对分离自山羊瘤胃的真菌分离培养液中甲烷菌进行16SrDNA扩增、DGGE分析、RFLP及测序分析,研究共存于真菌分离培养液中甲烷菌的种类及其多样性。DGGE结果显示:从厌氧真菌分离至第45代,甲烷菌多样性指数由1·32降至0·99,相似性最低为34·7%;第45代至62代,多样性指数由0·99升至1·15,相似性最低为89·2%。RFLP多态性分析69个克隆共得到5个操作分类单元,选择其中6个具有代表性的序列进行测序。序列及系统进化分析表明,属于其中3个操作分类单元的克隆最相似菌都是UnculturedarchaealsymbiontPA202,相似性均为95%,没有与这些克隆相似性较高的已培养甲烷菌;属于另外2个操作分类单元的克隆最相似菌都是Unculturedrumenmethanogen956,相似性均为97%,最相似已知菌为Methanobrevibactersp.NT7,相似性为97%。结果表明,真菌培养液中存在目前尚未分离培养的瘤胃甲烷菌。  相似文献

A rapid method for the quantitative determination of tyrosine (Tyr), phenylalanine (Phe), p-hydroxybenzoic acid (HBA), p-hydroxyphenylacetic acid (HPA), benzoic acid (BZA), p-hydroxyphenylpyruvic acid (HPY), phenylacetic acid (PAA), phenyllactic acid (PLA), tryptophan (Trp), indoleacetic acid (IAA), phenylpyruvic acid (PPY), phenylpropionic acid (PPA) and cinnamic acid (CNA) in goat rumen fluid was established by high-performance liquid chromatography (HPLC). The mobile phase used for isocratic elution was 50 mM sodium phosphate buffer (pH 6.5)–methanol (97:3, v/v). The flow-rate was 1.0 ml/min; column temperature 40°C and compounds were monitored at 215 nm with a UV absorbance detector after injection of 10 μl of filtered rumen fluid. Analysis was completed within 40 min. The minimum detectable limits of quantification (μM) of these compounds were Tyr, 2; Phe, 3; HBA, 1; HPA, 2; BZA, 2; HPY, 8; PAA, 3; PLA, 4; Trp, 2; IAA, 2; PPY, 15; PPA, 8 and CNA, 4. Detectable levels of Tyr, Phe, HPA, BZA, HPY, PAA, PLA, Trp and PPA were found in the deproteinized rumen fluid of goat fed a haycube and concentrate mixture. PAA was the predominant compound before and after feeding. The concentrations of HPA, BZA, PAA, PLA and PPA in the goat rumen fluid increased after feeding, while the concentration of Tyr decreased. Phe, HPY and Trp were minor components at all times. PPY, IAA and CNA were not detected and HBA was not completely resolved in the goat rumen fluid.  相似文献

AIMS: This study was carried out to determine whether bacterial and ciliate populations in goat rumen vary significantly between different goat species living in the same environment. METHODS AND RESULTS: Bacterial and ciliate communities in the rumen of three goat species were analysed at the molecular level using denaturing gradient gel electrophoresis. The microbial community varied considerably among goats living in the same environment. Interspecies variation in the bacterial population was noticeably greater than intraspecies variation. In contrast, there was considerable variation in the ciliate population among goats within the same species, and intraspecies similarities were no greater than those observed across species. CONCLUSIONS: Because environmental factors and diets were identical for all goats, differences in bacterial populations reflect species-specific differences in rumen microbes. SIGNIFICANCE AND IMPACT OF THE STUDY: Factors related to the host species have an important effect on determining the bacterial composition in the goat rumen.  相似文献

Aims:  To screen a pair of primers suitable for denaturing gradient gel electrophoretic (DGGE) analysis of ruminal methanogenic Archaea and to detect the archaeal communities in the rumen of goat.
Methods and Results:  Nine primer pairs for 16S rDNA of methanogenic Archaea , including six for directed polymerase chain reaction (PCR) and three for nested PCR were first evaluated by PCR amplification of the total DNA from rumen fluids and bacteria. The DGGE analysis of rumen fluids was then conducted with three primer sets (344fGC/915r, 1106fGC/1378r and 519f/915rGC) of the nine pairs tested. Good separation and quality of patterns were obtained in DGGE analysis with primer pairs 1106fGC/1378r and 519f/915rGC. A total of 40 DNA fragments were excised from the DGGE gels and their sequences were determined. All fragments belonged to methanogenic Archaea while primer pair 519f/915rGC had better amplification ranges than the other two primer pairs.
Conclusions:  The procedure of DGGE analysis with primer pair 519f/915rGC was more suitable for investigating methanogenic archaeal community in the rumen. The dominant methanogenic Archaea in the rumen of goat was Methanobrevibacter sp. and an unidentified methanogenic Archaea .
Significance and Impact of the Study:  One pair of primers suitable for DGGE analysis of ruminal methanogenic Archaea was obtained and the molecular diversity of ruminal methanogenic Archaea in goat was investigated by PCR-DGGE.  相似文献

A large amount of ammonia is produced in the rumen and some portion of the ammonia are absorbed into the portal blood through the rumen mucosa. Accordingly, it seems that ammonia detoxication is more necessary for the ruminant than for the non-ruminant. Activities of the urea cycle enzymes as principal instrument for ammonia detoxication in goat were investigated in this experiment.

The activities of the urea cycle enzymes of goat were found to be very similar to those of rat reported by other authors. The activities of the urea cycle enzymes were affected by the protein level of diet. Administration of magnesium aspartate increased the activities of argininosuccinate synthetase and arginase, and had some effect on the concentrations of citrulline, aspartic acid, and urea in the liver.  相似文献

Anaerobic ruminal fungi may play an active role in fibre degradation as evidenced by the production of different fibrolytic enzymes in culture filtrate. In the present study, 16 anaerobic fungal strains were isolated from ruminal and faecal samples of sheep and goats. Based on their morphological characteristics they were identified as species of Anaeromyces, Orpinomyces, Piromyces and Neocallimastix. Isolated Neocallimastix sp. from goat rumen showed a maximum activity of CMCase (47.9 mIU ml(-1)) and filter paper cellulase (48.3 mIU ml(-1)), while Anaeromyces sp. from sheep rumen showed a maximum xylanolytic activity (48.3 mIU ml(-1)). The cellobiase activity for all the isolates ranged from 178.0-182.7 mIU ml(-1). Based on the enzymatic activities, isolated Anaeromyces sp. from sheep rumen and Neocallimastix sp. from goat rumen were selected for their potential of in vitro fibre degradation. The highest in vitro digestibility of NDF (23.2%) and DM (34.4%) was shown for Neocallimastix sp. from goat rumen, as compared to the digestibility of NDF and DM in the control group of 17.5 and 25.0%, respectively.  相似文献

A rapid method for determination of phenylalanine (Phe), tyrosine (Tyr), benzoic acid (BZA), phenylacetic acid (PAA), phenyllactic acid (PLA), phenylpyruvic acid (PPY), phenylpropionic acid (PPR), and cinnamic acid (CNM) in goat rumen fluid was established by high-performance liquid chromatography (HPLC). The mobile phase used for isocratic elution was methanol-sodium acetate buffer (pH 6.5) (8:92, v/v). The compounds were monitored at 220 nm with a UV detector. A 5-μl portion of the filtrated rumen was analyzed and the analysis was completed within 20 min. The minimum detectable limits (μM) of these compounds were: 12 for Phe, 3 for Tyr, 3 for BZA, 9 for PAA, 12 for PLA, 15 for PPY, 20 for PPR, and 8 for CNM. The average contents of Phe, BZA, PAA, PLA, and PPR in the rumen fluid of three goats were 15.4, 73.7, 615.9, 51.1, and 39.9 μM before morning feeding, 17.0, 123.7, 650.4, 208.2, and 502.4 μM at 3 h after feeeding, and 18.4, 124.2, 510.0, 129.9, and 178.5 μM at 6 h after feeding, respectively. Of these compounds PAA was present at the highest concentration both before and after feeding. The content of PPR extremely increased especially at 3 h after feeding. The other three compounds, i.e. Tyr, PPY, and CNM, were not detected in goat rumen fluid.  相似文献

目的建立提取高质量的瘤胃微生物DNA的方法,为采用免培养技术研究山羊瘤胃微生物奠定基础。方法采集山羊瘤胃内容物,用SDS高盐法提取微生物总DNA,以通用引物扩增细菌和古细菌的16SrDNA。结果提取到的瘤胃微生物总DNA片段大于23kb,PCR能够扩增出细菌和古细菌的16SrDNA片段。结论用该提取方法得到的山羊瘤胃微生物总DNA能够满足后续实验的需要。  相似文献

采用免培养的rpoB和16S rDNA基因的变性梯度凝胶电泳技术(DGGE)对3种山羊(波尔山羊,内蒙古绒山羊,四川南江黄羊)瘤胃细菌优势菌群结构进行了比较分析。研究结果显示rpoBDGGE图谱中条带数目少于16S rDNA图谱,并且条带分离效果明显,更有利于分析瘤胃细菌群落组成。从两种DGGE图谱中均可以发现3种山羊瘤胃细菌具有一定的相似性,种内个体间相似性明显高于种间相似性,这说明寄主品种是影响瘤胃细菌种群构成的一个重要因素。同时进行了部分优势细菌16S rDNA基因V6-V8区序列的系统发育分析。基因序列分析表明,DGGE图谱中优势条带的16S rDNA基因序列中有4条克隆的序列与基因库最相似菌的相似性大于97%,余下的克隆序列相似性在89%~96%之间,其中13条序列的与之相似性最高的序列均来自于未被鉴定的瘤胃细菌。  相似文献

Bacteriological studies were made with in vitro sheep and goat ruminal fluids supplemented with several feeds (alfalfa hay, wheat straw, Agave americana, Opuntia ficus indica and Atriplex nummularia) during anaerobic incubation at 38-39 degrees C. Drastic changes in the bacterial population of sheep ruminal fluids occurred in the presence of different feeds, particularly with addition of feeds of low nutritional quality (wheat straw, A. americana and O. ficus indica). However, the bacterial population in goat rumen liquor was little affected by the addition of the same feeds. These results, which suggest that the rumen bacteria in goats are less affected by different nutritional conditions than the rumen bacteria in sheep, are discussed.  相似文献

The ability of rumen microorganisms to use fibrous plant matter plays an important role in ruminant animals; however, little information about rumen colonization by microbial populations after weaning has been reported. In this study, high-throughput sequencing was used to investigate the establishment of this microbial population in 80 to 110-day-old goats. Illumina sequencing of goat rumen samples yielded 101,356,610 nucleotides that were assembled into 256,868 reads with an average read length of 394 nucleotides. Taxonomic analysis of metagenomic reads indicated that the predominant phyla were distinct at different growth stages. The phyla Firmicutes and Synergistetes were predominant in samples taken from 80 to 100-day-old goats, but Bacteroidetes and Firmicutes became the most abundant phyla in samples from 110-day-old animals. There was a remarkable variation in the microbial populations with age; Firmicutes and Synergistetes decreased after weaning, but Bacteroidetes and Proteobacteria increased from 80 to 110 day of age. These findings suggested that colonization of the rumen by microorganisms is related to their function in the rumen digestive system. These results give a better understanding of the role of rumen microbes and the establishment of the microbial population, which help to maintain the host’s health and improve animal performance.  相似文献

Most studies present in the literature about the rumen microbiome have focused on cattle and sheep. This is the first report of the characterization of the bacterial and archaeal communities present in the liquid and solid-associated fractions of the rumen from free ranging Moxotó breed goats using 16S rRNA gene libraries. PCR was used to amplify the 16S rRNA gene with bacterial and archaeal universal primers and sequences from each library constructed were obtained. Sequences of Bacteria from the phyla Bacteroidetes and Firmicutes were predominant. The overall dominant classes in the rumen were Clostridia and Bacteroidia, which are known to play a role in plant fiber degradation in other ruminants. Unclassified Bacteria accounted for 4.7% of the liquid fraction sequences and 16.4% of the solid fraction sequences. From the archaeal libraries only sequences from the phylum Euryarcheota were identified and were assigned to the class Methanobacteria of the genera Methanobrevibacter and Methanosphaera. A group of Archaea not previously known to be associated with the rumen was identified: uncultured methanogens belonging to the "uncultured marine bacteria" groups II and III. The local water contained high salt concentrations and this may explain the presence of these groups in the Moxotó goat rumen.  相似文献

金属硫蛋白(MTs)是一类低分子量、金属和半胱氨酸含量高的细胞质蛋白, 哺乳动物的MTs包括MT-I、MT-II、MT-Ⅲ和MT-IV 4种亚型, 其中MT-IV只在磷状复层扁平上皮细胞中表达, 相关研究报道较少。本研究根据GenBank已公布的动物MT-IV基因序列, 设计出扩增山羊和绵羊MT-IV基因的特异性PCR引物MT-IVSP1和MT-IVSP2, 利用RT-PCR的方法, 分别从山羊和绵羊的瘤胃组织mRNA中, 克隆出山羊和绵羊的MT-IV基因编码区序列(均为189 bp), 序列登录GenBank, 获得序列号EF470251和EF624067。通过序列分析, 表明山羊和绵羊两个物种MT-IV基因编码区全编码均为189 bp、编码62个氨基酸, 其中绵羊的MT-IV含有20个半胱氨酸, 而山羊第61位保守的半胱氨酸被色氨酸所代替。两个物种的MT-IV均不含芳香族氨基酸, 含有MTs特有的C-X-C、C-X-X-C、C-C-X-C-C结构, 无明显的跨膜结构域, 无信号肽, 是一种细胞质蛋白。二级结构分析表明两个物种的MT-IV二级结构大多数为无规则卷曲结构, 分别在第7~9和第49~51氨基酸残基性存在折叠结构, 不存在螺旋结构。三级结构预测结果表明两个物种MT-IV的三级结构由a和b两个结构域组成, 其中β结构域相同, a结构域山羊少一个半胱氨酸残基, 其结构与绵间存在明显差异, 这一差异可能对山羊MT-IV的生理功能产生一定影响, 有必要深入研究。  相似文献

【目的】从饲喂高精料的本地山羊瘤胃内分离到一株利用乳酸并能产生大量丙酸的菌株L9,并进一步研究了该菌在调控瘤胃微生物发酵中的作用。【方法】采用厌氧培养技术,结合形态、生理生化特性和16SrRNA基因序列分析结果。【结果】该菌株被鉴定为反刍兽新月形单胞菌(Selenomonas ruminantium)。该菌株体外代谢特性研究表明,L9可利用乳酸作为唯一碳源,该菌在24h内可对90mmol/L的乳酸完全降解。体外摸拟瘤胃急性酸中毒的发酵试验结果表明,以淀粉为底物时,与对照组相比,添加菌株L9可显著降低瘤胃微生物体外培养体系中乳酸浓度,提高pH值,提高总挥发性脂肪酸和丙酸浓度,并显著降低乙酸与丙酸的浓度比(P〈0.05)。【结论】结果显示,菌株L9是一株可代谢乳酸,促进丙酸生成,提高总挥发性脂肪酸浓度的有益瘤胃细菌。  相似文献

The rumen parasite, Gastrothylax crumenifer (Platyhelminthes: Gastrothylacidae), is a highly pathogenic trematode parasite of goat (Capra hircus). It sucks blood that causes acute disease like anemia, and severe economic losses occur due to morbidity and mortality of the ruminant infected by these worms. The study of these rumen paramphistomes, their infection, and public health importance remains unclear in India especially in the western part of state Uttar Pradesh (U.P.), Meerut, India, where the goat meat consumption is very high. This paper provides the molecular characterization of G. crumenifer recovered from the rumen of Capra hircus from Meerut, U.P., India by the partial sequence of 28S rDNA. Nucleotide sequence similarity searching on BLAST of 28S rDNA from parasites showed the highest identity with those of G. crumenifer from the same host Capra hircus. This is the first report of molecular identification of G. crumenifer from this part of India.  相似文献

从内蒙古鄂尔多斯山羊瘤胃中分离筛选到1株产共轭亚油酸的瘤胃细菌RB111,该菌株的cis9,trans11-CLA和trans10,cis12-CLA总产量为269.2 mg/L,其中cis9,trans11-CLA占52.64%,trans10,cis12-CLA占47.36%。对菌株RB111进行了形态学观察、生理生化鉴定、脂肪酸组成分析以及16S rRNA基因序列分析。16S rRNA基因序列分析结果表明该菌株与婴儿链球菌(Streptococcus infantarius)的模式菌株NCDO 599的序列相似性为99%,该菌株的形态特征及生理生化特性与文献报道的Streptococcus infantarius一致。脂肪酸组成分析结果显示,菌株RB111的细胞脂肪酸主要成分是C16:0、C18:1ω9c和C18:0,3种脂肪酸占脂肪酸总量的60.64%。综合以上结果,菌株RB111被鉴定为婴儿链球菌Streptococcus infantarius。  相似文献
