Soil fungi of areas in the North-Eastern region of Saudi Arabia where truffles are native were surveyed. Forty-three species of fungi belonging to twenty genera were isolated. Most were recovered from soils underneath or around truffle ascocarps: thirty species from soil under the surface of Tirmania nivea ascocarps, twenty-four from Terfezia boudieri soil and twenty species each from Tirmania pinoyi and Terfezia claveryi soils. Rhizosphere soil of Helianthemum lippi, on the other hand, yielded twenty-four fungal species while only fourteen fungal species were found in soil without vegetation. The total counts of fungi/g soil were highest in soils from the under surface of truffles, followed by rhizosphere soil, with the lowest in soils without vegetation.  相似文献

Tuber spp. are filamentous ascomycetes which establish symbiosis with the roots of trees and shrub species. By virtue of this symbiosis they produce hypogeous ascocarps, known as truffles. Filamentous ascomycetes can reproduce by homothallism or heterothallism depending on the structure and organization of their mating type locus. The first mating type locus in a truffle species has been recently characterized in Tuber melanosporum and it has been shown that this fungus, endemic in Europe, is heterothallic. The availability of sequence information for T. melanosporum mating type genes is seminal to cloning their orthologs from other Tuber species and assessing their reproductive mode. Here we report on the organization of the mating type region in T. indicum, the black truffle species present in Asia, which is the closest relative to T. melanosporum and is characterized by an high level of morphological and genetic variability. The present study shows that T. indicum is also heterothallic. Examination of Asiatic black truffles belonging to different genetic classes, sorted according to the sequence polymorphism of the internal transcribed spacer rDNA region, has revealed sequence variations and rearrangements in both coding and non-coding regions of the mating type locus, to suggest the existence of cryptic species within the T. indicum complex. The presence of transposable elements within or linked to the mating type region suggests a role of these elements in generating the genotypic diversity present among T. indicum strains. Overall, comparative analyses of the mating type locus have thus allowed us to tackle taxonomical and phylogenetic issues within black truffles and make inferences about the evolution of T. melanosporum-T. indicum lineage. Our results are not only of fundamental but also of applied relevance as T. indicum produces edible fruit bodies that are imported also into Europe and thus may represent a biological threat for T. melanosporum.  相似文献

Although the truffle beetle, Leiodes cinnamomea, inflicts substantial damage to the ripe stage of fruiting bodies of the economically important black truffle (Tuber melanosporum), it is not attracted by ripe truffle odours. Rather, male beetles are attracted to infested truffles only in the presence of female beetles, suggesting that the former employ a pheromone to locate truffles over short distances. In contrast, female beetles show no attraction to infested or uninfested truffles, suggesting that they employ other cues, possibly linked to odours emitted by truffles prior to the ripe stage. We hypothesize that the chemical composition of truffle volatiles changes over the life of the truffle fruiting body, being attractive to insects early on and to mammals just prior to decomposition.  相似文献

This work presents DNA sequence motifs from the internal transcribed spacer (ITS) of the nuclear rRNA repeat unit which are useful for the identification of five European and Asiatic truffles (Tuber magnatum, T. melanosporum, T. indicum, T. aestivum, and T. mesentericum). Truffles are edible mycorrhizal ascomycetes that show similar morphological characteristics but that have distinct organoleptic and economic values. A total of 36 out of 46 ITS1 or ITS2 sequence motifs have allowed an accurate in silico distinction of the five truffles to be made (i.e., by pattern matching and/or BLAST analysis on downloaded GenBank sequences and directly against GenBank databases). The motifs considered the intraspecific genetic variability of each species, including rare haplotypes, and assigned their respective species from either the ascocarps or ectomycorrhizas. The data indicate that short ITS1 or ITS2 motifs (≤50 bp in size) can be considered promising tools for truffle species identification. A dot blot hybridization analysis of T. magnatum and T. melanosporum compared with other close relatives or distant lineages allowed at least one highly specific motif to be identified for each species. These results were confirmed in a blind test which included new field isolates. The current work has provided a reliable new tool for a truffle oligonucleotide bar code and identification in ecological and evolutionary studies.  相似文献

True truffles (Tuber sp.) that establish ectomycorrhizal symbiosis (ECM) with trees in the Mediterranean and temporal regions have species specific abilities to assimilate soil born elements. Suitable habitats are usually inhabited by few truffle species, while distinguishing their symbiotic potentials appeared very difficult. Two species that commonly inhabit riparian forests in Serbia are the most prized one, Tuber magnatum Pico (Piedmont white truffle) and not so highly valued Tuber brumale Vitt . In order to assess potential differences between their assimilation and accumulation abilities, the differences between contents of elements that may be the subjects of the symbiotic trade between the host plant and fungi were evaluated in accumulation target (ascocarps) and their source (the soil). Essential (K, Na, Ca, Mg, Fe, P, S, and Zn) and essential trace elements (Co, Cr, Cu, Mn, and Se) in truffles and soil samples were determined by means of inductively coupled plasma with optical emission spectrometry (ICP‐OES). Their concentrations (mg/kg) in ascocarps were in the range from 1.364±0.591 (Cr) to 10760.862±16.058 (K), while in soil ranged from 23.035±0.010 (Cr) to 20809.300±122.934 (Fe). Element accumulation potential (bioaccumulation factor) was calculated in the system truffle/soil. The statistical approaches were used for establishing the differences, while the possible differentiation between symbiotic potentials of two mycelia in the defined soil conditions was discussed.  相似文献

Tuber species produce highly sought-after truffles and host a wide diversity and high abundance of bacteria. It has been suggested that some of these bacteria contribute to the growth, maturity, and aromatic properties of truffles. Here, we characterized and compared the microbiomes of several species of truffles from the Sichuan Province in Southwest China using high-throughput sequencing of bacterial community 16S rRNA genes. Two T. pseudoexcavatum ascocarp samples had relatively similar bacterial communities, as indicated by PCoA analysis. In contrast, three T. indicum samples collected at different maturity stages did not contain similar communities, suggesting that the maturity stage of ascocarps affects community composition in addition to host phylogenetic background. Despite the variation seen among species and maturity stages, the Proteobacteria phylum dominated all communities, which is consistent with previous studies of Tuber-associated bacteria. Moreover, Bradyrhizobium was the dominant genus in most Tuber ascocarps, which is also consistent with previous studies, although the functional role of this genus within truffles is unclear. Notably, Serratia, which are essential producers of thiophene volatiles within T. borchii were dominant in all of our samples. This finding supports the hypothesis that the ability to produce thiophene volatiles is widespread among these bacteria.  相似文献

Tuber spp. are ectomycorrhizal ascomycetes that produce ascocarps known as truffles. Basic aspects of Tuber biology have yet to be fully elucidated. In particular, there are conflicting hypotheses concerning the mating system and the ploidy level of the mycorrhizal and truffle hyphae. We used polymorphic microsatellites to compare the allelic configurations of asci with those from the network of the surrounding hyphae in single Tuber magnatum truffles. We then used these truffles to inoculate host plants and evaluated the microsatellite configurations of the resulting mycorrhizal root tips. These analyses provide direct evidence that T. magnatum outcrosses and that its life cycle is predominantly haploid. In addition to its scientific significance, this basic understanding of the T. magnatum life cycle may have practical importance in developing strategies to obtain and select nursery-produced mycorrhizal plants as well as in the management of artificial plantations of this and other Tuber spp.  相似文献

The methanolic extracts of fresh fruiting bodies of Tuber melanosporum, T. aestivum, T. magnatum and T. borchii were evaluated for their free fatty acids (FFAs) content to test their contribution to truffle allelopathic activity. Linoleic acid was the major FFA detected in all truffle species. In addition to linoleic acid, oleic acid and palmitic acid were the other abundant FFAs in the Tuber spp. extracts. Looking at the total production, T. melanosporum and T. aestivum synthesized significantly higher amounts of all identified FFA than T. magnatum and T. borchii. The allelopathic activity, based on the specific activity (EC50) of the identified compounds, was evaluated in the growth of Arabidopsis thaliana, Lotus corniculatus, Melica ciliata and Silene vulgaris. Linoleic acid had the lowest specific activity and palmitic acid had the highest. The black truffle extracts had a higher inhibitory activity than white truffles, and this could be explained by the high relative amount of FFAs in them.  相似文献

Based on an assessment of soil and climatic conditions in British Columbia (BC), the Truffle Association of British Columbia (TABC) determined that the cultivation of Mediterranean Tuber melanosporum and Tuber aestivum might be possible in the warmer parts of the province. With the cooperation of independent truffle growers, TABC assessed the colonization of host tree roots collected from eight truffle orchards planted 2–7 years earlier using morphological and molecular criteria. Both Tuber species persisted on the roots of inoculated trees in six of the eight truffle orchards studied. The identity of Tuber ectomycorrhizas that had been characterized morphologically as differing from those of T. melanosporum and T. aestivum were determined using DNA sequence analysis to belong to three species of truffles native to the Pacific Northwest. One of those species, Tuber anniae, had been previously reported from BC, but the other two, Tuber menseri nom. prov. and Tuber beyerlei, are reported here from BC for the first time. Recently, production of three Périgord black truffles in one truffle orchard and one Burgundy truffle in another orchard demonstrates that these truffles are able to fruit in BC.  相似文献

Ecology of hypogeic mycorrhizal fungi, such as truffles, remains largely unknown, both in terms of their geographical distribution and their environmental niches. Occurrence of true truffles (Tuber spp.) was therefore screened using specific polymerase chain reaction (PCR) assays and subsequent PCR amplicon sequencing in tree roots collected at 322 field sites across the Czech Republic. These sites spanned a wide range of climatic and soil conditions. The sampling was a priori restricted to areas thought to be suitable for Tuber spp. inasmuch as they were characterized by weakly acidic to alkaline soils, warmer climate, and with tree species previously known to host true truffles. Eight operational taxonomic units (OTUs) corresponding to Tuber aestivum, T. borchii, T. foetidum, T. rufum, T. indicum, T. huidongense, T. dryophilum, and T. oligospermum were detected. Among these, T. borchii was the OTU encountered most frequently. It was detected at nearly 19% of the sites. Soil pH was the most important predictor of Tuber spp. distribution. Tuber borchii preferred weakly acidic soils, T. foetidum and T. rufum were most abundant in neutral soils, and T. huidongense was restricted to alkaline soils. Distribution of T. aestivum was mainly dictated by climate, with its range restricted to the warmest sites. Host preferences of the individual Tuber spp. were weak compared to soil and climatic predictors, with the notable exception that T. foetidum appeared to avoid oak trees. Our results open the way to better understanding truffle ecology and, through this new knowledge, also to better‐informed trufficulture.  相似文献

Abstract: We studied the effects of 6 green-tree retention levels and patterns on the diets of northern flying squirrels (Glaucomys sabrinus), Townsend's chipmunks (Tamias townsendii), Siskiyou chipmunks (T. siskiyou), western red-backed voles (Myodes californicus), and southern red-backed voles (Myodes gapperi) using fecal pellet analysis. These rodents are truffle spore dispersers and prey for forest predators such as the northern spotted owl (Strix occidentalis caurina). Pretreatment diets showed differences in truffle and plant consumption among genera. Tree harvesting, especially in the 15% aggregated retention pattern, reduced frequency of Rhizopogon spores in the diet of voles, which may reflect a reduced ability of these animals to forage for Rhizopogon truffles, a decreased access to these truffles, or a reduction in Rhizopogon truffle abundance or frequency. Habitat island effects and edge effects provide conceptual frameworks for the reduction in consumption of Rhizopogon truffles by voles in green-tree aggregates. Overall, small mammal consumption of truffles showed little change in response to the treatments. Animals may be compensating for a locally declining food source by altering their foraging behavior. The long-term effect of this postulated behavioral compensation on small mammal energetics and population dynamics is unknown. Forest managers may reduce the impact of tree harvesting on these key forest ecosystem components by including green-tree aggregates within a dispersed retention matrix.  相似文献

块菌是指Tuber属的真菌种类,属于子囊菌门(Ascomycota),生在地下,其中包含着世界上最美味和价值最昂贵的食用菌.中国食与药用真菌文化悠久,历代本草都不乏各种菌类的记载,但在本草中记载的与块菌有关系的菌类唯有1245年陈仁玉编撰的“菌谱”中称之为“麦蕈”和俗名为“麦丹蕈”的一种地下真菌.日本本草对“麦蕈”也屡有记载,并把“麦蕈”叫“地肾”和“松露”.这是“松露”一名的最早记载,“麦蕈”或“松露”可能是被现代菌物学称之为须腹菌(Rhizopogon spp.)的真菌种类.因而,“松露”一名虽是汉字,但是所指并非真正的块菌,应是须腹菌(Rhizopogon spp.),也称“假块菌”(false truffle),所以松露和块菌寓意各异.产于我国西南地区的食用黑块菌主要是印度块菌(Tuber indi-cum)或中华块菌(T.sinernse)、夏块菌(T.aestivum=T.uncinatu)和拟喜马拉雅块菌(T.pseudohimalayense=T.pseudoexcavatum),近年来身价倍增,成为中国野生食用菌出口种类中名列前三甲的菌类,在世界块菌市场上也颇受欢迎.喜马拉雅块菌(T.himalayense)在我国尚未发现它的踪迹.印度块菌和中华块菌形态上有差异,若把中国产的类似黑孢块菌的黑块菌统称为印度块菌(T.indicum)尚值得商榷.中华块菌或印度块菌形态和遗传变异多样,尚需进一步研究.2010年在云南和四川相继发现的大型香味浓郁的白块菌,近似于波氏块菌(T.borchii)组的块菌,颇具研究和巨大的商业价值,表明我国块菌资源比原有记载的要丰富的多.由于利益之驱动和疏于管理,掠夺式的采收方法造成了块菌产区生态环境毁灭性的破坏,一些商业化采集区的块菌已濒临灭绝,中国西南地区的块菌资源的保护问题迫在眉睫.本文对产自欧洲、美洲的块菌、沙漠块菌,以及块菌的生态意义和种植作了简要介绍和讨论.  相似文献

Twenty-nine yeast strains were isolated from the ascocarps of black and white truffles (Tuber melanosporum Vitt. and Tuber magnatum Pico, respectively), and identified using a polyphasic approach. According to the conventional taxonomic methods, MSP-PCR fingerprinting and sequencing of the D1/D2 domain of 26S rDNA, the strains were identified as Candida saitoana, Debaryomyces hansenii, Cryptococcus sp., Rhodotorula mucilaginosa, and Trichosporon moniliiforme. All isolates assimilated l-methionine as a sole nitrogen source and produced the volatile organic compounds (VOCs), 2-methyl butanol, 3-methyl butanol, methanethiol, S-methyl thioacetate, dimethyl sulfide, dimethyl disulfide, dimethyl trisulfide, dihydro-2-methyl-3(2H)-thiophenone and 3-(methylthio)-1-propanol (MTP). ANOVA analysis of data showed significant (P<0.01) differences in VOCs produced by different yeasts, with MTP as the major component (produced at concentrations ranging from 19.8 to 225.6 mg/l). In addition, since some molecules produced by the isolates of this study are also characteristic of truffle complex aroma, it is possible to hypothesize a complementary role of yeasts associated with this ecosystem in contributing to final Tuber spp. aroma through the independent synthesis of yeast-specific volatile constituents.  相似文献

Truffles (Tuber spp.) are ascomycete subterraneous fungi that form ectomycorrhizas in a symbiotic relationship with plant roots. Their fruiting bodies are appreciated for their distinctive aroma, which might be partially derived from microbes. Indeed, truffle fruiting bodies are colonized by a diverse microbial community made up of bacteria, yeasts, guest filamentous fungi, and viruses. The aim of this minireview is two-fold. First, the current knowledge on the microbial community composition of truffles has been synthesized to highlight similarities and differences among four truffle (Tuber) species (T. magnatum, T. melanosporum, T. aestivum, and T. borchii) at various stages of their life cycle. Second, the potential role of the microbiome in truffle aroma formation has been addressed for the same four species. Our results suggest that on one hand, odorants, which are common to many truffle species, might be of mixed truffle and microbial origin, while on the other hand, less common odorants might be derived from microbes only. They also highlight that bacteria, the dominant group in the microbiome of the truffle, might also be the most important contributors to truffle aroma not only in T. borchii, as already demonstrated, but also in T. magnatum, T. aestivum, and T. melanosporum.  相似文献

Truffles are abundant in some regions of China. Nevertheless, it was not until the 1980s that Tuber species were discovered by Chinese mycologists. In recent years, international truffle markets have shown an increasing interest in the import of Chinese Tuber. These truffles serve as a complement to European truffles due to their lower prices and their greater availability in a deficit market. However, Chinese truffles have been the subject of fraudulent commercial practices, and these could have a negative effect on truffle culture. These concerns have been behind numerous recent studies designed to obtain detailed information about Chinese Tuber species. Unfortunately, many of these studies are not published in English, and are dispersed throughout the specific local or national bibliography and proceedings of specialized truffle conferences. In view of the need to expand current knowledge of Chinese Tuber species, we present a comprehensive summary of the taxonomy, ecology, mycorrhizae, genetics, biochemistry, and cultivation of Chinese Tuber species. We also provide a synthetic taxonomy and morphological characterization of 16 Chinese Tuber species in order to assist in their verification and monitoring.  相似文献

The Chinese black truffle Tuber indicum is commercially valuable. The main factors influencing the success or failure of a truffle crop include the mycorrhizal colonization rate and host plant quality. The effects of a plant growth regulator (uniconazole) and plant growth management technique (root-tip cutting) on T. indicum colonization rate and Pinus armandii seedling growth were assessed under greenhouse conditions. The results indicated that 10 mg l−1 uniconazole or the combination of 5 mg l−1 uniconazole and root-tip cutting constitutes an effective method for ectomycorrhizal synthesis based on an overall evaluation of colonization rate, plant biomass, plant height, root weight, stem circumference and antioxidant enzyme activities (SOD and POD) of P. armandii. The abundance of Proteobacteria in the rhizosphere of colonized seedlings might serve as an indicator of stable mycorrhizal colonization. This research inspires the potential application of uniconazole and root-tip cutting treatments for mycorrhizal synthesis and truffle cultivation.  相似文献
