The structures of MornigaM and the MornigaM-mannose complex have been determined at 1.8 A and 2.0 A resolution, respectively. Both structures adopt the typical beta-prism motif found in other jacalin-related lectins and their tetrameric assembly closely resembles that of jacalin. The carbohydrate-binding cavity of MornigaM readily binds mannose. No major structural rearrangements can be observed in MornigaM upon binding of mannose. These results allow corroboration of the structure-function relationships within the small group of Moraceae lectins.  相似文献

The effect of 75% methanolic extract of the plant Phyllanthus amarus (P. amarus) was studied against cyclophosphamide (CTX) induced toxicity in mice. Administration of CTX (25 mg/kg b.wt, i.p.) for 14 days produced significant myelosuppression as seen from the decreased WBC count and bone marrow cellularity. Administration of P. amarus extract at doses 250 and 750 mg/kg b.wt significantly reduced the myelosuppression and improved the WBC count, bone marrow cellularity as well as the number of maturing monocytes. CTX treatment also reduced the activity of glutathione system and increased the activity of phase I enzyme that metabolize CTX to its toxic side products. P. amarus administration was found to decrease the activity of phase I enzyme. Administration of P. amarus also increased the cellular glutathione (GSH) and glutathione-S-transferase (GST), thereby decreasing the effect of toxic metabolites of CTX on the cells. Administration of P. amarus did not reduce the tumor reducing activity of CTX. In fact, there was a synergistic action of CTX and P. amarus in reducing the solid tumors in mice. Results indicated that administration of P. amarus can significantly reduce the toxic side effects of CTX and is not interfering with the antitumor efficiency of CTX.  相似文献

Changes in anatomical organisation of the leaf, photosynthetic performance and wood formation were examined to evaluate the temporal and spatial patterns of acclimatisation of micropropagated slow-growing black mulberry ( Morus nigra L.) plantlets to the ex vitro environment. Leaf structure differentiation, the rates of net photosynthesis (Pn), transpiration (E) and stomatal conductance (gs), and secondary xylem growth were determined in the course of a 56-day acclimatisation. Differentiation of palisade parenchyma was observed 7 days after transfer. At this stage, the rates of Pn, E and gs reached maximum values, after which the rates of all three gas exchange parameters gradually decreased. The highest proportion of woody area occupied by vessels was also observed 7 days after transfer. An important feature of developing woody tissue is the difference in patterns of vessel distribution from the characteristic differentiation patterns of earlywood and latewood vessels in mature wood of ring-porous trees. Vessels with lumen areas over 3000 μm2 were only differentiated in acclimatised plantlets, whereas vessels in stems sampled on days 0 and 7 had very small lumen areas of up to 560 μm2. Full acclimatisation, observed 56 days after transfer to the ex vitro environment, was associated with the rapid growth of new in vivo formed leaves, very low rates of E and gs, and much increased secondary xylem tissue within the stem area.  相似文献

Hepato-protective potential of carotenoid meso-zeaxanthin [(3R, 3'S)-beta, beta-carotene-3, 3'-diol] was studied using in vivo rat models. Paracetamol (3 g/kg body wt, orally), 20% ethanol (7.5 g/kg body wt, orally) and CCl4 (2.5 ml /kg, ip) were used as hepato toxins. Levels of marker enzymes of hepatic injury such as serum glutamate oxaloacetate transaminase, serum glutamate pyruvate transaminase and alkaline phosphatase, and serum bilirubin, which were drastically elevated by these hepato toxins were significantly decreased by meso-zeaxanthin pretreatment in a dose-dependent manner. Oxidative stress markers, tissue lipid peroxidation, conjugated dienes and tissue hydroperoxides, were high in the paracetamol treated control group animals, which were lowered by meso-zeaxanthin administration. Level of glutathione and antioxidant enzymes, superoxide dismutase, catalase and glutathione peroxidase, in liver tissue was increased by meso-zeaxanthin pretreatment compared to control group during alcohol and CCl4 induced hepatotoxicity. Hydroxyproline, an indicator of fibrosis in liver tissue, decreased remarkably by meso-zeaxanthin administration despite its notable elevation in ethanol treated rats. Histopathological analysis of liver tissue showed the hepatoprotective potential of meso-zeaxanthin.  相似文献

HgCl2 (5.0 mg/kg body weight) induced toxicity led to significant elevation of lipid peroxidation (LPO) level but decline in the glutathione content in liver of Swiss albino mice. In serum of HgCl2 treated mice there was significant elevation in serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) activities but significant decline in the alkaline phosphatase activity. Animals treated with O. sanctum extract (10 mg/kg body weight, po) before and after mercury intoxication showed a significant decrease in LPO level, SGOT and SGPT activities and increase in serum alkaline phosphatase activity and glutathione (GSH) content. Ocimum treatment alone did not alter SGOT, SGPT and alkaline phosphatase activities but significantly enhanced reduced glutathione. The results suggest that oral administration of Ocimum extract provides protection against HgCl2 induced toxicity in Swiss albino mice.  相似文献

Doxorubicin (DOX), a potent chemotherapeutic agent, is widely used for the treatment of various malignancies. However, its clinical uses are limited due to its dose-dependent adverse effects particularly cardiac and testicular toxicities. DOX-induced toxicity is mainly due to the induction of oxidative stress. Atorvastatin (ATV), a 3-hydroxy 3-methyl glutaryl coenzyme A reductase inhibitor, with lipid-lowering activity, acts as an antioxidant at lower doses. It possesses pleiotropic effects independent of cholesterol-lowering property usually shown at lower doses, which include antioxidant and anti-inflammatory activities. The present study was aimed to investigate the possible protection exerted by atorvastatin against oxidative stress and DNA damage induced by DOX in the heart and testes of mice. The protective role of ATV in the heart and testes of DOX-treated mice was evident from the amelioration of oxidative stress, DNA and cellular damage. The present study clearly indicates that ATV offers a significant protection against DOX-induced oxidative stress and DNA damage in the heart and testes of mice.  相似文献

In mulberry ( Morus alba L. cv. Shin-ichinose), shoot-tip abscission following the cessation of apical growth could be induced in different internodes, depending on the vigour of the shoot and its apex and other internal and external factors. In the lateral, short shoots of 1-year-old stems of low-pruned trees, the apical growth cessation and shoot-tip abscission (May–June) resulted primarily from the dominance of the upper, long shoots and intense competition among laterals along the stem. Decapitation of the laterals, before abortion of their apices took place (early May), readily caused adventitious abscission of the distal internode. Similar decapitation-induced, adventitious abscission of the distal internode of the upper, long shoots of 1-year-old stems of pruned trees also occurred (May–September), demonstrating that the abscission itself is not directly associated with photoperiod. In May and June, decapitation induced abscission primarily in parallel with or after sprouting of lateral buds and shoot elongation, while in July, August and September, the abscission was induced by decapitation and independently of sprouting. Shoot (stem) orientation positively affected the abscission, which is related to gravimorphic effects on buds and shoots on the lower and lateral sides of the horizontally trained stem. These results suggest that the vigour of shoots and apices is an important determinant of growth and apex abscission in mulberry.  相似文献

Patterns of spring development of lateral buds of mulberry (Morus alba L. cv. Shin-ichinose) coppice shoots on 11-year-old low-pruned stumps varied in response to girdling, pruning and arching. The erect controls showed a weak acrotonic (apex-favoring) growth habit, in which the majority of the buds, including the basal ones, sprouted and elongated in mid- and late April, and hence there was a prolonged imposition of dominance on the upper laterals in mid- and late May. In contrast, early spring girdling or pruning enhanced the activity of the upper buds of the proximal (lower) halves of the girdled stems or of the pruned stems, resulting in considerable dominance of the laterals from such buds in late April. Arching markedly inhibited buds on the under side of the arched stems, leading to poor shoots. By late April, the buds on the adaxial (upper) side readily grew into new vertical shoots, which dominated over the lateral ones. When studied by a multiple-node-cutting test, increased length of segments of post-dormant mulberry stems was accompanied by decreased bud activity of the segments and by decreased breaking ability of the lower buds within the segments, suggesting the importance of roots in the weak acrotonic habit of the erect stem in spring. By contrast, the acropetal influences of the attached stems can in part affect dominance relationships, perhaps mediated through competition for factors translocated from the roots. Continuous basal applications of abscisic acid inhibited bud break and shoot growth of the postdormant stem segments, but these inhibitory effects could be reversed by applied gibberellic acid A3 (GA3). Two phases of lateral bud dormancy in erect mulberry coppice shoots were identified. The first was characterized by a smaller breaking capacity in the upper buds than in the lower ones and hence by a basitonic (base-favoring) gradient in bud growth potential. The second phase corresponded to a restoration of these capabilities in the upper buds and to a change towards a linear gradient in bud growth potential, with disappearance of the dormant condition, in February and March. This gradient change during dormancy release may represent the physiological basis for the weak acrotonic habit of erect mulberry stems in spring.  相似文献

 A continuously growing callus was obtained from immature endosperm of Morus alba L Cv S-36 cultured on Murashige and Skoog medium containing 5 μm 2,4-dichlorophenoxyacetic acid. Shoot buds were produced when the callus was subcultured on a medium containing a cytokinin or a cytokinin and 1-naphthaleneacetic acid (NAA). The maximum number of shoots was formed on the medium containing thidiazuron (1 μM), or benzylaminopurine (5 μM) and NAA (1 μM). Shoots were multiplied by forced axillary branching and rooted in vitro. Endosperm-derived plants were established in soil. Each of the ten plants examined cytologically was triploid (3 n=42). Renal and hepatic subacute toxicity induced by the antihyperlipidaemic drugs: Bezalip-Pravastatin and Lopid was investigated in rats using serum biochemical parameters. Toxicological evaluation was performed in serum samples following the administration of the therapeutic dose regimens of the compounds that were previously shown to be effective in inhibition of 3-hydroxy-methylglutaryl coenzyme A (HMG CoA) reductase, the enzyme controlling the rate-limiting step in the synthesis of cholesterol, and acyl-CoA cholesterol acyl transferase (ACAT) which converts intracellular free cholesterol to cholesterol ester. Renal and hepatic subacute toxicity was evaluated by measuring enzyme activity or concentrations of: alanine aminotransferace, alkaline phosphatase, aspartate aminotransferase, gamma-glutamyltransferase, glucose, potassium, sodium, blood urea nitrogen, uric acid and creatinine. The use of the above serum biochemical parameters indicated that the overall toxicity impact of antihyperlipidaemic drugs was Bezalip = Pravastatin < Lopid. We have found that the Pravastatin--in contrast to the above antihyperlipidaemic drugs--only transiently affects the biochemical parameters associated with toxicity, but, it affects some of the biochemical parameters associated with hepatic and renal toxicity, up to a significantly lower extent than the antihyperlipidaemic drugs.  相似文献

The disease response and magnitude of genetic variability of 85 mulberry genotypes of different agroclimatic origin was studied against powdery mildew caused by Phyllactinia corylea. It was observed that there was a wide variation of disease severity among the test genotypes. Australian and France originating genotypes were found to be highly resistant to mildew followed by of Thai and Italian origin. Genotype wise, the lowest mildew disease severity was recorded in Thailand [lobed]) followed by M. malticaulis, M. australis and Italian. Genetic analysis of disease severity revealed that the estimate of phenotypic coefficient of variation (PCV) and genotypic coefficient of variation (GCV) were high and that PCV was greater than GCV. High estimate of heritability coupled with high genetic advance showed that the mildew disease resistant trait is governed by an additive gene action. Hence the highly resistant mulberry genotypes identified may be exploited through hybridisation followed by selection under epiphytotic conditions for the improvement of disease resistant traits in mulberry.  相似文献

Measurements of changes in the degree of dominance by upper laterals over lower ones in coppice shoots (1-year-old stems) of 12-year old low- pruned stumps of mulberry ( Morus alba L. cv. Shin-ichinose) were made by removal of upper stem sections (pruning) or of lateral buds (debudding.) before spring bud burst, as part of a study of the factors involved in dominance relationships between the developing buds and elongating shoots. Besides inhibition of lower laterals by the upper, leading shoots, there was evidence for mutual inhibition (competition) of neighboring laterals along the stem. Thus in stems in which every other bud, or 4 out of every 5 buds were removed, there was a delay in growth cessation of lower laterals and their greater elongation than in controls. Such competition was seen to exist even between the uppermost and sub-terminal laterals, since the former elongated more in the absence of the latter.
In contrast to high and middle pruned stems, the delay in sprouting of the buds in low-pruned stems resulted in limited elongation of the shoots from such buds. This inhibition was removed when all the stems on a stump were pruned to the same length, suggesting that it was associated with intact stems with actively growing laterals. Patterns of regrowth of the short shoots (lower laterals) after summer pruning (middle-pruned) depended on the extent of removal of other stems with vigorously growing, upper laterals. These results demonstrate that both acropetal and basipetal influences are important in bud and shoot dominance relationships.  相似文献

Liver injury was induced in female rats using tamoxifen (TAM). Grape seeds (Vitis vinifera) extract (GSE), black seed (Nigella sativa) extract (NSE), curcumin (CUR) or silymarin (SYL) were orally administered to TAM-intoxicated rats. Liver histopathology of TAM-intoxicated:rats showed pathological changes. TAM-intoxication elicited declines in liver antioxidant enzymes levels (glutathione peroxidase, glutathione reductase, superoxide dismutase and catalase), reduced glutathione (GSH) and GSH/GSSG ratio plus the hepatic elevations in lipid peroxides, oxidized glutathione (GSSG), tumor necrosis factor-alpha (TNF-alpha) and serum liver enzymes; alanine transaminase, aspartate transaminase, alkaline phosphatase, lactate dehydrogenase and gamma glutamyl transferase levels. Oral intake of NSE, GSE, CUR or SYL to TAM-intoxicated rats, attenuated histopathological changes and corrected all parameters mentioned above. Improvements were prominent in case of NSE (similarly SYL) > CUR > GSE. Data indicated that NSE, GSE or CUR act as free radicals scavengers and protect TAM-induced liver injury in rats.  相似文献

Genetic characterization of germplasm resources is necessary for their effective management and efficient utilization, especially for species like mulberry in which the available germplasm exhibits rich phenotypic diversity with almost no information about its genetic base. Here we present the first report on the isolation of six novel microsatellite markers of mulberry, developed from an enriched genomic library of Morus indica. These markers revealed a high degree of polymorphism (14–26 alleles per locus; polymorphic information content, 0.85–0.90) and a broad cross‐species affinity when tested on a set of 43 elite genotypes including 13 related Morus species. The data thus demonstrate their utility as potentially efficient genetic markers for germplasm characterization, crop improvement and molecular systematics of mulberry.  相似文献

Steroid binding has been studied in cytoplasmic extracts of normal mouse kidney, an androgen sensitive organ, and of kidney from mice affected with testicular feminization (Tfm mutant) that have inherited androgen resistance. Macromolecules that bind 3H-5α-dihydrotestosterone (DHT, the presumed active androgen in most testosterone target organs) and sediment in glycerol gradient at 8–9S can be observed in cytosol from kidney of mice of different sex, age, and hormonal history. The 8–9S component from normal females is heat labile, pronase sensitive, and dissociated by high salt to a lower molecular weight entity. The apparent equilibrium dissociation constant (Kd) for the DHT-receptor complex is 1.4 × 10?9M, and there are about 1500 binding sites per testosterone-sensitive kidney proximal tubule cell. Cytosol from Tfm/Y animals also shows a sharp peak of 3H-DHT-binding activity at 8–9S. The Tfm protein, however, has reduced affinity for DHT and binds only 10–25% as much 3H-DHT as wild-type receptor at 3H-DHT concentrations from 5 × 10?11M to 1.2 × 10?8M. Scatchard analysis, and studies involving competition with unlabeled steroids, relative binding of various androgens, and dissociation of the 3H-DHT-binding protein complex after extensive dialysis have led to the conclusion that Tfm kidney contains very little, if any, androgen receptor with properties similar to that found in normal kidney.  相似文献

Anthers of Morus indica L., with microspores at the uninucleate stage were cultured; and the influence of temperature and kinetin pretreatment on induction of androgenic calluses was examined. The effects of various pretreatments revealed that 24 h cold pretreatment increased the percentage of cultures inducing callus. First microspore division was observed after 16 to 20 days of culture. Th anthers split and developed embryogenic calluses on MB medium supplemented with NAA (0.5 mg l–1 and BA (1.0 mg l–1)) using 8% sucrose. Rhizogenesis was induced on medium supplemented with NAA and BA (each 0.5 mg l–1) with reduced myo-inositol (75 mg l–1). Cytological study of induced roots confirmed the haploid nature of calluses. Different type of embryos were initiated upon transfer of calluses to medium supplemented with NAA, BA (each 0.5 mg l–1), 2,4-d (1.0 mg l–1) and PVP (600 mg l–1). Abstract

The study reports the characterization of 10 isolates of mulberry black root rot causing fungus, Botryodiplodia theobromae obtained from the infected gardens of Karnataka, Andhra Pradesh and Tamil Nadu. The analysis based on cultural, morphological, pathogenicity and molecular markers (RAPD and SSRs) revealed significant variations among the isolates. Based on the disease reaction on susceptible V-1 variety, isolates were grouped as pathogenic (60%), moderate pathogenic (20%) and non-pathogenic (20%). Among all isolates, RAPDs revealed higher marker polymorphism however, based on Shannon’s Information Index (I) SSRs were more informative (0.781) compared to the former (0.444). Stepwise Multiple Regression Analysis (MRA) indicated a total of 5, 5 and 3 molecular markers were found to correlate with disease symptoms. Screening of germplasm using multiple strains of virulent isolates will enhance possibilities of locating diverse resistant genes. Pyramiding of these genes will aid in development of mulberry variety with durable resistance and sustainable sericulture.  相似文献

肌动蛋白在植物的各种生理活动中起着重要作用,是研究基因表达与调控模式的内标参考.通过染色体步移方法获得了桑树肌动蛋白actin基因1 612 bp的序列,该基因CDS长1 312 bp(GenBank登录号:HM623866),编码377个氨基酸残基,与水稻、葡萄等的同源基因的序列一致性在90%以上.基因内含子的数目及其在基因组上的位置也与水稻、葡萄等的相似.对来自不同物种的24个肌动蛋白基因进行聚类分析的结果显示,基因被分为Class Ⅰ和ClassⅡ两个明显的亚群.  相似文献
