Seasonal changes in the physiology of Antechinus stuartii result in complete male mortality after mating. The most important endocrine changes in males are large rises in plasma testosterone and cortisol concentrations. Glomerular filtration rate (GFR) in males declines coincident with high plasma testosterone and cortisol. In the present study GFRs were measured in males captured in May (when endogenous plasma testosterone and cortisol levels are low) and given depot injections of either saline, testosterone-only, cortisol-only or testosterone plus cortisol at doses designed to mimic plasma levels during the mating period. GFR decreased significantly with testosterone injection, independent of cortisol treatment. Urinary concentrations of sodium and chloride, and osmolality decreased significantly with cortisol treatment, although the addition of testosterone reversed the effect. Total urinary excretion of electrolytes was similar between groups. Plasma potassium levels significantly increased in testosterone plus cortisol treated males. Plasma sodium levels significantly increased and plasma chloride significantly decreased in all groups treated with cortisol. Water consumption significantly increased in all cortisol-treated males and food consumption significantly increased in all testosterone-treated males. The seasonal renal functional changes observed in A. stuartii were mimicked by testosterone administration. Accepted: 23 January 1998  相似文献

The detailed reproductive patterns and their associated endocrine characteristics have been documented only for a few species of bats. The objective of this study was to examine seasonal changes in plasma concentrations of leptin and compare it with the changes in body mass, circulating concentrations of testosterone, androstenedione and its correlation with prolonged survival of sperm during winter dormancy in the male sheath-tailed batTaphozous longimanus Hardwicke, 1825. Six bats were captured every month for three consecutive years during 2002 to 2005 from Varanasi, a subtropical part of India. The changes in the body mass were positively correlated with circulating concentration of leptin. Leptin concentration reached a peak (14 ng/ml) in November coinciding with peak body mass. Leptin levels declined during other months of the year except for a rise in March and August. Plasma leptin was positively correlated with androstenedione concentration, but did not show significant correlation with testosterone level. We noticed a significant increase in testosterone secretionin vitro in response to leutinizing hormone (LH) stimulation. However, we did not notice any increase in testosterone or androstenedione secretionin vitro in response to leptin stimulation. Plasma leptin concentration did not show any correlation with testis mass in this study. The higher concentration of testosterone and androstenedione may be responsible for the prolonged survival of sperm in the epididymidies and higher levels of leptin in November may be responsible for maintaining reproductive function during winter dormancy. We suggest that inT. longimanus, higher leptin concentrations in November may be responsible for the gonadal recrudescence and reproductive response during winter dormancy is modified by energy availability and by changing leptin concentrations during this period.  相似文献

Testosterone is important in mediating investment in competing activities such as territoriality, parental care, and maintenance behavior. Most studies of testosterone function have focused on temperate species and less is known about the role of testosterone in territoriality or variation in mating systems of tropical species. Results of studies of tropical species with year‐round territoriality indicate that territorial aggression during the non‐breeding season is maintained with low levels of testosterone, and increased levels of testosterone in males during the breeding season may increase mating opportunities or aid in competition for mates. We studied seasonal variation in testosterone levels of male Red‐throated Ant‐tanagers (Habia fuscicauda), a socially monogamous species with year‐round territoriality and with high levels of extra‐pair matings (41% of young), to determine if testosterone levels increased during the breeding season. We captured males during the non‐breeding and breeding seasons and collected blood samples for hormone analysis. We found that mean testosterone concentrations were low during the non‐breeding season (0.18 ± 0.05 [SD] ng/ml, range = 0.11–0.31 ng/ml), and significantly higher during the breeding season (2.37 ± 2.47 ng/ml, range = 0.14–6.28 ng/ml). Testosterone levels of breeding males were not related to aggression levels as measured by attack rates toward a stuffed decoy or singing rates during simulated territorial intrusions. These results suggest that the higher testosterone levels of breeding male Red‐throated Ant‐tanagers may be important in an extra‐pair mating context, possibly in display behavior or mate attraction, but additional study is needed to clarify the role of testosterone during the breeding season.  相似文献

Dominance hierarchies play an important role in access to mates or resources in many species. Rank is sometimes correlated with circulating testosterone levels or morphological traits such as body weight. The relationship of glucocorticoid secretion and rank, however, is less clear. In this study, we investigated the relationship of male rank to body weight, circulating testosterone and cortisol concentrations in captive possum triads (Trichosurus vulpecula). We carried out two experiments to examine hierarchy formation and the effects of castration of the dominant male during the non‐breeding season. A third experiment measured the effects of removal of the dominant male from a stable hierarchy during the breeding season. We found that dominant male rank was significantly correlated with higher circulating testosterone levels during periods of hierarchy formation and during the breeding season but not during periods of hierarchy stability in the non‐breeding season. Lack of correlation between plasma testosterone concentration and rank after male castration suggests that stable social rank is not dependent on hormone level and may be more dependent on behavioural traits. Any biocontrol measures that rely on manipulation of hormone levels may be unreliable when applied to unstable hierarchical situations, including the establishment of territories by subadult males, and the pre‐breeding season when circulating testosterone concentrations peak in males and a period of hierarchy establishment may occur.  相似文献

Summary Testicular development was studied in Tupaia belangeri (tree-shrew) from birth to sexual maturity. At birth the seminiferous cords contained peripheral supporting cells and centrally located gonocytes. Large foetal Leydig cells were prominent in the interstitium. The mitotic index of the gonocytes was low at birth and rose to peak levels at Day 20, following the regression of the foetal generation of Leydig cells, and during the nadir in circulating testosterone concentrations. Mitotic activity returned to low levels at Day 30 in association with the reappearance of differentiated Leydig cells and the first signs of increased androgenesis. The negative temporal relationship between mitogenesis and androgenic function suggests that the proliferation of the gonocytes does not require, and may be inhibited by, high titres of androgens. Post-mitotic development of the gonocytes occurred during a period of rising testosterone levels, and the first appearance of spermatogonia coincided with peak testosterone levels. This indicates that androgens may be specifically involved in the initiation of spermatogenesis. Spermatogenesis progressed to completion during a phase of declining testosterone levels. The precise temporal correlations established during post-natal development suggest that the tree-shrew is a suitable animal model for studies on the endocrine control of the initiation of spermatogenesis in primates.  相似文献

We investigated the effect of seasonal influence and geographic location on the testosterone concentrations of three adult male Formosan black bears kept at a Low Altitude Experimental Station, simulating natural breeding conditions. Estimations of fecal testosterone for males 1, 2, and 3 were made during January to October 2000, April 2001 to December 2003, and April 2002 to December 2003, respectively, using non-invasive enzyme-linked immunosorbent assays. Results indicated that fecal immunoreactive testosterone recrudesces after winter and ceases after summer. Mating behaviors were observed both before and after the fecal immunoreactive testosterone peak of March–May. In addition, fecal immunoreactive testosterone concentrations were significantly (p < 0.01) higher during January to June than from July to December. As expected, levels corresponded to the longer and earlier breeding seasons of subtropical zones. Formosan black bears appear to be seasonal breeders who breed primarily in spring, synchronized with sunlight and ambient temperature. The aim of this study was to determine if testosterone concentrations respond to seasonality thereby reflecting bear mating demands. Baseline parameters established in this study will help to evaluate endocrine information applicable for guidelines in future management and preservation of Formosan black bears in the field. Testosterone concentrations in serum samples collected weekly over a 5-year period from a young adult male Asian elephant (Elephas maximus) and a young adult male African forest elephant (Loxodonta africana cyclotis) were measured by radioimmunoassay. Testosterone profiles during this maturational period were compared between the two species and related to the occurrence of musth, a recurring physiological and behavioral condition exhibited by most mature Asian, and some African, bull elephants. Musth is characterized by secretion from the bull's temporal glands, dribbling urine, and increased aggression. Serum testosterone concentrations in the Asian bull were elevated substantially between April and September each year, coincident with the presence of temporal gland secretion, urine dribbling, and aggressive behavior. Testosterone levels from April through September averaged (± SEM) 41.2 ± 2.8 ng/ml, compared to 7.6 ± 1.0 ng/ml during the rest of the year. In contrast, the testosterone profile of the African bull showed greater variation and lower levels overall, the only pattern being a tendency for levels to be lowest from November to February (avg. 6.8 ± 1.5 vs. 10.3 ± 0.8 ng/ml during the rest of the year). Temporal gland secretion and other signs of musth were first observed in this bull in 1988, at age 17. While his testosterone profile did not show a pattern comparable to that in the Asian bull, average testosterone values were significantly greater in 1988 compared to previous years. The Asian bull showed sexual attention to preovulatory (estrous) cows whether in musth or not, and exposure to estrous cows did not appear to alter the highly consistent, annual pattern of musth as evidenced in temporal gland flow.  相似文献

Hydrolysis, extraction, and radioimmunoassay techniques for the estimation of excreted testosterone metabolites in the urine of saddle-back tamarins have been validated and are described. The steroids measured with the testosterone antiserum used are mostly present as glucuronides and sulfates. Immunoreactivity in high-performance liquid chromatography (HPLC) fractions of urinary extracts and in a standard mixture of cortisol, testosterone, and dihydrotestosterone (DHT) were compared. The fractions with the same retention data as testosterone accounted for the major part of the immunoreactivity. Several other immunoreactive compounds of unknown identity were present in low concentrations. These results suggest that testosterone conjugates are the major steroid metabolites measured with this method in Saguinus fuscicollis. Urinary testosterone levels of castrated males were much lower than those of intact males. Testosterone treatment of castrated males resulted in a temporary superphysiological increase in the levels of urinary testosterone and in an individually variable increase in the levels of the minor immunoreactive compounds. These results suggest that estimation of testosterone metabolite levels in urine is a valid method for the assessment of testicular activity in Saguinus fuscicollis.  相似文献

Peripheral concentrations of cortisol, growth hormone and testosterone were determined in two experiments which examined the endocrine and behavioral responses of sexually mature Angus bulls to an estrous female (Experiment 1) and to female exposure 5 hours following an adrenocorticotropin (ACTH) injection (Experiment 2). Sexual activity of bulls in Experiment 1 significantly increased levels of cortisol when compared with concentrations before exposure to a female. Administration of ACTH in Experiment 2 consistently elevated levels of cortisol by 30-fold (P<0.01) when compared with pre-ACTH concentrations. This heightened level of cortisol persisted throughout the period of exposure to an estrous cow, although a gradual decline in cortisol concentrations occurred over time (P<0.05). In Experiment 1, growth hormone profiles tended to increase in response to sexual activity (P<0.10), whereas in Experiment 2, growth hormone increased in response to ACTH administration (P<0.01) and to female exposure (P<0.01). Concentrations of testosterone were unaffected (P>0.10) by mating activity in Experiment 1. In Experiment 2, acute suppression (P<0.01) in testosterone concentrations 5 hours after ACTH administration coincided with the exposure period to the estrous female. Frequencies of mounting behavious (penis extension, mounting, intromission and ejaculation) exhibited by ACTH-treated bulls were significantly lower compared with the frequencies two days earlier. Exogenous ACTH administration suppressed reproductive behaviors of bulls and altered secretion of cortisol, growth hormone and testosterone. Furthermore, these data provide evidence that specific mating behaviors of the bull can be influenced by circulating steroids.  相似文献

This study addressed the question of whether maternal condition during egg laying or the rearing environment has a greater effect on offspring testosterone levels. We tested this in field experiments on a population of tawny owls Strix aluco in Duna‐Ipoly National Park, Hungary. In the experiments with females of poor condition, when broods were supplied with extra food none of the nestlings died, whereas in control broods, which were not supplied with extra food, some nestlings did die. Large differences in testosterone levels were correlated positively with hatching order both in experimental and control broods. However, it was only in control broods that the later‐hatched nestlings with low testosterone concentrations, died. In the experiments with females of good condition, the males were removed and females and their broods were supplied with restricted amounts of food. In these broods starving nestlings, whose growth had stopped, were considered as having died and were removed from the nest and hand‐reared. In control broods all nestlings fledged, and both in experimental and control broods testosterone concentrations were more even between siblings. Both types of trials confirmed a maternal influence on offspring testosterone concentration: large between‐sibling differences in concentrations in the broods of females of poor condition, where some nestlings died, could not be reduced with increased food supply, and the more even concentrations in the broods of females in good condition, where all nestlings survived, could also not be increased by restricting the food supply.  相似文献

Androgens are known to mediate aggressive and defensive behaviour in many vertebrate species. However, high concentrations of androgens might also conflict with the expression of nurturing behaviours and therefore a trade‐off can exist between aggressive and nurturing behaviours during parental care. We explored the role of testosterone in paternal care in bluegill sunfish (Lepomis macrochirus), where males provide both sole defence of the young from predators and sole nurturing behaviour such as fanning of the eggs. At the onset of parental care, we manipulated testosterone levels in males using testosterone propionate implants. We then observed the frequency of nurturing and aggressive behaviours displayed by the males over 6 d of parental care. Testosterone‐implanted fish were more aggressive when presented with a brood predator, performing more bites, opercular flares and lateral displays than control males. Testosterone‐implanted males, however, were not less nurturing than control fish, performing similar levels of fanning and nest‐cleaning behaviours. Thus, our results support a positive relationship between testosterone and paternal aggression but no testosterone‐mediated trade‐off between paternal nurturing and aggression.  相似文献

The purposes of the present study were to investigate the effect of conjugated linoleic acid (CLA) supplementation on testosterone levels in vitro on a cell line derived from Leydig cells (R2C) and in vivo in the blood of physically active subjects before and after a resistance exercise bout. In vitro R2C cells were treated with different CLA concentrations (0-30 μM) for 24 and 48 hours. After treatment, supernatant media were tested to determine testosterone secretion. The CLA increased the testosterone secretion only after 48 hours. In vivo, 10 resistance-trained male subjects, in a double-blind placebo-controlled and crossover study design were randomized for 3 weeks of either 6 g·d?1 CLA or placebo. Blood was drawn pre and post each resistance exercise bout to determine the total testosterone and sex hormone-binding globulin (SHBG) levels. No significant differences were observed for total testosterone or SHBG pre and post each resistance exercise bout; although after the resistance exercise bouts, total testosterone increased moderately (effect size = moderate), whereas after CLA supplementation, there was a large increase in total testosterone (effect size = large). CLA supplementation induced an increase in testosterone levels in Leydig cells in vitro after 48 hours but not in vivo before and after a resistance exercise bout. These findings suggest that CLA supplementation may promote testosterone synthesis through a molecular pathway that should be investigated in the future, although this effect did not have an anabolic relevance in our in vivo model.  相似文献

To assess diurnal fluctuations of serum androgens and cortisol in adult male Bolivian squirrel monkeys, these steroids were measured at predetermined times (0300, 0900, and 2300 hours) during two separate 24-hour periods in the breeding season (January 1983 and late November 1983). A significant diurnal change in serum cortisol was noted, with a nadir of 99.9 ± 11.9 μg/dl (x? ± SEM) at 2300 hours and a peak of 168.9 ± 7.8 μg/dl at 0900 hours. Conversely, a nadir in serum testosterone was noted at 0900 hours (117 ± 26.5 ng/ml) increasing to a peak of 328.5 ± 57.9 ng/ml at 0300 hours. Serum androstenedione and dehydroepiandrosterone followed a pattern similar to testosterone, with a serum androstenedione (176.4 ± 34.9 ng/ml) and dehydroepiandrosterone (11.7 + 1.8 ng/ml) nadir at 0900 hours and a plasma androstenedione (494.5 ± 55.4 ng/ml) and dehydroepiandrosterone (32.5 ± 4.1 ng/ml) peak at 0300 hours. Parallel changes of testosterone, androstenedione, and dehydroepiandrosterone suggest a significant contribution of all three androgens from a common site, the testes. In contrast to old world primates and humans, serum androstenedione levels exceeded serum testosterone levels in this species.  相似文献

Acute neuromuscular and endocrine adaptations to weight-lifting were investigated during two successive high intensity training sessions in the same day. Both the morning (I) (from 9.00 to 11.00 hours) and the afternoon (II) (from 15.00 hours to 17.00 hours) training sessions resulted in decreases in maximal isometric strength (p less than 0.01 and less than 0.05), shifts (worsening) in the force-time curve in the absolute scale (p less than 0.05 and ns.) and in decreases in the maximal integrated EMG (p less than 0.01 and less than 0.05) of the selected leg extensor muscles. Increases in serum total (p less than 0.05) and free testosterone (p less than 0.01) and in cortisol (p less than 0.01) concentrations were found during training session II. These were followed by decreases (p less than 0.001 and p less than 0.01 and ns.) in the levels of these hormones one hour after the termination of the session. The responses during the morning training session were different with regard to the decreases in serum total testosterone (p less than 0.05), free testosterone (ns.) and cortisol (p less than 0.05). Only slight changes were observed in the levels of luteinizing hormone and sex hormone-binding globulin during the training sessions. Increases (p less than 0.01) took place in somatotropin during both training sessions. The present findings suggest that high intensity strengthening exercises may result in acute adaptive responses in both the neuromuscular and endocrine systems. Latent toxoplasmosis is known to influence the morphology of infected persons and also increases the probability of the birth of male offspring in both humans and mice. All these traits can be related to the observed differences in the concentration of testosterone between Toxoplasma-infected and Toxoplasma-free subjects. However, it is not possible to decide, using the Toxoplasma-human model, whether toxoplasmosis influences the level of testosterone in the infected host or whether individuals with different levels of testosterone vary in the probability of toxoplasma infection. Here we studied changes in the testosterone levels in the latent phase of toxoplasmosis in laboratory mice artificially infected with cystogenic but relatively virulent strain T38 of T. gondii. We observed decreased testosterone levels in both female and male mice with latent toxoplasmosis in comparison to uninfected controls (P = 0.001). The present results indicate that Toxoplasma infection changes the concentration of serum testosterone in mice and human rather than changed concentration of testosterone influences the probability of the Toxoplasma infection. It is possible that the decrease of testosterone is an adaptive mechanism of infected mice aimed to compensate toxoplasmosis-induced immunosuppression observed during latent Toxoplasma infection.  相似文献

Cyproterone acetate was administered either orally or intraperitoneally to intact, adult male newts, Taricha granulosa. The number of males that exhibited the courtship behavior of clasping when tested with nuptial females was not altered by the antiandrogen treatments. In males which were unresponsive to nuptial females, the occurrence of clasping was not evoked by injections for 4 days of testosterone, dihydrotestosterone, or 11-ketotestosterone. Further, the incidence of clasping was not significantly elevated by injections of prolactin and/or testosterone for 30 days. The effect of sexual activity on testosterone and dihydrotestosterone levels in male newts was determined by radioimmunoassay of plasma collected from males which were: (1) isolated from females; (2) allowed to clasp a female for 2 min; or (3) allowed to clasp a female for 1 hr. The testosterone and dihydrotestosterone levels were unchanged during this period of clasping. In February and again in June, plasma androgen concentrations were measured in males which differed in their propensity to initiate courtship when paired with females. Androgen levels were similar for males that clasped a female and males that never attempted to clasp a female. Plasma androgen levels in the male newt are apparently not correlated with sexual responsiveness.  相似文献

ObjectiveTo determine the pharmacokinetics, bioavailability, and safety of a new formulation (1.62%) of testosterone gel that produces eugonadal serum testosterone levels with use of a lower amount of gel than the currently available 1% gels.MethodsIn an open-label, randomized, 3-way crossover study, 36 male patients with hypogonadism applied 5 g of 1.62% testosterone gel (81 mg of testosterone) once daily to the abdomen, to the upper arms/shoulders, or alternating between both sites per an established schedule for7 days. Serum levels of testosterone, dihydrotestosterone, and estradiol were measured and used to compare the pharmacokinetics and bioavailability of the 3 treatments.ResultsEach application method produced average serum testosterone concentrations within the eugonadal range (300 to 1,000 ng/dL), and steady-state testosterone concentrations were achieved after 2 days of gel application to either the abdomen or the upper arms/shoulders. When testosterone gel was applied to the abdomen, approximately 30% to 40% lower bioavailability (based on area under the serum concentration-time curve from 0 to 24 hours) was observed in comparison with application to the upper arms/shoulders. The 1.62% testosterone gel was found to be safe and well tolerated in men with hypogonadism.ConclusionAlthough lower testosterone bioavailability was observed after abdominal application of 1.62% testosterone gel in comparison with application to the upper arms/shoulders, application to either site yielded eugonadal levels of serum testosterone. The black‐handed spider monkey (Ateles geoffroyi) is a seasonal reproducer that requires a seclusiveness to copulate and has a fusion–fission social system. These features impose important restrictions to achieve reproduction of captive animals. We investigated if group composition in captive spider monkeys has any endocrine effects. We compared testosterone and cortisol concentrations during the mating season in all‐male and multifemale–multimale groups to study if the former condition impairs reproductive potential and increases stress. Concentrations of testosterone and cortisol of males living with females were higher than those of all‐male groups. In the multifemale–multimale condition, dominant males had the highest levels of testosterone, while the youngest males showed the highest concentrations of cortisol. Results show that males adjust well to isosexual grouping, this being an appropriate condition to keep animals when controlled reproduction is sought. Fecal steroid monitoring was applied as a non‐invasive method to investigate testicular cycles and seasonality in the Malayan Sun bear (Ursus malayanus), an endangered ursid from South East Asia. Fecal testosterone was analyzed by radioimmunoassay in samples collected from male Sun bears (n=8) housed in zoological parks in North America and New Zealand, over periods of <27 months. Testosterone levels were often, but not exclusively, elevated during mating periods with peaks accompanying breeding behavior and copulation. There was a significant effect of age with older bears having clearly higher concentrations of fecal testosterone (P<0.001). Testosterone concentrations fluctuated throughout the year, with no significant effect of season (P>0.05). All bears did, however, share a common pattern of annual excretion that suggests a potential role for non‐photoperiodic seasonal influences on testicular cycles. Levels were generally lower early in the year with regular increases occurring at 3–4‐month intervals.
