Quantitative Analysis of Cold Hardening and Dehardening in Lolium   总被引:3,自引:0,他引:3

The change in cold hardiness of three Lolium multiflorum Lam.varieties was followed in plants exposed to hardening or dehardeningconditions at a range of temperatures. Hardening and dehardeningwere analysed as quantitative processes dependent upon temperatureand time. Their time courses changed exponentially to an asymptotewhich was logistically related to temperature. Both componentsof the model were fitted simultaneously. Parameters of biologicalinterest, such as the initial rates of hardening and dehardeningfor a given temperature and the percentage of the process completedin a given time, were derived and compared for varieties ofcontrasting hardiness. The analysis demonstrated the importanceof dehardening in determining hardiness. Similar results wereobtained when the model was applied to hardening and dehardeningdata for Lolium perenne L. The potential of this quantitative analysis for distinguishingbetween possible mechanisms of cold hardiness is discussed andfurther experiments required to characterise the kinetics ofhardening and dehardening more fully are identified. Use of the self-thinning rule to describe size/density compensation(SDC) in defoliated swards is examined. It is shown that defoliationrelated variation in leaf area and associated morphogeneticchanges in plant structure necessitate slope corrections, designatedCa and Cr , respectively. The theory predicts that reduced leafarea in more heavily defoliated swards will result in SDC atslopes more negative than -3/2 (variable leaf area SDC), andthat there will be a transition to -3/2 (constant leaf area)SDC at higher herbage mass. Empirical data from previous experiments with Lolium perenneL. and Medicago sativa L. are examined, and appear to confirmthe theoretical predictions, including the slope change at thepoint of transition from variable to constant leaf area SDC.This transition point, designated di , is subject to interspecificvariation and is related to the mature shoot size of a particularspecies. Vapour from incubated Pinus radiata D. Don litter caused a depressionin the germination of Lolium perenne L. seeds and responsesvarying from stimulation to depression in germination of Trifoliumrepens L. seeds. These effects could be attributed to ethyleneand carbon dioxide. Experiments using ethylene and carbon dioxidesupported this conclusion. Lockyer, D. R. 1985. The effect of sulphur dioxide on the growthof Lolium perenne L., Lolium multiflorum Lam., Dactylis glomerataL., and Phleum pratense L.?J. exp. Bot. 36: 1851-1859. Fouragriculturally important grasses, Lolium perenne L., Loliummultiflorum Lam., Dactylis glomerata L. and Phleum pratenseL. were exposed to sulphur dioxide (SO2) in a system of exposurechambers. The plants were exposed for a total of 43 d to meanconcentrations of SO2 in the air of 0,87 or 448 (µg m–3and herbage was harvested twice. All four grasses showed chloroticlesions after exposure to the highest concentration of SO2.The effect of SO2 on the yield of herbage was statisticallysignificant only at the second harvest and at the highest concentration;the dry weights of shoots of D. glomerata and L. perenne werereduced by 33% and 16% respectively. Significant effects ofSO2 were also found on the 'transpiration coefficients' measuredfor D. glomerata and P. pratense. The grasses differed in theiruptake of sulphur from the atmosphere but this was not relatedto their sensitivity to SO2. Total–S concentration inthe shoots of L. perenne, L. multiflorum and D. glomerata increasedalmost linearly in response to increasing SO2 concentration;with P. pratense only the highest SO2 concentration raised total-Sabove the level in control plants. Mature leaves of Lolium temulentum L. were assayed for sucrosephosphate synthetase activity at different times during thephotoperiod. There was a rapid increase in activity at the onsetof illumination which was not observed in leaves maintainedin darkness. The activity prior to illumination was insufficientto catalyse the rates of sucrose synthesis observed in illuminateddetached leaves; after 15 min illumination the two processeswere of similar magnitude. Initiation and early development of the membranous ligule ofLolium temulentum L. was studied by light and transmission electronmicroscopy. The ligule appeared to be derived solely from theleaf adaxial epidermis and at this stage had a structure andultrastructure typical of a meristematic tissue. Effects of time of initial cut and subsequent cutting interval(1, 2 or 4 weeks) were examined in a glasshouse during summer1988 for reproductive plants of three prairie grass cultivars(Bromus willdenowii Kunth), Westerwolds ryegrass (Lolium multiflorumLam.) and perennial ryegrass (Lolium perenne L.). Measurementswere made of tiller and leaf numbers, sites of tillering, reproductivedevelopment, and herbage quality and yields. Effect of timeof initial cutting on regrowth appeared to be independent ofstage of reproductive development and unrelated to any of themeasured plant parameters. Characteristics for each cultivaras identified during undisturbed growth prior to the initialcuts, were modified by the subsequent cutting frequencies. Perennial ryegrass had the highest yields under frequent cuttingwith high herbage quality. Westenwolds ryegrass and the prairiegrass cultivars ‘Grasslands Matua’ and ‘Primabel’had the highest yields with infrequent cutting, but lower herbagequality than in perennial ryegrass. At each cut, tiller deathin prairie grass was determined by the number of reproductivetillers, and in the ryegrasses also by the numbers of elongatedvegetative tillers. Recovery of tiller numbers was rapid andprimarily from inhibited tiller buds at the base of reproductivetillers. In prairie grass, tiller numbers were relatively unaffectedby cutting frequency, but at frequent cutting, many axillarytillers originated from vegetative tillers rather than frominhibited tiller buds at the base of reproductive tillers. Effects of cutting to 3 or 6 cm stubble height at frequenciesof 1, 2 or 4 weeks were investigated in young, vegetative, spacedplants of prairie grass (Bromus willdenowii Kunth), a tetraploidWesterwolds ryegrass (Lolium multiflorum Lam.) and perennialryegrass (Lolium perenne L.). Plants were grown for 8 weekson nutrient solution in a growth cabinet at day/night temperaturesof 15/10°C and in a short photoperiod. Leaf appearance,site filling and tiller numbers were all reduced by both increasedcutting frequency and a lower cutting height. The extent ofthese reductions was least in prairie grass and greatest inWesterwolds ryegrass. Relative growth rates in all species respondedin a similar manner with more frequent cutting and a lower cuttingheight. Cutting frequency had the greatest effect on growthrates, although the effect of cutting height increased withtime. Results confirm general recommendations of long intervalsbetween defoliations to achieve high yields for vegetative prairiegrass, but the relative amounts of dry matter harvested underthe different cutting regimes may be no different to that obtainedin ryegrass. Stubble height was of lesser importance in determiningyields. A typical U-shaped curve for depletion and recovery in water-solublecarbohydrates occurred after defoliation in all species, butlevels remained low at frequent cutting. Water-soluble carbohydratelevels in stubble and roots were higher in prairie grass. Regrowthat the end of the experiment was highly correlated with totalstubble and root weights (r = 0.84), while regrowth per tillershowed a good correlation with water-soluble carbohydrate content,although the response varied between species. Vegetative plants of the long-day grass Lolium temulentum L.Ceres were exposed to threshold long days or light breaks. Protracteddarkness given just afterwards clearly promoted flowering andwas weakly inductive on its own. Understanding how environmental factors affect the longevityof roots is essential if root mortality linked nutrient cyclingprocesses within ecosystems are to be understood, and the impactof natural and anthropogenic climate change properly evaluated.In this study the longevity of roots at two geographically andclimatically distinct sites were compared to identify the scaleof change that can occur due to environmental differences. Minirhizotrontubes were inserted into swards sown with the same variety ofLolium perenne and Trifolium repens at sites in the UK and Italy.Roots were viewed using a video camera and digitized imagesof roots generated at intervals. From these images the lifehistory of individual roots was determined and compared. Therewere few differences in patterns of longevity between differentspecies at the same site. Major differences, however, were observedbetween roots of the same species at different sites. For example,73% of Trifolium repens roots failed to survive for 21 d inItaly compared to only 29% at the UK site. Similarly, over 84%of Lolium perenne roots failed to survive for more than 21 din Italy compared to 38% in the UK. The endomembrane system of the adaxial epidermal cells of themembranous ligule of the grass Lolium temulentum L. has beenstudied by conventional transmission electron microscopy, andby using zinc iodide-osmium tetroxide impregnation with examinationof thick sections at 100 kV. The components of the endomembranesystem are the nuclear membranes, rough endoplasmic reticulum(present in both cisternal and tubular forms), dictyosomes,dictyosome-derived vesicles, coated vesicles, plasma membraneand tonoplast. Direct continuity between tubular endoplasmicreticulum and the cis face of dictyosomes is demonstrated andthe applicability of the endomembrane concept to this putativesecretory tissue is discussed. Measurements on plants in a temperature gradient tunnel showthat diurnal accumulation of starch in illuminated leaves wasgreatly reduced at temperatures below 8 °C, whereas sucrosesynthesis was less affected under similar conditions. High chillingsensitivity for leaf starch accumulation was observed in a numberof chilling resistant temperate species. Enzymes of sucroseand fructan metabolism from mature leaves of Lolium temulentumwere less strongly inhibited at low temperatures than enzymesinvolved in starch synthesis. These results are discussed inrelation to carbon partitioning in species which grow and aremetabolically active at chilling temperatures. Response of vernalization to low temperature (2C) was studiedin a winter-annual form of Lolium temulentum L., using imbibedseeds, excised mature and immature embryos, shoot apical meristemsand callus tissue. Excised embryos, as early as 5 days afteranthesis, and excised shoot apices could be vernalized as effectivelyas imbibed seeds. Cold treatment of developing embryos withinthe ear, however, appeared to have no vernalizing effect. Plantsderived from callus by somatic embryogenesis showed varyingdegrees of vernalization response. The vernalization response in L. temulentum, as in winter annualcereals, appears to be located in the shoot apical meristemand the vernalized condition can be transmitted to new axillarymeristems formed from it. Daily analysis of anther samples during flower development hasenabled an estimation of the duration of defined developmentalperiods in pollen of the grass Phalaris tuberosa. A similarsequence of pollen development has been established for ryegrass,Lolium perenne, where changes in activity of wall enzymes havebeen followed using quantitative cytochemical methods. Acidphosphatase, an intine enzyme, showed two periods of activity:during the vacuolate period corresponding to deposition of theintine polysaccharides; and in the maturation period correspondingto cytoplasmic activity. Non-specific esterase showed greatestactivity in the parietal tapetal cells until their dissolutionearly in the vacuolate period when an increase in pollen-associatedactivity occurred. These changes provide additional evidencefor the transfer of tapetal proteins to exine sites. The relative assimilatory activity of the inflorescence, itsindividual components, and the leaves of flowering tillers ofPoa annua L. and Lolium perenneL. was determined over the periodfrom inflorescence emergence to seed shedding. The pattern of14CO2 fixation was similar for both species and the inflorescencewas by far the most important assimilatory organ of the reproductivetiller, particularly over the latter period of seed developmentas leaf senescence progressed. With the exception of the seedsall parts of the inflorescence showed significant assimilatoryactivity and the lemmas and paleas accounted for 40–50per cent of the total 14C fixed by the inflorescence in bothspecies. The importance of the grass inflorescence as a photosyntheticstructure is discussed in relation to similar studies on cereals. The effect of light quality on the extension growth of vegetativeshoots and on the final size of their leaves was investigatedin plants of Lolium multiflorum, Sporobolus indicus and Paspalumdilatatum. Three experimental approaches were used, (a) redor far-red end-of-day irradiations of sunlight-grown plants,(b) different red/far-red ratios of white light in a growthroom and (c) sunlight enrichment with radiation of differentred/far-red ratios or with different amounts of far-red lightduring the photoperiod. Plants treated with end-of-day far-redor low red/far-red ratios throughout the photoperiod developedlonger leaves and, as a result, longer shoots. This effect wasmore marked in leaf sheaths than in blades. Tiller extensionand leaf sheath length increased with the amount of far-redadded to sunlight in a simple hyperbolic relationship. Theseresults show that vegetative grass shoots respond to light qualityin a way similar to internodes of dicotyledonous plants. In order to assess the effect on microtubule arrays of slowlypressurising cells over 50 s from 0.1 MPa (atmospheric pressure)to 55 MPa, microtubules in epidermal cells of Lolium rigidumleaves were visualised by immunofluorescent staining and fluorescencemicroscopy. In both control and pressure-treated leaves cellshape, measured as the ratio of cell length and width, can becorrelated to the arrangement of cortical microtubules. Microtubulearrays change from random to organised in cells whose lengthis greater than their width. In untreated leaves, elongatedcells have microtubules aligned predominantly transversely.In pressure-treated leaves, elongated cells have microtubulesaligned predominantly longitudinally. Thus, pressure treatmentresults in the rapid reorientation of organised cortical microtubulesfrom a transverse to a longitudinal orientation. Cocksfoot (Dactylis glomerata L.). perennial ryegrass (Loliumperenne L.) and Italian ryegrass (L. multiflorum Lam.) plantswere grown on deep (75–95 cm) columns of soil in glasshousesand growth rooms with and without irrigation. The species inwhich growth declined least rapidly after water had been withheldwere those which transpired most slowly. During early establishmentin the glasshouse cocksfoot transpired least because of slowroot growth. In the growth room, when root systems were deeperand denser, perennial ryegrass transpired least because of lowleaf water conductance. Results are discussed in relation to(a) drought resistance in the three species; (b) breeding forincreased drought resistance through modifying root distributionand leaf water conductance; and (c) the use of isolated soilcolumns in water relations studies. Structure of the root cap ofLolium temulentum L. and of thechemistry of its secretory product were investigated with conventionaltransmission electron microscopy, zinc iodide–osmium tetroxideimpregnation, and use of protein and polysaccharide localizationtechniques at the ultrastructural level. Structural and functionalinformation concerning the root cap is compared with that previouslyobtained for the membranous ligule of this species. Similaritiesbetween root cap cells and adaxial epidermal cells of the liguleare described and discussed. It is concluded that both systemsare secretory and represent a similar response to the commonproblem of easing the passage of growing organs against thesoil or other plant parts.
