The ovipositor of Rhagoletis pomonella (Walsh) (Diptera : Tephritidae) has a dorsal process which terminates distally in a slightly upcurved tip and 2 ventral processes that are joined throughout their length by a membrane up to the region of the cloaca, where the ventral processes are not attached. The surface of the ovipositor contains both hair-like and campaniform mechanosensilla capable of monitoring both cuticular stress and mechanical contact of the ovipositor from the initial through the final steps of penetration of the fruit, egg-laying, copulation, and possibly dragging the ovipositor during deposition of the oviposition deterrent pheromone. Near the tip of the ovipositor are 2 longitudinally aligned grooves, each containing 3 pairs of chemosensilla. In addition, 1 pair of shorter chemosensilla are just distal to and outside the groove. All chemosensilla contain 3 or 4 chemosensitive neurons. During fruit penetration and egg-laying, the ovipositor is protracted, thus placing the chemosensilla in contact with various fruit chemicals. These chemosensilla are responsive to naturally occurring fruit chemicals. The possible role of the ovipositor chemosensilla in monitoring fruit quality, the deposition of the oviposition deterrent pheromone, the presence of chemical contaminants on the ovipositor, and the female's own accessory reproductive gland secretions remains to be established.  相似文献

The chemosensilla on the tarsi of Locusta migratoria were mapped using light microscopy, as well as scanning and transmission electron microscopy. Only chemosensilla chaetica were found on the tarsi. On the basis of their ultrastructure, these can be grouped into three main subtypes: short, long, and sunken sensilla chaetica. Short sensilla chaetica can be further divided into two groups containing 6 or 7 neurons. Long sensilla chaetica are mainly located on the lateral surface of the tarsi. Short sensilla chaetica were mainly found on the dorsal surface of the tarsi. Sunken sensilla chaetica were only found on the ventral surface, such as the pulvilli and arolium. Immunocytochemical localization of chemosensory protein (CSP) was performed on ultrathin sections of chemosensilla on tarsi. The antiserum against LmigCSP‐II intensively labeled all three types of sensilla chaetica. Gold granules were concentrated in the outer sensillum lymph surrounding the dendrite sheath, while the inner sensillum lymph containing dendrite branches was never labeled. Helicoverpa assulta and Helicoverpa armigera are sibling species with different host-plant ranges. We have previously reported electrophysiological and behavioral responses of H.armigera to sugars and amino acids. Here we describe a parallel study performed on H. assulta and compare the results obtained with the two species. In females, fourteen gustatory chemosensilla, identified on one ventrolateral side of the fifth tarsomere were stimulated with sucrose, glucose, fructose, maltose, myo-inositol, and the twenty common amino acids, using the tip-recording technique. The taste receptor neurons in eight chemosensilla were identified sensitive to the sugars, myo-inositol, Lys, Glu, Arg, Trp, and Ser which all induced proboscis extension reflex (PER) when tarsi were stimulated. There was a positive correlation between electrophysiological activities and PER responses triggered by sucrose. No stimulatory effect on oviposition was observed with sugar or amino acid mixtures. In males, three chemosensilla showed responses to the four sugars, but generally weaker than in females. The major difference of the two species was the variety of amino acids triggering electrophysiological responses. The stimulatory effect of sugars and amino acids on H.assulta was also generally weaker than that on H. armigera.  相似文献

Highly developed chemoreception allows insects to detect foods, find mates, and escape natural enemies. We described the structures and distributions of antennal chemosensilla in Adelphocoris fasciaticollis Reuter by scanning electron microscopy and transmission electron microscopy. Seven major types of antennal sensilla were identified in adults of both sexes. Types 1 and 2 are sensilla chaetica and have thick cuticular walls with conspicuous grooves at their surfaces. Types 3 and 4 are multiporous sensilla trichodea with 1–3 dendrites located at the sensillum lymph, indicating a putative olfactory function. Types 5 and 6 are typical sensilla basiconica but share different characteristics in both external morphology and internal ultrastructure, and may be involved in the perception of host-associated odorants. The last sensilla were Böhm bristles. In addition to the morphological characterization, electrophysiological responses of antennal chemosensilla to 51 semiochemicals were investigated based on electroantennogram (EAG) recordings. Results revealed that different chemical stimuli elicited significantly different dose-dependent EAG responses, in which potential sex pheromone components and green leaf volatiles showed relatively higher EAG responses, but neither monoterpenes nor sesquiterpenes can elicit favorable EAG values. The results provided direct morphological and electrophysiological evidence that the adult antennae of A. fasciaticollis could function in searching for mates and host plants.  相似文献

To allow an animal to behave appropriately, the location of sensorial structures is expected to be related to their function. As the different leg pairs of arachnids may have different functions (probing x supporting the body), one could expect them to have a different density of sensilla. Moreover, different regions of the same leg (dorsal, lateral, and ventral) would also be expected to have different densities of sensilla, according to the use of each region (e.g., the ventral part is often in contact with the substrate while the dorsal part is not). As cavernicolous animals are expected to be more sensitive than their epigean relatives, one could also expect a different density of sensilla when comparing cavernicolous and epigean animals. Using three epigean and three cavernicolous species of harvestmen (Arachnida, Opiliones), this study aimed at describing the morphology of hair sensilla on the legs and answering three questions: (1) Are there differences in the density of hair sensilla between the dorsal, lateral and ventral regions of each leg pair of the same individual? (2) Are there differences in the density of hair sensilla between the leg pairs of the same individual? (3) Are there differences in the density of hair sensilla when comparing the leg pairs of individuals of cavernicolous and non-cavernicolous species? The tarsi and metatarsi of all right legs of the six studied species were analyzed under a scanning electron microscope. The results (P < 0.05) showed that, in general: the ventral region of the tarsus was denser in sensilla trichodea than the lateral and dorsal regions, particularly on legs I and II; the density of sensilla chaetica did not differ on legs III and IV, but was greater on the dorsal region of legs I and II; the ventral part of legs I had the higher density of sensilla trichodea of the four pairs, whereas the second pair had the lower density; Holcobunus citrinus (Eupnoi) was the species with higher density of sensilla trichodea, on all legs; the cavernicolous species had a lower density of sensilla than the epigean species. The results are tentatively related to harvestmen behavior.  相似文献

Karyotypes of 24 diploid (2n=12)Tragopogon species are similar with one long pair of chromosomes (A), two medium-length pairs (B and C), and three short pairs (D, E, and F). These species may be divided into three karyotypic groups: 1) seven species with a satellite on A and on D; 2) 14 species with a satellite on A only; and 3) three species with a satellite on D only. Most species within karyotype groups may be separated from each other either by distinctive features of certain chromosomes or by statistical differences in length of chromosome arms or long arm: short arm ratios of chromosome A. Three tetraploid (2n=24) species had two long pairs (A,A′), four medium-length pairs (B,B′;C,C′), and six short pairs (D,D′;E,E′;F,F′). Suggestions are made as to the putative diploid parents of these presumed allotetraploids.  相似文献

It is well established that taste chemoreceptors in Drosophila are located on the tarsi of the first leg pair. In order to investigate the influence of the novel homeotic arista-tarsus transformation on behavior, an analysis of taste perception in the lawc-mutants, characterized by the transformation of the arista into tarsus elements, was carried out. The data were subjected to thorough statistical treatment. It was shown that elements of an additional leg that appeared as a result of homeotic transformation of the arista were sensitive to gustatory stimuli. Analysis of the innervation of the homeotic organs by means of cobalt staining of afferent projections showed that the afferents starting from the homeotic leg reached the thoracic ganglion.  相似文献

Evolution of the multidomain protein wheat germ agglutinin   总被引:1,自引:0,他引:1  
We compared the homologous amino acid sequences of hevein and each of the four domains (A, B, C, and D) of wheat germ agglutinin and used them to construct a pseudophylogenetic tree relating these sequences to a hypothetical common ancestor sequence. In the crystal structure of the wheat germ agglutinin dimer, six pseudo-two-fold rotational symmetry axes have previously been located in addition to the true twofold axis. Four of these relate two nonidentical domains to each other in each of the four possible pairs constituting the sugar-binding sites (A1D2, A2D1, B1C2, and B2C1). The remaining two relate contiguous unique pairs of sugar-binding sites to each other (A1D2 to B1C2, and A2D1 to B2C1). These latter two sets of pairs are related to each other by the true twofold axis. Side chains that mediate sugar binding in the interfaces of each of the four pairs were found to be largely conserved. The sequence homology, taken together with these pseudo-symmetry elements in the dimer structure, suggests a pathway for the evolution of the four-domain molecule from a single-domain dimer that can be correlated with simultaneous development of the saccharide-binding sites.  相似文献

The landing response of stationary flying houseflies Musca domestica has been recorded on video tape. The leg movements were quantitatively evaluated. It could be demonstrated that:
  1. only the first two pairs of legs are involved in the reaction (Fig. 1). Prothoracic tarsi are lifted beyond the head, mesothoracic tarsi are lowered and moved sidewards (Fig. 2a and b).
  2. the movement of the tarsal tips is mainly due to an opening of one single joint per leg, i.e. the femurtibia joint of the prothoracic leg (Fig. 2c), and the coxa-femur joint of the mesothoracic leg.
  3. the landing reaction is a fixed action pattern which does not seem to require further sensory input once it is released (Fig. 4d).
  4. the landing responses to a light-off stimulus and to expanding patterns with different angular velocities are indistinguishable (compare Fig. 3a-c with Fig. 2a-c). The only parameter that is obviously dependent on the stimulus conditions, is the latency of the reaction (Fig. 4a-c).

The foreleg claw sensilla of male D. variabilis, D. andersoni and A. americanum ticks indlude the receptors that perceive the female contact mounting sex pheromone (MSP). In all three tick species, the foreleg claw sensilla comprise six anteriorly-directed setae arranged in three symmetrical pairs, two each on the opposite sides of the apotele of the claw and one on the ventral side. Morphological study and behavioral bioassays of these setae revealed that only the dorsal and middle (=lateral) pairs of claw sensilla are mechanogustatory. While the ventral pair are strictly mechanoreceptors. The dorsal and middle sensory setae exhibit a single pore-like structure located at or near their tip, a feature characteristic of mechanogustatory sensilla. These setae are similar to those found on the palps that are believed to function as pheromone receptors. In all three tick species, male mounting and postmounting behaviors were suppressed only when the dorsal and middle pairs of claw sensilla were ablated or covered with gelatin; normal behavior was restored when the gelatin was removed. Doscresponse bioassays were conducted with D. variabilis males to authenticate the results of the gelatin tests. The results of these bioassays demonstrated that the gelatin coat was impervious to the pheromone. The characteristics of the ixodid tick mating system that distinguish it from mating processes in other arthropods are discussed.  相似文献

A complicated multicellular gland is situated in all the leg tarsi, occupying from one third to half the segment. The glandular cells form a single-layer sack; the inner surface of the gland cavity is covered with the multi-layer membrane. Cuticular rods (“sinews”) of muscles moving the claw pass inside the gland cavity. The glandular cells are characterized by the presence of numerous microvilli on their apical surfaces and by the presence of secretory vacuoles. The basal part of each secretory cell is characterized by accumulations of lipid vacuoles and glycogen granules. Problems concerning the possible role of tarsal gland in the production of the trace pheromone are discussed.  相似文献

Summary The responses of contact chemosensory sensilla on the tarsi of the apple maggot fly,Rhagoletis pomonella (Walsh), to a range of concentrations of salt, sucrose and oviposition-deterrent pheromone were studied (Fig. 1). Examination of dose-response curves (Fig. 2), short- and long-term adaptation (Figs. 3+4, Table 1) and impulse height and duration (Fig. 5, Table 2) indicated that each stimulant elicited a characteristic response. Stimulation with mixtures of the solutions demonstrated that salt, sucrose and the pheromone each excites a different sensory cell (Fig. 6). A cell which does not respond to a stimulant with impulses may, nevertheless, be affected by that stimulant (Fig. 7).  相似文献

Leg movements of stick insects (Carausius morosus) making turns towards visual targets are examined in detail, and a dynamic model of this behaviour is proposed. Initial results suggest that front legs shape most of the body trajectory, while the middle and hind legs just follow external forces (Rosano H, Webb B, in The control of turning in real and simulated stick insects, vol. 4095, pp 145–156, 2006). However, some limitations of this explanation and dissimilarities in the turning behaviour of the insect and the model were found. A second set of behavioural experiments was made by blocking front tarsi to further investigate the active role of the other legs for the control of turning. The results indicate that it is necessary to have different roles for each pair of legs to replicate insect behaviour. We demonstrate that the rear legs actively rotate the body while the middle legs move sideways tangentially to the hind inner leg. Furthermore, we show that on average the middle inner and hind outer leg contribute to turning while the middle outer leg and hind inner leg oppose body rotation. These behavioural results are incorporated into a 3D dynamic robot simulation. We show that the simulation can now replicate more precisely the turns made by the stick insect. The epoxydienyl sex pheromone of Ascotis selenaria cretacea can be detected only within a rod-like abdominal tip (RAT) of the female. To clarify which part of the RAT is the sex pheromone-producing region, the RAT was morphologically divided into three sections, defined positionally from the abdomen as sections A, B, and C. GC-MS measurements clearly showed that the sex pheromone compound levels in section B were four times greater than those of the other sections. Microscopic dissection analysis revealed that section B consists of four tissues: rectum, oviduct, musculature, and intersegmental membrane. GC-MS analysis of the individual tissues revealed that approximately 90% of the sex pheromone in section B is localized in the intersegmental membrane. A cell layer was found in the intersegmental membrane after staining with propidium iodide. Furthermore, incubation of tissues dissected from section B with a deuterated trienyl pheromone precursor revealed that the labeled epoxy pheromonal component was detected exclusively in the intersegmental membrane. We have determined that the sex pheromone-producing region of A. s. cretacea is on the terminal side of the intersegmental membrane located between the 8th and 9th abdominal segments.  相似文献

To ascertain the molecular mechanism that causes murine C5 deficiency, genomic and cDNA libraries were constructed from mouse liver DNA and mRNA employing the congenic strains B10.D2/nSnJ and B10.D2/oSnJ that are sufficient and deficient for C5, respectively. Genomic fragments were isolated which correspond to PvuII and HindIII restriction fragment length polymorphisms associated with C5 deficiency. Sequence analyses demonstrated that each of these polymorphisms resulted from single base pair substitutions and that neither substitution would probably cause or contribute to the C5 deficiency. Sequence analyses of C5 sufficient and deficient cDNAs revealed a 2 base-pair deletion in the deficient cDNAs. The "TA" deletion was located near the 5' end of the cDNA. This deletion shifts the reading frame of the C5 mRNA so that the termination codon UGA is present 4 base pairs downstream from the deletion. Genomic DNA was amplified and sequenced corresponding to the area surrounding the 2-base pair deletion. Six C5-deficient strains, A/HeJ, AKR/J, DBA/2J, NZB/B1NJ, SWR/J, and B10.D2/oSnJ, and four C5-sufficient strains, Balb/CJ, C57Bl/6J, DBA/1J, and B10.D2/nSnJ, were analyzed. The sequencing data revealed that the 2 base pairs were deleted from the C5 gene of each deficient mouse tested but not from the C5 gene of any sufficient mouse. These data demonstrate that: 1) there is an identical 2-base pair deletion in an exon of the C5 gene in several different C5-deficient mouse strains; 2) the mRNA transcribed from the C5D gene retains this deletion; and 3) this mutation should result in C5 protein deficiency.  相似文献

Data on 155 lambs computer tomography (CT) scanned at a mean age of 118 days and 41.5 kg were included in this study. The object was to examine in vivo composition in three major carcass regions, the leg, the mid-region and the shoulder of lambs of two genetic lines, a meat line (ML) and a control line (CL). Additionally, the study aimed to determine the best scan positions for estimating tissue weights in each region. Images were taken at 40 mm intervals, on average 23 scans per animal. Each CT scan was located to an anatomical position and to one of the three regions. Weight of lean, fat and bone was found for each region from all images located to a region. Tissue weights, and proportion of lean, fat and bone relative to the total weight of the region, was found by GLM adjusted for fixed effects and live weight (covariate). Stepwise regression procedure was used to find the best combination of scan sites for estimation of composition in each region.The ML had a significantly heavier weight and larger proportion of lean, and a lower proportion of bone in all regions compared to the CL. Furthermore, the ML had a lower proportion of fat in the leg and the mid-region than the CL (P < 0.05). Moreover, the ML had a larger weight of lean in the leg than in the shoulder compared to CL (P < 0.05). The latter indicated that selection for lean and introduction of Texel into the ML had increased the development of lean in the leg compared to the shoulder. Both lines had largest percentage lean (78–80%) in the leg and lowest in the shoulder (70–72%), whereas smallest percentage fat was found in the leg.A minimum of two scan sites should be recorded for estimations of the shoulder and the mid-region, and three for the leg. The best combination of scan positions for the shoulder was the 6th thoracic and the 7th cervical vertebra, for the mid-region the 4th lumbar and the 8th thoracic vertebra, and for the leg the 3rd and 4th caudal and the 4th sacral vertebra. Using these scan sites for estimation of tissue weights in each region, the R2 ranged from 0.894 to 0.978 for lean and fat and from 0.689 to 0.833 for bone.  相似文献

Mutants at 2 new loci which control mutagen-sensitivity are described. Mutants at both loci are female-sterile and are hypersensitive to killing by MMS; neither increases the frequency of sex-linked recessive lethals. A screen of previously described female-sterile and meotic mutants has revealed that a number of these are also sensitive to mutagens. In addition, several new mutants have been identified on the basis of sensitivity to either HN2 or MMS. An anlysis of complementation data suggests that all of the X-linked genes controlling sensitivity to MMS may now have been identified. Among the new mei-41 alleles are mutants which show verly little meiotic nondisjunction or loss. Cytogenetic mapping of previously known mutants is also described. The mutants mus(1)104D1 and mei-41D5 are located in th eregion 14B13±?14D1,2 on the polytene chromosome map, and they map very close to each other genetically. Cytogenetically mus(1)101D1 is between salivary chromosome bands 12A6,7 and 12D3, mus(1)103D1 is between bands 12A1,2 and 12A6,7, and mus(1)-109A1 is in section 8F3-9A2.  相似文献

Electrophysiological recordings of tarsal and labellar contact chemosensilla in Phormia regina females demonstrate activation of sugar-, cation-, and possibly anion-sensitive cells in response to 10% yeast extract. Yeast extract is a proteinaceous food source that supports full egg development. Though some protein is required for egg maturation in Phormia, a blowfly not normally autogenous, purified proteins alone neither activate the chemosensilla nor promote egg development. In experiments reported here, groups of females were given ad lib access to either 0.2 M sucrose or 0.2 M sucrose and 10% yeast for four days after eclosion. The sucrose diet induced a ‘protein’ preference but no egg development. Potassium and sodium salts were added to 0.2 M sucrose and presented to the flies on day six. The volumes of salt-sucrose mixtures taken were greater than those of 0.2 M sucrose for flies raised on 0.2 M sucrose, though yeast was preferred over all but one salt solution. For sucrose-and-yeast-fed flies, volumes of 0.05 and 0.1 M salt mixtures consumed did not differ from 0.2 M sucrose; however, sucrose alone was favoured over more concentrated salt mixtures. All salt-sucrose mixtures tested were significantly more effective stimuli for sucrose-and-yeast-fed flies than yeast. It is suggested that varying sensitivity in the salt-sensitive contact chemosensilla plays a role in reported food preference shifts associated with reproduction.  相似文献

The hypothesis that female sonic stimulus may evoke male pheromone release in a behavioural interaction analogous to the known male sonic stimulus of female pheromone release, was confirmed in Dendroctonus pseudotsugae, and also in D. brevicomis. In both species known male-produced substances collected from males stimulated by recorded female stridulation were identified by coupled gas chromatography/mass spectrometry. In a second test with D. pseudotsugae, male pheromone release during recorded female stridulation was evident in the change of male stridulation from the simple attractant chirp to the interrupted chirp, which is known to result from a medium concentration of 3,2-MCH. Also, the D. pseudotsugae male attractant chirp was synthesised with an electronic pulse generator and used to evoke pheromone release. It is concluded that the antiaggregative pheromone of this species is released by each sex at the sonic stimulus of the other sex.  相似文献
