The optimum temperature for multiplication of legionella strains in culture media is around 37°C. The effect of high temperatures on the growth of strains isolated from various environments is poorly known. We studied the growth (cell multiplication, respiration) of clinical and environmental Legionella pneumophila strains in liquid media at intervals of 0.5°C in the temperature range from 41.6 to 51.6°C using a temperature gradient incubator. Cell multiplication and CO2 production decreased markedly with all the strains at temperatures above 44–45°C. CO2 continued to be produced up to 51.6C even if cell multiplication generally stopped at around 48.4–50.0C. Thus, legionella retained its metabolic activity beyond the maximum temperature for cell multiplication. The CO2 production per bacterial cell (metabolic quotient, qCO2) increased with increasing temperature up to 45°C, whereafter it decreased, the turning point being almost at the same at which the rate of cell multiplication decreased. The difference in qCO2 between the strains may reflect their different physiological capacities for tolerating high temperatures.  相似文献

The role of high light stress in a natural environment was studied on spinach plants ( Spinacia oleracea L. cv. Wolter) grown in the field during the winter season. Fluorescence induction (at 293 K and 77 K) of leaves was used to characterize the stress effects. Night frost with minimum temperatures between – 1.5°C and –7.5°C (i.e. above the'frost killing point'at ca. –11.5°C) led to impaired photosynthesis. This was seen as increased initial fluorescence (Fo), decreased ratio of variable to maximum fluorescence (FV/FM) and lowered rates of O2 evolution. The freezing injury was reversible within several frostless days. Exposure to high light (about 900 mol m–2 s–1) at chilling temperatures in the field caused photoinhibition, manifested as decreased variable fluorescence (FV) and FV/FM ratio without changes in FO. The photoinhibitory fluorescence quenching was not stronger after frost than after frostless nights; synergism between light stress and preceding freezing stress was not observed. Fluorescence induction signals at 77 K showed that FV of photosystems I and II decreased to the same extent, indicating increased thermal deactivation of excited chlorophyll. Photoinhibition was fully reversible at +4°C within 1 h in low light, but only partially in moderate light. Preceding night frosts did not affect the recovery. The photoinhibition observed here is regarded as a protective system of thermal dissipation of excess light energy.  相似文献

The effects of 80% oxygen–20% carbon dioxide (O2–CO2) and 80% nitrogen–20% carbon dioxide (N2–CO2) atmospheres were compared with respect to the microbial and sensory characteristics of vacuum skin-packaged grain-fed beef steaks stored at −1 and 4 °C. In both N2–CO2 and O2–CO2 atmospheres, lactobacilli were predominant over Brochothrix , pseudomonads, enterobacteria and yeasts and moulds. The results of the current investigation showed that the O2–CO2 atmospheres did not yield total viable counts in excess of 105 cfu cm−2 on beef steaks after 4 weeks of storage. However, the sensory analysis and thiobarbituric acid (TBA) values (as a measure of oxidative rancidity) of the products were unacceptable at this time. In contrast, the N2–CO2 atmospheres yielded maximum total viable counts of approximately 107 cfu cm−2 and the sensory analysis and TBA values of the product were judged to be acceptable after 4 weeks of storage at −1 °C. These results indicate that sensory effects of the product were influenced to a greater extent by the chemical effects of high concentration of O2 on rancidity than by the high levels of lactobacilli.  相似文献

Five psychrophilic sulphate-reducing bacteria (strains ASv26, LSv21, PSv29, LSv54 and LSv514) isolated from Arctic sediments were examined for their adaptation to permanently low temperatures. All strains grew at −1.8°C, the freezing point of sea water, but their optimum temperature for growth ( T opt) were 7°C (PSv29), 10°C (ASv26, LSv54) and 18°C (LSv21, LSv514). Although T opt was considerably above the in situ temperatures of their habitats (−1.7°C and 2.6°C), relative growth rates were still high at 0°C, accounting for 25–41% of those at T opt. Short-term incubations of exponentially growing cultures showed that the highest sulphate reduction rates occurred 2–9°C above T opt. In contrast to growth and sulphate reduction rates, growth yields of strains ASv26, LSv54 and PSv29 were almost constant between −1.8°C and T opt. For strains LSv21 and LSv514, however, growth yields were highest at the lowest temperatures, around 0°C. The results indicate that psychrophilic sulphate-reducing bacteria are specially adapted to permanently low temperatures by high relative growth rates and high growth yields at in situ conditions.  相似文献

The effects of applying Lactobacillus plantarum and Lact. amylovorus at ensiling on wheat silage stored at 25 and41 °C was studied under laboratory conditions. The inoculants were applied at 106 cfu g−1.Silages with no additives served as controls. Three jars per treatment were sampled on days 2, 8 and 60 after ensiling, for chemical and microbiological analyses. After the ensiling period, the silages were subjected to an aerobic stability test. The control and Lact. plantarum inoculated wheat fermented faster at 25 than at 41 °C, whereas silages inoculated with Lact. amylovorus fermented faster at 41 °C. This was apparent from the rate of pH decrease and from the contents of residual sugars and lactic acid in the final silages. The numbers of lactobacilli in the control and Lact. plantarum silages at 41 °C after 2 and 8 days of ensiling were lower than in the corresponding silages at 25 °C. For the Lact. amylovorus silage the opposite held true. The control silages at both temperatures and the Lact. plantarum silage at 41 °C were the most stable silages under aerobic exposure.  相似文献

We investigated the thermal acclimation of photosynthesis and respiration in black spruce seedlings [ Picea mariana (Mill.) B.S.P.] grown at 22/14 °C [low temperature (LT)] or 30/22 °C [high temperature (HT)] day/night temperatures. Net CO2 assimilation rates ( A net) were greater in LT than in HT seedlings below 30 °C, but were greater in HT seedlings above 30 °C. Dark and day respiration rates were similar between treatments at the respective growth temperatures. When respiration was factored out of the photosynthesis response to temperature, the resulting gross CO2 assimilation rates ( A gross) was lower in HT than in LT seedlings below 30 °C, but was similar above 30 °C. The reduced A gross of HT seedlings was associated with lower needle nitrogen content, lower ribulose 1·5-bisphosphate carboxylase/oxygenase (Rubisco) maximum carboxylation rates ( V cmax) and lower maximum electron transport rates ( J max). Growth treatment did not affect V cmax :  J max. Modelling of the CO2 response of photosynthesis indicated that LT seedlings at 40 °C might have been limited by heat lability of Rubisco activase, but that in HT seedlings, Rubisco capacity was limiting. In sum, thermal acclimation of A net was largely caused by reduced respiration and lower nitrogen investments in needles from HT seedlings. At 40 °C, photosynthesis in LT seedlings might be limited by Rubisco activase capacity, while in HT seedlings, acclimation removed this limitation.  相似文献

Mean values ±95% CL of the upper incipient (TIL) and ultimate (TUL) lethal temperatures, determined at five acclimation temperatures ( TA ), increased for TIL from 19.2 ± 0.4° C ( TA 0.5° C) to 21.0 ± 0.4° C ( TA 20° C), and for TUL from 22.6 ± 0.1° C ( TA 0.5° C) to 26.6 ± 0.4° C ( TA 20° C). Mean values were close to those obtained for Arctic charr alevins from Windermere (north-west England). These comparative data for alevins, and previous data for 0+ year parr, indicate negligible geographical variation in the thermal limits of Arctic charr.  相似文献

The effect of temperature (1–34 °C ) on the maximum specific growth rate of Aeromonas salmonicida could not be described by the classical growth models ; for some strains, two optimal temperatures at 23 °C and 30 °C were observed, as well as an unexpected increase in the pseudolag time above 27 °C. This could be explained by the presence of two subsets, notably S-layer+ and S-layer sub-populations. The A cells had higher growth parameters (Topt and μopt) than the A+ cells and were selected by subcultures above 30 °C. Yet the relative proportion of A+ cells did not explain all the variation of μmax versus temperature, and the growth kinetics of an Aer. salmonicida isolate remained unpredictable.  相似文献

The effect of temperature and humidity on SO2–induced photosynthetic depression was determined in gas exchange experiments with leaves of Vicia faba , L. Stomatal behaviour was sensitive to humidity resulting in higher uptake rates of SO2 and stronger reductions of photosynthesis at low VPD (vapour pressure deficit). After a fumigation period of 2 h, when the photosynthetic rate had stabilized, photosynthesis of leaves exposed to SO2 at 8°C was reduced much more than at 18°C at the same rate of SO2 uptake. Data analysis with a mechanistic model revealed that this effect was due to the slower rate of S(IV) oxidation at lower temperatures, resulting in higher accumulation of S(IV) and thus stronger reduction of photosynthesis. These results were confirmed by experimental analyses of the S(IV) concentration in leaves following fumigation, which showed that more S(IV) accumulated in leaves exposed at a lower temperature. This may explain the high sensitivity of plants exposed to SO2 under winter conditions, when both VPD and temperature are low.  相似文献

Photosynthetic characteristics of leaves of tef, Eragrostis tef (Zucc.) Trotter, plants, grown at 25/15°C (day/night), were measured at temperatures from 18 to 48°C. The highest carbon exchange rates (CER) occurred between 36 and 42°C. and averaged 27 μmol m−2 s−1. At lower or higher temperatures, CER was reduced, but the availability of CO2 to the mesophyll, measured as internal CO2 concentration, was highest when temperatures were above or below the optimum for CER. In addition, CER and stomatal conductance were not correlated, but residual conductance was highly correlated with CER (r = 0.98). In additional experiments, relative 13C composition for leaf tissue grown at 25, 35 and 45°C averaged -14.4 per mille, confirming that tef is a C4 grass species. Dry matter accumulation was higher at 35 than at 25, and lowest at 45°C. Leaf CER rates increased hyperbolically with increased light when measured from 0 to 2000 μmol m−2 s−1 PPFD. The highest CER, 31.8 μ-mol m-2 s−1, occurred at 35°C and 2000 μmol m−2 s−1 PPFR. At high light, CER at 25 and 35°C were nearly equal because of higher stomatal conductance at 25°C. Residual conductance was, however, clearly highest at 35°C compared to 25 and 45°C treatments. Stomatal conductance and residual conductance were not correlated in either set of experiments, yet residual conductance was always highest when temperatures were between 35 and 42°C across experiments, suggesting that internal leaf photosynthetic potential was highest across that temperature range.  相似文献

The reasons for the decline in net CO2 assimilation ( A ) above its thermal optimum are controversial. We tested the hypothesis that increasing the ratio of Rubisco activase to Rubisco catalytic site concentration would increase the activation state of Rubisco at high temperatures. We measured photosynthetic gas exchange, in vivo electron transport ( J ) and the activation state of Rubisco between 15 and 45 °C, at 38 and 76 Pa ambient CO2, in wild-type (WT) and anti- rbc S tobacco. The Rubisco content of the anti- rbc S lines was 30% (S7-1) or 6% (S7-2) of WT, but activase levels were the same in the three genotypes. Anti- rbc S plants had lower A than WT at all temperatures, but had a similar thermal optimum for photosynthesis as WT at both CO2 levels. In WT plants, Rubisco was fully activated at 32 °C, but the activation state declined to 64% at 42 °C. By contrast, the activation state of Rubisco was above 90% in the S7-1 line, between 15 and 42 °C. Both A and J declined about 20% from T opt to the highest measurement temperatures in WT and the S7-1 line, but this was fully reversed after a 20 min recovery at 35 °C. At 76 Pa CO2, predicted rates of RuBP regeneration-limited photosynthesis corresponded with measured A in WT tobacco at all temperatures, and in S7-1 tobacco above 40 °C. Our observations are consistent with the hypothesis that the high temperature decline in A in the WT is because of an RuBP regeneration limitation, rather than the capacity of Rubisco activase to maintain high Rubisco activation state.  相似文献

S. CONDÓN, A. PALOP, J. RASO AND F.J. SALA. 1996. The influence of the incubation temperature on the estimated heat resistance for survivors after heat treatment was investigated. The survival curves and the D t values of spores of Bacillus subtilis heated at different temperatures in pH 7 buffer, obtained after incubating survivors at different temperatures (30, 37, 44 or 51°C), were compared. The incubation temperature influenced the profile of survival curves. Lower incubation temperatures led to bigger D t values and longer shoulders. D t values obtained after incubating at 30°C were higher (x3 approx.) than those obtained by incubating at 51°C. The incubation temperature did not modify z values ( z = 9.1). These results show that shoulders are not only due to the activation of dormant spores but also to heat damage repair mechanisms. From the profile of survival curves at different incubation temperatures it would seem that heat damage is accumulative. Cells can repair the initial heat injury, but the accumulation of injuries would eventually make the damage irreversible.  相似文献

Abstract.  The influence of temperature on the insect cell line, BmN, derived from the silkworm, Bombyx mori is investigated. These cells proliferate at an accelerated pace as the temperature increases from 22 to 30 °C, but the growth rate slows at 34 °C, and proliferation stops at 38 °C. At high temperatures, abnormal cellular morphology is observed. Cells treated at 38 °C have cytoplasmic bilateral protrusions and they gradually aggregate and float in the medium. BmN cells without proliferation at 38 °C are viable but have reduced DNA synthesis. At high temperatures, the cell cycle of BmN cells halts at the G2 phase. After heat treatment of the larvae, an accumulation of larval haemocytes with high DNA content is found, which suggests that the cell cycle arrest at G2 also occurs in the silkworm at high temperatures.  相似文献

Protoplasts were enzymically isolated from 2-week-old non-acclimated rye ( Secale cereale L. cv. Puma) seedlings. They were resuspended in isotonic sorbitol with different concentrations (0–10%) of dimethyl sulfoxide (DMSO). The survival of the protoplasts frozen in isotonic sorbitol solutions declined at temperatures below the freezing point with the LT50 being -8°C. Addition of DMSO to the osmoticum increased survival at freezing temperatures. The optimum concentration of DMSO was 4% and lowered the LT50 to -19°C. Freeze-fracture studies of the plasma membrane revealed aparticulate lipid lamellae at -4°C, but the first appearance of lateral phase separations, striations and inverted cylindrical micelles (hexagonal11-type structures) occurred at -6°C. At lower temperatures, -8 and -10°C, the occurrence of nonbilayer structures became more common. The addition of DMSO decreased the incidence of the ultrastructural changes. With 2 or 4% DMSO, non-bilayer structures were not observed at temperatures above -10°C. Instead, striations and H11-type structures were observed at - 15 and -20°C.  相似文献

Abstract Thymidine, leucine and acetate incorporation into soil bacterial communities extracted from two different soils using homogenisation-centrifugation were measured at different temperatures (0–28°C). Similar effects of temperature were found for both soils used. Optimum temperatures for incorporation of acetate into lipids were found between 20 and 24°C, while the incorporation of thymidine and leucine into cold acid insoluble material increased with temperature. A good fit to the square root model (Ratkowsky model) was found for all three methods, when only data below optimum was considered for the acetate incorporation. The apparent Tmin calculated from this model was −8.4 ± 0.77°C for thymidine incorporation. Tmin for acetate incorporation was slightly higher. Leucine incorporation had significantly higher Tmin (−6.0 ± 0.62°C), and the Q10 between 0 and 10°C was also higher than for the two other measurements. This resulted in a leucine/thymidine incorporation ratio which increased from 0°C up to about 15°C, but remained constant at temperatures above 15°C. The amount of leucine incorporated into hot acid insoluble material (protein) as a percentage of that incorporated into cold acid insoluble material (total macromolecules) was also constant above 15°C (about 40%), but decreased at lower temperatures to less than 25%. No effects were found of temperature on non-specific incorporation of thymidine into macromolecules other than DNA, or acetate incorporation into different lipid fractions (neutral, glyco- and polar lipids). The fact that the temperature relationships for soil bacterial communities appeared to follow the square root model will facilitate comparisons of such relationships between different soils, as well as recalculation of data to actual field temperatures.  相似文献

Picea sitchensis (Bong.) Carr. seedlings were exposed to SO2, NO2 and SO2+ NO2 during dormancy in controlled environments, and were taken to night temperatures of 4, 0, −5, −10 and −15 °C in a freezer. Conditions in the freezer were carefully monitored during the low–temperature treatments. In two experiments, different photoenvironments and temperature regimes were imposed prior to the cold treatments, and different effects were observed. In the first, only limited frost hardiness was achieved and night temperatures of −15 °C were lethal. Temperatures of −5 and − 10 °C led to poor survival of lateral buds, particularly in plants exposed to 45 ppb SO2. The poor bud break in plants exposed to SO2 and to − 5 °C resulted in a loss of the effectiveness of this temperature as a chill requirement. Pressure-volume analysis showed that the shoots of plants exposed to NO2 had greater elasticity (lower elastic moduli, e), so that loss of turgor occurred at lower relative water contents. In contrast, a hardening period (2 weeks in night/day temperatures of 3/10 °C and 8 h days at 50 μmol m−2 s−1 PAR) gave decreased elasticity and lower solute potentials of spruce shoots. In the second experiment, exposure to 30 ppb SO2 and SO2+ NO2 led to slight, but consistent, increases in frost injury to the needles of plants frozen to − 5 and − 10 °C. The results suggest that the main interaction of low temperatures and winter pollutants may be on bud survival rather than on needle damage, but that effects are subtle, only occurring with certain combinations of pollutant dose and cold treatment.  相似文献

Abstract The effect of temperature on CH4 production, turnover of dissolved H2, and enrichment of H2-utilizing anaerobic bacteria was studied in anoxic paddy soil and sediment of Lake Constance. When anoxic paddy soil was incubated under an atmosphere of H2/CO2, rates of CH4 production increased 25°C, but decreased at temperatures lower than 20°C. Chloroform completely inhibited methano-genesis in anoxic paddy soil and lake sediment, but did not or only partially inhibit the turnover of dissolved H2, especially at low incubation temperatures. Cultures with H2 as energy source resulted in the enrichment of chemolithotrophic homoacetogenic bacteria whenever incubation temperatures were lower than 20°C. Net photosynthetic CO2 exchange of 1-year-old plants of Coffea arabica L. was studied after the above-ground parts had been exposed once or repeatedly to night temperatures in the chilling range. Chill-reduced rates of CO2 uptake (measured at 24°C and at natural CO, level) were observed after a 12 h night exposure to about 6°C. After exposure to 4°C, activity was reduced to less than half of that of the controls, and after exposure to 0.5°C the leaves suffered visible necrotic injury and were no longer able to take up Co2 If the leaves were not lethally injured, net photosynthesis recovered completely within 2 to 6 days. About 25% of chill-induced reduction of CO2 uptake was due to reduced stomatal aperture and 75% to impairment of carboxylation efficiency.
Chilling on successive nights at 4–6°C reduced CO, uptake progressively on each day following treatment. After 10 nights, activity was decreased to less than 10% of initial performance. Conditioning at temperatures slightly above the chilling level (e.g. 15/I2°C) for 2 weeks led to almost complete impairment of photosynthetic activity without additional chilling stress instead of improving chilling tolerance.  相似文献

Abstract. C4 plants grown in optimum conditions are, by comparison to C3, capable of higher maximum dry-matter yields and greater efficiencies of water and nitrogen use, yet they are rare outside the subtropics. Both latitudinal and altitudinal limits of C4 distributions correlate most closely with a mean minimum temperature of 8-10°C during the period of active growth. The possibility that the C4 process is inherently incapable of functioning at low temperatures is examined. The reversible effects of chilling on the quantum efficiency of C4 photosynthesis and the functioning of the individual steps in the C4 cycle are examined. Chilling also produces an irreversible loss of capacity to assimilate CO2 which is directly proportional to the light received during chilling. It is suggested that the reversible reduction in capacity to assimilate CO2 and the lack of an alternative pathway for the utilization of lightgenerated reducing power may make C4 species more prone to chilling-dependent photoinhibition. Laboratory studies and limited field observations suggest that this damage would be most likely to occur during photosynthetic induction at the temperatures and light levels encountered on clear, cool mornings during the spring and early summer in cool climates. Even those C4 species occurring naturally in cool climates do not appear fully capable of tolerating these conditions; indeed their growth patterns suggest that they may be adapted by avoiding 'rather than enduring' such conditions.  相似文献

The seasonal changes in photosynthetic properties in 1-year-old needles of Sakhalin spruce ( Picea glehnii ) were measured using the chlorophyll fluorescence technique at various temperatures (5, 10, 20, 25 and 30°C). In the course of seasonal change, a temporary decrease in the quantum yield of PSII electron transport (ΦPSII) was observed just before budbreak. A decline in photochemical quenching ( q P) was observed at the same time as that of ΦPSII but only at the two lowest temperatures (5 and 10°C). Photochemical efficiency of open PSII ( F v'/ F m') also declined just before budbreak at 25 and 30°C. An increase in thermal energy dissipation as indicated by a decrease in F v'/ F m' before budbreak was not significant at lower temperatures (5 and 10°C) in spite of the declines in q P. This implies that thermal energy dissipation necessitated by the decline in ΦPSII might not be sufficiently strong to prevent a decline in q P at lower temperatures. On the other hand, at higher temperatures no decline was observed in q P because ΦPSII decreased to a relatively small extent, therefore thermal energy dissipation is sufficient in coping with the excessive energy accumulation in PSII. Seedlings of Sakhalin spruce exposed to ambient air temperature below 10°C before budbreak exhibited photoinhibition indicated by a decrease in the maximal photochemical efficiency of PSII ( F v/ F m) after an overnight dark adaptation. The present study suggests that 1-year-old shoots of Sakhalin spruce have an increased susceptibility to photoinhibition at low temperature just before budbreak.  相似文献
