The sex pheromone blend of the butterbur borer, Ostrinia zaguliaevi (Lepidoptera: Pyralidae) was analyzed by means of gas chromatography-electroantennographic detection (GC-EAD), GC-mass spectrometry and a series of wind-tunnel bioassays. Four EAD-active compounds were detected in the female sex pheromone gland extract, and these were identified as tetradecyl acetate (14:OAc), (Z)-9-tetradecenyl acetate (Z9-14:OAc), (E)-11-tetradecenyl acetate (E11-14:OAc) and (Z)-11-tetradecenyl acetate (Z11-14:OAc). The average amounts ± s.d. of the four compounds in a single sex pheromone gland were 7.9±3.7 ng, 10.1±3.2 ng, 1.1±0.5 ng and 11.6±5.1 ng, respectively. In a wind-tunnel bioassay, the ternary blend of Z9-, E11- and Z11-14:OAc at a ratio found in the sex pheromone gland (45:5:50) elicited the same behavioral responses from the males as did virgin females and pheromone gland extract. Removal of any single compound from the ternary blend significantly diminished the pheromonal activity, whereas addition of 14:OAc to the ternary blend had no effect on the males' behavioral responses. Therefore, it was concluded that the sex pheromone blend of O. zaguliaevi is composed of Z9-14:OAc, E11-14:OAc and Z11-14:OAc at a ratio of 45:5:50.

Four EAG-active components were detected in GC-EAG analyses of hexane extracts from virgin Etiella behrii (Zeller) (Lepidoptera: Pyralidae) females. These components were identified as dodecyl acetate (12:Ac), (E)-9-dodecenyl acetate (E9-12:Ac), either (Z)-9- or (E)-11-tetradecenyl acetate (Z9- or E11-14:Ac), and (Z)-11-tetradecenyl acetate (Z11-14:Ac) by comparison of retention indices on both nonpolar and polar GC columns. The available extract was insufficient for further GC-MS or other chemical analyses (<0.2 ng/female). In field tests carried out in East Java, a 10:90 mixture of E9-12:Ac and Z11-14:Ac showed attractiveness to male moths and addition of 12:Ac and/or E11-14:Ac significantly increased the trap catches while addition of Z9-14:Ac showed no significant effect. Maximum attraction was obtained with 5.35 or 10.7 g/rubber septum of a mixture of E9-12:Ac, Z11-14:Ac, 12:Ac and E11-14:Ac at the ratio of 10:90:0.7:6.3, respectively. The role of pheromone blends in species discrimination between E. behrii and the related E. zinckenella (Treitschke) is discussed.

Identification of the sex pheromone of Ostrinia palustralis
By means of gas chromatography with electroantennographic detection, gas chromatography-mass spectrometry and a series of bioassays, (E)-11-tetradecenyl acetate (E11-14:OAc) and (Z)-11-tetradecenyl acetate (Z11-14:OAc) at a ratio of 99:1 were identified as female sex pheromone components of Ostrinia palustralis. The average amounts of E11- 14:OAc and Z11-14:OAc in a single sex pheromone gland were 37.2±24.4 ng and 0.3±0.2 ng, respectively. In a wind-tunnel bioassay, the binary blend of E11- and Z11-14:OAc elicited the same male behavioral responses as did virgin females.

Previous attempts to shift the (Z)-11-/(E)-11-tetradecenyl acetate ratio of pheromone components in the redbanded leafroller moth (RBLR), Argyrotaenia velutinana, by several selection protocols showed that this ratio is strongly canalized. Analysis of the complete seven-component blend, however, showed that a Geneva laboratory stock of RBLR had a lower percent (20%) of the E9–12:OAc minor component compared to the E11–14:OAc component than a population of RBLR from North Carolina (31%). Hybrid populations from these two cultures were used in a two-way family truncation selection scheme in which families were selected for either the lowest (low line) or the highest (high line) ratio of E9–12:OAc/E11–14:OAc. After three generations of selection, the low line had 14% E9–12:OAc relative to E11–14:OAc and the high line had 42%. The selection pressure was removed in generations 4–9, and the low line remained unchanged at 14% E9–12:OAc; but in the high line, it drifted to 53%. Studies were conducted to estimate heritability and realized heritability. The realized heritability calculated for each generation of selection averaged 1.14 for the low line and 1.50 for the high line. These calculations, along with estimated heritability values of 0.416 and 0.644 for reciprocal crosses, indicate some plasticity in the E9–12:OAc/E11–14:OAc ratio. This ratio was positively correlated to the total amount of 12-carbon components to 14-carbon components, but was negatively correlated to the Z/E ratio of Δ11-tetradecenylacetates. The results of two studies on the canalization of various components of the RBLR sex pheromone blend indicate that there is limited potential in this insect for manipulation of the blend ratios in the laboratory.

Hellula undalis is a harmful insect pest of green mustard in the Mekong Delta of Vietnam. In order to establish a tool for a sustainable pest control program, the sex pheromone of H. undalis inhabiting the Mekong Delta was examined. GC-EAD and GC–MS analyses of pheromone gland extracts from the virgin females elucidated three new components, (Z)-11-tetradecenyl acetate (Z11-14:OAc), (Z)-11-hexadecenal (Z11-16:Ald), and (11E,13E)-11,13-hexadecadien-1-ol, in addition to the known pheromone component (11E,13E)-11,13-hexadecadienal (E11,E13-16:Ald). Double bond positions of the two monoenyl components were determined by GC–MS analysis of the pheromone extract treated with dimethyl disulfide. On the other hand, GC–MS analysis of the female body extract detected the unsaturated hydrocarbon (3Z,6Z,9Z)-3,6,9-tricosatriene (Z3,Z6,Z9-23:H). Field examinations of their synthetic compounds indicated the significant role of E11,E13-16:Ald as a major component and a clear synergistic effect of the two monoenyl compounds as a minor component. Although the 3:3:7 mixture of Z11-14:OAc, E11-16:Ald, and E11,E13-16:Ald captured the largest number of males among the tested mixtures, the activity was still quite a bit lower than that of virgin females. However, the 3:3:7:1 mixture, which was prepared by adding a small amount of Z3,Z6,Z9-23:H to the 3:3:7 ternary lure, succeeded in attracting males more powerfully than the females did. This strong synergistic effect was not observed when the triene was added to unmixed E11,E13-16:Ald, indicating important roles of not only the triene but also the two monoenyl compounds as natural pheromone components.

In moths, males can detect a distinct blend of several pheromone components by specialized olfactory receptor neurons (ORNs) on the antennae. Four candidate pheromone receptors (PR) with seven transmembrane domains were identified by homology cloning from the antennae of Spodoptera exigua (Sexi). Phylogenetic analyses reveal that all four odorant receptors (OR) belong to pheromone receptor subtypes. Expression patterns revealed that PRs were male-specific in the antenna except for SexiOR11, which was female antenna-biased. Functional analyses of these PRs were conducted using heterologous expression in Xenopus oocytes. SexiOR13 and SexiOR16 were all broadly activated by multiple pheromone components. SexiOR13 responded robustly to the critical pheromone component, Z9, E12-14:OAc and the minor pheromone component, Z9-14:OAc at a concentration of 10?4 M. Dose-response studies indicate that SexiOR13 was approximately 4 times more sensitive to Z9,E12-14:OAc (EC50 = 3.158 × 10?6 M) compared to Z9-14:OAc (EC50 = 1.203 × 10?5 M). While, SexiOR16 responded robustly to the secondary pheromone component Z9-14:OH with high sensitivity (EC50 = 9.690 × 10?7 M). However, similar tests of the five pheromones with SexiOR6 and SexiOR11 failed to elicit any response. These results provide basic knowledge to further advance research on the molecular mechanisms of pheromone reception.

The sex pheromones produced by virgin females of three species of small ermine moths occurring sympatrically on the European spindle tree were analysed by gas chromatography and the synthetic compounds tested against male moths in the laboratory and field. Pheromone gland extracts of Yponomeuta cagnagellus were shown to contain tetradecyl acetate, (E)-11- and (Z)-11-tetradecenyl acetate, tetradecanol, (E)-11- and (Z-11-tetradecenol and hexadecyl acetate in 30/3/100/6/0/7/42) ratio. Wind tunnel bioassays and field tests showed that (E)-11-tetradecenyl acetate primary pheromone components, and that tetradecyl acetate synergised their attractiveness. The alcohols were unimportant in these tests. Analysis of the pheromone gland extracts from Y. irrorellus showed the above components in 68/56/100/9/6/8/17 ratio, and a mixture of these attracted male moths in laboratory and field. Omission of either unsaturated acetate gave unattractive mixtures, and the alcohols were also found to be important for attraction. Pheromone gland extracts from Y. plumbellus were shown to contain the seven components in 46/148/100/20/20/13/25 ratio. Field tests showed that the two monounsaturated acetates are primary pheromone components, and removal of the alcohols had no significant effect. The titre of (Z)-11-tetradecenyl acetate in Y. plumbellus pheromone gland extracts was approximately 0.5 ng per female, one tenth the titre in the other two species. It is concluded that mixtures of three or more pheromone components with specific E/Z ratio are essential for full attraction and contribute towards premating reproductive isolation of the three species. Other factors such as rate of pheromone emission, time of flight and height of flight may also contribute to reproductive isolation.

In many moths, mate-finding communication is mediated by the female sex pheromones. Since differentiation of sex pheromones is often associated with speciation, it is intriguing to know how the changes in female sex pheromone have been tracked by the pheromone recognition system of the males. A male-specific odorant receptor was found to have been conserved through the evolution of sex pheromone communication systems in the genus Ostrinia (Lepidoptera: Crambidae). In an effort to characterize pheromone receptors of O. scapulalis, which uses a mixture of (E)-11- and (Z)-11-tetradecenyl acetates as a sex pheromone, we cloned a gene (OscaOR1) encoding a male-specific odorant receptor. In addition, we cloned a gene of the Or83b family (OscaOR2). Functional assays using Xenopus oocytes co-expressing OscaOR1 and OscaOR2 have shown that OscaOR1 is, unexpectedly, a receptor of (E)-11-tetradecenol (E11-14:OH), a single pheromone component of a congener O. latipennis. Subsequent studies on O. latipennis showed that this species indeed has a gene orthologous to OscaOR1 (OlatOR1), a functional assay of which confirmed it to be a gene encoding the receptor of E11-14:OH. Furthermore, investigations of six other Ostrinia species have revealed that all of them have a gene orthologous to OscaOR1, although none of these species, except O. ovalipennis, a species most closely related to O. latipennis, uses E11-14:OH as the pheromone component. The present findings suggest that the male-specific receptor of E11-14:OH was acquired before the divergence of the genus Ostrinia, and functionally retained through the evolution of this genus.

Variation in sex pheromone composition was studied in large fruit-tree tortrix. The pheromone contained cis-11-tetradecenyl acetate and trans-11-tetradecenyl acetate as macrocomponents as well as microcomponents cis-11-tetradecenol and trans-11-tetradecenol with the prevalence of cis-11-tetradecenyl acetate and cis-11-tetradecenol. The pheromone composition depended in part on the genotype of the male parent. Inbreeding increased the proportion of trans-11-tetradecenyl acetate and trans-11-tetradecenol.

The sex pheromones of Spilonota ocellana D. & S. and Spilonota laricana Hein. (Lepidoptera: Tortricidae) were identified by chemical analysis and field trapping. Female moths of the two species produce (Z)-8-tetradecenyl acetate, (Z)-8-tetradecen-1-ol and dodecyl acetate in almost the same proportions (98:1:1 and 97:3:1). Males of both species were best attracted to a blend of 10:1 to 1:1 Z8-14Ac:Z8-14OH. This indicates that mating barriers other than sex pheromones exist between sympatric populations.

ABSTRACT. The upwind flight response of individual male lightbrown apple moths, Epiphyas postvittana (Walker), to twenty combinations of the two pheromone components, (E)-11-tetradecenyl acetate (E11-14:OAc) and (E,E)-9,11-tetradecadienyl acetate (E,E-9,11–14:OAc), was observed in a sustained-flight tunnel. At the lowest dosage of E11-14:OAc tested (3 μg), a significantly greater percentage of males flew upwind to and landed at a source emitting 10% E1,E-9, 11–14:OAc than to all other sources. As the source dosage was increased, males showed decreased specificity of upwind flight to ratios of the two components. However, at the highest dosage of E11-14:OAc tested (300 μg), the response specificity of males shifted to blends containing lower percentages of E,E-9,11–14:OAc. The addition of tetradecyl acetate to a blend of the two components did not result in any detectable increase in male response. Analyses of the pheromone glands of individual female Iightbrown apple moths showed that females produced a range of ratios of E11-14;OAc:E,E-9,11–14:OAc from 100:2.2 to 100:11.4 with a median of approximately 100:7, reasonably paralleling the peak response of males. Pre-exposure of males to a blend of the two components, followed by exposure to E11-14:OAc alone (previously reported as a 'memory' effect) resulted in no significant response to E11-14:OAc alone.

ABSTRACT In field-trapping experiments the unsaturated analogues ( E )-6-, ( E )-12-, and ( Z )-12-tetradecenyl acetate were as attractive to male Yponomeuta rorellus Latr. as the native pheromone component tetradecyl acetate. All four analogues attracted more males than virgin females did, whereas ( Z )-6-, ( E )-11-, ( Z )-10- and ( Z )-11-tetradecenyl acetate were essentially non-attractive. Addition of 1–30% of ( Z )-11-tetradecenyl acetate to the pheromone tetradecyl acetate reduced the attraction to less than 2%. Flight tunnel experiments with Y. rorellus confirmed the activity of the ( E )-6- and ( E )-12-tetradecenyl acetates and demonstrated the activity of ( E )-7-tetradecenyl acetate as well. These analogues elicited orientation behaviour, upwind flight and landing at the odour source as frequently as the native pheromone did. Single sensillum recordings from male Y. rorellus showed two types of cells in most sensilla. A large spike amplitude cell was stimulated by tetradecyl acetate and the unsaturated analogues ( E )-11-, ( E )-6- and ( E )-12-tetradecenyl acetate, and to a lower extent by the ( Z )-6-, ( Z )-11- and ( Z )-12-isomers. A cell with medium spike amplitude was stimulated by ( Z )-9-tetradecenyl and ( Z )-11-hexadecenyl acetate. Some sensilla contained a third cell firing with a small spike amplitude which was activated by ( Z )-11-tetradecenol. Thus the tetradecyl acetate receptor was stimulated not only by the behaviourally active analogues, but also by behavioural antagonists. The interaction of ( E )-11-tetra-decenyl acetate and tetradecyl acetate with the same antennal receptor cell was also demonstrated in Y.cagnagellus . Electrophysiological discrimination between behavioural attractants and antagonists and the role of behavioural antagonists in the interspecific relations between Y.rorellus and sympatric closely related species are discussed.

Four compounds, tetradecyl acetate, (Z)-9-tetradecenyl acetate, (E)-11-tetradecenyl acetate and (Z)-11-tetradecenyl acetate were identified from female sex pheromone extracts of Hungarian and Egyptian lima-bean pod borers (Etiella zinckenella Tr., Lepidoptera: Phycitidae) by gas chromatography with flame ionization (FID) and electroantennographic (EAD) detection. In EAG studies these monoun-saturated acetates gave the best responses in a series of other tetradecenyl acetates and tetradecenols. The four component blend of the identified components in similar ratios as in the pheromone extract attracted significant numbers of male lima-bean pod borers in both Hungary and Egypt. In a preliminary subtraction test best capture was achieved by the ternary mixture of the monounsaturated acetates.

Abstract. In addition to the pheromone components (Z)-5-decenyl, (Z)-7-dodecenyl and (Z)-9-tetradecenyl acetate (Z5-10:OAc, Z7-12:OAc and Z9-14:OAc), it has previously been shown that the sex pheromone gland of the turnip moth, Agrotis segetum (Lepidoptera: Noctuidae Schiff) contains 10:OAc, 12:OAc, Z5-12:OAc, Z9-12:OAc, 11–12:OAc, Z5-14:OAc, Z7-14:OAc and Z11-16:OAc. To find out whether any of these additional compounds is involved in the sex pheromone communication in A. segetum, a comprehensive electro-physiological and behavioural investigation was conducted. Single-sensillum recordings on male antennae revealed three subtypes of sensilla among the previously so-called Z5-10:OAc sensilla. One subtype was identified having one receptor neurone (A) that responded to Z5-10:OAc with a large spike amplitude and another neurone (B) that responded to (Z)-5-decenol (Z5-10:OH) with a small spike amplitude. In another subtype the B neurone responded to Z5-12:OAc and sometimes also to 27-12:OAc and 10:OAc, in addition to responding to Z5-10:OH. In a third subtype the A neurone responded to all acetates identified from the female pheromone gland, whereas the small spike amplitude neurone was tuned to Z5-10:OH. A flight tunnel assay showed that blends composed of nine, eight or seven compounds were equivalent to the previously identified three-component pheromone blend in eliciting male behavioural responses. In field trapping tests, blends of eleven, nine or seven compounds did, however, catch significantly more moths than the three-component blend. Further assays showed that only 25- 12:OAc could significantly increase the catch numbers when added to the three-component blend, and thus qualified as a fourth pheromone component in A. segerum. The behavioural significance of additional female-produced acetates — for which males possess antennal receptors — is suggested, but may be impossible to confirm because of ‘diminishing returns’ when trying to refine a multicomponent pheromone further.

The upwind flight and landing responses of male Mamestra suasa (Den. & Schiff.) (Lepidoptera; Noctuidae) to various synthetic chemicals were studied in a wind tunnel. A mixture of (Z)-11-hexadecenyl acetate (Z11-16: Ac) and (Z)-11-hexadecenal (Z11-16: Ald) elicited upwind flight responses past the midpoint of the wind tunnel, and both components were necessary for landing of the males to occur at the pheromone source point. The best response to the mixtures tested was to a ratio 100:1 (ng) of Z11-16: Ac and Z11-16: Ald (blend B).The addition of (Z)-9-tetradecenyl acetate (Z9-14: Ac) (10 ng) to blend B resulted in a significant decrease of upwind flight and landing responses. The inhibitory effect of this chemical when it is mixed with blend B, and the presence of a specific receptor for this compound in sensilla trichodea of male M. suasa, suggest that Z9-14: Ac is involved in the chemical communication between M. suasa and other sympatric species. The addition of (Z)-11-hexadecenol (Z11-16: OH) (1 or 10 ng) to blend B had no effect on male upwind flight and landing responses. This compound does not seem to be involved in the chemical communication of this species.

Abstract

The sex pheromones of two endemic New Zealand tortricid species were investigated. Females of the first species, Cnephasia jactatana, a pest of kiwifruit, were found to produce (Z)-11-tetradecenyl acetate (Z11-14:OAc) in their sex pheromone gland. When tested in the field, this compound on its own elicited significant catches of male C. jactatana. However, the addition of even very small amounts (0.3%) of the geometric isomer, (E)-11-tetradecenyl acetate (E11-14:OAc), to Z 11-14:OAc virtually suppressed trap catches. The sex pheromone of the second species, Merophyas leucaniana, was found to consist of a mixture of Z11-14:OAc, E11-14:OAc, tetradecyl acetate, and (Z)-11-tetradecenol. This species is morphologically very similar to the congeneric species, M. divulsana, the Australian lucerne leafroller. The sex pheromone of M. leucaniana is also very similar to that of the so-called “Z-type” of M. divulsana, which is now found in New Zealand. The pheromone components of C. jactatana and M. leucaniana are discussed in relation to those found in other tortricids.

The electrophysiological and behavioural responses of male Sesamia nonagrioides Lefèbvre (Lepidoptera: Noctuidae) to several synthetic blends of its sex pheromone components were investigated. Electroantennogram (EAG) recordings showed the greatest response to a 77:8:10:5 blend of the four compounds found in the female glands: (Z)-11-hexadecen-1-yl acetate, (Z)-11-hexadecen-1-ol, (Z)-11-hexadecenal and dodecyl acetate. A logistic equation was fitted to the dose-response data and differences in relative EAG responses were analysed by deriving five parameters of this equation. Flight responses of males were also evaluated in a wind tunnel where the proposed blend (77:8:10:5) also elicited the best response. Field tests confirmed the laboratory findings as the 77:8:10:5 blend captured more S. nonagrioides males than other pheromone mixtures and showed more selectivity for male S. nonagrioides over other noctuid species, Discestra trifolii Rottenberg and Lacanobia oleracea L., but not for Mythimna unipuncta Haworth, a species with a sex pheromone composition almost identical to that of S. nonagrioides.
