ABSTRACT. The minimal copulation frequency of the reproductives of Macrotermes michaelseni (Sjöstedt) was investigated by removing the king from incipient laboratory colonies and from mature field colonies. Laboratory females stopped producing fertile eggs 56–154 days, and physogastric queens from field colonies less than 13 days after the removal of the king. Compared with young laboratory male reproductives, kings from mature field colonies are reported to have enormously enlarged testes, larger corpora allata (CA) and a higher juvenile hormone (JH) titre. However, JH application to young males did not increase the volume of the testes. The testes volumes only increased if the males were kept with additional workers or if their incipient colonies were allowed to grow continuously under unrestricted space conditions. These males also had bigger CA, and the soil in their midgut was replaced by food from the workers. It is concluded that in a growing colony the king is differently fed by the increasing number of workers and that trophic change in combination with higher CA activity induce enlargement of the testes. These enlarged testes are able to produce the sperm to fertilize the large number of eggs laid by a physogastric queen.  相似文献

Basal oöcyte length, corpus allatum volume and “in vitro” juvenile hormone biosynthesis were measured in isolated and crowded Locusta migratoria females at selected times during the first gonotrophic cycle. Using gas chromatography-mass spectrometry with selected ion monitoring, the juvenile hormone titre in the haemolymph of isolated and crowded females was also determined 1 and 4 days after fledging. The rate of oöcyte growth was more rapid in isolated females and a significant (P < 0.01) difference in mean length was apparent as early as 3 days after fledging. This early manifestation of a difference in rate of oöcyte growth was correlated with a difference in haemolymph juvenile hormone titre between isolated and crowded females. Whilst there was no difference in titre 1 day after fledging, by day 4 the juvenile hormone titre in isolated females was found to be approximately twice that in crowded females. There was no significant difference in the rates of juvenile hormone biosynthesis by corpora allata from isolated and crowded females on days 0 through to 6 after fledging. On day 8, however, the rates of juvenile hormone biosynthesis of corpora allata from isolated females were very high (mean value = 136 pmol/h/pair) and were significantly (P < 0.002) greater than those of corpora allata from crowded females. Day 8 was also the point in the first gonotrophic cycle at which the difference in the mean basal oöcyte length in isolated and crowded females was at a maximum. The mean volume of corpora allata from isolated females was greater than that of corpora allata from crowded females at all points at which measurements were taken during the first gonotrophic cycle.  相似文献

The corpora allata of castrated females of Nauphoeta grow only very slightly and do not reach a volume greater than that of the glands of normal females during gestation. These small corpora allata are, however, active and are responsible for the synthesis of vitellogenin (female specific protein) in large amounts. Besides vitellogenin the other haemolymph proteins are also synthesized and accumulated in the haemolymph in much higher concentrations than in normal females. Implanted oöcytes grow in castrated as well as in normal females at about the same rate until the tenth day of the oöcyte maturation period. Thereafter they only grow in castrated females. If castrated and normal females are decapitated, their protein content decreases. At the same time the growth stimulating capacity of their haemolymph decreases at a much faster rate. If oöcytes are implanted in castrated and decapitated females after 4 days they cannot grow any more although the vitellogenin titre of the haemolymph is still much higher than it is at any time in normal females. It can be concluded that vitellogenin alone cannot induce oöcyte growth and that juvenile hormone is necessary as well for vitellogenin synthesis as for its incorporation into the oöcytes. However, in insects rich in vitellogenin juvenile hormone leads to a more rapid oöcyte growth than in insects containing only small amounts of this protein.  相似文献

Morphometric studies were made on corpora allata of the cockroach Diploptera punctata from animals in which increasing gland size is not coupled to hormone synthesis (ovariectomized mated females; last-instar larvae) and in which gland size is coupled to hormone synthesis (normal mated and virgin females; penultimate-instar larvae). Cell number, gland volume, and juvenile hormone synthesis were measured. From electron micrographs, nuclear, cytoplasmic, and extracellular volumes; and cell membrane area were calculated; and fine structure described. Low-activity glands of ovariectomized mated females resembled high-activity glands from mated females in high cell number, large overall and cytoplasmic volume, and low nuclear-cytoplasmic ratio; they differed in having organelles typical of low-activity glands, mitochondria with dense matrices and large whorls of smooth endoplasmic reticulum. Inactive lastinstar larval glands resembled mated ovariectomized, female glands in increased cell number and organelles characteristic of inactive glands; however, their nuclearcytoplasmic volume ratio was much higher. Penultimate cytoplasmic volume ratio was much higher. Penultimate larval glands with high activity per cell resembled active glands of normal mated females. Ovariectomy did not change morphometric parameters of virgin female glands; thus mating results in increase in size of adult female glands whereas the growing ovary is needed for changes in mitochondria and endoplasmic reticulum associated with high juvenile hormone synthesis.  相似文献

When two-day-old female Leptinotarsa decemlineata were starved, their corpus allatum activity, as measured by the radiochemical in vitro assay, was significantly reduced after 24 hr. Such a reduction was not observed when the nerve connections between the central nervous system and the retrocerebral complex were severed and the beetles starved up to 5 days. In some experiments, the rate of juvenile hormone biosynthesis in vitro, was substantiated by measurement of the juvenile hormone titre in the haemolymph by physico-chemical methods. It is concluded that intact nervous connections between the central nervous system and the corpora allata are essential for restraining the juvenile hormone biosynthesis during the initial stages of starvation.Corpora allata from 1-day starved insects were considerably stimulated in vitro by farnesenic acid indicating that juvenile hormone synthesis is controlled enzymatically at a stage prior to the final two steps in the pathway. However, on day 5 of starvation, rate-limitation may occur after formation of this intermediate, since farnesenic acid stimulation was much less at this time.Corpora allata of adult females newly emerged from the soil were activated within 4 hr regardless of feeding.  相似文献

The factors responsible for the initiation of a second oöcyte maturation cycle were investigated by measuring oöcyte growth, vitellogenin titre, and corpus allatum activity after injection of juvenile hormone and/or removal of the egg-case from pregnant females and by performing ovary and corpus allatum transplant experiments.Egg-case removal in late pregnancy results in immediate oöcyte growth, whereas in early pregnancy oöcyte growth is resumed only after a lapse of time, even after injection of juvenile hormone. This, however, induces an immediate increase in the haemolymph vitellogenin titre. A single injection of 2 or 10 μg of juvenile hormone II first stimulates some oöcyte growth after this lapse of time and later activates the corpora allata, which in turn leads to completion of oöcyte maturation. A repeat injection of 10 μg stimulates continuous oöcyte growth without activating the corpora allata. In the presence of an egg case, activation of the corpora allata is suppressed, even after injection of 2 μg of juvenile hormone III, and the oöcytes do not grow. Injection of higher doses stimulates oöcyte growth and leads to expulsion of the egg case in up to 95% of the females. This, however, is not a direct consequence of the increase in size of the ovaries. Ovary transplant experiment show that in young pregnant females the second generation of oöcyte is not yet competent for growth and that ovaries which are competent can mature in young pregnant females, treated with juvenile hormone, whose egg case has been removed.The results are summarized in a model demonstrating the various factors involved in regulating corpus allatum activity in oöcyte maturation and pregnancy and after application of juvenile hormone. We prepose that the corpus allatum activating effect of exogenous juvenile hormone is mediated by the growing oöcyte and that this activation can be suppressed by the continuous presence of exogenous juvenile hormone.  相似文献

The effects of increased levels of dopamine (feeding flies with dopamine precursor, l-dihydroxyphenylalanine) and octopamine (feeding flies with octopamine) on ecdysone 20-monooxygenase activity in young (2 days old) wild type females (the strain wt) of Drosophila virilis have been studied. l-dihydroxyphenylalanine and octopamine feeding increases ecdysone 20-monooxygenase activity by a factor of 1.6 and 1.7, respectively. Ecdysone 20-monooxygenase activity in the young (1 day old) octopamineless females of the strain Tβh nM18 , in females of the strain P845 (precursor of Tβh nM18 strain) and in wild type females (Canton S) of Drosophila melanogaster have been measured. The absence of octopamine leads to a considerable decrease in the enzyme activity. We have also studied the effects of juvenile hormone application on ecdysone 20-monooxygenase activity in 2-day-old wt females of D. virilis and demonstrated that an increase in juvenile hormone titre leads to an increase in the enzyme activity. We discuss the supposition that ecdysone 20-monooxygenase occupies a key position in the regulation of 20-hydroxyecdysone titre under the conditions that lead to changes in juvenile hormone titre and biogenic amine levels.  相似文献

It is known that juvenile hormone plays an important role in the regulation of labour division and of the different life spans, and that the microclimate of the bee hive is characterized by its high CO2 concentration and its varying temperature depending on the presence of brood.We have investigated the influence of microclimates characteristic of breeding and broodless areas on the juvenile hormone titre in the haemolymph and whole body extracts, on the corpora allata in vitro activity, on the degradation of juvenile hormone and on the dry weight of the hypopharyngeal glands using bees of known ages. A microclimate of 35°C and 1.5% CO2, as observed in the breeding area, induces a rapid and pronounced increase in the juvenile hormone titre. On the other hand, this titre remains low in bees kept at 27°C and 1.5% CO2, a microclimate associated with broodless combs. Rates of juvenile hormone synthesis by corpora allata in vitro were found to be extremely low, even in the presence of farnesenic acid, and not related to the juvenile hormone titre. In vitro incubation of juvenile hormone in haemolymph revealed no degradation while injected juvenile hormone was found to be degraded and taken up by the gut at rates only weakly correlated with the juvenile hormone titre.We propose a hypothetical model for the regulation of the juvenile hormone titre as well as the course of labour division by the varying microclimates observed in the bee hive.  相似文献

When adult apterous viviparous females of Myzus persicae, reared in short night conditions at 21–23°C, are treated with the precocene analogue 6-methoxy-7-ethoxy-2,2-dimethylchromene, they deposit males towards the end of their reproductive lives. The first-born normal daughters of the treated females also deposit some males at various times in their reproductive lives. Karyotypic analysis was used to investigate the sequence of male and female embryos in the ovarioles of precociously metamorphosed aphids. The experiments support the hypothesis (Mittler et al., 1979) that juvenile hormone level controls the sex determination process in aphids. Since male aphids have an XO sex chromosome constitution, this implies that juvenile hormone level influences the behaviour of the X-chromosomes at or before the single maturation division of the egg. At this division one X-chromosome is eliminated from eggs which will develop as males. Aphids provide the first example of a specific endocrine influence on chromosome behaviour in sex determination.  相似文献

In the cricket, Teleogryllus commodus, eggs, haemolymph of 7th and 8th (last)-larval instars, and haemolymph of adults of both sexes contain only juvenile hormone III. While in the male the hormone titre is independent of previous mating experience, juvenile hormone concentration in haemolymph taken from females 36–38 hr after mating (an event which is followed by oviposition) is at a level 5 times higher than that of virgin females. Based on data gleaned from several research groups the identification of juvenile hormone III as the exclusive juvenile hormone in the Order Orthopteroidea is discussed.  相似文献

The in vivo control of corpus allatum (CA) activity in females of Leptinotarsa decemlineata was investigated. Evidence was obtained that CA activity is adjusted by negative feedback when juvenile hormone (JH) titres are changed experimentally. Conclusions are based on determination of the rate of in vitro JH synthesis by the CA, on changes in CA volume, and on JH titres in the haemolymph. These assay methods are used alternatively in some of the experiments.After unilateral allatectomy, the remaining CA had doubled its activity 7 days later. On the other hand, the activity of CA in young adults was suppressed after the JH titre was elevated by the implantations of 2 CA taken from active females. Similarly, in beetles treated topically with exogenous JH the CA atrophied and showed a much reduced activity after 5 days. Denervation of CA in 0-day-old long-day and 7-day-old short-day females did not change CA activity when measured 1 day later.  相似文献

There is an age-related division of labor in the honey bee colony that is regulated by juvenile hormone. After completing metamorphosis, young workers have low titers of juvenile hormone and spend the first several weeks of their adult lives performing tasks within the hive. Older workers, approximately 3 weeks of age, have high titers of juvenile hormone and forage outside the hive for nectar and pollen. We have previously reported that changes in the volume of the mushroom bodies of the honey bee brain are temporally associated with the performance of foraging. The neuropil of the mushroom bodies is increased in volume, whereas the volume occupied by the somata of the Kenyon cells is significantly decreased in foragers relative to younger workers. To study the effect of flight experience and juvenile hormone on these changes within the mushroom bodies, young worker bees were treated with the juvenile hormone analog methoprene but a subset was prevented from foraging (big back bees). Stereological volume estimates revealed that, regardless of foraging experience, bees treated with methoprene had a significantly larger volume of neuropil in the mushroom bodies and a significantly smaller Kenyon cell somal region volume than did 1-day-old bees. The bees treated with methoprene did not differ on these volume estimates from untreated foragers (presumed to have high endogenous levels of juvenile hormone) of the same age sampled from the same colony. Bees prevented from flying and foraging nonetheless received visual stimulation as they gathered at the hive entrance. These results, coupled with a subregional analysis of the neuropil, suggest a potentially important role of visual stimulation, possibly interacting with juvenile hormone, as an organizer of the mushroom bodies. In an independent study, the brains of worker bees in which the transition to foraging was delayed (overaged nurse bees) were also studied. The mushroom bodies of overaged nurse bees had a Kenyon cell somal region volume typical of normal aged nurse bees. However, they displayed a significantly expanded neuropil relative to normal aged nurse bees. Analysis of the big back bees demonstrates that certain aspects of adult brain plasticity associated with foraging can be displayed by worker bees treated with methoprene independent of foraging experience. Analysis of the over-aged nurse bees suggests that the post-metamorphic expansion of the neuropil of the mushroom bodies of worker honey bees is not a result of foraging experience. © 1995 John Wiley & Sons, Inc.  相似文献

In the subfamily Polistinae, caste dimorphism is not pronounced and differences among females are primarily physiological and behavioral. We investigated factors that indicate the reproductive status in females of Polistes ferreri Saussure. We analyzed females from nine colonies and evaluated morphometric parameters, ovarian development, occurrence of insemination, relative age, and cuticular chemical profile. The colony females showed three kinds of ovarian development: type A, filamentous ovarioles; type B, ovarioles containing partially developed oocytes; and type C, long and well-developed ovarioles containing two or more mature oocytes. The stepwise discriminant analysis of the cuticular chemical profile showed that it was possible to distinguish the three groups of females: workers 1, workers 2, and queens. However, the stepwise discriminant analysis of the morphological differences did not show significant differences among these groups. The queens were among the older females in the colony and were always inseminated, while the age of the workers varied according to the stage of colony development.  相似文献

Programmed cell death in the worker ovary of Apis mellifera reduces the number of ovarioles during metamorphosis from 150-200 primordia to less than 10. In contrast, practically all ovarioles in the ovary of queens survive to the adult stage. The correct formation and persistence of polyfusomes has been suggested as a critical factor for ovariole survival. We have analyzed the developmental dynamics of F-actin and alpha-spectrin in fusomes of queen and worker larvae, and in juvenile-hormone-treated worker larvae. Small fusomes containing actin and spectrin can be detected in the ovaries of fourth instar larvae in both castes. After molting to the fifth instar, the actin-spectrin association persists in the enlarged fusomes of queen ovarioles. In workers, actin dissociates from the fusomal and cortical alpha-spectrin. Coinciding with the appearance of apoptosis markers, large agglomerates of actin are detectable in worker ovarioles. Treatment of fourth-instar worker larvae with juvenile hormone rescues ovarioles from apoptosis and maintains the actin-spectrin association. Juvenile-hormone-dependent actin-spectrin interaction is thus one of the earliest steps in the differentiation of a polymorphic ovary. Plasticity in ovariole numbers as a result of hormone-dependent fusome formation may be a more widespread phenomenon in insects, extending beyond caste polymorphism in highly eusocial Hymenoptera.  相似文献

Penultimate-instar larvae of Bombyx mori were neck-ligated or ligated posterior to the prothoracic glands. Repetitive injections of 20-hydroxyecdysone every 3 or more hours elicited the gut purge in thorax-ligated animals. Single injections of 20-hydroxyecdysone up to 40 μg failed to induce the gut purge. However, a single injection of 20-hydroxyecdysone together with juvenile hormone analogue, resulted in larval moulting of thorax-ligated animals. Once the thorax-ligated larvae showed the gut purge, a single injection of 20-hydroxyecdysone was enough to provoke pupation. The change in ecdysteroid titre in those animals receiving repeated injections was compared with that observed in neck-ligated larvae that spontaneously underwent the gut purge followed by precocious pupation. These data indicate that the very low ecdysteroid titre found before the gut purge is important for the acquisition of competence to undergo the gut purge in response to a small ecdysteroid surge just before the gut purge.  相似文献

We have found that whether a female German cockroach, Blattella germanica (L.), is kept alone or in the presence of another female has a major impact on how fast it reproduces and how much it eats. By the sixth day of adulthood, females paired since adult eclosion had substantially larger o?cytes than did females isolated during the same time, and females paired with intact females, or with ones rendered incapable of feeding, consumed more rat chow in the first six days of adulthood than did isolated females. The stimulatory effect of pairing on reproduction was, however, partially independent of feeding because the o?cytes of solitary and paired females differed in size on day 6 even when they were given, and had consumed, the same amount of food. This result was confirmed with analysis of covariance using the total food intake of a female as the covariate in the analysis. A female's social condition probably influenced the development of its o?cytes by affecting the quantity of juvenile hormone synthesized by its corpora allata. The corpora allata of paired females produced more hormone than did those of isolated ones, even when all females had consumed an equivalent amount of food. Moreover, females treated with a juvenile hormone analog, fenoxycarb, reproduced more quickly than identically reared and fed control females, showing that juvenile hormone could influence reproduction independently of feeding. We conclude that both group rearing and food intake accelerate o?cyte development by diminishing the brain's inhibition on the synthesis of juvenile hormone.  相似文献

When the titre of juvenile hormone III in female Leptinotarsa decemlineata was elevated by the implantation of supernumerary corpora allata or by the injection of the hormone, the rate of endogenous hormone production by the host glands was significantly restrained, as determined by the short-term in vitro radiochemical assay. From denervation studies, it is suggested that during phases of elevated juvenile hormone titre, the corpus allatum activity is regulated via humoral as well as neural factors requiring intact nerve connections. Restrainment of gland activity appears to be mainly via the neural pathway. Isolated corpora allata were not influenced by 10?5 M juvenile hormone III added to the incubation medium in vitro.Studies with farnesenic acid revealed that the final two enzymatic steps in the biosynthetic pathway of juvenile hormone are also diminished during prolonged neural inhibition of the corpora allata.20-Hydroxyecdysone and precocene II had no apparent effect on the corpus allatum activity of Leptinotarsa decemlineata.  相似文献

Radioimmunoassay (RIA) of whole body extracts of Drosophila melanogaster males and females demonstrates that at eclosion all individuals contain high levels of ecdysteroid. Highly polar ecdysteroids (presumably metabolites) in the meconium represent approximately half of the total ecdysteroid RIA-activity present at this time and are subsequently eliminated. Ecdysteroids remaining after the elimination of the meconium are also highly polar as shown by reverse-phase high pressure liquid chromatography (RP-HPLC). The amount of ecdysteroid RIA-activity found in whole body extracts declines in both sexes until 18 h post-eclosion when levels begin to increase in the female and drop to undetectable levels in the male. In the female the ovaries are the major source of ecdysteroid. The increase in whole body ecdysteroid in the female coincides with the initiation of ovarian ecdysteroid production and accumulation. Topical application of methoprene, a juvenile hormone (JH) analog, stimulates ovarian ecdysteroid synthesis in apterous-ts 78j (apts 78j), a temperature-sensitive juvenile hormone-deficient mutant, corroborating previous results suggesting a role of juvenile hormone in ovarian ecdysteroid production. Stage 8–9 follicles, whose development is juvenile hormone dependent, are shown to be the most active in ecdysteroid production. The regulatory potential of these stages is discussed.  相似文献

Water loss rates (WLRs) varied across castes (workers > alate males > alate females) for desert ants in the genera Aphaenogaster, Messor and Pogonomyrmex. Exposure to soil caused increased WLRs in workers and foundresses (mated, dealate females), apparently because of cuticular abrasion caused by nest excavation. Moreover, field‐collected workers and foundresses from incipient nests (those exposed to soil) had similar WLRs, as did unabraded workers and alate females (those unexposed to soil). These data call into question previous adaptive scenarios for differences in WLRs across ant species and castes. For live alate females, WLRs increased over two stages. The first increase occurred immediately after mating, and the second occurred for foundresses collected 2 days later from incipient nests. By contrast, WLRs of HCN‐killed females were unaffected by mating, but increased significantly for foundresses collected from 2‐day‐old nests.  相似文献

Summary The changes in ovarian activity in the life cycle of the eusocial halictine beeLasioglossum (Evylaeus) duplex (Dalla Torre) was studied in both queens and workers by examining the stages of the terminal follicle in each of the six ovarioles. By this method, ovarian activities of both queens and workers were more quantitatively determined than by observation of gross ovarian features as usually conducted. Queen ovaries clearly exhibited two active periods, corresponding to the spring solitary phase and the summer eusocial phase, with distinctly greater activity in the latter. In ovaries of overwintering queens oosorption of young follicle was observed. Worker ovaries were found more active in orphan than in queenright colonies. The order of ovarian activity obtained from pooled data, summer queens>spring queens>orphan workers>queenright workers, was also recognized by comparison of individual females.
