The ability of 135 Staphylococcus strains isolated from Spanish dry-cured hams to produce enterotoxins in culture was investigated by the reversed passive latex agglutination method. A high percentage of enterotoxigenic Staphylococcus aureus strains (85.9%) was recorded, and 54.3% of these produced enterotoxin A. One of the two Staphylococcus epidermidis strains produced enterotoxin C. The reversed passive latex agglutination method yielded satisfactory results.  相似文献

Staphylococcus aureus strains were isolated from end-of-lay poultry carcases obtained from a plant at two different stages of processing before and after storage at different temperatures. These strains were supplemented with Staph. aureus strains isolated from poultry from a wide range of sources and biotyped, phage typed, and tested for production of enterotoxins A-E. The isolates were found to consist of poultry and human specific strains and each of these groups contained strains able to produce enterotoxin. Poultry strains produced only enterotoxin D whereas human strains produced enterotoxins A, C and D. The hen carcases used in storage experiments were found to be naturally contaminated with enterotoxin D producing staphylococci. No enterotoxin D could be detected on any of the carcases even after storage at temperatures which allowed multiplication of the organisms to occur (final Staph. aureus counts ranged from 102 to 107/16 cm2 of breast skin).  相似文献

The microbial flora of salt used in the production of Spanish dry-cured ham was studied. The results indicated that Micrococcaceae constituted the predominant flora. Identification of the 369 isolates belonging to the family Micrococcaceae revealed that 60% belonged to genus Staphylococcus, 25% to Micrococcus, 6% to Kocuria, 5.4% to Dermacoccus and 0.5% to Stomatococcus. The species most often isolated was Staph. xylosus (28.9%), followed by M. lylae (21.4%), Staph. equorum (18.55%) and D. nishinomiyaensis (5.4%). The results indicate that the salt during salting process of dry-cured hams offers an ecosystem suitable for the survival of the staphylococci and micrococci.  相似文献

The study of the enterotoxigenicity of S. typhimurium with the use of the skin test on rabbits (to detect the delayed permeability factor) has revealed that these strains produce an enterotoxin similar to Escherichia coli thermolabile enterotoxin (TLE). Study of the enterotoxic activity of lysates obtained from 39 S. typhimurium strains and 5 S. dublin strains by sonication has revealed that 87% of S. typhimurium strains and all S. dublin strains produce an enterotoxin similar to E. coli TLE, as demonstrated by all tests used in this investigation, while 59% of S. typhimurium cultures and all S. dublin strains have been positive when tested for the capacity of producing the rapid permeability factor. "Hospital" strains and polyresistant cultures isolated from the environment (phagovar 20) are characterized by a higher rate of producing an enterotoxin similar to E. coli TLE, detected by the tests used in this investigation (90%), than antibiotic-sensitive strains of different origin (78%).  相似文献

The paper presents results on the relatedness of Staphylococcus aureus strains colonizing the upper respiratory tract isolated from healthy persistent carriers. Genotyping was carried out using two methods—multiple-locus variable-number tandem-repeat fingerprinting (MLVF) and pulsed-field gel electrophoresis (PFGE). By comparison of the results obtained by both methods, good correlations between MLVF and PFGE genotyping of strains isolated from the asymptomatic carriers were observed. Further studies are needed to evaluate methods useful for genotyping of S. aureus strains circulating in the community.  相似文献

A total of 126 fungal strains belonging to the Aspergillus flavus group isolated from commercial poultry mixed feeds were studied. One hundred and twenty-five were identified as A. flavus and one as A. parasiticus. Forty nine strains (39%) produced aflatoxins on a crushed moist wheat medium (28 °C/10 days), whereas only sixteen (13%) showed specific fluorescence on Aflatoxin-Producing Ability Medium. In both media, mainly aflatoxins B1 and B2 were detected, the average concentration of aflatoxins being 4294+/–1083 g/kg in crushed moist wheat medium, and 877+/–257 g/kg in Aflatoxin-Producing Ability Medium.  相似文献

The study was carried out of 22 isolates of S. aureus isolated from 7 different incriminated food's samples from foodborne-disease outbreaks. The possibility of these isolates to producing of enterotoxins by commercial test SET-RPLA (Oxoid) was tested. The genotyping of these isolates was done by pulse-filed gel electrophoresis, acc. to Pfaller in own modification. On the basis of the DNA restriction patterns of the 22 isolates--5 strains were singled out, one of these strains--strain V (isolat nr 7) was not relationship to others. It was found that this strain V was one enterotoxin produced. Additionaly, all tested strains, in spite of the strain nr V, were isolated from the 2 or 3 samples of different kinds of foods. In the present study it has been shown too, that several similar colonies should be isolated for farther studies to assess microbiological contamination of the food products properly.  相似文献

An optimal sensitivity plate method for examining large number of staphylococcal strains for production of the known enterotoxins (A-E) is presented. Small volumes of relatively concentrated enterotoxin are produced by the semi-solid agar, cellophane-over-agar, or sac culture techniques. Detection of the enterotoxin in the supernatant fluid is accomplished with the optimal sensitivity plate method. In this method small plastic petri dishes (50 mm) were used for a modified Ouchterlony of high sensitivity.  相似文献

About half (49%) of strains of Staphylococcus aureus isolated from poultry were non-typable with the international human set of phages, and 55% were biotype B according to the biochemical identification scheme of Hájek & Maršálek (1971, 1973). A furthest neighbour clustering strategy and principal coordinate analysis based on 17 biochemical tests made clear distinctions between biotype B strains and a group of biotype A and intermediate strains. Overall 62% of strains were enterotoxigenic, the majority producing enterotoxin A. Significantly fewer intermediate strains than biotype A or B strains were enterotoxigenic. Starch gel zymograms of intracellular esterases showed a general correlation with the biotyping and phage typing results.  相似文献

The aim of study was the molecular characteristic of S. aureus and S. epidermidis isolates obtained from skin surface, wounds, deep tissues of hospitalized patients and from skin surface of non-hospitalized patients. Genes encoding virulence factors were examined using PCR reaction and specific primers. Genes encoding adhesinsfnbA and cna and gene eta for epidermolytic toxin were mostly present in S. aureus isolates coming from wounds and deep tissues compared to these from skin surface. Gene atlE encoding autolysin of S. epidermidis was detected in all studied isolates, whereas gene icaAB was present in almost all isolates. Comparison of results obtained by PCR and conventional method of the resistance to methicillin estimation showed discrepances suggesting the need for using of both methods in some clinically difficult cases of S. aureus infection.  相似文献

To determine whether staphylococci causing bovine mastitis are potential causes of human intoxications, 142 cultures identified as etiological agents of acute cases and 18 cultures causing chronic cases of staphylococcal mastitis were obtained from investigators in the United States and Canada, examined microscopically, and tested for carbohydrate utilization, terminal pH, catalase, coagulase, egg yolk hydrolysis, gelatin hydrolysis, cytochrome oxidase, urease production, nitrate reduction, micrococcal nuclease, phage type, and enterotoxin production. Three cultures were not confirmed as Staphylococcus aureus. Of the 157 S. aureus cultures, 23 produced staphylococcal enterotoxins. Although a direct relationship between staphylococcal mastitis and outbreaks of staphylococcal food poisoning was not proved, results indicated that staphylococcal infections of the bovine mammary gland represent a significant reservoir of enterotoxigenic strains of S. aureus.  相似文献

2545 strains of Staphylococcus aureus isolated from 24 Polish laboratories in the years 1994-95 were investigated. Phage typing was performed according to the method of Blair and Williams using the present basic set of typing phages and additional phages 88, 89 and 187. The phages were employed in concentrations of RTD and 100 x RTD. The predominance of phage group II, reported elsewhere since 1980-ties, was found in the present study (23.6%). Strains of group III were second in frequency (16.6%), whereas strains of groups I and V, as well as type 95 occurred in small percentage (7.6%, 4% and 3.4% respectively). Strains of groups II and V have been rarely lysed by phages belonging to other groups. The use of additional phages resulted in typability of strains by 7.9%. Percentage of non-typable strains was high and amounted to 22.0%.  相似文献

A surface antigen consisting of aminoglucuronic acid and N-acetyl-L-alanine was isolated from the culture filtrates of two human strains of Staphylococcus aureus. Double diffusion analysis in agar suggested that the antigen is immunologically similar to the alanyl-aminoglucuronic acid capsule of the Smith strain of S. aureus. Quantitative precipitin inhibition studies indicated that N-acetyl-L-alanine is the immunodominant determinant of the acidic antigen. In addition, conjugates consisting of N-acetyl-L-alanine coupled to bovine serum albumin gave a significant precipitin reaction with anti-staphylococcal serum which is rich in alanyl-aminoglucuronic acid polymer antibodies. Antibodies with N-acetyl-L-alanine specificity were isolated from N-acetyl-L-alanine-Sepharose immunoabsorbent columns. Double diffusion analysis in agar indicated that the eluted antibodies were serologically reactive and belonged to the IgG class of immunoglobulins.  相似文献

The VISA (vancomycin intermediate Staphylococcus aureus) and hetero-VISA strains were found among all isolates of S. aureus obtained in the years 1997-2000. The frequency of VISA was 0.3% and h-VISA was about 3%. Most but not all of the h-VISA and all VISA strains were methicillin resistant. Moreover the usefulness of different methods enabling recognition of the h-VISA strains was compared.  相似文献

Strains of Staphylococcus aureus which colonize defeathering machinery and become endemic within poultry processing plants appear to be resistant to the normal cleaning and disinfection processes. The resistance of endemic strains to chlorine was compared with that of isolates from the natural skin flora of poultry. Endemic strains were almost eight times more resistant and this was due primarily to their ability to grow in macroclumps but also to the production of an extracellular slime layer.  相似文献
