Survival of four intergravel development phases of rainbow trout, Salmo gairdneri Rich., in a dewatered environment varied with stage of development, duration of exposure, and relative humidity. Egg phases were considerably more tolerant than alevin phases. Cleavage eggs tolerated dewatering for 12 consecutive days (97% survival), the maximum period tested. Embryos tolerated dewatering until they began hatching on the eight day, when mortalities sharply increased. In contrast, eleutheroembryos had only 43% survival when dewatered 6h and pre-emergent alevins had only 36% survival when dewatered 2h. A few dewatered embryos died when relative humidity was 100%, but mortality increased significantly at 90 and 80% relative humidity. Survival of alevin phases, but not egg phases, in residual, non-flowing water was limited by formation of oxygen deficits.  相似文献

Floc destruction and its impacts on the dewatering properties in terms of capillary suction time (CST) in the process of using flat-sheet membrane for simultaneous thickening and digestion (MSTD) of waste activated sludge were investigated. A pilot-scale MSTD reactor was used to treat waste activated sludge, and extracellular polymeric substances (EPS), soluble biopolymers, dissolved cations and the dewatering properties were measured. The results indicated that the destruction mechanisms of the MSTD process could be divided into two phases due to the variation of dissolved oxygen (DO). Polysaccharides and proteins were released in both phases, but the release patterns were different. The concentration of polysaccharides was much greater than that of proteins in Phase 1, while the ratio of proteins to polysaccharides ranged from 1.5 to 1.7. The divalent cations in supernatant significantly increased in Phase 1, while the monovalent cations in supernatant rose in Phase 2. The dewatering properties in terms of CST significantly increased in the MSTD process, and the mixed liquor suspended solids (MLSS), particle size, and biopolymers in supernatant had significant effects on the dewatering properties.  相似文献

Temporary dewatering constitutes a drastic change in conditions for aquatic vegetation. Species’ sustained performance under these conditions relies partly on their ability to produce a terrestrial phenotype. Such adaptations may include the development of self-supporting aboveground organs with higher dry matter content enabling plants to withstand gravity and smaller leaves with thicker cuticle to reduce evapotranspiration, leading to lower specific leaf area, higher leaf-construction costs and consequently higher leaf life span. The ability of aquatic plant species to produce a terrestrial-adapted phenotype may differ according to growth form and evolutionary history. The objectives of this study were to (1) measure the effects of dewatering on aquatic plant performance, (2) determine how growth form and phylogenetic position affect performance, and (3) relate plant performance to plasticity. To meet these objectives, we experimentally studied aquatic plant responses to dewatering by measuring survival, growth, and a set of traits describing the morphology and leaf-resource economy of eight aquatic plant species with contrasting phylogeny and growth forms. The ability of aquatic plants to withstand dewatering differed according to phylogeny but not to growth form. The eudicots presented high survival and similar growth rates under terrestrial compared to aquatic conditions, while monocots generally did not survive dewatering. These species produced phenotypic adjustments, such as denser aboveground organs and leaf plasticity, which can explain the maintenance of similar growth rates under terrestrial conditions. The relatively strong plasticity and performance of eudicots in terrestrial habitats suggests that their optimal niche is the interface between aquatic and terrestrial ecosystems.  相似文献

The gravel commonly used to cover parking lots and roadways at retail agrichemical facilities may contain relatively large concentrations of pesticides that resulted from past management problems. These pesticides may threaten groundwater quality. Previous studies, however, suggested that the pesticides had not moved from the gravel in several sample profiles. Excavations at a closed facility revealed tremendous variability in pesticide distribution within the site. Pesticides were present below the gravel in two profiles, but the mechanism(s) for their movement were not clear. The objectives of this study were to investigate how the physical and chemical properties of the gravel influence the environmental fate of atrazine. All of the gravel samples collected and characterized contained atrazine and sufficient organic C to adsorb significant amounts of atrazine, thus retarding its movement through the gravel. Laboratory column leaching experiments, however, suggested that much of the atrazine should leach from the gravel within a year or two. A field-scale test plot was constructed to study how atrazine moves through the gravel under controlled conditions. Atrazine was “spilled” in the test plot. Atrazine moved from the gravel both vertically and horizontally. It appears that formulated product spilled on gravel will leach. A single discrete spill can give rise to phantom spills whose occurrence and distribution is not related to any specific pesticide-management practice. The apparent lack of atrazine leaching from gravel appeared to be a transient phenomenon and/or the result of sampling limitations in previous studies. The contaminated gravel clearly poses a risk to groundwater quality.  相似文献

Recent years have seen a dramatic rise in fermentation broth cell densities and a shift to extracellular product expression in microbial cells. As a result, dewatering characteristics during cell separation is of importance, as any liquor trapped in the sediment results in loss of product, and thus a decrease in product recovery. In this study, an ultra scale-down (USD) approach was developed to enable the rapid assessment of dewatering performance of pilot-scale centrifuges with intermittent solids discharge. The results were then verified at scale for two types of pilot-scale centrifuges: a tubular bowl equipment and a disk-stack centrifuge. Initial experiments showed that employing a laboratory-scale centrifugal mimic based on using a comparable feed concentration to that of the pilot-scale centrifuge, does not successfully predict the dewatering performance at scale (P-value <0.05). However, successful prediction of dewatering levels was achieved using the USD method (P-value ≥0.05), based on using a feed concentration at small-scale that mimicked the same height of solids as that in the pilot-scale centrifuge. Initial experiments used Baker's yeast feed suspensions followed by fresh Pichia pastoris fermentation cultures. This work presents a simple and novel USD approach to predict dewatering levels in two types of pilot-scale centrifuges using small quantities of feedstock (<50 mL). It is a useful tool to determine optimal conditions under which the pilot-scale centrifuge needs to be operated, reducing the need for repeated pilot-scale runs during early stages of process development.  相似文献

SUMMARY 1. Communities of aquatic macroinvertebrates and the terrestrial adult phases of aquatic insects were investigated from short stretches of English chalk streams with two different bankside vegetation types: simply structured grazed grass (grazed) and structurally complex herbaceous vegetation with scattered trees (ungrazed). Macroinvertebrates were sampled in spring, summer, autumn and winter 1996–97 from three aquatic habitats: mid-channel gravel, patches of the aquatic macrophyte Ranunculus and marginal emergent macrophytes. The terrestrial adult phases of aquatic insects were sampled in spring, summer and autumn from bankside vegetation.
2. Total macroinvertebrate abundance did not differ between stretches with different bankside vegetation. Taxon richness of mid-channel gravel was, however, significantly higher in ungrazed compared with grazed stretches and Shannon diversity ( H ') of mid-channel gravel and marginal vegetation was significantly higher in ungrazed compared with grazed stretches. Total abundance, taxon richness and Shannon diversity ( H ') of the terrestrial adult phases of aquatic insect were significantly higher from the bankside vegetation of ungrazed compared with grazed stretches.
3. Ordination of communities of aquatic macroinvertebrates and terrestrial adults demonstrated that individual families of both groups were generally more abundant in ungrazed stretches. Many more families were significantly associated with ungrazed stretches than with grazed stretches.
4. This investigation has shown that high structural diversity of bankside vegetation along lowland chalk streams is accompanied at the reach scale by increased diversity of both aquatic macroinvertebrates and the terrestrial adult phases of aquatic insects. The conservation potential of such streams may thus be lowered by management practices that result in the removal or simplification of bankside vegetation along extensive stream stretches.  相似文献

Pichia pastoris is becoming a desirable host in the biopharmaceutical industry for therapeutics production. It grows on methanol to high cell densities ≥100 g DCW/L and secretes foreign proteins at high titers. However, the culture conditions to reach high cell densities pose a challenge to the processability by primary recovery operations, in particular centrifugation, used for cell removal. This work aims to assess the impact of recombinant P. pastoris strain selection on centrifugal dewatering. Normally, the choice of P. pastoris recombinant strain is based on best target protein expression levels; however, it is unknown whether the choice of strain will have an impact on performance of centrifugation operation. To achieve this aim, a previously developed laboratory ultra‐scale down (USD) methodology that successfully predicted centrifugal dewatering of pilot‐scale disk‐type machines, was used in this work. Two recombinant P. pastoris strains, namely a X‐33 and a glycoengineered Pichia strain, were used to perform fermentations secreting different products. The resulting harvested fermentation culture properties were analyzed and the dewatering performances of a pilot‐ and a large‐scale disk‐type centrifuge were evaluated using the USD methodology. The choice of P. pastoris strain was found to have a considerable impact on dewatering performance, with P. pastoris X‐33 strain reaching better dewatering levels than the glycoengineered strain. The USD method proved to be a useful tool to determine optimal conditions under which the large scale centrifuge needed to be operated, reducing the need for repeated pilot‐scale runs during early stages of process development for therapeutic products. © 2012 American Institute of Chemical Engineers Biotechnol. Of the dewatering characteristics of activated sludges in our previous paper, the dewatering rate of sludge decreased in proportion to increasing amounts of extracellular polymer. As extracellular polymer in activated sludge was one of the important factors in the dewatering process, the change of the dewatering characteristics of thermochemically treated sludge (containing extracellular polymer) were compared with that of extracellular polymer extracted from sludge. The ultrafiltration rate of extracellular polymer flocculated by thermochemical treatment was much faster than that without treatment, indicating improved dewatering characteristics. Under the same treatment conditions, the dewatering characteristics of sludge were also much improved. The addition of the extracellular polymer treated thermochemically below pH 3 had no effect on the dewatering characteristics of the sludge. The addition of a flocculant to the thermochemically treated sludge was found to further improve the dewatering characteristics. The thermochemical treatment under low pH condition facilitated the flaking of the cake from the filter.  相似文献

Timing of breeding and offspring size are maternal traits that may influence offspring competitive ability, dispersal, foraging, and vulnerability to predation and climatic conditions. To quantify the extent to which these maternal traits may ultimately affect an organism's fitness, we undertook laboratory and field experiments with Atlantic salmon (Salmo salar). To control for confounding effects caused by correlated traits, manipulations of the timing of fertilization combined with intraclutch comparisons were used. In the wild, a total of 1462 juveniles were marked at emergence from gravel nests. Recapture rates suggest that up to 83.5% mortality occurred during the first four months after emergence from the gravel nests, with the majority (67.5%) occurring during the initial period ending 17 days after median emergence. Moreover, the mortality was selective during this initial period, resulting in a significant phenotypic shift toward an earlier date of and an increased length at emergence. However, no significant selection differentials were detected thereafter, indicating that the critical episode of selection had occurred at emergence. Furthermore, standardized selection gradients indicated that selection was more intense on date of than on body size at emergence. Timing of emergence had additional consequences in terms of juvenile body size. Late-emerging juveniles were smaller than early-emerging ones at subsequent samplings, both in the wild and in parallel experiments conducted in seminatural stream channels, and this may affect success at subsequent size-selective episodes, such as winter mortality and reproduction. Finally, our findings also suggest that egg size had fitness consequences independent of the effects of emergence time that directly affected body size at emergence and, in turn, survival and size at later life stages. The causality of the maternal effects observed in the present study supports the hypothesis that selection on juvenile traits may play an important role in the evolution of maternal traits in natural populations.  相似文献

Fluvial aquatic systems fluctuate naturally across a broad range of time‐scales. Land management activities impose further extreme fluctuations, particularly when water is abstracted for irrigation during summer. This study focused on the interaction between Atlantic salmon and brown trout parr during acute dewatering events. Brown trout are known to compete strongly for pool habitat whereas salmon can coexist by using riffle areas during normal flows. It is not known, however, how competition between the species affects their behavioural responses to extreme low flows when riffle areas decrease. Replicated groups of salmon were held in allopatry and sympatry with trout in sections of a large indoor stream, each of which was landscaped into riffles and pools. Space use and behaviours of the fish were recorded by direct visual observations and a network of Passive Integrated Transponder detectors. The use of a scroll decanter centrifuge for the removal and dewatering of affinity-flocculated yeast cell debris from a crude homogenate is described. Laboratory shear modulus measurements were used to compare the structure of flocculated and nonflocculated sediments and to indicate the dewatering conditions under which the sediment could be discharged from the centrifuge. The structure of the flocculated sediment was such that a dry beach could be used within the centrifuge while still being able to discharge the solids. The scroll decanter performance for recovery and dewatering of the flocculated homogenate was found to be independent of feed flow rate and differential scroll rate. Eighty-five percent of the solid material was recovered from the flocculated homogenate while the extent of sediment dewatering resulted in the loss of only 7% of the soluble protein in the sediment. The supernatant clarity matched that achieved by low-gravity laboratory centrifugation studies.  相似文献

为探究砾石对红壤工程堆积体边坡降雨侵蚀的影响,基于室内模拟降雨试验,在1.0、1.5、2.0、2.5 mm/min雨强条件下,以土质边坡为对照,研究了10%、20%和30%的砾石质量分数红壤工程堆积体边坡的径流特征、产沙过程及侵蚀动力机制。结果表明:1)砾石质量分数对稳定径流强度的影响存在阈值,在雨强 > 1.0 mm/min时稳定径流强度随砾石质量分数的增加先减小后增大,在1.0 mm/min雨强时则表现为先增大后减小,且均在10%砾石质量分数下达到极值;2)雨强1.0 mm/min时,径流为缓流,砾石促进径流流动,使弗汝德数增大24.5%-87.8%,雨强2.0、2.5 mm/min时,径流为急流,砾石延缓径流流动,使弗汝德数降低4.2%-13.0%;3)侵蚀速率随产流历时变化过程受砾石质量分数和雨强的影响,雨强越大,砾石质量分数越低,边坡越易发生细沟侵蚀且伴随重力崩塌现象,侵蚀速率呈多峰多谷变化趋势;4)1.0 mm/min雨强时,砾石存在加剧了土壤侵蚀,产沙增幅达28.7%-50.5%;雨强 > 1.0 mm/min时,砾石减沙效益为5.0%-64.4%;5)径流功率是描述红壤工程堆积体侵蚀动力机制最优参数,其可蚀性参数及发生侵蚀临界径流功率从大到小对应的砾石质量分数均为10%、0、20%和30%。研究成果可为红壤区开发建设项目水土流失治理及侵蚀预测模型提供科学依据。  相似文献

The meiofauna was studied from springs in Finland which wascompletely ice-covered during the last glacial epoch. Of the31springs that were examined, 15 were in areas considered to bein anatural state and 16 had catchment areas subject toanthropogenicdisturbance from one of four sources: agriculture,inhabitation,gravel extraction or winter road de-icing with NaCl. The taxarecorded were a mixture of those already described fromepigeicwaters or groundwaters and some which may be regarded ascharacteristic of springs Statistical analysis revealeddifferencesin the meiofaunal taxa in relation to the water quality orcatchment area variables. Of the major meiofaunal groups thenematodes favoured springs influenced by agriculture,oligochaeteswere most tolerant of road de-icing, cladocerans and copepodspreferred springs in a natural state, ostracods couldwithstand theinfluence of road de-icing and gravel extraction, whilstdipteranlarvae could withstand the influence of agriculture.Oligochaetescomprised 13.4% of meiofaunal numbers. The most abundantspecieswas Aeolosoma quaternarium, which was most numerous insprings in a natural state, while Dero digitatapreferredeutrophic conditions. Harpacticoids comprised 14.1% ofindividualsand clearly preferred a natural state, Bryocamptusechinatusand Moraria brevipes being the most abundant species.Ostracods, of which the most abundant species was Potamocyprispallida, were the most numerous animal group, constituting21% ofthe meiofaunal population, and their numbers were greatest insprings influenced by road de-icing or gravel extraction.Dipteranlarvae, of which the majority were chironomids, comprised14.9% ofindividuals. The ostracods Potamocypris pallida andthreespecies of Candona and the harpacticoid Bryocamptuscuspidatus are new records for Finland.  相似文献

Variations in distributions and behaviours of Atlantic salmon Salmo salar in allopatry (homogeneous) and in sympatry with brown trout Salmo trutta (mixed) were observed before, during and after 2 day periods of dewatering in a large glass-sided indoor stream at densities typical of Scottish upland streams. Brown trout utilized pools more than Atlantic salmon at normal flows and in both species the majority of fishes moved into pools during dewatering. There was no significant effect of brown trout, which was the more dominant species, on the overall ability of Atlantic salmon to use pool habitat as a refuge during dewatering. Within mixed and homogeneous groups, average feeding levels decreased during dewatering. The highest ranking fish, which was always a brown trout in mixed groups, predominantly monopolized the pool and other individuals in pools adopted a more cryptic, stationary behaviour. Dewatering effectively increased local population density with the result that dominance status became much more important in maintaining food intake, and polarization between the top ranking fish and others increased. During the first day of dewatering, there was extreme behavioural polarization such that the dominant fish exhibited most aggression and least feeding within the group. Among dominant fish on the second day of dewatering, aggression had largely abated and feeding had returned to pretreatment levels despite the reduced average feeding within the group. The main difference between mixed and homogeneous groups was in the behaviour of the most dominant Atlantic salmon, which was near-despotic in allopatry and subordinate to brown trout in sympatry.  相似文献

1. Flow conditions were modified over patches of river bed in three rivers in south-western Australia to determine the effects of turbulence on benthic invertebrate communities.
2. Artificial structures to increase downstream turbulence were developed in a laboratory flume. In the field, these increased turbulence intensity by 35% for a 20% reduction in velocity.
3. Patches of gravel were placed in each river and turbulence-generating structures allocated randomly to half of these, creating treatment patches. An acoustic Doppler velocimeter was used to measure flow conditions over both treatment and control patches at several heights above the bed. After 6 weeks, the invertebrate fauna of the gravel patches were sampled to examine the response to modified flow conditions.
4. The treatments increased relative turbulence intensity twofold for a reduction in velocity of between 3 and 5 cm s−1, but turbulence intensity was significantly higher in only one of the three rivers.
5. There were no significant effects of increased relative turbulence intensity on any aspect of the invertebrate assemblage. This may be a result of the fairly small increase in relative turbulence intensity created during the experiment, the spatial scale of the manipulation or the types of stream community studied.  相似文献

 The fast phases of latent/manifest latent nystagmus (LMLN) may either cause the target image to fall within (foveating) or outside (defoveating) the foveal area. We previously verified that both types are generated by the same mechanism as voluntary saccades and propose a hypothetical, dual-mode mechanism (computer model) for LMLN that utilizes normal ocular-motor control functions. Fixation data recorded during the past 30 years from 97 subjects with LMLN using both infrared and magnetic search coil oculography were used as a basis for our simulations. The MATLAB/Simulink software was used to construct a robust, modular, ocular motor system model, capable of simulating LMLN. Fast-phase amplitude versus both peak velocity and duration of simulated saccades were equivalent to those of saccades in normal subjects. Based on our LMLN studies, we constructed a hypothetical model in which the slow-phase velocity acted to trigger the change between foveating and defoveating LMLN fast phases. Foveating fast phases were generated during lower slow-phase velocities whereas defoveating fast phases occurred during higher slow-phase velocities. The bidirectional model simulated Alexander's law behavior under all viewing and fixation conditions. Our ocular-motor model accurately simulates LMLN patient ocular motility data and provides a hypothetical explanation for the conditions that result in both foveating and defoveating fast phases. As is the case for normal physiological saccades, the position error determined the saccadic amplitudes for foveating fast phases. However, the final slow-phase velocity determined the amplitudes of defoveating fast phases. In addition, we suggest that individuals with LMLN use their fixation subsystem to further decrease the slow-phase velocity as the target image approaches the foveal center.
