Two diseases of Oryctes rhinoceros caused by Metarrhizium anisopliae and a virus were introduced from Western Samoa into Tongatapu, Kingdom of Tonga, in 1969 and 1970, respectively. The fungus remained at a low incidence and did not significantly affect the beetle population. The virus disease developed to epizootic levels within 5 months and spread across the island in 15 months. The beetle population was monitored through palm damage surveys, which confirmed that the population was considerably reduced by the disease.The greater impact of the virus was attributed to its much higher power of dispersal through infected beetles.The experiment supports the view that the reduction in beetle numbers recorded previously in Western Samoa was caused by a virus epizootic.  相似文献

In view of the increasing and devastating damage by rhinoceros beetle (Oryctes rhinoceros) to coconut palms in the middle of last century, many efforts were made to find an efficient natural control factor against this pest, which could not be controlled by pesticides. The basic procedures of these monitoring programmes are outlined together with the final detection of a virus disease in oil palm estates in Malaysia in 1963. In extensive laboratory studies, the virus was isolated and identified as the first non-occluded, rod-shaped insect virus, morphologically resembling the baculoviruses. Infection experiments clarified the pathology, histopathology, and virulence of the virus and demonstrated that the virus was extremely virulent to larvae after peroral application. These findings encouraged the first pilot release of virus in 1967 in coconut plantations of Western Samoa where breeding sites were contaminated with virus. Surprisingly, the virus became established in the Samoan rhinoceros beetle populations and spread autonomously throughout the Western Samoan islands. As a consequence, there was a drastic decline of the beetle populations followed by a conspicuous recovery of the badly damaged coconut stands. This unexpected phenomenon could only be explained after it was shown that the adult beetle itself is a very active virus vector and thus was responsible for the efficient autodissemination of the virus. The functioning of the beetle as a 'flying virus factory' is due to the unique cytopathic process developing in the midgut after peroral virus infection. Pathological details of this process are presented. Because of the long-term persistence of the virus in the populations, rhinoceros beetle control is maintained. Incorporation of virus into integrated control measures and successful virus releases in many other countries are recorded.  相似文献

The incidence and distribution of the baculovirus of the coconut palm rhinoceros beetle was determined in Tongatapu, Tonga, 7 years after being first introduced. Surveys showed the virus to be spread throughout the beetle population, affecting 14.6% of breeding sites and over 84% of all adult beetles taken. Counts of damaged palms and of breeding site occupancy indicated that beetle numbers had remained at low levels. It appears that the virus has the potential for long-term control of beetle populations in these habitats.  相似文献

The rhinoceros beetle, Oryctes rhinoceros, has emerged as a serious pest of oil palm since the prohibition of burning as a method for maintaining estate hygiene in the 1990s. The abundance of beetles is surprising given that the Malay peninsula was the site of first discovery of the Oryctes virus, which has been used to effect good as a biological control agent in other regions. A survey of adult beetles was carried out throughout Malaysia using pheromone traps. Captured beetles were examined for presence of virus using both visual/microscopic examination and PCR detection methods. The survey indicated that Oryctes virus was common in Malaysia among the adult beetles. Viral DNA analysis was carried out after restriction with HindIII enzyme and indicated at least three distinct viral genotypes. Bioassays were used to compare the viral strains and demonstrate that one strain (type B) is the most virulent against both larvae and adults of the beetle. Virus type B has been cultured and released into healthy populations where another strain (type A) forms the natural background. Capture and examination of beetles from the release site and surrounding area has shown that the spread and persistence of the applied virus strain is accompanied by a reduction in palm frond damage.  相似文献

A follow-up of a Baculovirus oryctes release into the population of Oryctes monoceros in coconut plantations in Tanzania has been made to clarify important points which were not fully considered in an earlier publication. The low recovery of virus-infected field-collected beetles (29%) suggested that the release of B. oryctes virus was not a suitable method for the control of O. monoceros. The relatively dry conditions in the coconut growing areas were partly contributory since the virus is non-occluded. No significant and sustainable reduction in frond damage was obtained after the virus had established itself in the natural O. monoceros population. The status of O. monoceros as a serious pest of coconut is also revised since the beetle affects young plantations more than older plantations and control methods can be directed at picking up feeding insects by hooking and disposal of breeding sites.  相似文献

Abstract.
  • 1 Males and females of Oryctes rhinoceros (L.), a serious pest of coconut palms, congregate in decaying coconut trunks to mate, oviposit and prepare the wood for the young larvae. A baculovirus disease is often transmitted from beetle to beetle in this environment and transmission to and from larvae probably also occurs.
  • 2 Young beetles are mainly found feeding on palms, where they also frequently contract the virus disease, presumably through mating with diseased partners.
  • 3 Monitoring beetle populations in the Philippines with attractant traps and by collections from palms showed seasonal fluctuations which were often inversely correlated with the incidence of the virus disease. Disease prevalence matched closely the proportion of females in the population. Possible explanations for this correlation and for the seasonality of the disease prevalence are outlined.

Genetic pest management (GPM) methods involve releasing modified versions of a pest species to mate with wild pests in the target area. Proposed for a wide range of applications in public health, agriculture and conservation, most progress has been made with pest insects. Offspring of the released modified insects and wild pests carry the modification—which might be transgenes, artificially introduced Wolbachia or genetic damage from radiation, for example—but they also carry a complete haploid genome from their laboratory-reared parent, as well as one from their wild parent. Unless these F1 hybrids are completely unable to reproduce, further mating will lead to introgression of DNA sequences from the release strain into the wild population. We discuss issues around strain selection and the potential consequences of such introgression. We conclude that such introgression is probably harmless in almost all circumstances, and could, in theory, provide specific additional benefits to the release programme. We outline population monitoring approaches that could be used, going forward, to determine how background genetics may affect GPM.This article is part of the theme issue ‘Novel control strategies for mosquito-borne diseases’.  相似文献

The prospect of autocidal insect control was investigated in a cellar population of D. melanogaster using a compound-chromosome stock. The released stock was synthesized by irradiating virgin female progeny derived from the cellar and crossing to a second-chromsome compound laboratory stock. Incorporation of an appropriate genetic background into the compound stock was tested in laboratory studies. Larval development to adult emergence and adult survival studies indicated the compound release stock to be relatively similar to the wild population, while behavioral tests detected no mating isolation between wild or compound genotypes. An unstable equilibrium point of compound frequency 0.7 was observed in population cage experiments with the two genotypes.

Adults were released into the cellar at a 50:1 ratio in favor of the compound. Five hundred newly hatched compound-chromosome larvae were also released. Adult, larval and pupal samples were regularly made. The compound stock successfully bred in the cellar maintaining an adult frequency of at least 90% for 108 days after the release. The rapid decline in compound frequency after this period is thought to be due to the migration of inseminated wild-type females from wine storage areas adjacent to the cellar.

The results indicate that a compound stock may limit the rate of population expansion in an area and may be a useful mechanism of autocidal control. It cannot be overemphasized that the probability of a successful release will be related to the level of understanding of the adaptive strategy of the population into which the release is made.

Rhabdionvirus oryctes multiplies in adults of Oryctes rhinoceros. Adults and larvae of O. rhinoceros showed no significant difference in their susceptibility to the virus when it was inoculated into the hemocoel. Adults, collected from the natural population in Western Samoa, were found to be frequently infected with Rhabdionvirus. In the laboratory the infected adults had a shorter life-span and laid fewer eggs than the noninfected ones. Infected adults excreted virus material into the surrounding medium. This suggests a possible means of transmission of the virus in the field.  相似文献

The Korean insect industry is rapidly developing, with the Korean horn beetle Allomyrina dichotoma as one of the most popular insects kept as pets. Korean horn beetles reared in local farms are suffering from Oryctes rhinoceros nudivirus (OrNV). In a nationwide investigation on the early death of young larvae due to this virus, 70 to 77% mortality was found. It was also confirmed that several larvae collected from some wild horn beetle habitats were infected with the virus. Thus, it was concluded that the virus could be transmitted vertically from an infected adult to the offspring and that the OrNV disease is a very serious threat to the wild horn beetle in Korea. With local farmers expecting that an early detection technique would be helpful for the quick removal of infected larvae, an on‐site diagnosis method for the viral disease using loop‐mediated isothermal amplification (LAMP) assay was thus tested. To avoid the DNA extraction process, the LAMP assay used a 50‐fold diluted hemolymph was set for diagnosis standardization and convenient on‐site application to infected larvae from local farms. A. dichotoma larvae were assayed for the OrNV infectivity with LAMP primers targeting the OrNV_gp102 gene. To evaluate the LAMP specificity, two bacterial pathogens, Bacillus thuringiensis and Serratia marcescens, causing disease in A. dichotoma were tested along with OrNV. The positive results were detected only from the OrNV‐infected larvae.  相似文献

食品安全已成为当今社会广受关注的问题。尽管国内的一些学者就阿里山潜蝇茧蜂[Fopius arisanus(Sonan)]和橘小实蝇SIT技术分别开展了不同程度的研究,然而关于应用该蜂和不育雄虫对橘小实蝇[Bactrocera dorsalis(Hendel)]进行田间联合防治的研究还未见相关报道。为此,为探讨天敌昆虫和雄性不育技术对橘小实蝇的田间综合治理(IPM)技术,开展了阿里山潜蝇茧蜂和橘小实蝇不育雄虫的田间释放试验:首先应用性引诱剂降低田间野生雄虫数量;而后,释放橘小实蝇不育雄虫,进一步降低田间橘小实蝇种群数量;最后,释放阿里山潜蝇茧蜂对田间残余的橘小实蝇卵进行追踪寄生,以达到持续控制的目的。通过监测整个防控期田间橘小实蝇的种群数量以及果实受害率和防治效果作为评价指标评价该防治方法的综合防治效果。结果表明:应用阿里山潜蝇茧蜂、橘小实蝇不育雄虫并配合引诱剂的综合措施对橘小实蝇的种群数量有明显控制作用,田间总体防治效果可达90%左右。此外,提出完善橘小实蝇田间防控措施的建议,并提出该研究的不足以及有待进一步开展研究的方向。  相似文献

Background

A rapid rise in the number of captive African elephants (Loxodonta africana) used in the tourism industry in southern Africa and orphaned elephants in human care has led to concerns about their long-term management, particularly males. One solution is to release them into the wild at adolescence, when young males naturally leave their herd. However, this raises significant welfare concerns: little is known about how well released elephants integrate into wild populations and whether they pose a greater threat to humans than wild elephants. We document the release of three captive-raised adolescent male African elephants in the Okavango Delta, Botswana.

Methodology/Principal Findings

Despite having been part of a herd of working elephants for at least eight years, the three males progressively integrated into the complex fission-fusion society of wild bull elephants. In the three years following release, they showed no tendency to be closer to human habitation, and there were no significant differences between wild and captive-raised adolescent males in the total number of social interactions, size of ranges and habitat use. However, the captive-raised elephants sparred less and vocalised more, and spent more time alone and in smaller social groups. Thereafter the released elephants continued to expand their ranges and interact with both mixed-sex herds and males. One male was shot by farmers 94 months after release, along with ten wild elephants, on a ranch outside the protected area.

Conclusions/Significance

We show that captive-raised adolescent male elephants can integrate into a wild population. Long-term studies are required to determine the longevity, breeding success, and eventual fate of released male elephants, but we identified no significant short-term welfare problems for the released elephants or recipient population. Release of captive-raised mammals with complex social systems is a husbandry option that should be explored further.  相似文献

The introduction of Oryctes virus into outbreak areas of the rhinoceros beetle, Oryctes rhinoceros (Coleoptera: Scarabaeidae), has been a major success for "classical" biocontrol with a virus and has led to a dramatic reduction in palm damage in many areas of the Asia/Pacific region. In recent years, however, there have been new reports of high levels of rhinoceros beetle damage to palms. Damage has been especially intense in SE Asia following the introduction of no-burn polices for land clearance and replanting, but outbreaks have also been reported from some Pacific Islands where control seems to have diminished over time. SE Asian studies show that there is considerable genetic variation among endemic Oryctes virus isolates and studies in new island release areas have shown rapid evolution of the virus. The consequences of such genetic variation are in need of further study. Furthermore, the taxonomic position of the virus is unclear, with its removal from the Baculoviridae to an "unassigned' virus, reflecting its novel characteristics. Genomic sequencing could help resolve the taxonomy of the virus and provide a basis for studying strain variation. Oryctes virus has achieved wide success in the past without the benefit of molecular analysis and identification techniques. In order to fully take advantage of this unique pathogen for protection of palms, a renewed, coordinated effort centered on genetic selection and distribution of effective strains is required.  相似文献

The oil palm is an economically important crop cultivated in the North of Brazil. Damage caused by insects is one of the main causes of reduced productivity for the oil palm. Before this research, only the beetles of the family Curculionidae were considered to be oil palm pests in Brazil. However, for the first time, we report on the damage caused by a giant rhinoceros beetle to oil palm plantations in Pará, Brazil. The beetle was identified as Golofa claviger (Linnaeus, 1771), which has a single record in Brazil (Pará) but is widely distributed in South America. The species occurs in an unprecedentedly high abundance of local specimens. The attacks are concentrated on the central cluster of young palms. Feeding behavior is identified as the main cause of the damage as the beetles use their mouthparts to rip the plant tissues, causing wedge-shaped cuts on young fronds that have not yet unfurled. After an attack, the leaflets of the unfurled fronds are partially destroyed.  相似文献

The European hardwood ambrosia beetle (Trypodendron domesticum) and the striped ambrosia beetle (Trypodendron lineatum) are wood‐boring pests that can cause serious damage to lumber, resulting in a need for management of these pests in logging and lumber industries. Natural populations of ambrosia beetles exist throughout the world, but movement of ambrosia beetles into new habitats, particularly via international trade, can result in the establishment of invasive species that have the potential to spread into new territory. Efforts to monitor ambrosia beetle populations are time‐consuming and could be greatly enhanced by the use of molecular methods, which would provide accurate and rapid identification of potentially problematic species. Here, we present new real‐time PCR assays for the detection and identification of T. domesticum and T. lineatum. The methods described herein can be used with a variety of sampling strategies to enable timely and well‐informed decision‐making in efforts to control these ambrosia beetles.  相似文献

We tested the hypothesis that the population density of ambrosia beetles at the stand level influences the spatial distribution of infested trees. We evaluated the spatial distribution of the ambrosia beetle, Platypus koryoensis (Murayama) in three oak forest stands that varied in beetle population density using a multi-year trapping survey. We used these data to inform a clustering analysis based on aggregation indices using the SADIE software. Four important findings emerged: (1) the spatial distribution pattern of P. koryoensis at the stand level changed as the population density of the beetle varied; (2) at low population densities, beetle distribution was contagious at the stand level; (3) as beetle population densities increased, the spatial distribution of infested trees became random, potentially due to beetle avoidance of mass attacked trees; and (4) at high beetle population densities, the spatial distribution of infested trees became contagious, possibly due to temporal changes in location of the attack epicenter within the stand. Our results support the hypothesis that beetle population density has consequences for the spatial distribution of infested trees at the within-stand scale. We conclude that the spatial distribution of infested trees is flexible in response to beetle population density, suggesting that beetle attack behaviors are mediated by one or more density-dependent effects.  相似文献

As suitable habitat becomes increasingly fragmented the ability of a species to reach new areas may often dictate whether populations persist or perish. An understanding of dispersal ability is a prerequisite for informed management decisions. This is particularly true for species that have become restricted to one or a small number of sites, as is the case for several species of Cryptocephalus beetle in the UK. Using mark–release–recapture, host-plant marking and direct observations we investigated population size, movement through suitable habitat and the flight behaviour of Cryptocephalus decemmaculatus. In the UK this species is known from only two sites and it is listed as a priority species on the UK Biodiversity Action Plan. At the time of the study, the one known English site for this species supported a population of ~500 individuals. The adult beetles move through their habitat using suitable host-plants as ‘stepping stones’. There appears to be no interchange of adults between sub-populations that are separated by small areas of unsuitable habitat (e.g. small tracts of woodland or areas devoid of scrub), a result reinforced by studies of genetic differentiation between sub-populations. The small population size of this beetle and its association with, mid-successional habitats makes it vulnerable to local extinction. Furthermore, its limited dispersal ability means that other habitat patches are unlikely ever to be colonised naturally.  相似文献

Oryctes rhinoceros adults stopped feeding and flew less frequently, and the males mated less often after they became infected with the baculovirus of Oryctes. Monitoring the larval and adult population in Western Samoa over several years gave evidence that the incidence of the virus is dependent on the density of the O. rhinoceros population. Male beetles were more frequently infected than female bettles. Adding more virus-infected beetles to the O. rhinoceros populations of Nukunonu Atoll, where the virus had become established 5 years earlier, reduced the population significantly. The data indicated that the reduction was caused mainly by a decrease in the number of eggs deposited.  相似文献

The infestation levels of O. rhinoceros were investigated in the oil palm trunk heaps in three replanting blocks (Blocks A, B and C) at Sepang, Selangor, Malaysia. These blocks had different densities of pheromone trapping: high (11 traps per ha, Block B), normal (1 trap per 2 ha, Block C), while no pheromone trapping was conducted in (Block A. Without pheromone trapping, the infestation levels of O. rhinoceros can reach a maximum density of between 25 to 50 individuals per m2 (IPMS). The O. rhinoceros population was still detected in the heaps up to more than two years (at 26 months after replanting). With high density trapping, infestation levels were maintained below 10 IPMS and was not detected in the heaps after a period of 16 months of replanting. In the normal trapping density, the population was also maintained below 10 IPMS but had sustained itself in the heaps for up to 24 months. The mortality factors associated with the larval stages sampled within these replanting blocks was determined by visual symptoms with subsequent verification with microbial analysis. The analysis was narrowed down to detect only bacteria (Bacillus thuringiensis and B. popillae), virus (Oryctes virus) and fungus (Metarhizium). In Block A, the population of O. rhinoceros in the trunks was reduced drastically at 14 months after replanting (MAR), which could be due to the high mortality (88%) of the third instar larvae, 3 months earlier. In Block B, the percentage mortality for the larval stage was always highest at the third instars (between 21–82%). In Block C however, the mortality of the third instars was low, never exceeding 35%. In these sampling blocks, the infection of Metarhizium seem to be less dominant compared to virus or bacteria The k factor analysis indicates density dependence among the third instar larvae had contributed to the change in its population density in Block A. Therefore, with proper manipulation of pheromone traps and targeted application of disease agents (i.e virus, fungus and bacteria), the population of O. rhinoceros can be managed more effectively in an oil palm replanting.  相似文献
