Abstract By mixed inoculation experiments, the competitive ability of a nifD ::Tn 5 mutant of Bradyrhizobium japonicum was compared to its effective, isogenic parent strain. When the strains were inoculated in a 1:1 ratio at high concentration, the mutant was found to colonize almost as many nodules as the wild type. Thus, lack of expression of a functional nitrogenase system does not severely reduce competitiveness. In such experiments the majority of the nodules (> 60%) were infected by both wild-type and mutant strains. From statistical analysis it was concluded that a mean number of 2–4 bacteria have successfully elicited one nodule under the described conditions. Visual and microscopic observations of sections from mixed infected nodules revealed separated sectors containing effective or ineffective bacteroids.  相似文献

In order to identify physiological components that contribute to salinity tolerance, we compared the effects of Na+, Mg2+ and K+ salts (NaCl, Na2SO4, MgCl2, MgSO4, KCl and K2SO4), Ca2+ (CaSO4), mannitol and melibiose on the wild type and the single-gene NaCl-tolerant mutants stl1 and stl2 of Ceratopteris richardii. Compared with gametophytic growth of the wild type, stl2 showed a low level of tolerance that was restricted to Na+ salts and osmotic stress. stl2 exhibited high tolerance to both Na+ and Mg2+ salts, as well as to osmotic stress. In response to short-term exposure (3 d) to NaCl, accumulation of K+ and Na+ was similar in the wild type and stl1. In contrast, stl2 accumulated higher levels of K+ and lower levels of Na+. Ca2+ supplementation (1.0 mol m?3) ameliorated growth inhibition by Na+ and Mg2+ stress in wild type and stll, but not in stl2. In addition, under Na+ stress (175 mol m?3) wild-type, stll and stl2 gametopbytes maintained higher tissue levels of K+ and lower levels of Na+ when supplemented with Ca2+ (1.0 mol m?3). stl2 gametophytes were extremely sensitive to K+ supplementation. Growth of stl2 was greater than or equal to that of the wild type at trace concentrations of K+ but decreased substantially with increasing K+ concentration. Supplementation with K+ from 0 to 1.85 mol m?3 alleviated some of the inhibition by 75 mol m?3 NaCl in the wild type and in stl1. In stl2, growth at 75 mol m?3 NaCl was similar at 0 and 1.85 mol m?3 K+ supplementation. Although K+ supplementation above 1.85 mol m?3 did not alleviate inhibition of growth by Na+ in any genotype, stl2 maintained greater relative tolerance to NaCl at all K+ concentrations tested.  相似文献

A panel of monoclonal antibodies were generated against the surface polysaccharide antigens of the cell envelope of Salmonella typhi. Four clones (IgM) were specific for the capsular Vi polysaccharide, and one clone (IgG3) reacted selectively with the S. typhi lipopolysaccharide in enzyme immunoassay. On the basis of their reactivity pattern and binding affinity, MATy-V7 (IgM) and MATy-O9 (IgG3) antibodies were selected for further characterization of their antigenic specificity. In an inhibition enzyme immunoassay with rabbit factor-specific anti-Salmonella antibodies as the competing agents, the reactivity of MATy-V7 and MATy-O9 were significantly inhibited by the anti-Vi and anti-O9 antisera, respectively. Moreover, both the Vi- and O9-specific monoclonal antibodies were shown to be useful serotyping agents by correct identification in slide agglutination tests of 32 clinical isolates of all the S. typhi and other serogroup D salmonellae among a total of 140 bacterial isolates representing eight different Enterobacteriaceae genera tested.  相似文献

A modified methylation analysis is described which allows the elucidation of the structure of the inner core region [heptose/3-deoxy-D-manno-2-octulosonic acid (KDO)] of enterobacterial lipopolysaccharides (LPS) of Salmonella minnesota rough mutants (Re, strain R595; and Rd2P-, strain R4). Methylation, carboxyl-reduction, remethylation, hydrolysis, carbonyl-reduction, and acetylation of the Re-mutant LPS yielded the 2,6-di-O-acetyl and 2,4,6-tri-O-acetyl derivatives of partially methylated 3-deoxyoctitol in equimolar amounts, indicating the presence of a terminal and a 4-linked pyranosidic KDO residue. For Rd2P- LPS, the hydrolysis step involved 0.1M trifluoroacetic acid at 100 degrees for 1 h which cleaved ketosidic linkages, and the final products included the foregoing acetyl derivatives in the molar ratio of 1:02 and a partially methylated and acetylated 3-deoxyoctitol derivative which was substituted at O-5 by a methylated heptopyranosyl residue. Trideuteriomethylation of the latter product followed by methanolysis and acetylation gave 5-O-acetyl-3-deoxy-1,7,8-tri-O-methyl-2,4,6-tri-O-trideuteriomethyl++ +-D- glycero-D-talo/galacto-octitol and 1,5-di-O-acetyl-2,3,4,6,7-penta-O-methyl-L-glycero-D-manno-heptitol++ +. These results prove the presence of a (2----4)-linked KDO disaccharide in Re LPS and show that the core region of Rd2P- LPS contains a terminal alpha-L-glycero-D-manno-heptopyranosyl group and a non-substituted, a 4-O-, and a 4,5-di-O-substituted pyranosidic KDO residue in the molar ratios 1:1:0.2:1.  相似文献

Excretion of minerals by the NaCl-resistant and comparatively cadmium-resistant tree Tamarix aphylla (L.) Karst, was investigated. Cd2+ was excreted by plants exposed for 1–10 days to 9 or 45 μ M Cd2+ solutions. Excretion of this toxic ion increased considerably with time but was less than 5% of the quantities that had been accumulated in the shoots. Excretion of Na+ and Cl was positively correlated with NaCl concentration (1.5, 10, 50 m M ) of the medium. The Na+/Cl ratios of the excrete were positively correlated with the concentration of the treatment solution. Ca2+ excretion decreased with increasing NaCl concentrations of the solution. Excretion of K+ and Mg2+ was only little affected by NaCl. Excretion of Li+ occurred whenever this element was supplied in the uptake solution; daily excretion rates of Li+ increased with time. The ecological significance of excretion is discussed in relation to the low selectivity of the mechanism in T. aphylla .  相似文献

Monoclonal antibodies against the lipopolysaccharide (LPS) of the deep rough mutant I-69 Rd/b+ of Haemophilus influenzae were obtained after immunization of mice with sheep erythrocytes which had been coated with de- O -acylated LPS. Characterization of antibodies was performed by enzyme immuno assay (EIA) using LPS or neoglycoconjugates containing partial structures of LPS as solid-phase antigens and by haemagglutination with sheep erythrocytes coated with de- O -acylated LPS. Binding data were confirmed by EIA inhibition experiments using deacylated LPS or synthetic partial structures thereof. Three antibodies were specific for 3-deoxy- d - manno -octulopyranosonic acid- (Kdo) 5-phosphate, one for Kdo-4-phosphate, and one required, in addition to a Kdo-phosphate, parts of the phosphorylated glucosamine backbone of lipid A. All antibodies also bound in (i) Western blots to bacterial whole-cell lysates or isolated LPS separated by SDS–PAGE, (ii) bacterial colony blots, and (iii) immunofluorescence with live bacteria. The latter result indicated that Kdo-4- and Kdo-5-phosphate are synthesized by the bacteria and are not the result of phosphate migration.  相似文献

GM1 ganglioside binding to the crude mitochondrial fraction of rat brain and its effect on (Na+, K+)-ATPase were studied, the following results being obtained: (a) the binding process followed a biphasic kinetics with a break at 50 nM-GM1; GM1 at concentrations below the break was stably associated, while over the break it was loosely associated; (b) stably bound GM1 activated (Na+, K+)-ATPase up to a maximum of 43%; (c) the activation was dependent upon the amount of bound GM1 and was highest at the critical concentration of 20 pmol bound GM1 X mg protein-1; (d) loosely bound GM1 suppressed the activating effect on (Na+, K+)-ATPase elicited by firmly bound GM1; (e) GM1-activated (Na+, K+)-ATPase had the same pH optimum and apparent Km (for ATP) as normal (Na+, K+)-ATPase but a greater apparent Vmax; (f) under identical binding conditions (2 h, 37 degrees C, with 40 nM substance) all tested gangliosides (GM1, GD1a, GD1b, GT1b) activated (Na+, K+)-ATPase (from 26-43%); NeuNAc, sodium dodecylsulphate, sulphatide and cerebroside had only a very slight effect. It is suggested that the ganglioside activation of (Na+-K+)-ATPase is a specific phenomenon not related to the amphiphilic and ionic properties of gangliosides, but due to modifications of the membrane lipid environment surrounding the enzyme.  相似文献

Two monovalent ion porters, the putative Na+/H+ antiporter (NapA) of Enterococcus hirae and the putative K+/H+ antiporter (KefC) of Escherichia coli, are similar in sequence throughout their hydrophobic domains. These two proteins, which comprise a novel family of transporters unrelated to the previously characterized Na+/H+ exchangers of E. coli (NhaA and NhaB) are proposed to function by essentially the same mechanism.  相似文献

The classic compartment analysis of ion efflux from roots is often applied with the assumption that there is a system of 3 compartments in series. However, complex ion transport across the root tissues, as well as influences from the shoot, may complicate the picture. The present experiments were performed to study the immediate effects that excision of the shoot before the experiment exerts on the efflux of Rb+(86Rb+) and of K+(86Rb+) from 9-day-old roots of plants of barley (Hordeum vulgare L. cv. Salve). The efflux from high K+ and low K+ roots of intact and detopped plants were compared. After excision of the shoot of high K+ plants, a marked increase in efflux was observed after 2.5 h with a maximum at about 7 h. The increase in efflux was seen as a peak in plots of efflux versus time. Excision of the shoot from low K+ roots did not give rise to a consistent increase in efflux. Regular K+ ion efflux curves were observed from roots of intact plants of high or low K+ status. Furthermore, after a pulse treatment of 9-day-old roots of intact plants of high or low K+ status with a solution containing Rb+(86Rb+), the Rb+(86Rb+) transport to the shoots was not reduced during the following 3 h in unlabelled solution. It is suggested that both the peak appearing in the efflux plots and the maintained tracer transport to the shoots after transfer of the roots to an unlabelled solution indicate the existence of a K+/Rb+ transport system in the symplasm of the roots that has only a slow exchange with the bulk cytoplasm and vacuoles.  相似文献

Antiserum to the catalytic subunit of goldfish brain (Na+, K+)-ATPase has been employed at the electron microscopic level by means of the peroxidase-antiperoxidase immunohistochemical method. In optic nerve, antigenic sites are restricted to the nodes of Ranvier. No reaction product is detected in underlying internodal neurolemma. Outgrowing neurites for cultured retinal explants devoid of glial ensheathment exhibit a continuous distribution of the enzyme subunit. Antibodies against eel electroplax (Na+, K+)-ATPase cross-react with the goldfish brain enzyme and show a similar immunocytochemical distribution pattern.  相似文献

Shoot activity has been reported to affect rates of ion uptake by plant roots in other ways than merely through supply of assimilates. To elucidate the mechanisms by which a signal from the upper part of the plant controls the rate of K+ and NO3 uptake by roots, both uptake of K+ and NO3 and secretion into the xylem of young sunflower plants ( Helianthus annuus L.) were measured after changes in light intensity.
No close correlation was observed between the uptake of NO3 and that of K+; an increase in light intensity produced a much greater stimulation of NO3 uptake than of K+ uptake. On the other hand, secretion of NO3 into the xylem was tightly coupled to that of K+, and this coupling was strongly disturbed by excision of the root. The results suggest the involvement of the K2-malate shuttle on the regulation by the shoot of K+ and NO3 secretion in the xylem, which is linked to NO3 uptake, while K+ uptake is independent of this regulation mechanism.  相似文献

Tomato growth was examined in solution culture under constant pH and low levels of NH4+ or NO3?. There were five nitrogen treatments: 20 mmoles m?3 NH4+, 50 mmoles m?3 NO3?, 100 mmoles m?3 NH4+ 200 mmoles m?3 NO3?, and 20 mmoles m?3 NH4++ 50 mmoles m?3 NO3?. The lower concentrations (20 mmoles m?3 NH4+ and 50 mmoles m?3 NO3?) were near the apparent Km for net NH4+ and NO3? uptake; the higher concentrations (100 mmoles m?3 NH4+ and 200 mmoles m?3 NO3?) were near levels at which the net uptake of NH4+ or NO3? saturate. Although organic nitrogen contents for the higher NO3? and the NH4++ NO3? treatments were 22.2–30.3% greater than those for the lower NO3? treatment, relative growth rates were initially only 10–15% faster. After 24 d, relative growth rates were similar among those treatments. These results indicate that growth may be only slightly nitrogen limited when NH4+ or NO3? concentrations are held constant over the root surface at near the apparent Km concentration. Relative growth rates for the two NH4+ treatments were much higher than have been previously reported for tomatoes growing with NH4+ as the sole nitrogen source. Initial growth rates under NH4+ nutrition did not differ significantly (P≥ 0.05) from those under NO3? or under combined NH4++ NO3?. Growth rates slowed after 10–15 d for the NH4+ treatments, whereas they remained more constant for the NO3? and mixed NH4++ NO3? treatments over the entire observation period of 24–33 d. The decline in growth rate under NH4+ nutrition may have resulted from a reduction in Ca2+, K+, and/or Mg2+ absorption.  相似文献

We report on the interactions of Li+, a congener of K+ with the (Na+ + K+)-ATPase from E Electricus as measured by their effects on the rate of [3H]-ouabain binding to this enzyme. Like K+, Li+ slows ouabain binding under both Type I (Na+ + ATP) and Type II (P1) conditions, but with lower affinity. In contrast to K+, the Li+ inhibition curve is hyperbolic, suggesting interaction at an uncoupled site. Also differing from the complete inhibition by high K+, a residual ouabain-binding rate persists at high Li+. The interactions of Li+ and K+ are synergistic: the apparent K+ affinity increases 3 to 4-fold in presence of Li+. These results are consistent with the conclusion that Li+ interacts with only one of the two K+ sites and may be of interest in interpreting lithium pharmacology.  相似文献

The effects of phenytoin, a potent antiepileptic drug, on the active transport of cations within membranes remain controversial. To assess the direct effects of phenytoin on the Na+,K+ pump, we studied the drug's influence on the phosphorylation of partially purified (Na+,K+)-ATPase from mouse brain. (Na+,K+)-ATPase subunits were resolved by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Phenytoin, in vitro, decreased net phosphorylation of the (Na+,K+)-ATPase catalytic subunit in a dose-dependent manner (approximately 50% at 10(-4) M). When the conversion of E1-P to E2-P, e.g., the two major phosphorylated conformational states of (Na+,K+)-ATPase, was blocked by oligomycin or N-ethylmaleimide, phenytoin had no effect. The results suggest that phenytoin acts on the phosphatasic component of the reaction cycle, decreasing the phosphorylation level of the enzyme.  相似文献

Previous studies have shown that sloth bears are mainly myrmecophagous, but in this study, 39 and 83 percent (wet and dry season, respectively) of scats surveyed in South India contained fruits. Seeds from three of six plant species collected from scats germinated faster than seeds not passing through bears; germination percentage was not higher in any of the six species. These results suggest that the sloth bear may play an important role in the population dynamics of fleshy‐fruited species of the region.  相似文献

Abstract: The denatured catalytic polypeptide of (Na+, K+)-ATPase of goldfish brain was purified and identified as the 32P-labeled phosphoprotein. The protein served as immunogen for the preparation of rabbit antisera for immunohistochemical application to goldfish tissue sections, using the peroxidase-antiperoxidase indirect method. Labeling in brain cross-sections appears primarily in fibers of the optic nerve layer of the tectum. In optic nerve cross sections, labeling is restricted to fiber bundles.  相似文献
