Sorghum halepense L. (johnsongrass) is one of the world's most noxious weeds, and a paradigm for the potential dangers of crop-weed hybridization. Introduced into the southeastern United States about 200 years ago, S. halepense is a close relative of cultivated sorghum (Sorghum bicolor). Both artificial crossing and experimental field studies have demonstrated the potential for S. halepensex S. bicolor hybrid formation, but no prior study has addressed the long-term persistence of sorghum genes in johnsongrass populations. We surveyed 283 loci (on all 10 sorghum linkage groups) to identify 77 alleles at 69 loci that are found in US sorghum cultivars but are absent from a worldwide sampling of johnsongrass genotypes. These putatively cultivar-specific alleles were present in up to 32.3% of individuals in johnsongrass populations adjacent to long-term sorghum production fields in Texas and Nebraska. Lower frequencies of cultivar-specific alleles at smaller numbers of loci are found in johnsongrass populations from New Jersey and Georgia with no recent exposure to cultivated sorghum, suggesting that introgressed sorghum alleles may be dispersed across long distances. The number of cultivar-specific alleles and extensive multilocus patterns of cultivar-specific allelic composition observed at both linked and unlinked loci in the johnsongrass populations, are inconsistent with alternatives to introgression such as convergence, or joint retention of ancestral polymorphisms. Naturalized johnsongrass populations appear to provide a conduit by which transgenes from sorghum could become widely disseminated. 相似文献

The role of crop-to-weed gene flow is often controversial and overlooked. As a consequence, the likelihood of spontaneous crop-to-weed hybridization in most crop/weed systems is generally unknown. The lack of data relating to the formation of crop/weed hybrids has particular contemporary significance when considering the wide scale commercial release of transgenic crop plants and the potential for escape of engineered genes via crop-to-weed hybridization. We created an experimental system whereby we could examine the incidence and rate of spontaneous crop-to-weed hybridization between Sorghum bicolor and S. halepense, johnsongrass. An isozyme marker was used to identify hybrid plants through progeny testing. Incidence and rate of hybridization were highly variable with respect to weed distance from the crop, location of the study site, and year the study was performed. Crop/weed hybrids were detected at distances of 0.5–100 m from the crop. Interspecific hybridization can and does occur in this system at a substantial and measurable rate. Transgenes introduced into crop sorghum can be expected to have the opportunity to escape cultivation through interspecific hybridization with johnsongrass. Traits that prove to be beneficial to weeds possessing them can be expected to persist and spread. This is an issue that needs to be addressed when developing biosafety guidelines for the commercial release of transgenic sorghums. 相似文献

BACKGROUND AND AIMS: Transgene escape through gene flow from genetically modified (GM) crops to their wild relative species may potentially cause environmental biosafety problems. The aim of this study was to assess the extent of gene flow between cultivated rice and two of its close relatives under field conditions. METHODS: Experiments were conducted at two sites in Korea and China to determine gene flow from cultivated rice (Oryza sativa L.) to weedy rice (O. sativa f. spontanea) and common wild rice (O. rufipogon Griff.), respectively, under special field conditions mimicking the natural occurrence of the wild relatives in Asia. Herbicide resistance (bar) and SSR molecular finger printing were used as markers to accurately determine gene flow frequencies from cultivated rice varieties to their wild relatives. KEY RESULTS: Gene flow frequency from cultivated rice was detected as between approx. 0.011 and 0.046 % to weedy rice and between approx. 1.21 and 2.19 % to wild rice under the field conditions. CONCLUSIONS: Gene flow occurs with a noticeable frequency from cultivated rice to its weedy and wild relatives, and this might cause potential ecological consequences. It is recommended that isolation zones should be established with sufficient distances between GM rice varieties and wild rice populations to avoid potential outcrosses. Also, GM rice should not be released when it has inserted genes that can significantly enhance the ecological fitness of weedy rice in regions where weedy rice is already abundant and causing great problems. 相似文献

Some transgenic crops can introgress genes into other varieties of the crop, to related weeds or themselves remain as 'volunteer' weeds, potentially enhancing the invasiveness or weediness of the resulting offspring. The presently suggested mechanisms for transgene containment allow low frequency of gene release (leakage), requiring the mitigation of continued spread. Transgenic mitigation (TM), where a desired primary gene is tandemly coupled with mitigating genes that are positive or neutral to the crop but deleterious to hybrids and their progeny, was tested as a mechanism to mitigate transgene introgression. Dwarfism, which typically increases crop yield while decreasing the ability to compete, was used as a mitigator. A construct of a dominant ahasR (acetohydroxy acid synthase) gene conferring herbicide resistance in tandem with the semidominant mitigator dwarfing Delta gai (gibberellic acid-insensitive) gene was transformed into tobacco (Nicotiana tabacum). The integration and the phenotypic stability of the tandemly linked ahasR and Delta gai genomic inserts in later generations were confirmed by polymerase chain reaction. The hemizygous semidwarf imazapyr-resistant TM T1 (= BC1) transgenic plants were weak competitors when cocultivated with wild type segregants under greenhouse conditions and without using the herbicide. The competition was most intense at close spacings typical of weed offspring. Most dwarf plants interspersed with wild type died at 1-cm, > 70% at 2.5-cm and 45% at 5-cm spacing, and the dwarf survivors formed no flowers. At 10-cm spacing, where few TM plants died, only those TM plants growing at the periphery of the large cultivation containers formed flowers, after the wild type plants terminated growth. The highest reproductive TM fitness relative to the wild type was 17%. The results demonstrate the suppression of crop-weed hybrids when competing with wild type weeds, or such crops as volunteer weeds, in seasons when the selector (herbicide) is not used. The linked unfitness would be continuously manifested in future generations, keeping the transgene at a low frequency. 相似文献

The Corsican swallowtail butterfly, Papilio hospiton , is endemic to Corsica and Sardinia (France and Italy) and included in the list of endangered species by the Washington Convention, It is spread all over Corsica in scattered populations linked to diverse habitats. A study by enzyme electrophoresis showed that the genetic diversity of the species is of the same order of magnitude as that of Papilio machaon from continental France and Corsica. The differentiation between populations is rather low, which is consistent with the high vagility of the adults. Natural hybridization between P. hospiton and P. machaon is frequent, and laboratory crosses show that the hybrids are not sterile. However, developmental perturbations impair the viability of further hybrid progenies. Although limited introgression between the two species is likely to take place, enzyme electrophoresis and PCR-RFLP analysis of mitochondrial DNA show that their gene pools remain distincfTtienetic assimilation by P. machaon therefore does not seem to be a threat for P. hospiton. 相似文献

高粱(Sorghum bicolour)是世界上最重要的粮食、饲料、酿造和能源作物之一, 也是C4植物研究的模式植物。蚜虫是农业生产上的重要害虫, 几乎危害所有的栽培作物。危害高粱的蚜虫主要包括高粱蚜(Melanaphis sacchari)、麦二叉蚜(Schizaphis graminum)和玉米蚜(Rhopalosiphum maidis)。高粱的抗蚜资源尚不丰富且缺乏深入系统的研究。目前研究较多的是麦二叉蚜的抗性遗传方面, 已定位20个抗性QTLs, 单一QTL对抗性差异贡献率最高可达80.3%, 对高粱蚜和玉米蚜的研究尚需进一步加强。高粱的理化特性与其抗蚜性能相关, 故可与育种实践相结合。高粱和蚜虫(Acyrthosiphon pisum)的全基因组测序工作已经完成, 这将有助于蚜虫-植物间的相互作用关系及植物对蚜虫的抗性机制研究。目前已克隆到2个抗蚜基因, 且多个抗蚜基因(位点)已被定位在染色体上。该文重点综述了上述研究成果并对高粱抗蚜的研究前景进行了展望。 相似文献

of rhesus macaques (Macaca mulatta) from mainland Asia and long-tailed macaques (M. fascicularis) from mainland and insular Southeast Asia. The subjects were either recently captured in the wild or derived from wild-caught
founders maintained in captivity for biomedical research. A large number of alleles are shared between the 2 macaque species
but a significant genetic division between them persists. The distinction is more clear-cut among populations that are not,
or are unlikely to have recently been, geographically contiguous. Our results suggest there has been significant interspecific
nuclear gene flow between rhesus macaques and long-tailed macaques on the mainland. Comparisons of mainland and island populations
of long-tailed macaques reflect marked genetic subdivisions due to barriers to migration. Geographic isolation has restricted
gene flow, allowing island populations to become subdivided and genetically differentiated. Indonesian long-tailed macaques
show evidence of long-term separation and genetic isolation from the mainland populations, whereas long-tailed macaques from
the Philippines and Mauritius both display evidence of founder effects and subsequent isolation, with the impact from genetic

The role of hybridization in animal speciation is controversial and recent research has challenged the long-standing criterion of complete reproductive isolation to attain species status. The speciation-with-gene-flow model posits that the genome is semi-permeable and hybridization may be a phase in the process of divergence. Here, we apply these concepts to a previously identified zone of mtDNA introgression between the two strongly morphologically differentiated subspecies of red-tailed chipmunk ( Tamias ruficaudus ) in the US Inland Northwest. Using multilocus genotype data from the southern, older contact zone, we demonstrate that neutral gene flow is unusually low between the subspecies across the Lochsa River. This is geographically congruent with the discontinuity in bacular morphology, indicating that the cline of mitochondrial DNA (mtDNA) haplotypes is displaced. Furthermore, we elucidate the evolutionary forces responsible by testing hypotheses of lineage sorting and hybridization. We determined that introgressive hybridization is the cause of mtDNA/morphology incongruence because there are non-zero levels of migration and gene flow. Although our estimate of the age of the hybrid zone has wide credibility intervals, the hybridization events occurred in the Late Pleistocene and the divergence occurred in the Middle Pleistocene. Finally, we assessed substructure within and adjacent to the hybrid zone and found that the hybrid zone constitutes a set of populations that are genetically differentiated from parental sets of populations; therefore, hybridization in this system is not likely an evolutionary sink, but has generated novel combinations of genotypes. 相似文献

The roles of intra- and interspecific gene flow in speciation and species evolution are topics of great current interest in molecular ecology and evolutionary biology. Recent modelling studies call for new empirical data to test hypotheses arising from the recent shift from a 'whole-genome reproductive isolation' view to a 'genic' view of species and speciation. Particularly scarce (and thus of particular interest) are molecular genetic data on recently radiated, naturally hybridizing species in strongly structured and species-rich environments. Here, we studied four sympatric plant species (Pitcairnia spp.; Bromeliaceae) adapted to Neotropical inselbergs (isolated outcrops resembling habitat 'islands' in tropical rainforests) using nuclear and plastid DNA. Patterns of plastid DNA haplotype sharing and nuclear genomic admixture suggest the presence of both, incomplete lineage sorting and interspecific gene flow over extended periods of time. Integrity and cohesion of inselberg species of Pitcairnia are maintained despite introgression and in the face of extremely low within-species migration rates (N(e)m < 1 migrant per generation). Cross-evaluation of our genetic data against published pollination experiments indicate that species integrity is maintained by the simultaneous action of multiple prezygotic barriers, including flowering phenology, pollinator isolation and divergent mating systems. Postzygotic Bateson-Dobzhansky-Muller incompatibilities appear to contribute to isolation, as suggested by asymmetric introgression rates of single loci. Our results suggest that incomplete lineage sorting, hybridization and introgression form integral aspects of adaptive radiation in Neotropical inselberg 'archipelagos'. Inselbergs with multiple closely related co-occurring species should be of special interest to students of speciation in mountain systems, and to ongoing conservation programmes in the Atlantic Rainforest biodiversity hotspot. 相似文献

Genomic sequence data provide a rich source of information about the history of species divergence and interspecific hybridization or introgression. Despite recent advances in genomics and statistical methods, it remains challenging to infer gene flow, and as a result, one may have to estimate introgression rates and times under misspecified models. Here we use mathematical analysis and computer simulation to examine estimation bias and issues of interpretation when the model of gene flow is misspecified in analysis of genomic datasets, for example, if introgression is assigned to the wrong lineages. In the case of two species, we establish a correspondence between the migration rate in the continuous migration model and the introgression probability in the introgression model. When gene flow occurs continuously through time but in the analysis is assumed to occur at a fixed time point, common evolutionary parameters such as species divergence times are surprisingly well estimated. However, the time of introgression tends to be estimated towards the recent end of the period of continuous gene flow. When introgression events are assigned incorrectly to the parental or daughter lineages, introgression times tend to collapse onto species divergence times, with introgression probabilities underestimated. Overall, our analyses suggest that the simple introgression model is useful for extracting information concerning between-specific gene flow and divergence even when the model may be misspecified. However, for reliable inference of gene flow it is important to include multiple samples per species, in particular, from hybridizing species. 相似文献

Hybridization and introgression in Carpobrotus spp. (Aizoaceae) in California. II. Allozyme evidence
Morphological evidence has indicated that hybridization and introgression are occurring between Carpobrotus edulis L., a nonindigenous, invasive species in California (Bolus), and its putative native congener, C. chilensis. The identification of allozyme markers has enabled us to quantify hybridization and the extent of introgression between these species. Samples from 20 individuals of each of five morphological types (both parent species and three hybrid phenotypes) were collected from 39 populations along the coast of California from the Mexico to Oregon borders. Ten enzyme systems revealed a total of 17 resolvable loci, eight of which were polymorphic for the genus. Five of the polymorphic loci easily differentiate C. edulis and C. chilensis. Allele frequencies among the morphologically defined types are consistent with estimations of allele frequency changes due to first- and second-generation backcrossing. In comparison to long-lived, herbaceous perennials and widespread species, C. edulis and intermediate types have more variation in their populations (P = 41.73, Ap = 2.11, Ho = 0.246, and P = 38.13, Ap = 2.06, Ho = 0.216, respectively) than C. chilensis (P = 11.76, Ap = 2.00, Ho = 0.082). Indirect estimates of gene flow indicate four of the five morphological types are outcrossing. Gene flow between previously allopatric species may have broad implications if it results in an increase in fitness; further experimentation is needed to determine the ultimate ecological consequences of this phenomenon and its possible threat to limited, remaining natural habitat in California. 相似文献

Individuals of the invasive non-native plant species Carpobrotus edulis, its native or naturalized congener C. chilensis, and putative hybrids are found throughout coastal California. This study characterized morphological variation within the genus and assessed the extent of hybridization and direction of introgression between C. edulis and C. chilensis. Twelve randomly selected Carpobrotus individuals from each of 49 sample locations were classified as "C. edulis," "C. chilensis," or one of three intermediate morphotype classes, and compared for clonal, vegetative, and fruit characters. Univariate comparisons showed that C. edulis individuals had deeper mats with more cover within the mat boundary, longer, wider, more serrate leaves, and wider fruits with more carpels than intermediate and C. chilensis morphotype classes. Carpobrotus edulis and intermediates had more lateral branches than did C. chilensis, suggesting that hybrids tend to vary toward C. edulis. Discriminant function analysis (DFA) identified the following characters as discriminating significantly among Carpobrotus groups: leaf length, leaf width, leaf length/width, number of lateral branches, number of carpels, and fruit diameter. Using these characters, DFA correctly classified 98% of apparently pure C. edulis and C. chilensis, but only 76% of all Carpobrotus individuals when putative hybrids were included. Principal components analysis showed that C. edulis and C. chilensis cluster into distinct morphotypes, but that hybrids overlap with both groups, particularly with C. edulis. The genus Carpobrotus in California resembles a large hybrid swarm, with putative hybrids forming a large portion of the overall population and tending to vary toward C. edulis. 相似文献
