Effect of nitrogen (N) deficiency on antioxidant status and Cd toxicity in rice seedlings was investigated. N deficiency resulted in a reduction of shoot growth but not root growth. The contents of N-containing compounds such as nitrate, chlorophyll, and protein decreased in leaves of rice seedlings grown under N deficiency. Accumulation of abscisic acid and H2O2 in leaves was induced by N deficiency. The content of ascorbate and the activities of ascorbate peroxidase, glutathione reductase, and catalase in N-deficient leaves were lower than their respective control leaves. However, glutathione content was not affected and superoxide dismutase activity was increased by N deficiency. Cd toxicity in N-deficient seedlings was more pronounced than that in N-sufficient ones. Pretreatment with ascorbate or L-galactono-1,4-lactone, a biosynthetic precursor of ascorbate resulted in a reduction of Cd toxicity enhanced by N deficiency. N deficiency also resulted in an enhancement of Cd uptake in rice seedlings. The possible mechanism of Cd toxicity enhanced by N deficiency is discussed.  相似文献

The effects of exogenous abscisic acid (ABA) on lead tolerance in rice (Oryza sativa L.) seedlings were investigated. Pre-treatment with 0.1 g m3 ABA for 2 d restricted amount of Pb translocated from roots to shoots, decreased malondialdehyde and H2O2 contents in leaves, and alleviated Pb-induced decrease in plant growth and leaf chlorophyll content. Further, ABA pre-treatment adjusted leaf antioxidative enzyme activities (increased ascorbate peroxidase and catalase activities while decreased superoxide dismutase activity) and so alleviated oxidative stress.  相似文献

Rice (Oryza sativa L.) seedlings were grown hydroponically in Hoagland’s nutrient solution under controlled conditions to investigate the effects of NaCl pretreatment on their response to subsequent application of cadmium (Cd) alone and Cd + NaCl combination. The Cd stress caused growth retardation in all plants, significantly reduced pigment content, stomatal conductance (gs), and net photosynthetic rate (PN). Cd stress significantly increased malondialdehyde and proline content. Compared to Cd treatment alone, combination stress had more detrimental effects on the above parameters. However, the NaCl pretreatment was beneficial in improving the plant growth and plant tolerance to Cd alone or combination stress.  相似文献

The effects of Cd on changes in proline level and peroxidase activity in roots of rice seedlings were investigated. CdCl2 was effective in inhibiting root growth and in accumulating proline in roots. The inhibition of root growth by Cd is reversible. The reduction of root growth induced by Cd is closely associated with accumulation of proline in roots. External application of proline markedly inhibited root growth of rice seedlings in the absence of Cd. Ionically bound, but not soluble, peroxidase activity in roots was increased by CdCl2. Proline treatment also resulted in an increase in ionically bound peroxidase activity in roots. The relationship between growth inhibition of roots induced by Cd and changes of proline level and peroxidase activity is discussed.Abbreviations POX peroxidase  相似文献

Rice (Oryza sativa L.) seedlings stressed with CdCl2 (0.5 mM or 50 μM) showed typical Cd toxicity (leaf chlorosis, decrease in chlorophyll content, or increase in H2O2 and malondialdehyde contents). Rice seedlings pretreated with heat shock at 45°C (HS) for 2 or 3 h were protected against subsequent Cd stress. Rice seedlings pretreated with HS had similar Cd concentration in leaves caused by CdCl2 as those non-HS. The content of H2O2 increased in leaves 1 h after HS exposure. However, APX and GR activities were higher in HS-treated leaves than their respective control, and it occurred after 2 h of HS treatment. Pretreatment of rice seedlings with H2O2 under non-HS conditions resulted in an increase in APX, GR, and CAT activities and protected rice seedlings from subsequent Cd stress. HS-induced H2O2 production and protection against subsequent Cd stress can be counteracted by imidazole, an inhibitor of NADPH oxidase complex. Results of the present study suggest that early accumulation of H2O2 during HS signals the increase in APX and GR activities, which in turn prevents rice seedlings from Cd-caused oxidative damage.  相似文献

The role of hydrogen peroxide (H(2)O(2)) in abscisic acid (ABA)-induced anthocyanin accumulation in detached and intact leaves of rice seedlings was investigated. Treatment with ABA resulted in an accumulation of anthocyanins in detached rice leaves. Dimethylthiourea, a chemical trap for H(2)O(2), was observed to be effective in inhibiting ABA-induced accumulation of anthocyanins. Inhibitors of NADPH oxidase (diphenyleneiodonium chloride and imidazole), phosphatidylinositol 3-kinase (wortmannin and LY 294002), and a donor of nitric oxide (N-tert-butyl-alpha-phenylnitrone), which have previously been shown to prevent ABA-induced H(2)O(2) accumulation in detached rice leaves, inhibited ABA-induced anthocyanin increase. Exogenous application of H(2)O(2), however, was found to increase the anthocyanin content of detached rice leaves. In terms of H(2)O(2) accumulation, intact (attached) leaves of rice seedlings of cultivar Taichung Native 1 (TN1) are ABA sensitive and those of cultivar Tainung 67 (TNG67) are ABA insensitive. Upon treatment with ABA, H(2)O(2) and anthocyanins accumulated in leaves of TN1 seedlings but not in leaves of TNG67. Our results, obtained from detached and intact leaves of rice seedlings, suggest that H(2)O(2) is involved in ABA-induced anthocyanin accumulation in this species.  相似文献

Two contrasting rice (Oryza sativa L.) cultivars, i.e. Wuyujing 3 (WYJ3, Cd-tolerant) and Shanyou 63 (SY63, Cd-sensitive), were grown on a red soil (Ultisol) to study both individual and combined phytotoxicity of arsenic (As) and cadmium (Cd) in terms of Cd and As availability, their uptake and accumulation, antioxidant defense activity and oxidative damage. The antioxidant defense system examined in this study included enzymatic and non-enzymatic molecular antioxidants such as superoxide dismutase (SOD), peroxidase (POD), glutathione (GSH) and ascorbic acid (AsA). Results showed that As or Cd treatment decreased root and shoot biomass in both cultivars compared with their corresponding control (no Cd or As treatment), although less severe inhibition of plant growth was observed in WYJ3 than in SY63. Moreover, rice growth was inhibited more severely by Cd treatment than by As treatment, which could be explained by the higher amount of available Cd (60%) (0.1 M HCl-extractable Cd) compared to the lower amount of available As (15%) (0.5 M NaH2PO4-extractable As) in their postharvest soils. However, shoot biomass in cultivar SY63, and root and shoot biomass in cultivar WYJ3 were significantly higher in the As plus Cd treatment than in the Cd treatment alone, showing that the combined toxicity of these two heavy metals was not additive and on the contrary, As mitigated Cd-induced growth inhibition. The As plus Cd treatment also significantly decreased As or Cd concentrations both in roots and in shoots of the two rice cultivars compared with the As or Cd treatment alone, respectively. On the other hand, treatment with As or Cd alone significantly decreased the SOD and POD activities, and GSH and AsA concentrations, while the activities of these enzymes and the concentrations of GSH and AsA were significantly higher in the As plus Cd treatment than in the Cd treatment alone, resulting in less severe oxidative damage as indicated by the lower concentration of MDA in the As plus Cd treatment (P < 0.05). However, no significant difference was observed in the antioxidant defense activity between the As plus Cd treatment and the As treatment alone. These results suggest that the combined toxicity of As and Cd in rice is lower than that of individual Cd or As, which might be attributed to the decreased uptake and accumulation of Cd and As, and the less oxidative stress caused by the interactive effects of As with Cd both in rhizosphere and in plants.  相似文献

Plants experience low phosphorus (P) and high iron (Fe) levels in acidic lowland soils that lead to reduced crop productivity. A better understanding of the relationship between these two stresses at molecular and physiological level will lead to development of suitable strategies to increase crop productivity in such poor soils. Tolerance for most abiotic stresses including P deficiency and Fe toxicity is a quantitative trait in rice. Recent studies in the areas of physiology, genetics, and overall metabolic pathways in response to P deficiency of rice plants have improved our understanding of low P tolerance. Phosphorous uptake and P use efficiency are the two key traits for improving P deficiency tolerance. In the case of Fe toxicity tolerance, QTLs have been reported but the identity and role played by underlying genes is just emerging. Details pertaining to Fe deficiency tolerance in rice are well worked out including genes involved in Fe sensing and uptake. But, how rice copes with Fe toxicity is not clearly understood. This review focuses on the progress made in understanding these key environmental stresses. Finally, an opinion on the key genes which can be targeted for this stress is provided.  相似文献

In order to clarify the effect of abscisic acid (ABA) on anaerobic tolerance in alfalfa ( Medicago sativa L.), the seedlings were subjected to anaerobic stress after pretreatment with ABA. At concentrations> 1 μ M , ABA pretreatment increased the root viability of the seedlings to anaerobic stress and the viability increased with increasing ABA doses. At 100 μ M ABA, the viability was 2.5-fold greater as compared with that of control seedlings. Roots of the seedlings rapidly lost ATP under the anaerobic stress; however, the decrease in ATP was much slower in the ABA-pretreated seedlings than the control seedlings. At 12 h after onset of the stress, ATP concentrations in the roots of 100 and 10 μ M ABA-pretreated seedlings were 2.7- and 2.0-fold that of the control seedlings, respectively. During the period of ABA pretreatment under aerobic condition, ABA increased alcohol dehydrogenase (ADH, EC 1.1.1.1) activity in the roots until 12 h and then leveled off. The maximum ADH activities were 4.3- and 2.8-fold that in the roots of the control seedlings for 100 and 10 μ M ABA-pretreated seedlings, respectively. After being subjected to the anaerobic stress, both ADH activities in the roots of the ABA-pretreated and the control seedlings increased but the differences in their activity remained. These results suggest that ABA pretreatment may maintain ATP level due to induction of ADH activity, which may be one of the causes of increasing anaerobic tolerance in the seedlings.  相似文献

Cadmium (Cd) is one of the most dangerous environmental pollutants, among other things, affecting plant mineral composition. Thus, in this study, we investigated the changes in potassium (K) concentration in Cd-treated rice (Oryza sativa L.) seedlings of two cultivars. On treatment with 5 μM CdCl2, the Cd concentration increased in the shoot and roots of Cd-sensitive cultivar (cv. Taichung Native 1, TN1) but not or slightly in the Cd-tolerant cultivar (cv. Tainung 67, TNG67). The decrease in K concentration in the shoot and roots of TN1 caused by Cd was more pronounced than that of TNG67. Exogenous addition of KCl decreased Cd concentration and reduced Cd toxicity of TN1 seedlings. Evidence presented in this study suggests that the improvement of K status is able to reduce toxicity of rice seedlings to CdCl2.  相似文献

Rice ( Oryza sativa L.) seeds were soaked for 18 h in distilled water in the absence (–PBZ) or presence (+PBZ, a triazole) of 100 mg l−1 paclobutrazol and then air dried. These air-dried seeds were germinated in the dark and then cultivated in a Phytotron. Twelve-day-old –PBZ and +PBZ seedlings were treated or not with CdCl2. Cd toxicity was judged by the decrease in biomass production, decrease in chlorophyll and protein content, increase in NH4+ content and induction of oxidative stress. The results indicated that PBZ applied to seeds was able to protect rice seedlings from Cd toxicity. On treatment with CdCl2, the abscisic acid (ABA) content increased in +PBZ leaves, but not in –PBZ leaves. The decrease in the transpiration rate of –PBZ seedlings by CdCl2 was less than that of +PBZ seedlings. Exogenous application of the ABA biosynthesis inhibitor, fluridone (Flu), reduced ABA accumulation, increased the transpiration rate and Cd content, and decreased the Cd tolerance of +PBZ seedlings. The effects of Flu on the Cd toxicity, transpiration rate and Cd content were reversed by the application of ABA. It seems that the PBZ-induced Cd tolerance of rice seedlings is mediated through an accumulation of ABA.  相似文献

Salicylic acid (SA) is an important plant hormone, and its exogenous application can induce tolerance to multiple environmental stresses in plants. In this study, we examine the potential involvement of endogenous SA in response to chilling in cucumber (Cucumis sativus L.) seedlings. A low temperature of 8 °C induces a moderate increase in endogenous SA levels. Chilling stimulates the enzymatic activities and the expression of genes for phenylalanine ammonia-lyase (PAL) and benzoic acid-2-hydroxylase rather than isochorismate synthase. This indicates that the PAL enzymatic pathway contributes to chilling-induced SA production. Cucumber seedlings pretreated with SA biosynthesis inhibitors accumulate less endogenous SA and suffer more from chilling damage. The expression of cold-responsive genes is also repressed by SA inhibitors. The reduction in stress tolerance and in gene expression can be restored by the exogenous application of SA, confirming the critical roles of SA in chilling responses in cucumber seedlings. Furthermore, the inhibition of SA biosynthesis under chilling stress results in a prolonged and enhanced hydrogen peroxide (H2O2) accumulation. The application of exogenous SA and the chemical scavenger of H2O2 reduces the excess H2O2 and alleviates chilling injury. In contrast, the protective effects of SA are negated by foliar spraying with high concentrations of H2O2 and an inhibitor of the antioxidant enzyme. These results suggest that endogenous SA is required in response to chilling stress in cucumber seedlings, by modulating the expression of cold-responsive genes and the precise induction of cellular H2O2 levels.  相似文献

Cadmium (Cd) toxicity of rice (Oryza sativa L. cv. Taichung Native 1) seedlings was evaluated by the decrease in chlorophyll content and the increase in malondialdehyde (MDA) in the second leaves of rice seedlings. CdCl2 (5 μM) treatment was accompanied by a decrease in the contents of ascorbic acid (AsA) and AsA + dehydroascorbate (DHA) and in the ratios of AsA/DHA in leaves. However, CdCl2 treatment resulted in an increase in DHA content in leaves. Moreover, the decrease in AsA content was prior to the occurrence of chlorosis and associated with the increase in MDA content in the leaves of seedlings treated with Cd. Pretreatment with 0.5 mM AsA or l-galactono-1,4-lactone (GalL), the biosynthetic precursor of AsA, for 6 h resulted in an increase in the contents of AsA and reduced glutathione (GSH), the ratios of AsA/DHA and GSH/oxidized glutathione, and the activities of ascorbate peroxidase (APX) and glutathione reductase (GR) in the leaves of rice seedlings. Quantitative RT-PCR was applied to quantify the mRNA levels for OsAPX and OsGR genes from rice leaves to examine the effect of AsA or GalL pretreatment on the expression of OsAPX and OsGR genes in rice leaves. The expression of OsAPX2, OsAPX3, OsAPX4, OsAPX5, OsAPX6, OsAPX7, and OsGR1 was increased by AsA or GalL pretreatment. Rice seedlings pretreated with AsA or GalL were observed to reduce the subsequent Cd-induced toxicity. Our results suggest that AsA content may play a role in regulating Cd toxicity of rice seedlings.  相似文献

The effect of cadmium on growth parameters of seedlings of maize, rye and wheat as well as the role of phytochelatins in Cd detoxication in these species were studied. Cadmium was found to inhibit root growth and decrease fresh weight and water content in roots and shoots of the studied plants. Although a considerably lower Cd accumulation was shown in maize seedlings than in other species, they were characterized by the highest sensitivity to cadmium. Among γ-Glu-Cys peptides synthetized by plant species, phytochelatins — glutathione derivatives predominated. In maize they were synthetized in amounts sufficient for binding the total pool of the metal taken up, and the detoxication mechanism was localized in their roots. Larger amounts of cadmium were accumulated in roots of wheat and rye, but the quantity of the formed γ-Glu-Cys peptides seems insufficient for detoxication of the metal.  相似文献

A hydroponic experiment was carried out to study the physiological mechanisms of N-acetyl cysteine (NAC) in mitigating cadmium (Cd) toxicity in two barley (Hordeum vulgare L.) genotypes, Dong 17 (Cd-sensitive) and Weisuobuzhi (Cd-tolerant). Addition of 200 μM NAC to a culture medium containing 5 μM Cd (Cd + NAC) markedly alleviated Cd-induced growth inhibition and toxicity, maintained root cell viability, and dramatically depressed O 2 ·? and ·OH, and malondialdehyde accumulation, significantly reduced Cd concentration in leaves and roots, especially in the sensitive genotype Dong 17. External NAC counteracted Cd-induced alterations of certain antioxidant enzymes, e.g., brought root superoxide dismutase and glutathione reductase, leaf/root peroxidase and glutathione peroxidase activities of the both genotypes down towards the control level, but elevated Cd-stress-depressed leaf catalase in Dong 17 and root ascorbate peroxidase activities in both genotypes. NAC counteracted Cd-induced alterations in amino acids and microelement contents. Furthermore, NAC significantly reduced Cd-induced damage to leaf/root ultrastructure, e.g. the shape of chloroplasts in plants treated with Cd + NAC was relatively normal with well-structured thylakoid membranes and parallel pattern of lamellae but less osmiophilic plastoglobuli compared with Cd alone treatment; nuclei of root cells were better formed and chromatin distributed more uniformly in both genotypes. These results suggested that under Cd stress, NAC may protects barley seedlings against Cd-induced damage by directly and indirectly scavenging reactive oxygen species and by maintaining stability and integrity of the subcellular structure.  相似文献

Carbohydrate metabolism in growing rice seedlings under arsenic toxicity   总被引:7,自引:0,他引:7  
We studied in the seedlings of two rice cultivars (Malviya-36 and Pant-12) the effect of increasing levels of arsenic in situ on the content of sugars and the activity of several enzymes of starch and sucrose metabolism: alpha-amylase (EC 3.2.1.1), beta-amylase (EC 3.2.1.2), starch phosphorylase (EC 2.4.1.1), acid invertase (EC 3.2.1.26), sucrose synthase (EC 2.4.1.13) and sucrose phosphate synthase (EC 2.4.1.14). During a growth period of 10-20 d As2O3 at 25 and 50 microM in the growth medium caused an increase in reducing, non-reducing and total soluble sugars. An increased conversion of non-reducing to reducing sugars was observed concomitant with As toxicity. The activities of alpha-amylase, beta-amylase and sucrose phosphate synthase declined, whereas starch phosphorylase, acid invertase and sucrose synthase were found to be elevated. Results indicate that in rice seedlings arsenic toxicity causes perturbations in carbohydrate metabolism leading to the accumulation of soluble sugars by altering enzyme activity. Sucrose synthase possibly plays a positive role in synthesis of sucrose under As-toxicity.  相似文献

Ascorbic acid (AsA) is the most abundant antioxidant in plants and plays a role in responding to oxidative stress. It has been shown that AsA plays a role in protecting against abiotic stresses. Rice seedlings stressed with 5 μM CdCl2 showed typical Cd toxicity (chlorosis and increase in malondialdehyde content). Rice seedlings pretreated with heat shock at 45°C (HS) or H2O2 under non-HS conditions resulted in the increase in ascorbic acid (AsA) content and the AsA/dehydroascorbate ratio in rice leaves. Exogenous application of AsA or L-galactonone-1, 4-lactone (GalL), a biosynthetic precursor of AsA, under non-HS conditions, which resulted in an increase in AsA content in leaves, enhanced subsequent Cd tolerance of rice seedlings. Pretreatment with imidazole, an inhibitor of NADPH oxidase, under HS conditions significantly decreased H2O2 and AsA contents in leaves and reduced subsequent Cd tolerance of rice seedlings. We also observed that pretreatment with lycorine, which is known to inhibit the conversion of GalL to AsA, significantly inhibited HS-induced AsA accumulation in leaves and reduced HS-induced protection against subsequent Cd stress of rice stress. It appears that HS- or H2O2-induced protection against subsequent Cd stress of rice seedlings is mediated through AsA. The time-course analyses of HS in rice seedlings demonstrated that the accumulation of H2O2 preceded the increase in AsA. Based on the data obtained in this study, it could be concluded that the early accumulation of H2O2 during HS signals the increase in AsA content, which in turn protects rice seedlings from oxidative damage caused by Cd.  相似文献
