The effects of tomato and bean rhizospheres on hyphal spreading of the arbuscular mycorrhizal (AM) fungus Glomus mosseae were studied using a soil compartment system in combination with hydrophobic polytetrafluorethylene (PTFE) membranes. Both the nylon screen and the PTFE membrane were freely permeable to hyphae but not to roots. Furthermore, the hydrophobic PTFE membrane seemed to be a barrier to the flux of soil solutions containing root exudates. The results show that water soluble exudates of tomato and bean roots greatly stimulate hyphal growth in the soil compartment system used. Moreover, water soluble root exudates of bean exert a clear attractional effect on AM hyphal growth.  相似文献

【背景】植物体内普遍存在一定数量的内生放线菌,对植物的生长发育具有促生作用。【目的】从番茄根内分离、筛选并鉴定出能够有效促进植物生长发育的内生放线菌,为生物菌肥的开发奠定基础。【方法】采用组织研磨培养法和放线菌分离培养基对番茄根内放线菌进行分离,利用Salkowski比色法、钼锑抗比色法和CAS平板检测法进一步筛选出具有较强促生特性的菌株,通过番茄和黄瓜苗期盆栽试验验证其促生效果。结合形态、生理生化以及16S r RNA基因序列相似性和系统发育分析,对菌株进行鉴定。【结果】分离筛选出一株吲哚乙酸(Indole-3-acetic acid,IAA)产量达25.56 mg/L的内生放线菌NEAU-D1,能够产生铁载体并且对多种难溶性磷酸盐具有良好溶解效果,通过16S r RNA基因序列分析,该菌株属链霉菌属。番茄和黄瓜苗期盆栽试验结果表明,接种该菌株的番茄幼苗其根长、株高、植株鲜重和干重较对照分别显著增加了9%、23%、47%和92%,而接种该菌株的黄瓜幼苗根长、株高、植株鲜重和干重较对照分别显著增加了43%、47%、134%和58%。【结论】链霉菌NEAU-D1可以作为潜在的促生菌资源用于设施蔬菜多功能生物菌肥的研发。  相似文献

Wind disturbance as a green method can effectively prevent the overgrowth of tomato seedlings, and its mechanism may be related to root system mechanics. This study characterized the biophysical mechanical properties of taproot and lateral roots of tomato seedlings at five seedling ages and seedling substrates with three different moisture content. The corresponding root system-substrate finite element (FE) model was then developed and validated. The study showed that seedling age significantly affected the biomechanical properties of the taproot and lateral roots of the seedlings and that moisture content significantly affected the biomechanical properties of the seedling substrate (p < 0.05). The established FE model was sensitive to wind speed, substrate moisture content, strong seedling index, and seedling age and was robust. The multiple linear regression equations obtained could predict the maximum stress and strain of the root system of tomato seedlings in the wind field. The strong seedling index had the greatest impact on the biomechanical response of the seedling root system during wind disturbance, followed by wind speed. In contrast, seedling age had no significant effect on the biomechanical response of the root system during wind disturbance. In the simulation, no mechanical damage was observed on the tissue of the seedling root system, but there were some strain behaviors. Based on the plant stress resistance, wind disturbance may affect the growth and development of the root system in the later growth stage. In this study, finite element and statistical analysis methods were combined to provide an effective approach for in-depth analysis of the biomechanical mechanisms of wind disturbances that inhibit tomato seedlings’ growth from the root system’s perspective.  相似文献

Transformed roots of carrot were used to determine the effects of root metabolites on hyphal development from spores of the vesicular-arbuscular mycorrhizal fungus Gigaspora margarita. Hyphal growth of this obligately biotrophic symbiont was greatly stimulated by a synergistic interaction between volatile and exudated factors produced by roots. Root volatiles alone provided little stimulation, and root exudates alone had no effect. For the first time, carbon dioxide was demonstrated to be a critical root volatile involved in the enhancement of hyphal growth. C-labeled root volatiles were fixed by the fungus and thus strongly suggested that CO(2) served as an essential carbon source.  相似文献

Five strains of Trichoderma with known biocontrol activities were assessed for their effect upon pea growth and their antagonistic activity against large Pythium ultimum inocula. The effect of Trichoderma inocula upon the indigenous soil microflora and soil enzyme activities in the presence and absence of Pythium is assessed. In the absence of Pythium, Trichoderma strain N47 significantly increased the wet shoot weight by 15% but did not significantly affect the dry weight, whilst strains T4 and N47 significantly increased the root weights by 22% and 80%) respectively. Strains TH1 and N47 resulted in significantly greater root lengths. Pythium inoculation significantly reduced the root length and the number of lateral roots and nodules, and significantly increased the root and rhizosphere soil fungal populations. Pythium inoculation significantly reduced the plant wet and dry shoot weights and significantly increased the wet and the dry shoot/root ratio. All the Trichoderma strains reduced the number of lesions caused by Pythium and increased the number of lateral roots. The effect of the Pythium on emergence and shoot growth was significantly reduced by all the Trichoderma strains except strain To10. Inoculation with Trichoderma strains TH1 and T4 resulted in significantly greater wet root weights (62% and 57%, respectively) in the presence of Pythium compared to the Pythium control. Strain N47 significantly increased the shoot/root ratio compared to the Pythium control. Inoculation with Trichoderma strains T4, T12 and N47 significantly reduced Pythium populations. Pythium increased the activity of C, N and P cycle enzymes, whilst four Trichoderma strains reduced this effect, indicating reduced plant damage and C leakage. Overall, strains T4 and N47 had the greatest beneficial characteristics, as both these strains improved plant growth in the absence of Pythium and reduced plant damage in the presence of Pythium. The dual properties of these strains improve the commercial application, giving them an advantage over single action inocula, especially in the absence of plant pathogens.  相似文献

The colonization process of tomato roots inoculated separately or/and simultaneously by a pathogenic Fusarium oxysporum f. sp. lycopersici strain Fol8 and the protective F. oxysporum strain Fo47, genetically tagged with the red and green fluorescent protein genes, respectively, was studied in a hydroponic culture. Plants were coinoculated with Fol8 and Fo47 at two conidial concentration ratios of 1/1 and 1/100, in which biological control was not effective or effective, respectively. First observation of fungi on root was possible 48 h after inoculation at a high inoculum level and 5 days post inoculation at the lower concentration of inoculum. The pattern of root colonization was similar for both strains with the initial development of hyphal network on the upper part of taproot, followed by the growth of hyphae towards the elongation zone, lateral roots and root apices. Finally, the whole elongation zone and root apex were invaded by both strains but no specific infection sites were observed. When coinoculated, both strains could grow very closely or even at the same spot on the root surface. At the nonprotective ratio, Fol8 was the successful colonizer, but application of Fo47 at a concentration 100 times >Fol8 delayed vessel colonization by the pathogen.  相似文献

The survival and colonization patterns of Pseudomonas putida PRD16 and Enterobacter cowanii PRF116 in the rhizosphere of greenhouse-grown tomato plants and the effects of their inoculation on the indigenous bacterial community were followed by selective plating, molecular fingerprinting, and confocal laser scanning microscopy (CLSM) over 3 weeks. Both strains, which showed in vitro antagonistic activity against Ralstonia solanacearum, were previously tagged with gfp. Seed and root inoculation were compared. Although plate counts decreased for both gfp-tagged antagonists, PRD16 showed a better survival in the rhizosphere of tomato roots independent of the inoculation method. Analysis of 16S rRNA gene fragments amplified from total community DNA by denaturing gradient gel electrophoresis and CLSM confirmed the decrease in the relative abundance of the inoculant strains. Pronounced differences in the Pseudomonas community patterns for plants inoculated with PRD16 compared to the control were detected 3 weeks after root inoculation, indicating a longer-lasting effect. Analysis by CLSM showed rather heterogeneous colonization patterns for both inoculant strains. In comparison with seed inoculation, root inoculation led to a much better colonization as evidenced by all three methods. The colonization patterns observed by CLSM provide important information on the sampling strategy required for monitoring inoculant strains in the rhizosphere.  相似文献

AIMS: Four well-described strains of Pseudomonas fluorescens were assessed for their effect on pea growth and their antagonistic activity against large Pythium ultimum inocula. Methods and RESULTS: The effect of Pseudomonas strains on the indigenous soil microflora, soil enzyme activities and plant growth in the presence and absence of Pythium was assessed. Pythium inoculation reduced the shoot and root weights, root length, and the number of lateral roots. The effect of Pythium was reduced by the Pseudomonas strains. Strains F113, SBW25 and CHAO increased shoot weights (by 20%, 22% and 35%, respectively); strains Q2-87, SBW25 and CHAO increased root weights (14%, 14% and 52%). Strains SBW25 and CHAO increased root lengths (19% and 69%) and increased the number of lateral roots (14% and 29%). All the Pseudomonas strains reduced the number of lesions and the root and soil Pythium populations, while SBW25 and CHAO increased the number of lateral roots. Pythium inoculation increased root and soil microbial populations but the magnitude of this effect was Pseudomonas strain-specific. Pythium increased the activity of C, N and P cycle enzymes, while the Pseudomonas strains reduced this effect, indicating reduced plant damage. CONCLUSION: Strains SBW25 and CHAO had the greatest beneficial characteristics, as these strains produced the greatest reductions in the side effects of Pythium infection (microbial populations and enzyme activities) and resulted in significantly improved plant growth. Strain SBW25 does not produce antifungal metabolites, and its biocontrol activity was related to a greater colonization ability in the rhizosphere. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first critical comparison of such important strains of Ps. fluorescens showing disease biocontrol potential.  相似文献

Pochonia chlamydosporia (Pc123) is a fungal parasite of nematode eggs which can colonize endophytically barley and tomato roots. In this paper we use culturing as well as quantitative PCR (qPCR) methods and a stable GFP transformant (Pc123gfp) to analyze the endophytic behavior of the fungus in tomato roots. We found no differences between virulence/root colonization of Pc123 and Pc123gfp on root-knot nematode Meloidogyne javanica eggs and tomato seedlings respectively. Confocal microscopy of Pc123gfp infecting M. javanica eggs revealed details of the process such as penetration hyphae in the egg shell or appressoria and associated post infection hyphae previously unseen. Pc123gfp colonization of tomato roots was low close to the root cap, but increased with the distance to form a patchy hyphal network. Pc123gfp colonized epidermal and cortex tomato root cells and induced plant defenses (papillae). qPCR unlike culturing revealed reduction in fungus root colonization (total and endophytic) with plant development. Pc123gfp was found by qPCR less rhizosphere competent than Pc123. Endophytic colonization by Pc123gfp promoted growth of both roots and shoots of tomato plants vs. uninoculated (control) plants. Tomato roots endophytically colonized by Pc123gfp and inoculated with M. javanica juveniles developed galls and egg masses which were colonized by the fungus. Our results suggest that endophytic colonization of tomato roots by P. chlamydosporia may be relevant for promoting plant growth and perhaps affect managing of root-knot nematode infestations.  相似文献

The direct impact of fenpropimorph on the sterol biosynthesis pathway of Glomus intraradices when extraradical mycelia alone are in contact with the fungicide was investigated using monoxenic cultures. Bi-compartmental Petri plates allowed culture of mycorrhizal chicory roots in a compartment without fenpropimorph and exposure of extraradical hyphae to the presence of increasing concentrations of fenpropimorph (0, 0.02, 0.2, 2, 20 mg l−1). In the fungal compartment, sporulation, hyphal growth, and fungal biomass were already reduced at the lowest fungicide concentration. A decrease in total sterols, in addition to an increase in the amount of squalene and no accumulation of abnormal sterols, suggests that the sterol pathway is severely slowed down or that squalene epoxidase was inhibited by fenpropimorph in G. intraradices. In the root compartment, neither extraradical and intraradical development of the arbuscular mycorrhizal (AM) fungus nor root growth was affected when they were not in direct contact with the fungicide; only hyphal length was significantly affected at 2 mg l−1 of fenpropimorph. Our results clearly demonstrate a direct impact of fenpropimorph on the AM fungus by a perturbation of its sterol metabolism.  相似文献

We investigated the growth and nutrient uptake of the Lycopersicon esculentum symbiosis mycorrhiza-defective plant mutant rmc, challenged with arbuscular mycorrhiza (AM) fungal propagules, in the presence or absence of roots of the commercial wild-type tomato cv. Golden Queen (GQ). Two plants shared the middle (combi) compartment of a horizontal three-compartment split-root pot with one part of their root system; the other part was grown separately in an outer (solo) pot. Combinations of rmc and GQ plants were grown together in soil that was either mycorrhiza-free (-M) or prepared with AM fungal inoculum (+M). Surface colonization of rmc roots was strongly increased in the presence of (+M) GQ roots. AM fungal inoculation increased phosphorus uptake of GQ plants, but decreased growth and P uptake of rmc plants. Growth and P uptake of (+M) GQ plants were reduced when plants were grown in combination with rmc rather than another GQ plant. AM fungi in the (combi) compartment may have preferentially formed hyphae spreading infection rather than functioning in P uptake in (+M) GQ plants grown in combination with rmc. Surface colonization of (+M) rmc roots, in the presence of GQ roots, was probably established at the expense of carbohydrates from associated GQ plants. Possible reasons for a decreased P uptake of rmc plants in response to AM fungal inoculation are proposed.  相似文献

Summary Aqueous extracts of Western Hemlock roots were tested for the ability to stimulate initiation of growth from carpophore tissue of certain mycorrhizal-associated fungi. Of numerous species tested, onlyRozites caperatus andInocybe napipes responded slightly to this treatment, initiating a feeble mycelial development from gill tissue fragments.To determine whether exogenous NAD or young, excised roots would stimulate the growth of certain mycorrhizal fungi, species ofAmanita, Tricholoma, andXerocomus were subcultured in a rich basal medium, with and without added NAD or excised tomato roots. Spore germination tests on mycorrhizal-associated species not previously subcultured were conducted in a similar fashion.Added NAD stimulated the growth ofLeucopaxillus amarus but inhibited development ofArmillaria zelleri. It was without pronounced effect on the other cultures. In the presence of excised tomato roots, growth of theLeucopaxillus species was also stimulated, butArmillaria zelleri was inhibited slightly andXerocomus chrysenteron almost completely. Both treatments were without essential effect on the other species tested.No positive results were obtained with either added NAD or excised tomato roots in the spore germination studies.  相似文献

The objective of the current investigation was to develop a reliable method to obtain vesicular arbuscular mycorrhizae (VAM) in micropropagated plantlets and to determine their influence on growth. An in vitro system for culturing the VA mycorrhizal fungus Glomus intraradices with Ri T-DNA-transformed carrot roots or nontransformed tomato roots was used in this study as a potential active source of inoculum for the colonization of micropropagated plantlets. After root induction, micropropagated plantlets grown on cellulose plugs (sorbarod) were placed in contact with the primary mycorrhizae in growth chambers enriched with 5000 ppm CO2 and fed with a minimal medium. After 20 days of tripartite culture, all plantlets placed in contact with the primary symbiosis were colonized by the VAM fungus. As inoculum source, 30-day-old VA mycorrhizal transformed carrot roots had a substantially higher infection potential than 5-, 10-or 20-day-old VAM. Colonized plantlets had more extensive root systems and better shoot growth than control plants. The VAM symbiosis reduced the plantlet osmotic potential. This response may be a useful pre-adaptation for plantlets during transfer to the acclimatization stage.  相似文献

Summary Excised tomato roots (Lycopersicon esculentum Mill. cv Bonny Best) were cultured in the presence of mannitol to determine the effects of varying degrees of mild water deficit on their developmental growth. It was found that over the 7-d culture period, the cultured roots could regulate their own developmental responses to the water deficit such that elongation of the primary root axis was favored over that of the lateral roots. Higher degrees of water deficit proportionately decreased lateral root number and density, but lateral root primordia (visualized by clearing roots in chromium trioxide) continued to be formed in water-stressed roots. Measurements of water and osmotic (solute) potentials of the root tips showed that the cultured roots osmoregulated and did not suffer a loss in turgor pressure as a result of the mannitol treatments. However, reciprocal transfer experiments showed that root cultures were unable to resume growth after removal from water deficit conditions, thus indicating a probable requirement for the shoot for complete recovery.  相似文献

Root respiration has been shown to increase with temperature, but less is known about how this relationship is affected by the fungal partner in mycorrhizal root systems. In order to test respiratory temperature dependence, in particular Q (10) of mycorrhizal and non-mycorrhizal root systems, seedlings of PICEA ABIES (L.) Karst. (Norway spruce) were inoculated with the ectomycorrhizal fungus PILODERMA CROCEUM (Eriksson and Hjortstam, SR430; synonym: PILODERMA FALLAX: [Libert] Stalpers) and planted in soil respiration cuvettes (mycocosms). Temperature dependence of hyphal respiration in sterile cultures was determined and compared with respiration of mycorrhizal roots. Respiration rates of mycorrhizal and non-mycorrhizal root systems as well as sterile cultures were sensitive to temperature. Q (10) of mycorrhizal root systems of 3.0 +/- 0.1 was significantly higher than that of non-mycorrhizal systems (2.5 +/- 0.2). Q (10) of P. CROCEUM in sterile cultures (older than 2 months) was similar to that of mycorrhizal root systems, suggesting that mycorrhizae may have a large influence on the temperature sensitivity of roots in spite of their small biomass. Our results stress the importance of considering mycorrhization when modeling the temperature sensitivity of spruce roots.  相似文献

Excised root systems of tomato plants (early fruiting stage, 2nd flush) were subjected to a gradual transition from normoxia to anoxia by seating the hydroponic root medium while aeration was stopped. Oxygen level in the medium and respiration rate decreased and reached very low values after 12 h of treatment, indicating that the tissues were anoxic thereafter. Nitrate loss from the nutrient solution was strongly stimulated by anoxia (after 26 h) concomitantly with a release of nitrite starting only after 16 h of treatment. This effect was not observed in the absence of roots or in the presence of tungstate, but occurred with whole plants or with sterile in vitro cultured root tissues. These results indicate that biochemical processes in the root involve nitrate reductase. NR activity assayed in tomato roots increased during anoxia. This phenomenon appeared in intact plants and in root tissues of detopped plants. The stimulating effect of oxygen deprivation on nitrate uptake was specific; anoxia simultaneously entailed a release of orthophosphate, sulfate, and potassium by the roots. Anoxia enhanced nitrate reduction by root tissues, and nitrite ions were released into xylem sap and into medium culture. In terms of the overall balance, the amount of nitrite recovered represented only half of the amount of nitrate utilized. Nitrite reduction into nitric oxide and perhaps into nitrogen gas could account for this discrepancy. These results appear to be the first report of an increase in nitrate uptake by plant roots under anoxia of tomato at the early fruiting stage, and the rates of nitrite release in nutrient medium by the asphyxiated roots are the fastest yet reported.  相似文献

The effect of root temperature on growth and yield of rockwool-grown tomato plants infected with Phytophthora cryptogea was investigated. Measurements of shoot and root growth were taken at high (25oC) and low (15oC) root temperatures during the generative phase of growth. The growth of roots of healthy and P. cryptogea-infected tomato plants in rockwool blocks was higher in plants grown with roots at 25oC than at 15oC after 60 days and a similar effect was found in slabs after 98 days. Under sub-optimal conditions for growth the disease became severe when root temperatures were low. Growth of roots was greatest when roots were maintained at a high temperature in combination with an ambient air temperature of c. 15oC and the response was greater in cv. Counter than cvs Calypso and Marathon. Water-soluble carbohydrates of roots were higher in those produced in blocks than slabs and were reduced by infection compared to healthy plants with roots at 15oC and 25oC. Reduced transpiration rates were found 17 days after inoculation in symptomless plants grown at a root temperature of 25oC. Infection, regardless of the temperature of the roots or cultivar, led to reduced stem growth. The plants grown at 25oC were taller than those with a root temperature of 15oC. After 9 wk of harvest, the cumulative fruit yields in infected cvs Counter and Calypso grown at 25oC were comparable to that in healthy plants grown at either temperature and cumulative fruit numbers followed a similar pattern. High root temperatures led to delayed fruit ripening between weeks 3–10 and a larger number of unripe fruit. The weight of unripe fruit from infected plants grown at 25oC at the terminal harvest was higher than from healthy plants with roots maintained at 15oC.  相似文献
