Transgenic Arabidopsis thaliana has been developed which expresses the oryzacystatin mutant OCI86, which is an inhibitor of the major proteinase present in the digestive gland of the slug, Deroceras reticulatum. When fed on leaf tissue from plants expressing this inhibitor the growth of juvenile slugs was significantly reduced by 31% compared with those feeding on control leaf tissue. Furthermore, while surviving slugs did not individually consume less when feeding on leaf tissue expressing OCI86, the total amount of leaf tissue eaten was 50% less, due to reduced survival of slugs. The synthetic cysteine proteinase inhibitors E64 and leupeptin also significantly reduced slug weight gain (by at least 40%) and digestive gland cysteine proteinase activity when administered in an artificial diet, indicating that their antimetabolic effects are due to direct inhibition of gut proteolytic activity. These results suggest that transgenic crop plants expressing phytocystatins could be used to suppress the growth rates of slug populations in the field.  相似文献

Each of 36 observer rats was: (1) exposed to a demonstrator rat that had eaten an unpalatable, cayenne-pepper-flavored diet (Diet Cay), then tested to determine its willingness to eat Diet Cay and (2) exposed to a demonstrator rat that had eaten a palatable diet (Diet NPT) to which the observer had previously learned an aversion, then tested to determine its willingness to eat Diet NPT. In both instances, some observers ate substantial amounts of the diet that their respective demonstrators had eaten, while other observers did not. No consistency was found across the two situations in the relative susceptibility of individual observer rats to social influences on their food choices. In a second experiment, observer rats interacted, at 3 day intervals, with demonstrator rats that had each eaten different diets. After each interaction, all observers were given a choice test to determine their preferences for the diet that their demonstrators had eaten. Again, there was no consistency in the relative strength of individual observer rats' socially induced preferences for diets fed to demonstrators. Stable individual differences in magnitude of susceptibility to social influence on food preference did not account for a detectable proportion of observed variance in diet selection.  相似文献

The effects of adding the molluscicides methiocarb and metaldehyde to pelleted foods on the encounter, acceptance, feeding and post-meal stages of the foraging sequence of the slugs Deroceras reticulatum and Arion distinctus have been studied using two complementary laboratory techniques - video and acoustic recordings. Whereas non-feeding slugs encountered wheat grains at random, slugs which fed on wheat grains or pellets encountered them more frequently because of changes in locomotor pattern and olfactory attraction. Slugs would almost always feed on the first pellet they found, regardless of the presence or absence of molluscicide: thereafter they fed on only one pellet in every four encountered. Far fewer slugs accepted wheat grains. Slugs ate much less from pellets containing molluscicide, and meals were more irregular. The relative amounts of non-toxic pellets of various types which were eaten gave no indication of the relative amounts eaten when molluscicide was added. Meal length did not correlate well with meal size on different pellet types because softer pellets were eaten faster. Most Deroceras fed several times on non-toxic pellets or pellets containing methiocarb, although subsequent meals were shorter than the first meal. Most returned to shelters by dawn. In contrast, metaldehydefed slugs were rapidly immobilised; they seldom fed again, moved little, and few regained shelter. Arion were also inhibited after methiocarb meals but Deroceras were not. Arion distinctus moved less, ate less during a meal, and had fewer meals on nontoxic baits than Deroceras reticulatum.  相似文献

Background

Rats prefer energy-rich foods over chow and eat them to excess. The pattern of eating elicited by this diet is unknown. We used the behavioral satiety sequence to classify an eating bout as a meal or snack and compared the eating patterns of rats fed an energy rich cafeteria diet or chow.

Methods

Eight week old male Sprague Dawley rats were exposed to lab chow or an energy-rich cafeteria diet (plus chow) for 16 weeks. After 5, 10 and 15 weeks, home-cage overnight feeding behavior was recorded. Eating followed by grooming then resting or sleeping was classified as a meal; whereas eating not followed by the full sequence was classified as a snack. Numbers of meals and snacks, their duration, and waiting times between feeding bouts were compared between the two conditions.

Results

Cafeteria-fed rats ate more protein, fat and carbohydrate, consistently ingesting double the energy of chow-fed rats, and were significantly heavier by week 4. Cafeteria-fed rats tended to take multiple snacks between meals and ate fewer meals than chow-fed rats. They also ate more snacks at 5 weeks, were less effective at compensating for snacking by reducing meals, and the number of snacks in the majority of the cafeteria-fed rats was positively related to terminal body weights.

Conclusions

Exposure to a palatable diet had long-term effects on feeding patterns. Rats became overweight because they initially ate more frequently and ultimately ate more of foods with higher energy density. The early increased snacking in young cafeteria-fed rats may represent the establishment of eating habits that promote weight gain.  相似文献

Background

Obesity is associated with excessive consumption of palatable, energy dense foods. The present study used an animal model to examine feeding patterns during exposure to and withdrawal from these foods.

Methods

Male Sprague Dawley rats were exposed to standard lab chow only (Chow rats) or a range of cafeteria-style foods eaten by people (Caf rats). After 1, 4, 7 and 10 weeks of diet in their home cage, rats were subjected to 24-hour test sessions in a Comprehensive Lab Animal Monitoring System (CLAMS). In the first two test sessions, Chow rats were exposed to standard lab chow only while Caf rats were exposed to a biscuit and high-fat chow diet. In the final two test sessions, half the rats in each group were switched to the opposing diet. In each session we recorded numbers of bouts, energy consumed per bout, and intervals between bouts across the entire 24 hours.

Results

Relative to Chow rats, Caf rats initiated fewer bouts but consumed more energy per bout; however, their motivation to feed in the CLAMS declined over time, which was attributed to reduced variety of foods relative to their home cage diet. This decline in motivation was especially pronounced among Caf rats switched from the palatable CLAMS diet to standard lab chow only: the reduced energy intake in this group was due to a modest decline in bout frequency and a dramatic decline in bout size.

Conclusions

Exposure to a cafeteria-diet, rich in variety, altered feeding patterns, reduced rats'' motivation to consume palatable foods in the absence of variety, and further diminished motivation to feed when palatable foods were withdrawn and replaced with chow. Hence, variety is a key factor in driving excessive consumption of energy dense foods, and therefore, excessive weight gain.  相似文献

We examined the effect of methionine deficiency on iodothyronine 5’-deiodinase activity in selenium-deficient rats or selenium-sufficient rats fed sodium selenate or selenomethionine. Forty-two weanling male Wistar rats were divided into six groups and pair fed the respective purifiedl-amino acid-based diets for 4 wk.l-methionine concentrations in the diet were 8.0 g/kg for sufficient rats, and 2.0 g/kg for deficient rats. Selenium concentrations in the diet were 0.5 mg/kg (as sodium selenate or selenomethionine) for selenium-sufficient rats and less than 0.005 mg/kg for selenium-deficient rats. Type I 5’-deiodinase activities were significantly lower in liver and higher in kidney of methionine-deficient rats than in those of methionine-sufficient rats fed either the selenium-sufficient or the selenium-deficient diets. The type I 5’-deiodinase activity in brain was significantly lower in the methionine-deficient rats than in the methionine-sufficient rats fed the selenium-deficient diet. Type II 5’-deiodinase activity in brain was significantly higher in the methionine-deficient rats than in the methionine-sufficient rats fed selenium-sufficient diet as sodium selenate. Both thyroxine and 3,3’,5-triiodothyronine concentrations in plasma were significantly higher in the methionine-deficient rats than in the methionine-sufficient rats. It is suggested that the methionine deficiency affects the 5’-deiodinase activity and thyroid hormones level in the rats.  相似文献

Molecular detection systems used to analyse the gut contents of invertebrate predators have enhanced our understanding of trophic interactions, but do not distinguish between the methods of consumption. Many predators regularly scavenge, which could have profound implications for quantitative analyses of the dynamics of predation. We report the first quantified assessment of the potential error caused by scavenging in post-mortem measurements of predation in a slug-carabid system. An anti-slug monoclonal antibody was able to detect antigens from decayed slugs after surprisingly long periods, significantly longer on relatively sterile peat than on natural soil. On soil the half-life of antibody-detectable slug proteins was 8.2 days while on peat it was 11.5 days. When slugs that had decayed on soil for 100 h were fed to the carabid predator Pterostichus melanarius, slug proteins could still be identified after 6 h (but not 12 h) digestion. Fresh and decayed slug was eaten in equal quantities by the beetles suggesting no aversion to the latter. The results suggest that significant errors may be caused by scavenging leading to inaccurate interpretation of predation rates in the field.  相似文献

The pathogenesis and treatment of nonalcoholic steatohepatitis (NASH) are not well established. Feeding a diet deficient in both methionine and choline (MCD) is one of the most common models of NASH, which is characterized by steatosis, mitochondrial dysfunction, hepatocellular injury, oxidative stress, inflammation, and fibrosis. However, the individual contribution of the lack of methionine and choline in liver steatosis, advanced pathology and impact on mitochondrial S-adenosyl-l-methionine (SAM) and glutathione (GSH), known regulators of disease progression, has not been specifically addressed. Here, we examined the regulation of mitochondrial SAM and GSH and signs of disease in mice fed a MCD, methionine-deficient (MD), or choline-deficient (CD) diet. The MD diet reproduced most of the deleterious effects of MCD feeding, including weight loss, hepatocellular injury, oxidative stress, inflammation, and fibrosis, whereas CD feeding was mainly responsible for steatosis, characterized by triglycerides and free fatty acids accumulation. These findings were preceded by MCD- or MD-mediated SAM and GSH depletion in mitochondria due to decreased mitochondrial membrane fluidity associated with a lower phosphatidylcholine/phosphatidylethanolamine ratio. MCD and MD but not CD feeding resulted in increased ceramide levels by acid sphingomyelinase. Moreover, GSH ethyl ester or SAM therapy restored mitochondrial GSH and ameliorated hepatocellular injury in mice fed a MCD or MD diet. Thus, the depletion of SAM and GSH in mitochondria is an early event in the MCD model of NASH, which is determined by the lack of methionine. Moreover, therapy using permeable GSH prodrugs may be of relevance in NASH.  相似文献

In an attempt to study on metabolic changes in rats fed on an amino acid diet devoid of one branched chain amino acid and of niacin, rats were force-fed a leucine-free, isoleucine-free, valine-free or complete amino acid diet for 3 or 4 days and killed 3 hr after the feeding on day 4 or 5 to observe the body weight changes, the urinary nitrogen and N1-methylnicotinamide (MNA), and liver tryptophanpyrrolase (TPase) and tyrosine-α-keto-glutarate transaminase (TKase) activities.

The excretion of the urinary nitrogen and MNA, TPase and TKase activities, and fat content of livers of rats force-fed these amino acid deficient diets were higher than those fed the complete amino acid diet. It was further confirmed in the present study that changes in TPase activity of rats given diets devoid of one essential amino acid were in the same direction with changes in urinary MNA which was observed in the previous studies on rats given threonine-free, tryptophan-free, methionine-free, lysine-free and complete amino acid diets. However, such metabolic changes in rats fed the leucine-free diet were not so remarkable, compared with those of rats fed the other amino acid deficient diets.  相似文献

We have demonstrated that larvae of codling moth, Cydia pomonella (L) can successfully complete their first instar when fed apple leaves instead of fruit. Larvae fed leaves after hatching maximized their feeding intensity (about 320 g/larva/day) on day 2. Weight gain revealed a stereotypic sigmoid pattern that peaked on day 3. Although the maximum body weight of larvae fed leaves was 70–85% less than for larvae maintained on apples or on artificial diet, 100% of larvae fed leaves molted to the second instar 3–5 days after hatching. Our investigation revealed a diurnal pattern of leaf ingestion, and neonates' feeding intensity decreased significantly during the scotophase. We also demonstrated that monosodium glutamate (MSG) increased feeding on leaves by codling moth larvae. Depending on the duration of the bioassay, and larval age at time of initial exposure, 0.05 mg/ml and 0.1 mg/ml MSG increased apple leaf consumption by 25–60% over leaves alone. The effect of monosodium glutamate was best demonstrated during the first day following hatching. Exposure to MSG also accelerated molting to the second instar. Larvae exposed to MSG initiated consumption of leaf tissue significantly earlier than control neonates. The feeding stimulatory effect of MSG was not observed if exposure to this chemical was delayed until 3–4 h after hatch.The addition of feeding stimulants to pesticides that act via the alimentary tract may reduce the amount of active ingredients needed to maintain the efficacy of these formulations. Here, we postulate that first instar codling moth larvae are potential targets for treatment with pesticide formulations enhanced with monosodium glutamate.  相似文献

饵料对鱼类生长发育和繁殖具有重要影响。为了筛选稀有鲫(Gobiocypris rarus)幼鱼和成鱼阶段最适投喂方式, 实验将出膜5周末(日龄为35 day after hatching)的幼鱼随机分为5个组: A组投喂丰年虫; B组每周前6d (days)投喂丰年虫, 后1d投喂商业化微颗粒S3饲料; C组每周前3.5d投喂丰年虫, 后3.5d投喂饲料; D组每周前1d投喂丰年虫, 后6d投喂饲料; E组一直投喂饲料; 各组均采用饱食投喂策略。每2周统计生长、存活指标, 直至第21周(147 dah), 在17周(119 dah)取材用于观察性腺发育程度。在产卵后统计各组产卵量、孵化率和子代畸形率。结果显示: (1)E组存活率和特定生长率显著低于其他组(P<0.05); (2)从产卵量、孵化率和子代畸形率上看, B组产卵量显著高于其他组(P<0.05); (3)从性腺组织学上看, 不同投喂方法对精巢的成熟度无显著影响, 但投喂过丰年虫的稀有鲫卵巢发育成熟度显著优于E组。研究结果提示:适量加入丰年虫比单一投喂活饵或饲料更有利于稀有鲫的生长和繁殖。建议在标准化养殖过程中, 幼鱼和成鱼期的稀有鲫采取丰年虫与饲料投喂频次比值为6﹕1的方式最佳。  相似文献

The eating pattern is altered by high-fat diet-induced obesity. To clarify whether this is dependent on the fatty acid profile of the diet, the authors conducted two studies on adult female Sprague-Dawley rats fed normal-fat chow or high-fat diets with varying fatty acid composition. Eating pattern and body weight were assessed in rats fed canola-based (low in saturated fatty acids) or lard-based (moderate in saturated fatty acids) diets for 7 days, and in animals fed chow or canola- or butter-based diets (rich in saturated fatty acids) for 43 days. These parameters were also determined when restricted amounts of low-fat canola- or butter-based diets were consumed for 25 days. Early exposure to canola or lard high-fat feeding or prolonged access to canola- or butter-based fat-rich diets (relative to chow feeding) did not alter the normal light-dark distribution of food and energy intake. All animals ingested most of their food during the dark phase. However, feeding the high-fat canola- and butter-based diets produced an altered eating pattern during the light phase characterized by a smaller number of meals, longer intermeal interval, and enhanced satiety ratio, and consumption of shorter-lasting meals than chow-fed animals. Relative to canola or chow feeding, butter-fed animals consumed a lower number of meals during the dark phase and had a higher eating rate in the light phase, but ate larger meals overall. Only butter feeding led to overeating and obesity. When given a restricted amount of low-fat canola- or butter-based diet at the start of the light phase, rats ate most of their food in that phase and diurnal rather than nocturnal feeding occurred with restriction. Slugs feed on some plant species but not on others for reasons that are not yet fully understood. We re‐examined the two largest studies on the preference of Arion lusitanicus for different plant species. Briner & Frank (1998) measured the consumption index of slugs (CIB) based on the dry leaf mass eaten per day per gram of slug mass. Kozlowski & Kozlowska (2009) also measured a consumption index of slugs (CIK) but on the basis of the area eaten per day per gram of slug mass. The two indices showed a moderate positive rank correlation. Briner & Frank (1998) found that slugs preferred annual to perennial plants and crops rather than wild species. On the other hand, Kozlowski & Kozlowska (2009) found no statistically significant difference in slug preference for annuals, biennials or perennials. Using botanical databases we selected plant traits related to slug herbivory and analysed 141 species. We first analysed the two data sets separately and found that CIB was higher for seedlings with a higher relative growth rate although this result was only marginally statistically significant. In addition, we found that annuals were consumed more than perennials and that cultivated plants were consumed more than wild plants. This is in agreement with the conclusions of Briner & Frank (1998). For CIK slugs consumed less from plants with high dry matter content (DMC). The use of classification and regression tree models solves the problem of analysing datasets with missing values. Regarding CIB, the tree models indicated that life cycle was the major branch. Within perennials, cultivated/wild origin of plants was the most important predictor of slug feeding. Within wild perennials slugs ate more from plants with low specific leaf area (SLA), i.e. relatively thick leaves. Within annuals we found the unexpected result that slugs ate more from plant species with either alkaloids or glucosinolates than from species without one of these compounds. For CIK, the results of tree models showed that slugs preferred shade‐intolerant plants to shade‐tolerant plant species. Within the shade‐tolerant group slugs fed more on leaves with low DMC. We only partially understand which factors make plants acceptable to slugs. We discuss this issue along with the pros and cons of the two methods used to calculate CI.  相似文献

The effects of feeding a prepared surimi diet (fish-based) and a prepared pelleted diet (shrimpbased) on the survival, growth and feeding rate of the cuttlefish Sepia officinalis L. were evaluated during a 45-day experiment. One hundred and twenty juveniles of laboratory cultured cuttlefish (74.5 ± 12.36 g) were divided into three treatments and were fed thawed shrimp (control), pellets or surimi. Survival rates on these diets were 95.0%, 67.5% and 22.5%, respectively. Preliminary data indicated that the low survival of cuttlefish fed surimi may have been caused by low levels of copper in their blood (131 vs 244 μg/ml) since copper is required for their respiratory blood pigment, hemocyanin. Instantaneous growth rates were 2.71 % body weight BW/day for cuttlefish fed raw shrimp, 0.33% BW/day for cuttlefish fed pellets, and 0.54% BW/day for cuttlefish fed surimi. The feeding rate of cuttlefish fed shrimp was high (6–8% BW/day). The feeding rate on pellets increased with time (from < 1 to 3% BW/day) but never reached the level for raw shrimp. The feeding rate on surimi increased to equal the rate for raw shrimp during days 1–30 (8 to 9% BW/day) and thereafter decreased (<4% BW/day). In conclusion, there was a major distinction between the palatability of a prepared diet and the ability of that diet to support growth. Surimi was highly palatable but resulted in poor survival, suggesting low nutritional quality. In contrast, pellets were less palatable but produced maintenance growth. Development of practical surimi diets will require supplementation of the surimi with soluble micro- and macronutrients.  相似文献

Summary Maturation of the reproductive tract ofLimax maximus was studied in a natural population using a reproductive organ-to-body weight index. Male-phase maturation, denoted by enlargement of the hermaphrodite gland (gonad), took place mainly in June and July. Female-phase maturation, signaled by growth of the albumen gland, occurred primarily in September. When slugs were subjected to artificial photoperiods consisting of long (LD 168) or short (LD 816) days, female-phase maturation was not found to be significantly affected by photoperiod (Fig. 4). In contrast, male-phase maturation appeared to be induced by a short-day to long-day transition (Fig. 7). The aim of this study was to assess the biological aspects and food preferences of Helicoverpa armigera fed different sunflower genotypes and an artificial diet. Tests were performed under multiple-choice and no-choice conditions to evaluate the biological characteristics of H. armigera. In addition, the biological data obtained were used to determine parameters of fertility life tables. The results showed that H. armigera does not have a feeding preference among the sunflower genotypes tested. The larval period on sunflower ranged from 15.0 to 16.2 days. The maximum fecundity on sunflower was 542.6 eggs/female and that on the artificial diet was 794.5 eggs/female. In general, insects feeding on Helio 250 consumed greater quantities of leaves, had higher survival until the end of the pupal stage, displayed high population growth rates, and had low population doubling times, suggesting that in the field populations will achieve greater population densities when fed on Helio 250. Insects that fed on CF101 consumed smaller leaf areas, had lower survival until the end of the pupal stage, lower fertility rates, and lower population growth rates, and may also have displayed lower population densities and smaller reductions in field productivity. This insect developed best on an artificial diet in comparison to sunflower genotypes studied.  相似文献

The foraging range of adult snails, Helix aspersa, has beenstudied using radio-transmitters. Snails did not return to anexact roost site after foraging, but often remained in the foodpatch, or returned only to the general roost area. Time lapse video films of the slug Deroceras reticulatum madeunder infra-red lighting in arenas were analysed for tracklengthsand degree of turning, in order to simulate slug movements inan unbounded situation. The results suggest that many food itemsare found by random encounter. Slugs usually ate the first food item found, but often ignoredfood items encountered later. If food was scarce, the slugsfed almost every time. Electronic recordings of bites on a wheatflour pellet over 24 hours show that feeding is most intensein the first two and a half hours from starting to feed, andlater meals are both shorter and less regular. Starved slugsdiffered from fed slugs principally by taking a second mealshortly after the first. When given a choice of a more preferredfood (maize pellets) and a less preferred food (pea pellets)in different ratios, the slugs appeared to encounter pelletsat random, but they fed more from the preferred pellets unlessthe ratio was 1 maize: 7 pea.
