 dm 1 mutation has altered the expression of Hh antigens at least quantitatively, resulting in a relative loss of hybrid resistance with the retention of Hh determinants recognized by allogeneic recipient mice which are notH-2 d . Since the Hh determinants of B10.D2 marrow cells have been mapped 16 cM to the right ofH-2, this mutation atH-2D/H-2L may have affected a regulatory gene.  相似文献

Factors influencing host resistance to the growth of a tumor bearing many mismatched minor histocompatibility antigens (MiHA) were studied. BALB/c (H-2d) and several of its F1 hybrids were injected intraperitoneally with DBA/2 (H-2d) P815 tumor cells. Compared to BALB/c, which was moderately susceptible, F1 hybrids of BALB/c with CBA, AKR, C3H.OH, and BIO H-2-congenic strains were highly susceptible, whereas hybrids of BALB/ c with A, A.SW, and BALB.B strains were quite resistant. Susceptibility was observed only with the intraperitoneally injected tumor, since both BALB/c and (CBA x BALB/c)F1 were resistant to the same tumor injected subcutaneously, and survival times of DBA/2 skin grafts did not differ between susceptible and resistant strains. Susceptibility was in part a function of the number of MiHA incompatibilities between tumor and host although the specific loci involved could not be identified. For example, susceptible (CBA x BALB/c)F1 hybrids probably shared certain MiHA with DBA/2 which BALB/c lacked, and which therefore subtracted from the net antigenic strength of the tumor in the hybrid, compared to its strength in BALB/ c. This interpretation was supported by in vitro studies which confirmed that the susceptible hybrids shared more MiHA with DBA/2, than did the resistant hybrids. Resistance was at least partially regulated by the host H-2 genotype, as shown by the observation that (BALB/ c x BALB.B)F1 (H-2d/b) mice were significantly more resistant than BALB/c. Segregation studies of the resistant (BALB/c x A)F1 hybrids, indicated that in addition to H-2, a nonH-2 gene in the A background was operating to confer resistance. Thus the factors influencing susceptibility to the MiHA-incompatible tumor were: (i) site of injection; (ii) the combined strength of the disparate MiHA; (iii) the host H-2 genotype; and (iv) at least one host nonH-2 gene conferring increased responsiveness.  相似文献

The reactivity of H-2b-restricted murine T cells towards bovine insulin was reported to depend on the expression of Ia.W39, a private specificity of I-Ab, on antigen-presenting cells. Cells of male (CBA/N x B6)F1 mice carrying the mutation xid on the X chromosome lack Ia.W39 on the cell surface. These cells are unable to present bovine insulin to primed T cells derived from female (CBA/N x B6)F1 mice. We show here that spleen cells of male (CBA/N x B6)F1 hybrids served perfectly as accessory cells for the insulin-dependent induction of a proliferative response of long-term cultured T cells with (B10 x B10.BR)F1 genotype, restricted to recognizing insulin in the context of F1-unique I-A determinants. The epitope on the insulin molecule essential for stimulation was determined to depend on the glutamic acid residue in position 4 of the A chain of insulin. This contrasts with the H-2b-restricted response of B6 mice to bovine insulin, which appears to be directed at the A chain loop determinant (amino acids A8 and A10). These data suggest that distinct I-Ab-encoded structures, the expression of which is regulated independently, may serve as components of restriction elements for H-2b and (H-2b x H-2k)F1 restricted T cells, which are specific for different epitopes of bovine insulin.  相似文献

Irradiation with ultraviolet B (UVB; 290–320 nm) initiates systemic immunosuppression of contact hypersensitivity (CHS). UV dose-responses for suppression of CHS to trinitrochlorobenzene were established in 18 strains of inbred mice. Three phenotypes with significantly different susceptibilities to UV suppression were identified. The phenotypes were: high (HI) susceptibility, 50% suppression with 0.7–2.3 kJ/m2 UV (C57BL/6, C57BL/10, and C57L and NZB females); low (LO) susceptibility, 50% suppression with 9.6–12.3 kJ/m2 UV (BALB/c, AKR, SJL and NZW), and intermediate (INT) susceptibility, 50% suppression with 4.7–6.9 kJ/m2 UV (DBA/2, C57BR, C3H/HeJ, C3H/HeN, CBA/N and A/J). UV suppression was not correlated with skin pigmentation or with the magnitude of the CHS response in non-irradiated animals. Major histocompatibility complex (MHC) haplotype was not correlated with UV suppression in MHC congenic strains B10.D2/oSnJ, B10.D2/nSnJ, B10.BR/SgSnJ, and A.BY/SnJ. There were no sex differences in UV suppression in BALB/c, C57BL/6, or NZW animals. In the autoimmune NZB strain, however, male mice (LO) were seven times less sensitive to UV suppression than NZB female mice (HI). Both sexes of (NZB × NZW)F1 and (NZW × NZB)F1 mice were HI, supporting dominance of HI over LO. Thus there are genetic factors and interacting sex-limited factors determining susceptibility to UV suppression. These findings may be of relevance to UV-related diseases such as photosensitive lupus and skin cancer. Correspondence to: F. P. Noonan.  相似文献

The differential expression of H-2 specificities recognized by antibody and by cytotoxic T lymphocytes (CTL) has been studied using a clone (FY7) of the C57BL/6 leukemia cell line FBL-3 (H-2 b /H-2 b ). Unlike C57BL/10 spleen cells, EL-4 lymphoma cells and Y57-2C leukemia cells (allH-2 b /H-2 b ), FY7 failed to induce the primary in vitro generation of anti-H-2b CTL by (B10.A x A)F1 (H-2 a /H-2 a or (B10.D2 x BALB/c)F1 (H-2 d /H-2 d ) responder spleen cells. In addition, FY7 was not lysed by, and did not competitively inhibit anti-H-2b CTL. Quantitative absorption tests with H-2Kb and H-2Db antisera revealed that FY7 expressed these antigens in quantitatively similar amounts to EL-4. The H-2Kb product of FY7 appeared to be identical with that of C57BL/10 spleen cells both in apparent molecular weight and isoelectric point. Yet FY7 failed to inhibit anti-H-2Kb CTL competitively in a cold target inhibition assay. Possible mechanisms are discussed for the lack of T-lymphocyte recognition of the H-2Kb-gene product expressed by FY7.Abbreviations used in this paper CTL cytotoxic T lymphocytes - MHC major histocompatibility complex - MLC mixed lymphocyte culture - PAGE polyacrylamide gel electrophoresis  相似文献

The ability to produce an autoimmune response to F antigen in mice is underH-2-linked and non-H- 2-linkedIr-gene control. There is an absolute requirement for ak allele atH-2K orI-A in order to produce anti-F antibodies. Low and high responsiveness is controlled by a non-H-2-linkedIr gene which behaves in a similar fashion toIr-3, in that as the dose of F-antigen is lowered, low responders behave as high responders and vice versa. This conversion from low to high responsiveness also occurs within a month after ATX.— Most F1 hybrids derived from (responder x nonresponder) parents bearing identical F-types behave as dominant nonresponders. As a result of ATX, such F1 mice convert to high responders. This conversion occurs if the animals are not immunized before day 90. If they receive F antigen prior to that time, they remain nonresponders for 7–9 months. One F1 combination — AKD2 — behaves as a dominant high responder. Genetic analysis showed that in the presence of ak allele atH-2K orI-A, a non-H-2-linkedIr gene inherited from the AKR mice determined dominant responsivenss. No manipulation of the immune response or combination of genes converted nonresponders lacking ak allele into responders. Such complex genetic control suggests regulation by a number of independently segregating loci whose function it is to limit the autoimmune response to F antigen.  相似文献

Alloantibodies specific for non-H-2 histocompatibility antigens of the mouse have been produced. Immunization (BALB/cJ×DBA/2J)F1 anti-B10.D2/n was conducted, followed by hemagglutination, immunofluorescence, and mixed hemabsorption tests on absorbed and unabsorbed sera. The results indicate that antibodies specific for H-3a and H-8a antigens are present. In addition, H-8a antigenic determinants were detected on erythrocyte membrane surfaces, as well as on cells of other body tissues.  相似文献

EA, i.e., antigen-antibody complexes are able to induce an antigen-nonspecific suppressive factor(s) from FcR+ B cells by binding on FcR. This factor, termed “suppressive B-cell factor (SBF)” was only effective on H-2 compatible, but not on H-2 incompatible spleen cells in an adoptive cell transfer system. Furthermore, SBF, prepared from B10.A (H-2a) splenic FcR+ B cells, suppressed the adoptive primary response of B10.D2 mice (H-2d), in addition to A/J mice (H-2a) against DNP-DE, by the pretreatment of cells with SBF in vitro. Absorption with affinity columns demonstrated that active components) of SBF from C3H/He mice (H-2k) was eliminated by both B6 anti-CBA (H-2b anti-H-2k) and B10.D2 anti-B10.BR (H-2d anti-H-2k), but not B10 anti-B10.A (H-2b anti-H-2a). In contrast, the suppressive activity of SBF was eliminated neither by anti-mouse Ig nor by a heat-aggregated human γ-globulin column. These results indicate that SBF contains a product coded by the right-hand side of H-2 gene complex, but does not contain Ig determinants nor FcR. Thus, it is conceivable that a compatibility of the right-hand side of H-2 gene complex is required for inducing effective suppression of spleen cells by SBF. SBF was considered to be a trypsin-resistant and heat-labile substance with a molecular weight of 30,000–63,000. The target cells for SBF were FcR? B precursors, but not helper T cells.  相似文献

The immune response to the liver protein F antigen which, in the mouse, occurs in two allelic forms, is under sharp immunogenetic control in that only mice that possess the Ak molecule can respond to allo-F antigen. This response has been studied in a number of F1 hybrids between inbred strains and with recombinant inbred lines all of which express Ak, and which thus enable immune suppression effects to be detected. In the AKXL and AKXD sets, the hybrids with CBA are responders if H-2 k/H-2k, and usually nonresponders if H-2 k/H-2b or H-2 k/H-2d. Although this may be due to gene dosage effects, this cannot be the explanation for the low responsiveness of the H-2 k/H-2b relative to the H-2 k/H-2d mice found in CBA × BXD hybrids. For this, and other reasons, it seems likely that low responsiveness in any mouse possessing a responder A k allele is due to suppression, and that this is mediated by the immune suppression effects of the non-H-2 k haplotype. These H-2-mediated effects can be modified, both positively and negatively, by background genes. Thus, of the ten H-2k/H-2d members of the CBA × AKXD cross, seven are low responders and three are high responders. No other typed marker has the same strain distribution pattern at present. Major unresolved questions, therefore, concern the location and mechanism of action of the background genes and the mechanism of action of the H-2 immune suppression genes.  相似文献

(C57B1 × DBA)F1 hybrids were more resistant to the inoculation of 103 Rauscher virus-induced, serially propagated RBL-5 cells than syngeneic C57Bl recipients. Resistance was linked toH-2di in the C57Bl backcross. Spleen cells from nonimmune (C57Bl × DBA)F1 hybrids were significantly more reactive against RBL-5 in the natural killer (NK) cytotoxicity test in vitro than were C57Bl spleen cells. In the C57Bl backcross,H-2dH 2b heterozygotes were more cytotoxic than theH- 2B homozygotes. Along with RBL-5, they were also more cytotoxic to the Moloney virus-induced YAC lymphoma (of strain A origin) and to the human ALL-derived MOLT-4 line.  相似文献

Hybrid resistance of lethally irradiated (C57BL/6 × DBA/2)F1 and (C57BL/10 × C3H)F1 hybrid mice to the engraftment of parental C57BL/6 or C57BL/10 bone marrow cells is controlled by the H-2-linked Hh-1 locus. This resistance can be specifically blocked or inhibited by the injection of irradiated spleen cells from lethally irradiated, marrow reconstituted donor mice of certain strains. By testing the ability of regenerating spleen cells from various donor strains to block the resistance, we studied the genetic requirements for the expression of putative cell-surface structures recognized in hybrid resistance to H-2b marrow cells. Strains of mice bearing informative intra-H-2 or H-2/ Qa-Tla recombinant haplotypes provided evidence that the Hh-1 locus is located telomeric to the H-2S region complement loci and centromeric to the H-2D region class I locus in the H-2 b chromosome. Two mutations that affect the class I H-2D b gene have no effect on Hh-1 b gene expression. The H-2D region of the H-2 S haplotype contains an allele of the Hh-1 locus indistinguishable from that of the H-2D b region, as judged by the phenotypes of relevant strains and F1 hybrids. Collectively these data indicate that the Hh-1 locus is distinct from the class I H-2D (L) locus in the H-2 b or H-2 s genome, and favor the view that the expression or recognition of the relevant determinants is not associated with class I gene products.Abbreviations used in this paper BM(C) bone marrow (cells) - CML cell-mediated lympholysis - CTL cytotoxic T lymphocytes - FBS fetal bovine serum - HBSS Hanks' balanced salt solution - SC spleen cells from irradiated, bone marrow-reconstituted mice Address correspondence to: Dr. I. A newH-2 mutant, BALB/c-H-2 db , is described. This mutant originated in BALB/c, is inbred, and is coisogenic with the parental BALB/cKh strain. The mutation is of the loss type since BALB/c-H- db rejects BALB/c, but not vice versa. Complementation studies have localized the mutation to theD region of theH-2 complex. A cross between BALB/c-H-2 db and B10.D2-H-2 da failed to complement for either BALB/c or B10.D2 skin grafts, indicating that these are two separate mutations at the same locus (Z2). Direct serological analysis and absorption studies revealed that, with one exception, theH-2 andIa specificities of BALB/c and BALB/c-H-2 db are identical. In particular,H-2.4, the H-2Dd private specificity, is quantitatively and qualitatively identical in the two strains. The exception is that of the specificities detected by antiserum D28b: (k×r)F1 anti-h, which contains anti-H-2.27, 28, and 29. These specificities appear to be absent from theH-2 db mutant since they are not detected directly or by absorption. Other public specificities are present in normal amounts,e.g., the reaction with antisera to H-2.3, 8, 13, 35, and 36. The reaction with antiserum D28 (f×k)F1 anti-s, which contains antibodies to H-2.28, 36, and 42, is the same in both strains. Antiserum made between the two strains (H-2 db anti-H-2 d ) reacts like an anti-H-2 serum, in that it reacts with both T and B cells by cytotoxicity, but is not a hemagglutinating antibody. The serum reacts as does the D28b serum in both strain distribution and in cross-absorption studies. We conclude that theH-2 db mutation occurred at a locus in theD region, resulting in the loss of the H-2.28 public serological specificity and of a histocompatibility antigen. Whether these are one and the same antigen is not yet known. The data, in view of other evidence, imply that the public and private specificities are coded for by separate genes.Abbreviations used in this paper are as follows CML cell-mediated lysis - MLR mixed lymphocyte reaction - GVHR graft-versus-host reaction - RFC rosette-forming cells - RAM-Ig rabbit anti-mouse IgG  相似文献

The specificity of an antiserum directed againstI region associated (Ia) antigens is described. The serum was raised in (DBA/1×B10.D2)F1 mice against lymphocytes of AQR mice, differing from the responder for theI region only. The serum reacts with Ia antigens expressed on B cells (Iab) as well as with Ia antigens expressed on T cells (Iat). Absorption studies indicate that B cells possess at least two Ia antigens, and one of these is shared by T cells. However, this shared antigen is not present on the surface of lymphocytes of thymectomized mice. Analysis of the strain distribution of Iab and Iat antigens revealed that the Iab antigens are present on lymphocytes of mice carrying theIA k subregion and that the Iat antigens are present on lymphocytes of mice carryingI region genes of theH-2 k haplotype located between theIA andIB subregions. This conclusion is based on the analysis of the antiserum's reactivity with T and B cells of the strains B10.A(2R), B10.A(4R) and B10.HTT: the serum reacts with B and T cells of B10.A(2R) but only with B cells of B10.A(4R) mice and only weakly with T cells of B10.HTT mice.Abbreviations ALG antimouse lymphocyte globulin from rabbits - B cells bone marrow derived lymphocytes - B10 C57BL/10Sn mice - D1D2F1 (DBA/1×B10.D2)F1 hybrid mice - GVHR graft-vs-host reaction - Ia I region associated antigen - Iab on B cells - Iat on T cells - MLR mixed lymphocyte reaction - T cells thymus-derived lymphocytes - Thy-1 thymus antigen 1, formerly called theta - Tx-Lyc lymphocytes of thymectomized, ALG treated, lethally irradiated and anti-Thy-1 treated bone marrow reconstituted mice - 2R B10.A(2R)/SgSn mice - 4R B10.A(4R) mice  相似文献

Experiments were conducted to answer the questions related to (a) the role played by the antigen-presenting cells (APCs) present within the thymus and (b) the effect of radiation dose to the recipients on the H-2 restriction profile of TNP-specific cytotoxic T lymphocyte precursors (CTLP) recovered from spleens and/or thymuses of H-2 incompatible radiation bone marrow chimeras (BMC). The H-2 restriction profile of intrathymically differentiating TNP-specific CTLPs was also analyzed in order to test an argument that donor-H-2 restricted CTLP detected in spleens of H-2 incompatible BMC were due to the extrathymically differentiated T cells under the influence of donor-derived lymphoreticular cells. The results indicated the following: (i) splenic T cells from B10(H-2b)→ (B10(H-2b) → B10.BR(H-2k)) chimeras, which were constructed by irradiating primary BIO → B10.BR chimeras with 1100 R and reconstituting them with donor-type (B10) bone marrow cells as long as 8 months after their construction, manifested restriction specificities for both donor- and host-type H-2, (ii) splenic T cells from two types of (B10 × B10.BR)F1→ B10 chimeras which were reconstituted after exposure of the recipients with either 900 or 1100 R with donor-type bone marrow cells generated both donor- and host-H-2 restricted TNP-specific cytotoxic T cells, and (iii) the TNP-specific CTLPs present in the regenerating thymuses of B10.BR → B10 and (B10 × B10.BR)F1→ B10 chimeras 4 weeks after their construction were also shown to manifest both donor- and host-H-2 restriction specificities. The significance of these findings on the H-2 restriction profile of CTLP generated in BMCs is discussed.  相似文献

Parabiosis of adult DBA/2J (H-2 d ) mice with adult (DBA/2J× CSH/HeJ)F1 (H-2 d /H-2 k ) mice results in survival beyond 100 days in 44% of such pairs, induction ofin situ unresponsiveness to C3H/HeJ skin, and the complete takeover of the erythroid system of the F1 by parental cells. However, in vitro responsiveness to C3H/HeJ cells remains. Dye exclusion cytotoxicity assays establish the absence ofF 1 lymphoid cells in the spleens and bone marrow of both partners. The parental takeover of the erythroid system of the F1 partner requires immune recognition of the hybrid's alloantigens, because this takeover is not seen with tolerant parental cells. PartialH-2 differences (on the C3H background) influence both survival and the takeover reaction when incorporated into parabioses with DBA/2J partners. When only theK andI subregions ofH-2 were targets of the parental response, 58% of parabionts survived, with complete parental hemopoietic takeover. When onlyH-2D was the target, 83% of parabionts survived, with incomplete hemopoietic takeover. Changing the non-H-2 background of the F1 target did not significantly affect survival or takeover, while substituting a differentH-2 d parental strain (BALB/c) eliminated survival altogether. Thus the parabiont takeover reaction encompasses the lymphoid and hemopoietic systems, requires immunorecognition of the target alloantigens, and seems to require a strongH-2 difference for its induction.  相似文献
