The effects of superficial gas velocity (Ugr), gas entrance velocity (ν), and bubble size on the growth of Tisochrysis lutea was investigated in 600-mL photobioreactors operated with airlift pumps. Superficial gas velocities, calculated from measured air flow rates, ranging from 7 to 93 mm s?1 were created using a 1.6-mm diameter syringe. We tested the effects of sparger velocity over a range of 2.48 to 73.4 m s?1 and the effects of bubble size by using two styles of air stones and an open glass pipette, which created a bubble sizes in the range of 0.5 to 5 mm. We calculated oxygen mass transfer coefficient, kLa, values for all experimental conditions. Cell growth increased linearly with increased superficial gas velocity and decreased with increased sparger velocity. Results indicated that smaller bubble size leads to some initial cell damage, but after time, the increased gas transfer as reflected by the kLa value produced higher growth than larger bubbles. Two mechanisms were observed to correlate with cell damage in T. lutea: increasing velocity at the sparger tip and bubble bursting at the surface. These results demonstrate a method to test sensitivity of T. lutea to aeration, which is important for the design of airlift systems.  相似文献

Foam fractionation of bovine serum albumin (BSA) was studied as a model system for potato wastewater. The effects of feed concentration, superficial gas velocity, feed flow rate, bubble size, pH, and ionic strength on the enrichment and recovery of BSA were investigated in a single-stage continuous foam fractionation column. Enrichments ranged from 1.5 to 6.0 and recoveries from 5 to 85%. The feed concentrations were varied from 0.01 to 0.2 wt %, and enrichments were found to increase with lower feed concentrations. Enrichments also increased with lower superficial gas velocities and larger bubble sizes. At sufficiently low feed flow rates, enrichment was found to increase with an increase in the flow rate, eventually becoming insensitive to the feed flow rate at higher values. The pH was varied from 3.5 to 7.0 and ionic strength from 0.001M to 0.2M. The effects of pH and ionic strength were found to be coupled with bubble size. A minimum bubble size was found at pH 4.8, the isoelectric point of BSA, resulting in a minimum in the enrichment. Bubble size, and thus enrichment, was found to increase as the ionic strength decreased from 0.2M to 0.01M. Previous models(1,2) for the hydrodynamics of foam column were extended for a singlestage continuous foam fractionation column for the prediction of enrichment and recovery. The model assumed adsorption equilibrium, infinite surface viscosity, and bubbles of the same size. Though coalescence was formally accounted for in the model by considering bubble size as a function of foam height, calculations for the experimental runs were performed only for the case of no coalescence. Quantitative predictions of enrichment and recovery could not be made with a single representative bubble size because of the broad inlet bubble size distribution as well as broadening of the distribution as a result of coalescence. The experimental enrichments were higher and recoveries were lower than the model predictions, the discrepancy being more pronounced at lower feed concentrations because of increased coalescence. The higher enrichments are due to the predominant effect of internal reflux as a result of coalescence whereas the lower recoveries are a result of detrimental effects of broadening bubble size distributions.  相似文献

There is limited data on gas dispersion characteristics of fixed bed biofilm reactors under growth and non-growth conditions. In this paper, the gas–liquid dispersion of a bubble bed packed with a fibrous structured packing for biofilm application is studied. The reactor is operated with Pseudomonas putida aimed at aniline degradation in wastewater. Gas hold-up and bubble size distribution are determined. Running gas–liquid reaction conditions as well as non-reactive flow gas hold-up and bubble size distribution in the presence of surface-active and viscous components were measured. The properties of the gas dispersion proved to be stabilized by the fibrous bed presence and showed improvement of the dispersion parameter by the packing. Gas hold-up was found to increase monotonously with the rise of gas superficial velocity and viscosity and with surface tension fall. Liquid superficial velocity showed marginal effect. Apart from showing high gas hold-up and low bubble size due to surface-active and viscous dissolved elements, the biochemical reaction did not pose any significant additional effect. In agreement with the expected lack of bubble coalescence and break-up in the highly ionic solution practiced, the population size distribution and average bubble size were found to vary with the major operation factors opposite to their gas hold-up contribution. Gas hold-up was correlated with the specific bubble-to-channel size ratio and further with the variables considered. An empirical equation is proposed that relates gas hold-up with all studied variables. Assuming geometric similarity of the prototype and the real vessels, the equation as well as its corresponding range of fluid velocities can be used for bioreactor design and scale-up. The results concerning the gas hold-up are shown to be comparable with previous studies of mesh wire packing.  相似文献

Measurements of local gas phase characteristics are obtained in an external-loop airlift reactor filled with newtonian or viscous non-newtonian liquids. A double-optical fiber probe technique is used. It allows the determination of the axial and radial profiles of gas hold-up, bubbling frequency, bubble size and velocity. In the case of air-water system, the results show a strong effect of radial liquid velocity variation on the gas flow characteristics at the bottom of the riser. In the case of highly viscous non-newtonian solution, the gas flow is strongly affected by the gas distribution just above the gas sparger. This study also points out the bubble coalescence and the break-up phenomena in different liquids and levels in the reactor. Furthermore, the local measurements of bubble size and velocity allows to gain more detailed information on the dynamics of the bubble-flow and shows a tendency of large bubbles to circulate in the column center.  相似文献

The local properties of the dispersed gas phase (gasholdup, bubble diamater, and bubble velocity) were measured and evaluated at different positions in the riser and downcomer of a pilot plant reactor and, for comparison, in a laboratory reactor. These were described in Parts I and II of this series of articles during yeast cultivation and with model media. In the riser of the pilot plant reactor, the local gas holdup and bubble velocities varied only slightly in axial direction. The gas holdup increased considerably, while the bubble velocity increased only slightly with aeration rate. The bubble size diminished with increasing distance from the aerator in the riser, since the primary bubble size was larger than the equilibrium bubble size. In the downcomer, the mean bubble size was smaller than in the riser. The mean bubble size varied only slightly, the bubble velocity was accelerated, and the gas holdup decreased from top to bottom in the downcomer. In pilot plant at constant aeration rate, the properties of the dispersed phase were nearly constant during the batch cultivation, i.e., they depended only slightly on the cell concentration. In the laboratory reactor, the mean bubble sizes were much larger than in the pilot plant reactor. In the laboratory reactor, the bubble velocities in the riser and downcomer increased, and the mean gas holdup and bubble diameter in the downcomer remained constant as the aeration rate was increased.  相似文献

A pilot scale airlift reactor with multiple net draft tubes was developed. The reactor, 29?cm in diameter and 300?cm height, had four modules of double net draft tubes. Bubble size, bubble number, gas holdup, and volumetric mass transfer coefficient were measured under different superficial air velocities. The air velocity had little effect on bubble size but had significant influence on bubble number. A bubble column was also investigated for comparison. The airlift reactor had a higher gas holdup and volumetric mass transfer coefficient than those in the bubble column. The draft tubes in the airlift reactor substantially improved the reactor performance.  相似文献

Summary The following two-phase properties were evaluated in bubble column reactors with porous plate (5 m pore diameter) or perforated plate (1 mm and/or 3 mm hole diameter) gas distributors using distilled water or a 1% methanol solution: transverse profiles of the mean and Sauter bubble diameters, local gas holdups, true mean liquid and bubble velocities. An unexpected phenomenon named “reverse enrichment” was observed, in which, contrary to the normal behaviour, isoelectric hemoglobin coagula concentrated in the bottom stage of a multistage bubble separator. The phenomenon was observed when the hemoglobin particles were highly concentrated, the bubble diameter was larger than one third of the hole diameter of the partition plate, and the superficial air velocity at the hole was high. Transient and final behaviours of the multi-stage bubble separator with the reverse enrichment were analyzed by assuming the rejection of particles from upward thin liquid streams.  相似文献

Enrichment and recovery of bovine serum albumin has been examined in a continuous foam separation column. The effects of the operating factors, superficial air velocity, feed flow rate, feed concentration and pH on the above characteristics was investigated. The protein enrichment decreased with the increase in the value of each of these parameters. Protein recovery increased with increasing air velocity, decreased with increasing feed flow rate and did not change very much with increasing feed concentration. Maximum protein recovery was obtained at the isoelectric point (pH 4.8) of the protein. Maximum protein recovery was found to be a strong function of the air velocity in the range 0.05-0.15 cm/s. Further increase in air velocity did not have much effect on recovery because of very large bubbles formed as a result of coalescence. Bubble size was determined as a function of the above factors in the liquid and foam sections of the column. It was found to be dependent on protein concentration, feed flow rate and solution pH. The effect was more significant in the foam section of the column. The bubbles in the foam section were significantly larger (about 3-10 times) than those in the liquid, with a sharp change at the foam-liquid interface. The bubble size measurements were used to calculate the interfacial area and it was shown that the rate of protein removal increases with increasing interfacial area.  相似文献

Coalescence frequency has been measured in an air-sparged bubble column by observing the rate with which insoluble tracer gases were mixed within the dispersed gas phase. The effects of gas-sparge rate and salt concentration were examined. Coalescence frequencies increased with increasing gas-sparge rates approximately in proportion to the increase in gas-phase hold up. The coalescence frequency decreased with NaCI concentration to a minimum frequency about half the frequency observed in pure water. Bubble size distributions were also measured in the air-sparged column. The size distribution changed significantly over the same range of NaCI concentration as variations in the coalescence frequency were observed. The gas-sparge rate showed no effect on the bubble size.  相似文献

Summary Mean relative gas holdup, slip velocity, bubble size distribution, mean specific interfacial area, and volumetric mass transfer coefficient of oxygen were estimated in sparged columns 14 cm in diameter and 380 and/or 390 cm high with two different aerator types (porous plate and injector nozzle) in highly viscous Newtonian (glycerol solutions) and non-Newtonian (CMC solutions) fluids.For the Newtonian liquids the above properties were estimated as function of the viscosity of the liquid. For the non-Newtonian liquids they were determined as function of the fluid consistency index and flow behavior index. Significant differences between Newtonian and non-Newtonian systems appear. In Newtonian medium kL a drops with increasing viscosity and already approaches a constant value at =40 cP. In pseudoplastic medium kL a varies with the fluid consistency and flow behavior indexes in the entire investigated range.In both of these systems the primary bubble population changes into two or three populations along the reactor: the medium bubbles gradually disappear and small and large bubbles are formed.  相似文献

We describe a method by which the degree of bubble saturation can be determined by measuring the velocity of single bubbles at different heights from the bubble source in pure water containing increasing concentrations of surfactants. The highest rising velocities were measured in pure water. Addition of surfactants caused a concentration-dependent and height-dependent decrease in bubble velocity; thus, bubbles are covered with surfactants as they rise, and the distance traveled until saturation is reached decreases with increased concentration of surfactant. Pluronic F68 is a potent effector of bubble saturation, 500 times more active than serum. At Pluronic F68 concentrations of 0.1% (w/v), bubbles are saturated essentially at their source. The effect of bubble saturation on the interactions between animal cells and gas bubbles was investigated by using light microscopy and a micromanipulator. In the absence of surfactants, bubbles had a killing effect on cells; hybridoma cells and Chinese hamster ovary (CHO) cells were ruptured when coming into contact with a bubble. Bubbles only partially covered by surfactants adsorbed the cells. The adsorbed cells were not damaged and they also could survive subsequent detachment. Saturated bubbles, on the other hand, did not show any interactions with cells. It is concluded that the protective effect of serum and Pluronic F68 in sparged cultivation systems is based on covering the medium-bubble interface with surfaceactive components and that cell death occurs either after contact of cells with an uncovered bubble or by adsorption of cells through partially saturated bubbles and subsequent transport of cells into the foam region. Suspended gold nanoparticle in water medium starts to warm up under nanosecond laser irradiation and creates a bubble around itself. The present study aims at evaluating the amount of nanoparticle size reduction at boiling temperature, the temperature variations of the nanoparticle, and its medium and finally the bubble formation moment. To this aim, Mie theory was used to calculate the absorption cross section of the nanoparticle in proximity of the bubble. Heat transfer equations were applied to determine the temperature of the nanoparticle and water. In addition, hydrodynamic equations were initiated to evaluate the expansion of the bubble. Then, these three groups of equations were coupled together and solved numerically. Based on the results, the bubble forms at the critical pressure and consequently due to the slow bubble velocity, temperature gradient in the medium is observed. Further, slight pulse width variations play a significant role on the nanoparticle temperature. The calculation of the nanoparticle heating associated with the creation of the bubble helps in controlling nanoparticle size and understanding the nanoscale heat transfer processes.

Measurements of bubble and pellet size distributions are useful for biochemical process optimizations. The accuracy, representation, and simplicity of these measurements improve when the measurement is performed on-line and in situ rather than off-line using a sample. Historical and currently available measurement systems for photographic methods are summarized for bubble and pellet (morphology) measurement applications. Applications to cells, mycelia, and pellets measurements have driven key technological developments that have been applied for bubble measurements. Measurement trade-offs exist to maximize accuracy, extend range, and attain reasonable cycle times. Mathematical characterization of distributions using standard statistical techniques is straightforward, facilitating data presentation and analysis. For the specific application of bubble size distributions, selected bioreactor operating parameters and physicochemical conditions alter distributions. Empirical relationships have been established in some cases where sufficient data have been collected. In addition, parameters and conditions with substantial effects on bubble size distributions were identified and their relative effects quantified. This information was used to guide required accuracy and precision targets for bubble size distribution measurements from newly developed novel on-line and in situ bubble measurement devices.  相似文献

Decolourisation of the azo dye Reactive Black 5 by Geotrichum sp. CCMI 1019 was studied using stirred tank reactors (STR) and two types of bubble columns (porous plate (PP) bubble column and aeration tube (AT) bubble column). For the bubble columns, the kLa increased with the gas fractional hold-up (εG) and the aeration rate. A linear relationship between εG and superficial gas velocity was obtained for all reactors. At same aeration rates, the PP bubble columns showed higher kLa and hold-up values than the AT bubble column. In the STRs, large and dense aggregates were formed which adhered to surfaces whereas bubble columns gave smaller and less compact pellets.

Manganese peroxidase and laccase were detected in the extracellular media in all reactors. However, laccase was only detected after the onset of decolourisation, suggesting that additional enzymes may be involved. Mn peroxidase activity was detected (about 46 U/ml) in both the STRs and AT bubble columns but higher values (110 U/ml) were obtained with the PP bubble columns.

Out of the three reactor systems studied, the AT bubble columns gave the most favourable results for Reactive Black 5 decolourisation. Rapid and complete colour removal was obtained throughout the visible spectrum. Bubble columns are simple in design as well as operation and may be useful for the bioremediation of textile wastewater.  相似文献

A comprehensive study of the influence of plate geometry and the added packing on the holdup and slip velocity in reciprocating plate columns is presented. The use of packing in the interplate spaces of the column is of interest as packing alters the character of dispersion and also provides contact area for the phases. A model is proposed relating slip velocity to the single drop rise velocity taking into consideration the pseudo-hydrostatic effects and particle-to-particle interaction effects.  相似文献

Overcoming shear stress of microalgae cultures in sparged photobioreactors   总被引:1,自引:0,他引:1  
In the present work we identified and quantified the effect of hydrodynamic stress on two different microalgae strains, Dunaliella tertiolecta and D. salina, cultivated in bench-scale bubble columns. The cell death rate constant increased with increasing gas-entrance velocity at the sparger. Dunaliella salina was slightly more sensitive than D. tertiolecta. The critical gas-entrance velocities were approximately 50 and 30 m s(-1) for D. tertiolecta and D. salina, respectively. The effects of gas-flow rate, culture height, and nozzle diameter on the death rate constant were also studied. From these results it was concluded that bubble rising and bubble bursting are not responsible for cell death. Regarding nozzle diameter, small nozzles were more detrimental to cells. The bubble formation at the sparger was found to be the main event leading to cell death.  相似文献

The effects of scalariform perforation plate thickness and anglewithin vessel elements of Liriodendron tulipifera were studiedwith a computational fluid dynamical model. The pressure gradientand hence resistance to flow through the plate increased asthe perforation plate increased in thickness. Increasing theangle of the plate relative to the axis of the vessel (samenumber of pores) also increased the pressure gradient alongthe modelled cell. For the model matching the actual vesselelement, the plate contributed 8% to the flow resistance ofthe vessel element. This contribution increased only to 11%for doubled plate thickness, and to 14% for a plate at an angleof 60° to the vessel axis. The perforation plate alteredthe velocity profile across the vessel element, but to a differentextent depending on the angle of the plate. A plate at an acuteangle to the vessel axis has little effect on the paraboloidprofile as found upstream from the plate, while obtuse angleplates change the flow profile such that fluid through poresnear the wall is accelerated to a greater velocity than foundin the centre of the cell. Gas transfer and mixing were characterized in a 32-L bubble column reactor equipped with a commercially available rubber membrane diffuser. The performance of the membrane diffuser indicates that the slits in the membrane are best described as holes with elastic lids, acting as valves cutting off bubbles from the gas stream. The membrane diffuser thus functions as a one-way valve preventing backflow of liquid. Our design of the bottom plate of the reactor enabled us to optimize the aeration by changing the tension of the membrane. We thereby achieved mass transfer coefficients higher than those previously reported in bubble columns. A strong dependence of mass transfer on gas holdup and bubble size was indicated by estimates based on these two variables. The microalga, Rhodomonas sp. , sensitive to chemical and physical stress, was maintained for 8 months in continuous culture with a productivity identical to cultures grown in stirred tank reactors.
