Alkylated chitosans (ACSs) were prepared by modifying chitosan (CS) with alkyl bromide. The self-aggregation of ACSs in acetic acid solution was characterized by fluorescence spectroscopy and dynamic light scattering method. The results indicate that introducing alkyl side chains leads to the self-aggregation of ACSs, and CS with a 99% deacetylation degree shows no aggregation due to the electrostatic repulsion. The electrophoresis experiment demonstrates that the complex between CS and DNA was formed at a charge ratio (+/-) of 1/1; ACS/DNA complexes were formed at a lower charge ratio (+/-) of 1/4. A small amount of alkylated chitosans play the same shielding role as chitosan in protecting DNA from DNase hydrolysis. Differential scanning calorimetry (DSC) and atomic force microscopy (AFM) were employed separately to investigate the thermodynamic behavior of dipalmitoyl-sn-glycero-3-phosphocholine (DPPC)/CS and DPPC/ACS mixtures and the variation in topological structure of DPPC membrane induced by CS and ACS. It is shown that CS and ACS can cause the fusion of DPPC multilamellar vesicles as well as membrane destabilization. In contrast, the perturbation effect induced by ACS is more evident due to the hydrophobic interaction. CS and ACS were used to transfer plasmid-encoding CAT into C(2)C(12) cell lines. Upon elongating the alkyl side chain, the transfection efficiency is increased and levels off after the number of carbons in the side chain exceeds 8. It is proposed that the higher transfection efficiency of ACS is attributed to the increasing entry into cells facilitated by hydrophobic interactions and easier unpacking of DNA from ACS carriers due to the weakening of electrostatic attractions between DNA and ACS. 相似文献

With few exceptions, the evolutionary consequences of harmful algae to grazers in aquatic systems remain unexplored. To examine both the ecological and evolutionary consequences of harmful algae on marine zooplankton, we used a two-fold approach. In the first approach, we examined the life history responses of two geographically separate Acartia hudsonica (Copepoda Calanoida) populations reared on diets containing the toxic dinoflagellate Alexandrium fundyense . One copepod population was from a region, Casco Bay, Maine, USA, that has experienced recurrent blooms of highly toxic Alexandrium spp. for decades; whereas the other population from Great Bay, New Jersey, USA, has never been exposed to toxic Alexandrium blooms. The life history experiment demonstrated that when the copepod population from New Jersey was reared on a diet containing toxic A. fundyense it exhibited lower somatic growth, size at maturity, egg production and survival than the same population reared on a diet without toxic A. fundyense . In contrast, toxic A. fundyense did not affect the life-history traits of the Maine population. Fitness, finite population growth rate (), was significantly reduced in the New Jersey population, but not in the Maine population. These results are consistent with the hypothesis of local adaptation (resistance) of the historically exposed copepod population to the toxic dinoflagellate. In the second approach, we further tested the resistance hypothesis with a laboratory genetic selection experiment with the naïve New Jersey copepod population exposed to a diet containing toxic A. fundyense. This experiment demonstrated that the ingestion and egg production of adult females of naïve copepods fed A. fundyense improved after three generations of being reared on a diet containing the toxic dinoflagellate. The results of the present study have important implications for understanding how grazer populations may respond to the introduction of toxic algae to their environment, and suggest that grazer resistance may be a feedback mechanism that may lead to bloom control.Co-ordinating editor: Hurst 相似文献

A technique was developed for the semi-mass culture ofthe unarmored dinoflagellate, Gymnodiniumsplendens under laboratory conditions. A maximumcell density of 4600 to 6800 cells ml–1 was observedwithin 8 to 11 days of culture. An initial feedingtest for 8 days with three important marine finfishlarvae showed that red spotted grouper, Epinephelus akaara preferred G. splendensfed 200 cells ml–1 with 44% survival. The Japanesestripe knife jaw, Oplegnathus fasciatus,attained 22% survival fed a combination of G. splendensand rotifers (200 cells ml–1 and 5ind. ml–1, respectively). Red sea bream, Pagrusmajor larvae did not respond well to the initialfeeding of G. splendens alone. Red seabream were observed to be solely dependent on rotifers(5 ind. ml–1) as initial food. Gymnodiniumsplendens may be used as a live food in the initialfeeding of red spotted grouper larvae (E. akaara) toreduce mortality and to further enhancegrowth during the critical first few days ofrearing. 相似文献

Recent work has shown that benthic invertebrate assemblages may be influenced in an ongoing fashion by dispersal. Water-column movements of meiofauna, juvenile insects and marine postlarvae are common and can act to alter greatly local dynamics such as predator-prey and competitive interactions in marine and stream ecosystems. These findings are important because past research on the role of dispersal in invertebrate dynamics has focused almost exclusively on how planktonic larval supply influences the establishment and maintenance of local assemblages, on the colonization of newly opened sites, or on the settlement success of new recruits. The emerging framework is that dispersal needs to be viewed as a regional process that may routinely influence local benthic dynamics, because fauna can move to and from water-column dispersal 'pools' and may do so at frequent intervals. 相似文献

Blooms caused by some species belonging to the dinoflagellate genus Alexandrium are known to cause large-scale mortality of fish. Thus, the dynamics of these species is important and of concern to scientists, officials, and people in the aquaculture industry. To understand the dynamics of such species, their growth and mortality due to predation need to be assessed. The newly described dinoflagellate Alexandrium pohangense is known to grow slowly, with a maximum autotrophic growth rate of 0.1 d−1. Thus, it may not form bloom patches if its mortality due to predation is high. Therefore, to explore the mortality of A. pohangense due to predation, feeding on this species by the common heterotrophic dinoflagellates Gyrodinium dominans, Gyrodinium moestrupii, Luciella masanensis, Noctiluca scintillans, Oxyrrhis marina, Oblea rotunda, Polykrikos kofoidii, and Pfiesteria piscicida, as well as by the ciliate Tiarina fusus, was examined. None of these potential predators was able to feed on A. pohangense. In contrast, these potential predators were killed and their bodies were dissolved when incubated with A. pohangense cells or cell-free culture filtrates. The survival of G. moestrupii, O. marina, P. kofoidii, and T. fusus on incubation with 10 cells ml−1 of A. pohangense was 20–60%, while that at the equivalent culture filtrates was 20–70%. With increasing A. pohangense cell-concentration (up to 1000 cells ml−1 or equivalent culture filtrates), the survival rate of G. moestrupii, O. marina, P. kofoidii, and T. fusus rapidly decreased. The lethal concentration (LC50) for G. moestrupii, O. marina, P. kofoidii, and T. fusus at the elapsed time of 24 h with A. pohangense cells (cultures of 11.4, 13.3, 1.6, and 3.3 cells ml−1, respectively) was lower than that with A. pohangense filtrates (culture filtrates of 35.5, 30.6, 5.5, and 5.0 cells ml−1, respectively). Furthermore, most of the ciliates and heterotrophic dinoflagellates in the water collected from the coast of Tongyoung, Korea, were killed when incubated with cultures of 1000 A. pohangense cells ml−1 and equivalent culture filtrates. Shoe soles are possible vectors for infectious diseases. Although studies have been performed to assess the prevalence of infectious pathogens on shoe soles and decontamination techniques, no systematic review has ever occurred. The aim of this study was to perform a systematic review of the literature to determine the prevalence of infectious agents on shoe bottoms and possible decontamination strategies. Three electronic bibliographic databases were searched using a predefined search strategy evaluating prevalence of infectious pathogens on shoe bottoms and decontamination strategies. Quality assessment was performed independently by two reviews with disagreements resolved by consensus. Thirteen studies were identified that supported the hypothesis that shoe soles are a vector for infectious pathogens. Methicillin‐resistant Staphylococcus aureus, Clostridium difficile and multidrug‐resistant Gram‐negative species among other pathogens were documented on shoe bottoms in the health care setting, in the community and among food workers. Fifteen studies were identified that investigated decontamination strategies for shoe soles. A number of decontamination strategies have been studied of which none have been shown to be consistently successful at disinfecting shoe soles. In conclusion, a high prevalence of microbiological pathogens was identified from shoe soles studied in the health care, community and animal worker setting. An effective decontamination strategy for shoe soles was not identified. Studies are needed to assess the potential for contaminated shoes to contribute to the transmission of infectious pathogens. 相似文献

The Mediterranean fruit fly (Ceratitis capitata) is a cosmopolitan pest of hundreds of species of commercial and wild fruits. It is considered a major economic pest of commercial fruits in the world. Adult Mediterranean fruit flies feed on all sorts of protein sources, including animal excreta, in order to develop eggs. After reaching sexual maturity and copulating, female flies lay eggs in fruit by puncturing the skin with their ovipositors and injecting batches of eggs into the wounds. In view of the increase in food-borne illnesses associated with consumption of fresh produce and unpasteurized fruit juices, we investigated the potential of Mediterranean fruit fly to serve as a vector for transmission of human pathogens to fruits. Addition of green fluorescent protein (GFP)-tagged Escherichia coli to a Mediterranean fruit fly feeding solution resulted in a dose-dependent increase in the fly's bacterial load. Flies exposed to fecal material enriched with GFP-tagged E. coli were similarly contaminated and were capable of transmitting E. coli to intact apples in a cage model system. Washing contaminated apples with tap water did not eliminate the E. coli. Flies inoculated with E. coli harbored the bacteria for up to 7 days following contamination. Fluorescence microscopy demonstrated that the majority of fluorescent bacteria were confined along the pseudotrachea in the labelum edge of the fly proboscis. Wild flies captured at various geographic locations were found to carry coliforms, and in some cases presumptive identification of E. coli was made. These findings support the hypothesis that the common Mediterranean fruit fly is a potential vector of human pathogens to fruits. 相似文献

In this study we tested the hypothesis that after administration of a single intraperitoneal dose of concanavalin A (Con-A) to mice, the proportion of neutrophils and macrophages in the peritoneal exudate and their phagocytic and candidacidal activities should change with time. The number of neutrophils in the peritoneal exudate was greatly increased 6 h after administration of Con-A, and those cells were able to kill both intracellular and extracellular yeast and germ tube forms of Candida albicans. Addition of catalase to the culture medium reduced the killing of C. albicans, suggesting that the candidacidal activity depended on the myeloperoxidase system. The survival of mice pretreated with Con-A and submitted to an inoculum of C. albicans 6 h afterwards was twice higher than that of controls, which suggests that neutrophils were able to clear the experimental infection. One day after the treatment, the population of neutrophils in the exudate was about 45%, but after 2 days it was reduced to only 5% and the candidacidal activity was also reduced. After 4 days the exudate contained over 95% of macrophages, the candidacidal activity reached a maximum, and the phagocytosis mediated by both complement receptors and mannose receptors was increased. Uptake of FITC-mannose-BSA by macrophages was maximal on about the 4th day and was inhibited by mannan, suggesting that treatment with Con-A increased the activity of mannose receptors. These results support the hypothesis that activation of cellular immunity by Con-A occurred in two phases, one dominated by neutrophils, and the other by macrophages expressing increased activity of mannose receptors. 相似文献

Summary Within 24 h after the initial phagocytotic uptake of freshly isolated (from host tissue) symbiotic algae (Symbiodinium microadriaticum) by the endodermal cells of the polyp (scyphistoma) stage of the jellyfish Cassiopeia xamachana, the algal population was observed to decline despite evidence of algal cell division. Analyses of the frequency of phago-lysosome fusion as an indicator of possible attempts of the host to digest the algae indicated that, although phago-lysosome fusion did occur, the low frequency of occurrence is inconsistent with the interpretation that the animals digested the algae. Animal cell lysosomes were located predominantly at the apices of the endodermal cells, and the symbiotic algae were transported toward the bases of the endodermal cells.Within 3 days after initial infection, most endodermal cells with algae ceased to be phagocytotically active (with respect to the uptake of carmine particles). Many of these endodermal cells soon migrated into the mesoglea to become what are traditionally referred to as amoebocytes. Within amoebocytes the algae proliferated. The onset of strobilation by the scyphistomae was directly correlated with the increase in the algal population within these amoebocytes. 相似文献

[目的]以生物导弹为理念,应用寄生性昆虫携带病原物双重防治松墨天牛Monochamus alternatus是目前研究的新领域,更是松材线虫病Bursaphelenchus xylophilus防治的新途径.[方法]本文以川硬皮肿腿蜂Scleroderma sichuanensis为载体,通过测定4株松墨天牛致病菌的孢子粉和孢子悬浮剂对其毒力大小和川硬皮肿腿蜂对4株菌的2种剂型的初始携带量及存留量,分析不同菌种与不同剂型对川硬皮肿腿蜂的影响和川硬皮肿腿蜂体表孢子脱落趋势.[结果]川硬皮肿腿蜂雌成蜂携带粉剂孢子的死亡率高于携带悬浮剂的,但川硬皮肿腿蜂对粉剂孢子的携带量却远远高于悬浮剂孢子的携带量.当喷施孢子悬浮剂时,川硬皮肿腿蜂的累计死亡率随浓度增加而增加,同样携带量也随浓度增加而增加;白僵菌对川硬皮肿腿蜂的毒力较绿僵菌高,致死速率也较绿僵菌大,但川硬皮肿腿蜂对白僵菌的携带效果却比绿僵菌的效果好.并且不论剂型和菌种,川硬皮肿腿蜂体表的孢子在第1d后都脱落最为明显,而后均保持较缓慢脱落.[结论]本研究证明川硬皮肿腿蜂携带松墨天牛致病菌的方法是可行的,其中白僵菌孢子粉更适合携带. 相似文献
