The level of resistance to root-knot nematode, Meloidogyne incognita, in NemX, a new cultivar of the Acala-type upland cotton, was evaluated in relation to four resistant breeding lines (N6072, N8577, N901, and N903) and four susceptible cultivars (Maxxa, SJ2, Royale, and Prema). In growth pouch tests, an average of only 4 nematode egg masses was produced on roots of NemX or the resistant lines, compared to a significantly higher average of 21 on the susceptible cultivars. In pot tests, the nematode reproduction factor (RF = Pf/Pi) in NemX and the resistant lines averaged 0.7, compared to a significantly higher average of 10 on the susceptible cultivars. Root galling in NemX or other resistant cotton averaged 15%, compared to 74% on the susceptible cultivars, in either pot or field tests. In plots with low levels of nematode infestation (Pi ≤ 150 second-stage juveniles [J2]/500 g soil), lint yield of NemX averaged 1,370 kg/ha and was less than the yield of susceptible Maxxa (1,450 k g /h a ). However, in plots with medium or high levels of nematode infestation (Pi = 151-300 or >300 J2/500 g soil, respectively), yields of NemX decreased only slightly and averaged 1,300 or 1,050 kg/ha, respectively, whereas yields of Maxxa were severely reduced to 590 or 503 kg/ha, respectively. Fusarium wih symptoms were observed on both NemX and Maxxa, and percent occurrence increased with increasing preplant nematode density. In plots with the highest nematode densities, 22% of NemX and 65% of Maxxa plants were wilted. NemX was highly effective against five M. incognita isolates and moderately effective against a sixth isolate that had been exposed to resistant cotton over several seasons. These results showed that NemX is as resistant to M. incognita as the four breeding lines, and much more resistant than the tested susceptible cultivars of cotton.  相似文献

Quantities of free amino acids in segments of cotton roots resistant and susceptible to Meloidogyne incognita were compared. Following infection, the root-knot susceptible cultivar, M8, had greater percentage increases of certain individual free amino acids than the resistant cultivar, Clevewilt, but the sum total of free amino acids was greatest in the resistant cultivar. More free amino acids were present in infected than in noninfected plants of both cultivars. The overall concn of glycine declined over the I 0-day period following inoculation. The concns of the aromatic amino acids, tyrosine and phenylalanine, varied as functions of infection, cultivar, and time of harvest. Proline in susceptible M8 increased nearly 2000-fold 10 days after infection, when considerable thickening of syncytial walls is occurring.  相似文献

The timing and expression of resistance to four isolates of Plasmodiophora brassicae, collected from research sites where pathotypes 2, 3, 5 and 6 (Williams' system) had been dominant when characterised in 2006, were assessed in four new commercial cultivars of canola (Brassica napus) with resistance to clubroot. Each of the resistant cultivars was highly resistant to all four of the isolates, and there was no difference in their response to infection. Root hair infection occurred at high levels, but pathogen development occurred more slowly than in a susceptible cultivar (control). Secondary infection and development in cortical cells was severely inhibited in each of the resistant cultivars; only a few bi‐nucleated plasmodia were observed at 12 days after inoculation (DAI), and plasmodia were rarely observed at 18 and 24 DAI. In contrast, development in the susceptible cultivar had progressed to resting spores by 24 DAI. A dense ring of accumulated reactive oxygen species (ROS) was observed in the endodermis, pericycle and vascular cambium of non‐inoculated controls and inoculated plants of the resistant cultivars. However, the ROS ring disappeared rapidly in infected plants of the susceptible cultivar. Plasmodia invaded the stele of susceptible roots by preferentially colonising the xylem parenchyma cells. Expansion and enlargement of lignified xylem cells was observed by 35 DAI. The absence of any specific points of ROS accumulation or lignification of epidermal or cortical cells in the resistant cultivars indicates that a hypersensitive response is not the main mechanism of resistance in these lines. The uniform response of these resistant cultivars to the four isolates of P. brassicae indicates that the resistance in each cultivar may be conditioned by a gene(s) from a single source that confers broad resistance, because most sources of resistance to P. brassicae are pathotype specific.  相似文献

Ninety-six cultivars of Brassica oleracea were screened for clubroot resistance in a seedling test using two populations of Plasmodiophora brassicae. The most resistant cultivars were kales. Sixteen resistant marrowstem kale cultivars of diverse geographical origin were used to start a selection programme for clubroot resistance. Four generations of selection, involving single plants, half-sib and full-sib families, reduced a disease index averaged over six clubroot populations from 41.2 to 12.5. This was lower than the most resistant cultivar in the original population, cv. Mixti 28.8, and as good as a German landrace of cabbage noted for its resistance, Bohmerwaldkohl 10.5. In comparison, the mean of five kale controls, cvs Bittern, Canson, Condor, Kestrel and Merlin, was 61.1 and the value for the most susceptible control, cabbage cv. Septa, was 89.3. In the final assessment, there were no clubroot population x B. oleracea genotype interactions and in the initial assessment of cultivars there were only small interactions which could be removed by an angular transformation of the data. It was concluded that a high level of non-differential resistance had been achieved and that it may prove durable. It was also concluded from a small field trial that this level of resistance would prevent serious yield losses in practice.  相似文献

Plasmodiophora brassicae causes clubroot in the turnip, Brassica rapa L. We used organ cultures of adventitious roots from B. rapa seedlings to investigate the initial response of resistant and susceptible cultivars to P. brassicae infection. Primary plasmodia of P. brassicae were observed in root hairs of both susceptible and resistant cultured roots. On the other hand, secondary plasmodia were able to proliferate only in the susceptible root culture but not in the resistant one. Root cultures from the susceptible cultivar all developed clubroot 4 weeks after treatment with 104, 105 or 106 spores/ml, but roots from the resistant cultivar did not develop clubroot under the same conditions. Cell death, as measured by Evans blue and TTC dye methods, was observed in cultured roots from the resistant cultivar but did not occur in roots from the susceptible cultivar after exposure to P. brassicae spores. Cell death was inhibited almost completely by EGTA and verapamil but not by the calmodulin antagonist W7. These results suggest the involvement of Ca2+ in P. brassicae‐induced cell death. Alkalization of the root culture medium of the resistant cultivar was observed 2 days after treatment with P. brassicae spores but was not observed in root culture medium from the susceptible strain. We conclude that our root culture system must be a useful tool for further studies of the molecular mechanism of clubroot resistance.  相似文献

In a glasshouse study, 54 batches of cabbage seedlings were each grown for 6 wk in clubroot inoculated soil. The batches were sown at weekly intervals during the experiment which lasted 60 wk. Soil temperature and light were monitored throughout the study while other potential variables were maintained as constant as possible. Records of clubroot incidence, severity and plant weights were kept and causal relationships for disease severity and percentage infected plants sought among the light and temperature data by regressions. It was found that clubroot severity was influenced most by the light level in the second and third weeks, while the percentage of diseased plants was most influenced by temperature, that in the second week probably being the most important. The optimum light value in the second and third weeks for expression of maximum clubroot severity was c. 600 Wh m-2 day-1 and a mean daily temperature of not less than c. 19-5 oC was required to give close to 100% infection. Possible reasons for the findings are discussed.  相似文献

The effects of cassava mosaic virus disease (CMD) on yield in fully and partly infected stands of cassava were investigated in field trials in Uganda in 1990-91 and 1991-92. Three cultivars (Ebwanateraka, Bao and Bukalasa 1 l), each at three levels of cutting infection (O%, 50% and 100%) and harvested 510 and 15 months after planting (MAP) were used in a randomised block design with split-split plots and four replicates. Moreover, yield and growth data for individual infected and uninfected plants were considered in relation to the health status of their nearest neighbours. In each experiment, fresh tuberous root yields of plants from 100% infected plots gave sigdicantly lower yields than those from 0% or 50% infected plots at each harvest date and the losses were greatest in cv. Bao. Yields of plants from 0% and 50% plots for each of the three cultivars were not significantly different, 10 and 15 MAP. The loss in yield differed between cultivars and harvest dates. Fresh stem, leaf and root yields and the number of tuberous roots were influenced by the health status of the plants harvested and that of their nearest neighbours. Uninfected plants surrounded by infected ones had more roots and heavier total fresh root, stem and leaf weights than those surrounded by uninfected ones. Overall, 26% and 42% compensation was recorded in 1990-91 and 1991-92, respectively. The effects of CMD on cassava production and of compensation in mixed stands of infected and uninfected plants are discussed, especially in relation to control strategies such as roguing.  相似文献

The effects of infection with potato leafroll virus (PLRV) on the four crop processes leading to tuber fresh weight yield were examined in field plots of four cultivars (Montana, Pentland Crown, Maris Piper and King Edward) differing in tolerance to infection. Averaged across cultivars, infection decreased yield by 50%. This decrease was equally due to less light (total solar radiation) being intercepted, a lower efficiency with which intercepted light was converted into dry weight, and a smaller proportion of dry weight being partitioned to tubers. Dry matter content of the tubers was also diminished but to a lesser extent. The main difference between the cultivars in their response to infection was in the partitioning of dry weight. In Montana and Pentland Crown, harvest index was decreased by 15% in infected plants, whereas in the less tolerant cultivars, Maris Piper and King Edward, it was decreased by 25%. The decline in photosynthetic performance of Montana, a cultivar with slightly earlier maturity than the other three, was delayed in PLRV-infected plants. Effects on number of daughter tubers essentially reflected those on yield to the extent that average tuber weight did not change in Maris Piper, was one third less in King Edward, and the change was intermediate in Montana and Pentland Crown.  相似文献

Summary A field experiment was carried out to study the influence of vesicular-arbuscular mycorrhiza (VAM) on nutrient uptake and growth of pea at two levels of P fertilizer. Pea was grown in dazomet-fumigated and non-fumigated soil, either with or without addition of inoculum of VAM fungi. Dazomet efficiently eliminated infection by indigenous VAM fungi, whereas roots from untreated soil were extensively infected. Inoculation with VAM fungi increased P uptake by peas in fumigated plots, but had no significant effect on dry matter production in either soil treatment. However, VAM developed less in inoculated fumigated than in untreated plots. Seeds of uninfected plants in fumigated plots contained only 56 and 81%, respectively, of the P and dry matter content of seeds of infected plants in untreated plots. The addition of 60 kg P ha−1 also increased seed yields but to a smaller degree than VAM. The seed yield as a proportion of total shoot yield (harvest index) was higher in VAM infected than in uninfected peas but was unaffected by P fertilizer. Alternaria linicola survived as thick-walled chlamydospores in hyphal or conidial cells on infected linseed stem debris, either on the soil surface or buried in the soil, during the period between the harvest of linseed and sowing of the following crop (September - April). Conidia produced on these debris under favourable conditions were not only viable but also pathogenic to young linseed seedlings. Infected stem debris increased the incidence of infected seedlings which emerged from infected seed (incidence of A. linicola 1% to 28%), especially if the debris was on the soil surface rather than buried. A. linicola also survived between successive linseed crops on infected volunteer linseed plants which survived the low temperatures in winter and on the weed Veronica persica. A. linicola was more effectively transmitted from infected seeds to seedlings at temperatures 15–25°C than at 10°C. The incidence of the disease on seedlings which emerged from infected seed was positively correlated with the amount of seed-borne inoculum, whereas the proportion of seedlings which emerged was negatively correlated with the incidence of A. linicola on the seed.  相似文献

Pathogenecity and race classification of some isolates of V. dahliae from resistant tomato in Morocco In Morocco, the cv. H 204, resistant to Verticillium dahliae race 1 is widely used under plastic tunnels. Use of this cultivar controlled the disease for many years but recently a high incidence of Verticillium on this hybird was obsereved. V. dahliae was isolated from 90 % of the wilted plants.The other 10 % were infected by F. oxysporum f. sp. lycopersici alone (7 %) or in association with V dabliae (3 %). The inoculation of the Verticillium susceptible cv. Vemone and of the race 1 resistant cv. H 204 by 120 isolates obtained from the resistant hybird, showed that (69 %) of the isolates are race 1 and 31 % are race 2. There are different virulence levels of V. dahliae race 2 both on the susceptible cv. Vemone and on the race 1 resistant cv. H 204. Race 2 on the average was less virulenton the susceptible than on the resistant cultivar.  相似文献

Experiments were made on field plots with four soil partial sterilants applied at different dose rates or as combination treatments. The effects of treatments on clubroot were assessed by growing cabbages on the field plots or in a glasshouse in soil removed from the plots. In two experiments, no plants grown on plots or seedlings grown in soil samples from plots treated with Dowfume M.C.2 or Telone were infected with clubroot, whilst only one infected seedling was found in soil samples from those plots treated with chloropicrin. Although relatively few infected plants were recovered from Basamid treated plots, many infected seedlings were found in soil samples from those plots. The effectiveness of these treatments as indicated by seedling survival was Dowfume M.C.2 < chloropicrin < Telone < Basamid. At five sites a combination of Dowfume M.C.2 at 98 g/m2 with 40 g/m2 Basamid resulted in all cabbage seedlings grown in soil samples being disease free.  相似文献

Collar rot, caused by Rhizoctonia solani Kühn, is one of the most severe fungal diseases of opium poppy. In this study, heritability, genetic advance and correlation for 10 agronomic, 1 physiological, 3 biochemical and 1 chemical traits with disease severity index (DSI) for collar rot were assessed in 35 accessions of opium poppy. Most of the economically important characters, like seed and capsule straw yield per plant, oil and protein content of seeds, peroxidase activity in leaves, morphine content of capsule straw and DSI for collar rot showed high heritability as well as genetic advance. Highly significant negative correlation between DSI and seed yield clearly shows that as the disease progresses in plants, seed yield declines, chiefly due to premature death of infected plants as well as low seed and capsule setting in the survived population of susceptible plants. Similarly, a highly significant negative correlation between peroxidase activity and DSI indicated that marker-assisted selection of disease-resistant plants based on high peroxidase activity would be effective and survived susceptible plants could be removed from the population to stop further spread.  相似文献

大白菜是中国北方的重要蔬菜之一,根肿病是危害大白菜生产世界性病害,利用花药培养可以大大加速抗根肿病大白菜杂交育种工作的进程。对33份抗根肿病大白菜品种(品系)进行花药培养,有24个品种诱导出胚,品种诱导率为72.7%,以东方皇冠×C11的诱导率最高,为1.86胚/蕾。对大部分基因型来说,在培养基中添加0.4 g/L谷氨酰胺的诱导效果最好。研究还发现生长在温室中的供体植株更容易诱导出胚状体。变绿的子叶型胚转到B5+0.2 mg/L BA+0.1 mg/L NAA+0.1%活性炭的胚分化培养基上可诱导生芽。  相似文献

We have examined the effect of a strain of Banana streak virus (BSV‐Cav) on the growth and yield of dessert bananas (Musa AAA group, Cavendish subgroup cv. Williams) in north Queensland, Australia. Healthy and infected plants were compared in a replicated field experiment over plant and first ratoon crops. In both crops, symptom expression followed a similar pattern, increasing to a maximum near the estimated time of bunch initiation, then decreasing in the period prior to bunch emergence. There was no evidence of plant‐to‐plant spread of virus, but the rate of transmission through suckers was 100%. In the plant crop, the mean bunch weights of healthy and infected plants were not significantly different. However, BSV‐Cav infection resulted in an 18 day delay in harvest, causing a 6% reduction in yield per annum. In the ratoon crop, the mean bunch weight of infected plants was 7% less than that of healthy plants, and the interval between the harvest of plant and ratoon crops was delayed by 9 days, resulting in a 11% reduction in yield per annum. Also, the mean length of fruit from infected plants was 5% less than that of healthy plants, resulting in a smaller percentage of fruit in the extra large size category. We conclude that in horticulturally favourable conditions typical of the tropical Australian banana industry, the effects of BSV‐Cav infection on the growth and yield of Cavendish bananas are small.  相似文献

The effect of soybean mosaic virus (SMV) infection on symptom severity, yield, seed mottling and seed transmission in soybean in relation to the growth stage at infection and subsequent temperature was investigated using a susceptible (Harosoy), a moderately resistant (Evans) and a highly resistant (Merit) cultivar. Disease symptoms were more severe with early infection. A greater reduction in plant growth and seed yield, and higher percentages of mottled seeds and seed transmission of SMV also occurred with early infection. Virus titer was higher in younger plants than in older ones and also higher in plants infected at the ealier stage than at the later stage of growth. Merit (a highly resistant cultivar previously reported to be immune to seed mottling) inoculated at the early stage of plant growth resulted in infection and production of some mottled seeds. Temperature affected all parameters investigated. The effect of temperature was greater in the susceptible cultivar than in the resistant one. The optimal temperature for symptom severity, yield, seed mottling and seed transmission was 20 °C. Virus titer was highest at 30 °C in all three cultivars. Maturity of susceptible cultivar was delayed by infection.  相似文献

This study was conducted to determine if there were differences in competitive ability and yield stability on the growth of an old landrace of winter wheat (Triticum aestivum), Pinglang 40 (PL40), and a modern cultivar, Changwu135, (CW135), which differed in time of cultivar release, height, shoot and root biomass. A second aim was to investigate whether there is a relationship between competitive ability and yield stability. One pot and two field experiments were conducted to monitor changes in the competitive ability and yield stability of an old and a modern winter wheat cultivar grown in monoculture and mixtures using a de Wit replacement series. The pot study was conducted at two soil moisture levels: (a) well watered (WW), soil maintained at 85% field capacity, and (b) moderate drought (MD), soil maintained at 55% field capacity. The field experiments were conducted in 2 years that were drier than the average. In the second field experiment 40 mm of irrigation was applied to half of the plots at jointing, booting and anthesis. The results were similar in the pot and field experiments. In the mixture, the old cultivar PL40 had a greater relative yield with a larger number of spikes per plant when compared with those of the modern cultivar CW135. The dry matter per stem of CW135 tended to decrease with the increasing proportion of PL40, indicating a lower competitive ability of the modern cultivar than that of the old cultivar. The superior competition of PL40 was primarily due to the higher plant height, larger leaf area index, greater tillering capacity and larger root system. Our results showed that the modern cultivar CW135 produced a higher grain yield, yield components (except spike number), water use efficiency (WUEg) and harvest index under both water regimes in monoculture. However, the reduction in grain yield of CW135 when subjected to water-limited conditions was less than that of PL40 in the pot and field experiments. The greater grain yield of the CW135 was associated with a higher harvest index, thousand-kernel-weight and a lower root:shoot ratio. Water consumption over the entire growing period was significantly lower in CW135 under all soil moisture conditions, and the main difference in water consumption between the two cultivars was observed before anthesis. Post-anthesis accumulation of dry matter was greater in CW135 under water-limited conditions, but there was no difference between cultivars when water was adequate. The dry matter remobilization and contribution to grain yield of CW135 was lower than that of PL40. The results of the present study demonstrated that the higher competitive ability in the old landrace led to an increased sensitivity to environmentally-induced stress. As a result, there was a greater loss in grain yield by the old cultivar when the water supply was unfavorable. In addition, the differences in the life history strategy between the old and modern cultivars imply that reducing competitive ability in the modern cultivar has led to increased yield of the crop population and greater yield stability.  相似文献

Clubroot is a devastating disease caused by Plasmodiophora brassicae and results in severe losses of yield and quality in Brassica crops. Many clubroot resistance genes and markers are available in Brassica rapa but less is known in Brassica oleracea. Here, we applied the genotyping-by-sequencing (GBS) technique to construct a high-resolution genetic map and identify clubroot resistance (CR) genes. A total of 43,821 SNPs were identified using GBS data for two parental lines, one resistant and one susceptible lines to clubroot, and 18,187 of them showed >5× coverage in the GBS data. Among those, 4,103 were credibly genotyped for all 78 F2 individual plants. These markers were clustered into nine linkage groups spanning 879.9 cM with an average interval of 1.15 cM. Quantitative trait loci (QTLs) survey based on three rounds of clubroot resistance tests using F2 : 3 progenies revealed two and single major QTLs for Race 2 and Race 9 of P. brassicae, respectively. The QTLs show similar locations to the previously reported CR loci for Race 4 in B. oleracea but are in different positions from any of the CR loci found in B. rapa. We utilized two reference genome sequences in this study. The high-resolution genetic map developed herein allowed us to reposition 37 and 2 misanchored scaffolds in the 02–12 and TO1000DH genome sequences, respectively. Our data also support additional positioning of two unanchored 3.3 Mb scaffolds into the 02–12 genome sequence.  相似文献

Abstract:  Field studies were conducted in Harran Plain, Turkey, to investigate the infestation of spiny bollworm (SBW) and pink bollworm (PBW) in cotton blind bolls during the period 1999–2001. Blind bolls left on the cotton plants after the harvest were collected, and the PBW and SBW larvae, which are the causes of infection were counted in the laboratory by splitting the bolls. The statistical methods reveal that the infestation ratio variable plays an important role in reducing cotton yield. The infestation ratio variable was found to be statistically significant in all models used. Different soil types may have little effects on yield. We also estimate elasticity of yield with respect to infestation ratio variable. A 1% increase in infestation ratio would reduce about 2.5–6% of cotton yield. Although the infestation ratio of cotton yield is inelastic, which means that the yield is less responsive to the infestation ratio, the elasticity is however an economically important indication because additional cost spent on pests will increase total cost thereby increasing the level where marginal revenue equals the marginal cost.  相似文献
