Nanoparticles of cadmium selenide (CdSe) doped with europium, were synthesized as stabilizing agents using thioglycolic acid ligand. This method is based on the enhancing effect of CdSe quantum dots (QDs) doped with europium on chemiluminescence (CL) emission. This emission was generated by mixing CdSe QDs with manganese (II), iron (II) and chrome (II) sulfates as catalysts in the presence of hydrogen peroxide (H2O2). The structural characteristics and morphology of these nanoparticles were investigated by scanning electron microscopy, Fourier transform infrared spectroscopy, ultraviolet–visible absorption spectroscopy, X‐ray pattern and dynamic light scattering methods. The CdSe QDs doped with europium were used as the sensitizer in a luminol?hydrogen peroxide CL system. The sensitized CdSe QDs were analyzed for antibacterial activity against Gram‐positive or Gram‐negative bacteria. The results showed that the CdSe QDs are effective against all the studied bacteria, effectiveness was especially higher for Bacillus subtilis.  相似文献

In this paper, a novel chemiluminescence (CL) system was introduced based on the use of CdTe quantum dots (QDs) with the mixture solutions of fluorescein and potassium periodate (KIO4) in alkaline medium. The CL signal of an ultra‐weak system was strongly enhanced in the presence of QDs. The application of CdTe QDs–fluorescein–KIO4 system is reported for the first time. It was found that catechol had a diminishing effect on the CL reaction. Under optimal experimental conditions, CL intensity decreased linearly in a 1 to 100 μM catechol concentration range, with a 0.18 μM detection limit. A possible reaction mechanism was proposed according to the results of kinetic analyses, CL spectra, ultraviolet–visible and fluorescence spectra. The results pointed to an efficient energy transfer between the CL energy donor CdTe QDs and acceptor fluorescein. Finally, the CL method was successfully applied to the determination of catechol in environmental water samples.  相似文献

A novel and highly sensitive chemiluminescence (CL) method for the determination of ethanol was developed based on the CdS quantum dots (QDs)–permanganate system. It was found that KMnO4 could directly oxidize CdS QDs in acidic media resulting in relatively high CL emission. A possible mechanism was proposed for this reaction based on UV/Vis absorption, fluorescence and the generated CL emission spectra. However, it was observed that ethanol had a remarkable inhibition effect on this system. This effect was exploited in the determination of ethanol within the concentration range 12–300 µg/L, with detection at 4.3 µg/L. In order to evaluate the capability of presented method, it was satisfactorily utilized in the determination of alcohol in real samples. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

The chemiluminescence (CL) behaviour of the luminol–potassium periodate system enhanced by CdTe quantum dots capped with thioglycolic acid (TGA–CdTe QDs) was studied using kinetic experiments, CL spectra, UV–vis absorption spectra and fluorescence spectra. The production of oxygen‐containing reactant intermediates (O2?? and OH?) in the present CL system was verified by CL. The possible CL mechanism was discussed in detail. Furthermore, theophylline (THP) was determined based on its enhancement of the CL intensity of the CdTe QDs–luminol–potassium periodate system coupled with a flow‐injection technique. Under these optimized conditions, the linear range was found to be from 1.0 × 10?8 to 1.0 × 10?5 g/mL with a detection limit of 2.8 × 10?9 g/mL (3σ). The recoveries for the determination of THP in tablets were from 98.2 to 99.6%.  相似文献

High luminescence quantum yield water‐soluble CdTe/ZnS core/shell quantum dots (QDs) stabilized with thioglycolic acid were synthesized. QDs were chemically coupled to fully humanized antivascular endothelial growth factor165 monoclonal antibodies to produce fluorescent probes. These probes can be used to assay the biological affinity of the antibody. The properties of QDs conjugated to an antibody were characterized by ultraviolet and visible spectrophotometry, fluorescent spectrophotometry, sodium dodecyl sulfate–polyacrylamide gel electrophoresis, transmission electron microscopy and fluorescence microscopy. Cell‐targeted imaging was performed in human breast cancer cell lines. The cytotoxicity of bare QDs and fluorescent probes was evaluated in the MCF‐7 cells with an MTT viability assay. The results proved that CdTe/ZnS QD–monoclonal antibody nanoprobes had been successfully prepared with excellent spectral properties in target detections. Surface modification by ZnS shell could mitigate the cytotoxicity of cadmium‐based QDs. The therapeutic effects of antivascular endothelial growth factor antibodies towards cultured human cancer cells were confirmed by MTT assay. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

A pH‐sensitive and double functional nanoprobe was designed and synthesized in a water‐soluble system using thioglycolic acid (TGA) and mercapto‐acetohydrazide (TGH) as the stabilizers. TGA is biocompatible because the carboxyl group is easily linked to biological macromolecules. At the same time, the hydrazide on TGH reacts with the aldehyde on poly(ethylene glycol) (PEG) and forms a hydrazone bond. The hydrazone bond ruptured at specific pH values and exhibited pH‐stimuli‐responsive characteristics. As an optical imaging probe, the PEG–TGA/TGH‐capped CdTe quantum dots (QDs) had high quality, with a fluorescence efficiency of 25–30%, and remained stable for at least five months. This pH‐responsive factor can be used for the effective release of CdTe QDs under the acidic interstitial extracellular environment of tumor cells. This allows the prepared pH‐stimuli‐responsive nanoprobes to show fluorescence signals for use in cancer cell imaging. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

A simple and highly sensitive chemiluminescence (CL) method is reported for the determination of naphazoline hydrochloride (NH). It was found that the weak CL from the reaction of luminol and KIO4 in an alkaline medium could be highly amplified by cysteine‐capped cadmium telluride quantum dots (QDs) and the enhanced CL was effectively quenched by NH and this finding was utilized as a basis for the determination of NH. The QDs were synthesized in aqueous medium and characterized by X‐ray diffraction (XRD), transmission electron microscopy (TEM), and UV‐vis and photoluminescence spectroscopy. A possible mechanism was proposed for the CL system based on radical identification experiments, along with CL spectrum of the system. The experimental parameters were optimized by the reliable response surface methodology (RSM). Under the optimized experimental conditions, the proposed method allowed the determination of NH over the range of 5.0 × 10‐10–2.0 × 10‐7 mol/L (r2 = 0.9993, n = 10). The precision (RSD%) of the method, obtained from five replicate determinations of 2.0 and 150 nmol/L NH, was found to be 1.0% and 1.3%, respectively. The method was successfully applied to the determination of NH in pharmaceutical formulations and human urine and serum samples with results corroborated with the aid of those obtained from a standard method. Stable photoluminescence QD light‐emitting diodes (QD‐LEDs) were made based on hydrophilic CdTe quantum dots (QDs). A quantum dot‐inorganic nanocomposite (hydrophilic CdTe QDs incorporating dehydrated silica gel) was prepared by two methods (rotary evaporation and freeze drying). Taking advantage of its viscosity, plasticity and transparency, dehydrated silica gel could be coated on the surface of ultraviolet (UV) light LEDs to make photoluminescence QD‐LEDs. This new photoluminescence QD‐LED, which is stable, environmentally non‐toxic, easy to operate and low cost, could expand the applications of hydrophilic CdTe QDs in photoluminescence. A highly sensitive flow‐injection chemiluminescence (FIA‐CL) method based on the CdTe nanocrystals and potassium permanganate chemiluminescence system was developed for the determination of l ‐ascorbic acid. It was found that sodium hexametaphosphate (SP), as an enhancer, could increase the chemiluminescence (CL) emission from the redox reaction of CdTe quantum dots with potassium permanganate in near‐neutral pH conditions. l ‐Ascorbic acid is suggested as a sensitive enhancer for use in the above energy‐transfer excitation process. Under optimal conditions, the calibration graph of emission intensity against logarithmic l ‐ascorbic acid concentration was linear in the range 1.0 × 10?9–5.0 × 10?6 mol/L, with a correlation coefficient of 0.9969 and relative standard deviation (RSD) of 2.3% (n = 7) at 5.0 × 10?7 mol/L. The method was successfully used to determine l ‐ascorbic acid in vitamin C tablets. The possible mechanism of the chemiluminescence in the system is also discussed. We have previously shown that CdSe/ZnS core/shell luminescent semiconductor nanocrystals or QDs (quantum dots) coated with PEG [poly(ethylene glycol)]-appended DHLA (dihydrolipoic acid) can bind AcWG(Pal)VKIKKP9GGH6 (Palm1) through the histidine residues. The coating on the QD provides colloidal stability and this peptide complex uniquely allows the QDs to be taken up by cultured cells and readily exit the endosome into the soma. We now show that use of a polyampholyte coating [in which the neutral PEG is replaced by the negatively heterocharged CL4 (compact ligand)], results in the specific targeting of the palmitoylated peptide to neurons in mature rat hippocampal slice cultures. There was no noticeable uptake by astrocytes, oligodendrocytes or microglia (identified by immunocytochemistry), demonstrating neuronal specificity to the overall negatively charged CL4 coating. In addition, EM (electron microscopy) images confirm the endosomal egress ability of the Palm1 peptide by showing a much more disperse cytosolic distribution of the CL4 QDs conjugated to Palm1 compared with CL4 QDs alone. This suggests a novel and robust way of delivering neurotherapeutics to neurons.  相似文献

With excellent optical properties, quantum dots (QDs) have been made as attractive molecular probes for labelling cells in biological research. In this study high‐quality CdSe QDs prepared in a paraffin–oleic acid system were used as fluorescent labels in direct and indirect detection of carcinoembryonic antigen (CEA), a cancer marker expressed on the surface of HeLa cells. The primary antibody (Ab) (rabbit anti‐CEA8) and secondary Ab (goat anti‐rabbit IgG) were covalently linked to carboxyl‐functioned CdSe QDs, and both the QDs–antibody and QDs–IgG probes were successfully used to label HeLa cells. The present study demonstrates the practicability of CdSe QDs as an attractive type of fluorescent labels for biological applications such as protein probes and cell imaging. This paper aimed to study the possible involvement of adenosine triphosphate‐binding cassette (ABC) transporters in the detoxification of quantum dots (QDs) in human breast carcinoma (SK‐BR‐3) cells. The effects of QD sizes on such interactions were also evaluated. For this purpose, we used monodispersed MPA‐COOH‐CdTe QDs with different diameters (emission length at 560 and 625 nm, named as QD‐560 and QD‐625). Such QDs tended to accumulate in cells and cause significant toxicity. Using specific inhibitors of ABC transporters, the cellular accumulation and toxicity of QDs in SK‐BR‐3 cells were significantly affected. Moreover, treatment of QDs caused concentration‐ and time‐dependent induction of ABC transporters. Furthermore, the induction effects of smaller QDs were found to be greater than larger ones at equivalent concentrations, suggesting a size‐dependent recognition of substrates by ABC transporters. Overall, these results provided important support for the modulation of QDs toxicity by ABC transporters.  相似文献

A new class of zinc oxide quantum dots (ZnO QDs) was investigated as nanoprobes for targeting cancer cells in vitro. ZnO nanoparticles were synthesized using conventional sol–gel method and encapsulated using trimethoxy aminopropyl silane. Transferrin, the ligand targeting the cancer cells, was conjugated to the ZnO QDs. In vitro imaging studies using MDA-MB-231 showed the biocompatible ZnO nanoprobe selectively binding to the cell surface receptor and internalizing through receptor-mediated endocytosis. Time-lapsed photobleaching studies indicate the ZnO QDs to be resistant to photobleaching, making them suitable for long term imaging purpose. Investigation of the ZnO nanoprobe as a platform for sensitive bioassays indicates that it can be used as an alternative fluoroprobe for cancer cell targeting and sensing applications.  相似文献

In recent years, more and more nanomaterials‐based chemiluminescence (CL) systems have appeared to improve the sensitivity and expand the scope of the analytical applications with the explosive growth and development of nanomaterials and technology. As a fascinating class of luminescent carbon nanomaterials, carbon dots (CDs) are now substantially studied in fabricating CL based assays due to their unique optical and mechanical properties. Herein, we summarize and highlight the current developments of CDs‐involved weak or ultraweak CL systems, as well as the corresponding mechanisms and proper applications in some fields. CDs can take part in the CL reactions as oxidants, emitting species directly involved in redox oxidation, energy acceptors of CL energy transfer, or even catalysts involving other luminophores. In fact, they always have more than one role in many cases, owing to the formation of various excited species with short life in CL systems. Therefore, in this review article, the most recent progress of the different CDs‐assisted CL systems including the mechanisms and applications are presented. Finally, the conclusions and future prospects of this field are also discussed. The significant features of the CDs‐based CL systems may open up new prospects and challenges in a wider range of fields.  相似文献

In this article, we have examined the direct spectroscopic and microscopic evidence of efficient quantum dots‐ α‐chymotrypsin (ChT) interaction. The intrinsic fluorescence of digestive enzyme is reduced in the presence of quantum dots through ground‐state complex formation. Based on the fluorescence data, quenching rate constant, binding constant, and number of binding sites are calculated under optimized experimental conditions. Interestingly, fluorescence quenching method clearly illustrated the size dependent interaction of MPA‐CdTe quantum dots. Conformational change of ChT was traced using synchronous fluorescence measurements, circular dichroism and FTIR spectroscopic methods. Furthermore, the AFM results revealed that the individual enzyme molecule dimensions were changed after interacting with quantum dot. Consequently, this result could be helpful for constructing safe and effective utilisation of QDs in biological applications. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

The fabrication of a low reabsorption emission loss, high efficient luminescent solar concentrator (LSC) is demonstrated by embedding near infrared (NIR) core/shell quantum dots (QDs) in a polymer matrix. An engineered Stokes shift in NIR core/shell PbS/CdS QDs is achieved via a cation exchange approach by varying the core size and shell thickness through the refined reaction parameters such as reaction time, temperature, precursor molar ratio, etc. The as‐synthesized core/shell QDs with high quantum yield (QY) and excellent chemical/photostability exhibit a large Stokes shift with respect to the bare PbS QDs due to the strong core‐to‐shell electrons leakage. The large‐area planar LSC based on core/shell QDs exhibits the highest value (6.1% with a geometric factor of 10) for optical efficiency compared to the bare NIR QD‐based LSCs and other reported NIR QD‐based LSCs. The suppression of emission loss and the broad absorption of PbS/CdS QDs offer a promising pathway to integrate LSCs and photovoltaic devices with good spectral matching, indicating that the proposed core/shell QDs are strong candidates for fabricating high efficiency semi‐transparent large‐area LSCs.  相似文献

Functionalized CdTe–CdS core–shell quantum dots (QDs) were synthesized in aqueous solution via water‐bathing combined hydrothermal method using L‐cysteine (L‐Cys) as a stabilizer. This method possesses both the advantages of water‐bathing and hydrothermal methods for preparing high‐quality QDs with markedly reduced synthesis time, and better stability than a lone hydrothermal method. The QDs were characterized by transmission electronic microscopy and powder X‐ray diffraction and X‐ray photoelectron spectroscopy. The CdTe–CdS QDs with core–shell structure showed both enhanced fluorescence and better photo stability than nude CdTe QDs. After conjugating with antibody rabbit anti‐CEACAM8 (CD67), the as‐prepared l ‐Cys capped CdTe–CdS QDs were successfully used as fluorescent probes for the direct immuno‐labeling and imaging of HeLa cells. Novel, water‐soluble CdTe quantum dots (QDs) capped with β‐cyclodextrin (β‐CD) and ~ 4.0 nm in diameter were synthesized in aqueous solution, and characterized using transmission electron microscopy (TEM). A fluorescence‐sensing system based on the photoinduced electron transfer (PET) of (mono‐6‐thio‐β‐CD)–CdTe QDs was then designed to measure the interaction of phenothiazine dyes [methylene blue (MB) and methylene green (MG)] with herring sperm DNA (hsDNA). This fluorescence‐sensing system was based on a fluorescence “OFF–ON” mode. First, MB/MG adsorbed on the surface of (mono‐6‐thio‐β‐CD)–CdTe QDs effectively quenches the fluorescence of (mono‐6‐thio‐β‐CD)–CdTe QDs through PET. Then, addition of hsDNA restores the fluorescence intensity of (mono‐6‐thio‐β‐CD)–CdTe QDs, because hsDNA can bind with MB/MG and remove it from the as‐prepared (mono‐6‐thio‐β‐CD)–CdTe QDs. In addition, detailed reaction mechanisms of the (mono‐6‐thio‐β‐CD)–CdTe QDs–MB/MG–hsDNA solution system were studied using optical methods, by comparison with the TGA–CdTe QDs–MB/MG–hsDNA solution system. Cysteamine (CA)‐capped CdTe quantum dots (QDs) (CA–CdTe QDs) were prepared by the reflux method and utilized as an efficient nano‐sized fluorescent sensor to detect mercury (II) ions (Hg2+). Under optimum conditions, the fluorescence quenching effect of CA–CdTe QDs was linear at Hg2+ concentrations in the range of 6.0–450 nmol/L. The detection limit was calculated to be 4.0 nmol/L according to the 3σ IUPAC criteria. The influence of 10‐fold Pb2+, Cu2+ and Ag+ on the determination of Hg2+ was < 7% (superior to other reports based on crude QDs). Furthermore, the detection sensitivity and selectivity were much improved relative to a sensor based on the CA–CdTe QDs probe, which was prepared using a one‐pot synthetic method. Realizing efficient solid‐state luminescence is of great important to expand quantum dots (QDs) application fields. This work reports the preparation of CdTe@BaCO3 composite by a one‐pot precipitation method. Both steady‐state PL and PL decay characteristics in either solid‐state or colloid solution show no obvious difference, mainly benefited from the effective protection of BaCO3 on QDs from the external environment. By utilizing green and red CdTe QDs as dual‐color emission centers, precise emitting‐color control from green (0.312, 0.667) to red (0.691, 0.292) could be achieved in CdTe@BaCO3 composite by adjusting volume ratio of CdTe solution precursor. Our results demonstrate that this composite material shows bright solid‐state luminescence and facile adjustment of the emitting color in QDs‐based composite is feasible, which could offer new path to design color‐tunable luminescent materials for future optoelectronic applications.  相似文献
