The genus Chromobacterium encompasses free-living Gram-negative bacteria. Until 2007, the genus consisted of only one species but six species are now recognized. Chromobacterium violaceum is the type species of the genus and is commonly found in soil and water in tropical and sub-tropical regions. We have investigated a collection of 111 isolates displaying violet pigmentation from undisturbed aquatic and soil environments from Brazilian Cerrado ecosystem. The 16S rRNA gene phylogeny revealed that all isolates were allocated in a monophyletic cluster inside the Chromobacterium genus and formed few clusters related most closely with Chromobacterium piscinae. The two sets of isolates from water and soil were analyzed by the repetitive extragenic palindromic (rep)-PCR genomic fingerprinting technique using a BOX-AR1 primer. The antimicrobial susceptibility and the different carbon sources utilized by these isolates were also investigated. Physiological profiles of the isolates generated by BIOLOG GN2 plates showed great versatility in the substrate utilization, much higher than the C. violaceum ATCC 12472. All isolates exhibited a high minimum inhibitory concentration (MIC) to ampicillin (MIC > 512 μg/ml) and were inhibited by ciprofloxacin, tetracycline and mercury at the lowest concentration tested (MIC < 2 μg/ml). Thirteen BOX-PCR band patterns were identified from 33 individual fingerprints. Eleven patterns provided evidence for endemic distributions. Antimicrobial susceptibility and BOX-PCR fingerprint clustering showed a clear distinction between Chromobacterium isolates from the water and soil. The results suggested that microenvironment barriers such as water and soil can play an important role in the periodic selection and diversification of Chromobacterium population ecotypes.  相似文献

Aims: Polyhydroxyalkanoate (PHA) with enhanced physicochemical properties will be ideal for a wide range of practical applications. The incorporation of 3‐hydroxy‐4‐methylvalerate (3H4MV) into the polymer backbone is known to improve the overall properties of the resulting polymer. However, the most suitable micro‐organism and PHA synthase that can synthesize this monomer efficiently still remain unknown at present. Therefore, we evaluated the abilities of a locally isolated Chromobacterium sp. USM2 to produce PHA containing 3H4MV. Methods and Results: The ability of Chromobacterium sp. USM2 to synthesize poly(3‐hydroxybutyrate‐co‐3‐hydroxy‐4‐methylvalerate) [P(3HB‐co‐3H4MV)] was evaluated under different culture conditions. It was found that Chromobacterium sp. USM2 can synthesize P(3HB‐co‐3H4MV) when glucose and isocaproic acid were fed as carbon source. However, the highest molar fraction of 3H4MV, 22 mol% was detected in Chromobacterium sp. USM2 when isocaproic acid was provided as the sole carbon source. In addition, aeration was identified as a crucial factor in initiating the accumulation of high 3H4MV molar fractions. Conclusions: Chromobacterium sp. USM2 was able to synthesize broad comonomer compositional distribution of P(3HB‐co‐3H4MV). Significance and Impact of the Study: Compared with Cupriavidus necator and Burkholderia sp., Chromobacterium sp. USM2 was found to have better ability to bioconvert isocaproic acid to form 3H4MV unit.  相似文献

Different methods of clustering were applied to the results of a numerical study of the taxonomy ofRhizobium, Agrobacterium, Beijerinckia, Chromobacterium, andBacillus published by Graham (1964) (J. Gen. Microbiol.35: 511).The groupings obtained were compared with that of Graham and external information from published studies on flagellation and DNA-base-composition was used to judge the merits of the various groupings. It is shown that a more satisfactory grouping is obtained with furthest neighbour and flexible sorting than with either nearest neighbour sorting or with the modified method of Sneath (1957) used by Graham.The conclusions from this study are that (1)Agrobacterium radiobacter andA. tumefaciens belong to one taxon; (2)Agrobacterium is more closely related to fast-growingRhizobium than is slow-growingRhizobium; (3) fast growing and slow-growingRhizobium are more closely related to each other than toBeijerinckia andChromobacterium; (4)Bacillus, Beijerinckia andChromobacterium are more closely related to each other than toRhizobium. Of these conclusions (1) and (2) are in agreement with Graham.It is postulated that the genusRhizobum should not be split at the generic level and thatAgrobacterium should not be merged withRhizobium as was suggested by Graham. Instead it is proposed thatRhizobium be considered as consisting of two Sections, one withR. japonicum as the type species and the other withR. leguminosarum as the type species, and that taxonomic treatment at the species level be left until much more material has been studied, and thatAgrobacterium be retained as a generic name until more evidence has been produced.  相似文献

Summary Bacteria utilizing high concentrations of acetonitrile as the sole carbon source were isolated and identified asChromobacterium sp. andPseudomonas aeruginosa. Maximum growth was attained after 96 h of incubation andP. aeruginosa grew slightly faster thanChromobacterium sp. The strains were able to grow and oxidize acetonitrile at concentrations as high as 600 mM. However, higher concentrations inhibited growth and oxygen uptake. Degradation studies with (14C)acetonitrile indicated 57% of acetonitrile was degraded byPseudomonas aeruginosa as compared to 43% byChromobacterium. The isolates utilized different nitrile compounds as carbon substrates.  相似文献

Summary Native plants of Wisconsin seem to show considerable selectivity in the absorption of mineral elements. This was indicated by an unusually high or low concentration of a specific element in a species in comparison with other species growing under generally similar conditions, and by the fact that a species might have an unusually high concentration of one or several elements but an unusually low content of other elements. A capacity for selective uptake of a particular element was in general characteristic of samples of the same species obtained from different sites. The potassium content of some accumulator species was as high as 7.0 per cent when the average concentration in other species from the same sample area was only 2.0 per cent. Manganese seemed to be selectively excluded by some species.Cornus canadensis, for example, collected from a bog of pH 4.0 contained only 149 ppm manganese in comparison with an average concentration of 1061 ppm in other species from the same area.Nemopanthus mucronata showed an unusual capacity for selective zinc accumulation. Samples of this species contained from 300 to 700 ppm zinc while comparison species from the same sites contained less than 50 ppm. Some of the nutritional and ecological implications of the results are discussed.  相似文献

Karyotype analyses were conducted onCunninghamia konishii, Cunninghamia lanceolata, andTaiwania cryptomerioides, all members ofTaxodiaceae. The somatic chromosome number was found to be 2n = 2x = 22 in all species which concurrs with previous reports. The karyotypes are generally asymmetrical with the smaller chromosomes being more submedian than the larger ones. Chromosomes with unusual or specific structures, thought to be associated with the nucleolar organizing region, were found in each species.Cunninghamia species have a marker chromosome pair with an unusually long secondary constriction.Taiwania has an unusually long kinetochore region present in a submedian chromosome pair.  相似文献

A lectin has been isolated from rhizomes of ground elder (Aegopodium podagraria) using a combination of affinity chromatography on erythrocyte membrane proteins immobilized on cross-linked agarose and hydroxyapatite, and ion-exchange chromatography. The molecular structure of the lectin was determined by gelfiltration, sucrose density-gradient centrifugation and gel electrophoresis under denaturing conditions. It has an unusually high Mr (about 480000) and is most probably an octamer composed of two distinct types of subunits with slightly different Mr (about 60000). Hapten inhibition assays indicated that the Aegopodium lectin is preferentially inhibited by N-acetylgalactosamine. Nevertheless, it does not agglutinate preferentially blood-group-A erythrocytes. The ground-elder lectin is a typical non-seed lectin, which occurs virtually exclusively in the underground rhizomes. In this organ it is an abundant protein as it represents up to 5% of the total protein content. The lectin content of the rhizome tissue varies strongly according to its particular location along the organ. In addition, the lectin content changes dramatically as a function of the seasons. The ground-elder lectin differs from all other plant lectins by its unusually high molecular weight. Two new species of Myxozoa from the brain of the green knife fish Eigemannia virescens are described: Myxobolus inaequus sp. n. has an unusually large spore body and extremely unequal polar capsules, and Henneguya theca sp. n. has an attenuated spore encased in a sheath not previously described in other Myxozoa. Only spores of the two species were observed, and infections caused no obvious pathological changes in the brain.  相似文献

Chromobacterium violaceum is a Gram‐negative bacterium that causes fatal septicaemia in humans and animals. C. violaceum ATCC 12472 possesses genes associated with two distinct type III secretion systems (T3SSs). One of these systems is encoded by Chromobacterium pathogenicity islands 1 and 1a (Cpi‐1/‐1a), another is encoded by Chromobacterium pathogenicity island 2 (Cpi‐2). Here we show that C. violaceum causes fulminant hepatitis in a mouse infection model, and Cpi‐1/‐1a‐encoded T3SS is required for its virulence. In addition, using C. violaceum strains with defined mutations in the genes that encode the Cpi‐1/‐1a or Cpi‐2 locus in combination with cultured mammalian cell lines, we found that C. violaceum is able to induce cytotoxicity in a Cpi‐1/‐1a‐dependent manner. Characterization of Chromobacterium‐induced cytotoxicity revealed that cell lysis by C. violaceum infection involves the formation of pore structures on the host cell membrane, as demonstrated by protection by cytotoxicity in the presence of osmoprotectants. Finally, we demonstrated that CipB, a Cpi‐1/‐1a effector, is implicated in translocator‐mediated pore formation and the ability of CipB to form a pore is essential for Chromobacterium‐induced cytotoxicity. These results strongly suggest that Cpi‐1/‐1a‐encoded T3SS is a virulence determinant that causes fatal infection by the induction of cell death in hepatocytes.  相似文献

A new species of siphonostomatoid copepod, Asterocheres crinoidicola, is parasitic on two closely related comasterid crinoids (Nemaster grandis and Davidaster rubiginosus) in Belize, Central America. An unusually long terminal prolongation of the third segment of the endopod of leg 1 distinguishes this species from all congeners. This is the first report of a copepod parasitic on a crinoid in the Caribbean.  相似文献

Six new fossils of Micromalthus (Coleoptera: Archostemata) from Dominican amber are compared with extant and previously described fossil micromalthid beetles. The amber inclusions are well preserved and all important morphological features are visible. Taking into account the morphological variability of the extant species Micromalthus debilis LeConte, 1878 , it is not possible to find any morphological features that distinguish the fossils from the extant species. This also applies to the Dominican amber inclusion described as Micromalthus anasi Perkovsky, 2008, which therefore is considered a junior synonym of M. debilis. The lack of morphological change in M. debilis over time might possibly be explained by unusually stable environmental conditions, as this species occupies a very specialized ecological niche in decaying timber. Background  

Chromobacterium violaceum is a free-living bacterium able to survive under diverse environmental conditions. In this study we evaluate the genetic and physiological diversity of Chromobacterium sp. isolates from three Brazilian ecosystems: Brazilian Savannah (Cerrado), Atlantic Rain Forest and Amazon Rain Forest. We have analyzed the diversity with molecular approaches (16S rRNA gene sequences and amplified ribosomal DNA restriction analysis) and phenotypic surveys of antibiotic resistance and biochemistry profiles.  相似文献

Pyxicephalus adspersus , is exceptional among living frogs. Embryonic development, larval phase, and metamorphosis can be completed in 17 days at a temperature of 29°C. The metamorphosis only takes 5 days. The present study shows that, despite the unusually short larval phase in P. adspersus, the state of skeletal differentiation reached at the end of metamorphosis is similar to that of other frog species. There is no shift of cranial bone formation postmetamorphosis as could have been expected and is known from other species. The majority of compared species are particularly similar in the sequence of bone formation in the postcranial skeleton. However, there are clear differences among species in the timing of these events relative to the larval growth trajectory, absolute time, and certain developmental markers, such as external limb differentiation. For example, skeletogenesis and externally visible limb differentiation are only loosely integrated. Interspecific comparisons show that, in P. adspersus, the early onset of skeletal ossification is an unusual feature among frogs. Freshly metamorphosed froglets of P. adspersus are already distinct from comparable stages of other species in having strong jaws, fang-like teeth, and a squamosal-maxilla contact. The latter stabilizes the maxillary arcade and the suspensorium and might relate to the ability to catch and swallow very large vigorous prey, such as siblings, shortly after metamorphosis. The presence of a complete set of dermatocranial elements and postmetamorphic ossification of only the sphenethmoid and operculum are considered plesiomorphic features, whereas the much less completely ossified skulls of metamorphosed froglets, particularly in Bufo and Hamptophryne, are likely apo- morphic developmental traits within the Anura. In this paper we discuss the aerobic biodegradation of calcium libnosulfonate (CLS) in a beechwood sulfite waste liquor by means of a mixed culture of microorganisms consisting of two Trichosporn Years and bacteria in the Arthrobacter (two species), Psedomonas, and Chromobacterium genera. Under the established parameters 50% CLS was biodegraded in 24 hr accompanied by the demethylation of methoxyl groups, the splitting of sulpher–carbon bonds, and the appearance of carbonyl and carboxyl groups. The achieved results by determination of phenolic OH groups, as well as established changes of the absorption bands of IR spectra of the CLS molecule and the results of the shortening of the analyses of the C, H, O, and S, show that the degradation of the aromatic nuclei-culture biodegradation, which confirms the increase in the concentration of conjugated carbonyl groups and carboxyl groups.  相似文献

A rich source of markers may be overlooked by screening for polymorphism in the source species only. We screened 129 microsatellite loci isolated from the powerful owl (Ninox strenua) against two closely related species; Ninox connivens and Ninox novaeseelandiae. From the screening effort 20 polymorphic markers were isolated, including six loci which were originally discarded as they were monomorphic in the source species. Further cross-species amplification of all 20 loci across species from two families, Strigidae and Tytonidae, revealed unusually high levels of polymorphism within closely related species, and limited success within phylogenetically distant species. Routine screening of multiple species during the marker development phase can yield a wider range of polymorphic markers which can subsequently enhance cross-species amplification attempts.  相似文献

Summary The sensitivity of twenty six microbial lipases towards acetaldehyde (an unavoidable by-product in lipase-catalysed acyl transfer reactions with vinyl esters) was investigated. The sensitivity of an individual enzyme strongly depends on its properties such as microbial source, molecular weight and relative lysine content. Whereas the majority of enzymes (from Pseudomonas, Rhizopus, Chromobacterium, Mucor and Candida antarctica sp.) proved to be remarkably stable, lipases from Candida rugosa and Geotrichum candidum lost most of their activity when exposed to acetaldehyde.  相似文献

Laurencia Lamouroux (Rhodophyta) was recently separated into three genera–Laurencia, Chondrophycus (Tokida et Saito) Garbary et Harper and Osmundea Stackhouse – each of which was newly defined based on vegetative and reproductive structures. In this study, the previously unknown vegetative and reproductive morphology of two Australian endemic species of Laurencia, L. clavata Sonder and L. elata (C. Agardh) Harvey, was studied, particularly in the context of the revised generic delineation. These species exhibit vegetative axes with four pericentral cells and trichoblast-type spermatangial development. Tetrasporangia are abaxially produced from the existing third and fourth pericentral cells. L. clavata has terete thalli with distinctive verticillate branching and is similar to Chondria C. Agardh, rather than to Laurencia, in having an unusually marked constriction at the base of the branches and starch accumulation in subcortical and medullary cells. Compared to Laurencia, apical cells of this species exhibit a less oblique division ; the resulting recognizable axial cell rows extend somewhat below the branches, and particularly at a young stage they are also clear throughout branchlets. However, other vegetative and spermatangial structures show that L. clavata is more closely allied to Laurencia than to Chondria, and it is placed in Laurencia. By contrast, L. elata exhibits morphology typical of Laurencia and is characterized by large, robust, compressed thalli with fastigiately distichous branching and an extensive secondary cortex. Furthermore, it appears to be distinct from similar species in sometimes having a parasitic species of Janczewskia Solms-Laubach (Rhodophyta). Taxonomy of Laurencia is discussed on the basis of these and previous studies.  相似文献

 The cowpea [Vigna unguiculata (L.) Walp.] is a morphologically and genetically variable species composed of wild perennial, wild annual, and cultivated forms that are mainly used for edible seeds and pods. In this study, genetic variation in 199 germplasm accessions of wild and cultivated cowpea was evaluated using an allozyme analysis. The results from this survey showed that wild cowpea exhibits genetic variation perfectly fitted with the existing morphological classification. The cowpea gene-pool is characterized by its unusually large size. It encompasses taxa (ranked as subspecies) that could be considered as different species considering the high genetic distances observed between accessions belonging to different taxa. These subspecies can be classified into three groups characterized by their breeding systems: perennial outcrossers, perennial out-inbreds, and inbred annuals. Allozyme data confirm this grouping. Perennial outcrossers look primitive and are more remote from each other and from perennial out-inbreds. Within this large gene-pool, mainly made of perennial taxa, cultivated cowpeas (ssp. unguiculata var. unguiculata) form a genetically coherent group and are closely related to annual cowpeas (ssp. unguiculata var. spontanea) which may include the most likely progenitor of cultivated cowpeas. Chromobacterium lividum oxidizes gluconate to 2,5-diketogluconate via 2-ketogluconate. The conversion by either growing or resting cells proceeds best in an aerated medium at pH 5.9. The yield is nearly 50%. Diketogluconate is not further decomposed. The preparation of chromatographically pure diketogluconate using eitherChromobacterium orGluconobacter is described.
