In many clinical trials, it is desirable to establish a sequential monitoring plan, whereby the test statistic is computed at an interim point or points in the trial and a decision is made whether to stop early due to evidence of treatment efficacy. In this article, we will set up a sequential monitoring plan for randomization-based inference under the permuted block design, stratified block design, and stratified urn design. We will also propose a definition of information fraction in these settings and discuss its calculation under these different designs.  相似文献

The importance of the starting point for iterative robust regression is well known since the ordinary least square solution often used doesn't always give good results. We suggest a new method called “regression by the sectors” (RPS) as a starting point to an iterative windsorisation of the original data (ITER). We used artificial samples to compare RPS and other quick techniques of fit: regression by median, Hinich-Talwar's method, L1 norm minimization. Besides RPS-ITER algorithm is tested (examples and simulations) in relation to algorithms using different starting points associated to ITER. The results show the superiority of RPS-ITER.  相似文献

In classical linear regression model for analysis of medical data concerning hepatic extraction of insulin and c-peptide the fundamental assumption is that the subjects involved are of a similar nature. In reality, if this assumption is violated then the precision of the results is questionable. This paper suggests a robust alternative to overcome this problem. It is observed that the robustification may be a better option to determine an optimal quantity of insulin which minimizes the risk of damage associated with diabatic treatments.  相似文献

A new modification of Berkson's minimum logit chi-squared estimator in simple linear logistic regression is suggested in order to achieve reduction of first order bias of the estimator as well as in the model. Furthermore, unlike estimators currently available, our procedure is quite simple to apply in practice and is valid even in the presence of zero frequencies in the table.  相似文献

Summary .  Size-biased sampling arises when a positive-valued outcome variable is sampled with selection probability proportional to its size. In this article, we propose a semiparametric linear regression model to analyze size-biased outcomes. In our proposed model, the regression parameters of covariates are of major interest, while the distribution of random errors is unspecified. Under the proposed model, we discover that regression parameters are invariant regardless of size-biased sampling. Following this invariance property, we develop a simple estimation procedure for inferences. Our proposed methods are evaluated in simulation studies and applied to two real data analyses.  相似文献

Many modern genomic data analyses require implementing regressions where the number of parameters (p, e.g., the number of marker effects) exceeds sample size (n). Implementing these large-p-with-small-n regressions poses several statistical and computational challenges, some of which can be confronted using Bayesian methods. This approach allows integrating various parametric and nonparametric shrinkage and variable selection procedures in a unified and consistent manner. The BGLR R-package implements a large collection of Bayesian regression models, including parametric variable selection and shrinkage methods and semiparametric procedures (Bayesian reproducing kernel Hilbert spaces regressions, RKHS). The software was originally developed for genomic applications; however, the methods implemented are useful for many nongenomic applications as well. The response can be continuous (censored or not) or categorical (either binary or ordinal). The algorithm is based on a Gibbs sampler with scalar updates and the implementation takes advantage of efficient compiled C and Fortran routines. In this article we describe the methods implemented in BGLR, present examples of the use of the package, and discuss practical issues emerging in real-data analysis.  相似文献

We assessed complementary log–log (CLL) regression as an alternative statistical model for estimating multivariable‐adjusted prevalence ratios (PR) and their confidence intervals. Using the delta method, we derived an expression for approximating the variance of the PR estimated using CLL regression. Then, using simulated data, we examined the performance of CLL regression in terms of the accuracy of the PR estimates, the width of the confidence intervals, and the empirical coverage probability, and compared it with results obtained from log–binomial regression and stratified Mantel–Haenszel analysis. Within the range of values of our simulated data, CLL regression performed well, with only slight bias of point estimates of the PR and good confidence interval coverage. In addition, and importantly, the computational algorithm did not have the convergence problems occasionally exhibited by log–binomial regression. The technique is easy to implement in SAS (SAS Institute, Cary, NC), and it does not have the theoretical and practical issues associated with competing approaches. CLL regression is an alternative method of binomial regression that warrants further assessment.  相似文献

In his recent paper Liski (1989) derived conditions for superiority of the minimum dispersion estimator over another with respect to the covariance matrix when the parameter vector of a regression model is subject to competing stochastic restrictions. Linear regression formulae are given for converting 1H and 13C magnetic shielding constants calculated at common ab initio and density functional theory levels of calculation into chemical shifts relative to tetramethylsilane. Accuracies of roughly ±2.2 ppm (13C) and ±0.15 ppm (1H) or better are found for the training set for most levels. Stratified Cox regression models with large number of strata and small stratum size are useful in many settings, including matched case-control family studies. In the presence of measurement error in covariates and a large number of strata, we show that extensions of existing methods fail either to reduce the bias or to correct the bias under nonsymmetric distributions of the true covariate or the error term. We propose a nonparametric correction method for the estimation of regression coefficients, and show that the estimators are asymptotically consistent for the true parameters. Small sample properties are evaluated in a simulation study. The method is illustrated with an analysis of Framingham data.  相似文献
